    Notes: Individual culture supernatants of six cellulolytic fungi and their mixed enzyme systems were compared in hydrolysis of pure celluloses and cellulose-rich wastes. It has been observed that the hydrolysis rate of pure cellulosie substances in general increased by mixed enzymes as compared to the enzymes supernatant derived from only one microbial source. This effect was particularly high when Trichoderma and Penicillium enzyme systems were operated together.

    Notes: The study of a heat-shock process for RNA reduction was carried out for different yeast strains. Different results were obtained from each of them. Candida utilis NRRL Y-660 shows its best performance after a 8-s. heat-shock in the presence of 3% NaCl. For commercial baker's yeast Saccharomyces cerevisiae and Kluyveromyces fragilis L-1930, similar results were obtained with only 1% of NaCl. The latter needed longer heat-shock periods. e.g. 15s. to give such an RNA reduction. Biomass recovery ranged from 60 to 75%, being higher for C. utilis and K. fragilis while excessive losses were observed in S. cerevisiae cells.No significant protein deterioration was obtained in the best performance samples. The aminoacid profile appears to be improved in comparison to the starting material in these strains after RNA reduction.

    Notes: Ethanol synthesis was initiated in aerobically pre-cultivated synchronized yeast populations by interrupting aeration and adding carbon substrate. Synchronization of yeast-cell functions was carried out by aerobic phased cultivation.Ethanol synthesis was more effective if it was closely connected with aerobic cell-mass synthesis and cell proliferation. Thus, anaerobic continuous ethanol production with cell recycling could be intensified by replacement of aged cells in single-cell state by aerobically pre-cultivated budding cells.

    Notes: The strains E. coli C 600 and E. coli C 600 pL CR 665-58 were cultivated on a glucose containing nutrient medium. The cell states of the cell cycle were investigated by means of phased cultivation. The doubling time and the specific carbon-substrate consumption of cells were higher when E. coli C 600 pL CR 665-58 was used. During cell doubling cell states characterized by different specific carbon-substrate consumption coefficients were observed.By adaptation of carbon-substrates supply to the repetitive cell states during continuous cultivation of synchronized bacteria populations the efficiency of cell-mass production was increased by 15 percent.

    Notes: Xanthan overproduction only takes place if a sugar which may serve as precursor is present and transition to limited growth occured. This transient phase is characterized by a change in composition of macromolecular cell components (RNA, protein, carbohydrates). During the transient phase nearly no nitrogen is consumed, whereas glucose and oxygen consumption proceeds nearly undisturbed. The changes observed in the transient phase are discussed in context with physiological adaption.

    Notes: The influence of the osmotic pressure on the viability of yeast cells was studied in batch processes. It could be found, that the viability is a function of the total osmotic pressure obtained by adding the partial osmotic values of the principal ingredients saccharose, ethanol and salts dissolved in the medium. At optimum process conditions (pH, T, etc.) the cells can tolerate the osmotic pressure up to fixed value π1. Above π1 the viability decreases linearly and upward of a second threshold value π2 nonlinearly. In the case of the used strain Saccharomyces cerevisiae Hansen Sc 5 the values of the both points are π1 = 25 atm and π2 = 47 atm.

    Notes: Experimental kinetic data (initial rate and high conversion) on the hydrolysis of cellobiose by 1,4-β-glucosidace (Gliocladium sp.) have been analysed and a competitive inhibition by glucose has been proposed. The determination of kinetic parameters from integral data is based upon algorithms for non-linear optimization and numerical integration. The values of kinetic constants \documentclass{article}\pagestyle{empty}\begin{document}$(v_{\max } = 1.02\frac{{\mu {\rm M}_{{\rm glucose}} }}{{{\rm mg}_{{\rm protein}} \cdot \min }},K_M = 2.6{\rm mM/l, and }K_P = 1.2{\rm mM/l)}$\end{document} agree well with the initialrate results. An important distinction is the confidence limit of parameters. Linear regression analysis shows a virtual accuracy and can lead to wrong conclusions.

