    Notes: A method is described for the separation, identification and quantitation of endogenous hydrogenated metabolities of corticosterone in rat liver Corticosterone, 5α- and 5β-dihydrocorticosterone, 3α,11β,21-trihydroxy-5α-pregnane-3,20-dione, 3β,11β,21-trihydroxy-5α-pregnane-3,20-dione, 3α,11β,21-trihydroxy-5β-pregnane-3,20-dione, 3β,11β,21-trihydroxy-5α-pregnane-3,20-dione, 5α-pregnane-3α,11β,20α,21-tetrol, 5α-pregnane-3α,11β,20β,21-tetrol and 5α-pregnane-3β,11β,20β,21-tetrol are specifically quantitated in one analysis by mass fragmentography all as their O-methyloxime pertrimethylsilyl derivatives. A pronounced difference in the amounts of these nine metabolites between male and female adult rats was found, while in newborn rats only the two tetrahydro-compounds bearing a 5α-hydrogen were detected in both sexes. On the other hand 3α,11β,1 5α,21-tetrahydroxy-5α-pregnan-20-one was detected only in the adult female as its second prominent metabolite of corticosterone. Corticosterone metabolites being biochemical markers of the sex-linked differentiation of the hepatocytes, their mass fragmentographic assay is proposed as a tool for the quantitative analysis of this type of gene expression.

    Notes: A new metabolite, 6-ethylsalicylic acid, was shown to accompany 6-methylsalicylic acid in cultures of Mycobacterium phlei grown on media supplemented with propionate. Mass spectral analyses of the aromatic acid fractions obtained after growth in the presence of β,β,β-tridenteropropionate demonstrate that the ethyl group derives from propionate. It is postulated that this bacterium can use a propionate ‘starter’ unit in elaboration of a polyketomethylene chain.

    Notes: The difficulty of collecting volatile hydrocarbons, such as benzene, from street air in a form suitable for subsequent analysis is overcome by the use of a time-based elution procedure. By linking this technique to a gas chromatograph-mass spectrometer-data system, a rapid analysis is possible for low concentrations of collected material. Among advantages of this novel sample introduction procedure are the facility for collecting organic materials without interference from water vapour and their subsequent introduction into the g.c.-m.s. system without the use of any auxiliary solvents. Consequently the mass spectrometer can be used continuously on-line enabling the more volatile materials, normally masked by solvents, to be examined. This procedure is illustrated by the measurement of benzene and other hydrocarbon levels in street air (London). Following gas chromatographic separation the hydrocarbons can be identified by library search facilities attached to the mass spectrometer.

    Notes: The metabolism of cyclophosphamide-4-d2 (2-[bis(2-chloroethyl)amino]-tetrahydro-4,4-dideuterio-2H-1,3,2-oxazaphosphorine 2-oxide), was studied. The first detectable metabolite was a hydroxy derivative which was trapped with ethanol. Mass spectrometry of the resulting two ethoxy derivatives and of the deuterated acrolein 2,4-dinitrophenylhydrazones formed therefrom via reaction with acidic 2,4-dinitrophenylhydrazine, afforded evidence that the ethoxy substituents were at C-4, which was therefore the position of the original hydroxy substituent. The mass spectrum of the deuterated acrolein 2,4-dinitrophenylhydrazone obtained from the total reactive metabolites was used to estimate the ratio of 4- to 6-hydroxylation. The rate of metabolism and the antitumour activity of cyclophosphamide and its 4-d2 analogue were compared.

    Notes: The two major metabolites of [14C]niflumic acid, 2-(3′-[14C] trifluoromethylanilino)nicotinic acid have been isolated from human urine. Low and high resolution mass spectrometry indicate that both metabolites (M2a and M2b) are monohydroxylated derivatives of niflumic acid. The 100 MHz proton magnetic resonance spectrum of M2b demonstrates it to be 5-hydroxyniflumic acid, whereas M2a is either the 4′- or 6′-hydroxy derivative. The synthesis of 6′-methoxyniflumic acid methyl ester and comparison of its spectral data and melting point with that of the corresponding derivative of M2a reveal their non-identity. The structure of M2a is, therefore, 4′-hydroxyniflumic acid.

In the field ion spectra the molecular ions were the base peaks and structurally diagnostic fragment ions formed by direct cleavage were present, whereas the molecular ions were weak in the electron impact spectra and fragment ions formed by rearrangement were dominant.     Notes: The determination of drug metabolites is very important in the biochemical field. However, the procedure for doing this has been very complicated in many cases. The major reason is because of the need for fairly large amounts of the samples. We have been studying procedures for the determination and identification by use of mass spectrometry, which usually gives a lot of information rapidly from very small amounts of the samples (below μg). In this paper, the metabolites excreted in urine and bile following oral administration of 2-(diethyamino)ethyl tetrahydro-α-(1-naphthylmethyl)-2-furanpropionate oxalate (1) in rats, were investigated by means of mass spectrometry and other techniques. The drug was not excreted unchanged in urine. Of the eight metabolites suspected to be present in urine and bile, the structures of three metabolites were elucidated to be tetrahydro-α-(1-naphthylmethyl)-2-furanpropionic acid (3) and α-(1-naphthylmethyl)-2-perhydro-5-oxofuranpropionic acid (4) in urine, and diethylaminoethanol (2) in bile. These metabolites might also be present as their structural isomers or conjugates.

