    Notes: Abstract  Recent developments in the field of endocrine cell biology and pathology at both morphological and molecular levels are briefly outlined and discussed as a basis for endocrine tumor characterization. The main tools available for identifying the endocrine nature of the tumors, their pathogenetic interpretation, and experimental reproduction with special emphasis on tumor antecedents are reported. Based on this, classifications of endocrine tumors of the pancreas and gastrointestinal tract are developed, covering most clinical (hyperfunctional syndromes included), pathological, and biological patterns, with special emphasis on tumor prognosis.

    Description / Table of Contents: Summary Collodion baby is a clinical description for a transient condition of the newborn which is caused by various disorders of cornification. The majority of cases are due to autosomal recessive lamellar ichthyosis. In up to 10% of cases „self-healing” of the skin occurs. We report about a mature male baby who at birth was covered entirely with a shining, collodion-like membrane. There also were marked ectropion of the eyelids and eversion of the lips. After shedding of the membrane almost complete clearing of the skin occurred until the age of three months. In the case reported here skin biopsies taken on the 20th day of life showed discrete and non-specific ultrastructural changes, with no features of lamellar ichthyosis or other cornification disorders. Discussion: From these findings and those reported in the literature we conclude that the prognosis of a collodion baby is unpredictable on clinical evaluation. However an ultrastructural examination can contribute to early assessment of prognosis in this heterogeneous neonatal condition.
    Notes: Zusammenfassung Das klinische Bild des Kollodiumbabys wird durch verschiedene Verhornungsstörungen hervorgerufen; in der Regel entwickelt sich später eine lamelläre Ichthyose. Eine Selbstheilung wird in bis zu 10% der Fälle beobachtet. Wir berichten über ein männliches Reifgeborenes, welches bei der Geburt vollständig in eine glänzende, kollodiumähnliche Membran gehüllt war. Weiterhin fanden sich ektropionierte Augenlider und evertierte Lippen. In den ersten Lebenswochen löste sich die Membran gänzlich ab und bis zum Alter von 3 Monaten war die Haut des Patienten weitgehend erscheinungsfrei. Die elektronenmikroskopische Untersuchung einer am 20. Lebenstag entnommenen Hautbiopsie zeigte bei unserem Patienten keinen auf eine lamelläre Ichthyose oder eine andere Verhornungsstörung hinweisenden Strukturdefekt der Keratinozyten. Es fanden sich lediglich unspezifische Veränderungen im Sinn einer Abräumreaktion. Diskussion: Unter Berücksichtigung der Literatur läßt sich folgern, daß das klinische Bild eines Kollodiumbabys keine Rückschlüsse auf den weiteren Verlauf zuläßt. Dagegen kann die Elektronenmikroskopie prognostisch richtungsweisende Aussagen beim ätiologisch heterogenen Erscheinungsbild des Kollodiumbabys erlauben.

    Notes: Abstract Biofilms form on all implanted foreign materials. In venous access ports (VAPs), the biofilm with entrapped organisms may be the source of recurring bacteraemia. At present, little is known of the development of biofilms in VAPs. In this study light, scanning, and transmission electron microscopy were used to investigate the evolution of biofilms in VAPs implanted in 15 African children with Wilms' tumour. The VAPs were removed either emergently because of infection (n = 6) or blockage (n = 3), or electively at the end of chemotherapy (n = 6). Intact biofilms were obtained from lengths of the catheter attached to ports that had been in place for 11 days to 3 years. Each was prepared for light and electron microscopy. In infected ports, shortly after implantation biofilms were thin and comprised of apparently healthy erythrocytes (RBCs) and occasional platelets, leucocytes (WBCs), and bacteria enmeshed in a network of fibrin. Three weeks after implantation, RBCs had autolysed and large numbers of WBCs and bacterial colonies were present within and on the luminal surface. In 1 instance, the lumen of a VAP had been occluded by a “plug” of WBCs. In non-infected patients, the biofilms in long-standing VAPs were of varying thickness and primarily composed of an amorphous granular material. In most cases, healthy and necrotic bacteria were present both within the core and on the surface of the biofilms. The results suggest that while bacteria, per se, are an important factor, the presence and degradation of blood components may be an equally important factor in the development of biofilms in VAP catheters.

    Notes: Abstract  We report the first case of an extranodal follicular dendritic cell (FDC) tumour localized in the nasopharynx of a 44-year-old male patient. The tumour cells were characterized immunohistochemically by strong expression of CD21, HLA-DR and vimentin and focal expression of CD68 and cytokeratin. Electron microscopic examination revealed desmosomal cell junctions between adjacent cell processes. Molecular genetic analysis using polymerase chain reaction (PCR) showed germline configuration of immunoglobulin and T-cell receptor genes. Epstein-Barr virus (EBV) genomes were detectable by PCR. After complete surgical tumour removal and radiotherapy the patient is disease-free 20 months after the initial diagnosis.

    Notes: Abstract  Although pruritus is the cardinal symptom of atopic dermatitis, its mechanism is not well understood. Free nerve endings in the skin are involved in pruritus as itching receptors. We studied the cutaneous nerve fibres in lichenified lesions of 16 patients with adult atopic dermatitis. On immunohistochemistry, fibres immunoreactive for neurofilament, neuron-specific enolase, and protein gene product 9.5 were observed in the papillary dermis and dermoepidermal junctions as well as in the epidermis. In these areas, no fibres stained positively for substance P, neuropeptide Y, vasoactive intestinal peptide, beta endorphin, somatostatin or serotonin. On electron microscopy, the ultrastructure of subepidermal and intraepidermal free nerve endings appeared to be essentially normal. However, the distribution density of the cutaneous nerve fibres was much higher than in normal controls, and the diameter of these fibres was much larger, because of the large number of axons in each nerve fibre. Degranulation of mast cells was not seen. These findings suggest that pruritus in lichenified atopic skin is probably not caused by damage to the cutaneous free nerve endings. In such lesions, the number of the cutaneous free nerve endings is greatly increased, but they may have a normal function.

    Notes: Abstract  Tumour necrosis factor (TNF)-α-induced apoptosis is associated with several nuclear and cell surface alterations, in particular with the condensation of chromatin and the fragmentation of the cell nucleus, formation of blebs on the cell surface and breakdown of the plasma membrane. However, there is little information about the relationship between the cell surface alterations and the nuclear changes during apoptosis. To study this, cultured WEHI cells were exposed to TNF-α over different time periods. The cytological changes were studied using a correlative approach, which allowed observation of the same cell consecutively under light, scanning and transmission electron microscopy. The earliest sign of cell alteration was a reduction of the number of microvilli after 15 min of TNF-α exposure. This reaction was reversible (reappearance of microvilli) and took place during the first hour, in which neither nuclear alterations nor plasma membrane breakdown were observed. The changes in the nucleus began with condensation of chromatin after approximately 1 h of TNF-α-exposure. After 4–5 h the microvilli disappeared again, particularly in areas where the formation of blebs (blebbing) was observed. Strikingly, cell surface alterations (bleb formation) were detected only in those cells that presented with condensed chromatin, and not in cells with a normal chromatin pattern, proving at least a close correlation between nuclear and cell surface changes during the process of apoptosis.

    Notes: Abstract In almost all of the earlier reported cases of Kufs’ disease, the adult form of ceroid lipofuscinosis, the diagnosis was ascertained by cerebral tissue examination, while peripheral biopsy examination revealed an apparent poor diffusion of specific lipofuscinic deposits, the finger print profiles (FPs). We report the ultrastructural data from skin, muscle and rectal biopsy specimens from two siblings, both still living, who present clinical features of Kufs’ disease. We observed the presence of FPs in locations that differ from the previous classic reports. Our results emphasize the value of extracerebral biopsies for the diagnosis of Kufs’ disease in vivo, and suggest some physiopathological assumptions based on vascular wall involvement.

    Notes: Abstract  Guanylin, structurally related to the heat-stable enterotoxin of E. coli, is a 15-amino-acid peptide isolated from rat small intestine. We investigated the morphological effects of an intravenous injection of rat and human guanylin upon the rat intestine. Various doses of rat guanylin were injected intravenously in anesthetized rats. After 5, 10 and 30 min, rats were killed by intracardiac perfusion with aldehyde fixative, and specimens of the intestine were then prepared for light and electron microscopy. Intravenously injected rat guanylin rapidly induced mucus secretion from crypt goblet cells in the duodenum. About half of the crypt goblet cells secreted mucous granules by compound exocytosis within 5 min. The villus goblet cells, in contrast, were not sensitive to guanylin. Goblet cells in the jejunum were less responsive than those in the duodenum. This secretory response was rare in the ileum and colon. Human guanylin produced similar results. The mucus secretion induced by guanylin was inhibited by a prior-injection of atropine, but not hexamethonium. Moreover, guanylin induced intense edema in the mucosa and submucosa of the small intestine 5 min after the injection, which disappeared after 30 min. A prior-injection of atropine did not block the appearance of edema. In conclusion, the intravenous injection of guanylin induces two phenomena related to water movement: (1) compound exocytosis of mucous granules from crypt goblet cells in the rat duodenum and jejunum; (2) perineural, inter-epithelial and intra-epithelial edema in the rat small intestine.

    Notes: Abstract Insulin-dependent diabetes mellitus is a chronic metabolic disease that causes long-term secondary complications such as neuropathy. The occurrence of diabetic neuropathy has generally been thought of as being associated with hyperglycaemia. However, in a previous light microscopic examination of plantar nerves in diabetic BB/Wor rats treated with insulin implants we found that eu-/hyperglycaemic rats present a normal picture, whereas eu-/hypoglycaemic rats show severe changes. The aim of the present work is to supplement our previous light microscopic report with electron microsocpic data from the lateral plantar nerve of normal, eu-/hyperglycaemic and eu-/hypoglycaemic BB/Wor rats. Under the electron microscope lateral plantar nerves collected from eu-/hyperglycaemic rats presented a qualitatively normal picture. In addition, the fibre numbers and the size distribution of the myelinated fibres were normal. In contrast, specimens from eu-/hypoglycaemic BB/Wor rats showed severe qualitative changes, interpreted as signs of axonal de- and regeneration. The total number of axons was somewhat subnormal and the sizes of the myelinated fibres were strongly shifted towards smaller diameters. These data confirm our previous light microscopic observations. We conclude that eu-/hypoglycaemic BB/Wor rats treated with insulin implants, but not similarly treated eu-/hyperglycaemic animals, develop a neuropathy in their plantar nerves.

    Notes: Abstract Human spinocerebellar degeneration is one of the intractable diseases. We studied the detailed neuropathology of cats with hereditary cerebellar degeneration obtained from the experimental breeding. The findings included almost total loss of Purkinje cells with an increase in Bergmann’s glia in the cerebellar hemisphere, preservation of some Purkinje cells in the vermis and moderate neuronal depletion of the olive nucleus. Cerebellar and pontine nuclei were normal. The cerebrum and spinal cord as well as the peripheral nervous system appeared normal. Electron microscopic examination revealed swelling of the distal dendrites of Purkinje cells in the less-affected nodule of the vermis, and clusters of presynaptic boutons without any synaptic contact in the severely affected folia where Purkinje cell bodies and dendrites disappeared. Prolonged existence of presynapses in the molecular and Purkinje cell layers was confirmed by positive immunoreactivity to anti-synaptophysin. Quantitative analysis using electron microscopy demonstrated an apparent increase in the density and mean size of presynapses in the molecular layer of the severely affected folia. These findings indicate that degeneration of Purkinje cells started at the most distal part of the dendrite in this animal model of cerebellar degeneration, and that presynapses, axon terminals of the granular cells and basket cells can exist for a long time even after complete degeneration of the Purkinje cells. Further investigation of this novel animal model may promote a better understanding of pathogenesis of human hereditary cerebellar degeneration.

