    Notes: Abstract End-tidal $$P_{CO_2 } $$ $$\left( {P_{ET_{CO_2 } } } \right)$$ and ventilation of the rat anesthetized with halothane were measured. The $$P_{ET_{CO_2 } } $$ measured by an infrared analyzer agreed well with the simultaneously measured $$Pa_{CO_2 } $$ in the range from 20–60 mm Hg (2.7–8.0 kPa). When the level of anesthesia was deepened by increasing the halothane concentration from 0.9–3.0%, minute ventilation was decreased progressively accompanied by a rise in $$P_{ET_{CO_2 } } $$ and the CO2 output was reduced. Halothane induced a progressive decrease in frequency of respiration (f) with almost constant or even slight increase in tidal volume (V T). Decrease inf was caused largely by the prolongation of expiratory duration (T E). Changes in inspiratory duration (T I) were small and the meanV T/T I ratio remained unaltered at different levels of anesthesia. These changes in respiratory pattern induced by halothane anesthesia contrasted with the simultaneous decrease inV T,f andV T/T I ratio and apparent changes of bothT I andT E reported in other species. Species differences in effects of anesthesia on ventilation were discussed. Restraining the rat on a stereotaxic apparatus with head holders, especially with ear bars, elicited an initial transient stimulation of ventilation which was followed by a strong depression.

    Notes: Abstract The uptake ofl(+)lactate into rat renal cortical brush border (BBV) and basolateral (BLV) membrane vesicles, isolated through differential centrifugation and free flow electrophoresis, were studied using a rapid filtration technique. In contrast to the lactate transport into the BBV, that into the BLV: 1) was found to proceed only towards equilibrium, 2) showed Na+-independent coupling of the influx ofl(+)lactate and the efflux ofl(+) but not to the efflux ofd(−)lactate, 3) was not inhibited byd(−)lactate, 2-thiolactate or 3-phenyl-lactate, but 4) was inhibited by 3-thiolactate and α-hydroxybutyrate and 5) was accelerated by changes in inwardly directed ionic gradients or by increases in cation conductance both of which led to increased intravesicular positivity. The latter changes had the opposite effect on the uptake ofl(+)lactate by BBV. Thus, while thel(+)lactate transport system present in BBV showed the characteristics of Na-dependent electrogenic cotransport system, that in the BLV was consistent with a carrier mediated Na-independent, facilitated diffusion system.

    Notes: Abstract Plasma titers of erythropoietin (Ep) are known to increase initially during hypoxia and to return then towards prehypoxia values. To find out if this pattern of plasma Ep might be related to changes in the production of the hormone, I have compared plasma with kidney Ep titers in hypoxic rats. Rats were exposed to hypoxia in a hypobaric chamber at 0.42 atm for various time intervals for up to 4 days. Kidney Ep titers were assayed in extracts from kidneys that had been flushed free of blood in situ. It was found that kidneys of normal rats do not store significant amounts of Ep. Kidney Ep titers increased transiently during hypoxia. They reached maximum values after 6h and then declined to almost undetectable levels at continued hypoxia. In the plasma, maximum values were found after 12–18h of hypoxia. Additional studies were done on the effects of discontinuous hypoxia. It was found that, even after 3 days of previous hypoxia exposure, plasma and kidney Ep titers increased again in rats when these were maintained intermittently in normoxia for 18 h. It is concluded that the rise and fall in plasma Ep titers during hypoxia reflect similar changes in kidney Ep production.

    Notes: Abstract Mucus secretion into the rat colon has been measured in situ using a single perfusion technique. Protein, sialic acid and hexose concentrations in the perfusion solution were found to give reliable estimates of mucus output if samples were homogenized prior to analysis. Mucus output as indicated by an increase in the concentration of mucus constituents was higher when the solution was hypotonic (270 mosm·kg−1) or hypertonic (370 mosm·kg−1) than when isotonic solutions (320 mosm·kg−1) were used. The proportion of hexoses and sialic acid to protein was 23 and 14% at low, 23 and 11% at high osmolality, and 21 and 13% when isotonic solutions were used. Deoxycholic acid (DCA, 4 mmol·l−1) increased the net secretion of mucus constituents 3 fold, whereas short chain fatty acids (SCFA) had no effect. Mucus composition during all treatments did not change significantly, even when stimulated with DCA. When mucus was released from the epithelial surface by previous perfusion with a DCA containing solution, net water and SCFA absorption rates and mucus output were significantly lowered for 2 to 3 h. However, no correlation between mucus secretion and SCFA absorption was found, indicating that a role for mucus as a diffusion barrier to SCFA is unlikely. Mucus output, which indicates the amount of mucus released from the epithelial surface, probably depends on the direction of net water movement, which follows the osmotic gradient between colon lumen and blood.

