    Notes: Based on the partial differential equations for the transport of enthalpy and material a calculation model for the examination of the change of temperature and concentration in axial and radial direction was developed for the continuously working enzyme reactors. The pecularity of the kinetics of processes which are catalyzed by enzymes was considered due to the reaction velocity in modelling. In order to induce the numeric solution, an explicit method of differences was selected, the applicability of which was proved under consideration of appropriate initial and marginal conditions.

    Notes: The effective means of microbial culture monitoring is the measurement of low-inertial parameters (respiration rate, rates of supply of alkali for pH maintenance and the limiting substrate) and utilization of computer on line with fermenter for recalculation of these rates into the instant values of mass and energy cell yields, specific rates of cell growth and substrate and oxygen consumption, using the method of mass-energy balance. In this paper, the equations of mass-energy balance are presented both in general form and in the form of numerical algorythms for computer programming.The installation for automation of microbial cultivation experiment is described. Experimental data are presented which indicate the effectiveness of the method of indirect measurement of cell biomass yield and specific rates of physiological processes.

    Notes: Extracellular production of laccase (E.C. 1.10.3.2) by Polyporus versicolor was studied on lignin, complex and defined media. Although the production of enzyme was more on lignin the specific activity was more on malt extract. Laccase was produced on all the media tested i.e. phenolic compounds, lignins and sugars alone, and in combination with malt extract, excepting salicylic acid. On single source media maximum yields of enzyme were obtained on polyfon followed by resorcinol, reax and lignin. As compared to phenolic compounds the enzyme production was low on sugars. Addition of malt extract enhanced the enzyme yield which was maximum in the case of lignin followed by gallic acid.

    Notes: The ability of yellow streptomyces to synthesize lipids and carotinoids was examined.It has been established that the content of lipids in the mycelium varied from 9.7 to 57.1% depending on the species and the strains. Cultures belonging to the species Str. nephtigulosus, Str. ambofaciens, Str. citreus showed carotene-synthesizing abilities. In the content of the lipids and carotinoids of the examined cultures were found our fractions possessing biological activity. The fatty acid content of the examined cultures was characterized by the saturated and unsaturated acids containing an even and odd amount of the carbon atoms (from C11 to C18) and a normal and branched chain (iso-C16 and iso-C17).

    Notes: “Solid-substrate” fermentation developed in the Orient is a very useful fermentation method. It is presently used to produce a variety of foods, beverages and related products. Solid-substrate fermentation products utilizing fungi including soy sauce, miso and tempe, ontjom, sake, and bread have been produced for centuries at the home and village level. They are examples of economical methods of preserving and improving the flavor, texture and nutritive values of cereal/legume substrates. “Solid-substrate” fermentation is also applied to animal products such as milk to produce Roquefort and Camembert cheeses which diversify the food flavors available to man“Solid-substrate”fermentation has certain advantages. The substrate is concentrated; the product can be extracted with relatively small quantities of solvent; the product can be easily dehydrated; moisture level can be controlled favoring the desired organisms; enzyme concentration is generally higher than is submerged fermentation; product concentration is generally higher than in submerged cultures; it is the only technique that yields true mushroom fruiting bodies and it can be used not only for production of crude enzyme concentrates (koji) but also for raising the protein content of high starch substrates. It also can be used to increase the content of vitamins at low cost. Disadvantages of “solid-substrates”from the modern industrial processing view point are the greater difficulty of handling solid substrate and the greater difficulty of controlling the fermentation parameters, temperature, pH and oxygen, and rate of microbial growth compared with liquid submerged fermentations.

    Notes: Transformation of mass indexes of microbial growth efficiency (the maximal yield and maintenance coefficient) into energetic ones is desribed. Energetic yield is expressed in terms of ATP expenditures for constructive metabolism, cell maintenance and other physiologo-energetic coefficients. From experimental data obtained during H. polymorpha cultivation on glucose, ethanol and methanol, it has been found that the growth-independent fraction of ATP expendditure for cell maintenance and futile dissipation (CMFD) is the same in all the three cases. The coefficient of the growth-dependent CMFD on glucose and methanol is approximately the same. Maximal energetic biomass yields on these substrates have been measured and the background of their differences is discussed.

    Notes: Sephadex LH-20 was tested for a potential application in the enrichment of ubiquinone-9 from a lipid-hydrocarbon-extract. The lipid-hydrocarbon-extract was isolated from yeast grown on gas oil (b. p. 240 to 380°C) as carbon-source. The main components of the lipid-hydrocarbon extract were phosphatides, glycerides, fatty acids, and hydrocarbons of gas oil. By use of Sephadex LH-20 and CHCl3/CH3OH (2:1)as eluent it is possible to concentrate the whole ubiquinone-9 in a special fraction both directly from the lipid-hydrocarbon-extract and from the aceton-soluble part of the extractThe concentration of ubiquinone in the special fraction was ten times higher than in the raw-material and the hydrocarbons were separated completely.Ubiquinone-9 can be obtained by a simplified separation from the enriched fraction.