    Notes: Abstract  The innervation of the knee joint synovial membrane of the guinea pig, i.e., the synoviocyte layer, the subjacent connective tissue and the connective tissue region beneath, was analyzed with immunohistofluorescence and electron microscopy. A screening of the innervation with antibodies against the general axon marker – protein gene product (PGP) 9,5 – revealed the presence of nerve fibers distributed in various regions of the knee joint synovial membrane. Confirmating previous studies, some of these nerve fibers stained with antibodies to tyrosine hydroxylase (TH), neuropeptide Y (NPY), substance P (SP), calcitonin gene-related peptide (CGRP), and vasoactive intestinal polypeptide (VIP). In addition, dynorphin (DYN)-containing fibers were detected, which have not been reported previously in normal joints. In general, the immunoreactive fibers were observed close to the synoviocytes and at blood vessels. Fibers with colocalization of NPY- and TH-like immunoreactivities (LIs), as well as of DYN- and TH-LIs were demonstrated. In the electron microscope, bundles of unmyelinated fibers as well as single fibers were found in the connective tissue region below the synoviocytes. Varicose parts of the nerve fibers contained mainly small, clear vesicles. Small and large dense-cored vesicles were also seen, but less frequently. Denser portions of the plasma membranes of some axons were observed in these regions, facing the extracellular space. Myelinated fibers were also observed in some nerve bundles. These findings emphasize the complex innervation of the synovial membrane, with nerve fibers containing a host of neuroactive substances. Altogether, these fibers are probably involved in many functions such as vasoregulation and control of synovial secretion in addition to being a source of mediators in joint inflammation.

    Notes: Abstract  Specimens of human cerebral cortex were obtained during neurosurgical operations and studied by immunocytochemistry and electron microscopy, using antibodies to the metabotropic glutamate receptor subunit mGluR1a and the ionotropic glutamate receptor GluR2/3. A small number of non-pyramidal neuronal cell bodies were labelled for mGluR1a. Double immunolabelling with mGluR1a and GluR2/3 showed that most pyramidal cell bodies were labelled for GluR2/3 but not for mGluR1a. Despite the non-colocalisation of these two receptor subtypes in cell bodies, however, many dendrites and dendritic spines were double-labelled for mGluR1a and GluR2/3 at electron microscopy. As there is evidence that most neurons positive for GluR2/3 are pyramidal cells, this suggests that mGluR1a is present in dendrites of pyramidal neurons, despite absent or low levels of immunoreactivity in their cell bodies.

    Notes: Abstract The aim of this electron-microscopic study was to analyze the distribution of synaptic contacts on the cell bodies and dendrites of permanently axotomized adult cat spinal α-motoneurons. Following transection and ligation of the medial gastrocnemius nerve, the synaptic covering of the cell bodies and three different dendritic compartments of homonymous α-motoneurons was analyzed quantitatively at 3, 6, and 12 weeks postoperatively. The synaptic boutons were classified according to their size and the shape of their synaptic vesicles. On the soma, a transient increase in the number of boutons was noted at 3 weeks and 6 weeks postoperatively, while after 12 weeks the bouton number had decreased to half of its normal value. The transient increase was mainly due to an increase in the number of F-type boutons. At 12 weeks postoperatively, the synaptic covering was reduced by 83% on the soma and by 57% on the proximal dendrites. In the distal dendritic regions, the values for synaptic covering remained largely unchanged. In summary, axotomized motoneurons exhibit a reduction in synaptic covering which is maximal on the cell body and becomes less pronounced centrifugally along the dendrites. However, if also taking into account the loss of distal dendritic branches that occurs in axotomized motoneurons, the total loss of boutons is several times larger in the dendrites than on the soma.

    Notes: Abstract The morphological and physical characteristics of the capsule of Vibrio cholerae O139 were examined. An electron microscopic study using the freeze-substitution technique showed that all of the V. cholerae strains of the O139 serogroup examined have a very thin fibrous layer on the outside of the outer membrane. In contrast, the mutants of strain O139, strain MO10T4 (which lacks capsule synthesis), and strain Bengal-2R1 (which fails to synthesize both the capsule and the O-antigen of lipopolysaccharide) were all found to have lost the surface layer. In addition, the capsule layer could also not be observed on the surface of V. cholerae strain O1. To determine the biological characteristics of the capsule of strains of the O139 serogroup, we investigated the serum killing activity and bacterial phagocytosis by polymorphonuclear leukocytes. The O139 strains were more resistant to the serum killing activity than were the V. cholerae O1 strain and the O139 mutant strains, thus suggesting that the existence of the capsule gave a serum-resistant character to the O139 strains. The surface character of the O139 strains had the same hydrophobic character as did that of the O139 mutant strains and the O1 strain. In addition, all the V. cholerae O1 and O139 strains examined, including the mutant strains, were effectively ingested by the human polymorphonuclear leukocytes. The number of ingested bacteria was not significantly different among the strains, and the ingestion of the acapsular O139 mutants thus showed that the capsule does not play an antiphagocytic role. These data suggest that the capsule of V. cholerae O139 has a physiological function different from that of the ordinal hydrophilic capsule that is found in invasive bacteria such as Klebsiella pneumoniae.

    Notes: Abstract Natural enrichments of magnetic bacteria from the Itaipu lagoon near Rio de Janeiro were dominated by coccoid-to-ovoid morphotypes that produced unusually large magnetosomes. To determine the phylogenetic position of these unusual microorganisms, 16S rRNA genes were retrieved from bacteria magnetically separated from sediment of the Itaipu lagoon by in vitro amplification and cloning of PCR products into a plasmid vector. Partial sequencing of the obtained clones revealed two clusters of closely related sequences affiliated to a distinct lineage consisting exclusively of magnetic bacteria within the α-subclass of Proteobacteria. For a detailed phylogenetic analysis, several almost complete sequences of the 16S rRNA genes were determined. One representative clone of each cluster provided a PCR template for the in vitro transcription of group-specific polynucleotide probes complementary to a variable region of the 16S rRNA molecule. At least three different morphotypes of magnetic bacteria were reliably identified by post-embedding hybridization of ultra-thin sections. Electron microscopic analyses of hybridized cells enabled for the first time a detailed description of the morphological variety and ultrastructure of phylogenetically identified, uncultured magnetic bacteria. Two distinct coccoid bacteria were identified by the transcript probe complementary to the 16S rRNA sequence mabrj12, whereas the probe complementary to the sequence mabrj58 allowed the identification of an ovoid morphotype that displayed magnetosomes with the largest volumes observed to date.

    Notes: Abstract DNA bound on clay minerals, sand, and humic acids has been shown, both in vitro and in situ, to be capable of transforming bacteria and to resist degradation by nucleases, which could result in the crypticity of genes in soil and other natural habitats. To determine where DNA is bound on clay minerals, which may help to explain how bound DNA becomes resistant to degradation by nucleases but retains the ability to transform competent cells, chromosomal DNA from Bacillus subtilis bound on montmorillonite (M) and kaolinite (K) was examined by X-ray diffractometry and transmission and scanning electron microscopy. X-ray diffraction analysis showed that the basal spacings of M and K were not altered, indicating that this DNA did not significantly intercalate the clays. Scanning and transmission electron microscopy showed that the binding of this DNA was primarily on the edges of M and K, although some binding was also apparent on the planar surfaces. Based on the results of these studies, it is postulated that: 1.extension from the edges of the clays enables the unbound end of DNA to interact with receptor sites on competent cells and result in their transformation; and 2.binding on clays alters the electron distribution and/or conformation of DNA, which reduces its hydrolysis by nucleases.

    Notes: Abstract  In transmissible spongiform encephalopathies (TSE), such as scrapie in animals and Creutzfeldt-Jakob disease in humans, the central event is the conversion of a host-encoded amyloidogenic protein (PrPc) into an abnormal isoform (PrPsc) that accumulates as amyloid in TSE brain. PrPc is a membrane sialoglycoprotein synthesized in the central nervous system and elsewhere. We have examined the ultrastructural localization of PrPc in numerous hamster and some human extracerebral tissues, by means of a post-embedding electron-microscopic method combined with immunogold labeling. In stomach, intestine, lung, and kidney from hamsters, and in stomach, kidney, and spleen from humans, immunogold labeling specific for PrPc is observed on various cellular substructures related to secretory pathways: Golgi apparatus, secretory globules, and plasma membrane. In mucous epithelial cells of stomach and intestine, PrPc appears to be concentrated in secretory globules, suggesting a role for PrPc in the secretory function of the digestive tract. The secretory aspect of PrPc may be a key to understanding the physiopathological mechanisms underlying TSE.

    Notes: Abstract  The evidence for vesicular transport as a mechanism for secretion by human basophils is reviewed. Initially, direct electron-microscopic inspection of experimentally produced and sequentially biopsied contact allergy skin lesions revealed a unique form of secretion termed piecemeal degranulation, characterized by the slow emptying of secretory granule contents (with retention of empty containers) in the absence of extrusion of entire granules. Budding of small vesicles to/from secretory granules was observed, and cytoplasmic vesicles were abundant. A generalized degranulation model was proposed to unify classical regulated secretion and this new form of secretion. Investigation of the mechanism(s) of secretion from human basophils required the development of numerous tools and resources. Chief among these were: (a) isolation and purification of circulating basophils; (b) identification of specific growth factors to increase the supply of this rare granulocyte; (c) understanding of secretogogue mechanisms and reliable analyses of secreted basophil products; and (d) development of ultrastructural preparations allowing imaging of small vesicles and quantifiable small electron-dense tags for granule materials in small vesicles. Applications of these tools to well-defined models of basophil secretion have established a role for vesicles as a mechanism for effecting secretion of histamine and the Charcot-Leyden crystal protein from activated human basophils.

    Notes: Abstract Specific populations of hepatic sinusoidal cells were stained with monoclonal antibodies that recognize monocytes/macrophages (ED1), tissue macrophages (Kupffer cells) (ED2), MHC class II (Ia) antigen (MRC OX6), and dendritic cells/γ,δ T-cells (MRC OX62) and analyzed by light and electron microscopy. The majority of ED1+ and/or ED2+ cells were localized to the hepatic parenchyma, whereas OX6+ and/or OX62+ cells were more densely distributed within Glisson’s sheath than in the hepatic parenchyma. Double-immunoperoxidase staining of normal liver for ED1, ED2, and OX6 identified dendritic cells (DC) of two different phenotypes, ED1+ED2–OX6+ and ED1–ED2–OX6+. DC can be classified into three different types based on ultrastructural characteristics. The first type (type I) is characterized by one or more long cytoplasmic processes and a well-developed lysosomal system. The second type (type II) has an inconspicuous lysosomal system, abundant hyaloplasm, and characteristic short cytoplasmic processes. The third type (type I–II) has cytologic features intermediate between those of type I and type II DC. At the electron-microscopic level, these three cell types are found in the sinusoidal lumen, whereas the majority of type II DC are located in the space of Disse and Glisson’s sheath. Furthermore, some OX6-labeled elongated DC appeared to traverse the lumen of sinusoids through endothelial pores to enter the space of Disse. One hour after intravenous injection of latex particles (0.81 μm in diameter), numerous latex-laden dendritic cells (ED1+OX6+, type I and type I–II) were detected in the lumen of hepatic sinusoids, but not in the space of Disse or Glisson’s sheath. These findings suggest that normal rat liver contains resident dendritic cells which downregulate phagocytic activity and mature into potent accessory cells during migration from the portal vein toward the central vein. These DC then traverse the sinusoidal lumen to the hepatic lymph system via the space of Disse.

    Notes: Abstract Interaction of the Escherichia coli DnaA (replication initiator) protein with restriction fragments of phage λ DNA demonstrated differential binding of DnaA along the whole λ DNA. Interaction of DnaA with the λ replication region (from the promoter p R to the origin of replication, oriλ) demonstrated a strong binding of DnaA to the region around the p o promoter where synthesis of a short antisense oop RNA is initiated. The four sequences protected by DnaA (two 9mers and two 5mers) are not related even to a relaxed DnaA box. The pattern of protection of these four sequences and the location of three DNase I hypersensitive sites in the λ DNA r strand, together with results of mobility shift assays and electron microscopy studies, may indicate an interaction involving DnaA monomers bound to different DNA positions on one side of the helix and the formation of higher-order nucleoprotein structures. Therefore, it is tempting to suggest that DnaA, in addition to its activity in regulation of replication and transcription, could be considered as a factor which structures certain chromosomal regions.

