    Notes: The synthesis and optical activity of novel chiral dendrimers with axial chirality are reported. The dendrimers were constructed by coupling of the polyether dendritic bromides with (R)-(+)-1,1′-bi-2-naphthol (1). The uniform (2, 3, 4) and non-uniform double-O-alkylated (8, 9, 10) as well as mono-O-alkylated (5, 6, 7) products were thus obtained. These chiral molecules were characterized by 1H- and 13C-NMR, elemental analysis, optical rotation, adsorption spectra, and circular dichroism. It was found that the specific rotation decreases with the increase of the number of generation for each group of dendrimers (2-4, 5-7, and 8-10, respectively). In terms of the molar rotation, it was quite different; the molar rotation increased sharply for dendrimers, 2-4, but only slightly for dendrimers 5-7.     Notes: Almond β-glucosidase was used to catalyze the synthesis of alkyl β-d-glucopyranosides 1b-3b starting from either d-glucose (4) or phenyl-β-d-glucopyranoside (5) and the racemic alcohols 1a-3a. The enzymic reactions were provided in different acetonitrile/water mixtures [9:1 (v/v) for the reverse hydrolysis, and 1:9 (v/v) for the transglycosylation]. Both enzymic reactions, that is, the reverse hydrolysis and the transglucosylation, are enantioselective processes. The enantiomeric purity of products 1b-2b of the enzymic reactions varied between 75 and 86% ee, the values of which were based on the analysis of the aglyconic parts (1c-2c) of the alkyl β-d-glucopyranoside molecules (1b-2b).     Notes: The association equilibria for complex formation between serum albumin (bovine, rat) and the optical isomers of methamphetamine (MAMP) was determined using an ultrafiltration method. It was found that serum albumin/d-MAMP and serum albumin/l-MAMP complexes had distinctly different Scatchard plots with bovine and rat albumin. The binding parameters of each association equilibrium were estimated from the Scatchard plots by Rosenthal's graphic method. This distinguished two kinds of specific binding sites in terms of the association equilibrium between bovine serum albumin and d-MAMP, and one binding site for rat serum albumin and d-MAMP. One specific binding site was found between serum albumin and l-MAMP in both bovine and rat. Molar ellipticities, [θ], of peaks were decreased in the CD spectra of the complexes formed between bovine serum albumin and d-MAMP or l-MAMP when compared with the CD spectrum of bovine serum albumin alone. However, no difference in [θ] was found between the CD spectra of the enantiomers of MAMP in the measured wavelength range.     Notes: (±)-Pantoprazole ((±)-PAN), (±)-5-(difluoromethoxy)-2-[[(3.4-dimethoxy-2-pyridinyl)methyl]sulfinyl]-1H-benzimidazole is a chiral sulfoxide that is used clinically as a racemic mixture. The disposition kinetics of (+)-PAN and (-)-PAN given separately has been studied in rats. Serum levels of (+)- and (-)-PAN and its metabolites, pantoprazole sulfone (PAN-SO2), pantoprazole sulfide (PAN-S), 4′-O-demethyl pantoprazole sulfone (DMPAN-SO2), and 4′-O-demethyl pantoprazole sulfide (DMPAN-S) were measured by HPLC. Following single intravenous or oral administration, both enantiomers were rapidly absorbed and metabolized, resulting in similar serum concentrations, suggesting that the two enantiomers have approximately the same disposition kinetics. The major metabolite of both (+)- and (-)-PAN was PAN-SO2, while DMPAN-SO2 was also detected as a minor metabolite. Serum levels of PAN-S and DMPAN-S could not be quantified after intravenous or oral administration of either enantiomer.Significant chiral inversion occurred after intravenous and oral administration of (+)-PAN. The AUCs of (-)-PAN after intravenous and oral dosing of (+)-PAN were 36.3 and 28.1%, respectively of those of total [(+) + (-)] PAN. In contrast, the serum levels of (+)-PAN were below quantitation limits after intravenous or oral administration of (-)-PAN. Therefore, chiral inversion was observed only after administration of (+)-PAN, supporting the hypothesis that stereoselective inversion from (+)-PAN to (-)-PAN occurs in rats.     Notes: In order to elucidate the chiral recognition mechanisms of the enzyme cellobiohydrolase I (CBH I), its carboxylic groups were covalently modified. The synthetic modification was carried out either in the presence or absence of cellobiose, which has proven to inhibit the enzymatic activity and if present in the mobile phase impairs enantioselectivity of amino alcohols. Compared to the reference CSP (unmodified CBH-I silica and CBH-I core silica), the synthetically modified phases show differences both in enantioselectivity and retention. The enzymatic differences between the CSPs were also in line with the chromatographic results. The selectivity factors of propranolol are almost unchanged during the reaction periods in the presence of cellobiose, while they decreased rapidly without the inhibitor.     Notes: Proline and prolylproline dipeptide derived surfactants promote the asymmetric hydrogenation of (Z)-methyl α-acetamidocinnamate in water in the presence of the catalytic system [Rh(cod)2]BF4 + BPPM. Activity and enantioselectivity are enhanced significantly and the results in water are similar to those obtained with organic solvents. The possibility of a chiral induction was investigated in the presence of the optically active amino acid and peptide amphiphiles and an achiral rhodium catalyst [Rh(bdpb)(cod)]BF4. The analysis of the low optical induction gave some indications of the site where the reaction takes place within the micelle.     Notes: Chromatographic resolution of 12 derivatives in the 4a-methyl-2,3,4,4a-tetrahydro-1H-fluorene and 4a-methyl-1,2,3,4,4a,9a-hexahydrofluoren-9-one series differing by the framework around position 9 and substitution in position 6, are reported on Chiralcel OD, Chiralpak AD, and Chiralpak AS under two elution conditions and according to the two classes of enantiomers. Results from principal component analysis (PCA) as well as hierarchical clustering show a clustering of the actual compounds depending on properties around position 9, the effect of the substituent in position 6 (methyl, chloro or fluoro) not being strong enough to intermesh the data. Carbamate phases show very different properties when they are used in the separation of a series of ketones C and α-chloroketones D, which differ in basicity and the steric requirement around the carbonyl. Analysis of the effect of 2-PrOH content in hexane on the retention is consistent with a large difference in solvation of the carbamate moiety by 2-PrOH, in the order Chiralcel OD 〉 Chiralpak AD 〉 Chiralpak AS. 4a-Methyl-2,3,4,4a-tetrahydro-1H-fluorene derivatives, which lack hydrogen bonding sites, are not discriminated on these CSPs and show identical k′ responses to 2-PrOH content changes on the three CSPs.     