    Notes: A comparative study of 9 yeasts namely, Candida blankii,C.humicola,C.ishiwadae,C.rhagii,C.tropicalis,Hensenula subpelliculosa,Saccharomyces cerevisiae,Trichosporon cutaneum and Tr.pullulans was carried out for the production of extracellular and cell bound β-glucosidase using cellobiose as the substrate. Trichosporon cutaneum was found to be the best extracellular as well as cell bound β-clucosidase producer and the former activity was more than the latter. In the rest of the yeasts most of them showed more cell bound β-glucosidase as compared to the extracellular.

    Notes: The optimal parameters for the cultivation in 10-l fermenters of a mutant strain Humicola lutea 120-5 were established:temperature 30°C, inoculum size 6%, inoculum age 24 h, aeration rate 0,6 vol/vol · min, medium agitation 620 rpm and cultivation time 72 h. A maximal proteolytic activity of 2000 µg tyrosine liberated from 2%casein ml-1 culture filtrate min-1 at pH 3.0 and 40°C was obtained under the fixed conditions. α-Amylase biosynthesis during the cultivation of H. lutea 120-5 was observed but it was insignificant to the 72nd h. It is demonstrated that starch can be used as alternative to glucose carbon source. It is proved that the mutant strain H.lutea 120-5 produced two acid proteinases.

    Notes: The calculation and scale-up of fermentation processes need kLa as one of the most important engineering data. There are two methods to determine kLa depending on power input, aeration rate and the properties of the fermentation broth: static with a balance between air supply and exit, dynamic gassing out with following the changes of dissolved oxygen concentration during periods of air off and a following air on.Within early intervals of fermentation time the data from both methods agree well, while for later time intervals the dynamic method always gives much lower values for kLa than static. The only explanations for this phenomenon are quick changes in the oxygen metabolism or an enzymatic storage of oxygen. For both gassing out and saturation period it is possible to calculate the same absolute amounts of this additional oxygen.

The main components of the lipid-hydrocarbon extract were hydrocarbons of gas oil (B. p. 513 to 653 K)phosphatides, glycerides, and fatty acids.By adsorption on silicagel (1 part silicagel to 2-3 parts of lipid-hydrocarbon extract dissolved in hexane) were ergosterol and ubiquinone-9 enriched and nearly completely separarated from hydrocarbons and phosphatides, partly from glycerides and fatty acids. Ergosterol and ubiquinone-9 can be obtained by a simplified separation from the enriched fraction.

    Notes: Generally, methylotrophic bacteria grow optimally in a pH range between 6 and 7.2. The assimilation of methanol can take place via several pathways. Acetobacter methanolicus preiers an acidic pH range for growth, the pH optimum is about 4, and it uses the FBP variant for methanol assimilation.The latter is interesting from a regulatory point of view because phosphofructokinase disappears during growth on glucose, which is assimilated via the hexosemonophosphate pathway. Since Entner-Doudoroff enzymes and phosphoketolase are absent in A. `ethanolicus as well as in non-methylotrophic Acetobacter and Gluconobacter species phosphofructokinase becomes a key enzyme of the assimilation of methanol.Although A. methanolicus uses the hexulosephosphate pathway the growth yield on methanol is smaller than with other “hexulosephosphate pathway bacteria” e. g. with obligate methanol assimilating bacteria.At first sight it may appear that the acidic optimum pH is responsible for the smaller growth yield and the discrepancy between the experimental and predicted values.The relationship between the dependence on and the protection from, high external proton concentration on the one hand and the causes of the low growth yield on the other are discussed. Accordingly, A. methanolicus and another heterotrophic acidophiles seem to be acidoresistant above all, their machinery guaranteeing the protection from the high proton concentration is responsible for the acidophily and the low growth efficiency is caused by a simple respiratory chain.

    Notes: An universal consideration is proposed for calculation of the heat exchange of industrial fermenters at which the energy-intake is usual as main parameter in the formulas. On this way mathematical models developed, useful for heat transport during mixing by means of mixer with and without aeration, during boiling and for bubble column, are declared. With these models it is possible to describe Non-Newtonian fluids, which rheological conditions in the concrete apparatus is formed by the intaken energy. Stable rubbers are proposed for treatment of high viscous fluids and a half-empirical model is offered for its calculation.

