    Notes: Summary Seven cases of dog islet cell carcinomas were studied by conventional and immunohistochemical light- and electron-microscopy. Antisera to insulin, pancreatic polypeptide, somatostatin and glucagon were used. In 6 tumours several hormones were demonstrated. Glucagon never occurred. Insulin was the only hormone present in every tumour, thus it seems to be a good marker for these neoplasmas. Liver metastases contained less immunoreactive cells than primary tumours and cell types found in primary carcinomas were sometimes not present in liver metastases. In two cases a degenerative neuropathy occurred.

    Notes: Summary Lymph node biopsies from 24 male heroin addicts and 9 control patients were studied using immunohistochemical, and in $$\tfrac{1}{3}$$ of cases, quantitative methods. 5 out of these 24 patients were also homosexual. All presented diffuse lymphadenopathy but none had any signs or symptoms of opportunistic infections nor Kaposi's sarcoma. Histologically the lymph nodes showed a very peculiar follicular hyperplasia with atrophy of the paracortex. The germinal centers appeared irregular, ill defined fined and contained clusters of small lymphocytes. The mantle zone was atrophic. Immunolabelling of T lymphocytes by monoclonal antibodies showed that germinal centers were invaded by small nests of Leu 2a (and OKT8) positive lymphocytes, i.e. chiefly cytotoxic-suppressor phenotype; the number of these cells increased by about 100 times in the follicles. OKT4 (and Leu 3a) positive cells, i.e. chiefly helper-inducer phenotype, appeared to decrease. These histological and immunohistochemical changes are considered to be suggestive of drug addicts' lymphadenopathy, and also possibly of other conditions increasing the risk of AIDS.

    Notes: Summary Results of cell kinetic analyses on transurethrally obtained material from urinary bladder are compared with parallel immunohistochemical tests on carcinoembryonic antigen (CEA) and tissue polypeptide antigen (TPA), performed on the same material. Labelling index increases from 1.4% in slight to 20% in marked urothelial atypia. CEA reaction in slight atypia is slight or moderate, slight, moderate or distinct in atypia, and moderate to distinct in carcinoma in situ. TPA always shows moderate to distinct reactions. Cell kinetically, urothelial carcinomas mas yield similar gradations. They were positive for CEA in 70% and for TPA in 100%. In G0 and GI carcinomas, negative and slightly positive reactions predominate, poorly differentiated lesions yield predominantly distinct reactions. In all grades, TPA ranges from slight to distinctly positive. As in cell kinetic analyses, there is a relationship between differentiation grade and stage for CEA expression. This does not apply for TPA. The results permit us to draw conclusions on the different biological and histogenetical behavior of urothelial carcinomas. There are undoubtedly differences in the behavior of papillary-exophytical and solid invasive carcinomas in terms of both cell kinetics and immunohistochemistry.

    Notes: Summary Seventeen cases of malignant peripheral neuroectodermal tumour (MPNT) were studied by means of light microscopy, immunohistochemistry and electron microscopy. There were nine males and eight females. The mean age of the 17 patients was 10 years with a range of seven months to 20 years. The vast majority of tumours was located in the trunk. Histologically, they closely resembled Ewing's sarcoma, although minor differences were obvious. Special findings included ganglion cells and Flexner rosettes. In 10/11 cases positive staining for neuron-specific enolase (NSE) was obtained. Five of 10 tumours were positive for protein S-100. Three contained vimentin, two neurofilaments and one vimentin, neurofilaments and GFAP. Neurosecretory granules were noted in the three cases studied. Five patients died, three are alive with disease and five patients are alive without evidence of disease. It is concluded that these tumours form a homogeneous group, although the grade of differentiation varies. The prognosis in most cases is poor. Distinction from Ewing's sarcoma is possible by staining for NSE and by electron microscopy.

    Notes: Summary To elucidate the histogenesis of sclerosis haemangioma of the lung, we examined 7 cases with the immunoperoxidase method using antibodies against several useful marker antigens; secretory component (SC), cytokeratins, epithelial membrane antigen (EMA) for epithelial cells, factor VIII related antigens (factor VIII) for endothelial cells, vimentin and desmin for mesenchymal cells. The results were compared with those of histologically normal lung tissues. Both the characteristic round cells arranged in sheets, which are present predominantly in the solid area and are reported to be neoplastic, and the flattened cells lining blood lakes show positive staining for EMA only, with negative staining for the other marker antigens. These observations suggest that these cells are derived from epithelium rather than mesothelium or from endothelium, and are analogous to type I pneumocytes. This conclusion is supported by their immunohistochemical characteristics, in comparison with the localization patterns of the marker antigens in normal lung tissues. However, the lining epithelial cells of papillary projections in the papillary area and of ducts in the solid area stained for SC and cytokeratins as well as EMA, and their immunohistochemical characteristics are analogous to those of bronchiolar epithelial cells or type II pneumocytes in normal lung tissues.

    Notes: Summary By immunohistochemistry, the presence of lactoferrin was investigated in follicular adenomas (10 cases) and carcinomas of the thyroid gland (23 cases). Normal thyroid tissue was also tested as control. Follicular adenomas showed a consistant negativity, whereas follicular and papillary carcinomas exhibited various degrees of positivity for lactoferrin. Incorporated organoid structures observed in anaplastic carcinomas were strongly stained; the spindle cell parts of these cancers were always negative for this iron-binding protein. Medullary carcinomas were also unstained. These findings are discussed in relation to the distribution pattern of thyroglobulin. The authors emphasize the possibility that lactoferrin may be useful in clarifying some diagnostic problems in neoplastic thyroid pathology.

    Notes: Summary Immunohistochemical localization of theN-terminal fragment (1–76) (NTF) of human pro-opiomelanocortin (POMC) was studied in human adult and fetal pituitary glands, as well as in pituitary adenomas associated with Cushing's syndrome and in ectopic ACTH-producing tumors. Comparison of localization between NTF and ACTH was performed using mirror sections. Our results indicated concomitant localization of NTF and ACTH in the same cells, not only in normal adult and fetal pituitaries but also in pituitary adenomas and ectopic ACTH producing tumours. Specificity of the NTF staining was confirmed by immunoabsorption. Negative staining of the bovine pituitary gland indicated the immunohistochemical localization ofN-terminal (1–45) of human POMC as there is a known species difference in the sequence 1–45 between human and the bovineN-terminal fragment. Presence of NTF in cisterna of rough endoplasmic reticulum indicates its production by small cell carcinoma. These findings, together with the previous studies, suggest that the complete form of POMC is produced in the tumours as well as in normal pituitaries.

    Notes: Summary The microvasculature of lymph nodes of 55 cases of T-cell lymphoma was studied by light microscopy, immunohistochemistry and electron microscopy. A modified peroxidase-antiperoxidase (PAP) method was used for staining paraffin sections with lectin I of Ulex europaeus (UEA-I), which is a specific marker for vascular endothelial cells. The T-cell nature of each case was proven by immunohistochemistry, including immunoperoxidase staining of frozen sections with monoclonal T-cell antibodies. The cases were subclassified according to previously established criteria, but with the addition of a separate group showing a high content of clear cells. For the purpose of the present study, the small blood vessels were separated into two main variants, viz.: high endothelial venules (HEV) and all other types of vessels with flat endothelium (SVFE). The development of each of these variants and the extent of lymphocyte migration through the vascular wall were assessed semiquantitatively. The findings suggest that the blood microvasculature, as a whole, is similar in all types of T-cell lymphoma. There were distinct differences, however, in the development of the two main categories of small vessels between the various types. Chronic lymphocytic leukaemia of T-type (T-CLL) and Sézary's syndrome were poor in SVFE and rich in HEV, and there was considerable lymphocyte traffic through the latter. In contrast, T-immunoblastic and especially T-lymphoblastic lymphomas showed numerous SVFE, only a few or no HEV and minimal lymphocyte traffic. The appearance of the microvasculature varied markedly in the various subtypes of “pleomorphic T-cell lymphoma”. In the small cell subtype HEV predominated and SVFE represented only a small or moderate fraction of the microvasculature.     Notes: Summary About 200 human tumours and corresponding normal tissue samples were investigated by immunoperoxidase tests for the expression of MAM-3, MAM-5 and MAM-6 antigens, which had previously been defined by monoclonal antibodies to human milk fat globule membranes. All tissue specimens had been treated for routine histopathology, i.e. fixed in formalin and embedded in paraffin. One of the antigens, MAM-6, appeared to be an important epithelial marker, present in all normal and neoplastic breast tissue samples, in about 80% of non-mammary normal tissues and in more than 90% of non-mammary epithelial tumours. It could never be detected in normal and neoplastic mesenchymal and neuroectodermal structures. Direct comparison with the distribution of Carcinoembryonic Antigen (CEA), Tissue Polypeptide Antigen (TPA) and keratin provided clear evidence that MAM-6 is different from these well known epithelial markers. MAM-3 proved to be an additional important marker exhibiting a characteristic distribution pattern in those epithelial tissues investigated. In contrast to MAM-6, it could never be detected in renal cell cancers and carcinomas of the prostate gland, thus allowing differential diagnosis on the basis of immunohistochemistry. MAM-5, known to be associated with lactoferrin, was mainly detectable in secretory organs and their tumours. In the group of breast tumours, its expression was mainly seen in lobular cancers. These findings suggest a use for these new markers for routine histopathology.

