    Notizen: ABSTRACT Multiple sclerosis (MS) is an autoimmune demyelinating disease of the central nervous system. A complex genetic etiology is thought to underlie susceptibility to this disease. The present study was designed to analyze whether differences in genes that encode myelin proteins influence susceptibility to MS. We performed linkage analysis of MS to markers in chromosomal regions that include the genes encoding myelin basic protein (MBP), proteolipid protein (PLP), myelin-associated glycoprotein (MAG), oligodendrocyte myelin glycoprotein (OMGP), and myelin oligodendrocyte glycoprotein (MOG) in a well-characterized population of 65 multiplex MS families consisting of 399 total individuals, 169 affected with MS and 102 affected sibpairs. Physical mapping data permitted placement of MAG and PLP genes on the Genethon genetic map; all other genes were mapped on the Genethon genetic map by linkage analysis. For each gene, at least one marker within the gene and/or two tightly linked flanking markers were analyzed. Marker data analysis employed a combination of genetic trait model-dependent (parametric) and model-independent linkage methods. Results indicate that MAG, MBP, OMGP, and PLP genes do not have a significant genetic effect on susceptibility to MS in this population. As MOG resides within the MHC, a potential role of the MOG gene could not be excluded.

    Notizen: Abstract The last two decades have seen clinical and molecular delineation of the different forms of neurofibromatosis. Differentiation of these forms is not just an academic exercise: their natural history, management and genetic counselling are quite different. Of the numerical classifications of neurofibromatosis proposed in the past, only neurofibromatosis type 1 (Nf1) and neurofibromatosis type 2 (Nf2) are now well delineated clinically and have been shown to be distinct at the molecular level. For both forms of neurofibromatosis, patients with clinical generalised disease have been demonstrated to be mosaic at the molecular level, and features of segmental or mosaic Nf1 and Nf2 have been delineated. Other reported forms of neurofibromatosis are rarer; they include Watson syndrome, hereditary spinal neurofibromatosis, familial intestinal neurofibromatosis, autosomal dominant café-au-lait spots alone, autosomal dominant neurofibromas alone, and schwannomatosis, the latter believed to be a variant of Nf2. Further delineation is neeeded for individuals having overlapping features of Noonan's syndrome and neurofibromatosis (the so-called Noonan/neurofibromatosis syndrome) and the syndrome of “multiple naevi, multiple schwannomas and multiple vaginal leiomyomas”. In this article we review the forms of neurofibromatosis which we believe are true clinical entities. Particular attention is given to the neurological manifestations of neurofibromatosis.

    Notizen: Abstract The etiology of non-insulin-dependent diabetes mellitus (NIDDM) is usually explained as a combination of peripheral insulin resistance and impaired beta-cell function. Phosphofructo-1-kinase (PFK1) is a rate limiting enzyme in glycolysis, and its muscle subtype (PFK1-M) deficiency leads to an autosomal recessively inherited disorder known as glycogenosis type VII or Tarui’s disease. It was evaluated whether PFK1-M deficiency leads to NIDDM in humans. A core family of four was evaluated for PFK1-M deficiency by DNA- and enzyme-activity-analyses. All members underwent oral and intravenous glucose tolerance test (oGTT/ivgtt), as well as an insulin sensitivity test (IST) using octreotide. Results: Father (46 years, BMI 22.4 kg/m2) and older son (19 years, BMI 17.8 kg/m5) showed homozygous PFK1-M deficiency, while mother (47 years, BMI 28.4 kg/m5) and younger son (13 years, BMI 16.5 kg/m5) were shown to be heterozygously PFK1-M-deficient on enzyme activity levels. DNA analysis revealed an exon 5-missense-mutation at one allele of all four members, and an exon 22-frameshift-mutation at the other allele of the two homozygously affected individuals. By oGTT the father showed impaired glucose tolerance, and the mother clinical diabetes. By ivGTT both parents and the older son had a decreased first phase insulin secretion, and a diminished glucose disappearance rate. The IST showed marked insulin resistance in both parents and the older son, and moderate resistance in the younger son, previously not described. Conclusion: PFK1-M-deficiency leads to a metabolic state typical for early NIDDM in homozygously affected humans, especially concerning insulin resistance and loss of first phase beta-cell insulin secretion, and may contribute to the manifestation of NIDDM in a subgroup of patients.

    Beschreibung / Inhaltsverzeichnis: Summary A multifactorial etiology underlies the majority of cases of Alzheimer’s disease (AD). Both ill-defined environmental and genetic factors contribute to the development of the disease. Allele ɛ4 of ApoE is a genetic risk factor. Its presence increases the risk of developing AD. However, presence of e4 is neither necessary nor sufficient for the disease to arise. Apart from the common multifactorial forms of the disease, there are rare variants which are inherited as Mendelian traits. To date three genes are known that can be mutated in these rare forms of AD. Of these, mutations in the gene presenilin 1 on chromosome 14 are most frequent. In addition, mutations in the gene presenilin 2 on chromosome 1 and in the amyloid precursor protein gene (APP on chromosome 21) occur in autosomal dominant AD. This article reviews our present knowledge of the genetics of AD and discusses its relevance for patients with AD and their relatives.
    Notizen: Zusammenfassung Der Großteil der Fälle von Alzheimer-Krankheit (AK) hat eine multifaktorielle Ätiologie. Das bedeutet, bisher nicht genauer bekannte Umwelteinflüsse und genetische Faktoren spielen bei der Entwicklung der Krankheit eine wesentliche Rolle. Von seiten der Genetik unterscheidet man bei der AK gegenwärtig genetische Risikofaktroren und Mutationen. Der einzige bisher gesicherte genetische Risikofaktor ist das Allel ɛ4 des Gens für Apolipoprotein E auf Chromosom 19. Dieses Allel erhöht die Wahrscheinlichkeit, an der AK zu erkranken, ist jedoch weder eine notwendige noch eine hinreichende Bedingung. Neben den häufigen Formen mit multifaktorieller Ätiologie kommen seltene Varianten der Krankheit vor, die nach Mendelschen Regeln vererbt werden. Bisher sind 3 Gene bekannt, die bei diesen seltenen, in der Regel früh auftretenden und autosomal dominant vererbten Formen mutiert sein können. Am häufigsten findet sich bei den autosomal-dominanten Fällen eine Mutation im Gen präsenilin 1 auf Chromosom 14, seltener liegen Mutationen im Gen präsenilin 2 auf Chromosom 1 und im Gen des Amyloid- Vorläuferproteins auf Chromosom 21 vor. In diesem Beitrag geben wir eine Übersicht über gegenwärtige Befunde zur Genetik der AK und diskutieren die Bedeutung dieses Wissens für Patienten und deren Verwandte.

    Beschreibung / Inhaltsverzeichnis: Summary Schizophrenia is a genetic complex disease as it does not follow monogenic transmission while non-familial environmental factors have a strong additional impact. A heterogenous, continuous phenotype is transmitted in families which can now be more precisely characterized. Genes coding for proteins with presumed pathophysiological relevance are apparently not playing a major causal role. However, in the last three years several (currently seven) candidate regions have been identified in a replicable manner by linkage studies. These regions are likely to host susceptibility genes for schizophrenia, but none of them has been identified up to now. Given these findings, polygenic transmission has now become very likely. The candidate regions are currently being narrowed down by various promising techniques.
    Notizen: Zusammenfassung Die Schizophrenie gehört zu den genetisch komplexen Erkrankungen, die keinem monogenen Erbgang folgen und bei denen auch nichtfamiliäre Umgebungsfaktoren eine wichtige Rolle spielen. Dabei wird intrafamiliär ein heterogener, quantitativ variierender Phänotyp übertragen, der zunehmend genauer charakterisiert werden kann. Keines der bekannten Gene mit vermuteter pathophysiologischer Relevanz spielt nach den bisherigen Erkenntnissen eine substantielle Rolle. In den vergangenen drei Jahren ist es aber erstmals durch Kopplungsuntersuchungen gelungen, mehrere replizierbare Kandidatenregionen (derzeit sieben) auf dem Genom zu identifizieren, in denen vermutlich Suszeptibilitätsgene für Schizophrenie liegen. Keines dieser Gene wurde jedoch bislang identifiziert. Mit diesen Befunden ist eine polygene Übertragung der Schizophrenie sehr wahrscheinlich geworden. Verschiedene Techniken zur Eingrenzung der Kandidatenregionen werden derzeit erfolgreich angewandt.

