    Notizen: Process control strategies, e.g. the steady-;state fermation, which do not consider the cell-;state distribution of cell populations, cause a decrease of yield coefficient connected to an increase of heat-;production coefficient.Contrary to this, the dynamic process control adapts the carbon-;substrate supply to the different repetitive cell-;population states during fermentation. In order to recognize the different states, the cell population is synchronized. The optimum period ofchanging carbon-;substrate supply was found to be dependent on the cell-;doubling time. Dynamic process control causes a higher yield coefficient as well as a lower heat-;production coefficient compared to the steady-;state fermentation.

    Notizen: Phanerochaete chrysosporium cells were immobilized on the sintered porous glass support. Such a biocatalizer was used as a bed of the enzymatic reactor system for the continuous production of lignin peroxidase. From the after culture fluid the lignin peroxidase enzymatic activity was recovered and purified applying anion exchangers. Additionally, some physico-chemical properties of lignin peroxidases were determined.

    Notizen: It has been found visually and by spectrophotometric measurements that the infection of the algae culture of Sc. acuminatus used for biomass production has caused a changing of its color toward the large wavelengths of the spectrum. Experimental studies have proved that the color change was due to the reaction between the phytoplankton and the nitrous acid produced throughout the cultivation process. It has also been demonstrated that the coefficient of bathochromicy introduced in this study was efficient for quantitative estimation of algae biomass color, and could be used for control of algae culture color.

    Notizen: A fed-batch process is used for the production of concentrated pure fructose syrup and ethanol from various glucose/fructose mixtures by S. cerevisiae ATCC 36859. Applying this technique, glucose-free fructose syrups with over 250 g/l of this sugar were obtained using High Fructose Corn Syrup and hydrolyzed Jerusalem artichoke juice. By encouraging ethanol evaporation from the reactor and condensing it, a separate ethanol product with a concentration of up to 350 g/l was also produced. The rates of glucose consumption and ethanol production were higher than in classical batch ethanol fermentation processes.

    Notizen: The feasibility of using ultrafiltration to concentrate RNA was investigated. Ultrafiltration flux and solute retentivity were affected by membrane cut-off and inlet pressure.The RNA can be concentrated by selecting a hydraulically permeable membrane (PM 30) and then operating the system at a sufficiently high pressure (2.07 · 105 Nm-2) and a high recirculation rate (150 cm-1). The use of this procedure was limited to a high-flux regime. However, a very high retentivity of macrosolute was achieved. The effect of transmembrane pressure on the apparent mass transfer coefficient in the “gel polarization model” was discussed and related to ultrafiltration flux.

    Notizen: Two methane utilizing bacteria strains, GB 21 and WSB 874, were cultivated with methane as sole carbon and energy source in a submerged continuous fermentation process. The aim of the investigations was to reach high biomass concentrations (〉25 g BDM ·kg-1 medium) and high productivities (〉3 g BDM · kg-1 · h-1). A precondition for the high-performance fermentation process was to ensure a transfer rate of methane and oxygen in quantities required by the microorganisms. For this purposes a high performance stirred pressure fermenter was used. The fermentation process is characterized by the transfer of two gaseous substrates. In order to develop a technical process it was necessary to investigate both the correlations between the two gases and the productivity of the fermentation process and the influence of system pressure on the microorganisms.Within a pressure range of up to 0.7 MPa a biomass concentration of up to 50 g BDM · kg-1 medium and a productivity of up to 9 g BDM · kg-1 · h-1 was reached in a continuously running fermentation process under nonsterile conditions. Under these fermentation conditions the microbial population consisted of a dominating main culture GB 21 or WSB 874, respectively, and an accompanying flora (type II). The microbial population was very stable in its composition during the fermentation process running continuously for weeks and months.

