    Notizen: Summary Ultrastructural studies of the middle ear mucosa appear to be of significant value in better understanding the pathology of otitis media with effusion (OME). Our present study was undertaken in order to take advantage of the use of electron microscopy in investigating all areas of the middle ear mucosa. Tissues studied were obtained from the fresh postmortem temporal bones of three patients with OME and terminal head and neck malignancies. In the mucoid type of effusion (cases 1 and 2), goblet cells were seen to proliferate and secretory activity was greatly enhanced. In contrast, there was no evidence of secretory cell proliferation in the serous type of effusion. It was noteworthy that accumulated fluid was not homogeneous in the same ear, as exemplified by case 1, in which both mucoid and serous effusions were present. This occurrence was possibly the result of topographic diversity involving the secretory activity of the middle ear.

    Notizen: Summary A rat animal model was used to study the ultrastructure of submucosal calcifications induced in the middle ear following inoculation with Streptococcus pyogenes and high doses of parenteral vitamin D3. The morphological changes present in affected animals resembled the classical picture of tympanosclerosis. While calcification occurred about bacterial remnants and myelin structures, the most important calcification centers were lysosomal and non-lysosomal matrix vesicles in the extracellular spaces. These formed band-like calcifications close to the basal membrane without affecting the epithelial layer. This animal model offers the possibility of studying the effect of various therapeutic regimens in the treatment of the dynamic tympanosclerotic process.

    Notizen: Summary Mitochondrial (mt) DNAs from normal (N) and male sterile (S) cytoplasms of sugar been have been isolated and investigated by electron microscopy. The results showed that mtDNA was composed of a heterogeneous population of circular molecules. Their contour lengths varied from 0.28 to 51 μm, but unlike in the case of maize, a large difference was not observed in the distribution of molecular classes greater than 1.0 μm between N and S cytoplasms of sugar beet. On the other hand, N and S cytoplasms were shown to contain their own characteristic combinations of small circular mtDNA species with lengths between 0.28 μm and 0.6 μm. Mitochondrial DNAs from various sources of male-sterile cytoplasms were analyzed by agarose gel electrophoresis to determine the extent of cytoplasmic variation. Additional low molecular weight DNA bands appeared in all male-sterile lines examined, and as a result, three distinctive banding patterns were recognized. These data are in general agreement with those based upon restriction endonuclease digestion of mt and chloroplast DNAs and the genetic analysis of fertility restoration in test crosses.

    Notizen: Summary Immunohistological techniques demonstrate the alpha-subunit of glycoprotein hormones in the majority of endocrine-inactive, undifferentiated pituitary adenomas and pituitary oncocytomas. In about one-fifth of endocrine-active adenomas, the alpha-subunit is produced in combination with either adrenocorticotropic hormone or prolactin, and it is found in combination with growth hormone in about half of those adenomas causing acromegaly. Pure alpha-subunit-producing, endocrine-inactive adenomas characteristically have small secretory granules that are destroyed by direct osmium fixation, but are well preserved after prefixation with glutaraldehyde. As only a few atypical prolactinomas show similar secretory granules, and as they display a positive reaction for the alpha-subunit only exceptionally, this ultrastructural feature can serve as a guide to differentiate such adenomas.

    Notizen: Summary This paper deals with the paramyxovirus-like intranuclear inclusions observed in giant cells tumours of bone (GCTB). Twenty-one (49%) of 43 cases of GCTB (1977–1985), either fresh and/ or cultured, show these ultrastructural inclusions. Fifty samples of various bone lesions in which giant cell lesions occurred, including aneurysmal cysts, hyperparathyroidism, osteoblastoma, human and rat osteopetrosis, GCT of tendon sheaths, and non skeletal granuloma were used as controls. These, together with 20 samples of normal bone (osteoclasts) did not contain intranuclear or intracytoplasmic viral inclusions.

    Notizen: Summary An ultrastructural stereological analysis of Leydig cells of the guinea pig testis was carried out following surgically induced testicular torsion. Morphometric analyses of the Leydig cells of the experimental group of animals revealed an increase in the nucelar and mitochondrial volume and a decrease in the lipid volume, in comparison to those in the Leydig cells of the control group of animals. We believe that these changes in the Leydig cells of the experimental group of animals are indicative of cellular hypertrophy. The possible mechanisms of the Leydig cell hypertrophy in the guinea pig testis following the induction of spermatic cord torsion are discussed.

    Notizen: Summary an electron microscope study of needle biopsies from the quadriceps muscle was carried out in 11 non=selected patients (ten females and one male), with clinically and laboratory-diagnosed hyperthyroid disease. Alterations of the normal structure of muscle fibres were found in all cases. Changes in capillaries were found in ten patients, and ranged from an increase in basement membrane thickness with reduplication, to total destruction of the capillaries. The importance of the vascular involvement in the muscles of patients with Graves-Basedow disease is stressed.

    Notizen: Summary Folliculo-stellate cells (FS cells) in 40 pituitary adenomas and portions of anterior pituitary adjacent to the tumor in 26 cases were investigated immunohistochemically, using polyclonal antisera to S-100 protein (S-100) and glial fibrillary acidic protein (GFAP). The objective was to clarify the histological behavior of the FS cells. In most pituitary adenomas there were few or no S-100-or GFAP-positive cell, in comparison with numerous positive cells in the parts of the adenohypophyses compressed by adenomas. However, positive FS cells were observed in some types of pituitary adenomas. Growth hormone and prolactin producing adenomas frequently contained significant amounts of FS cells. In non-functioning adenomas, an unique case of FS cell adenoma was present. The adenoma was composed mainly of FS cells and immature glandular cells. The FS cells were sometimes located around follicles containing Periodic acid Schiff-positive material. Therefore, the FS cell adenoma is characterized by S-100- and GFAP-positive FS cells and PAS-positive follicles. In this type of adenoma, FS cells seemed to be the main proliferating component. In parts of the adenohypophyses adjacent to the adenomas, GFAP0-positive FS cells were numerous. In the pathological conditions FS cells may possess the potential of reactive proliferation.

    Notizen: Summary Ia antigen, encoded within the major histocompatibility complex, plays an important role in the activation of T lymphocytes. Since experimental allergic encephalitis is an essentially T cell-mediated disease, Ia antigen in the central nervous system (CNS) may be pathogenetically relevant. The occurrence of Ia antigen in the CNS of normal rats and of rats with experimental allergic encephalitis was studied by light and electron microscope immunocytochemistry using the monoclonal anti-Ia antibodies Ox 4 and Ox 6. In normal, unsensitized animals a distict population of stellate cells in the meninges and some perivascular mononuclear cells in the nervous tissue carried Ia antigen. In rats with experimental allergic encephalitis a dramatic increase of Ia-positive cells was found. In addition to the positive cells found in normal animals, monocytes, macrophages and many lymphocytes in the meningeal perivascular and parenchymal inflammatory infiltrates as well as “activated microglia” stained for Ia antigen. We did not find evidence for Ia expression on endothelial cells, astrocytes or other components of the CNS in either normal or diseased rats.

    Notizen: Summary In adult inbred rats of the AVN strain, branches of the crural interosseous nerve were dissected out from donors and transplanted into the brain of recipients, together with a cluster of Pacinian corpuscles, (either into a suction cavity or the cerebral cortex) into a slit 1–2 mm deep. The grafts were fixed and processed for electron microscopy 10 days to 6 months after the operation, and their ultrastructure was examined. Sporadic axons of small diameter grew into the nerve branches of some of the grafts from 11 days onward, and became myelinated during the 2nd month after the operation, but none of the transplanted Pacinian corpuscles became reinnervated. The corpuscles, however, survived denervation and grafting. Most of them retained a well-preserved inner core and an intact capsule, consisting of a normal complement of 29.2±1.0 (mean ±SE) capsular layers (n=8), as did the corpuscles previously examined after denervation in situ. Some of the corpuscles underwent degenerative changes, presumably due to a delayed or restricted revascularization. In this group of corpuscles, the inner core underwent disintegration and was gradually replaced by collagen fibrils, whereas the capsule remained preserved but the number of its layers eventually reduced by 40%. It is assumed that the lack of reinnervation of the grafted Pacinian corpuscles was due to the paucity of regenerating axons, and their failure to form correct projections along those Schwann cell columns connected with the corpuscles.

    Notizen: Summary Thirty-eight human pituitary adenomas (24 endocrine active and 14 endocrine inactive tumors) were studied immunohistochemically for the presence of the basement membrane component, laminin, and ultrastructurally for the presence of basement membrane. Immunoreactivity of laminin delineated staining of epithelial and endothelial basement membranes, the reaction product being confined mostly to the perivascular zones. Moreover, a hitherto undescribed presence of intercellular laminin-positive droplets was observed in ten of the active adenomas (nine patients with hyperprolactinemia and/or acromegalia and one patient with Cushing's syndrome). Concurrently, at the ultrastructural level, bunches of basement membrane-like material intermingled between the adenoma cells were demonstrated in seven of these ten active adenomas. Furthermore, secretory granules were entrapped occasionally in this intercellular matrix, indicating a mutual dependence between excessive hormone extrusion and an increase of “misplaced” deposits of basement membrane components, e.g., laminin.

    Notizen: Summary The nature of the cells in neurofibromas was studied by electron microscopy and immunoelectron-microscopic examination of S-100 protein. Ultrastructurally, all five neurofibromas studied were found to be composed of Schwann cells, perineurial cells, and intermediate cells, which had features of both perineurial cells and fibroblasts. The Schwann cells had complex, branched cytoplasmic processes and a continuous basal lamina. The perineurial cells were distinguishable from Schwann cells by the presence of numerous pinocytotic vesicles, unbranched slender cytoplasmic processes and a discontinuous basal lamina. The intermediate cells had no basal lamina, but were topographically related to Schwann cells and had a similar fine structure to that of perineurial cells. Thus, they seemed to be modified neoplastic perineurial cells. Immunoelectron-microscopic studies showed the presence of cells with and without S-100 protein in the neurofibromas: cells with S-100 protein resembled Schwann cells ultrastructurally, and those without S-100 protein were perineurial and intermediate cells. Some Schwann cells with S-100 protein in one neurofibroma had numerous pinocytotic vesicles characteristic of perineurial cells, suggesting that Schwann cells and perineurial cells, are functional variants of the same cell type. Thus this study showed that neurofibromas were composed of Schwann cells with S-100 protein and perineurial and intermediate cells, including socalled endoneurial fibroblasts, without S-100 protein. Morphological and functional transition seems to occur between Schwann cells and perineurial cells, and between perineurial cells and intermediate cells.

    Notizen: Summary Two siblings born from consanguineous tunisian parents are reported. They showed a severe form of Farber's disease with prominent involvement of the central and peripheral nervous system: low conduction velocity was noticed in both children. Macular cherry red spots were observed in one of them. The diagnosis for the girl investigated was confirmed by evidence of ceramidase deficiency in cultured fibroblasts. Here we report the pathological findings in the subcutaneous nodules using light and electron microscopy (one case), and in sural nerves using morphometric studies (both cases). Varying morphological aspects of intracellular inclusions, depending on the tissues involved, are described and discussed. A review of all cases reported since Farber's first paper in 1952 is given.

