    Notes: A magnetoplumbite type of Ba ferrite (BaM) thin layer was deposited on a 9 nm thick Pt underlayer, and excellent c-axis orientation was observed even for an 8 nm thick BaM layer, which corresponds to only three or four BaM unit cells. The grain size was almost in the same range of 60–85 nm even when the BaM layer thickness tBaM decreased from 60 to 17 nm, and tBaM should be reduced below 10 nm to make a grain size smaller than 50 nm. However, the perpendicular coercivity Hc⊥ and squareness S⊥ decreased drastically from 2.6 to 0.5 kOe and from 0.6 to 0.2, respectively, with the decrease of tBaM from 60 to 8 nm because of higher demagnetizing field and susceptibility to thermal fluctuation. On the other hand, the [BaM(5–24 nm)/Pt(9 nm)]3 multilayer exhibited higher Hc⊥ and larger S⊥ than the BaM/Pt bilayer for the same BaM layer thickness and Hc⊥ and S⊥ of the [BaM(8 nm)/Pt(9 nm)]3 multilayer was 2.0 kOe and 0.6, respectively. It was clarified that the deposition of the BaM/Pt multilayer was very effective for achieving a high perpendicular magnetic anisotropy constant even for the ultrathin BaM layer. © 2001 American Institute of Physics.

    Notes: Laser Raman microscope measurements in asymmetric double quantum wells with coupled narrow and wide quantum wells were performed to observe the nonequilibrium longitudinal-optical (LO) phonons that are generated by electrons during the phonon assisted tunneling. Both the Stokes and the anti-Stokes intensities show maxima at a certain applied voltage, where the calculated subband spacing between the wide and the narrow quantum well states is found to be equal to the LO phonon energy. This fact indicates that the population of nonequilibrium LO phonons becomes maximum when resonant LO phonon scattering occurs. A strong reduction in the photoluminescence intensity for the narrow quantum well is also observed at the same bias condition. © 1998 American Institute of Physics.

    Notes: NodD is the major regulator of nod genes expression in rhizobia. Previously, a HU-like protein in Rhizobium leguminosarum bv. viciae has been identified to bind specifically with nod promoters and be involved in in vitro nodD transcription, but its in vivo function remained unknown. In this work we have cloned and sequenced the R. leguminosarum bv. viciae gene, named hurL, for this HU-like protein. Using the E. coli-expressed HurL proteins, we proved that HurL had high affinity to several nod promoters and showed a stimulation effect on in vitro nodD transcription at appropriate concentration. The R. leguminosarum bv. viciae hurL gene was mutated by insertion of a kanamycin resistance cassette. The obtained hurL mutant strain M704 exhibited poor growth under free-living conditions and failed to induce nodules on Pisum sativum cv. Frisson and Vicia hirsuta. Further studies of NodD production and nod genes-lacZ fusions expression in the hurL mutant revealed that inactivation of hurL led to severe impairment in the nodD expression, repression in the inducible expression of nodA and nodF, and slight enhancement in the expression of px2, a gene identified earlier in this lab. These results suggested that hurL might be required for maintaining the normal expression of nod genes in R. leguminosarum bv. viciae.

    Notes: We previously designed a triple auxotrophic host-vector system in Aspergillus oryzae by isolating red-colored adenine auxotrophic mutants upon UV mutagenesis of a double auxotrophic host (niaD−sC−). In the present study an effort to exploit this system and construct a novel quadruple auxotrophic host was made by disrupting the argB gene involved in arginine biosynthesis. The argB gene-disruption cassette was generated by fusion PCR, which required only two steps of PCR to insert the selectable marker, adeA, into the target argB gene. The chimeric DNA fragment was transformed into the triple auxotrophic strain (niaD−sC−adeA−) and the argB disruptants were obtained with a high rate of efficiency (approximately 40%). The argB disruptants were characterized by normal colony color and reversal of arginine auxotrophy by introduction of the wild-type argB gene. Quadruple auxotrophic strains (niaD−sC−ΔargB adeA− or niaD−sC−ΔargB adeB−) were subsequently isolated upon UV mutagenesis of the triple auxotrophic strain (niaD−sC−ΔargB) followed by screening of red-colored colonies for adenine auxotrophy. The results obtained showed that the adeA gene served as an efficient selection marker in developing a novel host-vector system with quadruple auxotrophy in A. oryzae, thus providing a powerful tool to breed multiple auxotrophic mutants from a deuteromycete wherein sexual crossing is impossible.