    Notizen: Abstract Recently intronic and exonic mutations in the Tau gene have been found to be associated with familial neurodegenerative syndromes characterized not only by a predominantly frontotemporal dementia but also by the presence of neurological signs consistent with the dysfunction of multiple subcortical neuronal circuitries. Among families, the symptomatology appears to vary in quality and severity in relation to the specific Tau gene mutation and often may include parkinsonism, supranuclear palsies, and/or myoclonus, in addition to dementia. We carried out molecular genetic and neuropathological studies on two patients from a French family presenting, early in their fifth decade, a cognitive impairment and supranuclear palsy followed by an akinetic rigid syndrome and dementia. The proband died severely demented 7 years after the onset of the symptoms; currently, his brother is still alive although his disease is progressing. In both patients, we found a Tau gene mutation in exon 10 at codon 279, resulting in an asparagine to lysine substitution (N279K). Neuropathologically, widespread neuronal and glial tau accumulation in the cortex, basal ganglia, brain stem nuclei as well as in the white matter were the hallmark of the disease. These deposits were shown by immunohistochemistry and immunoelectron microscopy, using a battery of antibodies to phosphorylation-dependent and phosphorylation-independent epitopes present in multiple tau regions. In the neocortex, tau-immunopositive glial cells were more numerous than immunopositive neurons; the deeper cortical layers as well as the white matter adjacent to the cortex contained the largest amount of immunolabeled glial cells. In contrast, some brain stem nuclei contained more neurons with tau deposits than immunolabeled glial cells. The correlation of clinical, neuropathological and molecular genetic findings emphasize the phenotypic heterogeneitiy of diseases caused by Tau gene mutations. Furthermore, to test the effect of the N279K mutation and compare it with the effect of the P301L exon 10 mutation on alternative splicing of Tau exon 10, we used an exon amplification assay. Our results suggest that the N279K mutation affects splicing similar to the intronic mutations, allowing exon 10 to be incorporated more frequently in the Tau transcript.

    Notizen: Abstract  Acute leukemia is more common in males at almost every age, and this fact remains unexplained. A study was carried out in northeast peninsular Malaysia, where the population is predominantly Malay, to examine whether there was a difference in ABO blood group distribution between males and females with acute leukemia (AL). The ABO blood groups of 109 male and 79 female patients with AL (98 ALL, 90 AML) were compared with those of 1019 controls. In the control population, 39.7% were group O. Among males with AL, 39.4% were group O, whereas among females with AL, the proportion was 24.1% (p=0.03). The same trend to a lower proportion of group O among females was seen if the group was divided into adult/pediatric or lymphoblastic/myeloblastic groups, though these differences were not statistically significant. If these findings can be confirmed, they suggest the presence of a "sex-responsive" gene near to the ABO gene locus on chromosome 9, which relatively protects group O women against AL, at least in our population. The existence of such a gene might also partly explain why acute leukemia, and possibly other childhood cancers, are more common in males.

    Notizen: Abstract The human T cell leukemia virus type 1 (HTLV-1) Tax is a phosphoprotein, however, the contribution of phosphorylation to Tax activity is unknown. Previous studies have shown that phosphorylation of Tax occurs on serine residue(s), within one tryptic fragment, in response to 4β-phorbol-12β-myristate-13α-acetate, in both mouse and human cells. Studies were conducted in multiple cell lines to identify the specific phosphorylated serines as a prelude to functional analysis. The phosphorylation pattern of Tax was found to be different in 293T and COS-7 cells in comparison with MT-4 and Px-1 cells. However, one tryptic fragment remained consistent in comigration analyses among all cell lines. Using selected Tax serine mutants a tryptic fragment containing a serine at residue 113 believed to be the site of phosphorylation of Tax did not comigrate with the common phosphorylated tryptic fragment. Analysis of selected Tax mutants for ability totrans-activate the cytomegalovirus promoter demonstrated mutation of serine 77 to alanine reducedtrans-activation by 90% compared to wild-type Tax. However, examination of the phosphorylation pattern of the serine 77 mutant demonstrated that it is not the site of phosphorylation. These studies demonstrate the importance of using relevant cell lines to characterize the role of phosphorylation in protein function.

    Beschreibung / Inhaltsverzeichnis: Summary Rhabdomyosarcoma (RMS) is the most important and a very heterogeneous group of malignant soft tissue tumors of childhood and adolescence.The two major subtypes (embryonal and alveolar) share a common myogenic differentiation, but seem to be histogenetically not related. The so-called ’International Classification of Rhabdomyosarcoma’ includes, besides the two major subtypes, the botryoid and leiomyomatous subtypes of embryonal RMS which are associated with a better prognosis and are treated less aggressively according to current protocols. In addition, the solid variant of alveolar RMS is included in the alveolar group of RMS. The identification of the various subtypes is necessary and important because the treatment with the current protocols is also related to histology. Using conventional stains and immunohistochemistry, these subtypes are distinguishable. Genetic analysis can be helpful in the demonstration of t(2;13) or t(1;13) translocations in alveolar RMS. The identification of alveolar RMS with t(1;13) translocation might become important in the future, because this type of translocation seems to be related to a better prognosis as compared to tumors with a t(2;13) translocation.
    Notizen: Zusammenfassung Rhabdomyosarkome stellen eine heterogene Gruppe von ganz verschiedenartigen, histogenetisch wohl nicht zusammengehörenden Tumoren dar. Nach der heute verwendeten „Internationalen Klassifikation” der Rhabdomyosarkome werden neben der Unterteilung in embryonalen und alveoläre Rhabdomyossarkome auch Subtypen des embryonalen RMS identifiziert (botryoider und leiomyomatöser Subtyp), die durch eine günstigere Prognose und durch die Notwendigkeit einer weniger aggressive Therapie gekennzeichnet sind. Durch Einsatz von verschiedenen histologischen und immunhistochemischen Färbungen ist die Identifizierung der verschiedenen Typen der RMS heute möglich und auch zwingend notwendig, da die einzelnen Entitäten nach ganz unterschiedlichen Therapieprotokollen behandelt werden. Der Nachweis typischer molekulargenetischer Veränderungen kann in der Unterscheidung insbesondere von embryonalen und alveolären RMS hilfreich sein. In der Regel ist die Abgrenzung zwischen diesen beiden Entitäten auch an konventionell gefärbten Schnittpräparaten möglich. Die Identifizierung von alveolären RMS mit einer t(1;13)-Translokation könnte in Zukunft eine große Bedeutung haben, da diese genetische Veränderung möglicherweise mit einer günstigeren Prognose assoziert sein könnte als die t(2;13)-Translokation.

    Notizen: Abstract The purpose of our study was to describe the prevalence and the clinical spectrum of monosomy 22q11.2 in a population of patients with pulmonary atresia and ventricular septal defect. We examined all 44 patients with this conotruncal cardiac malformation who presented to our institution from January 1994 until December 1997. The type of collateral lung perfusion was recorded including anomalies of the pulmonary arteries as well as facial and immunological abnormalities. Molecular-cytogenetic testing for a 22q11.2 microdeletion was performed using the probes D22S75 and cHKAD26. Statistical differences were evaluated with the Fisher's Exact Test. Monosomy 22q11.2 was present in ten children (23%) with major aortopulmonary collateral arteries (group 1). The remaining 13 children (29%) with major aortopulmonary collateral arteries (group 2) and all 21 children (48%) with ductus arteriosus (group 3) were negative for this microdeletion. All children in group 1 had facial anomalies, six had mild immunological abnormalities including decreased CD 4+ or CD 8+ cells. Anomalies of the pulmonary vascular bed were significantly more frequent in children of group 1 (9/10) than in children of group 2 (4/13) or group 3 (0/21). Due to these pulmonary vascular anomalies, corrective surgery had been accomplished in fewer children with monosomy 22q11.2 (none in group 1) as compared to 7/13 children in group 2 and 14/21 children in group 3. Conclusion In children with pulmonary atresia and ventricular septal defect, monosomy 22q11.2 is preferentially associated with major aortopulmonary collateral arteries. Due to the higher incidence of pulmonary arterial abnormalities, successful surgical repair will require a different therapeutic approach in most patients with this microdeletion.

    Notizen: Abstract The fission yeast Schizosaccharomyces pombe provides a simple eukaryotic model for the study of cytokinesis. S. pombe cells are rod-shaped, grow mainly by elongation at their tips, and divide by binary fission after forming a centrally placed division septum. Analysis of mutants has begun to shed light upon how septum formation and cytokinesis are regulated both spatially and temporally. Some of the proteins involved in these events have been functionally conserved throughout eukaryotic evolution, suggesting that aspects of this control will be common to all eukaryotic cells.

