    Notizen: Abstract After a short summary on the ecology and rhizosphere biology of symbiotic bacteria and vesicular-arbuscular (VA) mycorrhiza fungi and their application as microbial inocula, results on competitiveness and communication are summarized. Stress factors such as high temperature, low soil pH, aluminium concentrations and phytoalexins produced by the host plants were studied withRhizobium leguminosarum bv.phaseoli andRhizobium tropici onPhaseolus beans. Quantitative data for competitiveness were obtained by usinggus + (glucoronidase) labelled strains, which produce blue-coloured nodules. ForPhaseolus-nodulating rhizobia, a group specific DNA probe was also developed, which did not hybridize with more than 20 other common soil and rhizosphere bacteria. Results from several laboratories contributing to knowledge of signal exchange and communication in theRhizobium/Bradyrhizobium legume system are summarized in a new scheme, including also defense reactions at the early stages of legume nodule initiation. Stimulating effects of flavonoids on germination and growth of VA mycorrhiza fungi were also found. A constitutive antifungal compound in pea roots, β-isoxazolinonyl-alanine, was characterized.

    Notizen: Summary In isolated rat pancreatic islets exogenous glutathione which is not able to penetrate into cells, augmented glucose (11.1 mmol/l)-mediated insulin release. In the presence of a non-stimulatory glucose concentration (2.8 mmol/l) glutathione had no effect. The half-maximal synergistic action of glutathione on insulin secretion was observed at approximately 8.0 μmol/l. This concentration of glutathione is similar to that found in the plasma of non-fasted anaesthetised rats (6.5 μmol/l). Oral ingestion of glucose increased the arterial plasma glutathione in rats. Our data provide for the first time indirect evidence for a modulating effect of plasma glutathione in postprandial glucose-mediated insulin secretion which appears to be located at the extracellular site of islet cells.

    Notizen: Summary Glycation of basement membrane collagen IV has been implicated as a major pathogenetic process leading to diabetic microvascular complications. To evaluate the relevance of carbohydrate-induced modifications on collagen IV in diabetic nephropathy, we isolated the cross-linking domains 7S and NC1 from the glomerular basement membrane (GBM) of patients with diabetes mellitus. Modifications characteristic for glycated proteins were identified when the domains from diabetic kidney were compared with the same domains from human placenta as an unmodified control. In both domains a marked formation of inter-and intramolecular cross links could be demonstrated by SDS-PAGE. Furthermore circular dichroism studies showed a decrease in helicity of the 7S domain from human diabetic kidneys of 13 %, indicating denaturation already at room temperature. Thermal transition profiles, showing a shift of the denaturation temperature towards a lower temperature, with loss of a distinct second melting point, confirmed this observation. Our data provide further evidence for a possible role of protein-modification by glycoxidative reactions in the onset of diabetic nephropathy in vivo. [Diabetologia (1998) 41: 1073–1079]

    Notizen: Abstract Background: The purpose of this study was to compare the performance of in-phase and out-of-phase gradient recalled echo (GRE) pulse sequences on paramagnetic contrast-enhanced magnetic resonance (MR) imaging of malignant liver lesions. Methods: Fifty patients (27 women, 23 men; mean age = 50 ± 27 years) with known or suspected focal liver lesions, nine of whom had a fatty liver, were examined at 1.5 T before and 60 min after injection of gadobenate dimeglumine at a dose of 0.05 or 0.1 mmol/kg using two GRE techniques: echo time of 2.3 ms (out-of-phase) or 4.6 ms (in-phase). Liver signal-to-noise ratio (SNR) and lesion–liver contrast-to-noise ratio (CNR) were calculated. Results: In patients with a nonfatty liver, liver SNR increased from 26 ± 9 to 41 ± 17 on in-phase images and from 28 ± 8 to 45 ± 14 on out-of-phase images. In patients with a fatty liver, in-phase images provided significantly higher (p 〈 0.01) liver SNR than did out-of-phase images predose (34 ± 8 on in-phase vs. 21 ± 8 on out-of-phase) and postdose (44 ± 13 on in-phase vs. 33 ± 14 on out-of-phase). In patients with a nonfatty liver, lesion–liver CNR was similar on in-phase and out-of-phase images, predose and postdose. In patients with fatty liver, lesion–liver CNR was significantly (p 〈 0.01) lower on out-of-phase images on predose and postdose images. Conclusion: In-phase GRE imaging is recommended for imaging focal liver lesions on paramagnetic contrast-enhanced MR imaging in patients with fatty infiltration of the liver.

Background: To demonstrate the radiologic–pathologic correlation of adenomyomatosis of gallbladder (GBA) and emphasize the role of high-resolution real-time ultrasound (RTUS) in the diagnosis of GBA. Methods: Ten (four male and six female, mean age = 49 years) patients with proven GBA (three diffuse, three segmental, and four fundal) diagnosed by histopathology or confirmed by oral cholecystography (OCG) were reviewed. Radiologic studies included OCG (n = 8), RTUS (n = 8), and computed tomography (CT; n = 4). Six patients subsequently underwent cholecystectomy. Results: Histopathologic correlation between pathologic specimens and OCG, RTUS, and CT was possible in six patients. The diagnostic criteria with ultrasound included numerous tiny intramural cysts containing echogenic foci with reverberation artifacts and associated segmental or diffuse gallbladder wall thickening. OCG with fatty meal demonstrated intramural diverticula. Localized fundal GBA was better visualized on RTUS and CT scan than on OCG. Conclusion: Accurate diagnosis of GBA may be made by either OCG or high-resolution RTUS preoperatively. CT scan may used as an alternative method to help make the diagnosis in equivocal cases.

