    Notes: Summary Horseradish peroxidase was injected into the biceps brachii muscle of rats at various stages of development, from 10 days to 50 weeks. The retrogradely labeled neurons were found in the ipsilateral ventro-lateral column of the cervical cord, C4–C8, of all stages studied, but the number of labeled neurons decreased according to exponential curve as the age advanced. A striking finding was that the contralateral ventral horn cells were also labeled in the 10- and 14-day-old rats.

    Notes: Summary The origin of the spinal cord noradrenaline (NA) has been investigated by means of the horseradish peroxidase (HRP) method, combined with monoamine oxidase staining (Glenner) to identify the NA neurons. Following the injection of HRP to the various levels of rat spinal cord, cervical to sacral cord, A1–3, 5–7 NA neuron groups were labeled with HRP. They showed almost the same distribution pattern regardless of difference in the injected segment. Labeled NA neurons in A6 were concentrated in the ventral division of the locus coeruleus, which continued to the labeled NA neurons in the subcoeruleus area. The HRP positive neurons in the pons outnumbered those of the medulla oblongata. As the NA neurons described above were considered to be the source of NA in the forebrain, such as the hypothalamus and preoptic area, the possibility that the same NA neurons might innervate both the forebrain and spinal cord has been presented.

    Notes: Summary Combination of glyoxylic acid perfusion and postfixation in permanganate was used in an electron microscopic study of the locus coeruleus (LC) of the rat to give good preservation of fine structure and a reproducible demonstration of noradrenaline (NA) storage granules. Medium-sized LC cells (18 × 30 μm) contained a moderate number of small granular vesicles (SGV) and a few large granular vesicles (LGV), mainly near the Golgi apparatus. Dendritic branches were identified by their SGV content up to the tip. Dendrites were occasionally in close contact with each other or with the soma of LC cells, forming dendro-dendritic or dendro-somatic contacts. Numerous axon terminals containing many SGV and some LGV were observed in the neuropil, and they tended to contact dendrites and somata of LC cells or dendrites of unknown origin. These neuronal contacts were devoid of synaptic specializations except for an array of dense perpendicular lines between the juxtaposed membranes. Small oval cells (10 × 15 μm) devoid of SGV occurred frequently in the peripheral part of the nucleus, and they were occasionally in direct contact with LC cells.

    Notes: Summary Attempts were made to determine the afferent projections to the anterior hypothalamus including the preoptic area from the lower brain stem by means of the horseradish peroxidase method combined with monoamine oxidase staining to identify noradrenaline (NA) neurons. In addition to this technique, a histofluorescence analysis was performed. NA fibers in the medial part of the anterior hypothalamus were mainly supplied by A1 and A2 NA neuron groups, while the lateral part and periventricular zone received NA terminals from both pontine and medulla oblongata NA neuron groups. Furthermore, the present study indicated that there were direct projections to the anterior hypothalamus from non-noradrenergic neurons in the lower brain stem: nuclei raphe dorsalis, centralis superior, cells in the mesencephalic and pontine central gray matter, nuclei parabrachialis lateralis and medialis, cells around fasciculus longitudinalis medialis.

The effects of SK&F 24260 administered intravenously or intraduodenally on the coronary sinus outflow, coronary arteriovenous oxygen difference (A-V O2), myocardial oxygen consumption (MVO2), systemic blood pressure, heart rate and atrioventricular (AV) conduction time were examined in open-chest dogs.2. SK&F 24260 in doses of 0.3–10 μg/kg, i.v., caused a dose-dependent increase in coronary sinus outflow, but the increase was smaller with 30 μg/kg, i.v., than with 10μg/kg, i.v.3. SK&F 24260 (0.3–30μg/kg, i.v.) decreased A-V O2 and MVO2 in a dose-dependent manner.4. SK& F 24260 (0.3–30μg/kg, i.v.) decreased systemic blood pressure and heart rate, and increased AV conduction time.5. Intraduodenal administration of SK&F 24260 (1 mg/kg) produced almost the same effects on coronary sinus outflow, A-V O2, MVO2, systemic blood pressure, heart rate and AV conduction time as did the intravenous administration of the compound (10μg/kg).6. The property of SK&F 24260 to increase the coronary blood flow and to moderately decrease MVO2, systemic blood pressure and heart rate indicates that this agent is a potential antianginal drug.

    Notes: SUMMARY  Contracture of jaw-closing muscles is one of the causes of limitations of jaw opening. In contracture patients who have no history of trauma or infection, it is not easy to distinguish contracture from temporomandibular joint (TMJ) closed lock (TCL). The purpose of this study was to clarify whether there is any difference between electromyographic (EMG) activities of jaw muscles during jaw opening in patients with TCL and patients with masseter muscle contracture (MMC). The MMC-patient group consisted of one male and 11 females with no history of trauma or infection. The TCL-patient group consisted of one male and 11 females. Ten of the MMC patients showed certain types of EMG activities in masseter muscles (and eight in temporal muscles) during jaw opening. However, particular EMG activities were not observed in most of the TCL patients. The integral values in masseter muscles and in temporal muscles at the maximum opening position were significantly higher in the MMC-patient group than those in the TCL-patient group. These findings demonstrate that the EMG pattern of MMC patients without a history of trauma or infection is different from that of TCL patients. Therefore, EMG analysis of jaw-closing muscles during jaw opening is expected to be useful for differential diagnosis between MMC and TCL.