    Notizen: Summary The question of whether the development of CNS glial cells requires the presence of axons or not can be studied with in vitro systems. In order to compare the differentiation of glial cells during development in vitro with that in situ, we have selected the optic nerve, which is anatomically as well as histotypically a well defined structure. For the in vitro investigations, small explants, called minisegments, of newborn rat optic nerves were cultivated taking four major conditions into account: (1) the regular size of the minisegments should guarantee a permanent exchange of the culture medium in order to avoid cell death, (2) neither mechanical nor enzymatic dissociation of the tissue were applied, (3) the minisegments were explanted into flasks without substrate for cell adhesion and (4) the minisegments were under constant gyratory agitation. The following in situ results were obtained: optic nerves of newborn rats are morphologically characterized by the presence of naked axons, astrocytes, glial precursors, and the absence of both differentiated oligodendrocytes and myelin. At postnatal day 5 myelin sheaths are still absent. Two weeks after birth, differentiated oligodendrocytes and microglial cells are present and numerous axons are surrounded by compact myelin. The in vitro experiments show the following main results, which were obtained after 14 h, 2 d, 5 d and 14 d in culture: during time in culture, the shape of minisegment of newborn rat optic nerves undergoes drastic changes, which indicate high cellular dynamics. After 14 h in vitro, axonal profiles, cells with pyknotic nuclei as well as clusters of astrocytes and glial precursors are present. After 2 days in culture the axonal profiles disappeared and the number of degenerating cells decreased drastically. Many large cells, probably phagocytes containing inclusions and more cells are differentiated. At the stage of 5 d in vitro 4 major types of cells can be distinguished: differentiated oligodendrocytes, which form compact and loose myelin, astrocytes, large and small glioblasts and phagocytes. Immunoprecipitates for myelin basic protein and/or myelin associated glycoprotein were found in oligodendrocytes, in their processes and associated to the myelin. Processes of some astrocytes showed immunoreactive products of glial fibrillary acidic protein. After two weeks in culture, the minisegments were mostly composed of astrocytes, whereas oligodendrocytes became rare and phagocytes disappeared. It can be concluded that CNS glial cells can attain their structural and immunocytochemical characteristics in the total absence of neuronal cell bodies and axons. However, it can be speculated that neurons (or neuronal factors) could regulate the number of astrocytes and oligodendrocytes and keep these glial cells in a physiological equilibrium.

    Notizen: Abstract We examined the morphological and functional characteristics of erythroblasts derived from marrow erythroid progenitor cells grown in a methylcellulose microculture, which were taken from a female child with rate atypical sideroblastic anaemia (SA) partially responsive to pyridoxine. Colony formation was within the normal range in three successive cultures (median values: 82.25 CFU-E and 16.4 BFU-E derived colonies/6.6×104 cells) compared to growth by normal cells (65-315 CFU-E and 9-40 BFU-E). We evaluated in vitro differentiation by biochemical microassay of a cytosol enzyme involved in the haeme pathway: uroporphyrinogen I synthase (UROS). The UROS values in the erythroid colonies from SA marrow were at the lower end of the normal range (median values: 6.7±0.3 and 14.4±3.8 pmol uroporphyrinogen/h in CFU-E and BFU-E-derived colonies respectively versus 17.4±7.3 and 25±7.2 pmol/h in CFU-E and BFU-E colonies from normal subjects. Ultrastructural examination of the SA erythroblasts from non-cultured bone marrow or derived from cultured BFU-E revealed the characteristic deposition of iron in mitochondria around the nucleus of most cells (ringed sideroblasts). However, the majority of cultured cells had marked dyserythropoietic featuress, with a large number of bilobulated or trilobulated crythroblasts, multiple cytoplasmic vacuoles, numerous abnormalities of the nucleus, and excessive membrane material beneath the plasma membrane, all features difficult to observe in non-cultured marrows.

    Notizen: Abstract Since allyl isothiocyanate has been reported to be a bladder carcinogen and benzyl isothiocyanate is a known anti-carcinogen, it is important to know the mode of their cytotoxic action. This was investigated in a RL-4 hepatocyte cell line by studying the morphological effects of increasing concentrations of the isothiocyanates and their glutathione and cysteine conjugates. These effects were compared with those induced by tert-butylhydroperoxide which supposedly has its primary effect upon the cytosolic glutathione status and thus upon the integrity of Ca2+-sequestrating mitochondria. The results agree with the previously postulated role of conjugation in the exposure of cells to isothiocyanates: Conjugates show effects similar to those produced by the free parent compounds because conjugates release free isothiocyanates in aqueous solution. The cytomorphological effects increase in a more or less dose-dependent manner with increasing concentrations of isothiocyanate or exposure time. Probably due to increased exposure, suspended RL-4 cells are more sensitive to the toxic action than cells growing on a substrate. No qualitative differences were found between the effects of allyl and benzyl isothiocyanate, indicating that their different effects in vivo are perhaps related to organ-specific differences in equilibrium between the conjugated and unconjugated forms of the test substances. The first cytomorphological effects of isothiocyanates consist of surface blebbing (zeiosis) and swelling of dictyosomal cisternae. At higher concentrations swelling extends to vesicles of endoplasmic reticulum. Mitochondria are not affected until the cells reach the necrotic phase of injury. In contrast, tert-butylhydroperoxide causes mitochondrial damage in an early pase of toxic injury. The cellular symptoms suggest that the primary target of isothiocyanates is in the plasma membrane and the cellular membrane system, affecting the monovalent cation and water balance of the cell organelles rather than the Ca2+ homeostasis as in cells exposed to tert-butylhydroperoxide. Differences in lipophilicity may be at the basis of this differenc in primary action.

    Notizen: Summary Electron microscopic data on the development of myonecrosis following cerebral vasospasm associated with subarachnoid haemorrhage, meningitis and trans-sylvian surgery are presented. The basic feature of myonecrosis was dissolution of myofilaments with resultant fine granular or filamentous material. The disintegrating cytoplasm often contained numerous glycogen granules, dense bodies, autophagic vacuoles and myelin-like membranous bodies. A well-developed sarcoplasmic reticulum was preserved despite myofilament dissolution, while mitochondria showed marked sweling. The nuclei showed either dilution of chromatin or pyknotic change. The basal lamina was remarkably thickened and maintained an irregular outline of the necrotic smooth muscle cells. Enlarged intercellular space contained abundant cellular debris, vesicular structures and connective tissue fibres. The pathogenesis of these changes is discussed.

    Notizen: Abstract The three dimensional organization of the complete photosynthetic apparatus of the extremely halophilic, bacteriochlorophyll b containing Ectothiorhodospira halochloris has been elaborated by several techniques of electron microscopy. Essentially all thylakoidal sacs are disc shaped and connected to the cytoplasmic membrane by small membraneous “bridges”. In sum, the lumina of all thylakoids (intrathylakoidal space) form one common periplasmic space. Thin sections confirm a paracrystalline arrangement of the photosynthetic complexes in situ. The ontogenic development of the photosynthetic apparatus is discussed based on a structural model derived from serial thin sections.

    Notizen: Abstract The genome of the slow-growing Bradyrhizobium japonicum (strain 110) was mutagenized with transposon Tn5. A total of 1623 kanamycin/streptomycin resistant derivatives were screened in soybean infection tests for nodulation (Nod) and symbiotic nitrogen fixation (Fix). In this report we describe 14 strains possessing a stable, reproducible Nod+Fix- phenotype. These strains were also grown under microaerobic culture conditions to test them for free-living nitrogen fixation activity (Nif). In addition to strains having reduced Fix and Nif activities, there were also strains that had reduced symbiotic Fix activity but were Nif+ ex planta. Analysis of the genomic structure revealed that the majority of the strains had a single Tn5 insertion without any further apparent physical alteration. A few strains had additional insertions (by Tn5 or IS50), or a deletion, or had cointegrated part of the vector used for Tn5 mutagenesis. One of the insertions was found in a known nif gene (nifD) whereas all other mutations seem to affect different, hitherto unknown genes or operons. Several mutant strains had an altered nodulation phenotype, inducing numerous, small, widely distributed nodules. Light and electron microscopy revealed that most of these mutants were defective in different stages of bacteroid development and/or bacteroid persistence. The protein patterns of the mutants were inspected by two-dimensional gel electrophoresis after labelling microaerobic cultures with l-(35S)methionine. Of particular interest were mutants lacking a group of proteins the synthesis of which was known to be under oxygen control. Such strains can be regarded as potential regulatory mutants.

    Notizen: Summary A single instillation of 1 mg chrysotile B with a fiber length between 0.05 and 0.2 µm in 0.1 ml tricaprylin was made via a polyvinyl catheter into the lower lobe of the right lung of 120 six-week-old Wistar rats under anesthesia. The animals were killed at intervals between five minutes and two years. The lower lobes of the right lung were investigated by light and electron microscopy. The process of pulmonary fibrosis induced by asbestos can be subdivided into four phases: these are the phase of phagocytosis (five to 15 min), the phase of granuloma formation (between one and two weeks), the phase of septal fibrosis (between two and six months) and finally the scar stage (after one year). After instillation of small asbestos fibers into the alveoli, a major proportion of these fibers is phagocytosed by alveolar macrophages after five minutes and leaves the lungs via the airways. A proportion of the fibers penetrates through the alveolar wall (mostly conveyed by type I pneumocytes) and reaches the interstitium of the lungs. There, the fibers are taken up by pulmonary tissue macrophages and giant cells. Within the phagolysosomes, the fibers are broken down into fragments less than 0.01 µm in length. Type II pneumocytes produce surfactant in excess. These cells become necrotic, tubular myelin and lamellar bodies pass into the alveoli and into the interstitium. Surfactant is phagocytosed by resident macrophages. These macrophages phages can break down. Besides asbestos and surfactant, mediators of fibrillogenesis are released. Macrophages following up from blood monocytes ingest surfactant and asbestos. This process is perpetuated up to complete scarring. After two years, small asbestos fibers less than 0.01 µm long are present in fibroblasts and pleural mesothelia.

    Notizen: Summary A comparative analysis of the Vicia faba mitochondrial genome in whole plants and in longterm suspension culture has been conducted. Restriction fragment patterns of the mtDNA isolated from these two sources were notably different. Electronmicroscopic analysis also revealed significant differences. Large circular mtDNA patterns shifted from a 37–80 kb subpopulation, which was predominant in whole plants, to 18–34 kb subpopulations although in both classes notable quantities of circular molecules of 80 to 120 kb and more were also found. Both in whole plant and suspension culture cells very large circular DNAs were observed. Some of them had lengths nearly 290 kb and could be considered as evidence of the existence of master chromosomes. The minicircular DNA population was also altered. In the suspension culture we observed a notable increase of percentage of minicircles with sizes near 1 kb. Simultaneously, the percentage of minicircles with sizes near 3.5–10 kb significantly increased in suspension culture cells. In addition, a new peak (10–12 kb) of minicircles appeared. Copy number alterations for some sequences homologous to CCC1A, CCC1B and CCC2 (Negruk et al. 1982, 1985) were shown. Southern hybridization revealed the existence of a family of minicircles having sizes 1.4–2 kb with predominance of CCC1A, CCC1B and CCC2. The copy numbers of CCC1B and some minor minicircles was changed in the suspension culture when compared with the whole plants.

    Notizen: Summary Biochemical and ultrastructural analysis of epidermis from the porpoise, Phocena phocena, revealed certain similarities and differences between cetaceans and terrestrial mammals. The predominant cell of cetacean epidermis, not found in normal terrestrial mammals, is a lipoker-atinocyte, which elaborates not only keratin filaments, but also two types of lipid organelles: first, lamellar bodies, morphologically identical to those of terrestrial mammals, are elaborated in great abundance in all suprabasal epidermal layers, forming intercellular lipid bilayers in the stratum corneum interstices: and second, non-membrane-bounded droplets appear and persist in all epidermal layers. Although the porpoise lipokeratinocyte morpologically resembles the sebokeratocyte of avians in certain respects, nonmembrane-bounded lipid droplets are not released into the intercorneocyte space as they are in avian stratum corneum. Whereas phospholipid/neutral lipid gradients are similar in porpoise and terrestrial mammals, PAS-positive glycoconjugates, specifically glycosphingolipids, are retained in porpoise stratum corneum, but lost from these layers in terrestrials. The novel, non-polar acylglucosyl-ceramides, which also are lost during cornification in terrestrial mammals, are retained in porpoise stratum corneum. The lipid components of porpoise lipokeratinocytes appear to subserve not only barrier function in a hypertonic milieu, but also underlie the unique buoyancy, streamlining, insulatory, and caloric properties exhibited as adaptations to the cetacean habitat.