    Notizen: Investigations simulating solid-state fermentation, SSF, were carried out to study the influence of a varying N supply on the growth of a mycelium.Experiments with two fungi and two N sources demonstrated the high adaptability of the mycelium to a deficit of nitrogen over a wide range of N supplies. The mycelium of the fungi Gliogladium sp. and Aspergillus sp. can grow up to the very low crude protein content of 11-15%. But protein-poor biomass could not make up the deficit of protein even under optimal cultivation conditions, that means the state of the mycelium remains irreversible. This showed the limit of the adaptability of filamentous fungi, which has to be taken into account for mycelium growth in SSF.

    Notizen: The low incidence of antigen-specific B cells in available material is one of the major problems in human monoclonal antibody production. A technique is described which permits the efficient production of specific hybridomas by the separation of specific B cells with magnetic beads prior to fusion.Cells with immunoglobulin γ chains on the surface were enriched from human peripheral blood lymphocytes by separation withmagnetic beads. After electrofusion of the pre-enriched fraction with CB-F7 heteromyeloma cells, the percentage of IgG-producing hybridomas was considerably enhanced. Similarly, B cells with antigen-receptors for α haemolysin from Staphylococcus aureus were enriched by separation with antigen-coated beads. In nine fusion experiments taken together, 153 out of 503 wells contained antigen-specific immunoglobulin after the application of this technique, compared with 37 out of 1584 wells after immortalization of unseparated lymphocytes. The ratio of IgG:IgM-producing antigen-specific hybridomas was slightly shifted towards IgM.Using this technique, antigen-specific hybridomas from lymphocytes of healthy normal donors were established without any artificial administration of antigen.

    Notizen: Conversion of starch to ethanol without its prior hydrolysis is of special interest in the treatment of the starch containing waste-waters. In this work the effect of calcium carbonate on the growth and ethanol production from soluble starch by Schwanniomyces castellii NRC 2676 was studied. When the medium was supplemented with 0.1% calcium carbonate the biomass yield and the total starch consumption were increased by 20% and 22%, respectively, while the ethanol yield decreased for 28% in comparison with the control. The rate of biomass production, measured for the period of time between 0 and 24 h, was the highest in the medium with 0.06% calcium carbonate while the rates of ethanol production and starch consumption decreased with and increase in the concentration of that salt; the media with 0.5% and 0.7% calcium carbonate had 3.5 and 5.5 times lower ethanol production rates, and 2.3 and 3.8 times lower rates of starch consumption than the control medium which was not supplemented with calcium carbonate, respectively. The obtained results also showed that the percentage of starch consumed was higher in the media supplemented with calcium carbonate than in the control.Empirical mathematical expressions are given for the relationship of the biomass and ethanol yields to the concentration of calcium carbonate in the medium.

    Notizen: Oats was used as a substrate in tempeh fermentation. The time needed to obtain sufficient mold growth was at least 30 hours at 31°C (Rhizopus oligosporus NRRL 2710). pH was decreasing during the first 32 hours of incubation reaching pH = 5.30. Fermentation of oats led to an increase in water soluble nitrogen, but it did not change protein nitrogen content. R. oligosporus proteinases of optimum pH = 5.50 are postulated to play an important role in oats tempeh fermentation. When a mixture of oats and soybean (1:1) was used, mold growth was faster and the cake tougher. Mixing cereals with legumes to produce good tempeh is recommended.

    Notizen: The Rhizobium sp. isolated from the root nodules of Clitoria ternatea L., a leguminous twiner, produced a high amount of IAA (16.4 μg/ml) from tryptophan in an unsupplemented basal medium. The production of IAA started simultaneously with the growth and had no different growth and production phase. The growth and production were parallel and increased up to 45-50 h. The IAA production by the Rhizobium sp. was increased by 520% when the medium was supplemented with fructose (1.5%), MnSO4 (1.0 μg/ml), riboflavin (0.10 μg/ml) and Triton X-100 (0.01%). The possible role of the rhizobial production of IAA on the rhizobia-legume symbiosis is discussed.