    Notes: The accumulation of nucleic and protein (amino nitrogen) components as decomposition products of the process of autolysis of Saccharomyces cerevisiae yeast cells was studied at 50°C under the effect of various membranotropic additives. The influence of the added n-alcohols, n-fatty acids and several peptides was investigated in the range of concentration of 0.1-0.5 M. The maximal acceleration of the autolysis has been demonstrated under the effect of additives with a hydrophobicity of 7.5-8.5 ccal/M. In all the investigated concentrations stearic acid and octadecyl alcohol have an inhibitory influence. The role of the hydrophobic influences and the mechanism of autolysis are discussed.

    Notes: The effect of different quantities and kinds of vegetable and animal oils and the importance of the time of the oil addition on growth and protease synthesis by T. vulgaris was investigated. The used oils stimulate the protease production if they are added in a suitable concentration. However, the stimulation effect of each oil is different. Culture inoculation with spores gives the best results, when oil was added to the medium 1 up to 2 hours after beginning of the fermentation. The enzyme activity is equal to or lower than the control, when the oil addition was carried out before or 3 hours after starting the fermentation.

    Notes: The splitting of vegetable and animal oils used as antifoam agents and the metabolization of fatty acids released were analysed during fermentation of T. vulgaris in a 30 l jar fermenter. During cultivation time of 22 hours the oil amount decreased by 60 to 70%. The metabolization of fatty acids is 50 to 90% of the starting level depending on the kind of acid. The eventual effect of the fatty acids released from antifoam oils on the protease production is discussed.

    Notes: This paper presents the application of a new developed measuring device for continuous determination of ammonium concentration especially in the field of biotechnology.     Notes: The primary objectives in biological wastewater and sludge treatment processes are to procedure a minimum quantity of solid, stable residues and a maximum quantity of carbon dioxide from the organisms present in the process feeds. As far as minimizing solids production in activated sludge type processes is concerned, endogenous activity in the recycled biomass is usually considered to be the most important mechanism. However, increased understanding of the growth characteristics of mixed microbial cultures suggests that lysis and “cryptic” growth are probably dominant mechanisms. For pathogen destruction in treatment processes, death and subsequent lysis of pathogens are clearly events that must be promoted.Here, the kinetics for death, lysis and “cryptic” growth in aerobic wastewater treatment and aerobic sludge stabilization processes are examined.

    Notes: The total amount of carbon dioxide (CD) produced by a microbial culture is evolved via an outlet gas stream, outflowing broth, and can lead to a rise of the CD content in broth. To analyze relations among the three amounts, the equilibrium is considered between different ionic forms of carbon dioxide at different pH values as well as between the gaseous and disolved CO2. Dependences of the equilibrium constants on temperature are found by a thermodynamic analysis. Numerical estimation of an error of the CD production rate measurement, originating from the neglect of dissolved CD, is developed. The gas analysis technique alone provides a sufficient accuracy at pH lower than 6. At higher pH the error can be estimated using equations presented below.

    Notes: At the investigation of some properties of the α-amylase and proteinase in the culture filtrat from Bacillus licheniformis MB 80 strain it has been established that the α-amylase activity is the highest at pH 6.0 to 6.5 and at 90°C, that the proteolytic activity is the highest at pH 9.5 to 10.0 and at 70°C and that the proteinase is inactivated at temperatures over 70°C.

It can be conducted from many experiments performed in our laboratory. that 10-100 phages per 1010 bacteria are released. However, the production of bacterial mass is not markedly influenced by lysogeny.Strains of Bordetella bronchiseptica used for production of vaccine against Rhinitis atrophicans of pigs have temperate phages too. These phages may cause a complete lysis during a submerse cultivation.The phages of Bordetella pertussis and Bordetella bronchiseptica can be propagted on Bordetella parapertussis.