    Notes: Aim: Various treatments for hypertrophic and keloid scars have been attempted including electron beam irradiation, local triamcinolone injection, oral tranilast administration, use of silicone sheets, and compression therapy using splints. Among them, we have been attempting silicone cushion patching on hypertrophic scar. Methods: Twenty cases (10 males, 10 females) with hypertrophic scar were treated with silicone cushion. Scar surfaces were kept in contact with silicone cushion for as long periods of time as possible every day. Results were assessed in scores using objective findings (redness, bulging, induration) and subjective symptoms (itching, spontaneous pain, tenderness). Results: As for objective findings, redness remained unchanged in one case, but bulging and induration were found improved in all cases. As for subjective symptoms, itching and tenderness were found improved in all cases and clinical course considered proving usefulness of the treatment was obtained. Conclusions: Silicone cushions have highly viscous silicone oil enclosed in the silicone pack designed to generate negative charge electrostatic fields. Generation of adequate and sustaining electrostatic fields is important for useful clinical effect to exhibit on hypertrophic scar recession. Generation of negative charge electrostatic fields was confirmed on potential determination data with an electrostatic field meter as well.

    Notes: Rabeprazole sodium is a proton pump inhibitor.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To evaluate the efficacy and safety of 1-week triple therapy with rabeprazole, amoxycillin and clarithromycin for the eradication of Helicobacter pylori.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:A total of 100 subjects with H. pylori were randomly divided into two groups of 1-week triple therapy with rabeprazole 10 mg b.d., amoxycillin 750 mg b.d. and either clarithromycin 200 mg b.d. (RAC400, n=50) or clarithromycin 400 mg b.d. (RAC800, n=50). Endoscopic examination with four biopsies (two specimens from the antrum and two from the gastric body) was performed. The status of H. pylori infection was determined using culture and histology (Giemsa stain) of the biopsy specimens. Sensitivity to clarithromycin was determined using the E-test: MIC 〉 8 g/mL was considered to be resistant, whereas MIC 〈 2 g/mL was considered to be sensitive. Cure was defined as no evidence of H. pylori infection 1 month after completion of treatment.〈section xml:id="abs1-4"〉〈title type="main"〉Results:There were no significant differences in the clinical characteristics of the two groups. Eradication rates (intention-to-treat and per protocol, respectively) were: RAC400: 86% (95% CI: 76–95%) and 89% (95% CI: 80–97%); RAC800: 94% (95% CI: 87–100%) and 97% (95% CI: 94–100%). There was no significant difference between the eradication rates of either regimen. Three subjects with failed eradication in the RAC400 group were all infected with a clarithromycin-resistant strain before beginning the therapy. Haemorrhagic colitis was the only severe adverse event, which was observed in one patient in the RAC800 group.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusion:One-week triple therapy with rabeprazole, amoxycillin and low-dose clarithromycin is effective for the eradication of H. pylori infection.

    Notes: Helicobacter pylori infection is a major cause of the progress of gastric glandular atrophy, a high-risk background factor in the development of gastric cancer. Regression of gastric atrophy is critical to prevention of cancer by H. pylori eradication treatment. However, it is controversial whether gastric atrophy regresses after H. pylori eradication.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To determine the most sensitive and appropriate biopsy site for evaluation of regression of atrophy after treatment.〈section xml:id="abs1-3"〉〈title type="main"〉Subjects and methods:Thirty-eight patients who showed regression of gastric atrophy in histology after treatment were investigated. Four biopsy specimens from the lesser and greater curvatures in the antrum and corpus were evaluated before and after treatment according to the Updated Sydney System.〈section xml:id="abs1-4"〉〈title type="main"〉Results:Regression of atrophy after treatment was seen in 30 of 38 biopsy specimens from the lesser curvature of the corpus (79%), and this site was most sensitive. Odds ratio of this site to the others was 8.28. Regression of atrophy in this site was observed at 12.2 months in the younger patients and 15.9 months in the elder patients.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusion:Biopsy sampling from the lesser curvature of the corpus is the most sensitive and appropriate for evaluation of regression of gastric atrophy after H. pylori eradication treatment.

In anaesthetized, open-chest dogs, 2-nicotinamidoethyl nitrate (SG-75) administered intravenously (0.3–1 mg/kg) or intraduodenally (3 mg/kg) produced decreases in systemic blood pressure, coronary resistance, heart rate and an increase in coronary sinus outflow, but virtually no change in myocardial oxygen consumption and atrioventricular conduction. The effects of SG-75 administered intraduodenally emerged within a few minutes after dosing and lasted over about 1 h.2. In isolated, blood-perfused sino-atrial node preparations of the dog SG-75 administered into the sinus node artery decreased slightly sinus rate at the dose which doubled blood flow through the artery.3. In isolated, blood-perfused atrioventricular node preparations of the dog SG-75 administered into the atrioventricular node artery did not impair atrioventricular conduction even in doses which increased blood flow through the artery to more than twice the basal level.4. In isolated, blood-perfused papillary muscle preparations of the dog SG-75 administered into the anterior septal artery scarcely affected force of contraction of the papillary muscle at the dose which doubled blood flow through the artery, although in further large doses it produced a transient decrease in the force of contraction. In extremely large doses it produced ventricular fibrillation.5. In anaesthetized, open-chest dogs in which cardiac input was kept constant SG-75 (0.01–1 mg/kg) administered into the ascending aorta increased venous return without changing systemic output.6. 2-Nicotinamidoethyl alcohol, a denitrated compound of SG-75, had no vasodilator action in doses comparable to those of the parent compound.7. These results indicate SG-75 to be a potential antianginal drug having no cardiodepressant actions but having properties uncharacteristic of the nitrates.