    Notizen: Seedings of Hypericum perforatum L. have produced multiple shoots on RM medium supplemented with 2.22 or 4.40 μM BAP. Regenerants were evaluated for variability in several morphological characteristics. A comparison with the control showed significant differences in fresh and dry matter and height of plants, but did not effect branching and the number of glands containing dianthrones.

    Notizen: Two calorimetric techniques are described allowing us to monitor and control microbial processes at relatively high cell densities: dynamic and continuous calorimetry. These techniques have been applied to study microbial systems, e.g. the dependence of thermodynamic efficiency and heat yield on specific growth rate, the heat yield of mesophilic and thermophilic microorganisms, the oxy-caloric coefficient during aerobic microbial growth, as well as the energetics of the yeast-cell cycle. Such studies are of importance for optimizing biotechnical processes.

    Notizen: The mechanism of carbon dioxide saturation of alkaline nutrient medium for the purpose of autotrophic cultivation of microalgae is studied. Carbon dioxide desorption from aqueous solutions of sodium hydrocarbonate is investigated and the influence of chemical equilibria in the solutions on the cultivation of carbon dioxide supply is discussed. It is pointed out that these solutions can be considered to be carriers of the assimilable carbon source for the microalgae cultivation, most efficient at pH 8.5. A comparative estimation of the carbon dioxide and the hydrocarbonic ion as assimilable components of the chemical equilibria under study was done, leading to the conclusion that the more probable component in an alkaline medium autotrophic microalgae cultivation is carbon dioxide. A scheme of bisource feeding of the photosynthetic process, with carbon dioxide for the autotrophic microalgae cultivation in a alkaline nutrient medium, providing a stable and economical cultivation process, is proposed.

    Notizen: The thermoacidophilic strain TAC1 was isolated from a sulphatara field. It grows heterotrophically on a synthetic medium, containing yeast extract and carbohydrates at 50 to 70°C and pH 2 to 5. The cells are motile spore-forming rods utilizing, for instance, glucose, lactose, or sucrose as carbon substrate. They also used concentrated whey as nutrient medium.The maximum specific growth rate calculated from batch culture data of the strain TAC1 on glucose is 0.9 h-1 at 65°C and pH 3. The yield coefficient determined in a chemostat culture of the strain TAC1 on glucose, is 0.15 to 0.31 grams of cells produced per gram of glucose consumed (63 to 70°C, pH 2.2 to 4.0, dilution rate 0.2 to 0.4 h-1).The lipid fraction extracted from the cells consists of 72 to 93% of ω-cyclohexyl C17 and C19 fatty acids. The composition of lipid fraction varied with the pH value and the dilution rate but not with the temperature.In regard to the morphology and physiology of the isolated strain as well as the high percentage of ω-cyclohexyl fatty acids of the cell material, the strain TAC1 is similar to Bacillus acidocaldarius.

    Notizen: The paper deals with the pectolytic enzymes immobilization on different acrylic supports, and the application of immobilized preparations in the apple juice pectinization process.The correlation between the protein content (Cp) and specific catalytic activities of immobilized enyzme preparations suggest a specific immobilization process only in the case of PONILEX ASH type acrylic supports.The active immobilization degree on PONILEX ASH type supports of Ultrazym 100 G and technical pectinase extract ranged from 99.80 to 296.40% for the Pectinesterase (PE) activity, and from 101.85 to 252.94% for the chain splitting (CS) activity, proving that the ionic immobilization process is a selective one. The simulated operational stability of the immobilized pectolytic enzymes tested by the PE and CS activity values proves the preservation of enzyme catalytic activity.