    Notes: Sorption of Penicillium cellulase onto cotton linters samples differing in physical structure and onto KLASON- lignin from spruce has been investigated by determining cellobiase, CMCL- and ‘linters-activity’ as well as protein content of a cellulase culture filtrate before and after contact with the sorbent at different temperature for different time intervals. CMCL- and cellobiase-activity were found to be reduced much less by the sorption process onto the cellulose samples than ‘linters-activity’ and protein content. Sorption increased in the order: untreated linters 〈 mercerized linters 〈 wet fibrillated (‘colloidal milled’) linters. With the KLASON-lignin also a considerable sorption of the cellulase system was observed. Results are discussed with respect to preferential sorption of enzyme components.

    Notes: The given paper represents formulae for calculation of oxygen consumption and of carbon dioxide formation in a microbial culture obtained from corrected measuring data of gas balance measurements. Basic relations between these measuring data the respiration quotient RQ were derived. The influence of measuring error to the accuracy of determination of oxygen consumption and of carbon dioxide formation at the analysis of gas concentrations and of air flow is discussed. It is shown that the determination of carbon dioxide formation rate is possible in many cultivation regimes with better accuracy than the determination of oxygen uptake rate.

    Notes: The toxic fractions were isolated from unsuitable molasses,not accepted for citrie acid fermentation. When the strains of A. niger: Z-208, R-8, P-16, R-65/4, all good producers of citric acid, were cultivated on the medium containing such molasses, the growth patterns were altered and the strains lost their acid-forming properties.After dialysis of the molasses samples fractions of low and high molecular weight were obtained but the latter had no effect neither on growth nor on fermentation patterns. The low molecular part, responsible for the toxicity of molasses against A. niger, was fractionated with Sephadex G-25.In such a way a low coloured fraction which inhibited the growth of A. niger strains could be separated. However there were no toxic compounds in the fractions with the highest values of absorbancy. The susceptibility of the strains,to some extent, depended on their ability for adaptation to the toxic fractions of molasses.

    Notes: Starting from cellulose samples prepared from cotton lintes and differing in lattice type, crystallinity and fibrillar morphology, enzymatic hydrolysis of fibre cellulose has been studied employing complete enzyme systems from Trichoderma, Sporotrichum, Gliocladium and Penicillium as well as isolated endo- and exo-1,4-β-glucanases from Trichoderma reesei and Sporotorichum pulverulentum. The effect of hydrolysis was characterized by content of reducing sugars (RS) and of glucose in the hydrolyzate as well as by DP and X-ray diffraction pattern of the residues. With all the complete enzyme systems investigated about the same order of degradability was found with a series of substrates differing in physical structure. The hydrolysis effect of cellulase from S. pulverulentum proved to be sensitive to the gas atmosphere above the system (N2 or O2), probably due to the interaction of an O2-atmosphere with the activity of the cellubiose-oxydase existent in the system. Isolated endoglucanase from S. pulverulentum and T.reesei still led to a considerable formation of RS and glucose, a corrosion of the fibre surface and a significant descrease in DP. Influence of substrate physical structure was rather small with regard to RS, but still considerable with regard to residue-DP.The effect of isolated exoglucanases depends largely on the chemical structure of the cellobiohydrolase in question, as demonstrated with the two samples “CBH I” and “CBH II” from T. reesei. With CBH I, rather resembling endo-glucanase behaviour, a considerable formation of RS and a significant corrosion of the fibre surface has been observed. On the other hand, only negligibly small amounts of RS were formed by CBH II.Results are discussed with regard to the complex mechanism of cellulase action on fibrous cellulose and with regard to the relevance of different parameters of physical structure of cellulose in connection with enzymatic hydrolysis. A remarkable acceleration of the Cellulose III → Cellulose I lattice transition due to chain fragmentations in the presence of cellulase can be concluded the experiments.

The main advantages of the instrument are a wide range from 0.1 to 200 g biomass/l the sterilizability, the self-cleaning of the cuvette-window, a favourable design and a little maintenance.     Notes: Short path distillation was used for the enrichment of ubiquinone-9 from a lipid hydrocarbon extract. The main components of the lipid hydrocarbon extract were hydrocarbons of gas oil (b. p. 513 to 653 K), phosphatides, glycerides, and fatty acids. Phosphatides were isolated by extraction with acetone before distillation. The acetone soluble fraction was distilled by a pressure of ≦ 0.5 kPa and a temperature of 433 to 453 K. The concentration of the ubiquinone-9 in the residue of distillation was three times higher than in the acetone soluble fraction.Ubiquinone-9 can be obtained by a simplified separation from the enriched fraction.