    Notes: Abstract The ventilatory response to CO2 in rats under sodium pentobarbital anesthesia has been measured using the rebreathing technique. The animal rebreathed through a tracheal cannula for a period of 4 min from an apparatus of 200–400 ml capacity, containing 5–6% CO2 in O2. $$P_{{\text{CO}}_{\text{2}} } $$ in the rebreathing apparatus (PappCO2), instantaneousV T,f, and $$\dot V_{\text{E}}$$ were monitored before, during, and after rebreathing. During the rebreathing run,PappCO2 andPa CO2 rose linearly from 35–40 to 65–70 mm Hg; there was no significant difference betweenPappCO2 andPa CO2 at any time during rebeathing.V T and $$\dot V_{\text{E}}$$ increased almost linearly with the rise inPappCO2, whilef increased to a maximum within 2 min of rebreathing. In the rat,V T regulation seemed to operate exclusively as a proportional control system in response to linearly increasing CO2 stimulus. The slopes ofPappCO2,V T or $$\dot V_{\text{E}}$$ response curves varied considerably during the time course of the experiment, depending upon the level of anesthesia, even though there was no large change in $$\dot V_{\text{E}}$$ in the control periods which were under hyperoxic conditions. However, a significant linear relationship was seen betweenf in the respective control period and the slope ofPappCO2-V T response at various levels of anesthesia. We concluded that the rebreathing technique can be applied in small experimental animals and that changes in the sensitivity of the respiratory control system to a CO2 stimulus by anesthesia can be easily monitored by repeating the rebreathing test.

    Notes: Abstract The in vivo rates of protein synthesis in the peripheral nervous tissues of methylmercury-treated rats (10 mg/kg/day, for 7 days) have been estimated with improved methods by the injection of a large amount of [l-14C]valine of low specific activity. Protein synthesis activity in the dorsal root ganglia was inhibited to the extent of 60% of the control as early as day 5 and this continued to the symptomatic period (day 15) on which crossing of hind limbs, a typical sign of organomercurial poisoning, was observed in the animals. The sciatic nerves and dorsal roots increased protein synthesis by 56% at the symptomatic period. These increases in protein synthesis may be due to the stimulation of reactivity of Schwann's cells. On the contrary, the protein synthesis in the ventral roots showed a gradual decrease as the intoxication proceeded and decreased to 73% of the control at the symptomatic period, being similar to the case of the brain. The double-labeling studies with sodium dodecyl sulfate/polyacrylamide gel electrophoresis exhibited that methylmercury inhibited the synthesis of the dorsal root ganglion proteins non-uniformly in various apparent molecular sizes, especially on day 10.

    Notes: Abstract Radioselenium concentration and excretion was studied after administration of 75Se-labelled selenite to male rats during ontogeny. The concentration of radioselenium in individual organs decreases with increasing age. The largest differences between young and adults were in the quantity and quality of excreted substances. During 2 h after the administration of 20 μmol selenite/kg young rats excreted 2.4% of the dose, essentially in the urine only, whilst adults excreted a total of 11%, distributed equally in breath and urine. The part excreted as methylated metabolites was 0.1% of the administered dose in young and 6.3% in adult rats. These results support the hypothesis that the differences in the sensitivity to the toxic action of selenite between young and adult rats can be due to ontogenic differences in selenium metabolism.