Notes: Identification and enantiodifferentiation of γ-lactones produced during the bioconversion of soy bean fatty acids by Penicillium roqueforti spores in the presence of an exogenous lipase was performed using gas chromatography-mass spectrometry (GC-MS) in selected ion monitoring (SIM) mode. It was shown that 4-dodecanolide and 4-hexanolide were first produced with an enantiomeric excess (99%) in favor of the (R) form, whereas an enantiomeric excess in favor of the (S) form (94%) is found for (6Z)-dodecen-4-olide, the major lactone produced by the fungus. If incubation was continued, mixtures of both enantiomers were found, more particularly for 4-decanolide (R/S:40/60), which was produced only after 120 hr of incubation. The results obtained can be explained by the stereoselective hydroxylation by a 10-hydratase and/or a 10-lipoxygenase of the unsaturated fatty acid precursors, oleic and linoleic acids, and by competition between different pathways. The results point out the limitations of chiral GC analysis as a criterion for the natural origin of relevant lactones, and the complexity of mechanisms involved in γ-lactone formation by microorganisms.     Notes: The β2-receptor agonist class of drugs is metabolized in humans almost exclusively by sulfate conjugation. The objective of this investigation was to determine the influence of chemical structure on the stereoselectivity of the sulfoconjugation of these chiral drugs. The pure enantiomers of six β2-agonists, including those clinically most widely used, were all effectively sulfated both by the cytosol of the human intestine and the recombinant human M-form phenolsulfotransferase (PST). Whereas the apparent Km values (Km,app) for the sulfation of the individual drug enantiomers by the intestinal cytosol varied widely, ranging from 4.8 μM for (S)-isoproterenol to 889 μM for (S)-albuterol, these Km,app values were highly correlated with those obtained with M-PST (correlation coefficient 0.994). In contrast, the M-PST Vmax,app values were similar for all drug enantiomers, ranging from 276 to 914 pmol min-1 mg-1 protein, implying that substrate binding to M-PST by far is the main determinant of the sulfation activity. For isoproterenol, the Km,app for M-PST was 6.1 times higher for the active (R)- than for the inactive (S)-enantiomer. For other β2-agonists, the stereoselectivity decreased towards unity as the Km,app increased. However, for albuterol, containing a hydroxymethyl substituent at the aromatic ring, the stereoselectivity was dramatically reversed, with 10 times higher Km,app for the inactive (S)- than for the active (R)-enantiomer.     Notes: A number of 2,3-epoxy alcohol derivatives (1-16), obtained either as racemates or through the Sharpless asymmetric epoxidation reaction, were studied on a Chiralcel OD column. Nearly all compounds exhibit good enantioselective resolution on this chiral support. The order of elution of enantiomers is reversed between nerol and geraniol compounds. For 2,3-epoxy alcohols bearing a remote alkoxy (or silyloxy) group, the order of the enantiomeric elution alternates with the number n (n = 1-3) of methylenic groups present between the epoxide ring and the terminal OR (R = p - BrBn or OSitBuPh2) functionality. In the case of trans 2,3-epoxy alcohols for the same number n, the order of elution is reversed when changing the terminal group -OSi to -OR.     Notes: The self-regulation of biological signalling receptors via homodimerization is discussed in relation to the symmetry changes occurring when these receptors bind their target ligand. The idea of positive and negative cooperativity between dimerization and ligand binding, mediated by changes in the symmetry of the system, as a source of signalling control is considered; and an analogy made with the homodimerization of a glycopeptide antibiotic, ristocetin A, which displays negative cooperativity. Finally, the regulation of the bacterial aspartate receptor and the human growth hormone receptor is discussed as a function of ligand-induced asymmetry.     Notes: An improved model-based method for the stereospecific assignment of prochiral centers in collagen-like triple-helical molecules is introduced. Using the concepts of reporter atoms and of ensemble NOEs, the proposed methodology extracts the stereochemical information contained in the chiral elements of triple-helices and transfers it to prochiral centers with nondegenerate proton resonances. The improved approach has been successfully validated using -(Gly-Pro-Hyp)n- triple-helices for which the stereospecific assignment was previously obtained with established techniques. We have applied our stereochemical characterization to novel peptoid containing triple-helices for which existing methods of stereospecific assignment can not be used for all the prochiral centers. In our approach, several different NOE measurements are employed to make a given stereospecific assignment. The multiple NOE comparisons allow internal cross checks, which reduce the chance of erroneous assignments caused by experimental artifacts including spin diffusion and bias from anisotropic rotational motions. In addition, the multiple NOE comparisons are useful in overcoming problems associated with resonance overlap often encountered in the 1H-NMR spectra of collagen-like molecules. Our stereochemical analysis is anticipated to improve the precision and accuracy of the characterization of collagen-like triple-helices through a better correlation of structures with their 1H-NMR spectra.     Notes: A class of polymers synthesized at DuPont in the late 1950's, the polyisocyanates, are the simplest analogs of the Nylons and have proven valuable as experimental models for theories of wormlike macromolecules. The macromolecular dimensional properties associated with all wormlike polymers arise from a strongly preferred local conformation of the chain and in the polyisocyanates this conformation is helical with an interesting additional property in that the mirror helical senses are of equal probability. Recent experiments have shown that discrimination between the helical senses can be accomplished with surprisingly small chiral influences indicating high cooperativity which arises from a conformational state in which long sections of one helical sense are separated from the other sense by infrequent helical reversals. This can be seen to be analogous to theoretical ideas about one-dimensional paramagnetic materials in which the spin states and the domain boundaries are analogs to the helical sense and the helical reversals in the polyisocyanates. The 2,3,5-trimethyl-thiadiazolium salt 9 does not condense with 6e, as other CH2-acidic compounds do, instead the vinylogous guanidinium salt 10a is produced. On heating, the ketenaminals 8d, aa cyclize to give the pyridone-carbonitriles 11a, b, resp. From 4-amino-coumarins 12 and the orthocarboxylic acid amideacetals 13a, b and the ketenaminal 16 resp., the amidines 14a-c and the heterocyclic compounds 15a-c resp., are formed. The enamines 17a-c, 19a, b react with the orthoamides 6a-f to give the pyridine derivatives 18a-1, 20a-h and 21a, b, resp. Analogously, from 6-aminouracil 22 and 6a, b, e, f are formed the pure 7-dimethylaminopyrido[2,3-d] pyrimidines 23a, b or mixtures of compounds 23c, d and the isomeric 4-dimethylamino-pyrido[2,3-d]pyrimidines 24a, b resp., which can be separated via their salts 25a,b/26a,b. The heterocyclic compounds 30a-d, 32a,b can be prepared from the pyrazole derivatives 28, 31 resp. and the orthoamides 6a-f.