    Notes: Studies of the biomass production during a continuous cultivation of yeasts on a nutrient medium, prepared from a hydrolysate from the production of microcrystalline cellulose, have been carried out. A new strain of yeasts has been used. Its cultivation has been achieved without addition of biostimulators to the nutrient medium in spite of their absence in the initial hydrolysate. Practically a complete assimilation of sugars has been achieved at high dilution rates (D = 0.25 to 0.50 h+1). The yield of biomass achieved is above 50% compared to the initial sugars and it contains 48.89% true protein. The results obtained offer the possibility of a complex utilization of the products of cellulose hydrolysis in the production of microcrystalline cellulose with a realization of a waste free technology.

    Notes: A systematic concept in bioprocess analysis and design is presented according to an integrating strategy as basis for a biotechnological methodology. A comparison illustrates analogies and significant differences between chemical and biological processes.The study of the interactions between environments and organisms includes influences of physical transport phenomena and also enzyme and metabolic regulations. In both cases the macroscopic principle with a formalkinetic approach is recommended for quantification purposes. The determination of the characteristic-time-regime with characteristic rate constants makesit possible to simplify mathematical modeling using the concepts of the rate-determining-step and quasistationarity. Finally, guidelines are summarized for the use of unstructured and structured kinetic models.

    Notes: The method of extended cultivation was studied mathematical-analytical by a simple mathematical model. It was reflected on the control algorithm of feeding rate. Because corresponding to the model the substrate is consumed only for the biomass growth, the feeding is to control proportional to the biomass variation. It is shown that the biomass concentration in this case is independent of outflow and approaches a limit value. Relations between biomass growth and feeding rate of limiting substrate are given for all cases. In particular for the cyclie fed batch, it is shown that assuming periodic cycles the cultivation volume at the end of the cycles approaches a maximum value defined only by initial volume, specific growth rate and cycle period.

    Notes: The extracellular complex of β-glucanases produced by the mould Trichoderma viride hydrolyzes β-1,3-; β-1,4- and β-1, 6-bonds of β-glucans, as well as mixed β-1,3- and β-1,4-glycosidic linkages. This complex contains also xylanase. The enzymes were isolated from liquid culture medium by centrifuge techniques, concentration and precipitation with acetone. Isolation and purification of β-1,3-glucanase was carried out according to a procedure involving filtration on Bio-Gel P-100, DEAE-Sephadex A 50 and CM-cellulose C-11 chromatography, ultrafiltration and selective adsorption on xylan. The homogeneity of the enzyme was determined by polyacrylamidegel electrophoresis. The purified homogeneous preparation of the isolated β-1,3-glucanase from Tr. riride was subjected to detailed characterization. Amino acid composition, molecular weight and optimum conditions for the enzymatic activity of the protein were determined. The isolated enzyme was shown to be highly specific to substrates with β-1,3-glycosidic linkages; the rate of degradation was found to be proportional to the degree of polymerization of the substrate.

    Notes: Because the interior scale of turbulence in reinforced bioreactors is essentially greater than the particles of bacteria or yeasts, the mass transfer occurs at the particles only in consequence of the molecular diffusion and depends on the concentration of the soluted material only. As a decided criterion for the transmission of scale in such processes is considered the volumetric oxygen transfer coefficient KLaThis article practises a theoretical analysis of the characteristic hydrodynamic conditions in bubble columns and in agitators: the content of gas, the specific area of mass transfer and the coefficient of mass transfer KL. These conditions are joined with the evaluation of the KLa value. Resultant the data of calculation hence it follows a computational determination of the KLa value by means of the physical matter values, the construction of devises and the process conditions.

    Notes: Abstract The growth of white clover (Trifolium repens L.) is severely inhibited by boron starvation, but a foliar treatment with boric acid can transitorily alleviate the deficiency symptoms. The 10B(n ,α)7 Li nuclear reaction has been used to study boron transport in the plant after foliar application. More than 98% of the boron supplied remained at the point where it was applied to the leaves, and less than 2% was useful to the growth of the treated plant. This small “efficient” portion of boron was quite mobile. It was distributed to the different parts of the plant, then was transferred from the oldest parts to the newly formed leaves. Physiological and agronomical implications of these data are discussed.

    Notes: The behaviour of the mycelium forming mold Aspergillus flavus in packed column culture in comparison to conventional fermentation systems (surface culture and submerged culture) was investigated.     Notes: Es wird die Immobilisierung eines Milchgerinnungsenzymes (als Präparat) aus der Kultur des niederen Pilzes Mucor pusillus Lab. sp. renninus durch Adsorption und kovalente Bindung an einen anorganischen Träger beschrieben. Die Adsorption wurde an Amino- und Karboxylsilochrom und die kovalente Bindung an Azid-Derivaten von Silochrom untersucht. Proben mit einer hohen Aktivität wurden sowohl durch Adsorption (1 200 Einheiten/g Präparat) als auch durch kovalente Bindung (890 Einheiten/g Präparat) gewonnen. die Möglichkeit des Einsatzes von Immobilisierten Präparaten zur mehrfachen Gerinnung von Milch wird erörtert. Die stabilisten Präparate wurden durch die Methode der kovalenten Immobilisierung hergestellt.

