    Notes: Summary Formaldehyde is a reactive chemical which undergoes spontaneous reactions with various cellular constituents. Mutagenicity data may be interpreted on the background of this behavior. Mice are better able to reduce the irritating effect of formaldehyde than rats and to reduce their ventilation rate when formaldehyde acts on the respiratory tract. Subacute exposure of rats to concentrations higher than 2 ppm inhibits mucociliary clearance of the nasal epithelium and leads to progressive histological and ultrastructural lesions at this site. The occurrence of squamous cell carcinomas of the nasal epithelium of rats after 2 years inhalation of 14.3 ppm formaldehyde (CIIT study) is probably the result of chronic and recurrent local toxicity; this is supported by species differences in susceptibility to the tissue damaging and carcinogenic effect of formaldehyde (rat, mouse, hamster). Data on formaldehyde-DNA interaction further support the argument that a direct risk extrapolation from the formaldehyde effects in rats to those expected for man is not possible.

    Notes: Summary A histochemical assay was routinely performed of myoadenylate deaminase (MAD) in muscle biopsy specimens. MAD was absent in 13 cases, i.e. 2.9% of the specimens. In 10 cases the deficiency was confirmed biochemically. The diagnoses in the 13 patients were: polyneuropathy (n=5), infantile spinal muscular atrophy (n=3), congenital myopathy with type 2 fibre atrophy, facioscapulohumeral myopathy, polymyositis, myotonic dystrophy and hyperornithinaemia with gyrate atrophy of the retina. In contrast, 35 unrelated patients presenting with exercise-related muscle cramps or pains showed normal histochemical MAD activity. The biopsy specimens in all of these patients were essentially normal and in none of them was the diagnosis of a neuromuscular disease made. The results failed to confirm the association of MAD deficiency with aches, cramps and pains or exertional myalgia.

    Notes: Computer programs have been developed or are under development for the IBM personal computer that enable their users to get information on atomic charges, electrostatic potentials, conformational and other properties of molecular systems containing H, C, N, O, F, Si, P, S, or Cl atoms. The zero-order wavefunction is constructed of strictly localized molecular orbitals with fixed atomic orbital coefficients. The wave function can be refined by optimizing these coefficients, i.e., considering inductive effects via a coupled set of 2 × 2 secular equations within the CNDO/2 approximation. Delocalization and exchange effects are accounted for by expanding the wavefunction on a basis of the aforementioned strictly localized orbitals, instead of conventional atomic orbitals, and solving the corresponding SCF equations. Our method has been applied to the study of large systems. We calculated the electrostatic field of the complex of β-trypsin and basic pancreatic trypsin inhibitor and it has been found that strong field regions more or less coincide with hydration sites. A further potential application of protein electrostatic fields is in NMR spectroscopy. We found a linear correlation between CαH or backbone NH proton chemical shifts and the protein field at the site of the corresponding proton. At last, we propose a simple method to mimic the bulk around atomic clusters modeling crystalline and amorphous silicon. Based on this method we found a linear correlation between atomic net charges and bond angle distortions in silicon clusters with 35 atoms.

    Notes: The method of Lagrangian multipliers is used to constrain torsion angles during molecular mechanics refinement for the purpose of plotting strain energy against a reaction coordinate. A complete two-dimensional analysis of the conformational interconversion from δ- to λ-[Co(ethane-1,2-diamine) (NH3)4]3+ reveals a mechanism in which the transition state geometry has an envelope conformation and an inversion barrier of 15.7 kJ mol-1. Substitution at the carbon atoms, variation of the metal-nitrogen distance, and replacement of the amine ligands with bidentate amines only slightly alters the inversion barrier. Substitution at the nitrogen atoms of the bidentate ligand increases the inversion barrier significantly to 24.6 kJ mol-1 for (N,N,N′,N′-tetramethylethane-1,2-diamine) [(NH3)4]3+.

    Notes: We have analyzed two approaches to reproduce the resonance expansion of the scattering matrix appropriate for the calculation of molecular resonance states. The first is based on the resonance theory of Siegert-Humblet-Rosenfeld (SHR) and the second on the Fano-Feshbach formalism. The direct method of calculating the resonance expansion characteristics, devised on the basis of the SHR theory, makes it possible to obtain the energies and partial widths (detailed decay rate constants) of resonances. The Fano-Feshbach formalism, on the other hand, elucidates the resonance state as a concept and facilitates the interpretation of calculation results. The use of computational methods is illustrated by the study of the decay of a model triatomic system and of gas-phase nucleophilic substitution reactions. Used in the latter case is the division of all degrees of freedom of the reacting system into the adiabatic and dynamic ones along with an algorithm of inclusion of the restricted dynamical treatment in the calculation of reaction rate constants.