    Notes: Abstract Both in the intestinal copper complex and hepatic zinc-thionein of the newborn rat provide immediately available, high-affinity binding sites for Hg2+. Retention of orally administered Hg2+ in the gastrointestinal tract is much greater in the 5-day old rat than in the weanling and accounts for most (70–90%) of the body burden at 18 h. At a dose level of 790 μg Hg2+/kg body weight, 41% of the Hg2+ retained in the gastrointestinal tract of the newborn animal is located in the soluble fraction of the mucosa and 48% of this is bound by the copper-complex. This complex, therefore, seems to limit the transfer of Hg2+ from the mucosa to the serosa. It does not appear, however, to be a primary acceptor of Hg2+ in the neonatal intestine since, after administration of 100 μgHg2+/kg body weight, its binding of Hg2+ is less (33% of the soluble Hg2+) than that (67% of the soluble Hg2+) of the high molecular weight protein fraction. At 18 h after the intraperitoneal administration of Hg2+ (790 μg Hg2+/kg body weight), whole body retention is greater in the adult (80% of the dose) than in the suckling or weanling animal (63–68% of the dose). In the suckling animal the inter- and intra-organ distributions of the retained Hg2+ also differ from those in the weanling and/or adult rat. Thus the ratio of the renal: hepatic Hg2+ contents, after either intragastric or intraperitoneal doses of Hg2+, increases with age. Also most (approximately 90%) of the Hg2+ retained in the liver of the newborn animal after an intragastric dose is bound by the metallothionein whereas, in the adult liver, only 4% of the total Hg2+ is in this form. The presence of the metallothionein, however, is not the cause of the increased Hg2+ uptake in the former, since induction of hepatic zinc-thionein in the adult by pretreatment with Zn2+ does not affect the distribution of Hg2+ between the liver and kidneys. These findings are discussed in relation to the known differences in intestinal absorption and tissue distribution of Cd2+ and Hg2+ in newborn and adult animals and the greater resistance of the former to Hg2+ nephrotoxicity.

    Notes: Abstract Dimethylphthalate (DMP), dibutylphthalate (DBP) and di(2-ethylhexyl)phthalate (DEHP) were given i.p. (3.8 mM/kg) to Sprague Dawley rats for 5 days. DBP increased significantly the liver concentration of cytochrome P-450, but decreased the lung concentration by about 40%. DBP decreased the lung concentration of cytochrome b5 and NADPH-cytochrome-c-reductase activity by about 30%. Only minor effects were seen after treatment with DMP and DEHP. The direction of B(a)P metabolism was changed and the formation of 2- and 3-hexanol metabolites were increased in liver microsomes after DBP treatment. All phthalate esters decreased the lung metabolism of B(a)P. The cytochrome P-450 enzyme system in the lung was ten times more effective than that in the liver as far as metabolism of n-hexane was concerned. Only minor effects were observed in serum enzyme activities, but a significant decrease in the serum level of albumin was observed after treatment with DBP. No relationship was found between the carbon chain length of the investigated chemicals and effects on microsomal enzymatic activities.

    Notes: Abstract The acute renotoxicity of HgCl2 and phenylmercuric acetate (PhHgAc) was compared at two intraperitoneal dose levels: 0.5 and 1.0 mg Hg/kg. There was no difference in the type of proximal tubular damage caused by the two mercurials, but 1.0mg Hg/kg as PhHgAc produced approximately the same degree of damage as 0.5 mg Hg/kg as HgCl2. At the selected dose levels only HgCl2, but not PhHgAc increased the urinary excretion of alkaline phosphatase. At 12 and 24 h after PhHgAc the content of mercury was higher in blood and lower in the kidneys and urine than after the administration of equimolar doses of HgCl2. As the difference in the renal mercury contents of HgCl2 and PhHgAc treated groups declined with time, difference in renotoxicity seems to relate only to renal mercury taken up within 24 h of administration. It is suggested that the slower renal extraction of mercury — as in regenerating kidneys (Tandon and Magos 1980) — was responsible for the lower degree of renotoxicity in phenylmercury treated rats.

    Notes: Abstract Two comparative experiments on the behavioural, audiometric and histological effects of kanamycin-induced cochleotoxicity in the Wistar rat are reported. In the first experiment kanamycin 400–1,500 mg/kg/day was injected subcutaneously for 20 days and the morphological damage to the organ of Corti assessed. In the second experiment the progression of damage to the organ of Corti was examined in animals given kanamycin 700 mg/kg/day for 4–20 days. Behavioural audiometric studies of threshold shift were undertaken throughout both experiments. In the first study, all the animals were killed after a recovery period of 20 days from the last injection, i.e., day 40, and in the second study groups of animals were killed at 4-day intervals between days 4 and 20 of dosing. One cochlea from each animal was critical point-dried, dissected to expose the organ of Corti and examined by scanning electron microscopy. A vertical section through the contralateral cochlea was examined by light microscopy. The results of the morphological examination of the cochleas were collated with the behavioural audiometry. The morphological damage to the organ of Corti followed a stereotyped pattern of degeneration, the extent of which appeared to be determined both by the number and concentration of the kanamycin administrations. The collateral audiometric examinations indicated that extensive damage had taken place before a shift in the behavioural auditory threshold could be detected by observation of the Preyer reflex.