    Notes: The effects of pyrccarbonic acid diethyl ester (PADE) on Lactobacillus casei Sl and its phage Jl was investigated in relation to the control of phages in the dairy industry and other technica fermentation processes.PADE exhibited a bacteriostatic effect at 0.5 to 8 mM and a bactericidal effect at 10 mM or higher. It inhibited the growth of the phage at its bacteriostatic and bactericidal concentrations. The growth inhibition of the phage was reversible at the bacteriostatic concentrations but complete and irreversible at the bactericidal concentrations. PADE inactivated the free phage within several minutes; 10 and 30 mM of PADE inactivated 90 and 100%, respectively, of the phage. It completely decomposed into ineffective components in several minutes. The bacteria grew almost normally when they were inoculated after the complete decomposition of PADE.These four characteristics of PADE - its bactericidal effect, its inhibitory effect on phage growth, its phage-inactivating effect and its decomposition - suggest a novel strategy for phage control in technical fermentation processes, including the dairy industry.

    Notes: Alcaligenes latus strain DSM 1123 has been tested for the production of Poly-D(-3)-3-hydroxybutyric acid using sucrose as the only carton source. The strain is capable of accumulating the polymer associated to the growth very fast and lup to high concentrations in the biomass.     Notes: Although the carbon/energy ratios of heterotrophic substrates for microbial growth are different this is not reflected in biomass. Nevertheless the macromolecular composition of cells may vary in dependence on growth conditions this does hardly influence the elementary composition and the growth yield. The energy requirement for synthesis of biomass starting from a central precursor, e.g. phosphoglycerate, can be assumed to be constant, hence any differences in carbon conversion efficiency must be attributed to carbon catabolism up to this precursor. This sequence determines if and to what extent an auxiliary substrate effect is possible. However, one has also to consider changes of the P/O ratio due to simultaneous utilization of substratesd which may account for the increase in growth yield with Hansenula polymorpha growing on a methanol/glucose mixture.

    Notes: The effect of dehydration on proteolysis and activity of proteases A, B and C in the cells of baker's yeast Saccharomyces cerevisiae was investigated. It can be concluded, that under investigated conditions of yeast Saccharomyces cerevisiae drying a decrease of proteases activity takes place. In cells a limited proteolysis takes place which is indicated by an increase in amino nitrogen content and a decrease of tryptophane synthase activity. Adding the protease inhibitor to yeast suspension prevents decrease of tryptophane synthase activity upon dehydration.

    Notes: Autohydrolysis was studied as a pretreatment to enhance sugar yields from enzymatic hydrolysis of wheat and rape straw, beech, birch and poplar sawdust. Reaction temperatures were 185°C to 212°C and the reaction time 20 min. The pretreated slurries were hydrolyzed with “Novo” cellulase and Fusarium sp. 27 cellulase at 45°C and pH 4.8 for 24 h with addition of Fusarium sp. 27 cellbound cellobiase. From 85% to 90% sugar content of substrates were converted to reducing sugars after 24 h enzymatic hydrolysis, with exception of poplar wood.10.8 g biomass was obtained after cultivation of Fusarium sp. 27 with water solution hemicellulose fraction from 100 g beech sawdust autohydrolyzed at 200°C during 20 min.

    Notes: The influence of sucrose concentration on the specific ethanol production rate was studied during batch processes using the yeast strain Saccharomyces cerevisiae Hansen Sc 5. From experimental data a model could be derived for the simulataneous effect of substrate and product inhibition. It was found that both the decreases of fermentation activity of the cells caused by sucrose and ethanol have an additional relation to each other. This model also takes into consideration the fact that the maximum ethanol concentration P′ can't be realized at high substrate concentrations in a batch process. Compared to it sucrose concentrations below 100 g/l did not inhibit the ethanol production by the strain used in this investigation.