    Notizen: Summary An ultrastructural study of the features and distribution of collagen fibrils was performed in the endometrium of virgin and pregnant (2nd to 11th day) mice. Collagen-containing structures were observed in the cytoplasm of fibroblasts on the 2nd day of pregnancy. Treatment of tissues with lanthanum nitrate established that these structures were intracytoplasmic. Their association with lysosome-like bodies suggested the occurrence of intracellular digestion of collagen, probably connected with remodeling of the endometrial stroma prior to decidualization. On the 4th day of pregnancy, very few collagen fibrils were present in the intercellular space. From the 6th day of pregnancy onwards, “thick” collagen fibrils were observed between decidual cells. The diameter of these fibrils measured up to 300 nm whereas the fibrils present in the endometrium of virgin mice measured 40–68 nm.

    Notizen: Summary The present study describes the ultrastructure of non-neuronal cells and their interrelationships with intracardiac neurones present in cultures dissociated atria and interatrial septum from newborn guinea-pig. When compared with the in situ preparation, most of these features in culture were similar to those observed in situ, but some differences were also apparent. Both mature and immature Schwann cells were observed in culture, and as in situ, the latter were closely associated with intracardiac neurones, whilst the former were more widely separated. The ultrastructure of satellite cells was more variable in culture than in situ: three general types were distinguished on the basis of their 10-nm filament content. This variation could be due to conditions of culture. Interstitial cells were present in culture and closely resembled those described in situ, although there was less space between cultured interstitial cells and their associated cells. Many fibroblasts, some myoblasts and a few mast cells were also found in the culture preparations.

    Notizen: Summary The ultrastructure of cultured intrinsic neurones and SIF (small intensely fluorescent) cells dissociated from the atria and interatrial septum of newborn guinea-pig heart has been studied for the first time and compared with these cells in situ. Mononucleate and binucleate neuronal somata and their processes were observed in the culture preparation; their ultrastructure was similar to that of neurones in intracardiac ganglia observed in situ. The number of neurites associated with neuronal cell bodies increased after the first week in culture. A subpopulation of intracardiac neurones showed abnormalities in culture, comparable to the changes previously described in neurones of the monkey heart after unilateral vagotomy in situ. Small granule-containing cells were observed in culture, corresponding to those described in the heart in situ. One type of large process in the culture preparation containing densely packed mitochondria has not been seen in situ, suggesting that changes in cell ultrastructure due to the conditions of culture cannot be discounted. However, the ultrastructure of the cultured cells was, for the most part, consistent with that of the same cell type in situ, indicating that the culture preparation may be a useful model for investigation of the roles and interactions of intramural neurones in the heart, which are inaccessible for such studies in situ.

    Notizen: Summary Various stabilization and extraction procedures were tested to demonstrate the ultrastructural organization of the cytoskeleton in normal, locomoting Amoeba proteus. Most reliable results were obtained after careful fixation in glutaraldehyde/lysine followed by prolonged extraction in a polyethylene glycol/Triton X-100 solution. Before dehydration in a graded series of ethanol and critical-point drying, the amoebae were split by the sandwich-technique, i.e., by mechanical cleavage of cells mounted between two poly-L-lysine-coated glass slides. Platinum-carbon replicas as well as thin sections prepared from such cell fragments revealed a cytoskeleton composed of at least four different types of filaments: (1) 5–7-nm filaments organized as a more or less ordered cortical network at the internal face of the plasma membrane and probably representing F-actin; (2) 10–12-nm filaments running separately or slightly aggregated through the cytoplasm and probably representing intermediate filaments; (3) 24–26-nm filaments forming a loose network and probably representing microtubules; and (4) 2–4-nm filaments as connecting elements between the other cytoskeleton constituents. Whereas microfilaments are responsible for protoplasmic streaming and other motile phenomena, the function of intermediate filaments and cytoplasmic microtubules in amoebae is still obscure.

    Notizen: Summary The surface of extracellular merozoites of P. knowlesi is covered with a coat 15–20 nm thick, made up of clusters of filaments standing erect on the plasma membrane. Filaments have stems 2 nm thick, the peripheral ends of which are complex, branching or ending in long trailing threads. Coat filaments occur on the surface of the parasite in regular rows at an early schizont stage, and persist until well after merozoite release. They are sensitive to trypsin and papain, and bind ethanolic phosphotungstate, indicating a proteinaceous nature. They are also removed by exposure to phosphate-buffered saline. Filaments bear negative charges, binding cationised ferritin throughout the depth of the coat and staining with ruthenium red. They cover the whole merozoite surface and mediate intercellular adhesion at distances of 15–150 nm, membrane to membrane. It is suggested that these filaments correspond to a major merozoite surface protein, and are important in the initial capture of red cells.

The projections of two different neurons into the nephridia are described: 1. The overall morphology of the receptor muscle, the sensory neuron and its dendrites was found to share many common features with other arthropod sense organs of that type with two important differences: (1) the connective tissue segment (= intercalated tendon) is extremely short compared to that of other muscle receptor organs; (2) the naked dendritic terminals of the non-ciliated, multipolar sensory neuron of the organ contain clusters of microtubules, interconnected by an amorphous matrix, that resemble the tubular bodies of ciliated, epithelial receptor cells.

    Notizen: Summary The median eminence (ME) of the adult frog, Rana temporaria, was studied by means of electron microscopy including quantitative electron-microscopic autoradiography. In frogs captured in May and June numerous peptidergic neurosecretory fibres extending via the internal zone to the pars nervosa display large swellings containing few granules, mitochondria, neurotubules and cisternae of the smooth endoplasmic reticulum. In addition, few secretory globules up to 1.5 μm in diameter occur in these varicosities. In animals collected during the autumn period many of these neurosecretory swellings filled with neurosecretory granules and polymorphic inclusions resemble Herring bodies. Three types of granule-containing neurosecretory fibres were observed in the external zone (EZ) of the ME of adult R. temporaria. Peptidergic A1- and A2-type fibres are characterized by granules 150–220 nm and 100–160 nm in diameter, respectively. Monoaminergic fibres of type B with granules approximately 100 nm in diameter represent ∼ 50% of all neurosecretory elements in the EZ of the frog ME; ∼12% of the total number of granule-bearing axons in the EZ actively taking up radiolabelled 5-hydroxytryptophan are thought to be serotoninergic terminals. Neurosecretory terminals of all types and glial vascular endfeet establish direct contacts with the perivascular space of the primary portal capillaries. Some neurosecretory terminals are separated from the lumen of the third ventricle by a thin cytoplasmic lamella of tanycytes. The possible physiological significance of this structural pattern is discussed.

    Notizen: Summary The differentiation of cells and synapses in explants of 9-day-old chick embryo retina has been studied by light and electron microscopy over a period of 35 days in vitro, and samples of retina from the 9-day chick foetus were directly fixed and prepared for study. At the time of explantation the retinae were poorly differentiated and no lamination was apparent. From day 14 onwards, (i) outer and inner nuclear layers (ONL, INL) separated by a layer of neuropil corresponding to the outer plexiform layer (OPL) and (ii) a layer of scattered large ganglion cells separated from the INL by a zone of neuropil resembling the inner plexiform layer (IPL) were apparent, and (iii) a well-differentiated outer limiting membrane was established close to the surface of the explants. In the oldest cultures some development of photoreceptor outer segments occurred but a distinct optic nerve fibre layer did not form. Although cell identification presented problems even in the oldest cultures, the major retinal cell types described in vivo could be identified. Photoreceptor cells developed pedicles in the OPL which became filled with synaptic vesicles and synaptic ribbons and established ribbon synapses (including triads) with and were commonly invaginated by processes from horizontal and bipolar cells. Processes of bipolar cells in the IPL formed simple and dyad synapses. At least two types of presynaptic amacrine cells were also identified in the INL, one of which contained large numbers of dense-core vesicles. The ganglion cells, though sparse, were large and well differentiated. These findings show that all the major neuronal types of the retina are capable of developing and differentiating in vitro, lagging behind the time-table of development and differentiation in vivo by approximately 7 days, but resulting in a histotypically organised retina with synaptic neuropil showing many similarities to the corresponding neuropil in vivo.

    Notizen: Summary In the genital tract of male and female mouse embryos cholinesterase activity is described that is independent from innervation. The enzyme activity is localized in the mesenchyme at the junction of Wolffian and Müllerian ducts with the urogenital sinus. During male development prostate buds and vesicular glands grow out into the cholinesterase-active mesenchyme. During female development the active mesenchyme participates in the downgrowth of the vaginal anlage. Ultrastructurally the cholinesterase activity is localized in the perinuclear cisterna and in smooth endoplasmic reticulum of the mesenchymal cells. The enzyme activity disappears with definitive differentiation of the tissue. The embryonic cholinesterase is a component of a primitive muscarinic system. Its relation to the morphogenetic action of testosterone and its possible general functions are discussed.

    Notizen: Summary Wheat roots from germinating seedlings of Chinese spring wheatTriticum aestivum grown for 36 hours at 20°C were examined by conventional thin-section electron microscopy. Virus-like particles were seen inside a large cytoplasmic intrusion into the nucleus having the appearence of a nucleolar vacuole. The particles were isometric and about 50 nm in diameter with a membrane-like coat and a small core. The cytoplasmic intrusion was bounded by nuclear envelope with pores apparent where it abutted nucleoplasm. The particles are similar to previously reported solitary particles “S bodies” from a range of plants but are also similar in size and morphology to the retroposon particles associated with copia like elements in other organisms. The position of the virus-like particles in the young wheat roots is discussed in relation to interactions with components of the cell skeleton.

    Notizen: Summary Internodal cells ofChara, grown in culture either at pH 5.7, 6.5 or 7.5, were studied to determine their chloride influx capability, the quantitative aspects of charasome morphology and the degree to which these two parameters could be correlated. In cells grown at pH 5.7 the charasomes were relatively small, were widely spaced on the plasma membrane, and contributed only a 0.6% increase to the surface area of the plasma membrane in the acid region of the cell. In contrast, the charasome membrane surface area of cells grown at pH 7.5 had increased × 19, the density of charasomes on the cell surface increased × 42, thus producing a × 3.57 increase in the acid region plasma membrane surface area. Chloride influx in cells grown at pH 7.5 was × 8.7–12.7 greater than in cells grown at pH 5.7. Cells that had been starved of chloride exhibited a × 2.4 average increase in the rate of chloride influx. Our observations establish the existence of a positive correlation between the rate of chloride influx and the increase in membrane surface area due to charasomes, although other factors, such as the effect of pH on transport-related enzymes, and the effect of charasome structure on chemical equilibria, may also be of importance.

    Notizen: Summary The possible role of inorganic phosphate (Pi) in depressing the contractility of oxygen-deficient myocardium was investigated by examining the effects of 30 mM Pi on the cardiac function and myocardial biochemistry and fine structure in normoxic and anoxic Langendorff-perfused isolated rat hearts. In normoxia, the intracellular Pi level increased three-fold, the ATP content remained normal, and there was moderate loss of glycogen only. Contractile performance (as assessed from systolic pressure recordings) was significantly depressed, as was the heart rate for the first 10 min. The myocardial fine structure showed persistent glycogen, marked relaxation of myofibrils, and a higher incidence of vacuolation than in hearts with normal Pi. In anoxia, the intracellular Pi level was comparable with that of the perfusate and both ATP and glycogen were severely depleted. Contractile performance and heart beat ceased completely at 15 min, although in anoxic controls both persisted at low levels for at least 25 min. In anoxia, Pi also depressed coronary flow rate. In the inner half of the ventricular wall of oxygen-depleted hearts, where flow became reduced after 15 or more min, Pi markedly reduced the formation of intramitochondrial densities and augmented mitochondrial swelling and ischaemic contracture, which extended out through the mid-myocardium. In the outer half of the wall, where flow remained high, it promoted severe dilatation of the sarcoplasmic reticulum vesicles and undifferentiated regions of the intercalated discs. The observed effects in normoxia are probably attributable at least in part to the lowering of the free Ca2+ concentration of the perfusate by the increased Pi level. The effects in anoxia may be related chiefly to the critical reduction of available intracellular Ca2+ and the more rapid and extensive development of ischaemic contracture.