    Notizen: Biotransformation of furfural by Saccharomyces cerevisiae 354 was performed in a sugar cane molasses medium containing salts. The furfural added was rapidly reduced to furfuryl alcohol until a final addition of 3%, without inhibition in the cells growth.     Notizen: Several thermophilic strains similar to Thermus sp. were isolated from a hot spring in Nha Trang (Vietnam), and a domestic hot water reservoir, respectively. They grow aerobically on a complex medium at a temperature of 55 to 70°C and a pH value of 7 to 8. The cells are gram-negative, non-motile and non-sporulating. They possess yellow or red pigments. The isolated strains are similar to Thermus sp. in regard to the composition of the intracellular fatty-acid fraction as well as the absorption spectra of the extracted cellular pigments.

    Notizen: Thermostable and hydrophobic β galactosidase of Bacillus sp. TP32 is fixed on zeolithes of controlled hydrophobicity. Thereby, the Si/Al ratio of the zeolithe is an important quantity for controlling the zeolithe hydrophobicity and with it the rate and efficiency of enzyme fixation. The fixation properties improve from NaY via Us(w) to mordenite and Us-Ex.Mordenite β galactosidase is highly active in SOERENSEN phosphate buffer, dodecanole, and ethylene glycole at 70°C using ortho nitrophenyl β D-galactopyranoside (ONPG) as substrate. Further preparations of zeolithe β galactosidase have more or less disadvantages in regard to hydrolysis of ONPG at elevated temperature, as well as rate and efficiency of enzyme fixation exept of Us-Ex β galactosidase in ONPG-containing dodecanole.

    Notizen: A new isolated Bacillus strain was cultivated continuously at 68°C and pH 3 for a long time. Extractable lipids represent about 3.6% of cell dry weight and are made up of 18% neutral lipids, 39.3% glycolipids, and 42% acidic lipids. Main components of the fatty acid fraction are ω-cyclohexylundecanoic acid and ω-cyclohexyltridecanoic acid, respectively. MK-7 and hop-22(29)-ene are components of the unsaponifiable fraction. The glycolipid fraction contains the pentacyclic triterpenoid tetrahydroxybacteriohopane.Based on the thermoacidophilic growth conditions, the morphologic and physiological properties, and the nature of lipids, it can assumed that the new isolated strain belongs to the species Bacillus acidocaldarius.

    Notizen: The sequence of the effects of delayed and immediate sealed silos as well as the role of nutrient additives such as molasses and/or urea in the ensiling process of rice hulls and the subsequent nutritive value of the product has been shown.There were considerable variations in the chemical constituents during the ensiling of rice hulls among the different treatments under both delayed and immediate sealed silos. In all cases, when compared with the controls, the crude protein, soluble sugars, lactic acid, and fat contents were markedly increased, while the carbohydrate polymers and silica contents were significantly decreased. It was concluded that the treatment of rice hulls + 2% molasses for two months ensiling period under anaerobic conditions can be considered as the optimum conditions for the production of good quality silage from the low grade roughages rice hulls. Changes in pH values also confirmed the improving effect of the combination of molasses addition and the immediate sealing of the silos.

    Notizen: The identification and enumeration of yeasts and the effect of chemical preservatives on the yeast load in Nigerian palm wine have been studied. Yeast found largely belong to the genus Saccharomyces. Other genera found were Candida, Endomycopsis, Hansenula, Kloeckera, Pichia, Saccharomycoides and Schizosaccharomyces. The viable yeast count ranged from 0.5 × 107 cfu/ml to 4.2 × 109 in both fresh Elaeis and Raphia spp. of palms. Yeasts in palm wine were least sensitive to sodium nitrate and most sensitive to sodium benzoate. The addition of these two chemical preservatives amongst others reduced the yeast count to 3.2 × 108 (in Raphia sp.), and 4.1 × 109 (in Elaeis sp.) with 0.025% sodium nitrate and 1.2 × 106 (in Raphia sp.) and 1.9 × 107 (in Elaeis sp.) with 0.1% sodium benzoate. The values of the yeast count in bottled and fresh palm wine were between 1.3 × 103 cfu/ml to 9.8 × 106 and 0.5 × 107 to 4.2 × 109, respectively. Maximum values of actual dry and theoretical yeast weights were 1.09 and 42 mg/ml, respectively.