    Notes: By use of a laboratory fermenter with a gastight circle aeration system all interesting values connected with the respiration of microorganisms can be determined.The advantage of the respiration fermenter is the simple control of the oxygen partial pressure with high accuracy. The aeration is carried out by inert gas-oxygen-mixtures. This system was used to avoid errors in oxygen-concentration measurement and regulation by oxygen sensitive sensors in fermentation fluids.For each selected oxygen partial pressure in the course of fermentation the following date in connection with all desired analytical values can be determined by manual or automatical means.Total oxygen consumption, momentary oxygen consumption rate, kinetics of oxygen consumption rate, kinetics of respiratory quotient.Several opportunities of measurement are presented and examples for application in various microbial systems are given.

    Notes: The cell pools of tryptophan and anthranilate, the excretion of indole-containing metabolites, and the levels of the enzymes of aromatic amino acid biosynthesis have been determined in regulatory mutants of Hansenula henricii. The strain Hg 48-2-M8 produces indoles with a maximum specific productivity of 0.37 mg/g · h at a maximum specific production value of 21 mg/g dry cell weight. This methyl-tryptophan resistant mutant possess an anthranilate synthase, whose inhibition by tryptophan is reduced. The best conditions for production of indoles are the following: 1% glucose as C-source; ammonium as N-source; pH value smaller than 4. We found that under various growth conditions 25-60% synthesized indole-containing metabolites consists of tryptophan.

NaOH treatment, gamma ray irradiation and inoculation of the material with the white rotting mold Sporotrichum pulvurulentum and combinations of above mentioned alternatives were evaluated by measuring the increase in water solubility and the susceptibility to cellulose degradation as well as by the change in the chemical composition of the material.NaOH treatment at 6% concentration and the combined treatment with NaOH at 2% and further fermentation with the mold gave the best results. Gamma irradiation up to a dose of 60 Mrad didn't show an important effect.

    Notes: The spectrophotometrical measurement of the diffuse reflectance in fermentation media has been proved useful for the analytical control of the microbial biosynthesis and transformation. A laboratory steril fermenter was fitted at an UV/VIS-spectrophotometer equipped with a diffuse reflectance attachment. The apparent absorbance ΔE8(328) of aromatic hydrocarbons and the reciprocal scattering E8(650) have been compared with parameters of the discontinuous fermentation of the yeast Lodderomyces elongisporus and the bacteria strain EB 10c on petroleum hydrocarbons as a substrat.

    Notes: During the submerged cultivation of Trichoderma sp. 414 on a medium with 2% cellulose containing plant substrats an enzyme system - exocellobiohydrolase (C1-enzyme), endogluconase (Cx-enzyme), β-glucosidase and xylase - catalizing the cellulose hydrolysis was synthesized. The process of enzyme biosynthesis by the microbial strain under the conditions of two-step cultivation in flasks was optimized.The influence of different sources of carbohydrate - avicel, micricel, maize stalk and straw - on the activity of the synthesized enzymes was studied. This activity depends on induction properties and the concentration of the used substrats. During the cultivation of Trichoderma sp. 414 on a medium containing avicel and wheat bran the activity of cellobiohydrolase reaches 40 U/ml and this of endogluconase - 520 U/ml. When the cultivation was performed on a medium containing wheat bran and straw the activity of xylanase reaches 240 U/ml.

    Notes: Some aspects of the kinetics of bioremoval of pollutants during biodegradation of swine-breeding waste waters are studied and a biphasic model is proposed. The concentration of nitrogen and phosphorus compounds during the process is followed. Following a previous contribution on the same subject, the aspects of dilution of those effluents are discussed, and it is proposed to treat raw effluents, in spite of the reduction of possible biostatic actions. Dilution will be acceptable only if strong inhibitory effects are observed.