Change of the Type of the Mesophase by Variation of the Acyl Chain of Amphiphilic Tetradecyl (N-acylamino)-2-deoxy-1-thio-β-D-glucopyranosidesThe homologous amphiphilic tetradecyl-2-(N-alkanoylamino)-2-deoxy-1-thio-β-D-glucopyranosides 5a-f were synthesized from the 2-(N-alkanoylamino)-2-deoxy-D-glucopyranoses 1a-f via the glucosyl chlorides 2a-f, the glucosyl isothiuronium chlorides 3a-f, and the acetylated thioglucosides 4a-f. On heating the amphiphiles 5a-f form different types of liquid crystalline phases in dependence of the length of the acyl chain, i.e. 5a and 5b form smectic phases, 5d-f form columnar mesophases. Polarisation microscopy, DSC-measurements and contact preparations were used to characterise the liquid crystaline phases.

    Notes: The partially diethylamino substituted phenyl-chloromono- and -disilanes Cl2Ph(NEt2)Si(2) Cl2Ph(NEt2)2Si(3), [ClPh(NEt2)Si-]2 (7) and Cl(NEt2)PhSi—SiPh(NEt2)2 (8) were synthesized. 3 and 8 are able to react with lithium powder to form the corresponding silyllithium compounds. The resulting species were characterized in solution by 13C and 29Si NMR spectroscopy. To prove the expected substitution patterns of the silyllithium compounds some coupling reactions with chlorosilanes were carried out and the resulting new oligosilanes were characterized by means of NMR spectroscopy and MS.

    Notes: Starting with monochiral 1,2,3,4-tetrahydro-3,6-dimethyl quinoline 14a,b, (Schemes 1 and 2), we have synthsized the new chiral, symmetrical and unsymmetrical, tri- and pentamethinium streptocyanine dyes 2a, (Scheme 8), 4a, (Scheme 5), and 5a,b, (Scheme 4), resp. 9a, (Scheme 6), 10a,b, (Scheme 4), and 11a, (Scheme 7) with one or two stereogenic centers in the two heterocyclic end groups. The absolute configuration of 14a,b, and thus the absolute configuration of all monochiral polymethinium dyes derived from 14a,b, has been determined by a single-crystal X-ray analysis of its 4-bromobenzenesulfonyl derivative 17a, (Scheme 1 and Fig. 1). The UV/Vis spectroscopic and chiroptical properties of the new polymethinium dyes have been studied for the first time and compared with that of similar streptocyanine dyes synthesized earlier (Tables 1 and 2) in order to find possible correlations between chiroptical properties and molecular structure.

Chlorides 17 of 14 easily cyclize to N-acylpyrazolium salts 18/19, which quantitatively afford esters 22-26 and amides 27-29 of 14. Enantiomers of the butanoic acid 14th, obtained via their diastereomeric cholesterol esters, differ in their stimulus to peroxisome proliferation. At 140 °C pyrazolium salts 18 undergo thermolysis to bicyclic β-oxa-γ-lactams 30-32. 3-Carboxyisoalkylamino-pyrazoles similarly give 1H-β-aza-γ-lactams 34. Reactions of 14 with surplus SOCl2 result in 6-chloro- 37 resp. 7-chloro-β-oxa-γ-lactams 38 via chlorosulfinylation and extrusion of SO, and in 4,4-bispyrazolyl-sulfoxide 39. A mild introduction of additional O-functions into pyrazoles affording 4,5-dihydro-3-hydroxy-5-oxo-1H-pyrazoles 52-57 is presented. Biological effects of the new pyrazoles are protection against shock and ADP-induced thromboembolism, reduction of serum lipids and improvement of blood flow.

Formation of the endo isomer 2 is favoured. Compounds 2 possess dopamine uptake inhibitory effect with an additional selective MAO-B enzyme inhibitory potential.     Notes: A new synthesis of the title crown via isoxazolo crown ether 7 and macrocyclic bis-β-eneaminoketone 10 is described. 7 can be synthesized in 14% yield by a non-template double-[3+2] cycloaddition of dinitrileoxide 5 prepared in situ from dinitropolyether 19 by dehydration with Ph-NCO and alkyne 6. The compounds 16, 17 and 18 are synthesized by the same synthetic strategy. Comparable IR and 1H NMR spectroscopic data of macrocyclic and non-cyclic compounds show, that macrocyclic conformation stabilizing effects can be ruled out. The structures of the macrocycles 1, 7, 10 and that of the Hg(II)-complex 25, synthesized by reaction of 1 with Hg(OAc)2 were established by single-crystal X-ray structure analyses. Both inter- and intramolecular hydrogen bonds are observed for the macrocyclic bis-β-eneaminoketone 10, whereas only intramolecular hydrogen bonds are formed by 1.In the Hg(II)-complex of 1 the mercury is bonded to two methylene groups. C—Hg—C is almost linear [177(1)°], the mean Hg—C distance amounts to 215(1) pm. In addition to the Hg—C bonds, each Hg makes a short contact to a carbonyl oxygen in a neighbouring molecule in the plane perpendicular to the C—Hg—C axis [Hg(1)—O(1) = 279(1) pm, Hg(2)—O(5) = 284(1) pm].