Recent studies have shown reduced levels of the mainly astroglial glutamate transporter EAAT2 in ALS motor cortex and spinal cord and multiple abnormal EAAT2 mRNA species in ALS brain tissue. One cause of the low EAAT2 levels may be that point mutations in the EAAT2 gene, EAAT2, result in an abnormal unstable protein. To test this hypothesis we analysed EAAT2 in 128 sporadic and 23 familial European ALS cases. No variants within the coding sequence of EAAT2 to affect the protein sequence nor in the consensus splice sites of the flanking intronic sequences were found in any cases, similar to findings in other reports. Frequent polymorphisms within the flanking intronic sequences of both exons 2 and 4 were seen but at similar frequencies in controls. Mechanisms other than mutations within the coding region of EAAT2 must therefore be responsible for the low levels of EAAT2 seen in most cases of ALS.

    Notizen: Abstract Inflammatory bowel diseases (IBD) are complex disorders. While the exact etiology of these diseases remains unknown, recent progress in the epidemiology and genetics of IBD has clearly demonstrated both environmental and genetic factors to play a role in the development of the disease, and it is expected that some risk factors are common for both Crohn's disease (CD) and ulcerative colitis (UC). The environmental factor(s) are associated with the Western way of life in the second half of the twentieth century. Cigarette smoking is presently the best known environmental factor. However, the effect of tobacco is opposite in CD and UC. A familial history of IBD is the most important risk factor for developing the disease, suggesting a genetic predisposition to IBD. This hypothesis has recently been confirmed by the localization of at least two susceptibility loci on chromosomes 12 and 16. These genes seem to play a role in both CD and UC. They must now to be identified.

    Notizen: Abstract  The genetic studies of syndromic craniosynostoses lead to the characterisation of genes that regulate the correct development of the bones of the skull. From these studies, it appears that FGF/FGFR signalling has a crucial role in this problem. Numerous mutations affecting the genes coding for FGFR1, 2 or 3 are responsible for these syndromes. It is interesting to note that some identical mutations produced various different phenotypes, suggesting that other genes modulate the phenotypic expressivity. The other involved genes in these syndromes code for such proteins as Msx2 or Twist that interact in the cellular pathways responsible for FGF action. From these genetic studies, it is now important to establish the role of these proteins during the development of the skull. Msx2 plays a repressive role in osteogenesis, whereas FGFRs act as promoting proteins. In the near future, it will be very important to improve our understanding of these phenomena in order to test specific treatments to prevent the development of such syndromes.

    Notizen: Abstract Alzheimer’s disease (AD) has a significant genetic background manifested as autosomal dominant inheritance in some early-onset families and as familial risk in late-onset cases. Three genes responsible for early-onset autosomal dominant AD have been identified, and one gene, apolipoprotein E, has been confirmed as a susceptibility gene for late-onset forms of the disorder. These findings raise the possibility of genetic testing, either for early diagnosis or prediction. For early-onset autosomal dominant AD genetic testing will have a limited but useful role in confirming diagnosis in established cases and in predictive counselling for relatives; a situation analogous to that for Huntington’s disease. For late-onset AD significant problems remain to be overcome before the advances in molecular genetics have a direct clinical application

    Notizen: Abstract Mitochondrial DNA (mtDNA) gene defects may play a role in the development of maternally inherited diabetes mellitus and deafness (MIDD). A family from Southern Italy who showed maternal transmission of type 2 diabetes mellitus with three individuals affected is described. A 10.4 kb deletion and mutations at nucleotide positions (np) 3243, 7445 and 11778 in the mtDNA of six relatives were sought. The mitochondrial np 3243 mutation of the tRNA Leu (UUR) gene was identified in a boy affected by optic atrophy and mental retardation, as well as in his diabetic mother. No other mutations or deletions were found. Our study points out the variable phenotypic expression of the np 3243 mtDNA mutation. This may suggest the presence of other mitochondrial or nuclear mutations required to modulate the phenotype. A clinical and metabolic follow-up of all family members was necessary to understand the role of the np 3243 mutation, especially in one child affected by optic atrophy and mental retardation. Further studies will be aimed at investigating the prevalence of mutations and deletions of mtDNA in type 2 diabetes mellitus.

    Notizen: Abstract  The self-incompatibility in yellow passion fruit was previously described as homomorphic sporophytic with monofactorial inheritance. Five progenies were obtained by bud-selfing. The plants of these progenies were selfed, reciprocally crossed within each progeny and crossed with known incompatible phenotypes to identify their phenotypic group. Fruit set was evaluated at the 7th day after pollination. Two progenies consisted of two self-incompatible groups, the other three formed three suck groups. The groups were identified as S1, S2, S3, S4, S5 and S6. The results provide evidence that the self-incompatibility of passion fruit is controlled by two loci, the S-gene and another, whose expression needs to be investigated.

    Notizen: Abstract  We report the sequences of 17 primer pairs of microsatellite loci, which we have cloned and sequenced from two genomic libraries of peach [Prunus persica (L) Batsch] ‘Redhaven’, enriched for AC/GT and AG/CT repeats respectively. For ten of these microsatellite loci we were able to demonstrate Mendelian inheritance in a segregating back-cross population; the remainder did not segregate. The polymorphism of the microsatellites was evaluated in a panel of ten peach genotypes, including true-to-type peaches, nectarines and one canning-peach. Fifteen microsatellites (88%) were polymorphic showing 2–4 alleles each. The mean heterozygosity, averaged over all loci, was 0.32 and significantly higher than that reported in the literature for isozymes and molecular markers, such as RFLPs and RAPDs. We have also assayed the cross-species transportability and found that ten microsatellite (59%) gave apparently correct amplification in all Prunus species surveyed, namely P. domestica (European plum), P. salicina (Japanese plum), P. armeniaca (apricot), P. dulcis (almond), P. persica var. vulgaris (peach), P. persica var. laevis (nectarine), P. avium (sweet cherry) and P. cerasus (sour cherry), with three of them also being amplified in Malus (apple). The remaining microsatellites gave less-extensive amplification. Because of their appreciable polymorphism and wide cross-species transportability, most of these new markers can be integrated into the linkage maps which are currently being constructed in peach, as well as in other stone fruit crops, such as almond, apricot, cherry and plum.

    Notizen: Abstract  Six almond progenies, each the product of a cross between a self-compatible and a self-incompatible parent, were analysed for stylar ribonucleases. Proteins were extracted and separated using non-equilibrium pH gradient electrofocusing (NEPHGE), and the gels were stained for ribonuclease activity. Most seedlings showed either two principal bands, interpreted as corresponding to two incompatibility alleles, or a single band. The seedlings were also bagged in the field at flowering time to determine fruit set after selfing, and some were also examined for the growth of pollen-tubes in selfed styles using UV fluorescence microscopy. With very few exceptions, those seedlings showing single-banded zymograms were found to be self-compatible according to field and microscope studies, and those with two bands were found to be self-incompatible. We conclude that the allele for self-compatibility in almond does not code for ribonuclease activity and that the ribonuclease isoenzyme assay is a convenient technique for predicting self-compatibility in segregating progenies. A novel band in two derivatives of ’Ferrastar’ was ascribed to a new incompatibility allele, S 10 .

    Notizen: Abstract  A fundamental requirement of task regulation in social groups is that it must allow colony flexibility. We tested assumptions of three task regulation models for how honeybee colonies respond to graded changes in need for a specific task, pollen foraging. We gradually changed colony pollen stores and measured behavioral and genotypic changes in the foraging population. Colonies did not respond in a graded manner, but in six of seven cases showed a stepwise change in foraging activity as pollen storage levels moved beyond a set point. Changes in colony performance resulted from changes in recruitment of new foragers to pollen collection, rather than from changes in individual foraging effort. Where we were able to track genotypic variation, increases in pollen foraging were accompanied by a corresponding increase in the genotypic diversity of pollen foragers. Our data support previous findings that genotypic variation plays an important role in task regulation. However, the stepwise change in colony behavior suggests that colony foraging flexibility is best explained by an integrated model incorporating genotypic variation in task choice, but in which colony response is amplified by social interactions.