    Notes: Summary Skin biopsies from 34 patients, presenting with a variety of bullous skin disorders were investigated, using routine light microscopy and immunohistochemistry. In bullous skin diseases characterized by deposition of complement factors (CF) and/or immunoglobulins (Ig), a monolayer of OKIa 1 + , OKT 6 - , OKM 1 - dendritic cells was found at the dermo-epidermal junction. Retrospectively, these cells were easily recognized on paraffin embedded, H & E stained material. In bullous skin disorders, showing no deposition of CF and/or Ig, this monolayer of dendritic cells was lacking. It is suggested that these OKIa 1 + , OKT 6 - , OKM 1 - dendritic cells at the dermo-epidermal junction represent some type of antigen presenting cells, not corresponding to Langerhans cells, veiled cells or indeterminate cells.

    Notes: Summary An immunohistochemical investigation of vimentin, an intermediate filament, was carried out on formalin fixed paraffin embedded sections of 44 malignant mesotheliomas of the pleura and 24 pulmonary adenocarcinomas, in order to assess its value in differential diagnosis. Seventy-five percent of the malignant mesotheliomas showed positive staining for vimentin. An unexpected finding was the presence of vimentin tin in 46% of the pulmonary adenocarcinomas. In either case there was no correlation between the presence of vimentin and the histological types or grades of differentiation. The overall level of vimentin staining was significantly greater in the malignant mesotheliomas than in the pulmonary adenocarcinomas but no single antibody dilution was found to provide clear cut separation of the two groups. Vimentin does not appear to be a simple discriminatory marker of malignant mesothelioma.

    Notes: Summary Seven large bowel carcinomas were examined by light and electron microscopy for the presence of five oncofoetal antigens. Ultrastructural investigations involved a novel method whereby thick sections of gluteraldehyde-fixed material were cut on a vibratome and then labelled using slight modifications of a standard unlabelled antibody-enzyme (PAP) technique, before further processing. Ultrastructural preservation, staining properties and the retention of antigen activity was seemingly better than that achieved by other investigators. Specific, positive labelling for carcinoembryonic antigen (CEA), colon specific antigen (CSA) and pregnancy-specificβ-1-glycoprotein (SP1) was seen in every case. Clear positive labelling for placental alkaline phosphatase (PLAP) and human chorionic gonadotropin (HCG) was seen in two cases. Extracellular labelling was found in areas of cell debris, free lying or in phagocytic cells and on tumour cell brush borders. The pattern of intracellular labelling, however, was different for each antigen and reflected the probable sites of synthesis and release from the cells. Thus CEA, a complex glycoprotein, was localised within the golgi apparatus, small apical cytoplasmic vesicles and mucous droplets in relatively well differentiated tumour cells. CSA, a chemically related glycoprotein, had a similar, but less dense distribution. SP1, by contrast, was localised within basally-located vesicles associated with the ribosomal endoplasmic reticulum and appeared to be released and persist as debris or taken up by phagocytic cells below the basal lamina. PLAP and HCG, both proteins, were found within simple single membrane-bound vesicles within relatively undifferentiated cells.

    Notes: Summary A mouse monoclonal antibody (mAB lu-5) was prepared using a lung cancer cell line as an antigen. The selected clone produces an IgG with a gamma-1 heavy chain and a kappa-light-chain. Immunohistochemical testing of mAB lu-5 on 117 normal tissue biopsies and 474 tumours revealed reactivity with an intracytoplasmic, formaldehyderesistant antigen present in most epithelial and mesothelial cells, but absent in mesenchymal cells. The antibody can therefore be used as a first order, pan-epithelial marker. It proved also useful for fast tumour diagnosis on frozen sections.

    Notes: Summary Peripheral neuronal tumours were studied by the peroxidase-antiperoxidase (PAP) method for the presence of the neurofilament protein (NFP) and neuron-specific enolase (NSE). All cases of ganglioneuromas and ganglioneuroblastomas were positive for NFP and NSE. Both markers were observed only in tumour cells showing differentiation towards ganglion cells. Of the 14 cases of neuroblastoma, 8 were positive for NFP and 12 were positive for NSE. NSE was detected in most neuroblastic tumour cells. However, NFP was found in neuroblasts with signs of differentiation, such as nuclear enlargement, but not in immature, small round cells. NFP was present in cell bodies as well as in cytoplasmic processes of partially differentiated neuroblasts. The majority of pseudorosettes showed no NFP stain. Thus, antibodies against both NFP and NSE are useful in the diagnosis of peripheral neuronal tumours. Moreover, the presence of NFP seemed to be related to the degree of tumour cell differentiation.

    Notes: Summary Immunohistochemical identification of αl-antitrypsin (αl-AT) and αl-antichymotrypsin (αl-ACh) in pleomorphic adenomas of salivary glands is reported in order to compare their distribution profiles with those of lysozyme and lactoferrin, already described elsewhere. Normal salivary glands indicated positive αl-AT staining in ductal segments and had no αl-ACh in any glandular cell. Pleomorphic adenomas displayed moderate positivity to αl-AT staining in duct-like, tubular and glandular epithelia which was particularly intense in luminal cells. The limited number of tumour cells which showed duct-like structures with a single cellular layer arrangement, displayed the highest staining to αl-ACh. Strongly αl-AT positive tumour cells located on the inner side of luminal cavities were also markedly positive to αl-ACh. Spindle shaped tumour cells existed outside tubular and ductal structures and were negative to αl-AT and αl-ACh. Distribution of αl-AT in salivary glands was similar to that of lysozyme as is usual in ductal segments or their transformed cells, and occurrence of αl-ACh localization rather resembled that of lactoferrin, with occurrence in acinar compartments and changed epithelia within acini. The biological role of a specific immunohistochemical distribution of αl-AT and αl-ACh in pleomorphic adenomas may be associated with a self regulating mechanism which inhibits degradation by tissue proteinases.

    Notes: Summary Tumour tissue from a lung cancer patient who showed elevated serum amylase and adrenocorticotropin (ACTH) was studied ultrastructurally, immunohistochemically and biochemically. Histologically the tumour was a small cell carcinoma. On electron microscopic examination the tumour cells contained large zymogen-like granules within the cytoplasm. Furthermore, cells which possessed many small dense core granules of the endocrine type were also observed. It was of interest that the large zymogen-like granule-containing tumour cells had microvilli at the apical border, connected by desmosomes and forming lumina showing adenocarcinomatous differentiation. Electrophoretic analysis of the serum revealed that the major elevated amylase was of the salivary type with minor components. Immunostaining clearly demonstrated that most of the tumour cells possessed immunoreactive ACTH, whereas salivary amylase was only found in occasional clusters of the tumour cells. The results seem to indicate that the tumour showed both endocrine and exocrine characteristics - an amphicrine carcinoma, expressing amylase and ACTH simultaneously.

    Notes: Summary 10 pleomorphic adenomas of the human parotid gland were transplanted on several groups of nude mice. For comparative reasons, 10 other pleomorphic adenomas, a neurinoma and a chordoma and transplants of squamous cell carcinomas and of normal salivary gland tissue were also analysed. In the primary tumours and in the transplants, the presence of keratin, carcinoembryonic antigen, tissue polypeptide antigen, lactoferrin, lysozyme, immunoglobulins, secretory component, amylase, fibronectin and of several lectin-receptors (PNA, WGA, HPA, Ulex europaeus) was sought. The immunohistological observations show that many of the features of a pleomorphic adenoma are constant under the conditions of transplantation. In the transplanted tumour, the same heterogeneity as in the primary tumours can be observed. Autoradiographic studies show little labelling with 3-H thymidine, which is in good accordance with the biological behaviour of the tumour. The distribution of fibronectin shows an interesting association with myoepithelial-like cells. Our results support the hypothesis that the histogenetic origin of the pleomorphic adenoma is a cell pool of the terminal ductal segment. A differentiation towards ductal cells (with production of secretory substances) and towards myoepithelial cells (associated with large amounts of basal membrane like substances) is observed.