    Notes: The present article addresses the problem of identifying the structure of a parent compound through its chemical fingerprints such as the various numbers of isomeric substitution patterns, along the lines of Kekulé when he arrived at his benzene formula. In a pioneering paper (1929), Lunn and Senior laid out the conceptual framework for the permutation group description of substitution isomerism. It remained, however, for Pólya's celebrated contribution (1937) to initiate the actual mathematical realization of their vision. Pólya supplied the tools for solving the isomer enumeration problem: given a (symmetrical) parent compound, enumerate its spectrum of substitution patterns. The converse problem, though ranked the more interesting one by Lunn & Senior, hardly received any mathematical attention. The present article offers a complete and effective solution.

    Notes: Stereochemistry deals primarily with distinctions based on rigid geometry, e.g., bond angles and lengths. But some chemical species have molecular graphs (such as knots, catenanes, and nonplanar graphs K5 and K3.3) that reside in space in a topologically nontrivial way. For such molecules there is hope of using topological methods to gain chemical information. Viewing a molecular graph as a topological object in space makes it unrealistically flexible; but if one proves that a certain graph is “topologically chiral” or that two graphs are “topological diastereomers,” then one has ruled out interconversion under any physical conditions for which the molecular graph still makes sense. In this paper, we consider several kinds of topological questions one might ask about graphs in space, methology and results available, and specific topological properties of various molecules.

The broken symmetry solutions for S2N3, S3N3-, and S4N42+ are of planar C2v type with one of the nitrogens stripped of its π electrons, producing a π hole.

    Notes: To study molecular decomposition pathways it is necessary to use ab initio multireference determinant-configuration interaction or MCSCF (multiconfiguration SCF) calculations. The MRD-CI (multireference double excitation-configuration interaction technique of Buenker and Peyerimhoff) calculations on the decomposition pathway of nitrobenzene were carried out using all of the occupied molecular orbitals in the region of the bond being dissociated, plus all of the virtual orbitals. An effective CI Hamiltonian was used into which were folded the effects of all of the occupied molecular orbitals from which excitations were not allowed. So far we have investigated the lowest 1A1, 3A1, 1A2, 3A2, 1B1, 3B1, 1B2, 3B2 states and are investigating the higher states. Our results show a wealth of structure in the potential energy surfaces for the various electronic states of nitrobenzene as a function of distance. A number of the states are predissociative and change dominant configuration one or more times along these potential energy surfaces.

Symp. 11, 347 (1984)] is applied to all consecutive first-order, single-step reactions, and to all reactions possessing one autocatalytic intermediate. The reaction Hamiltonians presented are shown to be consistent with the phenomenological rate equations and the relationship between reaction form and the form of the reaction potential is discussed. In particular, we show: (1) that the interaction between consecutive reactions manifests itself as a coupling term in the reaction potential, a term which may be eliminated via transition to “normal reaction coordinates” for the chemical system; and (2) that coupled sets of autocatalytic reactions give rise to coupling terms in the reaction Hamiltonian which are characteristic of the reaction mechanism.

    Notes: Ab initio electronic structure calculations have been carried out for the He(1S)-Li2 (X̃, 1∑g+) interaction both by the single-configuration SCF and correlated second-order MP2 methods using an extended basis set. From these calculations, an estimate of the isotropic (V0) and first two anisotropic (V2 and V4) terms of the He-Li2 potential surface has been obtained. An assessment of the leading induced-dipole-induced-dipole dispersion energy is presented from the MP2 energies.     Notes: A variety of basis sets have been used for geometric and electronic structure studies. Electronic effects were measured using integrated spatial electron populations (ISEP). The two largest basis sets used, 6-31G* and DZ+P, give significantly different results. Use of two d-orbital sets (6-31G*[dd]) or decontraction of the 2sp shell on phosphorus has little further effect. d-Orbitals on oxygen are required for consistent electronic structure results, and d-orbitals on fluorine have a small but significant effect. Use of diffuse functions, required for anions, is not recommended with small basis sets on neutral molecules. Large negative charges (≈-1.5) on oxygen are given by all of the larger basis sets by the ISEP procedure and indicate that the PO bond in these compounds is largely semi-polar. The best simple symbolic representation of phosphine oxide is H3P+—0-, rather than H3P=0.