    Notizen: Summary The Maudsley Reactive and Non-Reactive strains have been developed as a model for the study of individual variations in stress-reactivity, and many differences in biobehavioral systems have been found between them. This review discusses limitations of the ‘emotionality’ construct in accounting for differences between the Maudsley strains and offers an alternative, theoretical approach. Amaral and Sinnamon have proposed that the locus ceruleus (LC) plays a stress-attenuating role in mediating behavioral, physiological and neuroendocrine response to prepotent, emergency-provoking stimuli and, building upon this formulation, it is proposed that the LC has been an important focus for gene action in the Maudsley model. It is suggested that the LC of the Non-Reactive strain is more strongly activated by stressful stimuli than the LC of Reactive rats, and is the basis of many of the behavioral and physiological differences between them. Behavioral and biochemical evidence consistent with this proposition is reviewed. Identification of the LC as a target for gene-action in the Maudsley model has an important advantage. It substitutes variations at a specific anatomic location in the brain for a loosely defined construct like emotionality, and the hypothesis is amenable to empirical tests by a variety of experimental approaches.

    Notizen: Summary The variation in immune responses to standard inoculation of the hepatitis-B virus vaccine suggest that host factors influence response in ways that are not presently understood. We studied 25 low/nonresponding health care workers (anti-HBs titer 〈50 IU/l) after the third inoculation of an experimental hepatitis-B vaccine to determine their immune status (through lymphocyte phenotypes) and HLA type. After application of a fourth inoculation, the seroconverting subjects showed only low anti-HBs levels; three male subjects remained anti-HBs negative. Twelve months after the fourth inoculation only 9 of 25 subjects (36%) maintained anti-HBs titer 〉10 IU/l. Almost all subjects had normal B-cell and CD-4 and CD-8 counts and ratios. Relative to other European populations HLA-A-10 (P〈0.05), B-12 (P〈0.025), CW-5 (P〈0.05), DR-3 (P〈0.025), and DR-5 (P〈0.025) were increased, whereas DR-2 (P〈0.05) was decreased. However, after correction of theP-values for the number of HLA antigens determined, these differences were no longer significant. Furthermore, these HLA types were not the same as those reported in other studies (except for DR-3). We suggest that larger sample sizes or even not yet available immunogenetic markers will be required to prove an “immunogenetic background” in low/nonresponders, if it exists.

    Notizen: Summary In the last five years, genetic markers for a large number of diseases have been localised using linkage analysis of DNA polymorphisms in affected families. The site of the genetic defect or defects leading to Type 2 (non-insulin-dependent) diabetes mellitus, a common illness with a major genetic component, remains unknown. This is due, at least in part, to the lack of large well-defined Type 2 diabetic pedigrees suitable for linkage analysis. There are several features of the disease which make large pedigrees difficult to find. The late age of onset of most probands means that informative older generations are often dead, while there is difficulty in detecting disease in younger generations. The diagnostic criteria for diabetes are, as yet, dependent on an arbitrary cut-off along a continuum of plasma glucose. The high prevalence of the disease may also produce problems as, in any given family, diabetogenic genes may be contributed by more than one parent. Varieties of the disease with a well-defined inheritance, such as maturity onset diabetes of youth, are more suitable for linkage analysis but might be due to defects at a different gene locus. Despite these difficulties, once large well-defined pedigrees have been found, linkage analysis using both candidate genes and random highly polymorphic markers is the strategy most likely to find genetic markers for the disease.

    Notizen: Summary The possibility of linkage between the human insulin receptor gene locus and diabetes was examined in three Type 2 (non-insulin-dependent) diabetic families and one family with maturity onset diabetes of the young. Insulin receptor gene haplotypes were established using BglII, Rsal and Sstl restriction enzyme digests of genomic DNA from all available family members. The digested DNA was subjected to agarose gel electrophoresis, Southern blotted, and hybridised to 32P-labelled human insulin receptor gene cDNA. In the pedigree with maturity onset diabetes of the young, formal linkage analysis allowed exclusion of close linkage between the insulin receptor locus and diabetes (logarithm of the odds for linkage versus non-linkage was −5.35 at recombination fraction of 0.01). This confirms the absence of linkage between insulin receptor and diabetes which has been reported in two similar pedigrees. In the three Type 2 diabetic families there were a minimum of 4 recombinants between the insulin receptor locus and diabetes, which makes a direct role for insulin receptor defects unlikely. The importance of using realistic estimates of penetrance when performing linkage analysis in a disease with a late age of onset is emphasised. In contrast to the one previous linkage analysis study of the insulin receptor gene, no specific association of diabetes with the rare Sstl Sl(-) allele was observed in either the maturity onset diabetes of the young or the Type 2 diabetic families.

    Notizen: Abstract Two siblings with typical features of the Cockayne syndrome were studied at autopsy. Many glomeruli revealed a paucity of capillary loops and had thickened capillary walls. Some glomeruli with advanced lesions showed collapse of the glomerular tufts or complete hyalinization. Atrophy of tubules and interstitial fibrosis were also observed. There were no significant arteriosclerotic changes in the vessels. Ultrastructural studies demonstrated thickened glomerular basement membranes with bends and folds. These histopathological findings are different to those previously reported with the exception of the 1966 report by Ohno and Hirooka.

    Notizen: Abstract C57BL/6J and DBA/2J mouse strains have been characterized as morphine preferrers and avoiders, respectively (Horowitz et al. 1977). Previously, sweetened morphine solutions were presented with a water alternative, primarily with male subjects. Because sweetness may affect the endogenous opioid system and rodents have shown strain and sex differences in taste preferences, this study looked for strain- and gender-related taste preferences that might have affected opiate consumption. Preference for sweetened and unsweetened morphine and etonitazene was compared across gender and strain. In all choice tests, the control was a similar tasting quinine sulphate solution. Under these conditions, C57BL/6J mice continued to show strong preference for morphine. However, DBA/2J mice drank approximately equal amounts of morphine and quinine solutions, rather than avoiding morphine as when water was the alternative. Both strains appeared surprisingly indifferent to the synthetic opioid etonitazene, compared because it is potent at concentrations having barely perceptible bitterness. This raises the possibility of unexpected differences in post-ingestional effects between morphine and etonitazene. Contrary to reports of gender differences in sweet preference in rats, none were found in either strain of mouse. Neither were there any significant sex differences in opiate preference in either strain. C57 mice preferred sweetness more than did DBA mice.

    Notizen: Abstract Four strains of mice (C57, BALB, DBA, C3H) were used to determine whether genetic factors influence the effects of lithium on hypoactivity induced by a low dose of the alpha2-adrenoceptor agonist clonidine (0.2 mg/kg). Lithium was administered in the diet for 3–4 weeks at a dosage that produced average serum lithium levels of 0.58–0.66 mmol/l. Locomotor activity was reduced by either clonidine or by lithium given alone. When combined, however, lithium attenuated the activity-suppressant effects of clonidine, and that action was influenced by genetic factors. The findings suggest that genetic differences in alpha2-adrenoceptors play a role in behavioural effects of lithium.

    Notizen: Abstract Low doses of ethanol (EtOH) stimulate activity in an open field in many strains of laboratory mice. We are selectively breeding two lines of mice to exhibit a large (FAST) response on this test, and two other lines to exhibit a small (SLOW) response (Crabbe et al. 1987). The lines initially diverged in response to EtOH, but despite continued selection pressure, the difference between each pair of FAST and SLOW lines has not increased over generations as much as expected. Our practice has been to test animals on the 1st day after saline injection, and repeat the test after EtOH injection 24 h later. Lister (1987) recently demonstrated that the order in which an animal was exposed to EtOH and saline influenced the magnitude of the response to EtOH, with animals tested initially after EtOH having greater stimulation. Middaugh et al. (1987) recently demonstrated that the magnitude of EtOH stimulation was greater under conditions of relatively bright light than under dim light. Using non-selected Swiss mice, the current experiments essentially confirmed Lister's findings. Using FAST and SLOW mice, the predictions of both groups were tested. Both hypotheses were confirmed. Additionally, these experiments demonstrated that the magnitude of the difference between FAST and SLOW mice was greater under bright light than under dim light. The line difference was also greater when tested in the EtOH-Saline order. In experiments with Swiss mice, the possible role of peritoneal irritation in the EtOH effect was eliminated, and the optimal dose and time for demonstrating the effect was determined. These experiments confirm the importance of lighting condition, order of testing, dose, and route of administration in eliciting EtOH-stimulated open field activity in mice. They demonstrate a genotype-environment interaction, since the magnitude of difference between genetically selected lines varied as a function of the testing parameters chosen. Finally, they indicate that the differences between FAST and SLOW lines in sensitivity to EtOH generalizes to several environmental conditions. We interpret this to mean that the various EtOH-induced activation traits represented by these different environmental and testing conditions are genetically correlated.