    Notes: Differential values of the specific ethanol production rate \documentclass{article}\pagestyle{empty}\begin{document}$$ v_{(t)} = \frac{1}{{x_{(t)} }} \cdot \frac{{dP}}{{dt}} $$ \end{document} can be calculated exactly from experimental batch fermentation process data by use of a nonlinear regression programme. The method used is based on the fact, that the function P = f(t) can be approximated by an exponential equation. The specific ethanol production rate is calculated then from the first differential derivation of this equation using the appropriated values of actual biomass concentration. For two strains of Saccharomyces cerevisiae a linear and nonlinear kinetic pattern, respectively, was found for product formation. This result can be explained by a simple mathematical relation according to ν=ν0 - a . Pb,in which the exponent becomes 1 in the case of linear kinetic pattern.

The further underline the necessity of parallel short- or longtime measurements with several charges. Under these conditions, techniques on the basis of gasanalysers are profitable for the planning of measuring systems for photosynthesis with the ability to continuous, simulataneous in vivo measurements of photosynthetic rates of O2-output and CO2-fixation in open gas-stream with parallel charges. A measuring system, which meets these requirements, is demonstrated.

    Notes: A review of errors, which can influence the measured data of electrochemical oxygen sensors (OS) in fermentation technique is presented.The specifities of various sensor constructions are pointed out. References are given for selection of sensors to application in fermenters. Influences on the measured values can take place by fermentation conditions, arrangement of sensors in the fermenter and composition of the fermentation broth. The oxygen measurement in the multi-phase system of fermentation fluids can cause a remarkable deviation of the measured values to the real ones.Practical hints are given for use, calibration and sterilization of OS. Restricting conditions for measurements with this sensors are enumerated.

    Notes: When single-cell suspensions prepared from embroyonic day 8 (E8) chick sensory ganglia are incubated with nerve growth factor (NGF), anti-NGF antiserum, and complement, an NGF-dependent cytotoxic kill of 20 (±3)% of the ganglia cells is observed. This percentage is increased by a factor of two when only the neuronal cells are tested. No kill is observed on the nonneuronal cell population representing 50% of the ganglia dissociate. When E8 sensory ganglia cells are cultured in the presence of NGF following cytotoxic kill, the large, phase-bright NGF-reponsive neurons are missing from the culture. These results indicate that the cells recognized in the cytotoxicity assay have to carry NGF-binding sites of type I, which is the one with the higher affinity of the two types of NGF-binding sites (I and II) present on sensory ganglia cells. This conclusion is further supported by the following data: (a) half maximal cytotoxicity is reached already at a concentration of NGF which is below the KD of binding site I; (b) a washing step which removes all NGF bound to type II receptors while leaving a high percentage of type I receptors occupied has no effect on the percentage of ganglia cells killed.Using the cytotoxicity assay the presence of high-affinity binding sites of type I can be demonstrated on sensory ganglia cells from E8 chick embryos but not from E4 embryos and not on liver and heart cells from E8 embryos. Further, type I receptor-bearing cells were detectable in the brain using this assay. At E8, NGF receptors could be detected on cells of the forebrain and the tectum but not on brain stem cells. Cytotoxic kill of forebrain cells was found to be especially high at E8 and E9, and decreased by E10.

    Notes: To investigate the dynamics of membrane processes that may be integral components of specific transmembrane signaling events we have synthesized several novel paramagnetic probes and their photoreactive counterparts. The structure of these probes was designed to (1) restrict “flipping” across the membrane bilayer; (2) contain paramagnetic or photoreactive moieties that could be placed at specific depths within the bilayer; (3) provide information about membrane structure as well as dynamics of protein movement; and (4) in the case of the photoreactive probes, be of high specific radioactivity.The molecules described in this paper consist of amino acid, dipeptide, or carbohydrate groups attached to arylazide- or nitroxide-bearing fatty acids. The synthesis and initial characterization of these membrane probes is described.