    Notizen: Maximum production of single-;cell protein (SCP), biomass, chemicals or fuels from microorganisms demands the manipulation of environmental conditions. Consequently, single-;cell protein production was carried out using Candida sp. and pawpaw fruit pulp extract (substrate), under conditions of varying initial inoculum, different agitation rates, nitrogen sources, and heat treatments. The highest viable cell count of 8.00 ± 1.34 × 1010 colony forming units (cfu)/ml was obtained with substrate supplemented with ammonium sulphate and the least viable cell count of 7.10 ± 2.10 × 106 cfu/ml was observed using urea treated substrate. An optimum viable cell count occurred with an initial inoculum of 5.60 × 105 cfu/ml, conditions of non-sterilization and agitation at 200 r.p.m. Growth also peaked at 24, 48 and 72 h with varying treatments.

    Notizen: A recently described vesicular packing material (VP) representing clusters of microcapsules (derived from plant cells) was tested with respect to its application for protein purification. Protein elution behaviour was investigated with 28 defined proteins and several protein containing preparations and biological fluids. All proteins were eluted with a neutral buffer without retardation as peaks in the permeable or excluded fraction. Due to its sharp separation limits VP can be used for the separation of proteins with small differences in size. In special cases, proteins of nearly equal molecular weight (e.g. carboxypeptidase A and pepsin) may be separated due to differences in the electrical charge of the protein molecules and resulting differences in the electric in the electrical charge of the protein molecules and resulting differnces in the electric interaction with the negatively charged polygalacturonan matrix of the vesicle membrane (cell wall).Vesicle chromatography is a biocompatible process. The VP may be applied on a large scale. Complete separation between excluded and permeable proteins may be reached if the columns are loaded with concentrated protein samples (e.g., blood plasma). Size fractionation by the VP seems to be applicable in the following fields: 1Preparative separation of an excluded protein from an excess of permeable macromolecules, especially if the difference in STOKES' diameter is too small for an effective separation by gel chromatography or conventional membrane techniques.2Preparative separation of permeable proteins from an excess of excluded proteins.3Chromatography of proteins in the presence of alcohol, polyethylene glycol or detergents.

The specific activities of the synthesized enzymes were higher in immobilized protoplasts than in both free and immobilized mycelia.Immobilized protoplasts show an enhanced rate of exocellular β-glucosidase production compared to intact mycelia due to the lack of cell wall. The ratio of the exocellular/intracellular β-glucosidase was 5.9 for immobilized protoplasts and 0.32 for free mycelia.

    Notizen: The protein synthesis by Cephalosporium eichhorniae on substrates containing starch was evaluated at different pH, moisture content, ammonium sulphate and potassium dihydrogen phosphate supplementation. The optimum pH level was about 4.2 and the moisture content 65%. The optimum level of supplementation of medium containing sweet potato substrate with (NH4)2SO4 was smaller than that for cassava substrate (5.2 g/l 〈 6.7 g/l in submerged culture, and 4.0% 〈 5.2% in solid state fermentation). The total crude protein yields were about 7.6 g/l for submerged cultures and 12% DM for solid state fermentations.

    Notizen: The regulation mechanism governing the xylanolytic activity in the strain Cellulomonas sp. IIbc was studied. High levels of activity were detected during the cultivation on cellulose as the only carbon source. No activity was produced with glucose, xylose or cellobiose cultures, but in the last one, the synthesis was de-repressed when the sugar concentration dropped to 0.2%. The activity was not inhibited by glucose, cellobiose and xylose up to 1% concentration.A basal constitutive synthesis was detected in nutrient broth cultures. At the same time, xylose and cellobiose acted as inducers of the xylanase activity.