    Notizen: Summary To study the structure of in vivo mitochondrial DNA recombination intermediates in Saccharomyces cerevisiae, we used a deletion mutant of the wild type mitochondrial genome. The mtDNA of this petite is composed of a direct tandem repetition of an ∼4,600 pb monomer repeat unit with a unique HhaI restriction enzyme site per repeat. The structure of native mtDNA isolated from log phase cells, and mtDNA crosslinked in vivo with trioxsalen plus UVA irradiation, was studied by electron microscopy. Both populations contained crossed strand “Holliday” type recombination intermediates. Digestion of both non-crosslinked and crosslinked and mtDNA with the enzyme HhaI released X and H shaped structures composed of two monomers. Electron microscopic analysis revealed that these structures had pairs of equal length arms as required for homologous recombination intermediates and that junctions could occur at points along the entire monomer length. The percentage of recombining monomers in both non-crosslinked and trioxsalen crosslinked mtDNA was calculated by quantitative analysis of all the structures present in an HhaI digest. The relationship between these values and the apparent dispersive replication of mtDNA in density-shift experiments and mtDNA fragility during isolation is discussed.

    Notizen: Summary Plasmids carrying hydrogenase genes in Alcaligenes eutrophus wild type H 16 and in two transposon Tn5 —induced mutants have been investigated by electron microscopy. Besides the pHG1 megaplasmid (458±27 kb) carrying genes coding for structural and regulatory properties of hydrogenases, small plasmids of unknown significance have been detected. The sizes of EcoRI fragments obtained from pHG1 were measured from electron micrographs. They were significantly different from sizes determined previously by agarose gel electrophoresis. Plasmid pHG1 isolated from the wild type H 16 was shown to contain two inverted repeats (IR 16-1 and IR 16-2) with sizes similar to known transposons. From electron microscopic hybridization studies, it was deduced that the sites of insertion of Tn5 into a regulation gene on pHG1 for both soluble and membrane-bound hydrogenase, and of Tn5-Mob into the gene coding for structural properties of the soluble hydrogenase, are about 67.2 kb apart. One of the inverted repeats (IR 16-1) was localized in between these sites.

    Notizen: Summary We sought to clarify whether or not hyperlipoproteinemia induces auditory dysfunction. In so doing, we studied the general states and cochlear pathologies of guinea pigs after the administration of a hyperlipid diet for 3 months. Serum biochemistries indicated marked elevations of cholesterol, high density lipoprotein (HDL)-cholesterol and low density lipoprotein (LDL) levels. An increased auditory threshold varying from 10 to 20 dB was observed in 40% of the guinea pigs using auditory brainstem responses. Histochemical study of the inner ear revealed variations in lipid metabolism and partial disorders of the outer hair cells. Electron microscopic observations showed vacuolar and parenchymal protrusions on the surfaces of the stria vascularis and Corti's organ, and vacuolar degeneration was seen around the capillary vessels of the vascular stria. Our data has shown that the auditory dysfunction present in the inner ear was less marked than were the morphological changes seen. Our findings suggest that other factors besides hyperlipoproteinemia are involved in the development of severe auditory damage.

    Notizen: A freezing apparatus has been developed for bringing blocks of tissue into contact with a block of sapphire chilled to 17°K. A toggle linkage minimizes rebound by slowing the rate of approach of the tissue to the cold surface to a velocity of zero. A glove box limits condensation on the surface of the sapphire, and a miniature moist chamber protects the specimen from drying and premature freezing. About 50 blocks of tissue can be frozen in an hour and a half by using 5 liters of liquid helium. The tissue is then frozendried at controlled temperature, fixed with OsO4 vapor, and infiltrated with epoxy resin in a simple bench-top freeze-drier without breaking vacuum. About two-thirds of the blocks are useful for electron microscopy. Brain tissue frozen and dried by using these methods retains enough immunoreactivity for enkephalin in plastic sections to permit its detection with immunohistochemistry by using both the light microscope (with immunofluorescence) and the electron microscope (with colloidal gold).

    Notizen: Purified rat peritoneal mast cells contained 3.3 × 10-5 gm SO4 and 2.2 × 10-8 gm Ca/106 cells. The molar ratio of S/Ca in the whole cell was 600:1. Frozen thin sections of unfixed mast cells contained only sulfur (S) in the granules when examined by X-ray energy dispersive spectroscopy (EDS). Mast cells fixed in 3% glutaraldehyde and 1.5% formaldehyde in 75% ethanol (Et/Ald) or in mixed buffered aldehydes and embedded in Epon 812 or the low viscosity resin diepoxyoctane (DEO) contained S in all granules and Ca in some of the granules measured. Neither element was found in the nucleus, cytoplasm, or resin. Isolated, Et/Ald fixed and embedded granules also contained S. The presence of Ca in the granules was artifactual in that the Ca was absorbed from water in the trough of the diamond knife and/or from the filter paper used to blot the sections dry. This phenomenon was investigated further. Sections of Et/Ald fixed and embedded mast cells were incubated with 5 × 10-6 to 10-2 M CaCl2. Ca was detectable in 100% of the granules incubated at concentrations ≥ 10-4 M and reached a constant S/Ca ratio of 2.0 at concentrations ≥ 10-3 M. Ca was not detectable in the nucleus, cytoplasm, or resin at 10-2 M. A plot of S versus Ca counts from the granules of cells incubated with 10-2 M CaCl2 was linear with a slope of 2.0 and a correlation coefficient of 0.88. Et/Ald fixed cells incubated with distilled H2O had fewer granules containing Ca, 10%, than unincubated cells, 77%. Further, H2O removed all Ca from Et/Ald fixed cells embedded in DEO. These studies show that S, which is present as SO4 on the proteoglycan heparin, is readily detectable by X-ray EDS in fixed and embedded cells. An artifact of the technique is that weak anionic sites, which are most probably carboxyl groups on the proteoglycan, can bind the divalent cation Ca and cause spurious localization.

    Notizen: In osteochondrosis juvenilis Scheuermann, foci of various sizes in the cartilaginous end plates of the vertebral bodies display a loosening or complete interruption of the collagen fibers. These findings, together with an alteration and occasional absence of the growth zone, may result in the typical deformation of the vertebral bodies. Electron micrographs of the areas with optically absent collagen fibers reveal collagen fibrils. They are arranged in an irregular pattern. We conclude that a disturbance of collagen or ground substance biosynthesis is of importance in the pathogenesis of juvenile osteochondrosis.

    Notizen: Pyramidal tract (Pt) neurons in the sensory-motor cortex of cat were labeled by injection of HRP into the spinal cord. Ultrastructural and quantitative analysis of the synaptic covering of their soma and apical dendrite (up to 100 μm from soma) was undertaken. We intercalated a visual-manual treatment between composite electron micrographs and a fully automated computer system and developed specific programs for evaluation of the morphometric data. Programs are included. A total of 412 synaptic boutons were examined that were found in contact with large Pt neurons. The mean linear percentage of the surface area covered by boutons was 26.2 ± 8.4% and the mean contacting length and cross-sectional area of the bouton profiles were 1.28 ± 0.58 μm and 0.89 ± 0.59 μm2, respectively. All types of boutons with active zones accounted for 41.2% of the total. The distribution of two types of bouton (S- and F-type boutons, showing asymmetric and symmetric contacts, respectively) was examined quantitatively. The mean proportion of F-type boutons was 89.1% with a soma and S-type boutons contacting apical dendrites was 10.9%. In addition, GABAergic boutons were identified with the soma by immunocytochemistry with antibodies against glutamic acid decarboxylase. They formed symmetric synaptic contacts with the Pt cells that were identical to those formed by F-type boutons. The quantitative analysis revealed that synaptic clefts are narrower and synaptic vesicles are smaller in symmetric F-type boutons than in S-type boutons forming asymmetric contacts. These data establish that at least three parameters (postsynaptic density, synaptic cleft, and size of vesicles) can be utilized singly or in combination to identify GABAergic inhibitory synapses in neocortex.

    Notizen: To investigate possible structural changes in reassembled proteoglycan aggregates during cartilage mineralization, we examined the molecular architecture and dimensions of growth plate proteoglycan aggregates by electron microscopy. The ends of fetal bovine femurs and tibias were separated into three regions: the epiphysis; the cartilage growth plate, consisting of the proliferative zone and the unmineralized portion of the hypertrophic zone; and the calcified portion of the hypertrophic zone along with part of the metaphysis. Aggregates from all three regions had the same molecular architecture. They consisted of central hyaluronic filaments with multiple attached monomers. Monomers consisted of two segments: (a) a peripheral thick segment, which represents primarily the chondroitin sulfate-rich region, and (b) a thin segment attached directly to the hyaluronic acid filament. The length of aggregated monomers did not differ between the growth plate cartilage and the metaphysis, nor did the lengths of the thin and thick segments, indicating that the chondroitin sulfate-rich region of aggregated monomers is not degraded during cartilage mineralization. Between the growth plate cartilage and the metaphysis, aggregates became shorter and had fewer monomers and wider spacing between monomers. These structural alterations in proteoglycan aggregates may be one of the events that prepares the matrix for mineralization.

    Notizen: To be successful, today's electron microscopist must have modern, well-maintained facilities and be part of a team of experts with specific skills for research. Furthermore, in an ideal situation specific instruments should be dedicated to specific applications (high resolution, analytical, conventional, etc.), since in general the efficiency of a microscope varies inversely as its operations (and operators?). This is partly a reason for establishing national or regional electron microscopy centers, so that expertise and expensive equipment may be made available in central facilities.In this brief summary an attempt is made to illustrate how sophisticated methods of electron microscopy can be applied to practical problems by choosing representative examples from research at Berkeley using high resolution, high voltage, diffraction, and analytical techniques. More detailed examples have been given in some recent reviews (Thomas, 1984a, and Gronsky et al., 1985).

    Notizen: Ease of cutting thin sections with glass knives is markedly improved if the embedding resin contains a surfactant such as lecithin. With lecithin, it is possible to cut 50-100 thin sections from the same place on the knife edge even after facing off the block with 1-2-μm-thick sections. Image quality is similar to that of the unmodified resin if the resin formula is optimized. If not, some chatter or a “mottled” appearance of the tissue image may be present. Lecithin does not significantly affect sectioning with a diamond knife or the appearance of the section in the microscope. The increased ease of sectioning with glass presumably will be translated to diamond knives in the form of an increased useful life of the cutting edge.

    Beschreibung / Inhaltsverzeichnis: Résumé Un implant céramique non poreux est testé au niveau du fémur de rat en ce qui concerne son adhésivité à l'os. Un certain nombre de techniques morphologiques sont utilisées pour examiner le rapport entre l'implant et l'os néoformé. La microscopie électronique par transmission et la microscopie par fluorescence après marquage à la tétracycline ont donné les meilleurs résultats. Un rapport étroit entre l'os minéralisé et la céramique a été noté en microscopie électronique. Par marquage à la tétracycline, il semble que l'implant puisse stimuler la formation osseuse.
    Kurzfassung: Zusammenfassung Ein unporöses keramisches Implantat in Rattenfemora wurde auf seine Fähigkeit geprüft, sich mit Knochen zu binden. Eine Anzahl morphologischer Techniken wurde verwendet, um die Beziehung zwischen den Oberflächen von Implantat und neuem Knochen zu untersuchen. Transmissions-Elektronenmikroskopie und Fluoreszenzmikroskopie nach Tetracyclinmarkierung waren die erfolgreichsten Techniken. Eine enge Beziehung zwischen mineralisiertem Knochen und dem Keramikimplantat konnte mit der Transmissions-Elektronenmikroskopie nachgewiesen werden. Das Aussehen der Tetracyclinmarkierung im keramischen Implantat deutet darauf hin, daß dieses wahrscheinlich die Fähighkeit hat, Knochenbildung zu erhöhen.
    Notizen: Abstract A nonporous ceramic implant in rat femora was evaluated as to its ability to bond to bone. A number of morphologic techniques were utilized to examine the interfacial relationship of the implant to new bone. Transmission electron microscopy and fluorescence microscopy after tetracycline labelling were the most successful techniques. An intimate relationship between mineralized bone and the ceramic was demonstrated by transmission electron microscopy. The appearance of tetracycline labelling at the ceramic interface indicates that the implant may have capacity to enhance bone formation.