    Notes: In the presence of the spindle poison Colcemid in the culture medium to prevent anaphase, approximately 20% of Chinese hamster metaphase cells were converted to micronucleated cells during 7 h. In the micronuclei the chromosomes had become enclosed by a nuclear envelope (NE). In the light-microscope the micronuclei were of two kinds: with either visible chromatids or with decondensed chromosomes. In the electron microscope (EM) the spatial relationship of the NE to the chromatin was of two kinds only in the presence of Colcemid. In about 90% of the micronucleated cells the spatial relationship was normal, ie, the NE was immediately adjacent to the chromatin. In the remaining cells, the NE was distended so that the outer NE was separated from the inner one. In the presence of the drivalent cation chelator, (ethylenedinitrilo) tetraacetic acid (EDTA) or the Ca2+-chelator [ethylenebis (oxyethylenenitrilo)] tetraacetic acid (EGTA), in addition to Colcemid, the amount of cells with micronuclei increased to 40%. The light-microscope appearance was the same as that found in the absence of the chelating agents. However, after Colcemid plus EGTA, EM revealed that only about 50% of the micronucleated cells had NE that was immediately adjacent to the chromatin and about 10% of them had distended outer NE. In the remaining 40% a third kind of spatial relationship was seen: the NE was intact but most of it was not adjacent to the chromatin. Furthermore, this type of micronucleus often contained mitochondria within the confines of NE. Thus, Ca2+ and possibly Mg2+ may regulate the rate of formation of the NE and also its ultrastructural relation to the chromatin. Mitochondrial function also appears to be involved in this relationship. In the presence of chloramphenicol (CAP), an inhibitor of mitochondrial protein synthesis, in addition to Colcemid, only about 50% of the micronucleated cells exhibited the normal relationship. The outer NE was separated from the inner NE in about 46% of the micronucleated cells and the third kind of NE-chromatin relationship was observed only in 2%. In the case of the third kind of relationship produced by CAP, inclusion of mitochondria within the micronuclei was not observed, in contrast to the finding with EGTA.

    Notes: Irreversibly sickled cells (ISCs) are sickle erythrocytes which retain bipolar enlongated shapes despite reoxygenation and owe their biophysical abnormalities to acquired membrane alterations. Freeze-etched membranes both of ISCs produced in vitro and ISCs isolated in vivo reveal microbodies fixed to the internal (PS) surface which obscure spectrin filaments. Intramembranous particles (IMPs) on the intramembrane (PF) surface aggregate over regions of subsurface microbodies. Electron microscopy of diaminobenzidine-treated ISC ghosts show the microbodies to contain hemoglobin and/or hemoglobin derivatives. Scanning electron microscopy and freeze-etching demonstrate that membrane-hemoglobin S interaction in ISCs enhances the membrane loss by microspherulation. Membrane-bound hemoglobin is five times greater in in vivo ISCs than non-ISCs, and increases during ISC production, paralleling depletion of adenosine triphosphate. Polyacrylamide gel electrophoresis of ISC membranes shows the presence of high-molecular-weight heteropolymers in the pre-band 1 region, a decrease in band 4.1 and an increase in bands 7, 8, and globin. The role of cross-linked membrane protein polymers in the generation of ISCs is discussed and is synthesized in terms of a unified concept for the determinants of the genesis of ISCs.

This method has been applied to an investigation of the outer cell surface structure of chicken embryo and hamster fibroblasts. Coelectrophoresis in sodium dodecyl sulfate-polyacrylamide gels of probe-labeled membrane proteins purified from baby hamster kidney fibroblasts have shown that 7 major protein groups of different molecular weight are exposed on both control and Rous sarcoma or polyoma virus-transformed cells. Moreover, the transformed cells display a nonvirion component of 80-100 k daltons that is not labeled by the probe in normal cells. In fibroblasts transformed by a temperature sensitive Rous sarcoma virus mutant, that transforms at 37°C but not at 41°C, the expression of this component is related to the expression of the transformed phenotype.