Only those compounds 7 could be oxidized with DDQ to the corresponding octahydroacridine-7-carbaldehydes 9, which bear a meta-chloro or bromo substituent (with respect to the methyl group) in addition to the para-amino group (i.e. 7c,d → 9c,d). An additionally existing chlorine atom in the starting materials 12a,c-e, thereby, is simulataneously replaced by hydrogen. The preparative advantage of this method is demonstrated by the selective access to 6α-H-isomers 13b and 13e as members of the ensemble of bicyclo[3.1.0]hexanediyl-dimorpholine diastereomers. A strong buckled bicyclohexane unit is present in 3α,6αisomer 13e as indicated by 1H NMR spectroscopy and X-ray structural analysis.

Thus, the reaction of para-(cyanomethyl)dialkyl[(methylthio)phenylmethylene]ammonium tetrafluoroborates 13a and 13b with sodium hydride affords the para-quinodimethanes 14a and 14b. Poly(meta-phenylenevinylene) 14c can be obtained when starting from 13c. We report the synthesis of para-quinodimethanes 14a and 14b and poly(meta-phenylenevinylene) 14c and describe a successive approach towards meta-phenylenevinylenes of definite length 16b and 17b starting from 12d. Moreover, we compare the stability and reactivity of para-quinodimethane 14a and 14b with those of the ortho-quinodimethane 2 and we focus on the characterization of the resulting donor and acceptor substituted compounds.

Thus, deeply colored cyanovinyl, arylazo, and arylmethine substituted bisthiazole dyes 17, 18, and 19, resp., have been formed.

    Notes: The reaction of bis(vinamidinium salts) (5a, 6) and amidinium-vinamidinium salts (7, 21) with primary amines was studied. Whereas the condensation of 6 and 7a with p-phenylenediamine and 4,4′ diaminostilbene gave rise to polymeric vinamidines (12, 18), 5a and 7b reacted with aromatic amines and diamines to give pyrrole and imidazole derivatives. The reaction of p-aminophenylacetic acid with DMF-POCl3 produced a new amidinium-vinamidinium salt (21) which could be converted into a donor-acceptor substituted diazaterphenyl derivative (22) displaying a strong solvatochromism. - The crystal structure analysis of the 2,2′-bis(vinamidinium salt) 5b revealed a 73° twist angle between the planes of the vinamidinium moieties.

    Notes: 2,6-Dimethoxy-(4a), 2,6-bis(dimethylamino)-(4b), 2,6-dichloro- (4c) and 2,6-dimethoxycarbonyl-9,10-dihydroanthracene (4d) were prepared by conventional methods and used as hydrogen transfer donors to α-methylstyrene (5) between 290-350 °C. The mechanism followed second order kinetics and the rate constants were only slightly influenced by the solvent polarity and the type of substituents introduced. The activation parameters are also closely similar in the series with ΔS≠ values between -21 and -28 cal/mol K. These results, together with the observation of a large isotope effect (kH/kD = 1.4-2.0 at 310-350 °C), suggest that the mechanism involves a primary kinetic H-atom-transfer from the donors to α-methylstyrene (5) in the rate determining step. The compounds 4a-4d constitute a new probe for investigating polar effects on H-transfer reactions.

Compounds 3 with bulky trialkylsilyl groups 3a,b only exist in the silatautomeric form 3a′, b′ under usual conditions, 3c in the N-trimethylsilyl structure. Despite of the bulky silyl substituents in 3a′, b′ their reactivity is higher than of trimethylsilyltriflate. Alcohols, carbonyl compounds, nitroalkanes and carboxylic acid esters are silylated in good yields, especially by the more reactive triisopropylsilyl derivative 3b′ in presence of tertiary amines. tert.-Butyl carboxylates and benzylcarboxylates are cleaved. Monosubstituted alkines and electron-rich (hetero) aromatics are carbosilylated in presence of N-ethyl-diisopropylamine.

By contrast, the linear cyclopenta[b]anthraquinone 23 is isolated from 10 applying the usual alkaline Marschalk conditions (aqueous methanol, NaOH, Na2S2O4). The linearly condensed 5,12-naphthacenequinones 24-28 of different degree of saturation are obtained in good combined yield from the corresponding 1-hydroxy-3-(4-oxoalkyl)-9,10-anthraquinones 19 and 20 under the conditions of the Marschalk reaction.

Compounds 14, 15, and 17 can be transformed into 2-alkyl-benzopyrylium salts 16 or 2-[1-(2-hydroxyphenyl)-alken-2-yl]-benzopyrylium salts 23 by treatment with mineral acids. With aromatic aldehydes or the Vilsmeier reagent the compounds 14, 15, or 17 are transformed into deeply colored 2-(aryl-alkenyl)-benzopyrylium perchlorates 25 or 2-(2-dialkylamino)-alkenyl-benzopyrylium salts 26, respectively.

    Notes: Conventional organic molecules for applications in second-order non-linear optics are donor-acceptor substituted π systems that show only one intense charge-transfer (CT) transition. Thus, only a single element of the second-order polarizability tensor, β, is significant in these one-dimensional systems. The advantages and optimization strategies for two new classes of molecules with multiple CT transitions and two-dimensional second-order polarizability are reviewed. These are donor-acceptor substituted π systems that lack a dipole and have a molecular symmetry of C3 or higher, and dipolar molecules of symmetry C2v. A basic introduction to the field is also given.