    Beschreibung / Inhaltsverzeichnis: Résumé Du collagène d'os compact de mouton est préparé par décalcification dans I'EDTA et à partir de tendons de queux de rats, par extraction dans l'acide acétique et reconstitution dans NaCl. Le dépôt d'apatite dans le collagène osseux de mouton dans une solution de calcification métastable est étudié chimiquement et par microscopie électronique. Le collagène osseux est un bon catalyseur de nucléation pour le dépôt minéral, alors que le collagène de tendons de rat ne l'est pas. Le dépôt minéral du collagène osseux se produit en deux phases cinétiques séparées, une phase rapide de nucléation et une croissance cristalline, donnant naissance à de petits ilots calcifiés et une seconde phase lente de croissance dans des régions ne comportant pas de zones catalytiques. La seconde phase de dépôt minéral paraît être le résultat d'une diffusion inhibée d'ions à travers les fibrilles collagènes alignées, laissant de larges régions de collagène sans minéral, bien que le tampon reste hautement sursaturé. La microscopie électronique permet de penser que les zones de catalyse pourraient avoir un rapport avec la périodicité de 640 Å de collagène, mais l'importance d'un matériel noncollagènique, lié au collagène, n'est pas à exclure. L'activité catalytique faible du collagène reconstitué n'est pas liée à la présence d'inhibiteurs faiblement liés, bien que des inhibiteurs puissent être intimement liés à ce type de collagène, qui pourrait être absent du collagène osseux. La différence d'activité catalytique pourrait intervenir dans la calcification physiologique. Une hypothèse plus générale pour la nucléation de la phase minérale dans les systémes biologiques est nécessaire.
    Kurzfassung: Zusammenfassung Kollagen wurde aus kompaktem Schafsknochen mittels EDTA-Entkalkung und aus Rattenschwanzsehnen durch Essigsäureextraktion und Rekonstitution mit NaCl gewonnen. Die Apatitablagerung aus einer metastabilen Verkalkungslösung auf Schafsknochenkollagen wurde chemisch und im Elektronenmikroskop untersucht. Es zeigte sich, daß das Knochenkollagen ein guter Nukleationskatalysator für die Mineralablagerung ist, was beim Rattenschwanzkollagen nicht zutraf. Im Knochenkollagen erfolgte die Mineralablagerung in zwei getrennten kinetischen Phasen: einer raschen Phase der Nukleation und des Kristallwachstums, welche kleine verkalkte Inseln entstehen läßt, und einer zweiten langsamen Phase, welcher das Wachstum in Bezierken, die keine katalytischaktiven Stellen einschließen, zuzuschreiben ist. Diese zweite Phase der Mineralablagerung wird als Resultat einer verminderten Ionendiffusion durch die enganeinanderliegenden Kollagenfibrillen angesehen, wodurch weite Kollagenbereiche ohne Mineral bleiben, obwohl der Puffer stark übersättigt ist. Elektronenmikrographien ließen vermuten, daß die katalytischaktiven Stellen in einem gewissen Verhältnis zur 640 Å-Periodizität des Kollagens stehen; es konnte jedoch nicht ausgeschlossen werden, daß nicht-kollagenhaltiges Material, welches an Kollagen gebunden ist, ebenfalls eine Rolle spielt. Die schlechte katalytische Aktivität des rekonstituierten Kollagens konnte nicht auf die Anwesenheit von schwachgebundenen Hemmstoffen zurückgeführt werden, obwohl Inhibitoren stark an dieses Kollagen gebunden sein könnten, die jedoch im Knochenkollagen nicht vorhanden sind. Die Unterschiede in der katalytischen Aktivität können mit der physiologischen Verkalkung in Beziehung stehen. Eine allgemeinere Hypothese für die Nukleation einer Mineralphase in biologischen Systemen wäre erforderlich.
    Notizen: Abstract Collagen was prepared from compact sheep bone by decalcification with EDTA and from rat tail tendons by acetic acid extraction and reconstitution with NaCl. The deposition of apatite in sheep bone collagen in a metastable calcification solution was studied chemically and by electron microscopy. The bone collagen was shown to be a good nucleation catalyst for mineral deposition, while rat tail collagen was a poor catalyst. Mineral deposition in bone collagen occured in two separate kinetic phases, a rapid phase of nucleation and crystal growth, giving rise to small calcified islands, and a second slow phase, ascribed to growth in regions not involving the catalytic sites. This second phase of mineral deposition is considered to be the result of impaired ion diffusion through the closely-aligned collagen fibrils, thus leaving large areas of the collagen free of mineral even though the buffer remains highly supersaturated. Electron micrographs suggested that the catalytic sites might be in some relationship to the 640 Å periodicity of collagen, but a role for non-collagenous material bound to the collagen has not been excluded. The poor catalytic activity of reconstituted collagen was not due to the presence of loosely-bound inhibitors, although inhibitors could be strongly bound to this type of collagen and be absent from bone collagen. The differences in catalytic activity may have a bearing on physiological calcification. A more general hypothesis for nucleation of a mineral phase in biological systems is required.

    Notizen: Summary Quantitative electron microscopical observations have been made on the afferent synaptology of neurones in laminae V and VII of the cervical spinal cord of the cat. Both laminae contain large bouton-covered polydendritic neurones of the type seen in the brainstem reticular formation. Each lamina contains two other categories of neurone. One of these, in lamina V, is peculiar in having a very high proportion of afferent boutons filled with dense-core vesicles; similar neurones exist in lamina VII, but their presynaptic afferents do not contain unusual quantities of dense-core vesicles. The third type of neurone in laminae V and VII seems to be identical with an oligodendritic neurone type found in the brainstem reticular formation and parafascicular-centromedian complex.

    Notizen: Summary Electron microscopic examination of Purkinje Cells was performed in sections from the cerebellum of three albino rats aged 4 1/2 month, intoxicated with diphenylhydantoin for 51 days. Three untreated albino rats served as controls. There were no difference between the substructure of the Purkinje cells from the two groups of animals. It was concluded that diphenylhydantoin in toxic but sublethal doses does not change the substructure of the Purkinje cells.

    Notizen: Summary Following administration of psychotropic drugs, the brains of rabbits were examined by electron microscopy, with special references to glycogen granules in the hippocampus, and were compared with the normal materials. After seven days' intramuscular injections of chlorpromazine (15 mg/kg/day) or trifluoperazine (10 mg/kg/day), glycogen granules appeared diffusely in the perikaryal cytoplasm of the pyramidal cells and small nerve cells and in the myelinated nerve fibers, and were accumulated in the dendrites of pyramidal cells and in the astrocytic cytoplasm. Those animals showed neuroleptic symptomes and extrapyramidal ones throughout the period. The administration of the drugs might inhibit the glycolytic metabolism in neurons and glial cells of the hippocampus.

Ces cellules contiennent de nombreuses mitochondries de petite taille à crêtes lamellaires, des ribosomes libres, un reticulum endoplasmique lisse moyennement développé, plusieurs petits dictyosomes formant l'appareil de Golgi, des liposomes et des microtubules. Elles renferment aussi de nombreux corps denses de grande taille qui paraissent être de nature lysosomiale. Le glycogène a été particulièrement étudié. Il est formé de particules β dispersées au hasard dans le hyaloplasme. Des variations saisonnières dans la teneur en glycogène ont été notées. Chez les hypophysectomisés, les cellules de Sertoli contiennent de grandes quantités de ce métabolite dont les particules sont concentrées dans des petites plages, souvent autour des liposomes. Les rôles possibles des cellules de Sertoli sont discutés: soutien et apport de nourriture aux cellules germinales, production d'hormones et phagocytose des corps résiduels. Les variations de la teneur en glycogène sont également discutées.
    Notizen: Summary Sertoli cells of the testis of Lacerta vivipara have been studied electron microscopically in animals obtained between spring and autumn during two years and in animals hypophysectomized in autumn. These cells contain numerous small mitochondria with lamellar cristae, free ribosomes, smooth endoplasmic reticulum moderately developed, several small dictyosomes forming the Golgi complex, lipid droplets and microtubules. There are numerous dense bodies of large size with an heterogeneous content which seem to be of lysosomial nature. Glycogen consists of β particles dispersed at random in the hyaloplasm. Seasonal variations in the content of glycogen are noted. In hypophysectomized animals Sertoli cells contain large amounts of that metabolite whose particles are concentrated in small areas often around the lipid droplets. Possible role of the Sertoli cells concerning mechanical support and nutrition of the germinal cells, production of hormones and phagocytosis of residual bodies are discussed. The variations in the glycogen content are also discussed.