    Notes: Basis set expansion and correlation effects on computed hydrogen bond energies of the positive ion complexes AHn · AHn + 1+1, for AHn = NH3, OH2 and FH, have been evaluated. The addition of diffuse functions on nonhydrogen atoms is the single most important enhancement of split-valence plus polarization basis sets for computing hydrogen bond energies. Basis set enhancement effects appear to be additive in these systems. The correlation energy contribution to the stabilization energies of these complexes is significant, with the second order term being the largest term and having a stabilizing effect. The third order term is smaller and of opposite sign, while the fourth order term is smaller yet and stabilizing. As a result, computed MP4 stabilization energies are bracketed by the MP2 and MP3 energies. The overall effect of basis set enhancement is to decrease hydrogen bond energies, whereas the addition of electron correlation increases stabilization energies.

    Notes: With advances in computer architecture and software, Newton methods are becoming not only feasible for large-scale nonlinear optimization problems, but also reliable, fast and efficient. Truncated Newton methods, in particular, are emerging as a versatile subclass. In this article we present a truncated Newton algorithm specifically developed for potential energy minimization. The method is globally convergent with local quadratic convergence. Its key ingredients are: (1) approximation of the Newton direction far away from local minima, (2) solution of the Newton equation iteratively by the linear Conjugate Gradient method, and (3) preconditioning of the Newton equation by the analytic second-derivative components of the “local” chemical interactions: bond length, bond angle and torsional potentials. Relaxation of the required accuracy of the Newton search direction diverts the minimization search away from regions where the function is nonconvex and towards physically interesting regions. The preconditioning strategy significantly accelerates the iterative solution for the Newton search direction, and therefore reduces the computation time for each iteration. With algorithmic variations, the truncated Newton method can be formulated so that storage and computational requirements are comparable to those of the nonlinear Conjugate Gradient method. As the convergence rate of nonlinear Conjugate Gradient methods is linear and performance less predictable, the application of the truncated Newton code to potential energy functions is promising.

    Notes: Techniques from numerical analysis and crystallographic refinement have been combined to produce a variant of the Truncated Newton nonlinear optimization procedure. The new algorithm shows particular promise for potential energy minimization of large molecular systems. Usual implementations of Newton's method require storage space proportional to the number of atoms squared (i.e., O(N2)) and computer time of O(N3). Our suggested implementation of the Truncated Newton technique requires storage of less than O(N1.5) and CPU time of less than O(N2) for structures containing several hundred to a few thousand atoms. The algorithm exhibits quadratic convergence near the minimum and is also very tolerant of poor initial structures. A comparison with existing optimization procedures is detailed for cyclohexane, arachidonic acid, and the small protein crambin. In particular, a structure for crambin (662 atoms) has been refined to an RMS gradient of 3.6 × 10-6 kcal/mol/Å per atom on the MM2 potential energy surface.     Notes: The computer program PRODIS is used to find low energy conformations of flexible molecules by searching the potential energy surface(s) of one or more torsion angles via rigid rotation. The n-dimensional grid of energy versus torsion angles is then converted to a Boltzman probability distribution, with the probability being represented not as a function of torsion angle, but rather a distance between two atoms. These atoms are chosen by comparison with a known, active analogue in which certain atoms have previously been determined as requirements for drug activity. PRODIS produces a list of low energy conformations, their corresponding interatomic distances and the Boltzman probability for each distance ±0.125, as well as the total probability for each conformation. The user also specifies a target interatomic distance and range (usually derived from a more rigid analogue) for which PRODIS lists all conformations and their Boltzman probability that meet this distance.

    Notes: Myelin basic protein (MBP) is the major extrinsic protein of the myelin sheath in the central nervous system. We have examined the predicted structure of segments of MBP using the molecular mechanics program ECEPP83 developed by Scheraga and coworkers as modified by Chuman, Momany, and Schafer. We have focused upon segments containing the Pro-Pro-Pro sequence (residues 100-102), which have been predicted from standard algorithms to exist in a hairpin loop connecting anti-parallel beta-strands. Both the shorter (98-105, 99-105, and 100-105) and longer segments (87-109, 87-118, and 87-120) have been examined. These results indicate potential for a chain reversal in this region. The shorter segments have been studied by others using NMR techniques and the results are compared.