    Notes: Monofunctionalized Dendrons of Different Generations - as Reagents for the Introduction of Dendritic SubstituentsIn recent years dendrimers become more and more important not only in organic chemistry. They represent a new class of molecules with unique characteristic features. But dendrimers represent not only designed molecular architecture. They stand for a new concept in chemistry. They can be used to alter the properties of already existing molecular skeletons or they can be used to transfer new properties to a classical functional unit. This means that functionalized dendrimers and dendrons (dendritic building blocks) can be regarded as reagents for the preparation of new compounds with dendritic properties. In this article the synthesis and the practical use of appropriate dendritic reagents is explained. Furthermore we introduce the new technical terms „{n} dendryl-“ for dendritic substituents of n generations and „dendriagent“ which stands for dendritic reagents. Moreover we give a short outlook on future developments.

    Notes: The acid dissociation constants (Ka) of a series of 3,4-diaryl-1H-1,2,4-triazole-5-thiones (1) were determined and were found to correlated linearly with Hammett substituent constants; log Ka = 1.06 σx - 11.01. Such a result indicates that 1 exists essentially in one tautomeric form namely the thione form. Reactions of 1 with hydrazonoyl chlorides 2 gave the thiohydrazides 5. Similiar reaction of 3-phenyl-1H(4H)- 1,2,4-triazole-5-thione 1g with 2a gave the thiohydrazide 5h which was converted into 1,3,5-triphenyl-1,2,4-triazolo[3,4-c]-1,2,4-triazole (9). The latter was also prepared from 3-phenyl-5-methylthio-4H-1,2,4-triazole (6) and 2a. The mechanism of the reaction of 1 with 2 is discussed.

    Notes: The 1-aza-2-azoniaallene salt 8 prepared from 3-acetyl coumarin via the hydrazone 6 and the (chloroalkyl)azo derivative 7 reacts with nitriles to afford the 3-(3-chromenyl)-1,2,4-triazolium salts 11a-d. With diisopropylcarbodiimide the triazolium salt 13 and with norborene a tricyclic pyrazolium salt 14 are obtained. Concurrent to these cycloadditions the by-product 12 is formed by intramolecular cyclization of the cumulene 8. Similarly, the intramolecular cyclization product 18 is isolated as the sole product when the 1-aza-2-azoniaallene salt 17a (prepared from the ethyl carbazone of camphor by chlorination and treatment of the product 16a with SbCl5) was treated with nitriles, carbodiimides or alkenes. In contrast, 1,2,4-triazolium salts 20a-c, 23c, respectively pyrazolium salts 20d-f, and 1,3,4-thiadiazolium salts 23a,b are obtained by reaction of the 1-aza-2-azoniaallene salt 17b with nitriles, respectively alkenes, alkynes, diisopropylcarbodiimide, and isothiocyanates. The constitutions of two of these products (20e, 23a) were secured by X-ray structural analysis.