    Notizen: Summary This is an electron microscopic study of non-fixed and non-dehydrated normal human stratum corneum from the lumbar region. Non-stained sections have a low contrast. In sections examined 3 days after skin biopsy the cytoplasm of the cells shows a uniform contrast or exhibits dark and light areas. A single layer delimits the cytoplasm from the intercellular space. The latter is partly filled out with substance. In sections stained 2 to 4 days after skin biopsy the fibrils are distinct. On the basis of the variations in their opacity and ultrastructure three types of horny cells are clearly distinguishable. In cells of type 1 intensely stained keratohyalin and less opaque fibrillar substance occur. A distinct keratin pattern is not found. In cells of type 2 the fibrils show areas with distinct kerytohyalin and keratin pattern and transitional phases between these two stages of fibrillar differentiation. The keratin pattern representing the final stage of the fibrillar differentiation process is visualized through a successive “discoloration” of the filaments, whereas the interfilamentous substance retains the opacity of the keratohyalin. In cells of type 3 the entire fibrillar substance exhibits a keratin pattern. This consists of less opaque filaments with a diameter of 74 Å. The septa representing the interfilamentous substance are estimated as 30 Å at their thinnest points. These observations of the fibrils are completely comparable to the findings in fixed and dehydrated normal human stratum corneum. In sections stained particularly more than 18 days after skin biopsy the fibrils exhibit pronounced changes in their staining properties with concomitant decrease in distinctness or a complete extinction of the keratin pattern. The observations of the modified plasma membrane and the intercellular space in stained sections correspond to the findings in fixed and dehydrated normal human stratum corneum. The modified plasma membrane and the structures in the intercellular space appear with equal distinctness, whether the sections are stained 2 to 4, 6 to 12 or 14 to 21 days after skin biopsy.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die Fasern des roten und langsamen M. soleus von Ratte, Kaninchen und Katze und des roten, jedoch schnellen, M. vocalis des Kaninchens wurden licht- und elektronenmikroskopisch untersucht und mit den verschiedenen Fasertypen aus dem M. tibialis anterior der Ratte und dem M. gastrocnemius des Kaninchens und der Katze verglichen. M. soleus und M. vocalis (einschließlich M. thyreoarytenoideus) enthalten nur einen mitochondrienreichen Fasertyp. Im schnellen M. vocalis ist der Z-Streifen schmal (50–60 nm), das sarcoplasmatische Reticulum ist gut entwickelt. Die Anordnung von Reticulum und Mitochondrion ist ähnlich wie in Herzmuskelzellen. Wie auch in anderen langsamen Muskeln verschiedener Tiere ist im M. soleus der Z-Streifen breit (100–120 nm), Triaden und Reticulum sind selten, und die Filamente bilden unregelmäßige Areale anstelle von Fibrillen. Hierin gleichen die Fasern des M. soleus den (mitochondrienreichen) C-Fasern eines entsprechenden gemischten Muskels; dagegen zeigen die Zwischentyp-(B-)Fasern schmale Z-Linien (50–70 nm), isodiametrische Fibrillen und mehr Triaden als die C-Fasern. Entgegen der bisherigen Vermutung, die auf der histochemischen Zuordnung der SoleusFasern zum Typ B und der Vocalis-Fasern zum Typ C beruht, ist daher anzunehmen, daß die langsamen motorischen Einheiten eines gemischten Muskels aus C- und nicht aus B-Fasern bestehen. In einigen Muskeln sind die Sarcomere der C-Fasern länger als die der B-(und A-) Fasern. Im M. tibialis anterior der Ratte verschwindet der Unterschied von 8,5% bei 2,6 μm Sarcomerlänge bei der Dehnung auf 2,8 μm mittlere Sarcomerlänge; vermutlich weil die Ruhedehnungskurve zunehmend steiler wird. Die isometrische Extraspannung im Tetanus ist bei 120% der Ruhelänge, d.h. bei 2,7 μm Sarcomerlänge. am größten. Daher muß bei 2,6 μm mittlerer Sarcomerlänge die Kraft der C-Fasern die der B-Fasern übertreffen. Rote Muskeln sind besser vaskularisiert als weiße Muskeln. Für die Mm. soleus und gastrocnemius der Katze verhalten sich die Kapillardichten (Kapillaren/mm2 Muskelfaserquerschnitt) wie 2,7∶:1. Dieser Wert entspricht dem Verhältnis zwischen den Größen für die Durchblutung (ml/min × 100 g) in Ruhe und bei maximaler Gefäßerweiterung.
    Notizen: Summary Muscle fibres of the red and slow contracting soleus of rat, rabbit and cat and of the red however fast contracting thyreoarytenoid of rabbit are compared with different fibre types in the anterior tibial muscle of rat and in the gastrocnemius of rabbit and cat. With respect to fibre types soleus and thyreoarytenoid (including m. vocalis) are homogeneous and both being rich in mitochondria. The fast thyreoarytenoid shows a narrow Z-line (50–60 nm) and a well developed sarcoplasmic reticulum. The pattern of reticulum and mitochondria resembles more that of heart muscle cells than of skeletal muscle fibres. Like many slow contracting muscles of different animals the soleus fibres display a wide Z-line (100–120 nm), few triads, little reticulum and irregularly shaped areas of myofilaments instead of fibrils. In that soleus fibres equal fibres of type C (rich in mitochondria) in a corresponding heterogeneous muscle, whereas intermediate (type B) fibres reveal narrow Z-lines (50–70 nm), isodiametrically shaped myofibrils and more triads than C-fibres. Therefore it is far more likely that the slow motor units of a mixed muscle consist of C-fibres than of B-fibres. This is at variance with the histochemical designation of soleus fibres as type B and thyreoarytenoid fibres as type C. In some muscles in C-fibres the sarcomeres are longer than in B-(and A-)fibres. In the anterior tibial muscle of rat this difference is 8.5% at a mean sarcomere length of 2.6 μm, and disappears at a mean length of 2.8 μm, probably due to the steeper slope of the length tension diagram at rest. Since the isometric extratension in a tetanus is highest at 120% resting length (corresponding to about 2.7 μm sarcomere length), the force of C-fibres exceeds that of B-fibres at 2.6 μm but not at 2.8 μm sarcomere length. Red and white muscle differ with respect to vascularisation. The relation between the densities of capillaries in soleus and gastrocnemius of cat is 2.7∶:1 and equals the relation between the blood flows through these muscles during rest and maximum vasodilatation.

This developmental stage is characterized principally by the elaboration of secondary (specific) granules. In addition, there is a modest decrease in cell size, a decrease in the number and mean size of primary (azurophil) granules, a decrease in the number of polysomes, free ribosomes and mitochondria, a depletion of rough endoplasmic reticulum, an increase in cytoplasmic glycogen, an increase in chromatin aggregations and a loss of nucleoli, and the formation of a markedly indented nucleus. The myelocyte stage has been subdivided into three arbitrary phases based upon morphological and functional characteristics which relate to the onset, active production and cessation of secondary granulogenesis.

    Notizen: Summary The fine structure of amniotic and amniotic-plaque epithelia has been studied from normal term pregnancies. The columnar/cuboidal amniotic epithelial cells usually have apical or central nuclei, some free ribosomes, patches of granular endoplasmic reticulum, juxtanuclear Golgi complexes, rod-shaped mitochondria, lipid droplets and some glycogen granules. They have short, blunt microvilli which frequently branch and bathe in the amniotic fluid. The lateral plasma membranes enclose tortuous intercellular spaces which are always interrupted by variously folded processes and desmosomes. The epithelial cells rest on a basal lamina and exhibit highly folded basal processes. The amniotic epithelial cells are neither distinctly Golgi and fibrillar types nor “light” and “dark” in appearance. Amnion from near the umbilical cord contains many microscopic and several large plaques. Similar structures are not found on the reflected amnion. The microscopic plaques are whitish and translucent, whereas the large ones are opaque. The large plaques vary between 1–3 mm in diameter, and are over 15 cell layers thick. Each large plaque has a main central region and edges continuous with either the microscopic plaque or the simple amniotic epithelium. The main region shows four zones, namely, stratum basale, stratum spinosum, stratum granulosum and stratum corneum. Such zones are not distinct at the edges. The fine structure of basal cells compares with the amniotic epithelial cells, but the cells of spinosum and granulosum layers possess variable amounts of tonofibrils, keratohyalin granules, free ribosomes and other cytoplasmic organelles and inclusions. The corneum cells are keratinized and are frequently separated by intercellular spaces. They slough into the amniotic cavity singly or as a sheet, and contribute towards the composition of the amniotic fluid. The plaques are of amniotic origin, and are not formed by adhesion of either squamous cells or fetal skin cells (masses of keratinized squames). The present observations suggest that the occurrence of amniotic plaques is normal. The presence of plaques may not be necessarily associated with fetal abnormality. However, increase in numbers of plaques may be caused by conditions of fluid imbalance. The homology and significance of plaques in eutherian mammals have been discussed.

Characteristic features of all of these tissues are secretory granules measuring approximately 150–250 mμ, a distended endoplasmic reticulum, prominent Golgi regions and large numbers of free ribosomes. Unusual features of the turtle ultimobranchial body are an abundance of large cytoplasmic bodies measuring 800–1000 mμ and a dense, homogenous material within the lumina of the ultimobranchial follicles. The large cytoplasmic bodies usually occur near the luminal portion of the cells and are of similar electron density to the luminal contents, suggesting a possible functional relationship of these two glandular components.

    Notizen: Summary The fine structure of the pterin layer was investigated in both wild type Rana pipiens and Rana pipiens homozygous for the speckle mutant gene. No difference in morphology of the layer was noted between the wild type and mutant. The layer lines the outer surface of the stratum compactum of the dermis and separates this stratum from the stratum spongiosum. The pterin layer consists of extra-cellular material and contains membrane-bounded granules filled with fine spicules. Many of the spicules are somewhat similar in appearance to the initial calcification loci present in developing membrane bone. The layer first appears in the tadpole at approximately stage 14 (Taylor and Kollros, 1946); subsequent developmental stages are described.

    Notizen: Summary Tissues of the young chick and chick embryo were prepared in a relatively unaltered condition by the freeze-fracture technique. The ultrastructure of the microvasculature and surrounding interstitial region is compared with that seen in conventional thin-sectioned material. In the undifferentiated vessels of the 3-day chick embryo, no distinct basement lamina can be distinguished in either type of preparation. In the 3-week chick, a continuous basement lamina is present beneath the endothelium only in chemically fixed and sectioned tissue; it cannot be distinguished from the remaining interstitial substance in freeze-fracture preparations. Blood-tissue exchange may depend on permeability characteristics of the entire interstitial region rather than on the basement lamina alone.

    Notizen: Summary On the basis of the occurrence, at the light microscopic level, of alkaline and acid phosphatases, the pigment epithelium covering the posterior surface of the iris in the albino rabbit can be divided into two zones not previously described, viz. a central zone close to the pupil, approximately corresponding to the area occupied by the iridic sphincter muscle, and a peripheral zone extending to the ciliary body. The central zone which is in intimate relation with the lens was found to have a high content of both phosphatases. At the fine structural level it exhibits a marked pinocytotic activity in the epithelium at the interdigitations between adjacent cells. Electron microscopy revealed that acid phosphatase is localized to the walls of the pinocytotic vesicles. Alkaline phosphatase is in evidence at the surface membrane folds and at microvillous processes between the epithelial cells and the adjoining muscle cells. Unlike the distribution of the acid phosphatase, that of the alkaline phosphatase does not differ fundamentally in the two zones at the fine structural level. In a series of dehydrogenases studied, staining with a view to succinic-, isocitric- and glucose-6-phosphate dehydrogenases revealed an evenly distributed content of enzyme throughout the epithelium. As to the lactic- and β-hydroxybutyric dehydrogenases, contents seem to be lower in the pupillary than in the peripheral zone.

    Notizen: Summary This investigation is concerned with pineal organs of human embryos 60 to 150 days old. At every stage central nerve fibres enter the pineal organ by way of the habenular commissure, but are restricted to the pineal's proximal part. On about the 60th day of the development the sympathetic nervus conarii grows into the distal pole of the pineal organ from a dorso-caudal direction and plays the predominant part in the innervation of the pineal organ. After penetrating, it soon branches out and forms a network in the pineal tissue. Much later, not until the 5th embryonic month, sympathetic nerves appear accompanying the supplying vessels in the perivascular spaces. After a short time these nerves pierce the outer limiting basement membrane and penetrate the parenchyma. Towards the end of the 5th embryonic month the axons of the sympathetic nerves form varicosities containing clear and dense core vesicles. At this point large amounts of laminated granules appear primarily in cell processes, probably of pinealocytes. Isolated granules also occur in the varicosities of axons. The granules encountered here are most likely secretory granules.

Lebenswoche unter Normalbedingungen sowie nach s.c. Injektion von 3 IE ACTH/100 g Körpergewicht innerhalb von 2 min — 24 Std untersucht. Ab der 5. Lebenswoche unterscheiden sich die Nebennieren von Weibchen im Bereich der äußeren Zona fasciculata durch Kernvergrößerung, größere Mitochondrien und kleindisperse Liposomen von denen der Männchen. Zyklusbedingte Schwankungen in der Ultrastruktur der weiblichen Nebennierenrinde bestehen nicht. Nach ACTH-Applikation nähert sich das Nebennierenbild des Männchen innerhalb von 30 min dem des unbehandelten Weibchens. Außerdem kommt es bei beiden Geschlechtern durch ACTH zu einer Dispersion und Reduktion der Liposomen, Vergrößerung des Golgiapparates und der Zellkerne, Ausweitung des endoplasmatischen Retikulums und Vermehrung der Mikrovilli. Der Geschlechtsdimorphismus wird funktionell erklärt.
    Notizen: Summary 178 Sprague-Dawley-rats were studied by electron microscopy from the 1st to the 12th week of life under normal conditions and after s. c. injection of 3 I. U. ACTH/100 gbodyweight from 2 min to 24 hours. Beginning with the 5th week of life females differ from males by small dispersed liposomes and enlarged nuclei and mitochondria in the outer z. fasciculata. There is no change in the ultrastructure of the female adrenal cortex depending on the sexual cycle. After application of ACTH the adrenal cortex of males assimilates to the untreated females within 30 min. After ACTH-application both sexes show dispersion and reduction of liposomes, dilation of endoplasmatic reticulum, increasement of microvilli and enlargement of the Golgi apparatus. The sex-dimorphism is interpreted by functional differences.