    Notizen: Cellulase from four different fungi and β-glucosidase from almonds were immobilized on the inner surface of nylon tubing. The highest values of β-glucosidase activity retention on the support were obtained when P. funiculosum and N. crassa were used as the enzyme source. A comparative study of the thermal stability referring to β-glucosidase activity was developed using free and immobilized enzymes. The most stable β-glucosidases (from P. funiculosum and A. niger) did not show an appreciable change in its thermal stability after immobilization. An important increase in thermal stability was observed when less stable β-glucosidases (from T. reesei, N. crassa and almonds) were immobilized.

    Notizen: To understand and control the dynamics of microbial growth and metabolism, a theoretical background based on segregated models is necessary, and therefore flow cytometry is the suitable measuring method.It is shown that additional information to the usual mean-value information, and to the well-known cytometric methods for determining DNA content and size/shape measurement by scattered light, like the distribution of membrane sterol content of baker's yeast, leads to a more detailed knowledge of growth dynamics.We have found, that a low content of membrane sterols is a characteristic of cells with suppressed proliferation activity, but necessary for high fermentation activity. Also, a high content of membrane sterols seems to enhance the viability and survival capability of harvested and dryed baker's yeast cells. Therefore, we recommend harvesting cells at a point of medium content of membrane sterols.

    Notizen: The most frequently used sources of more stable enzymes are thermophilic bacteria, e.g. Bacillus, Closrridium, and Therrnus strains, occurring in natural as well as man-made habitats. They grow from 55 to 88°C with a specific growth rate of up to 2.6 h- and a yield coefficient of up to 0.4 gram of dry cell weight per gram of carbohydrate consumed. Several thermophilic strains, e.g. Bacillus sp. TP32, rapidly and effectively produce enzymes having a higher thermal stability and resistance to chemical denaturants in comparison to their mesophilic counterparts.Therefore, thermostable enzymes are of importance for bioorganic syntheses. For the further optimization of enzyme production, genetic engineering is applied.

    Notizen: This paper prsents the results of teh study of rheological behaviour of antibiotic biosynthesis liquids obtained by submerged aerobic cultivation of microorganisms belonging to the actinomycete and fungi classes, in stirred tank bioreactors with turbine impellers. These liquids have a non-Newtonian behaviour which follows the power-law rhcological model with a correlation index of over 0.95.The studied liquids are pseudoplastic, and alter their rheological properties, such as consistency index, (K), flow index, (n), apparent viscosity, (ηa), maximum Newtonian viscosity (η0), with the culture age, microrganism strain and batch conditions. Also, these liquids are time dependent, exhibiting thixotropy.The most viscous liquids are produced by Streptomyces aureofaciens and Streptomyces rimosus cultivation, while that produced by Streptomyces griseus is the least viscous. A higher pseudoplasticity appears after 30 hours culture age.Since all these biosynthesis are aerobic, a careful observation of the rhelogical behaviour dynamics is necessary to avoid the oxygen culture supply limitation and the decrease of the bioreactor performance during biosynthesis.

We reported the development of mycelial thick threads of A. bisporus in the casing soil.Moreover, we pointed out the protective role of the calcium salts crystals surrounding the hyphae during the incubation stage. On the electron-micrographies, the mycelial aggregates were surrounded by bacteria, and this bacterial growth seemed to be related to the calcium salts crystals disappearance.The positive influence of A. bisporus upon bacterial growth, and vice versal, was confirmed with PETRI dish cultures.

    Notizen: Logistic equations are suitable for describing microbial growth. By means of VERHULST'S logistic equation, the adaptation to sigmoid-shaped curves of growth improves with a falling ratio Cxo/Cx, max 〈 0.2, if there is no lag-phase. The known logistic equations do not take into account any lag-phase behaviour, so that noticeable deviations in the model adaptation result in this range. Therefore, an extended logistic equation of rate is proposed by which any occuring lag-time is expressed by a 1st-order lag-term. The corresponding time law allows a very good adaptation of curves of delayed growth behaviour, and changes into VERHULST'S logistic equation for a lag-time tL = 0. Application is facilitated by instructions for the numerical determination.