    Notizen: Summary Within the broader context of our investigations into the heredity of the human EEG, we analysed the EEGs of 28 pairs of monozygotic and 21 pairs of dizygotic twins who were separated as infants and reared apart. The principal goal of this study was to determine the degree to which environmental factors possibly influence the development of a person's EEG. Monozygotic twins reared apart were, with respect to their EEGs, only slightly less similar to each other (if there is any difference at all) than the same person is to himself over time. For dizygotic twins reared apart, we verified the findings of our previous study, namely, that the average within-pair similarity of EEGs estimated from a sufficiently representative sample of fraternal twins was significantly higher than the average inter-individual similarity of EEGs obtained from unrelated persons. The results on both monozygotic and dizygotic twins, yielded conclusive proof that the individual EEG pattern is predominantly determined by hereditary factors.

    Notizen: Summary A 4-year-search for spinal muscular atrophies (hereditary motor neuropathies, HMN) in Benghazi, Libya, yielded a total of 24 patients, among whom 18 were index cases. This group comprised 6 acute infantile, 12 chronic childhood, and 3 each with adult-onset proximal, and distal forms of the disorder. Distal HMN constituted 12.5% of the total cases. The crude average annual incidence of acute infantile HMN was 0.3/100,000 total population and 1/12,500 births in Benghazi. The crude prevalence rates of chronic childhood, adult-onset proximal, and distal types of HMN were 2.3, 0.6, and 0.6/100,000 respectively. The segregation ratios, 0.26 for acute infantile HMN and 0.24 for chronic childhood HMN, suggested autosomal recessive inheritance. The consanguinity rates among parents of cases and the population did not differ significantly.

    Notizen: Summary Restriction Fragment Length Polymorphisms (RFLP) have been identified between widely distant cultivars (‘Minsoy’ and ‘Noir 1 ’) of soybean Glycine max (L.) Merrill. Using as probes randomly chosen clones of DNA, one in five probes revealed a polymorphism. More than half of these polymorphisms appear to result from rearrangements of the genomic DNA. Twenty seven markers were analyzed for linkage in F2 plants. Eleven of these markers were contained in four linkage groups. Five cultivars were compared in a search for new alleles. When RFLP markers corresponding to low copy DNA were used to analyze three other cultivars — ‘Sooty’, ‘Forrest’ and ‘Mandarin (Ottawa)’ — few new alleles were found. Using these probes, five different markers could be used to differentiate the five cultivars. Complex probes, which correspond to repeated DNA, revealed different polymorphisms in different cultivars and a single such probe could be used to distinguish the five cultivars from each other.

    Notizen: Summary Little is known about the biochemical correlates of selection for growth in farm or laboratory animals, or the identity of the gene products affected or produced by ‘trait-genes’. Modern broiler chickens have about 8-fold greater breast muscle mass than layer chickens at 7 weeks of age and over 2-fold greater breast muscle mass than their 1972 counterparts. This increase in muscle mass is associated with over 20-fold higher levels of ornithine decarboxylase (ODC) in broiler chickens at 1 week of age as compared with layer strain chickens; there is a comparable increase in a relaxed-selection strain of broilers. The increase in ODC levels is larger than the differences in muscle or body weight between broilers and layers at 7 weeks of age, occurs at an age when there is no difference in weights between the strains and precedes the major growth spurt. Increases in ODC levels and hence polyamine synthesis have been associated with, and usually precede, rapid growth and cell proliferation in a wide range of cell types and organisms in response to many different stimuli. Therefore, the correlation of ODC levels with genetic differences in muscle growth make it worth investigating the control of ODC gene expression in these strains.

    Notizen: Summary Separations of kafirin and alcohol soluble glutelin proteins by reversed-phase high-performance liquid chromatography (RP-HPLC) from 7 inbreds and one hybrid of sorghum [Sorghum bicolor (L.) Moench] and one source of Johnsongrass [Sorghum halapense (L.) Pers.] were compared. Objectives were to assess the stability of protein profiles for seed sources produced at different locations and in different environments to examine the potential of RP-HPLC to provide genotypic profiles for sorghum. Analyses of variance data showed that levels of variation due to environments and locations were small; the majority of variation (93%) was among genotypes. Associations among inbreds revealed by multivariate and cluster analysis showed similarity with those that would be expected on the basis of pedigree. A chi-square analysis showed no deviation in the hybrid profile from the expected 2∶1 ratio of peaks from the female and male inbred parents, respectively. Improvements in the ability to correctly assign common peaks are necessary before associations among numerous sorghum genotypes can be reliably demonstrated by analysis of data from reversed-phase high-performance liquid chromatography (RP-HPLC).

    Notizen: Summary The effect of genotype on maternal haploid plant production in maize was studied. The frequency of gynogenetic plants when “Stock 6” was used as pollinator varied according to the female parent genotype. No simple relation was observed between genotypic aptitudes for gynogenetic and androgenetic development, which occured after pollination of “W23” plant carrying the “indeterminate gametophyte” gene. Furthermore, the population NS, a favorably responsive genotype to anther culture, does not exhibit exceptional ability for in vivo gynogenesis. The effect of inbreeding and the influence of maternal haploid origin suggest that specific genes control maternal haploid initiation and development. However, gynogenetic development is not limited to a particular genotype. The frequency of maternal haploids may be increased by using specific pollen parents. Attempts were made to select for a high haploidyinducing trait and the present study reports the successful development of lines that can be utilized as pollen parents to induce haploids for experimental purposes and breeding programmes. When an inbred line “WS14”, derived from the cross W23 x Stock 6, was used as pollen parent, 2%–5% maternal haploids were obtained according to the female parent genotype. A high haploidy-inducing potential is a heritable trait and may be controlled by a limited number of genes. Genetic determination of the haploidy-inducing character was examined in relation to the efficiency of the selecting method and the mechanisms involved in the origin of maternal haploids.

    Notizen: Summary A total of 70 cnx mutants have been characterized from a collection of 211 nitrate reductase deficient (NR-) mutants isolated from mutagenized Nicotiana plumbaginifolia protoplast cultures after chlorate selection and regeneration into plants. They are presumed to be affected in the biosynthesis of the molybdenum cofactor since they are also deficient for xanthine dehydrogenase activity but contain NR apoenzyme. The remaining clones were classified as nia mutants. Sexual crosses performed between cnx mutants allowed them to be classified into six independent complementation groups. Mutants representative of these complementation groups were used for somatic hybridization experiments with the already characterized N. plumbaginifolia mutants NX1, NX24, NX23 and CNX103 belonging to the complementation groups cnxA, B, C and D respectively. On the basis of genetic analysis and somatic hybridization experiments, two new complementation groups, cnxE and F, not previously described in higher plants, were characterized. Unphysiologically high levels of molybdate can restore the NR activity of cnxA mutant seedlings in vivo, but cannot restore NR activity to any mutant from the other cnx complementation groups.

    Notizen: Abstract The human-type ABO blood groups were determined for 94 families of the squirrel monkey which included 151 animals. Four phenotypes of ABO blood groups (A, B, AB, and O) were detected. Family analysis revealed that the human-type ABO blood groups in this species were governed by three alleles, codominantA andB and silentO. There were intraspecific differences in the distribution of phenotypes and gene frequency among three populations imported by different routes at different times. The usefulness of ABO blood groups for defining the genetic variability of a squirrel monkey breeding colony through successive generations is discussed on the basis of the difference in distribution of ABO blood groups between wild-originated parental and its first colony-born populations.

    Notizen: Conformational preferences of model modified nucleic acid base N6-(N-alanylcarbonyl) adenine, ac6Ade, have been investigated using the quantum chemical PCILO (perturbative configuration interaction using localized orbitals) method. The multidimensional conformational space has been searched using selected grid points formed by combining the various torsion angles that take favored values derived from energy variation with respect to each torsion angle individually.The preferred molecular structure is stabilized by an intramolecular hydrogen bond from N(11)H of the amino acid to N(1) of the adenine. The observed crystal structure conformations for the naturally occurring, anticodon adjacent, threonyl analogs, tc6Ade, correspond to the predicted most stable conformation for the model modified base ac6Ade. Three stable, low energy conformations differing in the orientations of the carboxyl group and the amino acid side chain are predicted within 1 kcal/mol of the most stable structure. The possible bifurcated hydrogen bonding of N(11)H with N(1) and either of the carboxyl oxygens is of minor significance.The indicated orientational flexibility for the carboxyl group and the amino acid side chain may enable convenient probing of the molecular environment, in the vicinity of the anticodon in tRNA, by the amino acid substituent, with only modest changes in energy stabilization.