    Notes: The energetic effects of the addition of diffuse functions to molecules with second-row elements are much less dramatic than those for their first-row counterparts. Although diffuse functions on second-row elements have little effect on the geometries and vibrational frequencies of neutral molecules, significant changes are found for anions. While the largest basis set, 6-31 + G*, generally performs best, the results at 3-21 + G* are comparable, and this basis can be recommended for practical applications.

In one option, the intrinsic reaction coordinate has been followed along the path of steepest descent from the transition state backward to reactants and forward to products.     Notes: The additional energy stabilization due to cooperative effects was calculated in extended hydrogen bonded systems O—H⃛O—H⃛O—H with unidirectional (homodromic) orientation of the O—H groups. Ab initio restricted Hartree Fock, MP2 and MP3 calculations with geometry optimization and BSSE correction have been performed using the GAUSSIAN 83 program package for the ground states of the linear water dimer with Cs symmetry and the cyclic water tetramer with S4 symmetry. The latter represents the smallest possible, experimentally observed cooperative structure. A new definition for a cooperativity parameter is proposed. The definition is based on the two-body, non-neighbour interaction energy, plus three- and four-body contributions, including one-body deformation terms in relation to the total interaction energy of the water tetramer. The advantage of this definition is its independence of the reference system, which is necessary in complicated molecular systems with an undefined number of hydrogen bonds, such as disordered or flip-flop systems. According to this definition the energy gain based on cooperativity in the S4 water tetramer is 29% with the MP3/6-31G** approximation, (30% with HF/4-31G* and 46% with HF/3-21G). The largest contribution of 18% is due to the three-body term on the MP3/6-31G** level, followed by the two-body, non-neighbour term with 11%. The four-body term and the deformation term are in the order of 1% and cancel each other because they have opposite sign.

    Notes: The hydrogen-hydrogen nonbonded potential which may be derived from the calculated interactions between hydrogen molecules has been obtained. The best three-parameter Buckingham function gave an RMS error of 0.18 kcal/mol in fitting Price and Stone's ab initio data for 130 pairs of hydrogen molecules, which may be compared with an RMS error of 0.74 kcal/mol using the parameters in the MM2 force field. Burton's basis set is also considered. A better fit to these data requires that the angular relationship between the bonds be included. The data for hydrogen as well as experimental data for chlorine show that these atoms appear “larger” normal to the bond axis than along the axis, and this is probably also the case for other atoms. When simple angular terms are added it is possible to fit the Price and Stone data set with an RMS error of less than 0.06 kcal/mol. The preferred function was: V = [a0 + a1(sin θ1 + sin θ2)4 + a2r]e-3r - [c0 + c1(1 + sinθ1sinθ2)]/r5. Deficiencies in the current ab initio data are discussed.

    Notes: We discuss the three fundamental issues of a computational approach in structure prediction by potential energy minimization, and analyze them for the nucleic acid component deoxyribose. Predicting the conformation of deoxyribose is important not only because of the molecule's central conformational role in the nucleotide backbone, but also because energetic and geometric discrepancies from experimental data have exposed some underlying uncertainties in potential energy calculations. The three fundamental issues examined here are: (i) choice of coordinate system to represent the molecular conformation; (ii) construction of the potential energy function; and (iii) choice of the minimization technique. For our study, we use the following combination. First, the molecular conformation is represented in cartesian coordinate space with the full set of degrees of freedom. This provides an opportunity for comparison with the pseudorotation approximation. Second, the potential energy function is constructed so that all the interactions other than the nonbonded terms are represented by polynomials of the coordinate variables. Third, two powerful Newton methods that are globally and quadratically convergent are implemented: Gill and Murray's Modified Newton method and a Truncated Newton method, specifically developed for potential energy minimization. These strategies have produced the two experimentally-observed structures of deoxyribose with geometric data (bond angles and dihedral angles) in very good agreement with experiment. More generally, the application of these modeling and minimization techniques to potential energy investigations is promising. The use of cartesian variables and polynomial representation of bond length, bond angle and torsional potentials promotes efficient second-derivative computation and, hence, application of Newton methods. The truncated Newton, in particular, is ideally suited for potential energy minimization not only because the storage and computational requirements of Newton methods are made manageable, but also because it contains an important algorithmic adaptive feature: the minimization search is diverted from regions where the function is nonconvex and is directed quickly toward physically interesting regions.