    Notes: A mathematic model of microorganism colony growth on solid media was worked out, the substrate diffusion taken into account.The sample being Sacch. cerevisiac yeast correspondence of experimental and calculated data was demonstrated. Sacch. cerevisiac yeast specific growth rates on various substrates were determined. Some parameter effect on colony growth kinetics on a solid medium surface was shown. Two phase of limitation were detected in microorganism colony development - kinetical and diffusional ones. On the basis of calculations, theoretical suppositions and experimental data a conclusion was made that solid fermentation required the finest grinding possible, taking into account growth limitation by diffusion. Besides, dense seeding of all the medium surface with cells was found important.

    Notes: The uptake of hydrocarbons by yeasts is measured by gravimetric determination of intracellular accumulated N-free compounds as well as by detection of tritium-labeled hydrocarbons. The aim of the investigations is to compare the uptake of n-alcanes and aromatics with respect to the conditions of yeast fermentation on gas oil. Aromatics in comparison to n-alcanes are taken up in very little amounts. The uptake of n-alcanes is dependent on the chain length. According to increasing chain length in the range of C10 - C16 the velocity of uptake is increased whereas the optimal pH-value is decreased.In the presence of dimethylsulfoxide the uptake of n-alcanes is markedly increased whereas the uptake of aromatics is decreased. A mechanism of hydrocarbon uptake is discussed characterized by obligate participation of metabolic active cells and hydrocarbon selectivity.

    Notes: On the basis of the combustion enthalpy of microbial biomasses carbon substrates for the aerobically heterotrophic growth of microorganisms can be divided into substrates with energy excess (e.g. ethanol, n-alkanes) and with energy deficit (or carbon excess, e.g. acetate, glucose). By combining substrates having different carbon energy ratios from both groups in a certain mixing proportion an improvement of the yield coefficients linked to a reduction of the specific heat production (kJ/g dry weight), an increase of the growth rate (h-1) and the velocity of the substrate utilization ought to be possible on condition that the co- or auxiliary-substrate does not cause inhibition or (catabolite-) repression effects.There are some examples corroborating the theoretical concept, e.g. the combinations ethanol/sucrose-Candida utilis, methanol/xylose-Hansenula polymorpha. In each case the specific growth rate and the yield coefficient on the mixture were higher than on any single substrate of the combination.Thermochemical methods are suitable for the optimizing the substrate mixing proportion.

    Notes: A heat-sensitive, trypsin-sensitive factor that promoted growth and spreading of cells in serum-free, hormone-supplemented medium was partially purified from human serum. The major portion of the proteins in these preparations migrated upon SDS-polyacrylamide gel electrophoresis with a mobility consistent with molecular weights between 60,000 and 90,000. The spreading activity, which we have termed serum spreading factor, stimulated growth and spreading of a wide variety of cell types. The serum spreading factor was similar to fibronectin in that it showed an affinity for the plastic cell culture substrate but was shown to be distinct from fibronectin by several criteria. This factor may prove useful in studies of cell attachment and spreading and in studies of the relationship of cell shape and cell proliferation.

    Notes: The histone compositions and average distance between nucleosomes have been determined for F9.22 and PSA1 murine embryonal carcinoma cell lines, for primary extra-embryonic endoderm derived from the in vitro differentiation of PSA1 embryonal carcinoma cells, and for two long-term extra-embryonic endodermal cell lines. A change in the relative proportions of two forms of the H1 histones (H1A and H1B) was found to correlate with the extra-embryonic endodermal differentiated phenotype. The embryonal carcinoma cells had a ratio of H1A/H1B of 1.49 or greater. In contrast, extra-embryonic endoderm from either cell lines or freshly isolated from differentiating embryonal carcinoma cell cultures had a ratio of H1A/H1B of less than 0.9. Partial peptide mapping of gel purified H1A and H1B suggest the two proteins differ in primary structure. The nucleosome repeat length of the embryonal carcinoma cell lines was 196 bp of DNA. Primary extra-embryonic endoderm was found to have a value of 205 bp, but the long-term extra-embryonic endodermal cell lines had an average nucleosome repeat length of 187 bp. Since both freshly isolated primary endoderm and the long-term endodermal cell lines express differentiated functions (basement membrane glycoproteins and plasminogen activator activity), there appears to be no simple correlation between the nucleosome repeat length and the expression of these differentiated functions.