    Notizen: By using the n + (1/2)l filling rule of the atomic Aufbau principle, where n is the principal quantum number and l is the azimuthal quantum number, a new periodic table is presented, its periods having, in order, 8, 18, 18, 32, 42, 50, … elements. The mentioned rule is proposed instead of the n + l rule (or Madelung's rule) which constitutes the quantum mechanical basis of the current periodic table and predicts periods having, in order, 2, 8, 8, 18, 18, 32, 32, 50, … elements. The new periodic table is called “regular” because its groups are formed according to a single rule (namely, the first elements of each period are placed in the same order as the elements of the preceding period), in contrast with the current periodic table, where no simple rule can be applied for the same purpose. The most characteristic feature of the regular periodic table is the fact that its groups are also related in a periodic manner.

    Notizen: A rigorous approach to the study of the quality of theoretical descriptions of atoms and molecules is proposed. Theoretical descriptions, i.e., collections of atomic or molecular properties obtained via quantum chemical methods, are associated with information quantities defined with respect to reference theoretical or experimental data. In this way the comparison of two theoretical descriptions is reduced to the comparison of the respective information quantities. An increase in information is interpreted as improvement of the theoretical description. The aforementioned approach should be useful in problems related to classifications and systematic improvements of approximate wave functions and consequently for comparing the performance of quantum chemical methods in calculations of atomic and molecular properties. This approach should also constitute a powerful tool for the construction of suitable basis sets for use in calculations of atomic and molecular properties.

This method is computationally very fast, with an execution speed proportional to N3, Where N is the number of orbitals present.     Notizen: In a dissociation attachment experiment of water, three peaks were observed at 7,9, and 12 eV. The origin of the third peak has been believed to be 2B2. However, the calculated energy of this state is 0.6 eV higher than the experimental value. This discrepancy is quite large compared with the case of the lower two peaks. In this study we propose new candidates for resonant states responsible for the third peak. The configurations considered are (3a1)-1(3pa1)2, (3a1)-1(3pb1)2, (3a1)-1(3pb2)2, (3a1)-1(3pa1)1(3pb1)1, (3a1)-1(3pb2)1(3pa1)1, and (3a1)-1(3pb2)1(3pb1)1 which have the parent state (3a1)-1(3pa1)1, (3a1)-1(3pb1)1, or (3a1)-1(3pb2)1. The energy levels arising from these configurations are calculated by a method of configuration interaction. A Few resonance states, which could be responsible for the third peak, are found. New decay process of these states are proposed.

This approach allows one to overcome some conceptual difficulties of ligand-field theory and to derive effective parameters of d-d electron repulsion of a bound transition-metal in a proper way. The two ligand-field mechanisms of the nephelauxetic effect - the central field covalency and the symmetry-restricted covalency - are clarified and generalized. In addition, a new factor, important for the nephelauxetic effect is found: its size-dependence on the region of the topological atom.

The employed SCF variants allow for the treatment of atomic species up to bromine under the inclusion of the first (i.e., 3d) transition metal series. Band structure investigations of 2D and 3D materials containing more than 30 atoms per unit cell are feasible by the present SCF HF CO formalism. The theoretical background of the computational scheme is given in this contribution. Special emphasis is placed on physically reliable truncation criteria for the lattice sums, the adaptation of the crystal symmetry in k space, as well as the suitable choice of domains in Brillouin zone (BZ) integrations required in the determination of charge-density matrices. The capability and limitations of the semiempirical SCF HF CO approach is demonstrated for some simpler solids by comparing the present computational results with those of ab initio CO schemes as well as conventional numerical methods in soid-state theory. The employed model solids are graphite and BN (2D and 3D networks for both solids) as well as diamond, silicon, germanium, and TiS2.

    Notizen: Quantum mechanical models to treat long distance electron transfer are being developed. The model is based on the theory of R.A. Marcus. Our contribution is in the calculation of the electron coupling factor k. Estimations of the latter number, as well as the bond and solvent relaxation energies, λi and λo, respectively, are necessary to be able to calculate the rate constant for a reaction of the conductivity in an electric field. k may be approximately calculated from orbital energy differences at avoided crossings between orbitals localized in different parts of the system. A novel spectroscopic NDO method is suggested in which one may include any atom of the periodic table. Another problem discussed is the inclusion of electronic relaxation effects of the solvent or protein in the calculation. Applications are made to systems where metal ions are connected by organic bridges of different kinds such as dipyridine with coplanar and perpendicular pyridyl groups. As expected the electronic factor depends strongly on the conformation of the bridge. A strong conformational dependence is also obtained for a saturated bridge of the type NH2 · (CH2)n · NH2. In another study we use an α helix as a bridge between two metal ions. If one glycine in this α-helix is substituted by phenylalanine the electronic factor increases by factors of 1.5-10. It is suggested, however, that larger enhancement factors are possible if an aromatic group is positioned in a favorable way. The CNDO/S method is used to study the charge separation process in a bichromophoric molecule and in the reaction center (RC) of Rhodopseudomonas viridis. In those cases where the electronic coupling is large enough for the charge transfer states to be seen in the spectrum, the calculated results agree well with the experimental ones, but suggest a novel assignment. The CNDO/S results verify that electron transfer is possible through saturated spacers. In the special pair of RC the S1 state is calculated at approximately the correct position. Like the ground state, it has a delocalized character.

    Notizen: A class of thromboxane antagonists exists where the prostaglandin side chain containing the C16 hydroxyl moiety is replaced by a phenyl ring, and the bridged six-membered pyranose moiety by cyclohexane, pyranose and dioxane ring systems. Analysis of antagonist potency data in terms of a binding constant model previously used for membrane bound receptor-drug interactions shows that the major patterns of antagonist potency are governed as much by axial/equatorial conformer preference of the phenyl moiety and its orientation as by electrostatic effects of the aliphatic ring oxygen atoms. The conformational restriction of the two substituted side chains of the σ-bonded 6-membered ring is shown to be a primary requirement for binding to thromboxane receptors, and a quantitative separation of electrostatic and conformational components in the potency data is attempted.

    Notizen: Ab initio calculations have been performed to investigate hydrogen bonding and ion-molecule association in complexes of H2O with the neutral, protonated, and Li+ complexes of N-formylformaldehyde and N-formylformamidine. In the complexes with the neutral bases, H2O assumes an in-plane bridging position in the amide and amidine regions. The most stable complex is the bridging N-formylformamidine-H2O complex in the amidine region, which has an MP2/6-31 + G(d,p) binding energy of -9 kcal/mol. Hydrogen bonded complexes of H2O with the oxygen-protonated bases have open structures with the protonated bases as proton donors, and binding energies ranging from -16 to -24 kcal/mol. Nitrogen protonation of N-formylformamidine leads to an equilibrium chelated hydrogen bonded structure with a stabilization energy of -21 kcal/mol. When Li+ associates with these bases at a carbonyl oxygen, hydrogen-bonded bridging structures with H2O reappear, and wobble complexes exist in the amide and amidine regions of N-formylformaldehyde and N-formylformamidine. These complexes have binding energies of -13 to -14 kcal/mol. However, the most stable comples has H2O directly bonded to Li+, with an MP2 binding energy of -30 kcal/mol. No hydrogen bonded structures of H2O with N-formylformamidine exist in the amide region when Li+ associates with this base at the C=N group. Hydrogen bond energies computed at the single-determinant Hartree-Fock level with the 6-31G(d) basis set approximate correlated MP2/6-31 + G(d, p) energies to within 1 kcal/mol for all of the neutral and charged complexes. However, when H2O is bonded to Li+, HF6-31G(d) association energies overestimate MP2/6-31 + G(d, p) energies by 3 kcal/mol.

    Notizen: Whereas localization of orbitals has long been a tool for a semiclassical interpretation of chemical properties, it is in fact electron delocalization that is a fundamental property of quantum mechanical molecules. A mathematically well-defined measure is suggested for the degree of delocalization of molecular orbitals. It is shown that an orbital set of maximum delocalization exists for which the degree of delocalization depends on the charge distribution of the molecule. Hartree-Fock canonical orbitals are definitely more localized than the most uniformaly distributed MO's giving an equivalent description of the molecule. The changes in the geometrical shape of molecular orbitals are studied passing (quasi-) continuously from the strongly localized description towards the most delocalized picture. In the case of charge-inhomogeneities even the most delocalized orbitals remain rather compact. The degree of maximum delocalization may be correlated with chemical properties such as reactivity. The shape distortion of MO's under the perturbing effect of other ions and small molecules is investigated in several examples.