    Notes: Summary Exines fromTypha angustifolia L. pollen were dissolved in hot 2-aminoethanol. The solubilisate was successively fractionated and reaggregated via a dialysis cascade with dialysis tubings of different exclusion volumina. Four fractions of reaggregated material with different molecular mass were obtained. Fraction 1 with a molecular mass above 25,000 Da, fraction 2 with a molecular mass between 10,000–25,000 Da, fraction 3 with a molecular mass between 5,000–10,000 Da, and fraction 4 of a molecular mass lower than 5,000 Da. The fractions were comparatively analysed by scanning and transmission electron microscopy and X-ray microscopy. The material of the fractions with a molecular mass above 10,000 Da exhibit high congruence to the initial material. Analysis of the reaggregated material with the lowest molecular mass revealed special distinct substructures which in form and size showed high similarities to substructures of exines described in literature. In detail, spherical substructures consisting of an electron-dense core surrounded by an electron-transparent corona and in addition elongated substructures with a distinctive surface sculpture were detected.

    Notes: Summary In the seed coat ofGasteria verrucosa the deposition of phytomelan takes place during seed development in three stages. Phytomelan is a black cell wall material which is chemically very inert. First the radial walls and part of the transverse cell wall of the outer epidermis of the outer integument become thickened by exocytosis of dictyosome vesicles. Callose is deposited at the tangential plasma membrane against those walls. After the callose deposition about two thirds of the original cell volume is filled with callose. During the second stage the callose is broken down, probably into glucose monomers or small polymers. At the same time cellulose is deposited at the outer tangential plasma membrane, forming a wall between the dissolving callose and the plasma membrane. In the third phase small granules appear in the solution of dissolved callose. which grow out and finally fuse to form a block of phytomelan, consisting of spherical 15-nm units. Remarkable is the function of the callose: it determines the size of the phytomelan block, and it probably functions as carbohydrate source for the phytomelan synthesis and/or for the cellulose inner layer. In this study transmission electron microscopy and cryo scanning electron microscopy are used to study the three developmental stages of the formation of the phytomelan layer.

    Notes: Summary The filamentous brown algaHincksia hincksiae can be infected by a large icosahedral double-stranded DNA virus (HincV-1). The virus shows extended latency and is replicated only in cells homologous to sporangia. Virus formation was studied by transmission electron microscopy, DAPI staining, and β-tubulin immunofluorescence. Inhibition of cytokineses results in multinucleate cells, which are the first indication of virus replication in productive cells; the microtubular cytoskeleton does not seem to be affected by the virus. Replication of viral DNA begins in the nuclei, which increase in size and eventually disintegrate. Virus assembly takes place in a mixed nucleo-/cytoplasm. Capsids bud from cisternae, which are interpreted as modified endoplasmic reticulum aggregated to virus assembly centres. The internal membranous component of the virus is thus derived from the endoplasmic reticulum. The particles are empty (electron translucent) when assembled, and the nucleoprotein core seems to be packaged subsequently through an opening in the capsid. A number of fine structural features not previously reported from brown algae and related to virus formation are described. Our results on Hincksia hincksiae virus are compared with observations made on various other icosahedral DNA viruses infecting eukaryotic algae and animals.

    Notes: Abstract In animal experiments, studies on the mechanisms involved in drowning were carried out using latex and gold tracers of defined size and concentration. The tracers were detectable by fluorescence microscopy (latex tracers) and by electron microscopy (gold tracers) in the lungs, kidneys and lymph nodes and were analysed further by X-ray microanalysis using a transmission scanning electron microscope. Tracers with small diameters were shown to penetrate intercellular gaps of the alveolar epithelium and the larger tracers were incorporated into the epithelial and endothelial cells by active pinocytotic mechanisms thus passing through the air-blood barrier. The detection and analysis of tracers in organs of the systemic circulation originating from the immersion fluid can assist in understanding the pathophysiology of drowning and in some selected cases, in making a more definitive diagnosis.

    Notes: Abstract Congenital hydrocephalus was induced by intraperitoneal administration of antipregnant rat kidney serum (APRKS) into rats at 9 days of pregnancy. To clarify a correlation between the oncogenes and expression of hydrocephalus, immunostaining of the fetal rat nervous system for p53 was carried out using Ab-1, a pantropic antirat p53 antibody, and Ab-3, raised against a mutant form of rat p53, Ab-1 staining was observed in cells of the neural plate of the fetus, the yolk sac, and the placenta. Because no staining occurred with Ab-3, it was likely that the p53 induced by antiserum injection was the wild-type protein. This result indicated that p53 may represent a reparative reaction to DNA damage from the APRKS; also, when DNA is irreparably damaged, p53 could induce apoptosis by favoring transcription of a gene encoding an inducet of apoptosis. Insufficient apoptosis, on the other hand, may induce failure of fetal development, contributing in particular to congenital hydrocephalus and other central nervous system anomalies.

    Notes: Abstract A 12-year-old Japanese boy noticed a red nodule on the distal portion of his right great toe about 2 months before entering our clinic. There was evidence of traumatic injury and infection before the appearance of the nodule. Histological examination showed the subcutaneous localization of bony spicules lined by osteoblasts. Haversian canals, which contained blood vessels and connective tissue, were also seen. Electron microscopic examination revealed normal bone structure, but, interestingly, mesenchymal cells that were localized around the tumor had intranuclear inclusions composed of filamentous bundles. From these findings, this case was diagnosed as subungual exostosis. It is suggested that the appearance of intranuclear inclusions, reported here for the first time, is related to the pathogenesis of subungual exostosis.

    Notes: Abstract The maxillary mucosae of chronic sinusitis patients were observed by transmission and scanning electron microscopy. Although the epithelium was severely damaged by the chronic inflammation and the ciliated cells and goblet cells were degraded and had peeled off, most of the basal cells survived at the surface of the mucosa. The surviving basal cells extended their cytoplasmic processes toward each other and finally covered the bare surface of the mucosa. Cells having a few cilia and many microvilli were occasionally observed. Immunohistochemistry for PCNA (proliferating cell nuclear antigen) was also performed. The nuclei of many basal cells in the nonciliated area and those of a few ciliated cells were positive for PCNA. These findings suggest that the maxillary mucosa of chronic sinusitis has the ability to regenerate and that basal cells play the role of precursors to ciliated cells.

    Notes: Abstract To examine the morphological changes of HL-60 cells undergoing apoptosis, we induced apoptosis by etoposide treatment, and compared the findings obtained with an electron microscopic cytochemical method with those obtained with in situ end-labeling (ISEL) of DNA and a DNA fragmentation assay. Apoptotic cells were detected by ISEL of DNA 1h before the ladder formation in the DNA fragmetation assay, and an accumulation of DNA was observed simultaneously around the nucleoli and nuclear membrane by DNA-staining electron microscopy. These findings revealed that the apoptosis-related changes occurred in the DNA before the appearance of ladder formation. DNA staining with osmium ammine B revealed, at higher magnification, a rough fiber formation of DNA before apoptosis and an accumulation of small particles with the course of apoptosis. The number of primary granules stained for peroxidase in the cytoplasm decreased as apoptosis progressed. An examination by the periodic acid-thiocarbohydrazide-silver proteinate (PA-TCH-SP) method revealed in number after the induction of apoptosis. Thus, apoptosis-related changes were observed not only in the nucleus but also in the cytoplasm.

    Notes: Abstract Biopsy specimens obtained from the liver of a 32-year-old woman, diagnosed with acute fatty liver of pregnancy, were examined by light and electron microscopy. The patient had become pregnant after artificial insemination. At the 36th week of gestation, she suffered simultaneous fetal distress and liver dysfunction, and a cesarean operation was done immediately. Liver biopsies were performed on the 7th and 38th day after the cesarean operation. The first biopsy revealed enlarged hepatocytes in pericentral areas. Many transformed mitochondria with tubular cristae, which were often seen in steroid hormoneproducing cells and adrenal cortical cells, and microvesicular fatty droplets were detected in some swollen hepatocytes, as revealed by electron microscopy. At the second biopsy, almost all hepatocytes had recovered normal morphology as the result of appropriate treatment.

    Notes: Abstract Morphological changes in the CA1 region of the hippocampus in the rat cardiac arrest model were studied with the in situ nick-end labeling (TUNEL) method and light and electron microscopy. The TUNEL-positive pyramidal cells first appeared on day 1, increased in number with time, and reached a peak at 7 days after recirculation. At the ultrastructural level, cell shrinkage, nuclear fragmentation, and an increased number of atuophagic vacuoles of the pyramidal cells were observed in the CA1 region. The brief ischemia activates the microglial cells in the CA1 region, and these cells were found to increase in number with time. The microglial cells were seen to adhere to degenerating pyramidal cells and to phagocytose the apoptotic neurons selectively.

    Notes: Abstract We examined a case of an 87-year-old man who had had a 1-month history of ileus of unknown cause. Postmortem examination revealed numerous foci of constriction and thickening throughout the small intestine, associated with perforation and panperitonitis. Histologically. diffuse infiltration of anaplastic atypical cells was found in the wall of almost the entire small intestine. The atypical cells were immunohistochemically positive for carcinoembryonic antigen and epithelial membrane antigen. In the electron microscopic examination, most of those cells had microvilli with filamentous core rootlets and terminal webs. There was no atypical cell infiltration in the stomach or large intestine. We concluded that this case was primary linitis plastica type carcinoma of the small intestine involving almost the entire small intestine.

    Notes: Abstract The intestinal absorption of a medium-chain triglyceride (MCT) was studied by electron microscopy and biochemical analysis. In jejunal absorptive cells of rats fed tricaprylin, the smooth endoplasmic reticulum in the apical cytoplasm appeared to increase in number and contained one or two particles about 40–80 nm in diameter that were less electron dense and similar in size and profile to very low density lipoprotein. Similar particles were also observed packed in the dilated Golgi sacs and in the extended intercellular spaces. These particles were remarkably increased in number as compared with those in fasted rats. Biochemical analysis of lymph from the main intestinal lymph duct showed that caprylate was apparently demonstrated only in the lymph of rats given tricaprylin at the maximum rate 3h after oral administration. The study strongly suggests that medium-chain triglyceride is at least in part transported via lacteal, possibly in the form of very low density lipoprotein.

    Notes: Abstract Primary ovarian sarcoma is a rare neoplasm. The diagnosis sometimes becomes difficult by light microscopic examination alone because of the rarity and heterogeneity of these tumors. Immunohistochemical and ultrastructural studies are very useful for diagnosis. Here, we describe two cases of ovarian sarcomas: fibrosarcoma and leiomyosarcoma, and a case of carcinosarcoma (homologous malignant mixed müllerian tumor). In addition to histological findings, immunohistochemical and ultrastructural observation was undertaken to make a final diagnosis. Clinical outcome was variable in the three cases. It was unlikely to be related to the disease stage or treatment, such as surgical excision or anticancer drugs, whereas the mitotic index may be an important prognostic indicator in ovarian sarcomas.

    Notes: Abstract To investigate the mechanism of synovial pannus formation in rheumatoid arthritis, immunohistochemical and immunoelectron microscopic studies with monoclonal antibodies against the adhesion molecules, CD54 (ICAM-1), CD 11a (LFA-1), CDw49a (VLA-1), CDw49b (VLA-2), CDw49c (VLA-3), Cdw49d (VLA-4) and CDw49e (VLA-5), were carried out to determine the pattern of distribution of these molecules at the rheumatoid synovial cartilage junction. Treatment with anti-ICAM-1 resulted in membrane staining of most of the macrophages and fibroblasts infiltrating the synovial tissue and bordering the pannus-cartilage junction, suggesting the possibility that ICAM-1 may function to facilitate the adhesion of synovial type A cells bearing ICAM-1 to type B cells of the pannus. ICAM-1-positive macrophages and fibroblasts were often found to be in contact with lymphoid cells, suggesting also that a cellular immune reaction occurs in the formation of the pannus. In addition, VLA-3, VLA-4 and, particularly, VLA-5 were the predominantβ1 integrins expressed by rheumatoid synovial pannus. Since these three integrins all function as fibronectin receptors, it is possible that the fibronectin-rich environment of the rheumatoid cartilage surface effectively traps pannus cells expressing high levels of these molecules. The VLA-5 molecule was found in a pericellular and interterritorial matrix distribution in the present study, strongly suggesting that a recepfor-ligand interaction between VLA-5 and cartilage matrix may occur at the early stage of pannus formation. Furthermore, an increase inβ1 integrin may be necessary for the growth of the pannus and also for the upregulation of the VLA molecules, leading secondarily to increased attachment.