    Notes: A direct relationship between the structures of two-dimensional (2-D) crystallites of 4-octadecyloxy-E-cinnamic acid and 4-octadecyloxy-E-cinnamamide amphiphiles at the air-water interface and their photochemical behavior, is presented. The detailed packing arrangements of the monolayers were determined, close to the atomic level, from the diffraction pattern measured, in situ on the water surface, by the grazing incidence X-ray diffraction (GID) technique using synchrotron radiation. The products of the photochemical reaction, performed by irradiating the monolayer films directly on the water surface, were collected and analyzed. While the acid crystallites yield a β-truxinic acid product together with the cis-isomer, the amide undergoes only trans-cis isomerisation, in full agreement with the 2-D crystalline packing arrangements of the two amphiphiles on the water surface.     Notes: Hematoxylin, C16H14O6, from the extract of logwood, crystallizes as a trihydrate with the water molecules inside helical channels. It was the first organic compound used to deliberately stain a salt crystal and as such played a central role in the general development of the subject of dyeing crystals. We discuss this development and, moreover, show that efforts to stain (NH4)H2PO4 crystals with hematoxylin in 1879 provided evidence of enantioselective crystal chemistry recognizable with the unaided eye. What motivated nineteenth century chemists to dye crystals? Why and how was hematoxylin selected as the first colorant, and what were the consequences of this selection? In order to answer these questions, we offer an integrated history of hematoxylin. We trace logwood extract from the height of the Mayan civilization, through the development of the baconian scientific method, during the Atlantic slave trade, and in supramolecular stereochemical associations while dyeing crystals.     Notes: Molecular imprinting is a technique for the fabrication of biomimetic polymeric recognition sites or “plastic antibodies/receptors” which is attracting rapidly increasing interest. By this technology, recognition matrices can be prepared which possess high substrate selectivity and specificity. In the development of this technology, several applications have been foreseen in which imprinted materials may be exchanged for natural recognition elements. Thus, molecularly imprinted polymers have been used as antibody/receptor binding mimics in immunoassay-type analyses, as enzyme mimics in catalytic applications and as recognition matrices in biosensors. The best developed application area for imprinted materials, though, has been as stationary phases for chromatography, in general, and chiral chromatography, in particular. This review seeks to highlight some of the more intriguing advantages of the technique as well as pointing out some of the difficulties encountered.     Notes: Two exceptionally crowded derivatives of octaphenylnaphthalene bearing ortho-methyls on the phenyl substituents - 2,3,5,6,7,8-hexaphenyl-1,4-di(o-tolyl)naphthalene (2) and 2,3,6,7-tetraphenyl-1,4,5,8-tetra(o-tolyl)naphthalene (3) - were prepared by the addition of tetraarylbenzynes to tetraarylcyclopentadienones. Both compounds exist as a mixture of conformational isomers which do not interconvert at ordinary temperatures. The yield of 2 and its distribution of conformers is strongly influenced by the choice of precursors in the final cycloaddition; one method gives a high yield of a 55:45 trans-2:cis-2 mixture, while another gives a very low yield of a 15:85 trans-2:cis-2 mixture.     Notes: Circular dichroism (CD) spectra were determined for the bioactive (+)-enantiomers of 1 · HCl, 3 · HCl, and 4 · HBr to characterize the chiroptical properties of these pyrroloisoquinoline antidepressants. The compounds showed a low-intensity negative CD band with much fine structure between 252 and 272 nm, a medium negative CD band with fine structure between 215 and 225 nm, and a high-intensity negative CD maximum between 198 and 203 nm. Except for amplitude variation, the three CD spectra were essentially superimposable in sign and position of the bands. The CD curves for the (-)-enantiomers of 1 · HCl and 4 · HBr were opposite in sign and comparable within 5% to the (+)-enantiomers. The results are consistent with the previously assigned (Maryanoff et al. J. Med. Chem. 30:1433-1454, 1987) absolute configurations of (6S, 10bR) for 1 and 3, and (6R, 10bR) for 4.     Notes: We present evidence of cell death by apoptosis during the development of bone-like tissue formation in vitro. Fetal rat calvaria-derived osteoblasts differentiate in vitro, progressing through three stages of maturation: a proliferation period, a matrix maturation period when growth is downregulated and expression of the bone cell phenotype is induced, and a third mineralization stage marked by the expression of bone-specific genes. Here we show for the first time that cells differentiating to the mature bone cell phenotype undergo programmed cell death and express genes regulating apoptosis. Culture conditions that modify expression of the osteoblast phenotype simultaneously modify the incidence of apoptosis. Cell death by apoptosis is directly demonstrated by visualization of degraded DNA into oligonucleosomal fragments after gel electrophoresis. Bcl-XL, an inhibitor of apoptosis, and Bax, which can accelerate apoptosis, are expressed at maximal levels 24 h after initial isolation of the cells and again after day 25 in heavily mineralized bone tissue nodules. Bcl-2 is expressed in a reciprocal manner to its related gene product Bcl-XL with the highest levels observed during the early post-proliferative stages of osteoblast maturation. Expression of p53, c-fos, and the interferon regulatory factors IRF-1 and IRF-2, but not cdc2 or cdk, were also induced in mineralized bone nodules. The upregulation of Msx-2 in association with apoptosis is consistent with its in vivo expression during embryogenesis in areas that will undergo programmed cell death. We propose that cell death by apoptosis is a fundamental component of osteoblast differentiation that contributes to maintaining tissue organization. J. Cell.     Notes: A 186-base pair fragment of ors8, a mammalian autonomously replicating DNA sequence isolated by extrusion of nascent monkey DNA in early S phase, has previously been identified as the minimal sequence required for replication function in vitro and in vivo. This 186-base pair fragment contains, among other sequence characteristics, an imperfect consensus binding site for the ubiquitous transcription factor Oct-1. We have investigated the role of Oct-1 protein in the in vitro replication of this mammalian origin. Depletion of the endogenous Oct-1 protein, by inclusion of an oligonucleotide comprising the Oct-1 binding site, inhibited the in vitro replication of p186 to approximately 15-20% of the control, whereas a mutated Oct-1 and a nonspecific oligonucleotide had no effect. Furthermore, immunodepletion of the Oct-1 protein from the HeLa cell extracts by addition of an anti-POU antibody to the in vitro replication reactioninhibited p186 replication to 25% of control levels. This inhibition of replication could be partially reversed to 50-65% of control levels, a two- to threefold increase, upon the addition of exogenous Oct-1 POU domain protein.Site-directed mutagenesis of the octamer binding site in p186 resulted in a mutant clone, p186-MutOct, which abolished Oct-1 binding but was still able to replicate as efficiently as the wild-type p186. The results suggest that Oct-1 protein is an enhancing component in the in vitro replication of p186 but that its effect on replication is not caused through direct binding to the octamer motif. J.     