A reaction time of 15 minutes at 100 °C ensures complete derivatization, the derivatives being volatile at source temperatures below 150 °C, and giving electron impact spectra characterized by abundant (or moderately abundant) peaks at or near the mass of the molecular ion.The analysis of amino acid mixtures by mass fragmentography on characteristic high mass peaks of their N-(N′,N′-dimethylaminoethylene) methyl ester derivatives, introduced on a direct insertion probe and volatilized during temperature programming of the source, is qescribed. The method is applied to the analysis of a mixture containing arginine, citrulline, cystine, histidine and tryptophan, traditionally difficult to analyse by other methods.

    Notes: The urinary excretion of butobarbitone and its metabolites has been studied quantitatively using combined gas chromatography-mass fragmentography, tuned to ions at m/e 169 and 184. After a single oral dose (200 mg) to healthy male volunteers, unchanged drug (7 to 9%), 3′-hydroxybutobarbitone (22 to 27%), 3′-oxobutobarbitone (14 to 18%) and the 3′-carboxylic acid (4 to 8%), were found. In all cases, the maximum rate of excretion was in or near the third 12 hour urine.

    Notes: The organic material in airborne particulate matter is being studied by high resolution mass spectrometry to determine if hydrocarbons typical of major sources of pollution can be detected. Carbon number distributions and hydrogen-to-carbon ratios of components found in airborne particulate samples and the corresponding data for compounds detected in fuel combustion products show great similarity. Samples collected near large power stations and major airports were compared with typical urban particulate samples. Particulates collected from jet engine exhaust and a condensate from the stack effluent from a coal-fired combustion system were studied. Mass spectral data were obtained for an auto exhaust tar, a coal tar pitch and several fuels. The mass spectra of two total particulate samples, compared to the spectrum of an ether extract of the organic matter, indicate that direct vaporization can be used and extraction is not essential for qualitative mass spectral studies.

    Notes: The high resolution mass spectrometric integrated ion current procedure has been used to identify and quantitate amphetamine, isolated as its dansyl derivative, as it is found after addition to urine and in tissues of the rat following intraperitoneal injection.     Notes: A method is described for the identification of the sapogenins of several Digitalis species. The steroids are separated by repeated development thin-layer chromatography and identified by means of high resolution mass spectrometry. The spectra of tigogenin, gitogenin, digalogenin and digitogenin are recorded. These spectra are of particular use in establishing the position of extra hydroxy moieties in the steroid nuclei. Several species of Digitalis were screened for their sapogenin content. D. purpurea, D. mertonensis and D. lutea were found to contain tigogenin and digitogenin in small amounts, the principal steroid being gitogenin, whilst D. ambigua and D. lanata contained all four steroids.

    Notes: Acetic acid isolated from cider vinegar and inorganically synthesized glacial acetic acid have markedly different intramolecular isotopic distributions of the stable carbon isotopes. Carbon-12 is concentrated in the methyl group relative to the carboxyl group in the biologically produced acid. The reverse distribution is observed in the particular sample of glacial acetic acid examined here.

    Notes: An analysis has been made of a sample of rain water collected in Washington, D.C. following a sustained photo-oxidant smog alert which was accompanied by haze and low visibility. The analysis was effected by purging the organic content from the water with helium gas and identifying the constituents by means of gas chromatography - mass spectrometry. The predominant compound was found to be 3-methylfuran, a possible intermediate or end product of the atmospheric decomposition of terpenoid hydrocarbons. The analysis suggests that the smog resulted from the air oxidation of hydrocarbons evolved from Appalachian forestation rather than from manassociated activities.

    Notes: Combined gas chromatography-mass spectrometry and isotope labeling techniques have been employed to demonstrate that the carboxamide nitrogen atom in 2-carboxamido-5-isopropoxycarbonylaminobenzimidazole, a metabolite of 2-thiazolyl-5-isopropoxycarbonylaminobenzimidazole (cambendazole), is derived wholly from the parent drug. [15N] Thiazole-labeled cambendazole was administered to a rat and the metabolite isolated from urine. Trideutero-2-carboxamido-5-isopropoxycarbonylaminobenzimidazole was employed as a carrier to isolate the metabolite in sufficient purity for isotope ratio measurements using multiple ion detection. The metabolite was converted to its tetramethyl derivative by on-column reaction with trimethylanilium hydroxide to permit satisfactory gas chromatography.

    Notes: Some of the known and important metabolites of cyclophosphamide and the parent drug itself were investigated by low and high resolution field desorption mass spectrometry. The potential of the relatively new analytical method for the determination of the molecular weight and elemental formulae of drugs and drug metabolites was exemplified. This holds for pure, synthetic compounds as well as for contaminated extracts from biological sources. In addition, by thermally-/field-induced fragmentation, diagnostic fragment ions could be produced, providing valuable structural information. Further, an indication on how to distinguish [M]+. and [M + H]+ ions in field desorption mass spectrometry is given. The ability to identify these ions unambiguously is critical for interpretation. Finally, the potential of the method as applied to drug and drug metabolite mixtures is evaluated. The results indicate that field desorption mass spectrometry is a promising tool in metabolism studies.