    Notes: Stimulation of bone healing through local application of growth factors from implantsmay improve the clinical outcome in fracture treatments. However, the growth factors in reconstructive application require supraphysiologic dosing and considerable expense while hampering their clinical application. Genetic modification of mesenchymal stem cells (MSCs) to both produce and respond to osteogenic factors may have potential for use in enhancing bone healing. In this study, MSCs were genetically modified by a recombinant adenoviral containing the gene for human bone morphogenetic protein 2 (hBMP-2). The gene-transduced cells wereincorporated with a porous beta-tricalcium phosphate (TCP) as a novel complex. We investigated osteogeneic potential of gene-transduced MSCs/ceramic and the ability of the complex on facilitating bone formation in a radius segmental defect of rabbits. In vitro results showed that there were apparent hBMP2 gene expression and protein synthesis in MSCs with hBMP2 stably transfection, whereas negative expression of hBMP2 in controls. Histological studies demonstrated that gene-transfected MSCs/ceramic composite appeared an ability of heterotopic osteogenesis. In the segmental bone defects, endochondrial ossification at fracture sites was found in both transfected and untransfected MSCs-ceramic composites. While the composite with hBMP2 transfection showed the earliest and the most effective healing of the segmental bone defects both radiographically and morphologically. Our results show that genetically modified MSCs/ceramicshad enhanced osteogeneic capacity relative to unmodified MSCs or only ceramic implants. This study suggests that use of cell-and gene-activated bioceramics may offer promise for molecular design of implants to induce osteogenesis and enhance bone regeneration

    Notes: The objective of this study was to evaluate the interface shear strength and the responses of osteoblast-like cells to titanium implants with a sandblasted and acid-etched surface modified by alkali and heat treatments (SLA-AH). The implants with machined and SLA surface served as controls. Each type of implant was characterized by scanning electron microscopy (SEM) and energy-dispersive x-ray (EDX) analysis. In vitro assays were made using human osteoblast-like cellculture on different surfaces. The rectangle plates were also transcortically implanted into the proximal metaphysis of New Zealand White rabbit tibiae. After 4, 8 and 12 weeks implantation, mechanical and histological assessments were performed to evaluate biomechanical and biological behavior in vivo. By SEM examination, SLA surface combined with AH treatments revealed a macro-rough surface with finely microporous structure. The in vitro assays showed that theSLA-AH surfaces exhibited more extensive cell deposition and improved cell proliferation as compared with controls. Pull-out test demonstrated that the SLA-AH treated implants had a higher mechanical strength than the controls at all interval time after implantation. Histologically, the test implants revealed a significantly greater percentage of bone-implant contact when compared with controls. The results of this study suggest that a useful approach by combined processes couldoptimize implant surfaces for bone deposition and produce distinct biological surface features

    Notes: Biomimetic nanoapatite coatings was developed by functionally modified methods with acombination of topographic, chemical and biomimetic treatments on the surface of titanium (Ti)substrate. The biological behavior and bioactivity of functionally modified SLA implants withchemical and biomimetic treatments (SCB-treated Ti) using body like solution were investigated tocompare with untreated Ti and SLA Ti plates as controls. The cell attachment, proliferation,alkaline phosphotatse (AKP) activity, cell morphology and differentiation were evaluated by usingMTT, RT-PCR, scanning electron microscopy (SEM) and confocal laser-scanning microscope(CLSM) analysis system. The results showed that the cell adhesion and proliferation was enhancedon functionalized titanium surface with nano-scale apatite compared to the controls. SEMmicrographs also revealed that the osteoblast-like cells spreadly grew along the surface. Cellmorphology and differentiation could be further observed distinctly by CLSM graphs. Moreover,mRNA expression of alkaline phosphotatse in nucleus on the SCB-treated Ti increased obviouslyon the third day compared with the controls. The in vitro results demonstrated the remarkableimprovement on cell adhesion and proliferation of the biomimetic nanoapatite on SCB-treated Ti,which could be used for orthopaedic/dental implants