    Notes: The ability of utilization of vegetal oils belongs to common properties of Streptomycetes. During the cultivation of Streptomyces rimosus and biosynthesis of oxytetracycline in 15 m3 fermenters, a fixed regime of soya bean oil feeds as antifoam agent and partial carbon source according to subjectively designed mechanism was investigated and further also feeding regime after exhausting starch as main carbon source and finally his dosing with ammonia sulphate in accordance with physiological activity of the culture. In the second case with semicontinuous supplements of soya bean oil it was possible to prevent premature autolysis of mycelia and to extend the biosynthetic process with keeping the oxytetracycline production rate. The third approach, the most progressive one, represents besides the automatisation of control loop the necessity of assurance continuous pH adjustment for elevation of the process production rate.

    Notes: A comparative response of specific trace elements and organic growth factors for the growth of five Hansenula species (H. anomala, H. beijerinckii, H. ciferrii, H. polymorpha and H. sydowiorum) has been studied. Out of twenty three trace elements tested, Fe, Zn, Mn and Cu were found to be essential for the growth of all yeast species studied here, whereas the rest of the elements exhibited variable essentiality. From fifteen organic growth factors tested, thiamine, biotin, pyridoxine and inositol are the most commonly required growth factors by the yeasts, whereas the rest of the organic growth factors showed variable essentiality. All species of yeasts investigated required different concentrations of trace elements and organic growth factors for their optimum growth. Concentrations higher than the optimum have been found to be inhibitory for the growth of all the yeasts studied.

    Notes: In the fodder yeast production from sulfite spent liquor (SSL) the content of yeast-poisons in the SSL is of particular importance. The negative influence of sulfur dioxide has been known for a long time.Furfural is also named as a pollutant. Our own works showed that there is only an influence on the RS-analysis and in this way a decrease of yield is simulated but not genuine.Practical results and theoretical calculations showed that 1.0 gram of furfural is equivalent to 1.88 g RS.This knowledge is of importance for carrying out the production process by simultaneous change of the furfural concentration. In this way the charge of the fermenter with carbohydrate can be too high or too low.

Yeast entrapped in alginate stabilized by polyethylenimine and glutaraldehyde were unable to hydrolyse whey owing to the inactivation of β-galactosidase by the stabilizing agents.     Notes: A method is introduced which makes a continuous oxidation of glucose to glucose acid possible. This method is based on the auxiliary-substrate concept and co-metabolism, respectively. Micro-organisms (e.g. Acinetobacter calcoaceticus), which cannot assimilate glucose, but merely oxidize it, are grown continuously on a heterotrophic substrate (e.g. acetate). While growing they simultaneously synthesize gluconic acid. The productivity of the gluconic acid synthesis with a given strain depends on the dilution rate and the mixing proportion.Since growth and product synthesis are closely connected and growth yield is very much higher due to an auxiliary substrate effect in the presence of glucose than on the heterotrophic substrate alone, this method is suitable for SCP production as well. The productivity of gluconic acid production is controlled at a certain dilution rate by the mixing proportion of the growth substrate and glucose.

    Notes: A process for biological removal of iron from quartz sands, kaolins and clays was developed in which these industrial minerals were leached at 90°C with lixiviant produced as a result of the cultivation of acid-producing heterotrophic microorganisms, mainly strains of Aspergillus niger, at 30°C in a nutrient medium containing molasses as a source of carbon and energy. The lixiviant, i.e. the fermentation fluid, contained oxalic and citric acids as main components and after the cultivation was acidified to a pH of 0.5 by means of hydrochloric acid. The leaching was carried out in mechanically stirred acid-resistant vats for a period of from 1 to 5 hours. The iron content of some sands treated by this method was lowered from 0.035-0.088 to below 0.012% Fe2O3 making them suitable for the preparation of high quality glass. The iron content of different kaolins was lowered from 0.65-1.49 to 0.44-0.75% Fe2O3 and as a result of this their whiteness was increased from 55-87 to 86-92%. The iron content of a clay was lowered from 6.25 to 1.85% Fe2O3 and this increased the fireproofness of the clay from 1 670 to 1 750°C. Similar process was used for leaching of aluminium from aluminosilicates, mainly clays and kaolins. However, after the cultivation the fermentation fluid was acidified either by means of sulfuric or hydrochloric acid or by means of different mixtures of inorganic acids. For enhancing aluminium solubilization the aluminosilicates were heated before leaching at 600-650°C for 1-2 hours. Over 90% of the aluminium present in different clays and kaolins was leached within 3-6 hours in this way.“Silicate” bacteria related to the species Bacillus circulans and B. mucilaginosus were used to leach silicon from low-grade bauxite ores containing aluminosilicates as impurities. The bacterial action was connected with the formation of mucilaginous capsules consisting of expolysaccharides. The solid residues after leaching were characterized by higher values of alumina content and were suitable for processing by means of the BAYERprocess for recovering aluminium.Heterotrophic bacteria were used to leach manganese from oxide ores using different organic compounds as reducing agents.