    Notes: Summary We describe four lymph node based tumours in which numerous neoplastic cells and some mitotic figures were characterized by staining affinity for Lectin I of Ulex europaeus (UEA-I). The patients had no vascular or epithelial tumours and presented symptoms suggestive of a systemic lymphoproliferative disease. Histologically, the tumours were composed of large, cohesive, cells which were mainly located in the paracortex. UEA-I reactivity was more evident in the Golgi area and was present in large mononucleated cells often arranged to delimit vascular-like spaces. The neoplastic cells were weakly muramidase-positive in one case, and were ANAE+/AP + in two other cases. Large dots of UEA-I reactivity were detected in S-100+/muramidase-negative Langerhans-like cells present in one case of Letterer-Siwe disease. UEA-I staining was consistently negative in 20 cases of B cell- or T cell lymphoma and in 9 other cases of histiocytic lymphoma. It is suggested that UEA-I+ tumours of the lymph nodes are part of a distinct subset of histiocytic malignancies whose neoplastic cells present some morphological and phenotypic properties normally associated with endothelial cells.

    Notes: Summary Keratin was identified with the aid of polyclonal antisera in the cytoplasm in over 90% of the transitional cell carcinomas investigated. The intensity of staining increased with the degree of dedifferentiation. Detection of cytokeratin with monoclonal antibodies was successful in over 80% of samples. All squamous cell carcinomas of the bladder were strongly positive for keratin and cytokeratin. CEA was found in 20% of the G1 and 40% of the G2 and G3 carcinomas of the bladder. Both the prostatic epithelium markers PSA and PAP and the monoclonal antibody Ca1 were negative in all cases.

    Notes: Summary Dopamine-β-hydroxylase (DBH) immunohistochemistry was used to demonstrate the noradrenergic fibers and terminals in the anterior column of the rat lumbosacral spinal segments. PAP-positive varicose fibers were widely distributed in the gray matter with preferential accumulation in the nuclear regions containing motoneurons involved in the contraction of perineal striated muscles. Unmyelinated DBH fibers were composed of nodular enlargements (varicosities, 0.4–3.0 μm in diameter) and very fine, short intervals (intervaricose segments, 0.1–0.2 μm in diameter and 1.0–4.0 μm in length). DBH-positive dense products were electron microscopically often confined within small granular particles and less frequently within large granules. Additionally, in order to characterize the innervation pattern of noradrenergic fibers on dendritic bundles organized in the motoneuronal pools innervating the pelvic small muscles, semi-quantitative analysis was done in the area of the dorsolateral nucleus endowed with especially well-developed dendritic bundles. DBH terminals contacting with unreactive dendrites were more common (67.9%) than those with neuronal somata (15.1%), and the remainder (17%) had no contacts with surrounding neuronal elements. Furthermore, specialized synaptic formations were observed in only 20.1% of these nodules. The results suggest that bulbospinal descending noradrenergic neuron systems influence the functioning of pelvic muscles principally via the neuronal contacts with dendritic bundles in the spinal cord.

    Notes: Summary The objective of the present immunohistochemical study was to determine whether the close spatial relationship between hormone-producing cells as described in rats also exists in the mouse adenohypophysis. In both immature and mature mice, GH cells were the only cell type that had a round shape throughout the pars distalis. All other types of secretory cells had angular or irregular shapes and were closely apposed to round GH cells. Thus, between GH and ACTH cells the same intimate relationship pertains as in rats. Unlike in rats, however, the juxtaposition of LH and Prl cells was observed only occasionally in mature female mice. The salient features of the mouse adenohypophysis were that most LH cells closely surrounded GH cells. These findings show that the cytoarchitectural interrelationship between adenohypophysial cells of mice differs from that of rats.

    Notes: Summary The distribution of serotonin-containing nerve fibers in the corpus striatum of the rat, cat and monkey was studied with modified peroxidase-antiperoxidase method using serotonin antiserum without any pretreatment. In the neostriatum (caudate nucleus and putamen) of all mammalian species investigated, the immunoreactive fibers were distinct varicose fibers in a fine network. The concentration of these fibers was high in the ventral, medial and caudal neostriatum. Especially in the area bounded by the globus pallidus, serotonin fibers were abundant and compactly arranged along the nucleus. In this area of the monkey, a few thick fibers (tract fibers) were intermingled; they ran along the lateral medullary lamina. Such tract fibers were also observed outside the medial medullary lamina and in the central portion of the medial pallidal segment. The paleostriatum (globus pallidus and entopeduncular nucleus) of the rat and cat as well as the medial pallidal segment of the monkey was diffusely innervated with serotonin fibers composed of numerous varicosities and fine intervaricose segments, while in the lateral pallidal segment of the monkey, the distribution of fibers was scantly and partial. Our results hint at the morphological basis of the serotonergic regulation of the extrapyramidal system in mammals.

    Notes: Summary The distribution of substance P (SP) immunoreactive nerve cell bodies and preterminal processes was studied in the rat brain by using several anti-SP-antibodies in combination with immunohistochemical techniques. In normal rats and in rats pretreated with colchicine, SP immunoreactive preterminal processes were found in the hippocampal region, but SP positive cellbodies could be detected only after colchicine pretreatment. Medium-sized to large, multipolar cells immunoreactive for SP were found in stratum oriens of the hippocampal subfield CA3 and in the hilus of the area dentata. Medium-sized to small, round or fusiform cells were detected in the pyramidal layer of the ventral subiculum and in layers III–VI of the ventral entorhinal area. The SP stained preterminal processes were of two types. Numerous fine, varicose axons were stained in different parts of Ammon's horn, while in the retrohippocampal structures, the SP immunoreactivity was present in small distinctly stained puncta. These frequently formed pericellular arrangements around unstained cells, indicative of axosomatic contacts between SP terminals and cells in the hipocampus. In Ammon's horn, the densest SP innervation was found in strata oriens, radiatum and moleculare of subfields CA3a and CA2. Scattered fibers were also present in the stratum oriens of CA3a-c and in the hilus, in particular at ventral levels. In retrohippocampal structures, the SP innervation predominated in the deep pyramidal layer of the subiculum, the second layer of the presubiculum and in layers VI and IV of the medial and lateral entorhinal area. Many of these terminals may arise from local interneurons as well as from sources outside the hippocampal region. Taken together, these studies demonstrate a far more extensive innervation by SP, or a closely related peptide, of the rat hippocampal region than was previously recognized. This suggests that SP may play an important role in neurotransmission within the hippocampal region.

    Notes: Summary The morphology and innervation of the testicular artery and pampiniform plexus of the guinea-pig was investigated using light immunohistochemistry as well as transmission and scanning electron microscopy. The tortuous, spiraled testicular artery embedded within the epididymal fat pad is totally encompassed by a thin-walled sinus-like labyrinthine structure comprising the pampiniform plexus. Characteristic features of this lacunar system are: 1. Endothelial bridges, strands or trabeculae of various length, width and thickness which project into the lumen, occasionally branch, and attach to the opposite or adjacent area of the venous wall. 2. A frequent discontinuous smooth muscular layer. Nerve fibers were localized by use of antibodies against dopamine-β-hydroxylase (DBH-IR), neuropeptide Y (NPY-IR), and substance P (SP-IR). A great abundance of NPY-IR and DBH-IR axon bundles are seen surrounding the testicular artery. Fibers emanating from this dense plexus travel into the interstitium to finally innervate the walls of the sinus-like system, including the cross-luminal trabeculae. In contrast, larger varicosities are distinctive for SP-IR fibers which are also located at the media-adventitia border of the arterial and venous walls but to a far lesser extent than that seen with DBH-IR or NPY-IR. The axon varicosities supplying the arterial and venous walls contain a heterogeneous population of various types of vesicles, mostly including small agranular and granular ones as well as large granular vesicles of various size and density. The most conspicuous feature concerning the innervation pattern of the venous wall is the occurrence of numerous neuroendothelial contact zones. The findings of the investigated ‘rete mirabile’ are discussed with respect to rheology and temperature control for the maintenance of normal spermatogenesis.

    Notes: Summary Focal seizure was induced in rat and cat neocortex by the topical application of aminopyridines. The epileptic character of the focal events was followed by surface and intracellular recordings of seizure activity. The pathologic alterations in the neurons, the glial cells, and the protein permeability of the neocortical blood vessels were investigated by means of light microscopy, using standard histological stainings and immunohistochemical detection of endogenous serum albumin. Diffusion of [3H] 4-aminopyridine in the neocortex was studied by light-microscopic autoradiography. The spreading of the neuropathologic changes strictly followed the diffusion of the tritiated compound, suggesting the gradual involvement of neocortical layers in the seizure process.