    Notes: The amino acid sequences of the thirteen tryptic peptides of the cowpea strain of tobacco mosaic virus (Cowpea TMV) protein were determined by a combination of the mass spectrometric and Edman degradation procedures. In order to complete the amino acid sequences, a number of the tryptic peptides were further digested with chymotrypsin, thermolysin and carboxypeptidases A and B. The peptide fragments were separated and purified by the use of ion exchange chromatography. Sephadex chromatography and paper electrophoresis.The total number of amino acid residues in the Cowpea TMV protein was found to be 161 as opposed to the 158 found for the type TMV protein, and asparaginyl-arginine was the only tryptic peptide common to both proteins. All peptides used for mass spectrometry were first N-acetylated and then permethylated using methyl iodide and methyl sulphinyl carbanion or potassium t-butoxide dissolved in dimethyl sulphoxide.

    Notes: Representative phenylalkylamines (six enantiomeric pairs) and amino acid methyl esters (five enantiomeric pairs) have been examined by gas chromatography-mass spectrometry, in the form of their N-(R)-α-phenylbutyryl, N-(S)-α-phenylpropionyl- and N-(R)-α-chlorophenylacetyl derivatives. In most cases, satisfactory gas chromatographic resolution of the diastereomeric amides has been achieved on both SE-30 and OV-17 stationary phases. The phenylbutyramides and phenylpropionamides gave the more informative mass spectra (for characterization of the amines) but molecular ions were observed in all instances.

    Notes: Characteristic mass spectroscopic data for the identification of ten nitrosophenols is presented. The use of TMS derivatives for gas chromatography-mass spectrometry of 4-nitrosophenols is discussed, together with the use of copper chelates for obtaining direct insertion spectra of 2-nitrosophenols. The fragmentations of nitrosopher ols also show ions derived from their quinonemonoxime tautomers, even under gas chromatography-mass spectrometry conditions, indicating that isomerization occurs in the mass spectrometer. An anomalous elimination of nitrogen from these nitrosophenols has been shown to be a facile reduction of the nitroso moiety which takes place in the ion source of the mass spectrometer.

    Notes: A mass fragmentographic method for the estimation of the estimation of free and bound morphine and free 6-monoacetylmorphine in blood has been developed. It uses stable isotope internal standards and has a sensitivity of about 1 ng per ml or about 100 times better than ordinary gas chromatography. The focusing on specific high molecular fragments of the molecules gives high specificity to the assay and the stable isotope internal standards allow correction for losses occuring in the preparative steps of the analysis. A sensitive, specific method for monoacetylmorphine is particularly valuable since this compound seems to be a specific metabolite of heroin.

    Notes: Nonvolatile biological materials, such as biopolymers or even whole cells, can be made accessible to mass spectrometric analysis by pyrolysis techniques. In principle, this enables differentiation, classification and identification of these materials. Moreover, pyrolysis mass spectrometry can provide a general picture of the chemical composition and structure of the sample analysed. During the past few years we have investigated several pyrolysis techniques in combination with different mass spectrometric methods. These investigations included Curie point pyrolysis, infrared laser pyrolysis, low voltage electron impact ionization, field ionizationIn co-operation with Professor Dr H. D. Beckey and Dr H.-R. Schulten, University of Bonn, Germany. and pyrolysis field desorption. Pyrolysis reactions were performed either at the gas inlet or directly at the ion source. The relative merits of these different experimental approaches are discussed. Additionally, examples of various applications, such as computer identification of bacteria and qualitative analyses of biopolymers, are given.

    Notes: Quantitative methods based on chemical ionization mass spectrometry are described for the determination of tolbutamide, a sulfonylurea, and its metabolites from human plasma. The sulfonylureas are extracted from the acidified plasma and then N-1 methylated with diazomethane. The methylated compounds can be subjected to gas chromatography where they are partially cleaved to the corresponding N-1 methylated sulfonamides. Alternatively, the methylated compounds can be introduced into the ion source by means of a solid sampler. The sulfonylureas upon evaporation from the solid sampler or the formed sulfonamide and the parent sulfonylurea upon elution from the gas chromatographic column are subjected to chemical ionization with methane as the reactant gas. The [M + H]+ ions formed are measured quantitatively with respect to the deuterium substituted internal standards. It is suggested that the technique of introducing biological extracts into the ion source via a solid probe may be applied for the quantification of compounds that are unstable under gas-liquid chromatographic conditions. Both the gas chromatographic and the non gas chromatographic methods provided excellent and comparable results. The plasma level-time profile of tolbutamide and its metabolites in a diabetic patient following the administration of 1.0 g tolbutamide intravenously is presented.