    Notes: Abstract The effect of neonatal administration of zearalenone on the female reproductive system was studied in the rat. A single subcutaneous injection of 1.0 mg zearalenone to 3- or 5-day-old rats caused persistent vaginal estrus in adulthood. Ovaries in these animals contained many large follicles but no newly formed corpora lutea. The same effects were observed in rats which had received 100 μg estradiol-17β in the neonatal period. Most rats which had received 100 μg zearalenone or 10 μg estradiol-17β showed regular 4-day estrous cycles and had newly formed corpora lutea in their ovaries. These results demonstrate that neonatal exposure to zearalenone produces persistent anovulatory estrus in the rat, the potency being about one tenth that of estradiol-17β.

    Notes: Abstract Various trialkylphosphorothioates occur as impurities in many organophosphorus pesticides. In addition to the immediate, cholinergic symptoms usually associated with the toxicity of organophosphorus compounds an oral LD50 dose of O,S,S-trimethyl phosphorodithioate or O,O,S-triethyl phosphorothioate to rats, resulted in the delayed development of lung lesions and often death within 4 days. This study attributes the late crisis to impaired lung function resulting from massive alveolar oedema and cellular responses to the selective destruction of type I pneumocytes. Proliferation of type II pneumocytes produced an epithelium of very large cells which resulted in a thick blood/alveolus barrier and also occluded much of the alveolar lumen. Debris from the type I pneumocytes was liberated into the alveolar lumen resulting in leukocytic infiltration of the interstitial tissue and an increase in the number of alveolar macrophages. The lung morphology of animals surviving the crisis period returned to normal within a further 4 days with the transformation of many hypertrophic type II pneumocytes into type I alveolar lining cells.

    Notes: Abstract Male and female Wistar rats treated orally with 0.1, 1.0 or 10.0 mg/kg/day aristolochic acid as its sodium salt (AA) developed a high incidence of tumours dependent on dose and time. After 3 months' treatment 1.0 and 10.0 mg/kg AA led to severe papillomatosis of the forestomach with occasional signs of malignancy. Three to 6 months later without further treatment the rats developed squamous cell carcinomas in the forestomach with formation of metastases. At the same time anaplasia of the tubular epithelium and mainly adenomas appeared in the renal cortex. The transitional epithelium of the renal pelvis and the urinary bladder showed hyperplasia, papillomas or carcinomas. For the low dose (0.1 mg/kg) the treatment with AA varied between 3 and 12 months. No tumours were observed in the first 6 months of the study. After 12 and 16 months, however, papillomas or squamous cell carcinomas also occurred in the forestomach. In addition, hyperplasia of the transitional epithelium of the renal pelvis was found while the renal cortex and the urinary bladder remained normal.

    Notes: Abstract The distribution and elimination of di-2-ethylhexyl phthalate (DEHP) and mono-2-ethylhexyl phthalate (MEHP) after a single oral administration of DEHP (25 mmol/kg) were studied. A gas-liquid Chromatographic method was used for the simultaneous determination of MEHP and DEHP. The compounds were extracted with methylene chloride and the monoester was alkylated to the hexyl derivative by solid-liquid phase transfer catalysis in methylethyl ketone. The coefficients of variation of this method for determination of DEHP and MEHP were 8.3% and 11.4% respectively. The concentration of DEHP and MEHP in blood and tissues increased to maximum within 6–24 h after dosing, while the highest levels observed in the heart and lungs occurred within 1 h. At 6 h after administration, the highest ratio of MEHP/DEHP (mol%) were recorded in testes (210%) while the other tissues exhibited less than 100%. MEHP disappeared exponentially with t 1/2 values ranging from 23 to 68 h; DEHP t 1/2 ranged from 8 to 156 h and the t 1/2 values of MEHP in several tissues were slightly longer than DEHP. The t 1/2 values in blood were 23.8 h and 18.6 h for MEHP and DEHP, respectively.