These specimens were obtained from brains with Alzheimer's disease, Pick's disease, progressive supranuclear palsy, Alzheimer's type parkinsonism, parkinsonism-dementia complex on Guam, and normal aging. Under these neurologic conditions the affected cate-cholamine neurons with Alzheimer's neurofibrillary changes were stained positively with antisera against tyrosine hydroxylase. The results suggested that in these neurons, Alzheimer's neurofibrillary changes seemed to develop independently before the reduction of tyrosine hydroxylase protein synthesis.

    Notes: Summary Two methods of determining intermediate filament protein (IFP) expression by primitive brain tumors of childhood were compared using a panel of monoclonal antibodies to three classes of IFP. In addition to a controlled immunohistochemical study, a group of these tumors was subjected to direct immunologic assay of tumor-extracted IFP using the western blot method. Western blots of IFP extracted from ten prospectively microdissected brain tumors revealed no NF200 or NF150 in any tumor. Traces of NF68, VFP, and GFP were detected by this sensitive method in four, three, and six cases, respectively. Immunohistochemistry, using the same monoclonal antibodies on adjacent tumor sections, yielded results significantly different from the immunoblotting method: no NF proteins or VFP were detected, but immunoreactive GFP could be seen in a small percentage of cells in each case. A retrospective study of 46 primitive tumors, using only immunohistochemistry, showed GFP to be the most common source of immunopositivity (38 cases), followed by VFP (15 cases), but most positive cells were judged to be reactive astrocytes. NF protein was not detected except in three cases in which extremely rare cells had morphological features of neurons. Cells which were clearly malignant, and which constituted the majority of cells in a microscopic field, were devoid of any IFP immunoreactivity. The advantages and limitations of each method of IFP detection in this group of primitive tumors and the implications of the apparent paucity of mature neural IFP in these tumors are discussed.

    Notes: Summary The histogenesis of the cerebral neocortex and early allocortex of the sheep has been described and, using an immunohistochemical technique, five plasma proteins have been identified in the telencephalic wall and their distribution followed during its differentiation. The development of the neocortex was studied from 18 days gestation, when the neural tube was still open, to 120 days, when the adult structure was established. A primordial plexiform layer was formed above the ventricular zone by 25 days and by 35 days this layer was divided by the differentiating cortical plate into an outer marginal zone and an inner subplate zone. The appearance of the subventricular and intermediate zones by 50 days gestation completed the formation of the neocortical layers. The differentiation of the allocortex was generally less advanced than the neocortex up to 40 days gestation, when the primordium of the pyramidal layer was beginning to develop. The five plasma proteins identified, fetuin, α-fetoprotein, albumin, transferrin and α1-antitrypsin, are quantitatively the most important in the csf and plasma of the sheep fetus. Fetuin was the earliest plasma protein to be detected in the brain and it was also the most widespread; positive staining for this protein was seen in cells and fibres of all layers as they differentiated and could still be identified in some mature neurons at 120 days. α-Fetoprotein and albumin had a limited distribution, appearing in cells in the developing cortical plate for a short period early in gestation (35–40 days), but mainly confined to the ventricular zones later and barely detectable by 80 days gestation. Transferrin appeared to have a different distribution, being detected in fibres first in the primordial plexiform layer and then in the marginal and subplate zones, only later being identified in cells of the cortical plate. From their distribution it is suggested that fetuin and transferrin may play an important role in the differentiation of the cortex and the establishment of correct connections between fiber systems and migrating cells at certain stages of development. α1-Antitrypsin was only found in a few cells during a restricted period of gestation. All five plasma proteins were identified in precipitated csf and plasma at most ages examined, although at 18 days gestation albumin, transferrin and α1-antitrypsin and at 120 days, α-fetoprotein, could not be detected. The pattern of distribution of plasma proteins in the telencephalic wall suggests that they could originate either by uptake from csf and subsequent migration of protein containing cells or by local synthesis within some cells during a limited period of differentiation.

    Notes: Summary In the present study we have used the indirect antibody peroxidase anti-peroxidase technique to analyze the development of serotonin in the cerebellum of pouch young opossums ranging in age from birth (postnatal day (PD) 1) — to PD 47. The pathways by which serotoninergic axons enter the cerebellum appear to change during development. Between PD 1 and PD 11 varicose serotoninergic fibers course though the continuity between the tectum and the dorsal medial aspect of the cerebellar plate and distribute primarily to the intermediate zone. By PD 11 serotoninergic fibers enter the cerebellum via a connecting band between the cerebellum and the dorsal lateral aspect of the rostral medulla. Fibers entering the cerebellum via this later route course around the lateral recess of the fourth ventricle and initially distribute to ventral and lateral areas of the cerebellum. At later developmental ages (PD 14–PD 47) serotoninergic fibers are present in: 1) the cellular zone of migration between the Purkinje cell layer and deep nuclei; 2) the Pukinje cell layer; 3) the internal granule cell layer; and 4) the deep cerebellar nuclei. The external granule cell and the molecular layers rarely contain serotoninergic fibers. The present study has shown that serotonin is present in the cerebellar anlage on PD 1 (within 13 days of conception). This is prior to the arrival of other major afferent systems (King et al. 1982; Bishop et al. 1983; Martin et al. 1983; Morgan et al. 1983). Further, serotoninergic axons reach the cerebellum via different routes at different stages of development. The presence of serotonin in the intermediate zone early in development supports the hypothesis that it may influence neuronal migration and differentiation (Lander and Krebs 1976). Axons containing serotonin and arriving later in development may function in synaptic transmission, a role proposed for this indoleamine in the adult (Strahlendorf et al. 1979).

The fibers could be traced laterally directed towards the amygdaloid complex and some varicosities were also observed in the lateral parts of the nucleus periventricularis. It is postulated that brain CDD represents a new neuropeptide and that these CDD-IR neurons are involved in specific functions related to the modulation of the cardiovascular centers.

A three-dimensional reconstruction from serial sections of adult chicken heart was made to verify whether Purkinje cells, that can be recognized by a number of wellknown histological criteria, form specialized tracts in the adult chicken atria. This reconstruction revealed a loosely arranged network of Purkinje cells connecting the two atria. This network has not been described before. No tracts could be detected between the sinoatrial and the atrioventricular nodes. 2. These atrial Purkinje cells express the atrial and ventricular myosin isoform, as determined by the use of monoclonal antibodies that were prepared against atrial and ventricular myosin isoform, respetively. 3. Some atrial myocytes that are topographically closely related to the Purkinje cells and that cannot be distinguished from the surrounding myocytes with conventional histological criteria, express, apart from the atrial myosin isoform, also the ventricular myosin isoform. 4. The similar expression pattern of these two cell types and their close topographical relationship suggest the presence of a more elaborate system specialized in conduction than the well-known conductive system found with conventional histological techniques.

    Notes: Summary The distribution of serotonin immunoreactivity in the superior colliculus (SC) of the rat, hamster, chipmunk, cat, and monkey was studied using a sensitive immunohistochemical method. In all of these animals, serotonin immunoreactivity formed a dense network of varicose fibers throughout the SC. These fibers had a characteristic arrangement corresponding to the laminar structures of the SC. Except in the chipmunk, serotonergic fibers were more dense in the stratum griseum superficiale than in the other layers. In the SC of the chipmunk, these fibers appeared evenly distributed. To explore the degree of scrotonergic innervation in each layer, a semi-quantitative assay of serotonin immunoreactive varicosities was conducted in the rat, chipmunk, cat, and monkey. Peaks in varicose density were seen in the stratum griseum superficiale, the stratum griseum intermedium and the stratum griseum profundum. In the rat, cat, and monkey, the highest density of these varicosities was in the stratum griseum superficiale. On the other hand, the stratum griseum intermedium of the chipmunk SC received the greatest innervation of serotonergic varicose fibers.

    Notes: Summary The immunohistochemical distribution of laminin in 70 cases of primary and secondary brain tumors has been studied. The sections were fixed in Carnoy, embedded in paraffin, and treated with collagenase before applying the specific immune serum. In normal neural tissue and in gliomas, the distribution of laminin overlaps with that of basement membrane, as is known from observations on ultrastructure in the literature. The overlapping was observed also in neurinomas, papillomas of the choroid plexus, ependymomas, and meningiomas. Problems arise in the interpretation of the results in the other oncotypes. In hemangiopericytomas and hemangioblastomas, laminin demonstrates a basement membrane around the vessels only; it is absent around stromal cells. In desmoplastic medulloblastomas, laminin forms a network among the tumor celis. In primary lymphomas, concentric rings of laminin are evidenced around some vessels. In discussing the results, the ultrastructural distribution of basement membranes is taken into account.