    Notizen: The lysine content of the biomass of the acidophilic facultatively methylotrophic bacterium Acetobacter methanolicus MB 58 was increased by genetic manipulations.A homoserine auxotroph, MB 58.196, and a threonine auxotroph, MB 58.195, were obtained from Acetobacter methanolicus MB 58 by N-methyl-N′-nitro-N-nitrosoguanidine treatment.Investigations of enzyme activities revealed that the homoserine auxotroph lacks homoserine dehydrogenase activity, and the threonine auxotroph lacks homoserine kinase activity.Concerning the lysine-producing ability, only the homoserine auxotrophic mutant accumulates lysine in the intracellular pool. The intracellular lysine content of this mutant was increased 40-fold.An excretion of amino acids into the medium was not detected. A homoserine resistant mutant, MB 58.196.10, isolated from MB 58.196 by UV-irradiation, was able to excrete lysine. About 95% of free lysine were found in the culture medium. Altogether, the free lysine concentration was increased 800-fold in comparison to the wild-type strain.By these genetic manipulations the total lysine concentration of MB 58.196 was increased to 2.7% and of MB 58.196.10 to 56% in comparison to the wild-type strain.

    Notizen: The last decade saw remarkable progress in the quantitative and qualitative improvement of microorganisms applied in the foodstuff industry because methods of genetic engineering were used. Compared to the classic methods of mutagenesis and hybridization, genetic engineering facilitates the changing of genes coding for certain products or certain qualities and their new combination with other genes in either the donor or any other host cell.Success achieved so far in the changing of microorganisms by mutagenesis, hybridization and genetic engineering is explained with some examples. These refer to microorganisms used in the production of enzymes, amino acids, edible organic acids, aromatic and similar products.

    Notizen: The effects of temperature, pH and ethanol on the stability of killer wine yeasts were studied. Laboratory and collection strains as well as the strains prepared by the protoplast-fusion technique were used and a decline of killing activity was observed in the complete medium and in grape juice. The relationship between the loss of killer properties in fusant strains and the stability of the original donor strain was established. The negative effect of the ethanol addition on the killer character of the strains used and the possibilities of its stabilization are discussed.

    Notizen: Two hybridoma cell lines of human origin and two of murine origin were encapsulated using the cellulose sulfate-polydimethyldiallylammonium chloride procedure. All lines could be cultivated in capsules, but the time of growth and product synthesis were limited.The membranes formed were impermeable to albumin and transferrin over a period of 240 minutes. Therefore, the stop of cell proliferation is assumed to be caused by the lack of at least these two proteins. The cell densities inside the capsules reached a maximum of 4.1 × 106/ml (human line) and 3.9 × 107/ml (murine line). The maximum product concentration in the capsules measured by IgG- and IgM-ELISA was 0.17 mg/ml (human line) and 7 mg/ml (murine line). The calculated corresponding retention was 93% (human IgM) and 52% (murine IgG). The characterization of the product showed that the immunoglobulin was not intact and might have been destroyed during harvest. Using the same cell lines the method was compared with the alginate-PLL procedure originally developed by LIM and SUN [1].

    Notizen: The effect of formaldehyde on the cytochrome system of the methylotrophic yeasts Candida tropicalis, Candida boidinii, Candida methylica, Hansenula capsulata, Hansenula polymorpha, Pichia pastoris and nonmethylotrophic yeasts Saccharomyces cerevisiae was studied. In the investigated concentration range of 0.58 - 2.5 mol · 1-1, formaldehyde decreases the level of the reduced cytochrome c in the above-mentioned microorganisms. The kinetics of the change in the reduced cytochrome c content has two phases: a phase of slow decrease and a phase of complete disappearance of the reduced cytochrome c in the cells. The duration of the first phase shortens with increasing concentrations of formaldehyde.