    Notizen: Summary Electron-microscopic examination of the human fetal testis between 10 and 20 weeks gestation reveals the presence of two distinct cell types within the tubules: Sertoli cells and germ cells. The latter are distinguished by their spherical shape, smooth nuclear membranes, globular mitochondria and paucity of cytoplasmic organelles. The gonocytes, or primitive germ cells, occur as single cells in the central portions of the tubules. Their chromatin is finely granular and evenly dispersed. Nucleoli are centrally placed and of uniform electron density. Various stages in the migration of gonocytes to the tubular periphery are indicated by the extension of cytoplasmic processes toward the basal lamina. Bands of microtubules are present within the processes. Spermatogonia are arranged in pairs and groups at the tubular periphery. They lack the nucleolar and mitochondrial characteristics of adult spermatogonia. Except for slight changes in chromatin density and nucleolar structure, the fetal spermatogonia retain the ultrastructural characteristics of gonocytes. Intercellular bridges connect adjacent spermatogonia. Degeneration affecting large numbers of germ cells, but primarily gonocytes, begins with nuclear infolding and chromatin condensation and eventually involves both nuclear and cytoplasmic structures. The degenerated cells are removed by phagocytosis by adjacent Sertoli cells. Large phagosomes are present in the cytoplasm of many of the Sertoli cells.

    Notizen: Summary Receptor cells in the epithelium and the basiepithelial nerve net of the prostomium of Lumbricus terrestris were investigated with electron microscope with special regard to the presence of monoamines. The receptor cells are found in groups of about 40 intermingled with supportive cells. After pretreatment with α-methyl-noradrenaline and fixation with potassium permanganate a few receptor cells in each group and some nerve fibres in the basiepithelial nerve net contain small granular vesicles (about400 Å) characteristic for monoaminergic neurons. The distribution and relative number of these receptor cells and nerve fibres coincide well with previous reports on fluorescent receptor cells and varicose fibres. That the monoamine-storing small granular vesicles not are visualized until pretreatment with α-methyl-noradrenaline is in accordance with recent microspectrofluorometric analysis, which shows that dopamine is the only primary monoamine present in the epithelium. In the epithelium there are occasional receptor cells and nerve fibres containing large vesicles (1000–1800 Å) which resemble the neurosecretory vesicles in the central nervous system. Photoreceptor cells having an intracellular cavity with microvilli and cilia have infrequently been observed at the base of the epithelium. No synapses on the mucous cells have been noticed. Nor have any synaptic specializations been observed in the basiepithelial nerve net. The morphological conditions necessary for the existence of possible axo-axonal synapses are briefly discussed.

Die Abgrenzung der Acinusspitze gegen das interacinäre Gewebe hin bildet die Basalmembran des wandständigen Begleitzellepithels. Die Oocyten werden follikelartig von den ineinander verzahnten und durch Desmosomen verknüpften Begleitzellen umgeben. Nur in der Acinuskuppe liegen sie der hier stark verdickten Basalmembran unmittelbar auf. Die Spermatiden sitzen nur mit ihrem anterioren Zellpol den Begleitzellen apikal auf und sind durch Desmosomen mit ihnen verknüpft. Veränderungen der Ultrastruktur der Spermatiden während der Spermiohistogenese werden an drei gegeneinander abgrenzbaren Spermiohistogenesestadien aufgezeigt. Dabei finden die Kernstruktur, das Auftreten von Tubulikörpern und das Abstreifen des Restplasmas vom Mittelstück besondere Beachtung. Den recht uneinheitlich strukturierten Begleitzellen kommen für Oocyten und Spermatiden Ernährungs- und Transportfunktionen zu. Sie phagocytieren überfällige Geschlechtszellen. Es können jedoch trotz ihrer heteromorphen Struktur keine prinzipiell verschiedenen Begleitzelltypen mit jeweils nur einer spezifischen Funktion unterschieden werden. Das in früheren lichtmikroskopischen Arbeiten als Begleitzellprodukt beschriebene „Kinoplasma“ erweist sich als kernwärts wanderndes Restplasma der Spermatiden.
    Notizen: Summary Three species of cells always coexist in the tips of hermaphroditic gonad-acini of fertile Planorbarius corneus: oocytes, spermatids and auxiliary cells. The basement membrane of the auxiliary cell epithelium separates the acinus tips from the interacinary tissue. Like follicles the oocytes are enclosed by interlocked and desmosomically attached auxiliary cells. Only in the utmost tips of the acinus the oocytes are in direct contact with the here dilated basement membrane. The spermatids are attached to the auxiliary cells only with their anterior cell-pole and connected with these by desmosomes. Alterations of the spermatid-ultrastructure during the spermiohistogenesis can be studied in three separate stages of the spermiohistogenesis. Particular attention is given to the nuclear structure, the tubular bodies and the shedding of residual plasma from the middle-piece. The rather irregularly structured cells serve oocytes and spermatids as mediators for nutrition and transport. Occasionally occurs phagocytosis of germ-cells. Basically, even though their structures vary, auxiliary cells are not restricted to one specific function. The “Kinoplasma”-described in previous light microscopic studies as a product of the auxiliary cells, proves to be spermatidic residual plasma moving towards the nucleus.

    Notizen: Summary Umbilical vessels of guinea-pig fetuses were studied shortly before birth. In all umbilical cords investigated an innervation of the umbilical vessels is lacking. The intrafetal parts of the umbilical vessels on the other hand are richly innervated. A marked difference in the amount of nerve fibres and the pattern of innervation is found between artery and vein. The artery is supplied by a dense nerve plexus which spins around the media and which originates from nerve bundles within the outer adventitial layers. The comparatively scanty innervation of the vein exhibits a more coarsely meshed net pattern. The nerve bundles in the vein exhibit a close affinity to the vasa vasorum. Number and type of the close contacts between the muscle cells are different in the various sections of the umbilical vessels. Similar to the distribution of nerves they are almost absent in the vessels of the umbilical cord, numerously, however, in the intrafetal parts. Contrary to the innervation, the close contacts in the vein are developed more numerously and more broadly than in the corresponding artery.

    Notizen: Summary The organization of the cerebral ganglion of the shore crab Carcinus maenas, is investigated by conventional histological and electronmicroscopic techniques. This study forms part of a comprehensive survey of the blood-brain interface, particularly interesting in this group, as decapod Crustacea are unusual among invertebrates in possessing an intracerebral blood supply. Apart from the intracerebral blood vessels, tissue organization is closely similar to that observed in insect central neural ganglia. The ganglion is surrounded by the neural lamella, an acellular connective tissue sheath, probably containing mucopolysaccharide and collagen. A layer of specialised glia, the perineurium, immediately underlies the neural lamella, and appears to contribute to its formation. Large glia occupying a conspicuous cortical zone below the perineurium may be involved in glycogen metabolism and storage. Further morphologically distinct glial types are observed associated with neurones and blood vessels, but all neuroglia within the ganglion are probably of common origin. Neurone cell bodies are generally situated peripherally in groups, and send axons into neuropil (synaptic) areas in the ganglion core. Large lacunae in the cortical region and narrower 20 nm clefts deeper in the ganglion, constitute the interstitial space, and contain deposits of fibrillar material. Possible physiological implications are discussed.

    Notizen: Summary The structure of the cerebral ganglion of the shore crab, Carcinus maenas, is investigated by conventional electron miscroscope techniques, with particular emphasis on the relation of intracerebral blood vessels to other elements in the brain. The ganglion is permeated by a continuous network of channels which may be interpreted as invaginations of the ganglion surface. The afferent vessel (cerebral artery) is of mesodermal origin, but apparently terminates as an open-ended vessel soon after entering the brain, where it runs within the invaginated channels. The greater part of the cerebral vasculature, therefore, has no mesodermal endothelial lining. Tissue components in the diffusion path between blood and brain which could conceivably restrict diffusion, are the thick glial basement membrane, junctions between perivascular and between interstitial glia, and polymeric material in the extracellular space. However, apart from a barrier to large colloidal particles at the basement membrane, the present EM observations do not decisively pinpoint sites of diffusional restriction, nor can they be interpreted as evidence that such restriction exists.

    Notizen: Summary Studies of cardiogenesis in the chick embryo focus attention upon the intercellular junctions of epicardial, myocardial, and endocardial cells, and the role they play in diffusion across the cardiac wall. Cell membranes of apposed epicardial cells approach as close together as 40 Å; those of the endocardium additionally form focal tight junctions. In the myocardium focal tight junctions are restricted to the apposed membranes of the superficial layer of cells. The majority of close appositions in all parts of the myocardium are 40 Å gap junctions. Desmosomes and fascia adherens are distributed throughout the myocardium. Diffusion of horseradish peroxidase through the epicardium and endocardium occurs primarily through the intercellular junctions. The width of the cleft between cells, 200–300 Å, also permits the diffusion between cells of the larger ferritin particles. Pinocytotic activity, responsible for ferritin transfer across mesothelial and endothelial cells in the adult, is not significant. Tracers injected into the pericardial cavity or vasculature can be observed passing through the heart in the direction of their respective diffusion gradients. Unlike the apical junctions of epithelial cells, to which they have been compared, membrane specializations of the superficial myocytes do not form a seal separating the pericardial cavity, or subepicardial space, from the extracellular spaces of the myocardium.

Ein mit dieser Beobachtung völlig übereinstimmender Befund ist unlängst von anderer Seite an Nervenzellen des Nucleus entopeduncularis und der orbitofrontalen Großhirnrinde der Katze erhoben worden. Bei den „dunklen Zisternenfeldern“ dürfte es sich um Bereiche des endoplasmatischen Retikulums handeln, die sich in einem besonderen Funktionszustand befinden. Sie kommen wahrscheinlich schon normalerweise bei verschiedenen Tierarten in bestimmten Regionen des ZNS vor.

The finding corresponds exactly to the unique lamellar configurations recently described in nerve cells of the entopeduncular nucleus and orbitofrontal cortex of the cat. These “dark cisternal fields” are regarded as distinct districts of the endoplasmic reticulum in a special functional state. They seem to occur normally in certain regions of the CNS in different animal species.

Die Tatsache, daß die Häufigkeit heller Vesikel mit fortschreitendem Lebensalter abnimmt, findet ihre Parallele in der von anderer Seite getroffenen Feststellung, daß die Bildung eines humoralen Faktors im Thymus im Laufe der Zeit eingeschränkt wird. 3. Diese Koinzidenz läßt an die Möglichkeit denken, die hellen Vesikel könnten ein morphologisches Äquivalent der Bildung eines Thymuswirkstoffes sein. 4. In entodermalen Thymus-Retikulumzellen 13 Monate alter Ratten finden sich große Vakuolen mit granulärem Inhalt. Ihre Bildung beginnt schon vor der Thymusinvolution. Sie entstehen wahrscheinlich durch Phagozytose oder Aufnahme zelleigenen Materials, das nicht weiter abbaufähig ist. 5. Da diese Einschlüsse mit fortgesetzem Alter an Zahl und Größe zunehmen, scheinen sie als Ausdruck einer Beeinträchtigung der Stoffwechselvorgänge im Zytoplasma ein erstes morphologisch faßbares Symptom der frühzeitigen physiologischen Involution des Thymus zu sein.