Gas chromatograms of the reaction products show up to four additional peaks. These have been shown by mass and proton magnetic resonance spectrometry to be produced by the two isomeric N,O-dialkylbarbituric acids (1,4- and 1,2-dialkylbarbituric acids) and the two O,O′-dialkyl isomers.

    Notes: The precise isotopic analysis of carbon by means of differential comparison of CO2 samples is applied to neutral, acid c and basic extract fractions of human urine. It is shown that the standard deviation of the analytical procedure, including sample preparation steps, is about 1% or 0.001 atom % excess carbon-13, but depends some what on the fraction considered. Day to day variations (expressed as standard deviations) in the isotopic composition of the urine fractions are generally less than 2.8%, 1.4% and 3.9% for the neutral, acidica and basic fractions, respectively, although the effect of unusual dietary inputs can be recognized. The ingestion of 23 μg excess carbon-13 in the form of isotopically labelled aspirin is shown to perturb significantly the isotopic composition of the acidic urine fraction which, for a 24 hour collection period, had a mass of 570 mg C. Because only 0.01% of the fraction was consumed by isotopic analysis, further extensive analysis would be possible. It is concluded that carbon-13 tracer experiments generally should be arranged to provide at least 5 × 10-5 g excess 13C/g carbon in any fraction which is to be used for lable detection.

    Notes: Two different mass spectrometric techniques have been used to analyse the products resulting from Curie-point pyrolysis of nucleic acids: low voltage ionization quadrupole mass spectrometry and high resolution field ionization mass spectrometry. The spectra obtained with both techniques are in good agreement with one another. As already indicated by the low resolution spectra, the high resolution spectra proved that most of the peaks originate from the carbohydrate moiety of the nucleic acid. Micrograms of DNA/RNA can be distinguished very easily by use of these key fragments, although the bases themselves are not found. Some significant pyrolysis mechanisms are proposed.

    Notes: The mass spectra of synthetic methoxychlorobiphenyls have been investigated. These substances are of interest as reference compounds in studies of the metabolism of chlorobiphenyls. Some characteristic fragmentation patterns have been found, useful for differentiation between the 2-, 3- and 4-methoxychlorobiphenyl isomers. Thus, mass spectra of 2-methoxychlorobiphenyls show large fragments at [M - CH3Cl]+., while those of 4-methoxy compounds differ from 3-methoxy isomers by the presence of significant [M - CH3]+ fragments in the former.

    Notes: Piperidine is a putative neurotransmitter. Its sensitive determination is one of the prerequisites for the evaluation of its assumed physiological role. For its derivatization 5-di-n-butylaminonaphthalene-1-sulphonyl chloride, a new reagent which improves methods applying 5-dimethylaminonaphthalene-1-sulphonyl chloride, is used. Reaction of the perchloric acid tissue extract with this reagent, thin-layer chromatographic separation of the fluorescent derivatives and mass spectrometric evaluation of the di-n-butylaminonaphthalenesulphonyl piperidine spot by a version of the integrated ion current technique allows the determination of piperidine in tissues in picomole quantities. Standard deviation of the determinations of 2 to 16 pmol of piperidine added to 50 mg of brain tissue was ±8%. The method was applied to the analysis of different tissues. Brain concentrations of piperidine in different species were found to be lower by a factor up to 100 in comparison with published data.

(1971) die äußere Zona fasciculata von Sprague-Dawley-Ratten bei 5 Gruppen zu je 4 Tieren verglichen: Männchen, Weibchen im Diöstrus, Weibchen im Östrus, männliche und weibliche Kastraten (Kastration am 4. Lebenstag). Die Nebennierenrindenzellen der äu\eren Fasciculata sind bei Weibchen gegenüber denen normaler Männchen hypertroph: Weibchen im Diöstrus zeigen gegenüber Männchen größere Einzelzellen mit vermehrtem glattem endoplasmatischem Reticulum, erhöhten Einzel- und Gesamtvolumina der Mitochondrien sowie einem höheren Lipoidgehalt bei stärkerer Dispersion der Liposomen. Dieselben funktionsspezifischen Strukturen nehmen bei den Weibchen im Östrus noch weiter zu. Die Kastration der Weibchen am 4. Lebenstag führt zu einer Reduktion der funktionsspezifischen morphometrischen Parameter unter die Werte der Diöstrus-Tiere. Männliche Kastraten gleichen die Werte ihrer funktionsspezifischen Strukturen weitgehend denen der Weibchen im Diöstrus an. Nach Kastration am 4. Lebenstag zeigen weibliche Tiere somit etwas niedrigere morphonietrische Parameter in der äußeren Zona fasciculata als die männlichen Kastraten.
    Notes: Summary With the method of Rohr et al. (1971) the outer zona fasciculata of Sprague-Dawley-rats was compared ultramorphometrically in 5 groups of 4 animals each: males, females during diestrus, females during estrus, male and female castrates (castration on 4th day of life). Adrenal cortical cells of the outer fasciculata in females are hypertrophic in comparison with those of normal males: females during diestrus show in comparison with males larger individual cells with increased SER, increased volume of the individual mitochondrium and total volume of mitochondria as well as an increased lipoid content together with a more pronounced dispersion of liposomes. The same function-specific structures show a further increase in females during estrus. Castration of females on the 4th day of life leads to a reduction of the function-specific morphometric parameters below the values of the diestrus animals. Male castrates show adaption of the values of their function-specific structures towards those of females during diestrus. Following castration on the 4th day of life, female animals thus show somewhat lower morphometric parameters in the outer zona fasciculata than male castrates.