    Notes: AQR mice were immunized with H-2K and H-2 I encoded alloantigens presented by (Ax6R)F1 splenocytes. Spleen cells from these alloimmune mice were subsequently restimulated in vitro with B10.A lymphocytes and/or B10.T(6R) lymphocytes, thus presenting them with the immunizing H-2K and H-2 I alloantigens independently. When stimulated with B10.A lymphocytes, alloimmune lymphocytes develop significant cytotoxicity against the immunizing H-2K target antigens. When stimulated with a similar number of B10.T(6R) spleen cells, alloimmune lymphocytes undergo a prominant proliferative response, but develop little, if any, cytotoxicity against the immunizing H-2 K target antigens. The most efficient restimulation of cytotoxicity occurs when the alloimmune spleen cells are simultaneously restimulated by B10.A and B10.T(6R) lymphocytes. Stimulation with the immunizing H-2 I alloantigens alone is not sufficient for regeneration of detectable cytotoxic responses from alloimmune spleen populations. Stimulation with the immunizing H-2K alloantigens alone appears to be both necessary and sufficient to stimulate alloimmune cytotoxic responses. Although the immunizing H-2 I alloantigens are apparently not required to generate alloimmune cytotoxic responses, they markedly potentiate the cytotoxic responses induced by the immunizing H-2K alloantigens.

    Notes: In sub-confluent cultures of Balb/c-3T3 cells, pinocytosis rates were increased after exposure to specific growth factors (serum; platelet-derived growth factor, PDGF; epidermal growth factor, EGF). Conversely, as cells became growth-inhibited with increasing culture density, there was a corresponding decline in pinocytosis rate per cell. In order to test whether density-inhibition of pinocytosis was influenced either by the growth cycle or by cell contact independently of growth, cells were induced into a quiescent state at a range of subconfluent and confluent densities. Under such conditions, cell density did not significantly inhibit pinocytosis rate. When confluent quiescent cultures in 2.5% serum were exposed to 10% serum, the resulting round of DNA synthesis was accompanied by enhanced pinocytosis per cell, even though the cells were incontact with one another. Furthermore, in a SV40-viral transformed 3T3 cell line, both the growth fraction and the pinocytosis rate per cell remained unchanged over a wide range of culture densities. These studies indicate that density-dependent inhibition of pinocytosis in 3T3 cells appears to be secondary to growth-inhibition rather than to any direct physical effects of cell-cell contact.

    Notes: Replication of Gross strain N-tropic type C retrovirus was markedly restricted in a pluripotential undifferentiated embryonal cell line (PCC4) of murine teratocarcinoma, whereas the same virus could cause productive infection in a myoblast-derived differentiated line (PCD1) of the same tumor origin. To investigate the restriction mechanism, we compared the initial viral DNA formation in these two cell lines. Analyses by means of a modified Hirt extraction procedure and a modified Southern gel transfer method indicated that PCC4 and PCD1 cells supported the synthesis of viral DNA intermediates after inoculation of the Gross virus. In both cells, a linear DNA duplex (form III viral DNA) appeared at 4 hr, reached a maximal level at 8-9 hr, and declined rapidly thereafter, while two closed-circular supercoiled DNA duplexes (form I viral DNA) showed their appearance, increase and decline in the 8-24 hr period. During the period from 34 to 78 hr after virus inoculation, another burst of viral DNA synthesis occurred in PCD1 cells, presumably due to secondary virus infection, while at this period both form III and form I viral DNAs became undetectable in PCC4 cells. The Hirt supernatant DNAs prepared from PCD1 and PCC4 cells 10 hr after virus inoculation were equally infectious for NIH3T3 cells in a DNA transfection assay. Both PCD1 and PCC4 cells were very poor recipients for DNA transfection, although one positive result with PCD1 cells might suggest a difference between the two cell types in this aspect. These results indicate that restriction of type C retrovirus in undifferentated embryonl carcinoma cells occurs at a step subsequent to formation and maturation of viral DNA intermediates.

    Notes: The BALB/3T3 clone A31 mouse embryo cell line has been used by many investigators as a model “normal” “fibroblast” line for a variety of in vitro studies. It has been shown, however, that these cells are not “normal” because they will produce tumors within 2-4 months if 3 × 104 cells are implanted subcutaneously in BALB/c mice attached to 0.2 × 5 × 10-mm plastic plates. Previous studies also suggested that these cells were not fibroblasts because they gave rise to tumors with the characteristics of vascular endothelium not fibroblasts. We now report that BALB/3T3 (clone A31), BALB/3T3-T, a proadipocyte subclone of clone A31 cells, and six recent subclones of BALB/3T3-T cells show additional differentiation patterns when tumors derived by implantation of these cells attached to plastic plates are examined. Differentiation into pericytes, chondrocytes, and fibroblasts was observed. We conclude that the BALB/3T3 clone A31 cell line and related lines are multipotent mesenchymal cells which are capable of differentiation into a variety of cell types.