    Notizen: White-rot basidiomycete Phlebia radiata grown in the nitrogen-poor liquid medium was tested for the inducing effect of 2,5-xylidine and veratric acid on laccase production and properties of its preparations. Also lignin peroxidase and manganese-dependent peroxidase were assayed under the same conditions. The maximum of laccase activity for veratric acid as a stimulator was done much earlier than that of xylidine. It was very sharp and disappeared quickly. At that time only weak lignin peroxidase and manganese-dependent peroxidase activities were noticed. The maximum of laccase induced by xylidine was observed much later and kept longer. Both laccase preparations showed the same properties. For biotechnological reasons, the production of laccase induced by the nontoxic veratric acid is much more economic and better acceptable than that induced by xylidine.

    Notizen: In the colonization experiments of yeast cells on stainless steel surfaces the analyzed minimum of the colonization rate was found in a roughness interval between a highly polished sample and a very rough surface. This phenomenon is a problem of the shear forces and of adhesion.On the basis of reaction kinetics it was possible to analyze the change between an unstable and a stable cell-cluster. The analysis of the energy needed for forming a stable cluster tends toward values known from the stochastic simulation of cluster formation in an undisturbed region. But the potential function is not clear which describes the interactions between the cells and the forming cluster in the reaction field. This is a problem of special materials-testing which is not established.For producing plants it is of high interest to note that it are not the most highly polished surfaces that are the best.

    Notizen: The nutritional conditions for the germination of spores of Streptomyces galbus 5ME-13 were determined under laboratory conditions. The germination of the spores was intiated by the emergence of 1-2 germ tubes after the second hour of incubation and attained its maximum at the sixth hour. This was accompanied by a steady rise in the optical density of the germinating spore suspension. A malt-extract yeast-extract medium was found to be the best medium for the germination of the spores.Glycerol as the sole source of carbon was the best supporter for spore germination while, as N source, L-alanine was preferred. The optimum pH and temperature for spore germination were 7.2 and 30°C, respectively.

    Notizen: High-loaded pectate gels containing more than 1014 cells/l of gel (viability 〉 60%) were obtained. Immobbilizates of Lactobacillus sp. IMET 11466 showed a maximum mean gel activity of 120 g of lactate/l of gel * h. With the strain Streptococcus thermophilus Č 129 a value of 139 g of lactate/l of gel * h was attained. Using these immobilizates the conversion of 100 g of glucose/l with a yield of 90-95% can be realized in only 6-8 h. On the basis of a detailed analysis of the time course of the lactate-forming activity of the immobilizates during batch fermentations evidence is given of an analogy in the lactate-inhibitory behaviour of immobilized and free cells.

    Notizen: The recovery of bacterial extracellular alkaline and neutral proteinases was investigated using mineral hydrocolloid bentonite in combination with the organic flocculant Hernostan B for the removal of solid compounds and cells from fermentation broth. The flocculation was effective in the pH range of 7.0-8.5 at concentrations of bentonite of 2.0-5.0 g · dm-3 and Hernostan B of 1.0-5.0 g · dm-3. The results show that, by means of this technique, it is possible to achieve a yield of proteinases above 90% on a laboratory, and up to 85% on a pilot plant scale.

In batch culture the revertant showed characteristics which distinguished it both from B. stearothermophilus strains lld-15 and NCA 1503.

    Notizen: In fodder yeast production from sulphite spent liquor, sulphur dioxide is well-known as a yeast poison. The present paper deals with the detailed influence of sulphur dioxide.The experimental work was done in a laboratory fermenter under technical conditions. A decrease in biomass yield, and an increase in oxygen consumption, caused by increasing carbon dioxide production, were found.During the fermentation, there is no destruction of the carbonyl-bisulphite adduct to be observed. Sulphur dioxide has no influence on the crude protein content of yeast cells, and there is also no influence on the morphology of cells.