These vesicles do not occur in reticulum cells of 13 months old rats. 2. The frequency of these clear vesicles decreases with advancing age obviously in the same way as the production of a humoral thymic factor reported by several investigators. 3. This observation agrees with the hypothesis that the clear vesicles are the morphological equivalent of the production of a humoral thymus factor. 4. Entodermal reticulum cells of 13 months old Wistar rats contain large vacuoles filled with electron dense, granulated materials. The formation of these vacuoles starts already before the beginning of the thymus involution. Possibly they are produced by phagocytosis or necrobiosis and contain deposits of material that cannot undergo further degradation. 5. The increase of these vacuoles in number and size with advancing age may be considered as equivalent of the deterioration of metabolism responsible for early involution of the thymus.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die V. portae der weißen Ratte wurde licht-, fluoreszenz- und elektronenmikroskopisch auf ihre Innervation untersucht. 1. Paraldehydbedampfte Venenpräparate und Häutchenpräparate der gesamten Wandung (Falcksche Fluoreszenzmethode) lassen einen überwiegend längsorientierten äuβeren Nervenplexus erkennen, der den äußersten Muskelzellen aufliegt. Er ist leberseitig weitmaschig, darmseitig sehr engmaschig. Ein subendothelial gelegener innerer Plexus ist vorwiegend zirkulär orientiert. Er entspringt dem äußeren Plexus der darmseitigen Gefäßpartien. 2. Der Nachweis der Acethylcholinesterase (Gomori-Methode) bringt lichtmikroskopisch einige Nervenbündel in der bindegewebigen Adventitia zur Darstellung. Im übrigen findet sich die Aktivität des Enzyms nur in den interzellulären Spalten der Muskelschicht. Der elektronenmikroskopische Nachweis der Acethylcholinesterase (Karnovsky-Methode) läßt aber erkennen, daß sich die Enzymaktivität auf die Muskelzellmembranen beschränkt. 3. Die elektronenmikroskopische Untersuchung bestätigt den fluoreszenzmikroskopischen Befund. a) Lebernah finden sich nur vereinzelte Axonbündel, die der äußeren Muskellage aufgelagert sind. Die Einzelaxone sind vollständig von den Schwannschen Zellen umgeben. Nur wenige, den Muskelzellen benachbarte Axone enthalten agranuläre Vesikel. Sehr selten sind Ausfaltungen der vesikelhaltigen Axone zu sehen, deren Abstand zur Muskelzelle aber immer noch 1000–2000 Å beträgt. b) Auf über eintausend Dünnschnitten wurde kein Axon innerhalb der dicken Muskelschicht gefunden. c) Subendothelial verlaufende Axone (innerer Plexus) sind teilweise oder völlig aus den Schwannschen Zellen ausgefaltet. Sie sind dicht besetzt mit leeren Vesikeln (300–650 Å) und enthalten wenige kernhaltige Vesikel in der Größenordnung 800–1600 Å. Synaptische Endigungen werden nicht beobachtet. d) Eine dichte Häufung vesikelhaltiger Axone, die teils völlig, teils nur an der muskelzellnahen Seite aus den Schwannschen Zellen ausgefaltet sind, finden sich am Übergang der V. mesenterica superior zum Pfortaderstamm, deren einschichtiger Muskellage angelagert. Von diesen Bündeln stammende kleinere Bündel und Einzelaxone ziehen zwischen den Muskelzellen hindurch und erreichen das Endothel. Typische Synapsen werden nicht beobachtet. Kein vesikelhaltiges Axon nähert sich mehr als 1000 Å den Muskelzellen. 4. Die ausgefalteten vesikelbesetzten Axone werden als vegetative Überträgerstrecken angesehen. Die Erregung der Effektorstrukturen durch Transmittersubstanzen wird im Zusammenhang mit der postmortalen autonomen Gefäßkontraktilität diskutiert.
    Notizen: Summary The innervation of the portal vein of the white rat was examined with light-, fluorescence-, and electronmicroscopic techniques. The results are as follows: 1. Paraldehyde treated vein preparations (Falck's fluorescence method) demonstrate a predominantly longitudinally orientated external nerve plexus, being situated on the outermost muscle cells. Near the liver the nerve net is characterized by broad meshes, near the intestinal tract by narrow ones. The circular subendothelial inner plexus originates in the outer plexus of the intestinal vascular bed. 2. Nerve bundles in the fibrous adventitia were demonstrated with Gomori's Acethylcholinesterase method. In other respects, the enzyme activity was only observed in the intercellular spaces of the muscle layer. The electronmicroscopic demonstration of Acetylcholinesterase (Karnovsky's method) further showed that the enzyme activity is restricted to the muscle cell membrane. 3. The electronmicroscopic examination verified the results obtained with fluorescence microscopic techniques. a) In the proximity of the liver, only isolated nerve bundles occur on the outer muscle layer. The individual nerves are entirely surrounded by Schwann cells. Only a few of the axons in the vicinity to the muscle cell have agranular vesicles. Evaginations of the vesicular axons occur infrequently. Their distance from the muscle cell amounts to 1000–2000 Å. b) In more than one thousand thin sections, no axons were found inside the thick muscular layer. c) Subendothelial axons (inner plexus) are either partially or totally evaginated from the Schwann cells. They are densely filled with empty vesicles (350–650 Å) and contain a few dense core vesicles of 800–1600 Å in diameter. Synaptic endings were not observed. d) A dense collection of vesicle-containing axons, that were partially in their entirety and partially only from the muscle cell proximal side evaginated from the Schwann cells, were observed in the single muscle layer at the junction of the superior mesenteric and the portal vein. From these bundles, smaller bundles and individual axons pass between the muscle cells and reach the endothelium. Typical synapses were not observed. No vesiclecontaining axon was nearer than 1000 Å to the muscle cell. 4. Those axons possessing vesicles and being evaginated are considered to be vegetative conducting pathways. The excitation of the effector structures by transmitter substances is discussed in connection with the post mortem autonomic vascular contractility.

    Notizen: Summary The ultrastructural localization of peroxidase activity has been studied in the cells of normal human bone marrow using the diaminobenzidine peroxidase technique. Peroxidase activity has been localized within the primary (azurophil) granules of the neutrophilic series as well as in the cytoplasmic granules of eosinophils, basophils and monocytes. Peroxidase activity appears within the cisternal system (nuclear envelope, Golgi complex and rough endoplasmic reticulum) of these cells during the period of peroxidase-containing lysosome production. With the cessation of granulogenesis, peroxidase activity disappears from the cisternal system and does not reappear in subsequent developmental stages. In cells incubated in peroxide-free media, staining of granular components, but not of cisternae, is reduced. The inclusion of catalase in peroxide-free media eliminates all staining. This indicates that an endogenous peroxide is present within the cisternae and granules of these cell types.

    Notizen: Summary The ultrastructure of the developing chicken liver has been reexamined on specimens fixed by perfusion with glutaraldehyde. This fixation technique gives a preservation of ultrastructural detail superior to that of earlier investigations. Among others the following observations may be pointed out: 1. Bile canaliculi with well-developed microvilli and adjacent tight junctions are present already at the 4-day-old stage and then remain essentially unchanged during development. 2. A subendothelial space of Disse is not present until about 16 days of incubation. 3. The Golgi apparatus does not assume its adult appearance until about 8 days of incubation. 4. Glycogen is first observed in the 6-day-old specimens and then continuously increases throughout development. Glycogen particles often accumulate in membranelimited bodies reminding of the glycogen-filled lysosomes found in a certain type of glycogenosis (Pompe's disease). 5. The mitochondria increase in size and number during development with a conspicuous change from rounded towards more rodshaped and elongated forms.

Das MA ist ein inverses Becherauge. Ein Pigmentbecher aus zwei Pigmentzellen umschließt drei Sehzellgruppen. Die Sehzellen bilden verzweigte Rhabdome vom geschlossenen Typ. Ihr Zytoplasma enthält zahlreiche Vesikel, tubuläre Mitochondrien, kleine Golgiapparate, Mikrotubuli, variable multivesicular und lamellated bodies und Lipideinschlüsse, die von endoplasmatischem Retikulum spiralig umgeben werden. 2. Die Pigmentzellen enthalten dicht gepackte Pigmentkörner, einen gelappten Kern, Mitochondrien vom Cristatyp, wenig endoplasmatisches Retikulum, Mikrotubuli, lamellated und vesicular bodies. Gegenüber den rhahdombildenden Anteilen der Sehzellen ist ihre Zelloberfläche glatt, im übrigen bilden sie lange fingerförmige Ausstülpungen. 3. Artemia salina besitzt zwei Arten von Frontalorganen. Die „dorsalen PO“ sind möglicherweise neurosekretorische X-Organe. Das ventrale FO wird als Sehorgan gedeutet. Es besteht aus zwei Gruppen von Sehzellen ventral vom MA, die eigene Nervenfortsätze zum Protocerebrum senden. Die optische Achse ist der des ventralen Augenbechers entgegengesetzt. Zwischen MA und FO verläuft ein Nerv, der wahrscheinlich dem MA angehört. 4. Übereinstimmungen der Feinstruktur von MA, FO und den Retinulazellen der Komplexaugen betreffen insbesondere die Binnenstruktur der Rhabdommikrovilli und Äquivalente unterschiedlicher Funktionszustände (Hell-Dunkel-Adaptation) hinsichtlich der Ausbildung von perirhabdomalen Vakuolen und des Ausmaßes von pinocytotischen Vorgängen an der Basis der Rhabdome.
    Notizen: Summary The median eye (MA) and the ventral frontal organ (FO) of Artemia salina L. (adult specimens) have been investigated with the electron microscope. 1. The MA is an inverse cup-shaped eye. A pigment cup, consisting of two pigment cells, surrounds three groups of photosensory cells, which form ramified rhabdoms of the closed type. Their cytoplasm contains numerous vesicles, tubular mitochondria, small Golgi fields, microtubules, variable multivesicular and lamellated bodies and lipid inclusions, which are surrounded by spirals of endoplasmic reticulum. 2. The pigment cells contain densely packed pigment granules, an indented nucleus, crested mitochondria, small amounts of endoplasmic reticulum, microtubules, lamellated and vesicular bodies. Opposite the rhabdomeric surface of the visual cells their cellular surface is smooth, otherwise it bears long fingershaped projections. 3. Artemia salina possesses two types of frontal organs. The “dorsal FOs” are possibly neurosecretory X-organs. The ventral FO is interpreted to represent a photosensory organ. It consists of two groups of sensory cells located ventrally of the MA, which possess own nerve-processes leading to the Protocerebrum. Their optical axis is opposite the one of the ventral eye cup. Between MA and FO a nerve occurs, which presumably belongs to the MA. 4. Considerable finestructural similarities between MA, PO and the retinula cells of the compound eyes exist as far as the internal structure of the rhabdomeric microvilli and the equivalents of different functional stages (Light-Dark-Adaption) are concerned, namely perirhabdomeric vacuoles and the degree of pinocytotic processes at the base of the rhabdoms.

    Notizen: Summary Employing a non-ionic detergent as a colloidosmotically active substance 10 fully isolated rat kidneys were perfused for 60 min. A constant glomerular filtration rate (GFR) of 1.0 ml × g−1 × min−1 and a mean Na+-net transport of about 92 μmol × g−1 × min−1 was obtained. The mean O2-consumption of 0.17 ml × g−1 × min−1 was rather high. However, addition of the decoupling agent 2,4-dinitrophenol led to an increase of the O2-consumption by 98%, thus speaking against spontaneous uncoupling. Glomerula, proximal and distal convoluted tubules of kidneys perfused with plasmaexpander-solutions were studied electronmicroscopically. The most striking morphological deviations were found in the proximal tubules of the Haemaccel®-perfused kidneys. In the Pluronic®-kidneys significantly less morphological alterations were found. These morphological changes were of a type which was previously described by other investigators after periods of hypoxia.

    Notizen: The topography and the composition of a surface are in many cases of equal importance (catalysis, electroplating, pretreatment of foils and sheet metal, corrosion, passivation, adsorption, coating of fibers, etc.), and this explains the great interest in methods of investigation that reveal both. If the demands on the resolving power, the analytical possibilities, and the thickness of the surface layer are not too exacting, combined devices like the scanning electron microscope and its analytical accessories can be used. When it is necessary to avoid the compromises involved in simultaneous imaging and analysis, the investigations must be carried out with separate equipment. As an example of a method for the analysis of surfaces we consider briefly photo- and Auger electron spectroscopy (ESCA).