    Notes: Bacillus stearothermophilus cells vary the lipid fatty acid composition of cytoplasmic membranes with growth temperature. Spin label studies of such membranes have been interpreted to indicate lateral lipid phase separations at the growth temperature. We have now used freeze-fracture electron microscopy to confirm the spin label studies. Freeze-fracture faces of protoplasts indicate slight but distinct protein aggregation at the growth temperature. Aggregation increases rapidly with decreasing quench temperature in wild-type cells. In contrast we were unable to demonstrate extended protein segregation in membranes of a temperature-sensitive mutant that contains more than 58% branched fatty acids.Storage of protoplasts for prolonged times below the lipid phase transition results in the appearance of corrugated fracture faces with 300- to 500-Å repeat patterns, although this organism does not synthesize lecithins.

    Notes: Isolated melanophores of the angelfish, Pterophyllum scalare, have been used in a morphometric analysis and a quantitative study of their microtubule system. Using transverse sections spaced at regular intervals, the changes associated with the process of pigment aggregation have been determined. Upon the concentration of pigment granules in the central cell region, almost half of the cytoplasmic portion is also withdrawn from the peripheral cell regions. Counts of microtubules within a cell sector in cells with pigment aggregated and dispersed, respectively, reveal (a) a constancy of the number of microtubules in this sector regardless of the distance from the cell center, and (b) a reduction of microtubule number in cells with pigment aggregated by about 58%. On the basis of these counts, the total number of microtubules has been calculated. In the dispersed state, about 2,400 microtubules extend between the center and the periphery of the cell, while their number is about 1,000 in the aggregated state.Using a 13-protofilament model of a microtubule and relevant data on size and molecular weight of microtubule subunits, the amount of tubulin present as microtubules is calculated. In the average, the cells contain 1.95·108 monomers corresponding to 1.78·10-8 mg tubulin. A tentative estimation of the concentration of tubulin inside a melanophore yields values of 6.1 mg/ml for the whole cell and 16.5 mg/ml for the cytoplasm alone (excluding membrane-bound organelles). Based on this estimation, a comparison, with microtubule assembly in vitro is made.

    Notes: The red cell membrane has an asymmetric arrangement of phospholipids. The amino-phospholipids are localized primarily on the inner surface of the membrane and the choline phospholipids are localized to a large extent on the outer surface of the membrane. Evidence is presented based on the use of covalent chemical probes in sequence that the red cell membrane contains heterogeneous domains of PE and PS and that the transport systems for Pi and K+ are asymmetrically arranged. Certain amino groups of PE, PS, and/or protein localized on the outer membrane surface are involved in Pi transport and certain amino groups of PE, PS, and/or protein localized on the inner surface of the membrane are involved in K+ transport.Cross-linking studies with DFDNB show that the cross-linked PE-PE molecules are rich in plasmalogens. This suggests that clusters of plasmalogen forms of PE occur in the membrane. Both PE and PS are cross-linked to membrane protein. These PE and PS molecules contain 24-28% 16:0 and 18:0 fatty acids and 12% fatty aldehydes. PE and PS molecules are cross-linked to a spectrin-rich fraction. It is proposed that the binding of spectrin to membrane PE and PS may help anchor spectrin to the inner surface of the membrane and regulate shape changes in the cell.K+-valinomycin forms a complex with TNBS and converts it from a non-penetrating proble to a penetrating probe. Valinomycin enhances K+ leak and Pi leak in the red cells. SITS inhibits completely the valinomycin-induced Pi leak and inhibits partially the valinomycin induced K+ leak. Valinomycin and IAA have additive effects on Pi leak. Ouabin has no effect on basal or valino-mycin-induced Pi leak. These data suggest that Pi leak and K+ leak occur by separate transport systems.In summary, the amino-phospholipids in the red cell membrane are asymmetrically arranged; some occur in clusters and some are closely associated with membrane proteins. Amino-phospholipids also are believed to bind spectrin to the inner surface of the membrane and also may play a role in cation and anion leak.