    Notizen: Summary Immunoglobulin (Ig) concentrations were investigated in white matter samples of two adrenoleukodystrophy (ALD), threee multiple sclerosis (MS), two systemic lupus erythematosus (SLE), one rheumatoid arthritis, and three control brains obtained at autopsy. “Free” Igs were extracted at pH 7.4; subsequently, bound Igs were extracted at pH 2.5 and 10.8, respectively. Igs were quantified by radial immunodiffusion. In ALD material there was an increase of free IgG and IgA, in one sample also of IgM, as compared to controls. No significant amounts of Igs were detected in the pH 2.5 and 10.8 extracts of ALD brain. Similarly to ALD, an increase of free IgG and IgA was a characteristic finding in MS brain; in contrast to ALD and control material, significant amounts of bound Igs (IgG) extractable at acid or alkaline pH, respectively, were present in MS tissue. In both SLE brains increase of free IgM was conspicuous. Preliminary studies on binding of Igs extracted at pH 7.4 from brain to frozen sections of normal human and bovine brain tissue revealed different binding properties of Igs from ALD, MS, SLE, and control brains. Immunochemical findings in ALD indicating pathologic accumulation of Igs in brain tissue were paralleled by immunocytochemical observations demonstrating accumulation of lymphoid cells staining for IgG, IgA, and IgM, respectively, mainly in areas of recent demyelination. Participation of Igs in the pathogenesis of ALD lesions may be considered but needs further confirmation.

The hemorrhages of the lung were accompanied by fever and roentgenologically slight infiltrations of the middle and lower compartments of the lung with simultaneous decrease of hemoglobine. The disease having lasted 16 months the patient has been free of complaints for the last 5 months. The biopsy of the kidney resulted in the diagnosis of a momentarily inactive proliferative focal glomerulitis. This could be confirmed by electron microscopical investigation, although there were found traces of activity. of the cell elements. Deposits of iron faintly indicated under the light microscope, were confirmed by electron microscopy. The total picture corresponds to a typical Goodpasture' syndrome.
    Notizen: Zusammenfassung 30jähriger Mann mit rezidivierender Hämoptoe. Die Lungenblutungen waren von Fieber und röntgenologisch flüchtigen Lungeninfiltraten in Mittel- und Untergeschossen, gleichzeitigem Abfall des Hämoglobins begleitet. Bisherige Krankheitsdauer 16 Monate, die letzten 5 Monate war der Patient völlig beschwerdefrei. Die Nierenbiopsie ergab eine momentan inaktive proliferative Herdglomerulitis, die auch elektronenmikroskopisch nachgewiesen werden konnte. Allerdings fanden sich ultrastrukturell noch deutliche Zeichen für Aktivität der Zellelemente. Die lichtmikroskopisch nur angedeutete Eisenablagerung im Deckepithel der Glomerula konnte elektronenmikroskopisch bestätigt werden. — Das Gesamtbild entspricht einem typischen Goodpasture-Syndrom.

Three different methods are described for the visualisation of the Thomsen-Friedenreich (TF) antigen on cell suspensions, formalin-fixed and paraffin embedded or frozen tissue sections: a) Rosette formation with chicken and sheep erythrocytes. b) Fluorescence-microscopy with fluorescein labelled peanut agglutinin. c) Autoradiography with3H-labelled peanut agglutinin. 2. The TF antigen was shown, as far as presently investigated, to be exposed on various blood cells, glomerula of the kidney and normal mammary gland after neuraminidase treatment. Mammary gland was also shown to possess TF receptors without prior treatment with neuraminidase. 3. The exposure of this cryptantigen can be brought about by bacterial or viral neuraminidase and is followed by an antigen/antibody reaction, which can lead to possible pathological consequences.

Thomsen-Friedenreich (TF)-Antigenen in Zellsuspensionen, an Formalin-fixiertem und in Paraffin eingebettetem Gewebe sowie in Gefrierschnitten werden beschrieben: a) Rosetten-Bildung mit Hühner- und Schaferythrocyten. b) Fluorescenzmikroskopie mit Fluorescein-markiertem Erdnußagglutinin. c) Autoradiographie mit3H-markiertem Erdnußagglutinin. 2. Nach Neuraminidasebehandlung konnten TF-Antigene — soweit bisher untersucht — auf verschiedenen Blutzellen, in Nierenglomerula und in normalem Brustdrüsengewebe nachgewiesen werden. Ein Teil derartiger Antigene lag im Brustdrüsenparenchym auch bereits als freie Rezeptoren vor, d. h. waren nicht von Neuraminsäure bedeckt. 3. Die Freilegung dieser Kryptantigene kann im Rahmen von Infektionen durch bakterielle und virale Neuraminidase erfolgen. Dabei kommt es zu einer Antigen/Antikörper Reaktion mit unter Umständen klinisch relevanten Folgen.