    Notes: Summary Several recent studies suggested that serotonergic neuron-like elements are present in the guinea pig ileum. The present paper reports an extensive study of the digestive tract of the rat with the use of a histofluorescence technique. Administration of the serotonin precursor, tryptophan, associated with a monoamine oxidase inhibitor, did not allow histochemical demonstration of rapidly fading, yellow fluorescent, 6-hydroxydopamine-resistant neurons; conversely such neurons were readily detected in the brain. It is concluded that serotonergic neuron-like elements cannot be detected histochemically in the rat myenteric plexus area after chemical sympathectomy.

    Description / Table of Contents: Zusammenfassung Die Neurohypophyse und die neurosekretorischen hypothalamischen Kerngebiete von normalen männlichen und weiblichen Ratten, von trächtigen und durstenden Tieren wurden elektronenmikroskopisch und morphometrisch mit der Fragestellung untersucht, ob sich in den Häufigkeitsverteilungen der mittleren Granula-Durchmesser, bezogen auf die Anzahl der Axone, eine zweigipflige Verteilung ergibt. Bei Kontrolltieren zeigen die Perikarya des Nucleus supraopticus zahlreichere und größere Granula als die Zellen des Nucleus paraventricularis, wobei der Unterschied in den Durchmessern der Granula bei den weiblichen Tieren ausgeprägter ist als bei den männlichen. Beide Kontrollgruppen zeigen in der Neurohypophyse ein zweigipfliges Verteilungsmuster der Granula, wobei die beiden Gipfel bei den männlichen Kontrollen dichter beieinander liegen als bei den weiblichen. Bei den trächtigen Tieren weisen beide Kerngebiete Zeichen gesteigerter Aktivität auf und die Granula in den Zellen des Nucleus paraventricularis sind deutlich größer als bei Kontrolltieren. Zu Beginn der Tragzeit werden relativ große, wahrscheinlich vasopressinhaltige Granula in der Neurohypophyse gespeichert, während sich im letzten Drittel der Tragezeit die beiden Populationen dort nicht mehr unterscheiden lassen. Nach 48 Std. Wasserentzug erscheinen beide hypothalamischen Kerne aktiviert. Auffallend ist, daß die Granula im Nucleus supraopticus deutlich kleiner sind als bei Kontrolltieren. Entsprechend fehlen in der Neurohypophyse große Granula, während Axone mit kleinen Granula vermehrt sind. Nach zehntägiger Durstperiode lassen sich im Nucleus supraopticus, neben sehr typischen morphologischen Veränderungen, morphometrisch ebenfalls deutlich kleinere Granula nachweisen. Im Hypophysenhinterlappen dieser Versuchsgruppe sind die granulahaltigen Axone sehr stark vermindert, und in der Häufigkeitsverteilung fehlen die Granula mit großem Durchmesser während der dem Oxytocin zugeordnete Gipfel unverändert erscheint. In Verbindung mit den elektronenmikroskopischen Befunden lassen sich also aus den Verschiebungen der Verteilungsmuster zwischen den sechs Versuchsgruppen die Granula mit kleinerem Durchmesser oxytocinhaltigen Axonen zuordnen und Granula mit größerem Durchmesser zu den Endigungen, die Vasopressin enthalten. Die morphometrischen Befunde in den neurosekretorischen hypothalamischen Kerngebieten bestätigen diese Zuordnung. Die elektronenmikroskopischen und morphometrischen Befunde werden besonders im Hinblick auf die Hormonverschiebungen zwischen dem “reserve pool” und den “easily releasable pool” diskutiert.
    Notes: Summary A morphometric evaluation of electron micrographs has been carried out from neurosecretory terminals in the neurohypophysis and from the supraoptic and paraventricular nuclei in normal male and female rats as well as in pregnant and water deprived rats. The task of this investigation was to find out whether frequency distribution diagrams of the mean diameter of the neurosecretory granules, plotted versus the number of axons, reveal a grossly bimodal distribution. In normal rats the nerve cells of the nucleus supraopticus (S.O.N.) show more numerous and larger granules than the cells of the nucleus paraventricularis (P.V.N), this difference with respect to the diameter being more pronounced in female than in male animals. In the posterior pituitary lobes the neurosecretory granules exhibit a bimodal distribution, the second peak being situated more closely to the first one in male controls. In pregnant animals both nuclei appear to be activated, and the granules of P.V.N. are distinctly larger as compared to those of female control animals. In early pregnancy larger, i.e. presumably vasopressin-containing granules are stored in the neurohypophysis, while in the last phase before delivery oxytocin-containing neurons and vasopressin-containing ones cannot be differentiated any more. After 48 hrs of thirst both hypothalamic nuclei exhibit signs of increased activation, the diameters of the granules in the S.O.N. being distinctly smaller than in normals. In the neurohypophysis granules with large diameters are lacking, while axons with small granules are more frequent. In the S.O.N. of animals, water deprived for 10 days the diameters of the granules are smaller than normal and in the neurohypophysis the number of granule-containing fibres is strongly reduced. In the frequency distribution diagram granules with larger diameters are lacking whereas the peak, attributed to oxytocin, is not essentially altered. However, from the observed alteration on the distribution patterns of the six experimental groups and in connection with the electron microscopic results the granules with smaller diameters can be attributed to oxytocin-containing axons and the larger ones to vasopressin-containing terminals. Analogous morphometric data have been obtained from the neurosecretory nuclei as well. The electron-microscopic and morphometric observations are discussed with regard to hormone-shifts between the reserve pool and the easily-releasable pool.