    Notes: Abstract The activities of neurotransmitter-metabolizing enzymes were determined in the nervous tissues of rats treated with methylmercury chloride (10 mg/kg/day, for 7 days). The activity of choline acetyltransferase was lowered consistently in the cytosol and synaptosomal fractions of the brain in methylmercury-treated rats, while changes in acetylcholinesterase activity in the brain subcellular fractions were small. In peripheral nerves, decreases in activities of choline acetyltransferase in the sciatic nerve and of acetylcholinesterase in the dorsal root were most profound. Decreases in these enzyme activities started at an early phase and increased markedly with the progress of intoxication. The activity of acetylcholinesterase in the dorsal root ganglion and in the sciatic nerve was also inhibited significantly at the latent period and more profoundly at the symptomatic period at which time crossing of hind limbs, a typical sign of organomercurial poisoning, was observed in the animals. Activities of tyrosine hydroxylase and monoamine oxidase were elevated in the brain homogenate and especially in the synaptosomal fraction with respect to the former enzyme after methylmercury treatment. Effects of methylmercury in vitro on the activities of these enzymes revealed that a much higher amount of methylmercury was required to produce in vitro an inhibitory action equivalent to that observed in vivo. These results suggest that the neurotoxic action of methylmercury could be mediated, at least in part, through the level of neurotransmitter enzymes.

    Notes: Abstract The acute toxicity of trimethyl and triethylphosphorothioates present as impurities in technical organophosphorus insecticides has been examined in rats. Although the majority of these compounds caused cholinergic signs, in some a second phase of toxicity affecting the lung was also seen. Rats after recovering from the cholinergic phase of poisoning showed respiratory distress and died 3–6 days after dosing with a massive increase in lung weight, associated with progressive diffuse interstitial thickening maximal on day 4. For two compounds no cholinergic toxicity was observed and narcosis was the predominant sign of poisoning. Pretreatment with phenobarbitone, piperonyl butoxide or SKF525A protected rats against lung toxicity caused by trimethylphosphorothioate and dithioate but only phenobarbitone was effective against OOS-triethylphosphorothioate lung toxicity. Direct intrabronchial instillation of OSS-trimethylphosphorodithioate and OOS-triethylphosphorothioate, both of which had been shown to cause lung damage after other routes of dosing, did not greatly increase the toxicity of these two compounds, suggesting that they are not directly toxic to the lung and probably require metabolism.

    Notes: Abstract The morphology and ultrastructure of the thermophilic cyanobacteriumMastigocladus laminosus were examined by scanning and transmission electron microscopy. Mature cultures consisted of relatively old, wide filaments that branched frequently to form younger, thinner filaments. The cells of the younger filaments had a consistently cylindrical morphology, while those of older filaments were rounded and pleomorphic. The internal ultrastructure of the cells depended somewhat on their age. As young cells became larger and wider, their thylakoids underwent slight rearrangement and spread out toward the center of the cytoplasm. Polyphosphate bodies, carboxysomes (polyhedral bodies), and lipid-body-like structures increased in number as the cells aged, but ribosomes and cyanophycin granules were depleted. Cell division involved septum formation followed by ingrowth of the outer membrane and sheath. Cells in older filaments were separated from each other by a complete layer of sheath material. Septum formation in older cells was also seen to occur parallel to the long axis of the filament, thereby confirming that true branching took place.

    Notes: Summary Covalent binding of 3H-labeled adriamycin metabolites to bovine serum albumin and microsomal protein is demonstrated in an aerobic incubation system with rat liver and rat heart microsomes, respectively, using exhaustive organic solvent extraction and gel chromatography. Covalent protein binding was dependent on active microsomes, NADPH, and oxygen and was inhibited by reduced glutathione and other sulfhydryl compounds. The anthracycline moiety was spectrophotometrically evidenced in the adriamycin metabolite(s) covalently bound to protein. Thus, enzymatic activation of adriamycin in the heart with consecutive covalent protein binding of reactive adriamycin semiquinone radicals may contribute to adriamycin cardiotoxicity.