Notes: Heparin-binding epidermal growth factor-like growth factor (HB-EGF) is an activating ligand for the EGF receptor (HER1/ErbB1) and the high-affinity receptor for diphtheria toxin (DT) in its transmembrane form (proHB-EGF). HB-EGF was immunolocalized within human benign and malignant prostatic tissues, using monospecific antibodies directed against the mature protein and against the cytoplasmic domain of proHB-EGF. Prostate carcinoma cells, normal glandular epithelial cells, undifferentiated fibroblasts, and inflammatory cells were not decorated by the anti-HB-EGF antibodies; however, interstitial and vascular smooth muscle cells were highly reactive, indicating that the smooth muscle compartments are the major sites of synthesis and localization of HB-EGF within the prostate. In marked contrast to prostatic epithelium, proHB-EGF was immunolocalized to seminal vesicle epithelium, indicating differential regulation of HB-EGF synthesis within various epithelia of the reproductive tract. HB-EGF was not overexpressed in this series of cancer tissues, in comparison to the benign tissues. In experiments with LNCaP human prostate carcinoma cells, HB-EGF was similar in potency to epidermal growth factor (EGF) in stimulating cell growth. Exogenous HB-EGF and EGF each activated HER1 and HER3 receptor tyrosine kinases and induced tyrosine phosphorylation of cellular proteins to a similar extent. LNCaP cells expressed detectable but low levels of HB-EGF mRNA; however, proHB-EGF was detected at the cell surface indirectly by demonstration of specific sensitivity to DT. HB-EGF is the first HER1 ligand to be identified predominantly as a smooth muscle cell product in the human prostate. Further, the observation that HB-EGF is similar to EGF in mitogenic potency for human prostate carcinoma cells suggests that it may be one of the hypothesized stromal mediators of prostate cancer growth.     Notes: The involvement of serine/threonine protein phosphatases in signaling pathways that control the expression of the cyclooxygenase-2 (COX-2) gene in human chondrocytes was examined. Okadaic acid (OKA), an inhibitor of protein phosphatases 1 (PP-1) and 2A (PP-2A), induced a delayed, time-dependent increase in the rate of COX-2 gene transcription (runoff assay) resulting in increased steady-state mRNA levels and enzyme synthesis. The latter response was dose dependent over a narrow range of 1-30 nmol/L with declining expression and synthesis of COX-2 at higher concentrations due to cell toxicity. The delayed increase in COX-2 mRNA expression was accompanied by the induction of the proto-oncogenes c-jun, junB, junD, and c-fos (but not FosB or Fra-1). Increased phosphorylation of CREB-1/ATF-1 transcription factors was observed beginning at 4 h and reached a zenith at 8 h. Gel-shift analysis confirmed the up-regulation of AP-1 and CRE nuclear binding proteins, though there was little or no OKA-induced nuclear protein binding to SP-1, AP-2, NF-κB or NF-IL-6 regulatory elements. OKA-induced nuclear protein binding to 32P-CRE oligonucleotides was abrogated by a pharmacological inhibitor of protein kinase A (PKA), KT-5720; the latter compound also inhibited OKA-induced COX-2 enzyme synthesis. Calphostin C (CalC), an inhibitor of PKC isoenzymes, had little effect in this regard. Inhibition of 32P-CRE binding was also observed in the presence of an antibody to CREB-binding protein (265-kDa CBP), an integrator and coactivator of cAMP-responsive genes. The binding to 32P-CRE was unaffected in the presence of excess radioinert AP-1 and COX-2 NF-IL-6 oligonucleotides, although a COX-2 CRE-oligo competed very efficiently. 32P-AP-1 consensus sequence binding was unaffected by incubation of chondrocytes with KT-5720 or CalC, but was dramatically diminished by excess radioinert AP-1 and CRE-COX-2 oligos. Supershift analysis in the presence of antibodies to c-Jun, c-Fos, JunD, and JunB suggested that AP-1 complexes were composed of c-Fos, JunB, and possibly c-Jun. OKA has no effect on total cellular PKC activity but caused a delayed time-dependent increase in total PKA activity and synthesis. OKA suppressed the activity of the MAP kinases, ERK1/2 in a time-dependent fashion, suggesting that the Raf-1/MEKK1/MEK1/ERK1,2 cascade was compromised by OKA treatment. By contrast, OKA caused a dramatic increase in SAPK/JNK expression and activity, indicative of an activation of MEKK1/JNKK/SAPK/JNK pathway. OKA stimulated a dose-dependent activation of CAT activity using transfected promoter-CAT constructs harboring the regulatory elements AP-1 (c-jun promoter) and CRE (CRE-tkCAT). We conclude that in primary phenotypically stable human chondrocytes, COX-2 gene expression may be controlled by critical phosphatases that interact with phosphorylation dependent (e.g., MAP kinases:AP-1, PKA:CREB/ATF) signaling pathways.     Notes: Monoclonal antibodies were generated to vesicular membranes of clathrin coated vesicles enriched for acetylcholinesterase (AChE). One of these, C172, recognizes vesicles which accumulate in muscle cells around nuclei associated with acetylcholine receptor AChR clusters. Immunoblots of muscle extracts and brain purified clathrin coated vesicles show that C172 recognizes a 100 kd band in muscle, but a 180 kd band in brain. Western blots of purified AP180 protein stained with the two antibodies AP180.1 and C172 displayed the same staining pattern. Tryptic digests probed with peptide antibodies (PS26 and PS27) generated to known sequences of AP180 were used to map the epitope for C172 within the brain AP180 sequence. On immunoblots of digested AP180, all AP180 antibodies and C172 recognized a 100 kd tryptic fragment, however only C172 recognized a smaller 60 kd. Our results suggest that the C172 epitope is located within amino acids 305-598 of the AP180 sequence. Confocal fluorescence microscopy of myoblasts and myotubes stained with the C172 antibody gives a punctate immunofluorescence pattern. Myoblasts stained with C172 revealed a polarized distribution of vesicles distinct from that observed when cells are stained with γ adaptin antibody which is known to localize to trans Golgi network. Myotubes stained with C172 antibody reveal a linear array of vesicular staining. Quantitative analysis of C172 reactive vesicles revealed a significant increase in number of vesicles present around the nuclei associated with the acetylcholine receptor clusters. These vesicles did not colocalize with the Golgi cisternae. These results indicate that a protein with homology to the neuron-specific coated vesicle protein AP180, is present in muscle cells associated with vesicles showing significant concentration around postsynaptic nuclei present in close proximity to AChR clusters.     Notes: Endochondral bone formation involves the progression of epiphyseal growth plate chondrocytes through a sequence of developmental stages which include proliferation, differentiation, hypertrophy, and matrix calcification. To study this highly coordinated process, we infected growth plate chondrocytes with Rous sarcoma virus (RSV) and studied the effects of RSV transformation on cell proliferation, differentiation, matrix synthesis, and mineralization. The RSV-transformed chondrocytes exhibited a distinct bipolar, fibroblast-like morphology, while the mock-infected chondrocytes had a typical polygonal morphology. The RSV-transformed chondrocytes actively synthesized extracellular matrix proteins consisting mainly of type I collagen and fibronectin. RSV-transformed cells produced much less type X collagen than was produced by mock-transformed cells. There also was a significant reduction of proteoglycan levels secreted in both the cell-matrix layer and culture media from RSV-transformed chondrocytes. RSV-transformed chondrocytes expressed two- to- threefold more matrix metalloproteinase, while expressing only one-half to one-third of the alkaline phosphatase activity of mock infected cells. Finally, RSV-transformed chondrocytes failed to calcify the extracellular matrix, while mock-transformed cells deposited high levels of calcium and phosphate into their extracellular matrix. These results collectively indicate that RSV transformation disrupts the preprogrammed differentiation pattern of growth plate chondrocytes and inhibit chondrocyte terminal differentiation and mineralization. They also suggest that the expression of extracellular matrix proteins, type II and type X collagens, and the cartilage proteoglycans are important for chondrocyte terminal differentiation and matrix calcification.     