Der Granulagehalt der juxtaglomerulären Zellen nahm ab; die Kern-Plasma-Relation änderte sich und die Leiomyofibrillen in den Epitheloidzellen nahmen zu. An den übrigen Anteilen des juxtaglomerulären Apparates, insbesondere der Macula densa, wurden keine morphologischen Veränderungen beobachtet. Die DOC-Behandlung führte darüber hinaus zu einer Bildung von Einschlüssen in den Podocyten, den Mesangiumzellen und den Zellen der Bowman-Membran. Die verschiedenen Möglichkeiten, über welche Faktoren die juxtaglomerulären Zellen nach DOC-Behandlung degranuliert werden, werden diskutiert. Das ultrastrukturelle Bild der Epitheloidzellen nach DOC-Behandlung entspricht deutlich einer gehemmten Sekretion, da das Ergastoplasma und der Golgi-Apparat ganz besonders stark zurückgebildet werden.
    Notes: Summary Quantitative morphological analysis of the Juxtaglomerular apparatus of rats treated with deoxycorticosterone (DOC) and salt revealed hypoplasia of the epithelioid cells as a concomitant of the inhibition of renin synthesis. The granular content of the Juxtaglomerular cells was significantly decreased, while the nucleus-to-cytoplasm ratio and the volume density of leiomyofilaments in the epithelioid cells increased. No morphological alterations were observed in the remaining constituents of the Juxtaglomerular apparatus, including the macula densa. DOC-treatment also induced the appearance of inclusions in the podocytes in the cells of the mesangium, and in the cells of Bowman's capsule. The possible mechanisms of degranulation of the Juxtaglomerular cells after treatment with DOC are discussed. The ultrastructural appearance of the epithelioid cells after treatment with DOC corresponds to an inhibition of secretion, since the rough endoplasmic reticulum and the Golgi apparatus undergo marked regressive changes.

    Notes: Summary Adrenal glands of adult female rats are heavier than the glands of corresponding male rats. Postpubertal orchiectomy increases the adrenal weight, an effect restored by testosterone replacement. Under the same conditions ovariectomy of 8 weeks duration does not change the adrenal weight while estradiol replacement enhances the relative adrenal weight. Karyometric studies have shown that nuclei in the female zona fasciculata cells are larger (app. 18%) than those of the male. Similar but only slight differences (2%) were observed in the zona reticularis. Orchiectomy results in enlargement of cell nuclei within all zones of the adrenal cortex; testosterone replacement has the opposite effect. Ovariectomy of 8 weeks duration slightly enhances the volume of nuclei of the zona glomerulosa cells, has no effect on the nuclei of the zona fasciculata and reduces the volume of nuclei in the zona reticularis. Estradiol replacement reduces the volume of nuclei of the zona glomerulosa cells but increases nuclear volume in the zona fasciculata and in the zona reticularis. Thus testosterone has an inhibitory effect on the adrenal cortex of the rat while the physiologic effect of estradiol on the morphology of this gland, particularly on the zona fasciculata cells is rather dubious. The stereologic analysis of the volume fractions of particular adrenocortical zones have shown that the volume fraction of the zona glomerulosa in females is smaller as compared with males. On the contrary, the volume fractions of the fasciculata and reticularis zones are higher in female than in male rats. In both sexes gonadectomy and gonadal hormone replacement has an effect on the volume fractions of particular adrenocortical zones, these changes being more pronounced due to orchiectomy or testosterone replacement. The adrenal cortex of all experimental groups contains a similar quantity of parenchymal cells. The cytoplasm volume of a single adrenocortical cell is higher in all zones of the female adrenals as compared with the male. Orchiectomy increases the volume of the cytoplasm in fasciculata and reticularis cells. This effect was reversed by testosterone. Neither ovariectomy nor estradiol replacement has an effect on the volume of the cytoplasm in the fasciculata and reticularis cells. These studies have shown that sex differences with regards to rat adrenal size and weight depend upon the “physiologic atrophy” of the male adrenal cortex, while neither ovariectomy (8 weeks after operation) nor estradiol replacement has a conspicuous effect on the morphologic changes of this gland.