    Notes: Abstract A case of cervical juxtamedullary meningeal melanocytoma is presented along with results of careful neuroradiological and pathological examinations. The authors review the previous literature and discuss the problems related to the intriguing diagnosis of this very rare entity. CT and MRI show variable appearances due to different degrees of melanization and do not reliably allow us to distinguish meningeal melanocytomas from other pigmented tumors. Pathological examination of the tumor is critical for diagnosis, but is absolutely must comprise electron microscopy and immunohistochemistry. Immunohistochemical and ultrastructural criteria are thus proposed to distinguish meningeal melanocytoma from pigmented meningioma or schwannoma and malignant melanoma (primary or secondary).

    Description / Table of Contents: Anteroposterior and tangential traction on the central retina is an important factor in the pathogenesis of idiopathic macular hole formation. Histological studies have shown that macular holes of different stages can be associated with epiretinal membranes. Such membranes can be removed during surgery for macular holes. We investigated such tissue samples of 11 patients with macular holes in stages II – IV. Light microscopially, the tissue consisted of a thin collagen layer mostly covered by a thin layer of cells. Ultrastructural analysis revealed glial cells and macrophages as cellular components. The collagen can be ascribed to vitreous, inner limiting membrane and newly formed collagen. According to the morphological findings a multilayered tissue structure can be assumed. Macrophages were found on the retinal side of the inner limiting membrane and at the vitreal side of the tissue. Therefore, the macrophages probably originate from the retina as well as from the vitreous as so-called resident hyalocytes. Glial cells covered the inner limiting membrane forming pericellular collagen to which outer vitreous collagen fibrils can be attached. The multilayered membrane structure might possibly be the cause for only partial laminar surgical extraction so that contractile or potentially proliferative tissue residues might be one of the reasons for surgical failures after incomplete membrane peeling.
    Notes: A.-p. und tangentiale Traktionen an der zentralen Netzhaut spielen für die Entstehung idiopathischer Makulaforamina eine wichtige Rolle. Durch histologische Untersuchungen ließen sich bei Makulaforamina unterschiedlicher Stadien epiretinale Membranen nachweisen, die für die tangentiale Traktion auf die Foramenränder verantwortlich gemacht werden. Von 11 Patienten konnten wir derartige, von uns chirurgisch entfernte Membranen licht- und elektronenmikroskopisch untersuchen. Die Membanen bestehen lichtmikroskopisch aus einem azellulären Kollagenband mit einem schmalen, teilweise nur einschichtigen Zellbesatz. Elektronenmikroskopisch lassen sich als zelluläre Komponenten Gliazellen und Makrophagen differenzieren. Die Kollagenanteile sind Glaskörperkollagen, neugebildetem Kollagen und der Lamina limitans interna zuzuordnen. Aus den Befunden läßt sich eine Schichtung der Membranen rekonstruieren mit sublaminären Makrophagen, der Lamina limitans interna und epilaminären Gliazellen und vereinzelten Makrophagen, die am ehesten residenten Hyalozyten entsprechen. Auf der vitrealen Seite können auch Verbindungen der Membran zum äußeren Glaskörperkollagen bestehen. Die Membranen zeigen Charakteristika, die in ihrem Aufbau epiretinalen Proliferationsmembranen entsprechen. Die topographische Schichtung der Membranen könnte zur Folge haben, daß chirurgisch teilweise nur Lamellen und nicht die gesamte Membran abgeschält werden. Kontraktions- oder auch proliferationsfähige Membranreste können dann einer der Faktoren für operative Mißerfolge sein.

The particles on the filters were analysed for their elemental content using energy-dispersive X-ray analysis. In addition, the appearance of the silicone tubing used in the pump over 3 and 72 h was assessed and compared to that of flow-only and unused tubing. More particles were found on the microfilter when fluid had been delivered via the pump than on those through which non-pumped fluid had passed or that were unused. Elemental silicon-containing particles were only found on the filter when a pump had been attached to the IV line. The flow-only and unused tubing were found to have adherent particles on the inner surface that were not seen once the tubing had been used for 3 h in the Ivac 560 pump. Also, after 72 h use, the silicone tubing had a deformed inner layer. The clinical significance of these findings is yet to be determined, but it does appear that silicone embolisation occurs during pump-assisted infusions in children.

    Notes: Abstract Papillary adenoma of type II pneumocytes is a rare tumour. It is considered to be a benign neoplasm and is derived from immature cells in the bronchioloalveolar epithelium, however, its biological nature has not been elucidated. We report a case of an adenomatous tumour; a papillary adenoma of type II pneumocytes, which we regard as possessing malignant potential. Light microscopically, as well circumscribed, papillary tumour of predominantly cuboidal cells resembling type II pneumocytes was found, but Clara type and ciliated cells were also present. Immunohistochemically, the tumour cells reacted positively with antibodies to surfactant apoproteins (A, B), carcinoembryonic antigen, cytochrome P-450 1A1-2 and 2B1-2. Ultrastructurally, many osmiophilic lamellar bodies and electron-dense granules were demonstrated. Semi-serial sections revealed signs of transbronchial dissemination and vascular invasion. Morphometry using 12-dimensional cluster analysis disclosed features of the tumour cells which resembled those of pneumocyte type II adenocarcinoma. These findings suggest that the present case has some malignant characteristics and originates from immature bronchiolar or alveolar cells, with a potential to develop into both type II pneumocyte and Clara cell type adenocarcinomas.

    Notes: Abstract This study describes ultrastructural alterations of the interstitial stroma in the rat bladder epithelium during N-butyl-N-(4-hydroxybutyl)nitrosamine (BBN)-induced bladder carcinogenesis using scanning (SEM) and transmission (TEM) electron microscopy with NaOH treatment. The results obtained with SEM demonstrated the occurrence and development of stroma protrusions which exhibited pipe-like structures in the rat bladder epithelium following administration of BBN. Number and size of blood vessel sections also gradually increased both in the stroma and within the layer of the proliferated epithelial cells as examined by light microscopy (LM) and TEM. In this study stroma alterations were not only observed in malignant lesions of rat bladder, but hyperplastic lesions were also accompanied by stroma alterations. It is suggested that: (1) the interstitial stroma of the rat bladder epithelium may exhibit pathological changes in structure and these changes may correlate with the progression of epithelial cell proliferation following administration of BBN and (2) one of the most important alterations in the stroma is the occurrence of neovascularization, which may induce structural modification of the stroma in the processes of bladder tumor growth and development.

    Notes: Abstract Rat neonate vomeronasal organs were transplanted into the parietal cortex of littermates to examine their survival and the behavior of axon growth into the surrounding host brain parenchyma. After survival times of 10–100 days the brains were processed for ultrastructural examination. The transplanted vomeronasal organs (VNO) formed several vesicles lined with a sensory epithelium. From these sensory epithelia, VNO neurons leave the epithelium and enter the host brain. Transplant neurons grew axons that fasciculated into bundles surrounded by sheath cell processes and formed one or more fiber plexuses containing distinct globose or spherical-shaped glomeralar-like structures. The glomeruli consisted of nerve terminals between which existed asymmetric synaptic contacts. Rarely did we observe clear reciprocal synapses. The glomeruli also contained terminals that showed signs of degeneration, such as increased density of the terminals, clumping of mitochondria and multivesicular bodies. The glomeruli were not partitioned or subdivided by glial septa; however, glial profiles were interspersed among the sensory terminals. Transplant glomeruli also lacked periglomerular cells and had no definitive glial envelope. These results suggest that glomerular formation is not dependent on dendrite contribution of second order neurons or glial support, but rather on a complementary population of receptor neurons.

    Notes: Abstract Electron microscopy has been employed to analyze the normal maturationai sequence that characterizes the postnatal development of synaptic circuits in the ventrobasal (VB) and reticular (Rt) thalamic nuclei of rats at different ages (from birth to the end of the third postnatal week). Throughout the first postnatal week, similar signs of immaturity are observed in both nuclei, mainly consisting in scarcity of cytoplasmic organelles, presence of wide extracellular spaces, and absence of myelinated fibers. Several synaptic terminals are however present from birth, thus indicating that some of the afferents have already reached and contacted their thalamic target during embryonic life. Most of the terminals are small and contain only a few round, clear vesicles, and therefore their cytological features do not allow the identification of their origin. In particular, in both nuclei, terminals with flat vesicles and symmetric specialization are only rarely observed, and in VB the ascending terminals are not distinguishable from terminals of other sources as they are in adults. During the second postnatal week, progressive maturationai changes in VB and Rt lead to neurons having well-developed cytoplasmic organelles and to an elaborate neuropil containing myelinated fibers and synaptic terminals that are morphologically heterogeneous and resemble the adult ones. The permanence of growth cone-like profiles and of numerous somatic and dendritic protrusions, often contacted by synaptic terminals, indicates that a certain degree of reorganization is still taking place in both nuclei. By the end of the third postnatal week the synaptic organization of VB and Rt is indistinguishable from that observed in adults. This ultrastructural study shows that the appearance of the neuropil of VB and Rt and the morphological complexity of the synaptic arrangements characteristic of the adult rat are not present in neonates, but are gradually acquired during the first three postnatal weeks, and that they result from progressive modifications in circuit organization involving both pre- and postsynaptic elements.

    Notes: Abstract A light and electron microscopic study of the brain of an 18-week fetus with a prenatal genetic diagnosis of Fukuyama-type congenital muscular dystrophy revealed a widespread mantle of abnormal neurogliomesenchymal tissue that covered a dysplastic cerebral cortex. In this area alone, the glia limitans that adjoined the abnormal mantle via one or two layers of basal lamina had frequent breaches, through which neuroglial elements extruded. In the most severely affected cortical region, which had only a rudimentary and fragmentary glia limitans, the majority of cortical neurons had migrated into the neurogliomesenchymal tissue. The massive overmigrated neurons still maintained a somewhat columnar arrangement, and the marked dysplasia abruptly shifted to a neurogliomesenchymal tissue-free normal cortical structure with an intact glia limitans, thus indicating essentially vertical overmigration of neurons without significant tangential migration of them. Together the above findings imply that breaches in the glia limitans may be the primary cause of the micropolygyria seen in this genetic disorder.

    Notes: Abstract We have identified two central neurocytomas which contained cells co-expressing glial fibrillary acidic protein and synaptophysin defined by double-label immunostaining. Dual-positive cells were mostly polygonal in shape and with a morphological appearence similar to that of reactive astrocytes. This distinct morphology could be used to distinguish cells expressing glial fibrillary acidic protein from cells with round and clear cytoplasm which did not express glial fibrillary acidic protein and which composed the majority of the tumor. Samples containing polygonal cells were selected for electron microscopy from toluidine blue-stained semithin sections. Ultrastructural findings were similar in both neurocytomas, with both being composed predominantly of round cells with clear cytoplasm corresponding to the clear cells identified by light microscopy. Dense-core vesicles and clear vesicles were frequently observed in the cell processes. Apart from these clear cells, polygonal cells with electron-dense cytoplasm were noted. Paralleling the results of double immunostaining, these polygonal cells contained both dense-core vesicles and intermediate, presumably glial filaments. Microtubules and lipofuscin granules were also observed. These results suggest that cells expressing glial fibrillary acidic protein in central neurocytoma include tumor cells with both neuronal and glial characteristics.