Calculations were performed on the Be atom using LIP bases containing several of the natural spin orbitals from an accurate nonrelativistic 650-term configuration interaction (CI) wavefunction. Calculations were also performed using a basis set of 6 s-type orbitals optimized so that the lowest eigenvalue, d1, of the overlap matrix of products is maximized and the energy of a small CI wavefunction is minimized. The value of d1 in the optimized basis is 1 × 10-7. The density and the potential energy obtained from the 650-term CI wavefunction were accurately reproduced by the density matrix expressed in the optimized LIP basis, but the kinetic energy was somewhat less accurate.

    Notizen: The need to perform a numerical integration of the exchange-correlation functional because of its non-analyticity severely complicates the accurate application of local-density functional methods to molecules and clusters. The optimal choice of grid points for this integration and the estimation of the error made by the choice are subtle considerations. In particular, because the position and/or weighting of each grid point must change when the nuclear positions change, these errors are most noticeable when different geometries are compared. We have determined a method of grid point selection and weighting that reduces these errors. We have also determined a simple method of estimating the extent of the error made in the particular density of points used for the grid. These results are illustrated for a selection of small molecules.

    Notizen: The COLUMBUS program system is a collection of Fortran programs for performing general multireference single- and double-excitation configuration interaction (MRSDCI) wave function optimization based on the graphical unitary group approach. The program system also includes integral generation, SCF and MCSCF orbital optimization, integral transformation, and wave function analysis programs. The original program system was written in 1980 to 1981. Since that time, it has evolved into one of the most popular MRSDCI program systems used in the computational chemistry community. The discussion of this evolution will include the exploitation of efficient matrix-matrix and matrix-vector product computational kernels, the use of generally contracted symmetry-adapted orbital basis sets, general Hamiltonian diagonalization procedures, energy-based internal walk selection, flexible DRT specification, improved coupling-coefficient evaluation methods, coupled-pair functional and multireference CPF capabilities, and density matrix construction. The numerous versions of the program system that are maintained at different sites and on different computers are now in the process of being merged. The source code for this combined version will be made available to the computational chemistry community. The source code for a specific computer may be generated from the source code for another computer by a single pass through a simple filter utility that is included with the program system. The directly supported computers will initially include various models of VAX, Cray, FPS, IBM, CDC, and ETA machines with the addition of other machines shortly thereafter. The ongoing developments of the COLUMBUS system that are discussed include a new method for computing analytic energy gradients for MRSDCI wave functions. This effective-density-matrix based method avoids the “coupled perturbed MCSCF” solutions for each coordinate direction, avoids the transformation of any derivative-integral quantities from the AO to the MO basis, avoids the transformation of the coupling coefficients from the MO to the AO basis, allows a subset of the MCSCF doubly occupied orbitals to be frozen in the CI wave function, and allows the MRSDCI wave function to be generated from general reference CSFs that are not necessarily related to the MCSCF expansion CSFs.

    Notizen: The semiempirical Atoms-in-a-Molecule (AIM) hardness matrix, η, is defined, using the usual finite difference formula, ηii = Ii - Ai, for the diagonal AIM hardness and the Ohno formula, ηij = 1/(a2 + R2ij)1/2, for the off-diagonal AIM hardness. The Ohno formula is shown to exhibit the correct asymptotic behavior and satisfies the relevant stability criterion. The normal displacements in the AIM electron populations are examined for pyrrole and N-methyl pyrrole, and their relation to the polarization channels is briefly discussed. The new AIM hardness matrix is also tested by comparing the predicted global hardnesses with the corresponding experimental finite difference data for selected diatomics and triatomics. Finally implications of the hardness combination rules are examined and the corresponding softness combination rules are used to calculate the regional and global softnesses of selected molecules. We examine how the regional softnesses reflect known trends in selectivity of protonation of five membered heterocycles, and comment on the signs of the AIM fukui function and the Hard-Soft-Acids-and-Bases principle.

    Notizen: The application of molecular ab initio methods to investigate the electronic structure of localized impurities in semiconductors requires the study of the convergence of the results with increasing cluster size. Here we compare results for interstitial Ti in silicon, obtained with clusters of increasing size: TiSi10H16, TiSi30H40, and TiSi66H64. These clusters contain one, two, or three shells of silicon atoms, respectively, centered around Ti at a Td interstitial site. The hydrogen atoms serve as saturators of the dangling bonds. The Si core electrons are replaced by an effective potential. The calculations are based on open shell RHF theory and limited CI extensions. The charge distribution in the central part of the three clusters is very similar. In the clusters the partially occupied orbitals are much more delocalized than the 3d orbitals in the free ions. The total impurity-induced electronic charge, however, is quite localized, due to the compensating response of the Si closed shell density. Ionization of the impurity also causes a compensating response of the Si closed shells: only about 10% of the density difference is in the impurity region and the major part is behind the outermost shell of Si atoms. Transition metal associated (3d-like) excitation energies are not very dependent on the cluster size, and the relative ordering of the lowest lying states remains unchanged. Impurity associated ionization energies decrease considerably due to the extra relaxation offered by the additional shells of Si atoms. Our results indicate that a reliable description of interstitial transition metals in silicon can be provided by calculations on reasonably small clusters: Si30H40 is sufficiently large.

    Notizen: The titled isomerization has been studied at the HF/3-21G level. The changes in thermodynamic functions, equilibrium constant K(T), conversion efficiency C(T), and the rate constant k(T) for the isomerization have been calculated using our energies of the reactant (R), transition state (TS), and product (P), their harmonic vibrational frequencies, and the classical and corrected barrier (by vibrational zero-point energies). In the range of temperature, 300-1200K, ΔH 〈 0, ΔS 〉 0 and ΔG 〈 0; i.e., it is exothermic, increasing in the entropy, and spontaneous reaction. ΔS≠ 〈 0, log A is about 12.0 ±0.15 (s-1), but the k(T) is from 2.627 × 10(-33) to 6.933 × 10(8) (5-1); i.e., it passes through a tight TS, the A factor changes little, but the rate constant varies rapidly with the temperature. These results agree with the fact that the isomerization occurs readily at high temperature. Furthermore, Newman's mechanism, 2SiH2 → (H2SiSiH2)* → (HSiSiH3)* → Si2H2 + H2, is supported. Similarly, the isomerization, HDSiSiH2 → HSiSiDH2, is also studied.

    Notizen: A complete active space MCSCF (multi-configuration self-consistent field) scheme (CASSCF) using relativistic effective potentials followed by configuration interaction and relativistic CI calculations provides a very good description of the electronic states and potential energy surfaces of transition metal dihydrides. Such calculations are of considerable value not only in our understanding of the transition metal-hydrogen bonding but also in the prediction of the barrier to insert the metal atom into the H2 bond. All-electron CASSCF/CI calculations are carried out on twelve electronic states of CoH2. Comparable RECP-CASSCF/CI calculations are also carried out to show that the RECP-CASSCF/CI calculations provide an accurate method for the investigation of transition metal compounds. The general method of RECP-CASSCF/CI calculations for molecules containing heavy atoms is described. The calculations on CoH2 are compared with similar calculations on ScH2, YH2, PtH2, and PdH2. A critical comparison of all these transition metal hydrides reveals that in general the low-spin excited metal atom inserts into H2 spontaneously while the high spin ground state atom has to surmount a large barrier.

    Notizen: The problems involved in determining the mechanisms of reactions by quantum mechanical calculations are discussed. Various precautions must be taken if the results of any calculation are to be chemically meaningful. Ab initio studies of reactions must also be carried out at a high level, using large basis sets and allowing for electron correlation. Such calculations are usually restricted to the simplest example of a reaction, which is often untypical. More information can usually be obtained at far less cost through AM1 calculations for a number of examples of a reaction for which experimental data are available. These points are illustrated by recent studies of the Cope rearrangement.

    Notizen: The aim of this article is to illustrate the use of symbolic computation in solving some problems in quantum theory. Emphasis will be given to the possibility of manipulating expressions symbolically and to the interface of numerical and algebraic languages. The illustrate the use of the scientific computation language Maple as a research tool, we present our work on lower energy bounds for anharmonic oscillators and on the spherical Zeeman effect. Another example consists in the perturbation calculations of the N-dimensional hydrogen atom, keeping the quantum labels in general form. Further, we describe the interface of Maple, Fortran, and the typesetting language, Troff. The basic feature is illustrated on the application of the inner projection technique and of the method of intermediate Hamiltonians on the benzene molecule as well as on the geminal approach to the butadiene molecule; the main calculations were first done in Maple and then transferred into Fortran's notation so that Fortran subroutines could be used. Finally, the results either in Maple or in Fortran can be transferred to a Troff file.