    Notes: Conditions are described for an assay that allows the percent inhibition of α-bungarotoxin binding to acetylcholine receptors by antisera and monovalent antigen-binding fragments of antibody molecules (Fab) to be determined. Anti-Torpedo californica acetylcholine-receptor antisera, prepared in New Zealand White rabbits and Lewis rats, were tested for the ability to inhibit [125I]-α-bungarotoxin binding to membrane-associated and detergent-solubilized T californica acetylcholine receptors. Similar inhibition studies were performed using rabbit antisera and antigen-binding fragments prepared against each of the four acetylcholine receptor subunits. Antisera and antigen-binding fragments prepared against intact receptor could inhibit a maximum of 50% of the α-bungarotoxin binding to solubilized receptor. The results using monovalent antigen-binding fragments indicated that the inhibition was not due to antibody-mediated aggregation of receptor molecules. Rabbits and rats immunized with receptor denatured by sodium dodecyl sulfate all produced antisera that could bind to nondenatured receptor, but none of these animals developed experimental autoimmune myasthenia gravis. These results suggest that the antigenic determinants present on acetylcholine receptors responsible for induction of experimental auto-immune myasthenia gravis are lost with sodium dodecyl sulfate denaturation. A strong correlation was also observed between the presence of experimental autoimmune myasthenia gravis in rats and rabbits and the ability of the antisera from these animals to inhibit 50% of α-bungarotoxin binding to solubilized acetylcholine receptors.

    Notes: The synthesis of the leucine-specific and LIV-binding proteins was examined in vitro in a coupled transcription/translation system using the hybrid plasmids pOX7 and pOX13 as templates. Plasmid pOX7 contains the livK gene coding for the leucine-specific binding protein, and pOX13 contains the liv J gene coding for the LIV-binding protein. Both binding proteins were synthesized in vitro as precursor forms with molecular weights approximately 2,500 greater than their respective mature forms. Conversion of the precursor forms to their mature forms occurred during post-translational incubation following synthesis in the presence of membrane. The precursor of the LIV-binding protein was processed more rapidly than the leucine-specific binding protein precursor. Processing activity could be removed from the in vitro synthesis system by centrifugation, suggesting that the processing activity was membrane associated. Restoration of post-translational processing activity was achieved by adding inside-out membrane vesicles to membrane-depleted reaction mixtures.

    Notes: A small quantity of the 125I-EGF (epidermal growth factor) bound specifically to EGF receptors on the human epidermoid carcinoma cell line A431 associates covalently. The direct linkage complex formed migrates during gel electro-phoresis as a single diffuse band of MW = 160,000-170,000. In contrast, direct linkages complexes of 160,000, 145,000, and 115,000 daltons are formed when EGF is incubated with membranes isolated from these cells; these arise from EGF receptor modification during membrane isolation. None of these modifications affected the affinity of the EGF binding site for 125I-EGF.The electrophoretic mobilities of the MW = 160,000 and 145,000 direct linkage complexes were similar to those of the major 32Pi-labeled products of the EGF-stimulated phosphorylation reaction described by Carpenter et al [Nature 276:409-410, 1978], indicating that proteolytic fragments of EGF receptors are the major phosphate acceptors in this reaction. EGF receptors on intact A431 cells accepted phosphate effectively from γ-32Pi-ATP only when the cells were permeabilized with lysolecithin. This shows that the EGF binding and phosphate acceptor sites lie on opposing faces of the membrane. When the 145,000 dalton form of receptor is labeled with EGF or 32Pi and the labeled peptides subjected to tryptic hydrolysis under identical conditions, all phosphates is lost from high molecular weight products under conditions where the EGF-receptor covalent complex is converted largely to a 115,000 dalton form. This suggests that the phosphate acceptor site lies on the cytoplasmic side of the membrane on a region of receptor extending 30,000 daltons from the 115,000 dalton fragment containing the EGF binding site.

    Notes: In this paper we have examined the growth and differentiation of the embryonal carcinoma cell line, F9, in the defined medium EM-3 at low density. We show that the growth of F9 and their differentiated cells (F9-diff) in EM-3 is strongly density dependent. At low cell densities the growth of both cell types is severely limited and most of the cells do not survive. Although this poses a problem for working with F9 and F9-diff in EM-3, it provides a convenient assay for identifying molecules that support their growth at low density. Using this assay, we have determined that laminin, a newly isolated glycoprotein of basement membranes, significantly improves the growth and short-term survival of both F9 and F9-diff. However, addition of laminin to EM-3 is insufficient to promote the clonal growth of these cell types. Our findings also indicate that laminin promotes the attachment of F9 and F9-diff in defined media. On the basis of our results, we propose an attachment function for laminin during the early stages of mammalian development.