    Notes: The object of the presented studies was to determine the conditions of protein biosynthesis from lignocellulosic raw material using filamentous fungi.It was found that on beet pulp + 50% wheat straw, strain A. niger (A.n./61) shows the highest effectivity of protein biosynthesis. Using this strain, after 48-hour incubation the following results were obtained: increase in protein 4.43 g/100 g·DM, daily protein increase coeffizient 2.22 g/100 g DM x 24 h, protein yield (based on the decrease in dry matter) 22.3% and cellulose utilization 37%.Upon use of a new strain CEL 2 and milled wheat straw as the substrate, the best results obtained after 74-hour incubation were as follows: increase in protein 1.82 g/100 g DM, daily protein increase coefficient 0.61 g/100 g DM x 24 h, protein yield 15,8% and cellulose utilization 22.0%.For rape straw and strain Sporotrichum pulverulentum, after 192-hour cultivation the following results were obtained: increase in protein 5.4 g/100 g DM, daily protein increase coefficient 0.68 g/100 g DM x 24 h, protein yield 14% and cellulose utilization 50.2%.

    Notes: The influence of long-term storage on the stability of the cellulolytic enzyme and protein biosynthesis ability, was determined.Microorganisms belonging to genera Aspergillus, Fusarium, Penicillium, Sporotrichum, Allescheria and Mucor were used. The strains were stored on wort agar slants, on SAUNDER'Smedium and on a medium according to MANDELS-WEBER, under paraffin oil or without it. After 3 years storage no influence of the storage method on the biosynthesis of cellulolytic enzymes (strain A. niger 33/2 and 33/20) was found. A systematic increase in the celluloslytic activity was exhibited by strains A.n. 33/2 and A.n. 33/20 on wort agar (40 and 98%, respectively) and on SAUNDER'Smedium (79 and 66%, respectively), as well as by the new strain CEL 2 on Saunder's medium without paraffin oil.

    Notes: By the example of the Penicillin G production, it is shown, how the production process can be improved by using pellet suspension instead of highly viscous filamentous mycel. In order to optimize the pellet geometry, dissolved oxygen concentration profiles were measured, and the overall mass transfer resistance was calculated in the pellets; histological investigations were carried out with pellets also. All measurements indicated that the optimal pellet diameter is 400 μm. When using this pellet, the specific power input was reduced from 4-5 kW/m3 to 0.8 kW/m3. The losses during the recovery of Penicillin G can be reduced below 1% by using reactive extraction. The thermodynamics, kinetics and reaction technique of reactive extraction of Penicillin G are also taken into account.

    Notes: Employing anionic and non-ionic cellulose ethers, differing in type of substituent and degree of substitution, as substrates, the pH-profile of enzyme activity and the parameter Km and Vmax of the MICHAELIS-MENTEN kinetics have been determined with Penicillium citrioviride cellulase in an homogeneous system. Within rather wide limits, a linear correlation was found between the DS of the substrate and Km or Vmax, respectively. Also the pH-profile was found to depend on DS mainly, being bimodal at higher DS. On the other hand, no significant difference was observed between anionic and non-ionic cellulose ethers of about the same DS with regard to the parameters determined. It is assumed, that substrate-enzyme interaction is governed mainly by the length of non-derivatized chain sequences mainly and not by Coulomb interactions.

    Notes: Some original observations have been made on the process of cassava fermentation to produce “foofoo”, a local nigerian diet. During the period of fermentation the pH of the fermenting liquor decreases from 6.1 to 3.4 at the end of the 6th day. The change in pH is uniform throughout the fermentation period. Decreases in dry weight of the fermenting cassava have been recorded; there is a very rapid decline during the third and fourth days of fermentation. Free reducing sugars decrease drastically within the first and second days. Total sugar concentration which is an indication of the starch content of the cassava also declines with fermentation time, and more so during the third and fourth days. Protein concentration in the liquor increases very rapidly during the first and second days of fermentation. It is believed that cassava protein is converted to microbial protein.