    Notizen: The potential energy curve describing the fragmentation of a diatomic dication AB2+ is considered as arising from an avoided crossing between an attractive diabatic curve (correlating with A2+ + B) and a repulsive diabatic curve (correlating with A+ + B+). The simplest avoided-crossing (AC) model neglects diabatic coupling and polarization and leads to useful predictions of the transition structure bond length (rTS) and the kinetic energy released (T) in fragmentations of dicationic systems in which the difference (Δ1) between the ionization energies of A+ and B is small. When Δ1 is not small, it is necessary to include diabatic coupling and polarization in the treatment. The resultant ACDCP (avoided crossing with diabatic coupling and polarization) model provides very satisfactory estimates of rTS and T for both small and large Δ1. Its implementation requires only atomic ionization energy and polarizability data and comes at virtually no computational cost. Both the AC and ACDCP models are readily generalized to fragmentations of more highly charged cations.

    Notizen: The effect of electron-electron repulsions is assessed, primarily in the expanded fluid alkali metals, by confronting experimental data on these systems with the predictions of theory. This is usefully divided into two types of approach: (i)full inclusion of the long-range Coulomb interaction e2/rij between electrons i and j, but perturbative treatment of electron-ion interaction and(ii)the simplified on-site interaction described by the Hubbard U.The main conclusion is that both electron-ion and electron-electron correlations are of major importance in the expanded fluid alkali metals, and especially in Cs. The momentum distribution of the conduction electrons is changed in a very important quantitative way from the predictions of the jellium model at the same density by the electron-ion interaction. This leads to a metal-insulator transition at a much higher electron density than the jellium prediction. The relevance of this conclusion to crystalline alkalis is finally referred to.

    Notizen: Classical valence bond theory is recast in a spin-free form which provides a practicable route to ab initio calculations of molecular electronic structure. The approach is simple and direct and requires only efficient algorithms for the generation and processing of permutations and the handling of Rumer diagrams: it makes modest demands on computing power and pilot calculations have indeed been performed entirely within the fast memory of a personal computer, which should be sufficient for dealing with systems possessing up to 10 electrons outside a closed shell. Simple applications confirm the conclusion of Cooper et al. [1] that, by using strongly overlapping orbitals, a small number of classical (nonpolar) structures can give results close to those obtained in a “full CI” calculation.

    Notizen: A set of 20 nitropolycyclic aromatic hydrocarbons, whose mutagenicity has been determined in the Ames test, has been studied using octanol-water partition coefficients (P) as a measure of relative hydrophobicity and molecular orbital energies to account for variation in their electronic characteristics. A good structure-activity relationship was found using log P and ∊LUMO. The latter were taken from the results of ab initio calculations performed by Maynard, Pedersen, Posner, and McKinney [7] and were also calculated by the MNDO method. The dependence of mutagenicity on hydrophobicity was found to be similar to that observed for triazenes [2]. ∊LUMO values calculated by MNDO and STO-3G were found to be strongly correlated, and the role of hydrophobicity in correlating mutagenicity was not significantly affected by the molecular orbital model employed.

It is based on a graph associated with the van der Waals surface, defined by the detailed information on the interpenetration of van der Waals spheres of the constituent atoms. This “van der Waals graph” describes the three-dimensional body of the molecule, and it does not coincide in general with the less informative bond graph obtainable from the molecular skeleton.The description in terms of the graph reveals clearly the changes in molecular shape induced by conformational rearrangements.     Notizen: A systematic collection of spatial domains for reciprocal space integrations is derived for all possible crystal symmetries. This set can be used as a simpler alternative to the conventional Brillouin zones. The analysis is restricted to integrations where the function in the integrand satisfies inversion symmetry in k space. In this case only 24 different spatial domains have to be defined in order to allow for k space integrations in the 230 different crystal symmetries. A graphic representation of the asymmetric unit for each of the 24 integration domains is given.     Notizen: In this paper we have addressed an important problem: how to develop a strategy for identification of the topological indices which are most important in establishing a model for structure-activity analysis. In particular we consider a rational selection of graph theoretical (topological) indices in QSAR. The approach is illustrated by the study of the toxicity of ethers on mice. The indices examined include the connectivity indices based on adjacency matrix, the conceptually related graph distance indices, and an information theoretic index based on partitioning of electrons in various domains. We have outlined the similarities and differences and discuss advantages and limitations among different approaches. Our emphasis is on the problem how to select in a rational way appropriate graph-theoretical descriptors and how to introduce higher “corrections” in order to improve derived correlation.

    Notizen: The reaction field (RF) model of solvent effects, implemented within the SCF-CNDO/2 scheme of calculation, has been applied to analyze the proton transfer in the NH3…HCOOH system in the presence of several polarizable media. The aim of such a study was to characterize the tatutomeric equilibrium between the neutral and zwiterionic forms of H-bonded amino acids in aprotic solvents. Qualitative results concerning the energetics of this equilbrium show the stabilization of two different H-bonded complexes, corresponding to two separate minima in the free energy surface. These well known double minima potentials are found to be dependent on both the intermolecular N—O distance and the strength of the reaction field. The behavior of this model is qualitatively consistent with experimental observations of nitrogen-substituted amino acids in solution: both show, for low values of the dielectric constant, tautomeric equilibria where the H-bonded complexes appear to be more stable than the corresponding monomeric forms. The charge transfer process associated with the proton migration along the H-bond is also discussed. It is found that the amount of charge transferred increases with the N—O distance and with the RF strength, In order to test the general approach and compare it with previous work, calculations on the real monomeric systems glycine, β-alanine, and γ-amino butyric acid was also performed.

    Notizen: Analytical intermolecular potentials for the Fe+-H2O and Feo-H2O systems have been determined from ab initio calculations. Interaction energies for a lot of points along the two potential energy surfaces were calculated using Huzinga's MINI-2 basis set. The results obtained were fitted to an analytical function containing 11 adjustable parameters that we have already used with success for the Fe2+-H2O system. The goodness of the generated intermolecular potentials is discussed.

    Notizen: The quasirelativistic CNDO/1 method has been in investigating the geometry, electronic structure, and redox stability of metal complexes. The systems of [Co(NH3)6]q and [MCl4]q, M being Ni, Pd, and Pt, have been studied. A modified Germer model of solvation has been implemented into the method. This yields reliable results on the redox stability of complexes in aqueous solutions. The calculated excitation energies resemble the electronic spectra of [MCl4]2- complexes.

    Notizen: In this note we discuss the variational forms of the energy derivatives and the method of obtaining them. We show that perturbation theory can be formulated in terms of stationary energy derivatives and that this formulation extends the idea of the Hylleraas functional to infinite order and excited states.

    Notizen: We examine various perturbation-variational approximations for molecular electronic energy when the fifth-order perturbational energies are available. Such theories require very little additional computation once the sequence of perturbation energies are known yet supply a bound even when the peturbation sequence is poorly convergent. We choose for computational examples results obtained very rapidly from a zeroth order wave function consisting of doubly occupied localized bonds and examine polarization within these bonds, delocalization, and bond breaking. In general, we find that the fifth-order energy itself, and especially the [2, 1] Padé approximant on this sequence, are especially accurate in estimating the total energy and more accurate than any variational scheme when the zeroth order localized wave function is a good description of the electronic structure. The variational results, however, are nearly as accurate, and a [1, 0] Padé on the sequence of variational results is remarkably robust, even in those cases where the perturbation sequence is poorly defined.We also examine several scaling techniques, or partitionings of the Hamiltonian. Although these scaling techniques do accelerate convergence of the perturbation sequence, none that we examine give better results, than the [2, 1] padé, which is independent of any scaling.

    Notizen: The pair correlation energies for some nl4f pairs of the ground state of the Yb atom are calculated for the first time. The partial wave (PW) increments to the second-order pair energies are generated using numerical first-order radial pair functions obtained as the solution of two-dimensional differential equations. The analysis of the PWs contributions shows the dominant role of the df, fg, and gh PWs for the 4d4f pair, of the pf and dg PWs for the 4p4f and 5p4f pairs, and of the sf and pg PWs for the 4s4f, 5s4f, and 6s4f pairs. A discussion of the similarities and differences of the structure of the correlation energy found in this paper with those calculated earlier for smaller atoms is given.