Notes: Cordyceps sinensiscontains a factor that stimulates corticosteroid production in the animal model. However, it is not known whether this drug acts directly on the adrenal glands or indirectly via the hypothalamus-pituitary axis. In the present study, we used primary rat adrenal cell cultures to investigate the pharmacological function of a water-soluble extract of Cordyceps sinensis(CS) and thesignaling pathway involved. Radioimmunoassay of corticosterone indicated that the amount of corticosterone produced by adrenal cells is increased in a positively dose-dependent manner by CS, reaching a maximun at 25 μg/ml. This stimulating effect was seen 1 h after CS treatment and was maintained for up to 24 h. Concomitantly, the lipid droplets in these cells became small and fewer in number. Immunostaining with a monoclonal antibody, A2, a specific marker for the lipid droplet capsule, demonstrated that detachment of the capsule from the lipid droplet occurs in response to CS application and that the period required for decapsulation is inversely related to the concentration of CS applied. The mechanism of CS-induced steroidogenesis is apparently different from that for ACTH, since intracellular cAMP levels were not increased in CS-treated cells. However, combined application with calphostin C, a PKC inhibitor, completely blocked the effect of CS on steroidogenesis, suggesting that activation of PKC may be responsible for the CS-induced steroidogenesis.     Notes: During early postnatal development, cardiomyocytes, which comprise about 80% of ventricular mass and volume, become phenotypically developed to facilitate their contractile functions and terminally differentiated to grow only in size but not in cell number. These changes are due to the expression of contractile proteins as well as the regulation of intracellular signal transduction proteins. In this study, the expression patterns of several protein kinases involved in various cardiac functions and cell-cycle control were analyzed by Western blotting of ventricular extracts from 1-, 10-, 20-, 50-, and 365-day-old rats. The expression level of cAMP-dependent protein kinase was slightly decreased (20%) over the first year, whereas no change was detected in cGMP-dependent protein kinase I. Calmodulin-dependent protein kinase II, which is involved in Ca2+ uptake into the sarcoplasmic reticulum, was increased as much as ten-fold. To the contrary, the expressions of protein kinase C-α and ι declined 77% with age. Cyclin-dependent protein kinases (CDKs) such as CDK1, CDK2, CDK4, and CDK5, which are required for cell-cycle progression, abruptly declined to almost undetectable levels after 10-20 days of age. In contrast, other CDK-related kinases, such as CDK8 or Kkialre, did not change significantly or increased up to 50% with age, respectively. Protein kinases implicated in CDK regulation such as CDK7 and Wee1 were either slightly increased in expression or did not change significantly. All of the proteins that were detected in ventricular extracts were also identified in isolated cardiac myocytes in equivalent amounts and analyzed for their relative expression in ten other adult rat tissues.     Notes: Activin A, a member of the TGF-β superfamily, plays roles in differentiation and development, including hematopoiesis. Our previous studies indicated that the expression of activin A by human bone marrow cells and monocytes is highly regulated by inflammatory cytokines and glucocorticoids. The present study was undertaken to investigate the regulation of activin A gene expression in the human bone marrow stromal cell lines L87/4 and HS-5, as well as in primary stromal cells. Northern blots demonstrated that, like primary stromal cells, the cell lines expressed four activin A RNA transcripts (6.4, 4.0, 2.8, and 1.6 kb), although distribution of the RNA among the four sizes varied. The locations of the 5′ ends of the RNAs were investigated by Northern blots and RNase protection assays. The results identified a transcription start site at 212 nucleotides upstream of the translation start codon. In addition, luciferase expression assays of a series of deletion constructs were used to identify regulatory sequences upstream of the activin A gene. A 58 bp upstream sequence exhibits promoter activity. However, severalfold higher expression requires a positive element consisting of an additional 71 bp of the upstream region. Promoter activity was also identified between 2.5 and 3.6 kb upstream of the start codon.     Notes: Eukaryotic cells achieve complexity by compartmentalizing a subset of cellular functions into membrane-bound organelles. Maintaining this high level of cellular organization requires precise regulation of traffic between membranes. This task is accomplished, in part, by rab proteins. How these small GTPases regulate membrane traffic between cellular compartments is not clear. Here we report the characterization of a novel rab GTPase from the soil amoebae Dictyostelium discoideum. The predicted coding sequence of the new rab gene, Dictyostelium rab11b, encodes a protein of 25 kD containing all the structural hallmarks of a rab GTPase. Comparison of the sequence with the GenBank database and cladistic analysis demonstrated Dictyostelium rab11b to be a divergent member of the rab11 branch of rab proteins. Southern analysis revealed the presence of related genes in Dictyostelium. RNAse protection assays showed the Dictyostelium rab11b gene to be expressed at uniform levels throughout growth and development. Gene deletion experiments revealed that Dictyostelium rab11b was not essential for growth or development.     Notes: The intracellular mechanisms underlying the action of the endogenous vasodilators such as NO/EDRF, adenosine, and prostacyclin acting through cGMP and cAMP, respectively, are not well understood. One important action of cyclic nucleotides in smooth muscle relaxation is to lower the cytosolic Ca2+ concentration by enhanced sequestration into the sarcoplasmic reticulum. The present study was undertaken to elucidate the potential role of phosphorylation of phospholamban, the regulator of sarcoplasmic reticulum Ca2+ pump, for the control of coronary vascular tone by NO/EDRF, adenosine, and prostacyclin. Phospholamban was identified in pig coronary artery preparations by immunofluorescence microscopy, Western blotting and in vitro phosphorylation. Segments of pig coronary artery, with either intact or denuded endothelium, were precontracted with prostaglandin F2α (PGF2α). In endothelium-denuded preparations 3-morpholinosydnonimine (SIN-1), 5′-N-ethylcarboxiamidoadenosine (NECA), and iloprost (ILO) caused both relaxation and phospholamban phosphorylation with the potency: SIN-1 〉 NECA 〉 ILO. The regulatory myosin light chain was significantly dephosphorylated only by SIN-1. In endothelium-intact pig coronary artery, L-NAME caused additional vasoconstriction and a decrease in phospholamban phosphorylation, while phosphorylation of myosin light chain remained unchanged. An inverse relationship between phospholamban phosphorylation and vessel tone was obtained. Our findings demonstrate significant phospholamban phosphorylation during coronary artery relaxation evoked by NO, prostacyclin, and adenosine receptor activation. Because of the close correlation between phosphorylation of phospholamban and vessel relaxation, we propose that phospholamban phosphorylation is an important mechanism by which endogenous vasodilators, especially endothelial NO/EDRF, control coronary vascular smooth muscle tone.