    Notes: Summary Two cases of medulloblastoma are described which show positive immunostaining for glial fibrillary acidic protein (GFAP) in many cells. The surgical and autopsy specimens were examined by the indirect immunoperoxidase method. Positive staining for GFAP was demonstrated in the small, round to polygonal cells in both surgical specimens and in the small spindle cells in the autopsy specimen of one case. In the small, round to polygonal cells positive GFAP was shown as a perinuclear brown rim or intracytoplasmic brown droplet. In the small spindle cells, the cytoplasm and the polar processes were stained. Except for GFAP staining, these positive cells were morphologically indistinguishable from the accompanying unstained cells, indicating that GFAP was expressed in the medulloblastoma cells. Considering that GFAP is specific for astrocytes, these findings suggest the potential of astrocytic differentiation in the neoplastic cells of these medulloblastomas. The findings obtained in other 28 medulloblastomas examined in parallel are also discussed briefly.

    Notes: Summary Cellular localization of aldolase C isozyme was examined in the normal human brain and in nervous tissue tumors by the indirect immunoperoxidase method using an antibody to aldolase C. In normal brain tissues, staining was most prominent in astrocytes and Purkinje cells, although faint staining was also occasionally observed in some other neurons. Oligodendroglia and ependymal cells showed no distinct staining. The nervous tissue tumors we examined included 34 gliomas (ten astrocytomas and 24 anaplastic gliomas), 30 medulloblastomas, and seven neuronal tumors. Positive staining was observed in some gliomas, but not in the medulloblastomas and neuronal tumors examined. In gliomas, nine of ten astrocytomas and six of 24 anaplastic gliomas showed positive staining in their main constituent cells. In the remaining gliomas, most constituent cells were unstained or positive cells were only sporadically present. These findings indicated that aldolase C was expressed in some of astrocytic glioma cells as in the normal counterpart, more frequently in more differentiated form.

    Notes: Summary A case of a 14-year-old girl is reported in whom an alveolar rhabdomyosarcoma occurred in the soft tissues of the left forearm 4 years prior to death. Despite extensive surgery as well as chemotherapy and radiotherapy the tumor recurred locally and produced extensive metastases including a metastasis to the brain. Cerebral metastases have not yet been reported in the literature despite extensive reports on the pathology of alveolar rhabdomyosarcoma. The morphological diagnosis of rhabdomyosarcoma was supported by the immunohistochemical demonstration of desmin, myosin, and myoglobin in the tumor cells.

    Notes: Summary Serotonin (5-HT)-like immunoreactive nerve fibers were investigated in gut tissue obtained from seven Japanese children with Hirschsprung's disease. In the control untreated tissues, 5-HT-like immunoreactive fibers were observed neither in the normoganglionic nor in the aganglionic regions. After pargyline treatment, 5-HT-positive neuropils were consistently detected in association with the myenteric plexus in the normoganglionic segment, while in the aganglionic segment immunoreactive fibers could not be demonstrated through the entire layer of the bowel tissue. The occurence of 5-HT-like immunoreactive neuropils by pargyline treatment strongly suggests that the infant bowel is innervated with serotoninergic elements. After treatment with 5-hydroxy-l-tryptophan (5-HTP), the immunoreactive neuropils were localized in the myenteric and submucous plexuses of the normoganglionic segment. On the other hand, another type of 5-HT-positive fibers was characteristically demonstrated in aganglionic segments following the drug treatment; moderate numbers of 5-HT-like immunoreactive fibers appeared in the intermuscular zone and within the circular and longitudinal muscle layers. Several 5-HT-positive fibers were present in the hypertrophic nerve bundles seen in a diseased bowel. A possible origin of this type of 5-HT-positive nerve fibers was discussed.

    Notes: Summary Immunohistochemical localization of S-100 protein, its α andβ subunits, and glial fibrillary acidic protein (GFAP) in normal and neoplastic salivary glands was studied by the peroxidase-antiperoxidase method and immunoblot analysis. Positive immunostaining for S-100 protein was observed in pleomorphic adenoma, adenolymphoma, tubular adenoma, adenoid cystic carcinoma, acinic cell tumour, adenocarcinoma and carcinoma in pleomorphic adenoma. S-100 protein was localized in myoepithelial cells, epithelial cells of intercalated ducts and serous acinar cells of normal salivary gland. Both α andβ subunits of S-100 protein showed almost identical distribution in normal and neoplastic salivary glands, but skeletal muscle cells were α-positive/β-negative whereas Schwann cells and fat cells were α-negative/β-positive in the stroma and neighbouring tissue. GFAP was only found in pleomorphic adenoma and its malignant counterpart. Immunoblot analysis showed that the GFAP-related antigen consisted of several polypeptide bands with a molecular weight ranging between 35,000 to 50,000 daltons.

    Notes: Summary The appearance of S-100 protein-positive Langerhans cells was studied in 90 cases of various lung cancers by an immunohistochemical method. S-100 protein-positive dendritic cells were frequently observed in many adenocarcinomas, especially in those subclassified as bronchiolar cell or type II alveolar cell type. However, no S-100 protein-positive cells were found in “goblet cell type” adenocarcinoma. In some cases of squamous cell carcinoma and large cell carcinoma, these dendritic cells were also observed though they were fewer in number. In all cases of small cell carcinoma, however, S-100 protein-positive dendritic cells were rare. Electron microscopic study of two adenocarcinomas clearly demonstrated many Birbeck granules in the cytoplasm of S-100 protein-positive dendritic cells and confirmed that S-100 protein-positive cells in lung cancer were identical with Langerhans cells.

    Notes: Summary Twenty-four cases of hepatoblastoma, 14 cases of hepatocellular carcinoma and three cases of malignant mesenchymoma out of a total of 54 primary liver tumours were studied by light microscopy and immunohistochemistry. A remarkable finding in one case of hepatoblastoma and one case of hepatocellular carcinoma was a sarcoid-like reaction in the tumour tissue. Three cases of hepatoblastoma presented a macrotrabecular pattern. Among hepatocellular carcinomas, three cases corresponded to the fibrolamellar variant. By immunohistochemistry, the proportion of cases with positive staining for alpha1-fetoprotein was higher in hepatoblastoma than in hepatocellular carcinoma. HBs-antigen could be demonstrated in non-neoplastic liver cells in two cases of hepatocellular carcinoma, but not in the tumour cells. No strong correlation between histological pattern and prognosis could be established in hepatoblastoma. However, there was a tendency to more aggressive biological behavior in cases with pronounced mitotic activity. The number of mitoses in hepatoblastoma varied widely. As in previous studies, patients with the fibrolamellar variant of hepatocellular carcinoma fared better than those with the classical type of this tumour. Prognosis in malignant mesenchymoma was not as poor as suggested from previous studies.

    Notes: Summary Immunohistochemistry and fluorescence histochemistry were applied to the skin of the frog, Bombina orientalis, to detect the localization of bombesin and 5-hydroxytryptamine. The dermal layer contains three types (type 1, 2 and 3) of cutaneous glands but only the type-1 gland simultaneously contains bombesin and 5-hydroxytryptamine. This result suggests for the first time the coexistence of a bioactive peptide and monoamine in a cutaneous gland of the frog.

    Notes: Summary Immunohistochemical localization of corticotropin-releasing factor (CRF)-like immunoreactivity in the brain of the Japanese quail was studied by means of the peroxidase anti-peroxidase (PAP) method. CRF-immunopositive perikarya of parvocellular neurons were observed mainly in the nucleus praeopticus medialis and nucleus paraventricularis. Additional perikarya were also detected in the nucleus hypothalamicus posterior medialis in the hypothalamus and in the non-hypothalamic nucleus accumbens, nucleus septalis lateralis and nucleus dorsomedialis and dorsolateralis thalami. No CRF immunoreaction was found to coexist with the vasotocin (Vt)-containing system in comparative examination of consecutive sections treated with anti-vasopressin (Vp) serum. The CRF-immunoreactive fibers were detected mainly in the external layer of the anterior median eminence but not in its posterior division. Unilateral adrenalectomy induced the marked reduction in number of the CRF immunopositive fibers in the anterior median eminence.

    Notes: Summary Ontogenesis of gastrin cells was studied in the pyloroduodenal mucosa of the mouse using anti-human G17 serum, R-1301, and anti-human G34(1–15) serum, R-2703. R-1301-immunostained cells first appeared in the pyloric mucosa of 14-day-old fetuses. Cells stained with both R-1301 and R-2703 appeared immediately after birth, and gradually increased in number to the adult level. Most R-1301-reactive cells were also reactive to R-2703, whereas some cells that reacted with R-1301 exhibited very weak or no reaction with R-2703. The discrepancy between these two immunoreactivities is discussed. In the duodenum, a considerable number of R-1301-reactive cells were present from the perinatal stage and through out adult development. A few R-2703-reactive cells were seen in the duodenum of young mice but not of the adult.