    Notes: Abstract The origin and differentiation of the oxyntic cell lineage during physiological cell renewal was investigated by light and electron microscopy in the abomasal mucosa of adult cattle. The morphologically heterogeneous oxyntic cell population exhibits various developmental subtypes depending on the position within the oxyntic unit. Pre-oxyntic cells of the isthmus and neck represent the immature precursors. Though heterogeneous with respect to the degree of canalicular and tubulovesicular membrane development, they all contain secretory granules resembling those of either isthmus cells, immature surface mucous cells, neck cells or young chief cells. A secretory granule-free stem cell is not present in the bovine. Downward to the gland base genesis of canalicular as well as tubulovesicular membranes is gradually completed; thus pre-oxyntic cells give rise to mature oxyntic cells. Older degenerative oxyntic cells, primarily located within the gland bottom, are characterized by progressive involution of canalicular and tubulovesicular membranes. Towards the pit, differentiation of pre-oxyntic cells is associated with atypical and incomplete development of canaliculi and tubulovesicles. In consequence, these superficial oxyntic cells have a reduced secretory capacity from a morphological point of view.

    Notes: Abstract Hemidesmosomes are junctional complexes involved in the attachment of epidermal basal keratinocytes to the basement membrane. To try to understand better the sequence of events in the morphogenesis of hemidesmosomes, we undertook an ultrastructural analysis of hemidesmosome formation in fetal and neonatal digit skin. Hemidesmosomes, defined as membrane-associated densities or plaques, were counted and scored for three morphological characteristics: (1) the presence of a sub-basal dense plate, (2) association with anchoring filaments within the lamina lucida and (3) contacts with intermediate filaments. No hemidesmosomes were evident at 7 weeks’ gestational age. Between 9 and 15 weeks the number of hemidesmosomes increased by about fourfold (from 20.6 ± 3.8 (SD) to 95.5 ± 8.4 per 40 μm of basal cell plasma membrane; P 〈 0.01). The association of hemidesmosomes with intermediate filaments and anchoring filaments also increased after 15 weeks ( P 〈 0.05). Early attachment plaques first appeared as triangular focal densities on the basal plasma membrane with the appearance of sub-basal dense plates, which later became both larger and more electron dense. By 15 weeks, an inner plaque could be distinguished from the outer plaque, which coincided with a closer association with intermediate filaments. Hemidesmosomes appeared fully developed by 15 weeks’ gestation. This study illustrates the structural relationship of hemidesmosomes to both intra- and extracellular filaments, suggesting close functional interactions. The complexity of the hemidesmosome plaque is also revealed early during development.

    Description / Table of Contents: Summary Confluent and reticulated papillomatosis (CRP) is a rare dermatosis of unknown aetiology. Recent electron microscopic studies suggest that CRP is a disorder of keratinisation. In our case we could not confirm the previously reported ultrastructural findings. CRP is generally resistant to therapy. We treated a 19-year-old patient with typical CRP with oral minocycline. Within a few weeks the eruption resolved completely. A mild relapse 7 months later responded promptly to a repeated course of minocycline. Twelve months after discontinuatin of therapy there is no evidence of recurrence. In CRP minocycline should be preferred to systemic retinoid therapy because of its minor side effects.
    Notes: Zusammenfassung Die Papillomatosis confluens et reticularis (PCR) ist eine seltene Erkrankung unklarer Ätiopathogenese. Neuere elektronenmikroskopische Untersuchungen stützen die Vermutung, daß eine Keratinisierungsstörung vorliegt. Anhand unseres Falles konnten wir diese Befunde allerdings nicht bestätigen. Die PCR ist ausgesprochen therapieresistent. Wir behandelten einen 19jährigen Patienten mit klinisch und histologisch typischer PCR systemisch mit Minocyclin. Innerhalb weniger Wochen heilten die Effloreszenzen vollständig ab. Ein 7 Monate später auftretendes leichtes Rezidiv konnte durch einen zweiten Behandlungszyklus schnell und erfolgreich behandelt werden. Der Patient ist jetzt bereits 12 Monate nach Absetzen der Therapie erscheinungsfrei.     Notes: Abstract Specimens of human cerebral neocortex were obtained during neurosurgical operations and studied by immunocytochemistry and electron microscopy, using antibodies to the glutamate receptor subunit GluR1 and gamma-aminobutyric acid (GABA). Many GluR1-positive pyramidal neurons and fewer GluR1-positive nonpyramidal neurons were present in the cortex. Non-pyramidal neurons were more heavily labelled for GluR1 than pyramidal neurons. Most GABAergic neurons were labelled for GluR1. The white matter was unstained, except for occasional labelled neurons. This pattern of GluR1 immunostaining is similar to that in rat cerebral cortex, but is different from that in the hippocampus and amygdala, where large numbers of pyramidal or projection neurons, but few non-pyramidal or GABAergic neurons, were labelled for GluR1.

    Notes: Abstract The distribution of the [3H]alpha-amino-3-hydroxy-5-methylisoxzalepropionic acid (AMPA) receptor subunit GluR4 was studied in frontal, parietal and temporal cerebral cortex, subcortical white matter and corpus callosum of neonatal, immature and mature rats. In 1-to 2-day-old rats, a few oligodendrocyte progenitors and amoeboid microglia in the supraventricular part of the corpus callosum were immunolabelled for GluR4. At 7 to 10 days, the number of amoeboid microglia and oligodendrocyte progenitors in white matter increased; many neurons in cortex, including pyramidal neurons, were also moderately labelled for GluR4. The pattern of GluR4 immunostaining in 14-day-old rats was different from that in 7-to 10-day-old rats, but similar to the adult, in that there was no immunoreactivity in microglia and oligodendrocyte progenitors in subcortical white matter. A proportion of non-pyramidal neurons in cortex were moderately labelled, while some pyramidal neurons were lightly labelled. A population of small glial cells with features of oligodendrocyte progenitors were densely labelled in cortex.

The two index cases presented with morning fatigue, had ketotic hypoglycaemia when fasting which rapidly disappeared after eating, and hepatic glycogen deficiency and absent or very low hepatic glycogen synthase activity. Metabolic profiles comprising glucose, lactate, alanine, and ketones in blood were typical for hepatic glycogen synthase deficiency. Symptoms were rapidly relieved and chemical signs corrected by introducing frequent protein-rich meals and night-time feedings of suspensions of uncooked corn (maize) starch. The discovery of oligosymptomatic and asymptomatic siblings suggests that there are more persons with undiagnosed hepatic glycogen synthase deficiency. Conclusion Liver glycogen synthase deficiency is likely to be more common than is believed today. It should be sought in children who, before the first meal of the day, present with drowsiness, lack of attention, pallor, uncoordinated eye movements, disorientation or convulsions and who have hypoglycaemia and acetone in urine.

    Notes: Abstract Phospholipid/cholesterol vesicles were solu-bilized by 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate (CHAPS). Above 30 mol% cholesterol (Ch) in the lipid vesicles several remarkable changes of the solubilization process were observed. (i) Two modes of solubilization: The effective detergent to lipid ratio Rc(M) for the formation of mixed micelles decreased from Rc(M) = 43 ± 3 at low lipid concentrations, [L]≤ 0.15 mm, to Rc(M) = 2.4 ± 0.3 above [L] = 0.5 mm (40 mol% Ch, T = 20 °C). (ii) At subsolubilizing CHAPS concentrations, filamentous and helical microstructures were formed, similar to those which were observed in native and model bile. (iii) The number of observed fibers was about two orders of magnitude higher in the presence of the negatively charged lipids phosphatidylglycerol (PG) and phosphatidic acid (PA) compared to the zwitterionic phosphatidylcholine (PC). Fiber formation began after 16–18 h using PG and PA compared to 3–4 days in the presence of PC. Screening of the charged lipids by NaCl effectively reduced the formation of fibers. Assuming binding of Na+ to the charged lipid aggregates, an intrinsic binding constant Kint = 0.6 M–1 was determined by applying the Gouy-Chapman theory. After the addition of CHAPS to PG/Ch vesicles, a fast initial solubilization of the vesicles (〈1 min) to mixed micelles (rh = 2.3 ± 0.2 nm) and small vesicles (rh = 23 ± 1 nm) was observed, followed by an intermediate period of 2 h, after which the formation of fibers occurred (〉15 h). The microstructures are visualized by darkfield and electron microscopy. The method of vesicle solubilization is compared to the dilution of concentrated micellar solutions, which is usually applied to model bile systems.

Histologically, these tumors appeared as cancerous out-grown lateral-root primordes and were thus comparable with stem nodules of the legume Sesbania rostrata. Azorhizobium caulinodans (a diazotroph known as a specific endophyte of Sesbania rostrata) was introduced and became established inside rice para-nodules and in root tissues around tumor bases. The infection with A. caulinodans followed a typical “crack-entry” invasion at places where para-nodule tumors had emerged through the root cortex and epidermis. The bacteria settled with high cell densities in intercellular spaces of the induced tumors and betwen root cortical cells. Infection of plant cells took place both in the epidermis and in cortical tissue. Intracellularly established A. caulinodans was found inside the cytoplasm, surrounded by membrane-like structures. N2 fixation by tumor-inhabiting Azorhizobium sp. was increased at low O2 tensions (1.5–3 kPa) compared with an untreated control. Only a little activity remained at O2 tensions of 5 kPa and above. The present results confirm that root-tumor induction offers a suitable method of establishing diazotrophs endophytically in the roots of gramineous crops.

    Notes: Abstract Rhizomelic chondrodysplasia punctata (RCDP) is clinically characterized by symmetrical shortening of the proximal limbs, contractures of joints, a characteristic dysmorphic face, and cataracts. In the classical form an impairment of several peroxisomal functions and enzymes (plasmalogen synthesis, phytanic acid oxidation, 3-oxoacyl-CoA thiolase) has been repeatedly shown. Recently a variant involving only the peroxisomal dihydroxyacetonephosphate acyltransferase (DHAP-AT) has been described. We present a patient with isolated DHAP-AT deficiency and all clinical, radiological, and pathological features of classical RCDP. For the first time, microscopy and immunocytochemistry of hepatocytes could be performed. Conclusion In contrast to studies on classical rhizomelic chondrodysplasia punctata which have shown enlarged peroxisomes in numbers varying from hepatocyte to hepatocyte, the peroxisomes in our patient seem to be normal in size, number and shape.

    Notes: Abstract The fine structure of the cell surface of seven enterotoxemic Escherichia coli (ETEEC) O139:K12 strains isolated from piglets with edema disease were examined electron microscopically using both the negative-staining method and the freeze-substitution fixation method. Densely packed, fine fibers were observed; they consisted of a capsule layer approximately 25 nm thick around the cell surfaces of strains 107/86, IW-2, ED-3, ED-43, and ED-61, all of which have a capacity to adhere strongly to HEp-2 cells. In contrast, no such structure was observed on the surface of strains RK-O139 or ED-1, both of which adhere only weakly to HEp-2 cells. These results suggest that the capsule structure might be associated with the ability to adhere to HEp-2 cells and, as a result, also potentially play some role in ETEEC infection.

    Notes: Abstract This review covers the recent progress in the elucidation of the structure of photosystem II (PSII). Because much of the structural information for this membrane protein complex has been revealed by electron microscopy (EM), the review will also consider the specific technical and interpretation problems that arise with EM where they are of particular relevance to the structural data. Most recent reviews of photosystem II structure have concentrated on molecular studies of the PSII genes and on the likely roles of the subunits that they encode or they were mainly concerned with the biophysical data and fast absorption spectroscopy largely relating to electron transfer in various purified PSII preparations. In this review, we will focus on the approaches to the three-dimensional architecture of the complex and the lipid bilayer in which it is located (the thylakoid membrane) with special emphasis placed upon electron microscopical studies of PSII-containing thylakoid membranes. There are a few reports of 3D crystals of PSII and of associated X-ray diffraction measurements and although little structural information has so far been obtained from such studies (because of the lack of 3D crystals of sufficient quality), the prospects for such studies are also assessed.