    Notes: The specific growth rate of the ethanol producing bacterium Zymomonas mobilis was lower in the presence of oxygen than under anaerobic conditions. Aerobically, considerable amounts of acetaldehyde and acetic acid were formed in addition to the normal fermentation products, ethanol and carbon dioxide. This bacterium contains considerable amounts of pentacyclic triterpenoids, mainly 1, 2, 3, 4-tetrahydroxypentane-29-hopane. It seems that stability and permeability of the cytoplasmic membrane of this rather ethanol tolerant organism is achieved by the hopanoid content. A continuous culture of Zymomonas mobilis produced 60 g/l ethanol over a test period of 39 days. This strain was used for ethanol production from an enzymatically hydrolyzed wheat starch fraction on an industrial scale of 100 m3.

    Notes: The wild strain Aspergillus niger VKMF-2092 forms β-glucosidase on the basis of glucose as the sole carbon source. The formation of β-glucosidase is initiated after the glucose concentration in the medium has reached a low level since the formation is subjected to catabolite repression. The β-glucosidase is mainly cell-bound and only released after a longer period. Under non-repressed conditions the total formation of β-glucosidase is not associated with growth.By supplying glucose in a fed-batch-technique, maintaining a low actual glucose concentration in the medium, the formation of β-glucosidase is enhanced in comparison to simple batch fermentation. Using a fed-batch-technique with a frequently intermittent addition of glucose it is possible to increase the formation of β-glucosidase with regard to both productivity and the activity related to the mycelium and the fermentation broth as well. The increase of productivity is about two to four times greater than at constant feed rate of the same overall amount of glucose.The reason for this increase will be discussed below. A method is presented which permits to investigate the influence of the substrate concentration and other parameters on the enzyme formation in short periods of one and the same fermentation run.

    Notes: The probable effect of increasing levels of ammonium nitrogen on the growth, efficiency of nitrogen fixation, and main cellular constituents of Azotobacter vinelandii was studied under shaking and static culture conditions.The presence of NH4+-N up to 50 mgl-1 level has no harmful effect on the multiplication as well as the yield efficiency ratio of the tested organism. A. vinelandii was able to fix dinitrogen in the presence of NH4+-N when both nitrogen sources were available in the culturing medium. The efficiency of nitrogen fixation was affected by the initial presence of NH4+-N in the medium, it was quite low at the highest level. The crude protein efficiency ratio was correlated inversely with the initial NH4+-N concentration, whereas the total carbohydrate efficiency ratio as well as the total lipid efficiency ratio were positively correlated with the NH4+-N concentration. The presence of NH4+-N in the culturing medium has no essential influence on the qualitative composition of the amino acids in the Azotobacter cells.

    Notes: Growing and resting cells of microorganisms are able to accumulate metal ions.These reactions are based on a storage within the cells as well as sorption at the cell wall.The intracellular storage takes place preferably by growing cells.The stored amounts of metals by resting cells depend on the concentration of metal ions in the aqueous phase, the pH-value, and, in some cases on the temperature.Maximum concentrations for some metals and strains of microorganisms are given.The representation of the uptake reaction as an adsorption process using a FREUNDLICH-Isotherm demonstrates straight lines with different clopes.The removal of the metals from the biomass by a desorption reaction is possible in some cases, other possibilities are a reducing process or the formation of an ash of the biomss containing compounds of metals.These reactions may be used simultaneously for detoxification and removal of metals from waste water.

    Notes: In the commonly used sulfite method the consumption of sulfite is determined by iodometry. Since however, the addition of organic substances may interfere with iodometry (e.g. due to chemical reactions with iodine) the gasometric measurement of sulfite oxidation has been developed for analysis of how different culture media may influence the oxygen transfer rate. The striking decrease of sulfite oxidation rate due to addition of culture media to the sulfite solution suggests that adsorption of orgnic components in the gas liquid interface may account for an additional diffusion barrier and thus for a decrease of the oxygen transfer coefficient which in addition gives an explanation for differences between values found by the sulfite method and by aerobic cultivations. Consequently identical values of oxygen transfer rate have been obtained for both systems whenever the sulfite system has been properly adjusted to the aerobic cultivation conditions. In so far, the gasometric sulfite method proved to be a unique tool for rapid determination of factors influencing oxygen transfer rate in fermentation processes which may give rise to a reappraisal as to the relevance of the sulfite method for oxygen transfer optimization.