    Notes: Summary A study on the localization of fetal and neonatal brain macrophages of mice from embryonic day 10 (E10) to postnatal day 21 (P21) was carried out immunohistochemically using a monoclonal antibody against a macrophage differentiation antigen (Mac-1) and the labeled avidin-biotin technique. In the central nervous system, the macrophages recognized first were mainly located in the choroid plexuses of the fourth and lateral ventricles at E14. Their number increased at E17–P3 and gradually decreased thereafter. In the cerebral parenchyma, a few macrophages appeared at E14 in the matrix cell layer. They were also detected in the migrating zone at E15, E17 and in the cortical plate at E19. Mapping of positive cells at the stage of neuroblast formation (E15, E17, E19) disclosed the precise distribution of cerebral macrophages. The macrophages that appeared first in the choroid plexuses at E15 may be derived from the subarachnoid vessels, which extend into the stroma of the choroid plexuses when the matrix cell layer invaginates into the lateral ventricle to form the choroid plexuses. Almost all of the macrophages recognized in the cerebral parenchyma disappeared at P9 when the cytoarchitecture seemed to be completed. In the cerebellum, which develops later than the cerebrum, macrophages appeared after birth and were located mainly in the internal granular layer. The brain macrophages always appeared in the regions where cell proliferation and brain remodeling are most active at each stage. These findings suggest that fetal and neonatal brain macrophages may play an important role in scavenging degenerated cells and cell debris during histogenesis of the central nervous system.

    Notes: Summary The innervation of the ductuli efferentes and seven zones of the guinea-pig epididymis was investigated using immunohistochemical, histochemical and electron-micro-scopical techniques. Nerve fibers were localized by use of antibodies against substance P (SP-IR), vasoactive intestinal polypeptide (VIP-IR) and dopamine-beta-hydroxylase (DBH-IR). In the ductuli efferentes and all zones of the epididymal duct, SP-IR is consistently observed in the interstitial tissue and perivascular areas. Histochemistry reveals a significant amount of acetylcholinesterase-containing fibers in the interstitial, perivascular and periductal smooth muscles of the ductuli efferentes and zones V, VI and VII. In contrast to the homogeneous distribution of SP-IR within all zones of the epididymis, VIP-IR is seen only in zones VI and VII. Within these zones, VIP-IR is detected in large amounts in the subepithelial and muscular layers as is a sparse number of SP-IR varicosities. DBH-IR is also seen throughout all zones in the interstitial and perivascular regions with a tendency to increase in zones VI and VII. Transmission electron microscopy (TEM) reveals evidence of a cholinergic (agranular vesicles, AGV), adrenergic (small granular vesicles, SGV) and peptidergic (large granular vesicles, LGV) innervation throughout the interstitial connective tissue of the ductuli efferentes and all epididymal zones. Furthermore AGV are localized in the subepithelial layer, and also co-stored with LGV in the muscular layer of zones VI and VII. No nerve profiles were encountered within the epithelium. Correlation of immunohistochemical findings to TEM counterparts as well as their possible functional role are discussed.

    Notes: Summary The cellular localization of renin was examined in the kidneys of some amphibians of the genus Bufo by immunoperoxidase and immunofluorescence techniques with an antiserum to renin isolated from the submandibular gland of the mouse. Immunoreactivity could be demonstrated in the media cells of the afferent arterioles (juxtaglomerular cells) close to as well as at great distance from the glomeruli. Occasionally, media cells of larger arterial vessels were also stained. The immunohistochemical data seem to be in accordance with earlier results obtained with a modified silver impregnation technique (Movat's staining procedure) used for the visualization of juxtaglomerular cells in non-mammalian vertebrates. Mouse kidney tissue, studied for purposes of comparison, showed renin-immunoreactivity as described by earlier investigators, i.e., immunoreactive staining in the afferent arterioles near the glomeruli and in the proximal tubule cells.

    Notes: Summary The present study deals with the localization and development of S-100 protein-like immunoreactivity in the retina, ciliary body and iris of human fetuses. In the retina, numerous astrocytes, densely distributed in the nerve-fiber layer and ganglion-cell layer, were stained strongly with the S-100 antiserum. The first immunoreactive astrocytes occurred at the posterior pole of the retina and spread gradually outward and toward the ora serrata with increasing age. Müller cells were not immunoreactive for S-100 during development, except in the retina of the latest fetus examined. S-100 immunoreactivity was also found in the nonpigmented ciliary epithelium and posterior epithelium of the iris, both of which are developed from the inner wall of the optic cup. On the other hand, the pigmented epithelium extending from retina to iris, derived from the outer layer of the optic cup, was free of S-100 immunoreactivity.

    Notes: Summary The midgut of Blaberus craniifer is principally made up of columnar epithelial cells which are derived from small regenerative cells found grouped in nidi. Between them, small sparsely granulated cells with clear cytoplasm can be observed lying on the basal lamina. Mainly based on the size, shape and texture of their secretory granules, at least ten types of such endocrine cells have been identified. Five cell types contain a uniform population of dense granules: (1) medium-sized, round to oval granules; (2) small elongated granules; (3) large irregular granules; (4) oval granules with a highly osmiophilic core; (5) oval, haloed granules. Five others are characterized by a heterogeneous population of granules: (6) small, round to oval, variably electron-dense granules; (7) oval medium-sized granules of variable electron density; (8) large irregular granules of variable electron density; (9) small dense granules and large vesicles with filamentous material; (10) small dense granules and very large pale vesicles. In addition, near the regenerative cells, large cells characterized by very large, irregular, dense granules (up to 4 μm), lack contact with the lumen, and reach the basal lamina only by slender cytoplasmic processes. Several antisera raised against mammalian peptides and amine were used to reveal axonal fibers and endocrine cells. Serotonin-like immunoreactivity is localized in a profuse innervation of the muscle layers that surround the epithelium, whereas cholecystokinin and methionine-enkephalin antisera stain a more moderate number of axonal fibers. Cholecystokinin-, methionine-enkephalin-, substance P-, vasoactive intestinal peptide-, somatoliberin-, and gonadoliberin-like immunoreactivities were detected in endocrine cells of the epithelium. While most of the cells appear pyramidal, oval, fusiform or bowl-shaped, and seem to lack contact with the lumen, cells reaching it have been detected reacting with antisera to cholecystokinin, substance P, vasoactive intestinal peptide, somatoliberin and gonadoliberin.

    Notes: Summary Non-ciliated SER-rich cells of the tracheal epithelium of normal, phenobarbital-treated and 3-methylcholanthrene-treated mice were studied ultrastructurally and immunohistochemically. The apical portion of these cells protrudes into the tracheal lumen, especially in the mice treated with the two compounds, and the apical cytoplasm is filled with numerous tubular elements of SER. Besides, the non-ciliated cells of 3-methylcholanthrene-treated mice show a strong positive reaction to the antiserum against microsomal cytochrome P-450 of liver. These findings support the concept that the non-ciliated tracheal cell may be involved in the metabolism of endogeneous and exogeneous chemical compounds.

    Notes: Summary The immunoreactive CRF-neurons of the rat hypothalamus have been examined immunohistochemically employing anti-rat CRF serum. These neurons are confined to the paraventricular nucleus, dorsomedial-lateral hypothalamic area, and suprachiasmatic nucleus, and are, respectively, also immunoreactive to anti-Met-enk, -alpha-MSH, and -VIP sera. Intraventricular administration of colchicine (50 μg/5 μl/rat) induces a dramatic enhancement of the immunostainability of the cell somata, and also accelerates the development of immunoreactivity of other stored peptides, especially in the paraventricular nucleus. The CRF-neurons respond to adrenalectomy by showing increased immunoreactivity and an increase in the number of cell bodies; in the dorsomedial-lateral area and suprachiasmatic nucleus, there is also an enhanced immunoreactivity for alpha-MSH and VIP, respectively. CRF-cells in the paraventricular nucleus become markedly hypertrophied, but do not show any enhanced immunoreactivity for Met-enk. Since the axons of the paraventricular neurons run to the median eminence, it is probable that they are involved with the endocrine control of hypophysial ACTH release. It is concluded that the CRF-containing neurons in rat hypothalamus consist of three types which are functionally and morphologically different.