    Notes: Summary The dorsal lobe and the coagulating gland of the rat prostate were studied after administration of the anti-androgenic compound Cyp A for varying periods (7–20 days). There was a general involution of the organs with macroscopic atrophy and reduced amount of secretory material. Ultrastructurally the cells were pale, contained less organelles as compared with the controls, loss of cytoplasm and reduction both of cell height and width. The major changes also included reduction of the Golgi area, the rough endoplasmic reticulum and the number of free ribosomes. Within the dorsal lobe prominent nuclear changes were found, comprising different types of intranuclear inclusions of membranous configurations. The alterations observed in the present study are similar to the changes which were found in castrated animals and are consistent with atrophy. The results are discussed in relation to the available biochemical and autoradiographic data on the different lobes.

    Notes: Summary Electron microscopical evidence for the existence of an important serotonergic input to the rat median eminence is presented. This evidence is based on the demonstration of degenerating nerve terminals in the external layer of the rat median eminence following the application of 5,6-dihydroxytryptamine or 5,7-dihydroxytryptamine, drugs known to exert more or less selective toxic effects on central serotonin neurons. The finding of a serotonergic innervation of the median eminence in mammals has important implications for 5-hydroxytryptamine as a modulator of synthesis and/or release of hypothalamic, hypophysiotropic hormones.

    Notes: Summary Palisade-shaped nerve endings of the small normal hairs of the rat snout were examined with the electron microscope (fixation by perfusion). The terminals are located inside the ‘glassy membrane’ in the area of the neck of the hair root. The 10–20 radially arranged terminal axons are in direct contact with the basement membrane of the epithelium of the external root sheath. The axons are surrounded on all sides by leaf-shaped processes of the Schwann cells. The surfaces of these cell processes are marked by numerous vesicle-like invaginations (approx. 1000 Å dia.). Transverse sections from several areas of the palisadeshaped nerve endings are compared with longitudinal sections. In the upper area ‘empty’ vesicles (approx. 500–600 Å in diameter) occur, along with electron-dense vesicles (approx. 800–1100 Å in diameter); in the middle area, the axons are distended and contain accumulations of mitochondria.

    Notes: Summary Electron microscopic studies of ultrathin serial sections of the perikarya and adjacent neuropil of neurons of the supraoptic nucleus (SON) of the adult male rat revealed varying forms of two types of somatic spines. One type forms synapses with axons passing the cell, the other, without synapses, appears to serve as a buttress or clasp for adjacent neuronal and glial processes. The synapse-bearing spines lack the usual spine apparatus but contain the flocculent substance often seen in spines. The other spines do not exhibit either of these structures. These somatic spines were also seen in Golgi impregnated preparations but the types could not be distinguished. Certain axons synapse either on a somatic spine of the perikarya or penetrate the glial sheath of the neuron and synapse, usually repeatedly, on the soma in an en passant manner.

    Notes: Summary The rat anococcygeus muscle, which is known from previous functional and histochemical investigations to be a smooth muscle with a dense adrenergic innervation, was studied by electron microscopic methods. The muscle was found to be organized in small bundles of 2 to 8 muscle cells, which were interconnected by desmosome-like junctions and by gap junctions. Small Schwann cell-axon complexes with 1 to 5 axons appeared to represent the functional terminal nerve pathway. Such axon complexes were located at the periphery of the small smooth muscle bundles, they never penetrated into them. A ratio of approximately 6 small axon complexes per 100 muscle cells was found. After glutaraldehyde fixation axon varicosities contained mainly small empty vesicles. After treatment with 5-hydroxydopamine and permanganate fixation mainly small vesicles with a dense core were found. The neuromuscular distances ranged from 550 to 5000 Å, with a mean of about 2600 Å. True close neuromuscular contacts at a distance below 500 Å with a fusing of basal laminae were absent. The present observations provide a morphological base for the findings of previous electrophysiological and pharmacological studies on the anococcygeal muscle.

    Notes: Summary The pineal glands of Wistar rats were fixed in glutaraldehyde and prepared by the freeze-etching technique. With regard to the form and internal structure of pineal cells this technique has revealed nothing essentially new. Synaptic ribbons are very rarely found. A regular diagonal arrangement of membrane attached particles between the Schwann cell and the axon of an autonomic nerve terminal was observed. Three types of contact areas between pineal cells are described. In the first two types, which occur more frequently, the diameter of the area varied from 1000 to 2500 Å, with irregularly aggregated membrane associated particles. The third type, larger but less frequently found, has a surface area of 0.4 μm2 and much more regularly distributed membrane associated particles (ø 88 Å). These contact areas can be considered to be electric coupling areas between pineal cells.

    Notes: Summary Mast cells in the tongue and lamina propria of the duodenal mucosa, representing two cell types with different morphological, histochemical and functional properties have been studied under the electron microscope. Both cell types were found to contain similar moderately electron dense and homogeneous granules surrounded by a tight membrane constituting the basic ultrastructural characteristics of mast cells. The mucosal mast cells, however, contained fewer granules per cell and the individual granules often exhibited considerable variation in size. In addition the mucosal mast cells, unlike those of the tongue, peritoneum and skin, had a smooth plasma membrane lacking microvilli and possessed irregularly shaped or lobulated nuclei. In the tongue mast cells, administration of Compound 48/80 caused the formation of vacuoles around the granules, expulsion of granules from the cells and disintegration of the granular matrix, but no changes that could be attributed to 48/80 could be found in the mucosal mast cells.