    Notes: Abstract We report a case of pancreatic acinar cell carcinoma which contained a large number of pleomorphic inclusions with fibrillary internal structures and mature zymogen granules. To clarify the significance of fibrillary inclusions in the differentiation of acinar cells of the pancreas, we further investigated fetal pancreases (gestational weeks 16, 17, 19, 20 and 28). We found two types of inclusions: type A, corresponding to fibrillary inclusion of neoplastic acinar cells, was observed only in a 19-week fetus; type B showed a homogeneous density similar to that of zymogen granules. Type B was observed in all the fetuses after the 17th gestational week. Although the type A inclusion might be generated throught a different mechanism than the type B inclusion, the appearance of a large number of fibrillary inclusions in neoplastic acinar cells may represent a transient form of zymogen granule.

    Notes: Abstract Alveolar adenoma of the lung is a poorly characterized, uncommon pulmonary lesion with proliferation of alveolar epithelium and septal mesenchyme. We describe the electron microscopy, immunohistochemistry and DNA flow cytometry in a case of alveolar adenoma in a 55-year-old woman. Alveolar adenoma appears to be a distinct benign neoplasm of the alveolar structures. Our findings further suggest that it is not a precursor of bronchioloalveolar carcinoma or other type II pneumocyte lesions of presumed malignant potential.

    Notes: Abstract. Desmin protein is an abundant constituent of the intermediate filaments in the electrocytes of the electric organ of the electric eel Electrophorus electricus. Polyclonal antibodies were raised against purified desmin from the electric organ and used for immunolabeling of the protein in reconstituted filaments. In thick sections of the main electric organ that has been stained with fluorescein-labeled desmin-specific antibodies, light microscope revealed a diffuse meshwork of desmin filaments dispersed in the cytoplasm of electrocytes. In the region under the membrane, the immunostaining was slightly more intense than elsewhere. The meshwork of intermediate filaments composed of desmin was examined by electron microscopy of the main electric organ. Immuno-gold labeling demonstrated a widespread meshwork of desmin filaments in the cytoplasm and in close association with the plasma membrane. These observations suggest that intermediate filaments play a role in the maintenance of the morphology of electrocytes and, as an intracellular meshwork spanning the width of the cell, they may contribute to the organization of the intracellular compartments.

    Notes: Abstract. The nitric oxide synthase-immunoreactivity and NADPH-diaphorase activity of intracardiac neurones in the rat and guinea-pig was studied at the ultrastructural level. While some nitric oxide synthase-containing intracardiac neurones were very heavily labelled, with electron-dense immunoprecipitate distributed throughout the neuronal cell bodies and their processes, most of the labelled neurones exhibited a lighter and more patchy distribution of nitric oxide synthase-immunoreactive material. Synapses made by nitric oxide synthase-negative nerve fibres with labelled intracardiac neurones were seen. Conversely, many nitric oxide synthase-containing nerve fibres that made synaptic contacts with unlabelled intracardiac neurones were also observed. Some small granule-containing cells were nitric oxide synthase-immunoreactive and were associated with unlabelled nerve terminals, while non-immunoreactive small granule-containing cells that were innervated by nitric oxide synthase-immunoreactive nerves were also seen. Small patches of osmiophilic electron-dense material were observed in the cytoplasm of NADPH-diaphorase-positive intracardiac neurones. This is the first description of the ultrastructural distribution of nitric oxide synthase-immunoreactivity and NADPH-diaphorase activity in a subpopulation of intracardiac neurones of rat and guinea-pig heart and provides further evidence in support of a role for nitric oxide in the local control of the heart by intrinsic neurones.

We studied the structure of ocellar photoreceptor cells of Drosophila melanogaster, particularly the subrhabdomeric cisternae which our previous studies have shown to be essential structures for turnover of photoreceptive membranes in compound eyes. Each ocellus contained elongated photoreceptor cells with rhabdomeres positioned distally. In the subrhabdomeric regions, endocytotic invaginations were frequently observed, suggesting active turnover of photoreceptive membranes. In the vicinity of the photoreceptive microvilli, membranous structures similar to the subrhabdomeric cisternae in compound eyes were observed. These membranous structures were immunopositive for the rdgB protein, a phosphatidylinositol transfer protein that is localized to the subrhabdomeric cisternae in compound eyes. The ocellar photoreceptor cells of the retinal degeneration mutants (rdgA,B) were also studied. In these mutants, retinal degeneration has been reported to start, in compound eyes, with the disappearance of the subrhabdomeric cisternae. We found that the ocellar subrhabdomeric cisternae also disappear during the initial stage of retinal degeneration. From these observations, we conclude that the mechanism of photoreceptive membrane turnover in ocellar photoreceptor cells involves the rdgB and probably the rdgA proteins which are associated with subrhabdomeric cisternae, as is the case for photoreceptive membrane turnover in compound eyes.

More than 50 chordotonal sensilla, or scolopidia, embedded entirely in the integument were found in each side of the genital chamber wall in the female cricket, Teleogryllus commodus. Their cell bodies lie among the epidermal cells, and the tips of their dendrites terminate in the cuticle. About half of them contain two sensory cells (two-cell scolopidium), the others only one (one-cell scolopidium). The sensory cell in the one-cell scolopidium is the type-1 cell. In the two-cell scolopidium one is type-1 and the other type-2. Regardless of the number of sensory cells, they are all amphinematic. In the two-cell scolopidium only the type-2 dendrite, rich in microtubules, penetrates into the cuticle, bifurcates and terminates in the tube enclosed by an attachment cell; the type-1 never extends into the cuticle. On the other hand, the type-1 cell in the one-cell scolopidium projects its apex into the cuticle. The unique topography and structure of these scolopidia lead to the following hypothesis about the phylogenetic relationship between the scolopidia and other kinds of sensilla: the type-1 scolopidial sensory cell buried in the integument may be the original model, which through the loss of the long regular axoneme has given rise to type-2 cells. Modification of the apical region, the tubular body or ramification, may have lead to the cuticular sensilla corresponding to the development of the cuticular apparatus, and the scolopidia may have been withdrawn into the body cavity to form ordinary chordotonal organs.

    Notes: Abstract In order to study morphological effects on the nasal and sinus mucosa, New Zealand White rabbits underwent either unselective, regional sectioning of sensory and parasympathetic nerve branches or topical treatment of the mucosa with capsaicin. Ten days after treatment, mucosal specimens were analyzed by light and electron microscopy. Immunohistochemistry was used to evaluate neuropeptides present, in particular substance P, calcitonin gene-related peptide, vasoactive intestinal peptide and neuropeptide Y. In surgically denervated rabbits, mucosal glands were found to be enlarged and contained an increased number of zymogen granules having a bipartite substructure. Topical capsaicin application caused localized epithelial changes in the sinus mucosa and maxilloturbinal region of the nose, including clotting of cilia and an increased number of goblet cells. Reduced amounts of all neuropeptides investigated were found in the surgically denervated animals, while topical capsaicin treatment had only marginal effects on the mucosal neuropeptide content. The morphological changes observed after surgical denervation suggest an imbalance between neural stimulation and secretory capacity of the mucosal glands. These findings could explain the difference in clinical effect noted between sectioning of the vidian nerve and topical treatment with capsaicin in patients with perennial rhinitis.

    Notes: Abstract Numerous, small spherical inclusions with laminated horn-like material were observed in an epidermal cyst from the left earlobe of a 52-year-old Japanese man. They were examined by light and electron microscopy. Positive reactions obtained by staining with both oil red-O and anti-pankeratin antibody led to the conclusion that the inclusions consisted of lipid-rich keratin spherules. These were thought to have been formed in the hydrophobic regions of keratin proteins and non-polar lipids in the hydrophilic milieu of the cyst. Electron microscopy revealed cholesterol crystals within the spherules, suggesting that cholesterol had been slowly isolated from the lipoproteins and the membrane components of the cornified cells included in the lipid-rich keratin spherules, and had gradually become concentrated, finally appearing in the spherules as cholesterol crystals. With further study, a more detailed understanding of this process may throw some light on the development of gallstones and atheroembolism.

    Notes: Abstract To our knowledge, this is the first report of electron microscopic findings in the facial nerve sheath in Melkersson-Rosenthal syndrome. We performed decompression of the right facial nerve on a patient with Melkersson-Rosenthal syndrome. A part from the facial nerve sheath was obtained and observed with a transmission electron microscope. There were some fibers 180 to 200 nm in diameter which had obscure cross bandings, and also accumulations of these degenerated fibers among normal collagen fibers.

    Notes: Abstract A rare placental site trophoblastic tumor (PSTT) in a 39-year-old female was studied. This tumor, protruding into the uterine cavity, was histologically similar to tumors in previously reported cases of PSTT. Ultrastructurally, the characteristic finding was the presence of perinuclear filaments. Also, the tumor cells were strongly positive for hPL by immunohistochemical method. These findings suggest that this was a tumor caused by neoplastic proliferation of the extravillous intermediate trophoblast.

    Notes: Abstract We examined the incidence and morphological features of long-spacing collagen (LSC) in the intramuscular nerves of biopsied muscle tissues from patients with various neuromuscular diseases. LSC was found in 12 of 55 cases (21.8%). The muscles in all cases with LSC showed typical neurogenic changes at the light microscopic level. At the electron microscopic level, LSC was seen either in the vicinity of the basement membranes of the perineurium or Schwann cells, or running among the collagen fibrils. A close relationship between the form (compact or solitary type) and the periodicity of collagen was not confirmed.

    Notes: When testing theoretical models that quantitatively describe the sieving of macromolecules during gel electrophoresis, investigators have been limited by absence of control of the heterogeneity of the size of pores in the gel. In a recent study performed by electron microscopy of thin sections (G. A. Griess et al., J. Struct. Biol. 1993, 111, 39-47), pore size heterogeneity has been increased for agarose gels by a combination of both derivatization and molecular weight reduction of the polysaccharide chains of agarose. In the present study, pore size heterogeneity is increased by a mechanism that appears to have an origin different from the origin of this previously observed increase in heterogeneity: Pore size heterogeneity is increased by addition of a polyethylene glycol (PEG) of high molecular weight (18 500) to molten agarose before gelation. In contrast, the use of a lower molecular weight PEG (either 4 000 or 7 500) causes the formation of micron-sized precipitates within a gelled network of agarose fibers. Thus far, the PEG-induced heterogeneity of pore size occurs primarily in 100-1000 μm scale zones separated from each other by interzone regions of decreased agarose fiber density. More uniform gels are needed for the study of sieving.

    Notes: Summary The epithelium of human chorion laeve from pregnancies complicated by diabetes mellitus was examined under the light and electron microscopy. In comparison with normal chorion laeve, the epithelium of chorion laeve in diabetes mellitus shows the following morphological changes: 1. The trophoblast cells display more microvilli and cell processes on their surface. 2. Trophoblast cells with different cytoplasmic characteristics can be seen. Some display few organelles and large glycogen depositions; other are rich in organelles. Furthermore, some cells with electron dense filaments and few cell organelles can be found. 3. Necrotic cells are frequently present in the outer epithelial layers. 4. The intercellular spaces in the epithelial layers near the basement membrane are narrower. Since vasculopathy of the decidual arteries with disturbance of blood circulation is often found in pathological pregnancies, we assume that the morphological changes found are due to deficiency in nutrient and oxygen supply.

    Notes: Summary Stereological techniques on electron microscopy micrographs were used to evaluate the morphological changes of cultured isletβ cells that had been exposed to alloxan or 6-hydroxydopamine. Trypan Blue exclusion by cells cultured for 3 days indicated that the cells were 100% viable. Electron microscopy revealed that nearly all of the surviving cultured cells wereβ cells. Exposure to 5 mmol/l alloxan or 1–5 mmol/l 6-hydroxydopamine for 10 or 30 min caused a general swelling of the cultured cells with a concomitant swelling of mitochondria and nuclei. The size of the secretory granules was not affected by the drugs. Only 3–10% of the cells excluded Trypan Blue after exposure to 5 mmol/l alloxan or 6-hydroxydopamine. The data conform with the hypothesis that a primary action of alloxan and 6-hydroxydopamine is at the plasma membrane level ofβ cells.