    Notes: Summary Immunohistochemical studies were performed by use of specific rabbit antisera and purified antibodies to human Tg on cephalic and body sections of Eisenia foetida and on cephalic sections of Lumbricus terrestris. Secondary antisera, either fluorescein- or peroxidase-conjugated, were used to identify the immunoreaction. Immunoreactive perikarya and some immunoreactive nerve fibres were detected in both the cerebral ganglion and the ventral nerve cord of E. foetida. From 8 to 19 Tg-like positive neurons per frontal section were observed in the brain, mainly in the dorsal zone. From 2 to 4 positive perikarya per ganglion were found in sagittal sections of the ventral nerve cord with a repetitive distribution. Numerous positive neurons were also found in the cephalic segments of L. terrestris. The present results indicate that a substance immunologically related to mammalian Tg is synthesized in earthworms. This suggests that some conservative sequences of Tg structure arose very early in evolution and supports the idea of a common evolutionary origin for endocrine and nervous systems.

    Notes: Summary The postnatal development of the various cell populations in the rat popliteal lymph node was investigated applying enzyme-histochemical and immunohistochemical techniques. From birth, T-lymphocytes and interdigitating cells were demonstrable. During the development of the young lymph node, T-lymphocytes of the helper phenotype outnumbered the T-cells with a suppressor phenotype; they account for approximately 70% and 30% of all T-lymphocytes, respectively. At the very first day of postnatal life, post-capillary venules were already present. B-lymphocytes occurred later than T-cells during ontogeny; they were found on the second day after birth, most of them being IgM- or IgG-bearing lymphocytes. The first primary follicles occurred at day 18 and contained principally membrane-stained IgM cells and, to a lesser extent, membrane-stained IgG cells. The appearance of follicular dendritic cells correlated with the formation of primary follicles. With respect to the macrophages, it appeared that the ED1- and ED3-positive subpopulations were present with a similar distributional pattern as seen in adults, but in considerably lower numbers. The expression of ED2, however, showed a sudden increase in the third week of life. Findings of the present study are discussed in relation to those obtained in other investigations dealing with the ontogenetic development of lymphoid organs.

    Notes: Summary The distribution of substance P-immunoreactivity (SP-IR) in the brainstem and spinal cord of normal and colchicine-pretreated cats was analysed using the peroxidase-antiperoxidase (PAP) technique. Numerous SP-IR fibers are present in the nucleus solitarius, nucleus dorsalis nervi vagi and nucleus spinalis nervi trigemini, various parts of the formatio reticularis, substantia grisea centralis mesencephali, locus coeruleus and nucleus parabrachialis. SP-IR perikarya occur in the substantiae gelatinosa and intermedia of the spinal cord, the nucleus spinalis nervi trigemini-pars caudalis, the nucleus dorsalis nervi vagi, and the nucleus solitarius, as well as in the adjacent formatio reticularis and the medullary nuclei of the raphe. In addition, SP-IR cell bodies are located in the nuclei raphe magnus and incertus, ventral and dorsal to the nucleus tegmentalis dorsalis (Gudden), nucleus raphe dorsalis, substantia grisea centralis mensencephali, locus coeruleus, nucleus parabrachialis and colliculus superior. The results indicate that SP-IR neurons may be involved in the regulation of cardiovascular functions both at the central and peripheral level. A peripheral afferent portion seems to terminate in the nucleus solitarius and an efferent part is postulated to originate from the nucleus dorsalis nervi vagi and from the area of the nuclei retroambiguus, ambiguus and retrofacialis.

    Notes: Summary The presence and distribution of regulatory peptides in nerves and endocrine cells of the stomach, intestine and rectum of a urodele amphibian, the mudpuppy, Necturus maculosus, was studied immunohistochemically in sections or whole-mount preparations of the gut wall. The effect of the occurring peptides on gut motility was studied in isolated strip preparations of circular and longitudinal smooth muscle from different parts of the gut. Bombesin-, neurotensin-, substance P- and VIP-like immunoreactivity was present in abundant nerve fibres in the myenteric plexus of both stomach, intestine and rectum. Single fibres or bundles were present in the circular muscle layer and in a well-developed deep muscular plexus in the intestine and rectum. Immunoreactive nerve cells were found in the myenteric plexus of the stomach, intestine (neurotensin only) and rectum. Gastrin/CCK-like immunoreactivity was observed only in a few fibres in stomach and rectum. Endocrine cells containing bombesin-, met-enkephalin-, gastrin/CCK-, neurotensin-, somatostatin- or substance P- like immunoreactivity were present in the mucosa. The effect of bombesin was an inhibition of the rhythmic activity in circular muscle preparations and in longitudinal muscle from the rectum, while longitudinal muscle from the stomach usually responded with a weak increase in tonus. Neurotensin, like bombesin, was inhibitory on the spontaneous rhythmic activity of circular muscle throughout the gut, while the effect on longitudinal muscle was an increase in tonus. Met-enkephalin and substance P increased the tonus of all types of preparations, and often, in addition, initiated a rhythmic activity superimposed on this maintained tonus. VIP had a general inhibitory effect on the preparations, decreasing tonus and/or abolishing rhythmic activity. It is concluded that bombesin-, neurotensin-, substance P- and VIP-like peptides are present in nerves throughout the urodele gut and may have physiological functions in regulating the motility of the gut. The gastrin/CCK-like peptide present in nerves of the stomach and rectum may affect the function of these parts of the gut. The regulatory peptides present in endocrine cells may, perhaps with the exception of the somatostatin-like peptide, affect the motility humorally.

    Notes: Summary Location, distribution and density of nerve fibers immunoreactive to neuropeptide tyrosine, vasoactive intestinal polypeptide and substance P were studied in the reproductive tract of the female rat and compared with acetylcholinesterase-positive (“cholinergic”) and noradrenergic nerves. Plexuses of all types of fibers were present in the vagina, uterine cervix, uterine horn and oviduct. In the tubular reproductive organs all of these types of nerve fibers appeared to innervate vascular and non-vascular smooth muscle and nearly all types of fibers formed plexuses subjacent to the epithelium lining the organs. Individual fibers of all classes appeared to innervate fascicles of smooth muscle in the mesometrium of the uterine horn. A few acetylcholinesterase-positive and substance P-immunoreactive fibers were present in the ovary but no vasoactive intestinal polypeptide-immunoreactive nerves were observed. Noradrenergic and neuropeptide tyrosine-immunoreactive nerves were numerous in the ovary where they were seen in the interstitial gland tissue and associated with follicles and blood vessels. It is suggested that these nerves may influence hemodynamic events and non-vascular smooth muscle in such functions as transport of sperm and ova and parturition. Substance P-immunoreactive nerve fibers are likely to be sensory fibers that could have roles in neurohormonal reflexes.

A sub-population (6–10%) of the gastrin-containing endocrine cells (G-cells) was found to react with antisera to met-enkephalin, met-enkephalin-Arg6-Phe7 and met-enkephalin-Arg6-Gly7-Leu8. About 30% of all the enkephalin-containing cells were identified as G-cells. The results indicate that a fraction of G-cells produces both enkephalin-like peptides and gastrin.

    Notes: Summary The distribution of serotonin in the hypothalamus and the mesencephalon of guinea-pigs pretreated with both pargyline and L-tryptophan was investigated immunohistochemically using monoclonal antibodies to 5-HT. 5-HT-positive fibers and varicosities appeared distributed throughout the hypothalamus. Some areas showed a greater density of immunoreactivity: the suprachiasmatic nucleus, the region of the supraoptic crest, the area of the medial forebrain bundle, the ventral part of the nucleus ventromedialis, the median eminence and the ventral part of the mammillary bodies. 5-HT nerve fibers were also scattered in the posterior lobe of the pituitary. An extensive supraependymal plexus of immunoreactive axons was observed in most ventricular regions. No 5-HT positive cell bodies were present in the hypothalamus of the guinea-pig under our experimental conditions, whereas an intense serotonin immunoreactivity was detected in perikarya of the brain stem. 5-HT cell bodies were found predominantly in the raphe region including the nucleus raphe dorsalis and raphe medianus, nucleus interpeduncularis, reticular formation and dorsal area of the medial lemniscus.

    Notes: Summary The distribution of immunoreactive neuropeptides was investigated in the retina of three species of skates (Raja clavata, R. radiata, R. oscellata), elasmobranch fish often used in electrophysiological work on the retina. Enkephalins, neuropeptide Y (NPY), substance P and glucagon were found in different types of amacrine cells. All four peptides appeared in cell bodies in the innermost part of the inner nuclear layer. Processes from the cells containing enkephalins were numerous and ramified throughout the inner plexiform layer. Processes from the cells containing glucagon were thick and rare, and were found throughout the inner plexiform layer, at times with a predominance in sublaminae 1 and 4. NPY-immunoreactive fibres appeared mainly in sublamina 1 but also in 2 or 3, and substance-P-immunoreactive fibres in sublaminae 1,4 and 5. Antisera against somatostatin, VIP or neurotensin did not show any immunoreactivity in the skate retina.