    Notes: Cells of Chlorella vulgaris, BEIJ. Greifswald 9, were grown on autotrophic and mixotrophic conditions using glucose and acetate as organic substrates. It was shown that these C-sources applicated in a suitable range of concentrations increase the growth rate and the productivity of the algal cultures. The cells grown on mixotrophic conditions have a higher total pigment content and exhibit variations in the ratio chlorophyll a/chlorophyll b.In addition the contents of proteins, lipids, carbohydrates, and nucleic acids of the biomass were shown to be dependent on the kind of the organic substrate used.

    Notes: Cassava waste peels may constitute up to 55% of the original tuber. These waste peels were found to contain 41.8% carbohydrate, 1.1% protein, 12.5% ether extract and 4.9%, 4.9% total ash. and 20.8% crude fibre. Studies were conducted to formulate a fermentation medium to convert the waste peels to reducing sugars and to enrich the peels with microbial protein. Amylase producing microorganisms were isolated from rotten cassava tuber discs buried in the soil at different locations. The microorganisms isolated were Aspergillus fumigatus, A. flavus, A. niger, and a Pseudomonas sp. and A. niger; the level of reducing sugar was 20.5 mg/ml. The lowest was by B. subtilis an isolate from fermenting locust bean. Generally the levels of saccharification were higher when the waste media were supplemented with different nitrogen sourses. The crude protein yield in the cassava peel waste media by different microorganisms varied from 5.6% to 17.5%. The highest protein yield was in the waste medium fermented by A. fumigatus followed by A. niger, B. subtilis, Pseudomonas sp. in decreasing order.

    Notes: Candida lipolytica was cultured on a methanol containing medium as the only C source. The influence of different concentrations of methanol and ammonium sulphate and the effect of addition of some biological active materials to the methanol containing medium were studied.Dry cell yield of C. lipolytica and protein content were determined to be 23.1 g of cells/100 g of methanol added and 40.2%, respectively. Total lipids, phospholipids, keto acid and amino acid composition were estimated. The amino acid composition of the protein was comparable to the composition of the other reported single cell proteins (SCP). These results indicate that C. lipolytica is potentially important for fodder use.

    Notes: The review characterizes the submerged and solid state fermentation processes (SF and SSF) for obtaining of microbial biomass protein (MBP) on cellulose- and lignin-containing (CL) agricultural wastes (straw, wine, branches of fruit trees). There are discussed other alternatives as well. The main technological parametres are presented (protein (biomass) yield, degree of available substrate utilization, process efficiency, etc.). The use of a special stirring system of the authors' design for mycelial cultures results in an increase of CL substrate concentration in the initial nutrient medium from 2% to 8%of dry matter (DM) and in a threefold increase of the specific productivity (from 0.07 g biomass/l · h to 0.21 g biomass/l · h). The technological parametres can be increased also in the processes with substrate addition. The obtained preparations are analyzed as to their usefulness for the feeds of ruminants.

    Notes: When glucose is used as C-source extracelluar citric-acid accumulation does not start at the point of N-source exhaustion. A transition phase lasting for about 2-3 hours is observed. It may be assumed that the continuation of growth after the N-source exhaustion coincides with the delay of the citric-acid accumulation. During the course of citric-acid production characteristic interruptions of the glucose uptake and the citric-acid accumulation are found. The findings are discussed in more detail.

The cell shape was heterogenic at growth rates from 0.05-0.8 h-1. In some cases the cell size varied depending on the growth rate, evidently due to osmotic phenomena which do not pertain to the growth proper.

    Notes: Effects of exogenous applied lipids being of microbial origin on the water use efficiency (WUE) and the biomass production have been studied in pot and field experiments using crop plants.Lipid mixtures extracted from the cells of Candida maltosa EH 15 increased the WUE (9…16%) and the grain yield (÷x = 10%) of the wheat and barley plants under dry conditions. The fatty acid fraction separated from the lipid mixtures also improved the WUE especially under stress (20…100%) and the biomass production. The phosphatide fraction emulsified with sulfonate containing solvents was not physiologically active.