    Notes: Interactions between spectrin and the inner surface of the human erythrocyte membrane have been implicated in the control of lateral mobility of the integral membrane proteins. We report here that incubation of “leaky” erythrocytes with a water-soluble proteolytic fragment containing the membrane attachment site for spectrin achieves a selective and controlled dissociation of spectrin from the membrane, and increases the rate of lateral mobility of fluorescein isothiocyanate-labeled integral membrane proteins (〉 70% of label in band 3 and PAS-1). Mobility of membrane proteins is measured as an increase in the percentage of uniformly fluorescent cells with time after fusion of fluorescent with nonfluorescent erythrocytes by Sendai virus. The cells are permeable to macromolecules since virus-fused erythrocytes lose most of their hemoglobin. The membrane attachment site for spectrin has been solubilized by limited proteolysis of inside-out erythrocyte vesicles and has been purified (V). Bennett, J Biol Chem 253:2292 (1978). This 72,000-dalton fragment binds to spectrin in solution, competitively inhibits association of 32P-spectrin with inside-out vesicles with a Ki of 10-7M, and causes rapid dissociation of 32P-spectrin from vesicles. Both acid-treated 72,000-dalton fragment and the 45,000 dalton-cytoplasmic portion of band 3, which also was isolated from the proteolytic digest, have no effect on spectrin binding, release, or membrane protein mobility. The enhancement of membrane protein lateral mobility by the same polypeptide that inhibits binding of spectrin to inverted vesicles and displaces spectrin from these vesicles provides direct evidence that the interaction of spectrin with protein components in the membrane restricts the lateral mobility of integral membrane proteins in the erythrocyte.

    Notes: The D-glucose transporter from human erythrocytes has been purified and reconstituted by Kasahara and Hinkle (J Biol Chem 252:7394-7390). Using a similar purification scheme, we have isolated the protein with 65% of the extracted phospholipid at a lipid-protein ratio of 14:1 by weight. The KD (0.14 μM) and extent (11 nmoles/mg protein) for binding of 3H-cytochalasin B was determined by equilibrium dialysis. Glucose was a linear competitive inhibitor of binding of cytochalasin B, with an inhibition constant of 30 mM. To further characterize the protein, samples were filtered in the presence of sodium dodecyl sulfate (SDS) through Sepharose 6B to remove 95% of the lipid followed by filtration of Sephadex G150 to remove the remaining lipid and a contaminating amount of a minor, lower-molecular-weight protein. This preparation contains only 24% acidic and basic amino acids. The protein also contains 5% neutral sugars (of which 3% is galactose), 7% glucosamine, and 5% sialic acid.

Radioactive peptides have been prepared by 14C-carboxymethylation and chloramine T-catalysed 125I iodination. Maps of both sets of peptides demonstrate a marked similarity between the two parent polypeptides.

    Notes: Swiss 3T3 and C3H-M2 cells have a greater mitogenic response to epidermal growth factor (EGF) than do C3H-10T1/2 cells. The latter cell line, however, has a number of EGF receptors per cell intermediate between the two cell lines that have a more vigorous response to EGF. Scatchard analysis of binding data indicate that all three cell lines have one class of EGF receptor, with indistinguishable affinity for the ligand. When exposed to 10-nM EGF all three cell lines “down-regulate” their EGF receptors with the same time course, and to the same precentage of initial receptors.

    Notes: The effects of calcium ion on the adenylate cyclase system was studied in isolated, renal basal-lateral plasma membranes of the rat. Bovine parathyroid hormone (bPTH) and a guanyl triphosphate analogue, Gpp(NH)p were used to stimulate cyclase activity.Under conditions of maximal stimulation, calcium ions inhibited cyclic adenosine monophosphate (cAMP) formation, the formation rate falling exponentially with the calcium concentration. Fifty percent inhibition of either bPTH- or Gpp(NH)p-stimulated activity was given by approximately 50 μM Ca++. Also the Hill coefficient for the inhibition was close to unity in both cases. The concentration of bPTH giving half-maximal stimulation of cAMP formation (1.8 × 10-8 M) was unchanged by the presence of calcium.These data suggest that calcium acts at some point other than the initial hormone-receptor interaction, presumably decreasing the catalytic efficiency of the enzymic moiety of the membrane complex.