    Notes: Summary After an exposure of 24 h to synthetic LHRH (100 ng/ml) in vitro, the anterior pituitaries of 4-day-old rats show a notable loss of immunoreactive material in most LH cells in males, but not in females. When radioimmunoassayed without incubation, the pituitary LH content of 4-day-old female rats is 2.8 times higher than that of males of the same age. LHRH treatment stimulates a higher rate of LH discharge in females than in males, but if LH release is expressed as a percentage of the initial pituitary LH content, there is no apparent difference. In both sexes, more than 70% of the initially stored LH is discharged into the medium after 24 h of LHRH stimulation. In males, this discharge produces a pronounced depletion, but in females, the pituitary still contains 78.2% of the initial LH content despite the large amount of hormone released. From these results, it is concluded that in newborn rats the LH synthetic rate in females is higher than that in males. This high synthetic activity, together with the large store of LH, may explain why prolonged LHRH treatment fails to cause LH depletion in females. At 4 days of age LHRH had no stimulatory effect on pituitary synthesis of LH in either sex.

    Notes: Summary The objective of this in-vitro study was to examine whether the diencephalic floor or the mesenchyme is involved in differentiation of LH cells in the developing rat adenohypophysis. Overall growth of the adenohypophysial tissue was retarded when the adenohypophysial primordium was cultivated after enzymatic removal of the diencephalic floor on days 11.5 and 12.5 of gestation. This malgrowth was more marked when the brain was separated on day 11.5; most expiants retained a simple cystiform structure that consisted of a few layers of undifferentiated cells. Removal of the brain also caused a highly significant decrease (P 〈 0.001) in the number of immunoreactive LH cells, if it was performed on day 11.5 but not day 12.5. Mesenchyme had little effect on the adenohypophysial growth or the number of immunopositive cells. Cultivation of the adenohypophysial primordium with the diencephalic floor resulted in the appearance of many immunoreactive LH cells. The number of LH cells significantly decreased, however, when the co-cultivated brain completely surrounded the adenohypophysial tissue. These results indicate that in 11.5-day-old fetal rats the diencephalic floor is indispensable for the initial proliferation of adenohypophysial primordial cells and for the early determinating process of LH cells. Once determined, the development of LH cells may proceed without the surrounding tissues. The cytodifferentiation seems to be rather inhibited when in contact with the brain. The significance of the intimate spatial relationship between developing LH cells and the surrounding mesenchyme is also discussed.

    Notes: Summary Physiological compartmentation in carp (Cyprinus carpio L.) gonads was investigated after intracardial injection of horseradish peroxidase (HRP) and two mouse anti-carp-sperm monoclonal antibodies. Immunohistochemistry revealed that a physiological barrier exists in carp testis for HRP and mouse IgG monoclonal antibody around the central lumina of the tubules in which the spermatozoa are located, but not around the cysts containing the precursor germ cells. The results with HRP were confirmed by electron microscopy. Mouse IgM monoclonal antibody did not penetrate the spermatogenic cysts. Probably because of its large size, it was almost exclusively located inside blood capillaries and only sparsely in the interstitial tissue. In the ovary, HRP was regularly distributed in the gonadal tissue, whereas the IgG antibody was predominantly localised on oogonia and early prophase oocytes. The results indicate that in contrast with the testis, no barrier around germ cells exists in the carp ovary.

    Notes: Summary Numerous immunochemical and immunohistochemical studies have shown a wide distribution of several families of neuropeptides in invertebrates as well as vertebrates. There are relatively few data available for Annelida: Polychaeta. Therefore, we undertook an immunohistochemical investigation in the marine worm Nereis. Among the vertebrate type antibodies tested, those against met-enkephalin, LH-RH, vasopressin, oxytocin and ACTH had negative or only very slight effects. Slight to moderate reactions were obtained for VIP, SRIF, CRF, GRF, and leu-enkephalin. Moderate to very strong responses were found with anti-CCK/gastrin, -substance P, and -β-MSH sera. Immunopositive reactions were usually observed in the entire CNS (except, until now, in neurosecretory cells, type II, in nuclei 20, and in nerve fibres located in the infracerebral neurohemal area). The immunoreactivity was, however, more or less abundant according to different CNS regions. For example, it appeared that the immunostaining for CRF is more important in the VNC while the leu-enkephalin family is more abundant in the brain (particularly in fuchsinophilic neurosecretory cells, type I, in nuclei 20). Moreover, several vertebrate type peptides (such as CRF/GRF and CCK/gastrin) may coexist in a single neurone. Several antisera may elicit a positive reaction in some specific area (for example, substance P in the nuchal organ; SRIF in oocytes; CCK/gastrin in the gastrointestinal tract). Nothing is known about the role of the different substances immunologically detected in Nereis. It is suggested that CCK/gastrin-, β-MSHand substance P-like materials transmit external stimuli to neurosecretory centres located in the caudal part of the brain.

    Notes: Summary Ontogenetic development of GRF-containing neurons in the rat hypothalamus was studied employing antisera which were generated against hpGRF (1–44)NH2 and rhGRF(1–43)OH: anti-hpGRF-C and -rhGRF sera recognize the species-specific C-terminal portions of the peptides, and anti-hpGRF-MC and -N sera recognize hpGRF(27–44)NH2 and the N-terminal portion of hpGRF(1–44)NH2, respectively. The anti-hpGRF-C and-rhGRF sera stained different neuronal cell bodies, which were localized in distinct hypothalamic areas. The former serum did not stain the axonal terminals in the median eminence, but the latter stained them strongly. The antihpGRF-MC and -N sera stained neuronal cell bodies, some of which corresponded to those immunolabelled with antihpGRF-C or -rhGRF serum. The anti-rhGRF serum first demonstrated immunoreactive perikarya in the ventral-lateral border of the arcuate nucleus of 19.5-day-old fetuses that had received an intraventricular colchicine administration 24 h previously. The immunoreactive fibers were recognized first in the external layer of the median eminence of untreated fetuses on day 19.5 of gestation, and then they increased in amount with development. No immunore-active fibers, however, were found in the median eminence of colchicine-treated animals during the fetal period. It is concluded that in rats GRF may be synthesized in the perikarya on day 18.5 of gestation and conveyed to the median eminence without delay via axonal flow.

    Notes: Summary In the four species of elasmobranchs examined (Triakis scyllia, Heterodontus japonicus, Scyliorhinus torazame, Dasyatis akajei), all identifiable caudal neurosecretory cells and their corresponding neurohemal areas showed urotensin II (UII)-immunoreactivity with varied intensity. To localize urotensin I (UI) in the caudal neurosecretory system of the dogfish, Triakis scyllia, h-CRF (1–20) antiserum that cross-reacts with UI was used in place of UI antiserum. CRF/UI-immunoreactivity was demonstrated in the neurosecretory cells and neurohemal areas. A considerable number of neurons showed both UII- and CRF/UI-immunoreactivities, suggesting that UII and UI are produced in the same neurosecretory cells. However, some neurons exhibited UII-immunoreactivity, but no CRF/UI-immunoreactivity. Cells immunoreactive only to CRF antiserum were not detected. At least two populations of neurons exist in the dogfish caudal neurosecretory system: (i) cells immunoreactive for both CRF/UI and UII, and (ii) cells immunoreactive for UII. The dorsal cells of the lamprey, Lampetra japonica, did not react with either UII or CRF antiserum.

    Notes: Summary Bone resorption in the temporal bone of rats was induced by peripheral immunizations of heterogeneous type II collagen. Bone resorption was found at both the tympanic bone and the petrous bone of the otic capsule. Macrophages, fibroblasts and osteoclasts were found in the enlarged spaces of bone. Immunohistochemical studies demonstrated that macrophages, fibroblasts and osteoclasts produced collagenase and prostaglandins in the bone resorption processes, suggesting that these cells are responsible for the resorption observed.

    Notes: Summary Gelatinous and telangiectatic types can be differentiated among the human vocal fold polyps. Telangiectatic polyps are characterized by eosinophilic deposits consisting of fibrin and cellular blood constituents. Labyrinthine vascular channels are characteristic of these polyps, and are partially or completely lined by a single layer of flat cells. Using electron microscopy and immunohistochemical stainings (antibodies against factor VIII-related antigen, Ulex europaeus I lectin, and antibodies against lysozyme), we found that the lining cells are true vascular endothelial cells and are not organizing histiocytic cells that are arranged in an endothelial-like pattern.

It was demonstrated that intermicroscopic correlation of images between light microscopy (LM), scanning electron microscopy (SEM), and transmission electron microscopy (TEM) is easily and reliably done by means of the PEG method. In particular, the exact correlation of immuno-LM with SEM is shown to be of potential value.