    Notes: Summary An example of elephantiasis neuromatosa, an unusual variant of neurofibromatosis, coexistent with lipomatosis is presented. This dual cell population produced a diffuse swelling of the gluteal sulcus of a young female. The neurogenous origin of the spindle cell component was substantiated by immunoreactivity for S-100 protein as well as by the fine structural observation of a regular basement membrane coating and a profusion of Luse bodies. As far as can be ascertained, this is the first reported immunohistochemical and ultrastructural study of this particular variant of peripheral nerve sheath lesion.

    Notes: Summary A calcifying epithelial odontogenic tumor, simultaneously located in the two jaws (maxilla and mandible) was examined by histochemical and electron microscopic methods. Squamous tumor cells without secretory polarity were different from those of common ameloblastoma. High activities of alkaline phosphatase and ATPases were demonstrated by light and electron microscopy on the cytoplasmic membrane, findings similar to those in the stratum intermedium cells of the normal dental germ from which these tumor cells seem to arise. The tumor cells, like preameloblasts of the dental germ, also produce a granulo-filamentous material in intracytoplasmic vesicles and discharge it into the stroma. This “pseudo-amyloid” substance represents an abnormal protein of the enamel matrix and calcification, mainly occurring in that substance, might be an attempt at mineralization.

    Notes: Summary Primary granular cell tumours of the breast in 35 2and 55 year old women were studied by light microscopy, transmission electron microscopy and immunohistochemistry. Light and electron microscopy revealed a neural origin of the tumours and this was further substantiated by immunohistochemical studies, with positive S-100 protein reaction and negative reactions for surface heavy and light chains, CEA, alfa-1-antitrypsin, muramidase and GFA-protein. Granular cell tumour of the mammary gland is a very rare tumour. Clinically it sometimes simulates carcinoma because of its fibrous consistency, fixation to pectoral fascia and skin retraction. The diagnosis of granular cell tumour should be included in the differential diagnosis of carcinoma of the breast. The granular cell tumour is derived from neuro-ectodermal tissue. Whether it represents a neurogenic cell-confined metabolic disturbance with lysosomal activation, or a true neoplasm remains to be elucidated.

    Notes: Summary Material in urinary calculi is prone to irradiation damage during electron microscopy and this suggests the need for care in the interpretation of data. It is shown, however, that minimum-dose transmission microscopy is feasible for single-crystal electron diffraction work, and that although internal damage in severe, morphological artefacts are unlikely in the SEM unless the incident electron flux is greater than 10-13 A nm-2. During EDX micro-analysis, the detection of light elements is impaired by irradiation effects unless a minimum-dose procedure is used. For the preparation of SEM samples, artefacts can be created by cleaving air-dried material-and it is therefore important to consider more lengthy preparation methods such as cryogenics.

    Notes: Summary We used concanavalin A (con A)-peroxidase-iron dextran-diaminobenzidine (DAB) technique for the electron microscopic detection of con A binding sites on cell membranes. Normal bladder mucosa showed a sparse distribution of con A binding sites with both transmission (TEM) and scanning (SEM) electron microscopy, but bladder tumors showed a higher concentration in the distribution of con A binding sites in proportion to the histopathological grade of transitional cell carcinoma. Quantitative estimation of the con A binding sites was attempted using scanning X-ray pulse analysis of iron elements contained in the reaction complexes. Con A binding sites were quantitatively the smallest in normal mucosa, increasing proportionate to the grade of the bladder tumor. Some specimens were compared by the ferritin-labelled method and the pattern of ferritin conjugates distribution was similar to that seen with the con A-peroxidase-iron dextran method.

    Notes: Summary A study of urinary stones obtained from patients after surgery in the Medical College Hospital, Trivandrum, under the scanning electron microscope showed the presence of calcium oxalate and calcium biphosphate crystals as the main constituents. However, the pattern of the different phases of crystal growth was not uniform. Within the crystal lattice, fibrous structures, possibly of protein matrix, were invariably observed. Electron microscopy may be usefully adapted as a particularly suitable method for ultramicroscopic investigation of the fine structure of urinary stones including single crystal surface structure, section of urinary calculi and for possible presence of hitherto unknown components within the calculus.

    Notes: Summary In the canine circumanal gland, the morphological alterations of peroxisomes during differentiation and maturation of the glandular cells were studied by electron microscopy, cytochemistry and freeze-etch technique. Each of the following three cell types has its own characteristic peroxisomal population: 1) The basal cell contains only a few small peroxisomes, which appear as spherical and tubular profiles showing strong DAB reaction. In the differentiating basal cells, these are joined by a few dilated, hemispherical organelles with intensely stained small marginal plates. 2) In the intermediate cell, additional to spherical and tubular peroxisomes, numerous clongated organelles with distinct marginal plates are observed, displaying weak catalase activity. 3) In the mature cell, dumbbell-shaped peroxisomes with enlarged marginal plates predominate. Serial section analysis and freeze-etching studies reveal that these dilated particles are of erythrocyte-like shape. They exhibit very weak catalase activity or do not contain any visible DAB reaction product. In their flattened, thin central portions, the memoranes enclose the marginal plates and form straight cisternae, which are closely associated with adjacent fenestrated cisternae of ER on both sides, referred to as paramarginal cisternae. Dumbbell-shaped peroxisomes with their corresponding paramarginal cisternae form large peroxisome-ER-complexes. Furthermore, three to five dumbbell-shaped particles are often stacked in parallel. Only at their flat poles are the organelles in close contact with paramarginal cisternae. The observation of continuities, in particular between erythrocyte-like organelles and tubular peroxisomes in mature glandular cells, indicates the existence of a peroxisomal compartment composed of two segments in the mature stage. In freeze-etch replicas of mature glandular cells, only the dilated segments of the peroxisomal compartment can be easily recognized because of their unusual size and erythrocyte-like shape. Additionally, on the E-face of their central portion, a straight, square or rectangular area with a distinct crystalline pattern is seen, which corresponds to the marginal plate. These findings indicate that the circumanal gland of the dog offers unique possibility to analyze the biological properties of a well-defined peroxisomal compartment.

    Notes: Summary The ultrastructure of the wall of the Dandy-Walker cyst has been described rarely. A boy aged 2 years was confirmed clinically, neuroradiologically, and operatively as having a Dandy-Walker cyst in the posterior fossa. The cyst wall obtained during surgery consisted of an outer arachnoid cell layer, intermediate interwoven neuroglial strands, and an inner layer of cells which lacked the characteristic appearance of ependyma. An unusual finding was a small, buried island of ependymal cells in the intermediate layer of the neuroglial tissue. Ultrastructural study of the cyst wall provides a better understanding of the pathogenesis of the Dandy-Walker syndrome.

    Notes: Summary In spite of several cases reported in the literature, the exact pathogenetic mechanism of neuropathic changes in porphyric neuropathy remains uncertain. Various authors have ascribed the neuropathologic findings to either a dying-back axonal degeneration or segmental demyelination. In recent years, the hypothesis of an axonal and myelinic disorder has received support by the demonstration of a combined and simultaneous involvement of both these structures. Such different opinions are also a consequence of the reduced number of detailed bioptic observations in the different forms of acute porphyria not only during acute phases but also between attacks. In this paper we report the results of light- and electronmicroscopic examination of two sural nerve biopsies from subjects with hereditary coproporphyria. The first was performed 6 months after an acute attack, the second specimen was obtained from a patient without acute attacks, who had clinical and electrophysiologic signs of a chronic progressive neuropathy. In both cases a dying-back axonal degeneration is considered the primary change. The pathogenetic mechanism of peripheral nerve lesions in porphyric neuropathy will be discussed finally.

    Notes: Summary Anterior tibial muscle biopsies of the hemiplegic side of 16 patients with a cerebrovascular accident in the middle cerebral artery region were analyzed qualitatively and quantitatively by enzyme histochemistry and electron microscopy. Patients grouped according to the time lapsed as from the occurrence of the accident (1–17 months) demonstrated a progressive decrease in the fiber diameter and changes in fiber type distribution with predominant type II atrophy and type I predominance. Nuclear internalization, myopathic alterations, and perifascicular fatty infiltrations were observed constantly. In the affected fibers the ultrastructural findings were myofibrillar alterations with the formation of rods and cytoplasmic bodies. There was accumulation of lipofuscin, glycogen, and lipid droplets. Microvascular changes were observed frequently. Biopsies from the asymptomatic legs were either normal or showed age-related muscle alterations. Correlation was noted between the clinical and functional status of the patients and the morphological aspects seen in muscle biopsies.

    Notes: Summary Adult Wistar rats were exposed to carbon disulphide (CS2) vapour at a concentration of 2.4 mg/l of air for 5 days a week (6h a day), and the ultrastructure of peripheral nerves, neuromuscular junctions and muscles was investigated after 6 months of exposure to CS2. Numerous giant axons, i.e. paranodal or internodal swellings, were seen in the peripheral nerves. At the swollen paranodes, the myelin sheath was thinned, in other regions large intramyelinic vacuoles indicative of more dramatic demyelination were observed at axonal enlargements. Axonal enlargements consisted essentially of whorls of tightly packed neurofilaments. A number of nerve fibres underwent complete degeneration, but at the same time there was evidence of nerve regeneration. Nerve terminals were affected in a similar way following CS2 exposure. At neuromuscular junctions, filamentous swellings of nerve terminals preceded their degeneration and eventual denudation of synaptic gutters. As a rule, the postsynaptic part of neuromuscular junctions remained unimpaired by CS2 treatment. Muscles were affected by both atrophy and degeneration. Clusters of dense and lamellar bodies and numerous autophagosomes indicative of direct myotoxic effect of CS2 were frequently encountered in the investigated muscles. Some muscle fibres apparently underwent necrosis judging from the occurrence of myotubes characteristic of muscle degeneration and regeneration. The pathomorphology of CS2 neuropathy resembles that of other toxic neuropathies which presumably have a common origin in impaired energy metabolism.

    Notes: Summary The ultrastructure of peroxisomes in partially differentiated cells of the mouse preputial gland was investigated using serial thin sections and three-dimensional reconstruction as well as the alkaline diaminobenzidine technique for visualization of the peroxidatic activity of catalase. An analysis of serial sections indicates that the different types of intensely stained peroxisomal profiles, classified according to their shape, represent random planes through highly complex peroxisomes. These organelles exceed 4 μm in length and exhibit a focal heterogeneity with respect to their size, shape and enzyme distribution. The graphical three-dimensional reconstruction demonstrates that the most intricate peroxisomes are characterized by tortuous, elongate, and branched tubular segments of varying diameter equipped with enlarged terminal hollow-spherical structures which engulf areas of cytoplasm. A close spatial relationship is established between adjacent peroxisomes and peroxisomes and mitochondria, the latter two of which synchronously develop into highly complex structures. A close association is also observed between peroxisomes and the endoplasmic reticulum, whereby membrane continuities between the two compartments cannot be demonstrated. These observations are inconsistent with traditional concepts concerning peroxisomal shape and size, the number per cell, as well as their biogenesis from the endoplasmic reticulum. The functional significance of individual highly complex peroxisomes and their assemblage forming an extensive netlike membraneous system throughout the cell is discussed with respect to intracellular energy transport and trans-membrane electron exchange.

    Notes: Summary Ultrastructural studies were undertaken to investigate the temporal relationship between loss of the zona pellucida around the blastocyst and the appearance of decidual changes in the endometrial stroma during normal implantation in rats. Blastocyst-free and blastocyst-containing sites of pregnant uterine horns were studied and compared with control sites from contralateral salpingectomized horns and horns of pseudopregnant animals from 24.00 h on Day 4 and onwards. There were no membrane contacts between the blastocyst and the uterine epithelium at 10.00 h on Day 5 and earlier because of an intervening zona pellucida. From 14.00 h onwards, however, such contacts were present and at 18.00 h, the zona pellucida had disappeared and the blastocyst had attached onto the uterine epithelium. The stromal cells of pregnant and control horns were indistinguishable from each other at 24.00 h on Day 4, but from 06.00 h on Day 5 onwards specific changes were noted in the stromal cell nucleoli of the pregnant horns. The results therefore suggest that the first morphological sign of decidualization occurs about 12 h before the Pontamine Blue reaction and is initiated by the blastocyst early on Day 5 while it is still encased by the zona pellucida.