    Notes: This paper deals with a finite element formulation and solution of the boundary value problem of a concentrated force acting on a semi-infinite micropolar solid. A stiffness matrix is derived for a flat micropolar rectangular element in a plane state and use is made of this element to solve the case of the semi-infinite micropolar solid both in a state of plane stress and plane strain. An immediate application of the plane strain case is the important practical problem of a long line load acting on a soil mass which may be simulated as a micropolar body.

    Notes: A finite element formulation is presented for the equations governing the steady thermohydrodynamic behaviour of liquid lubricated bearings. This formulation permits application of the iterative solution scheme to bearings of arbitrary geometry.A generalized Reynolds equation resulting from the combination of the mass and momentum conservation equations is cast into variational form and used to derive general finite element equations. The method of weighted residuals with Galerkin's criterion is used to generate finite element matrix equations for the thermal energy equation. In addition to the finite element formulation, a discussion of appropriate finite difference techniques is also given for problems without complex geometry.As an example, the formulations are applied to obtain numerical solutions for a three-dimensional sector thrust bearing operating in the thermohydrodynamic regime. Pressure, velocity and temperature distributions are give, and the thermohydrodynamic solutions are compared with the results of classical isothermal theory.

    Notes: When large, progressive, deformation occurs under plastic or visco-plastic conditions, elastic deformations are negligible and the material flows in a viscous manner. The flow is non-Newtonian and the viscosity is a function of the current strain rates. An effective numerical treatment of such non-Newtonian flow by the finite element method is indicated here and is illustrated by examples of an extrusion process for which approximate slip line solutions are known.The methodology is extended to the solution of transient, quasi-static situations and coupling with thermodynamic equations is discussed.A stream function formulation of the problem is used.

    Notes: By representing the time-dependent material properties by mechanical models, i.e. the generalized Maxwell and/or Voigt (or Kelvin) models, it is possible to formulate a consistent procedure to analyse the effect of material damping on dynamic response. As a continuation of the previous work which concerned the creep and relaxation of viscoelastic materials, the present paper applies this representation to the harmonic response analysis. The harmonic solution can be obtained by decomposing the relevant forces and displacements into real and imaginary parts and it is shown that the modal analysis by an appropriate eigensolution procedure is useful for obtaining the dependence of vibration amplitude on frequencies of sinusoidal external excitations.

    Notes: Body oriented co-ordinates are defined for three-dimensional regions bounded by six surfaces. From this definition of body oriented co-ordinates, two transformations are derived: 1Body oriented surface co-ordinates to the Cartesian co-ordinates, and2Body oriented co-ordinates for a three-dimensional region to Cartesian co-ordinates.With these transformations a mesh generator is developed based on the principle that the mesh required to describe a region will be calculated according to the placement of the discrete points that describe the boundary edges. This is accomplished with two versions of the mesh generator: the surface generator and the volume generator. The surface generator maps the four boundaries of the surface onto the four boundaries of a unit square. Next, the interior points for the surface mesh are established in the mapped space. Then, using the first transformation, the co-ordinates for the surface nodal points are calculated. Similarly, the volume generator maps the six surfaces of the body onto the surfaces of a unit cube, establishes the interior nodal points in the mapped space, and using the second transformation, calculates the co-ordinates for the interior nodal points.

    Notes: By use of the least squares error criterion, an alternate finite element formulation is presented. The method is based on the discrete or element-wise minimization of square and weighted differential equation residuals which are expressed in terms of element nodal quantities. In order to overcome the stringent inter-element continuity requirement, a major stumbling block, on the element trial functions two practical schemes are proposed. One is the reduction of the original governing differential equation to a system of equivalent first order differential equations; the other is a method of smoothing discontinous trial functions. The latter essentially relaxes the continuity requirement and yields efficient non-conforming finite elements. This paper also demonstrates the use of constant weights which significantly improves the rates of convergence. Several numerical examples illustrate the proposed method. From these examples, it may be concluded that the use of constant weights and the relaxation of the inter-element continuity requirement are two indispensable features of the weighted discrete least square method.

    Notes: In a two-dimensional space of points on an equilateral triangle grid, a single difference equation holding at each node is found from the finite element formulation of the wave equation for all published polynomial orders of the functional expansion.     Notes: Through the use of complex series representations and the properties of adjoint differential operators, a semi-analytical finite element procedure is developed which can analyse the steady state and transient temperature fields of thermally anisotropic axisymmetric bodies with complete circumferential properties.     Notes: A finite element weighted residual process has been used to solve transient linear and non-linear two-dimensional heat conduction problems. Rectangular prisms in a space-time domain were used as the finite elements. The weighting function was equal to the shape function defining the dependent variable approximation. The results are compared in tables with analytical, as well as other numerical data. The finite element method compared favourably with these results. It was found to be stable, convergent to the exact solution, easily programmed, and computationally fast. Finally, the method does not require constant parameters over the entire solution domain.