    Notes: Nerve growth factor (NGF) is likely to exert its trophic action on dorsal root ganglion (DRG) and on sympathetic ganglion neurons by controlling a crucial function of these cells. This function would in turn regulate other cellular machineries and, ultimately, lead to the traditional NGF consequences, such as survival and neuritic growth. A corollary of this view is that the key to NGF action must lie in short-latency events, occurring within minutes of NGF administration. Chick embryo DRG dissociates have proved to be an effective experimental system to investigate short-latency responses to NGF, in that (1) measurable functional deficits develop over 6 h of NGF deprivation in vitro and (2) delayed presentation of NGF promptly and fully restores the defective function. The first deficit observed in this experimental system, a decline in RNA-labeling capability, led to the recognition that NGF controls the transport of selected exogenous substrates, all of which are Na+-coupled and depend on an Na+ gradient across the neuronal membrane. Subsequent work showed that NGF controlled such transport systems by actually regulating the neuronal ability to control intracellular Na+. Under NGF deprivation, the DRG cells accumulate Na+ to levels that reflect, and presumably equate, the extracellular Na+ concentrations. Conversely, on delayed NGF administration, the accumulated Na+ is actively extruded to an extent and at a speed that depends on the NGF concentration. The Na+ response is elicited by both Beta and 7S NGF, but not by other proteins tested. All ganglionic systems that display a requirement for exogenous NGF in culture have also displayed the Na+ response to NGF. The Na+ response is grossly paralleled by a K+ response. DRG dissociates, in which intracellular K+ has been pre-equilibrated with extracellular 86Rb+, lose their 86Rb+ over 6 h of NGF deprivation and restore it on delayed NGF administration. The regulation by NGF of mechanisms controlling intracellular Na+ and K+ levels in their target neurons is likely to occupy an early and fundamentl place in the sequence of events underlying the mode of action of this factor.

    Notes: SJL mice were injected intraperitoneally with adipocyte plasma membranes or with intrinsic membrane proteins obtained by extraction of plasma membranes with dimethylmaleic anhydride. Three days after the boost injection, the spleens were removed and fused with NS-1, a thioguanine-resistant myeloma cell line derived from P3X63 Ag8 (Balb/c). Following selection for hybrids with hypoxanthine, aminopterin, and thymidine, medium of the hybrid cells was tested for its ability to bind to the plasma membrane of the adipocyte and to stimulate the oxidation of D-(1-14C) glucose to 14CO2. Approximately 40% of the wells containing hybridomas derived from splenocytes of SJL mice immunized with plasma membranes produced immunoglobulin that bound to adipocyte plasma membranes. About 30% of these mimicked the ability of insulin to stimulate the oxidation of D-(1-14C) glucose to 14CO2 in adipocytes. Media from 51% of the wells containing hybridomas derived from splenocytes of SJL mice immunized with intrinsic membrane proteins produced immunoglobulin that bound to the plasma membrane and 48% of those stimulated glucose oxidation. The bioactivity of the hybrid cell media could be blocked by adsorption with intrinsic membrane proteins or by the removal of immunoglobulins using formalin-fixed Staphylococcus aureus. The hybrids generated in this study can be divided into three categories: (1) hybrids that secrete antibodies that can bind to plasma membranes and mimic insulin action of glucose transport; (2) hybrids that secrete antibodies that bind to plasma membranes but do not stimulate the oxidation of D-(1-14C) glucose to 14CO2; and (3) hybrids that produce no antimembrane antibodies. The data suggest that interaction of immunoglobulins with specific membrane proteins is essential in mimicking the action of insulin on glucose transport and oxidation in the rat adipocyte.

    Notes: Long-term bone marrow cultures established from C57Ks/J mice have been shown to spontaneously release endogenous ecotropic RNA type-C virus (retrovirus). C57Ks/J marrow cultures produced granulocyte-macrophage progenitor cells (GM-CFUc) and immature and mature granulocytes for over 45 weeks. In contrast, NIH Swiss mouse marrow cultures failed to release detectable ecotropic virus and generated GM-CFUc and granulocytes for 25-35 weeks and established WEHI-3 conditioned medium (CM) dependent cell lines in vitro and did not establish permanent cell lines. To determine whether viral and/or cellular genes regulated the longevity of C57Ks/J marrow cultures, groups of cultures were established from the marrow of (NIH-Swiss × C57Ks/J) F1 hybrid, F2 hybrid, and (NIH Swiss × C57Ks/J) X NIH Swiss backcross generations. Release of endogenous ecotropic virus was measured weekly in each culture as was the duration of production of immature granulocytic cells and GM-CFUc over a 58-week period. The results demonstrated a complex pattern of inheritance of longevity of long-term in vitro hemopoiesis. Increased longevity did not absolutely correlate with detectable replication of the C57Ks/J N-tropic virus.