    Notes: Summary During the embryonic development of the intestine, programmed deletion of epithelial cells plays a vital role in the formation of secondary lumina and of villi. Electron microscopy shows that this deletion is effected by apoptosis, a distinctive mode of cellular death recently recognised as being implicated in normal tissue involution in the adult, as well as during intrauterine development. The process involves cellular condensation and budding to form membrane-bounded apoptotic bodies containing well-preserved organelles. These are then either engulfed by neighbouring principal cells or sloughed off into the intestinal lumen. Engulfed apoptotic bodies that show various degrees of lysosomal degradation have previously been reforred to as “meconium corpuscles”.

    Notes: Summary Considering the possibility of a paracellular route for edema resolution we studied the microvasculature of the subependymal and subcortical white matter in hydrocephalic rats. Normal adult rats were used as controls. After injection of kaolin suspension into the cisterna magna, the animals were killed at intervals of 1, 2, 4, and 8 weeks. In hydrocephalic rats at 1 week after kaolin injection, widening of the interendothelical cleft between the tight junction (dehiscence) was seen in 27 of 76 (35%) vessels. At 2 weeks after kaolin injection, the number of the dehiscences had increased (39/7:56%) and some were enlarged, forming interendothelial blisters. At 4 weeks in hydrocephalic rats, both dehiscences and blisters were still prominent (45/73∶63%) and at 8 weeks the dehiscences were still prominent, but the number of the blisters had decreased (25/81∶31%). The blisters and dehiscences were most pronounced in the corpus callosum and occipital regions. Following i.v. injection of horseradish peroxidase, the interendothelial dehiscences and blisters were completely devoid of the marker substance. These findings indicate that in obstructive hydrocephalus the tight junctions may constitute part of a paracellular pathway for the resorption of interstitial edema fluid.

    Notes: Summary Photochemotherapy (PUVA)-induced freckles were found in 25 patients (41%) who had received more than 1,000 J/cm2 of PUVA. The patients had been treated with PUVA for more than 2 years, with more than 150 exposures before PUVA lentigines appeared on the thighs, the upper arm, the mid-lower arm, the waist, and the buttocks. The histopathology of these freckles was analyzed by light and electron microscopy. Light microscopy showed an increased amount of pigment and melanophages and increased numbers and size of melanocytes. The keratinocytes often displayed atypical features such as enlarged nuclei, giant size, or fibrillar degeneration. Homogenization of the papillary dermis was observed in 11 patients. The activation of melanocytes was confirmed electron microscopically, and pathological features such as large amounts of lipid droplets and lysosome-melanosome complexes within the melanocytes were seen. The Langerhans cells were mostly normal, whereas the keratinocytes showed cytolytic changes, fibrillar degeneration, and vacuolization. A close follow-up of patients with prolonged PUVA treatment is recommended.

    Notes: Summary To understand contact hypersensitivity, it is important to know the kinetics of Langerhans cells (LC) and related cells in the lymph node (LN), as well as in the skin. For this purpose, we tried experimentally to induce increased numbers of LCs, Birbeck granule-like structure (BgS)-containing cells, and interdigitating reticulum cells (IDC) in DNCB-sensitive mice and studied them by means of electron microscopy with the following results: (1) cytologically, LC, BgS-containing cells and IDC were closely related; (2) BgS seemed to arise from rough-surfaced endoplasmic reticulum (r-ER), and BgS-containing cells were midway in nature between LC and IDC from the morphological view point. From these findings, it appears that IDC, BgS-containing cells, and LCs were simultaneously involved in the contact hypersensitivity reactions of LNs.

    Notes: Summary Ten autosomal recessive/sporadic cases of hereditary motor and sensory neuropathy type I (HMSN I), nine of which originated from the northern part of Sweden, were included in the study. Parents were free from neurologic symptoms. Motor and sensory conduction velocity was normal when recorded, i.e., in 19 and 17 parents, respectively. Sural nerve biopsies from the ten cases revealed a varying degree of onion bulb formation. In eight of the cases the onion bulbs consisted of abundant basement membranes, whereas the Schwann cells were few and sometimes lacking. There were in some cases considerable differences between separate fascicles as to the loss of myelinated nerve fibers. In the six biopsies in which teasing was performed signs of present and previous demyelination were noticed. Numerous internodal segments were abnormally thin with reference to their length. In many such segments there were marked local thickenings of the nerve fiber. In cross sections the probable counterparts to these thickenings were nerve fibers with unduly thick myelin sheaths and complex folding of the myelin. Ultrastructural axonal changes were seen in the majority of the cases. The pathogenetic and diagnostic implications of the present findings are discussed.

    Notes: Summary A significant reduction in the myelinated nerve fiber population was observed during quantitative electron-microscopic examination of peripheral nerves in chronic alloxan diabetic rats. Dystrophic axonal abnormalities and regenerating fibers were more numerous in diabetics than age-matched controls. Schwann cells showed reactive changes including prominent pi granules of Reich and intracytoplasmic filament accumulation. Enumeration of these alterations, however, revealed no singificant difference from controls. Endoneurial macrophages in diabetic rats were also filled with lamellar intracytoplasmic inclusions characteristic of a chronic neuropathy. Quantitation of pathologic lesions in teased nerve fibers confirmed the preponderance of axonal over demyelinative disease and showed demyelination to be segmental. Microangiopathy was noted throughout the vasa nervorum of diabetic rats, and quantitative electron microscopy showed endothelial proliferation with doubling of the number of endothelial cells and proportional capillary mural thickening. Swollen, reactive endothelial cells appeared to effece the vascular lumen and may impair capillary perfusion. These microcirculatory changes, in the presence of biochemical and rheologic disturbances may contribute to tissue hypoxia and underly the loss of axons in experimental diabetic neuropathy.

    Notes: Abstract Cyanobacterin is a secondary metabolite produced by the cyanobacterium, Scytonema hofmanni. Highly purified cyanobacterin was found to inhibit the growth of many cyanobacteria at a minimum effective dose of 2 μg/ml (4.6 μM). The antibiotic had no effect on eubacteria including the photosynthetic Rhodospirillum rubrum. The site of action of cyanobacterin was further investigated in the unicellular cyanobacterium, Synechococcus sp. Electron micrographs of antibiotic-treated Synechococcus cells indicated that cyanobacterin affects thylakoid membrane structure. The antibiotic also inhibited light-dependent oxygen evolution in Synechococcus cells and in spheroplasts. These data support our conclusion that cyanobacterin specifically inhibits photosynthetic electron transport. This activity is similar to herbicides such as 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU). The anhydro analog of cyanobacterin had no biological activity.

    Notes: Abstract Electron microscopy of negatively stained isolated restriction enzyme EcoRI revealed particle projections with triangular or square outlines, indicating that the enzyme, in its tetrameric state, is tetrahedron-like. The two dimers making up the tetramer appear to be arranged in two planes orthogonal to each other. Complexes formed by EcoRI with the plasmids pBR322 or pGW10 were investigated by electron microscopic spreading techniques. In the presence of Mg2+, EcoRI was bound to the DNA molecules to form pearl necklace-like aggregates. The number of bound EcoRI particles was much higher as the sum of EcoRI-and 5′..AATT..3′ sites (with exceptions, the 5′..AATT..3′ sites may function as one type of EcoRI* sites) along the DNAs, indicating unspecific binding. In the absence of Mg2+, EcoRI was bound to the DNA only at the recognition site for EcoRI and the sites where the tetranucleotide sequence 5′..AATT..3′ was present. A direct correlation of the local concentrations of the bases A and T within the flanking sequences of the binding sites with the frequency of EcoRI to the DNA was observed. Dimers and tetramers of the enzyme was found to bind to the DNA. Tetramers occasionally exhibited two binding sites for DNA as indicated by the observation of DNA loops originating at the sites of bound tetrameric EcoRI particles.

    Notes: Summary A patient with prolymphocytic leukemia is described. The peripheral blood and bone-marrow cells contained nuclear pockets, bridges, and appendices, as well as cytoplasmic inclusions that were not membrane bound or connected with the endoplasmic reticulum. X-ray microanalysis of the cells showed them to contain large amounts of phosphorus, sulfur, chlorine, and calcium, as well as a smaller amount of sodium and magnesium in comparison with control lymphocytes. When compared with lymphocytes of a patient with chronic lymphocytic leukemia (CLL), the patient's cells showed higher amounts of magnesium, sulfur, and chlorine, while the sodium content was decreased. The usefulness of electron microscopy and X-ray microanalysis in the diagnosis of this type of leukemia is discussed.

    Notes: Summary A study of ellipsoids (EL) in the human spleen was done on 25 surgically resected specimens in order to assess the number of EL, their structure, the nature of the cuff cells and the problem of lymphocyte traffic through their wall. The results show that an average spleen of 200 g contains 1.6 × 107 EL. They are localized in a constant segment of the vascular tree, just at the transition of the penicillary arterioles into capillaries and they consist of a capillary (not an arteriole) surrounded by a cuff of cells. These cells are neither endothelial nor smooth muscle cells, display lysosomal activity and are considered as fixed specialized pericapillary cells capable of cytoplasmic expansion. Since their number is inconstant in different individuals and in different species they are considered to be dynamic histological structures. Because of the constant presence of lymphocytes in their walls we consider the EL to be a transit system for lymphocytes and the splenic equivalent of the high endothelial venules in the lymph nodes.

    Notes: Summary Collagen types I and III were examined immunohistochemically in 32 cases of gastric carcinoma classified as poorly differentiated adenocarcinoma with scirrhous stroma, well differentiated adenocarcinoma with intermediate stroma, or poorly differentiated adenocarcinoma with medullary stroma. In the stroma of scirrhous carcinoma, types I and III collagens were distributed abundantly in fibrillar or granular patterns with little difference in the intensity of staining. In well differentiated adenocarcinoma, type I collagen was diffusely distributed in the stroma with type III collagen distributed sparsely. In poorly differentiated adenocarcinoma with medullary stroma, the two types of collagen were only found around capillaries, constituting the tumor interstitium. Electron microscopic examination of scirrhous carcinoma showed tumor cells partially covered with fibroblasts, and discontinuous basal lamina, collagen fibers and microfibrils present between tumor cells and fibroblasts. In well differentiated carcinoma, tumor cells were surrounded by fibroblasts, and well developed basal lamina was observed beneath the tumor cells. In poorly differentiated carcinoma with medullary stroma, the stroma consisted of capillaries and very few fibroblasts with discontinuous basal lamina occasionally being present between tumor cells and fibroblasts.

    Notes: Summary The structure of guppy (Poecilia reticulata) spermatogonia and spermatocytes has been studied using electron microscopy. The spermatogonia, situated at the apex of the seminiferous tubule, are almost all surrounded by a network of Sertoli cells; they have very diffuse chromatin and one or two large nucleoli. The cytoplasm contains relatively few organelles, although annulate lamellae are found. The mitochondria have few cristae and are concentrated at one pole of the cell; they are sometimes found with intermitochondrial cement. These spermatogonia are separated from each other, having no intercellular bridges or inclusion in Sertoli cells, and are relatively undifferentiated; they correspond to stem cells. The spermatogonia beneath the apex are organized into cysts. First-generation spermatogonia are more dense and heterogeneous, their nuclei becoming smaller and their chromatin becoming denser during successive generations. In spermatocytes, the synaptinemal complex exists as a modified form until metaphase. The concentration of organelles in the cytoplasm increases and the organelles become more diversified as spermatogenesis progresses. Many cytoplasmic bridges are observed (several per cell), indicating that the cells remain in contact after several divisions. These changes in germ cell structure have been related to some of the characteristic features of spermatogenesis in guppy, e.g. the large number of spermatogonial generations and the complexity of spermiogenesis.