    Notes: Summary The anti-histaminic drug methapyrilene hydrochloride, which induces liver tumors in rats, was labeled with tritium by exchange and administered at a dose of 21 mg containing 700 μCi to each of 15 male Fischer rats. At 1 h, 6 h, 14 h, 24 h, and 44 h after treatment three rats were killed and their livers, pancreas, kidneys, and lungs were removed. The pooled organs were homogenized and DNA, RNA, and soluble protein were isolated from each. The extent of interaction of radioactive methapyrilene with liver nucleic acids was exceedingly small and did not differ significantly from the binding to nucleic acids in kidney, lung, or pancreas, which are not target organs of this carcinogen in rats. Binding of radioactivity to soluble proteins of the liver was considerable and substantially greater than in the other organs. If the mechanism of carcinogenic action of methapyrilene involves covalent interaction with DNA this must be at a very low and highly specific level.

    Notes: Summary Bleomycin was injected i.p. in rats, and the amount of expired ethane which indicateds lipid peroxidation was followed up for 78 h. Compared to controls neither 1x30 mg/kg and 2x30 mg/kg nor 1x70 mg/kg bleomycin led to increased ethane expiration, although body weight loss indicated toxicity. That pulmonary toxicity had been developed due to the acute bleomycin treatment could be demonstrated by histological examinations of lungs of the animals of the highest dosage group. The combined treatment of rats with bleomycin and ferrous ions neither resulted in an increase of ethane expired compared to that of the ferrous iontreated animals. Rather a decrease was observed. Our results indicate that acute bleomycin toxicity is not associated with increased lipid peroxidation. Furthermore, our data suggest that the bleomycin-ferrous-complex does not initiate lipid peroxidation in vivo.

    Notes: Summary Autochthonous neurogenic tumors of the rat induced by transplacental application of ethylnitrosourea were used for the first time to study their suitability as tumor models for experimental chemotherapy. Of 189 transplacentally treated rats, 87% developed neurogenic tumors. After the initial clinical diagnosis of a neurogenic tumor, additional malignant tumors often occurred. The mean number of neurogenic tumors from 62 untreated control rats increased from 1.0 per rat at the time of randomization to 1.2 as revealed by autopsy and 1.5 tumors by histological examinations. Out of all neurogenic tumors, tumors of the brain were observed in 31%, tumors of cranial nerves in 36% (90% tumors of trigeminal nerve), tumors of spinal cord in 21%, and tumors of peripheral nerves in 10%. The median survival time until natural death of 62 control rats was 228 days. Rats with tumors of peripheral nerves lived shortest, followed by rats with tumors of cranial nerves, tumors of the spinal cord, and brain tumors. Brain tumors were mainly astrocytomas and oligodendrogliomas. The survival time of untreated rats from randomization to natural death was longest for those with brain tumors, followed by tumors of peripheral nerves, cranial nerves, and tumors of the spinal cord. There was great variation in survival time from a few days to more than 6 months. To study the responsiveness to chemotherapy, 62 rats received BCNU as a single intravenous dose of 9 and later 10 mg/kg. Sixty-two untreated control rats had a median survival time of 36 days (95% confidence interval 26–52 days), the treated rats 43.5 days (26–62 days) The difference was not statistically significant. BCNU produced a remission or a no change of neurologic symptoms in 60% (37 out of 62) in comparison to 39% (24 out of 62) in the control group (p〈0.05). The advantages and disadvantages of the present models are discussed. Due to methodical problems and the marginal response to BCNU, autochthonous neurogenic tumors of the rat are not suitable as models for chemotherapeutic studies.

Die Ultrastruktur der quantitativ meist noch geringen Lipidspeicherprozesse in Gehirn oder viszeralen Organen glich qualitativ jener der postnatalen Speicherprozesse: GM2-Gangliosidose Type 2; Nachweis von „menbranous cytoplasmic bodies“ lysosomalen Ursprungs in Fortsätzen von Nervenzellen des Groß-hirns. Morbus Krabbe; Auftreten von einkernigen und mehrkernigen Speicherzellen, z. T. mit Gefäßbeziehung, im Rückenmark. In den Speicherzellen traf man auf spieß- oder lamellenförmige, teils auch fädig strukturierte Einschlußkörper. GM1-Gangliosidose Typ 1; in Nervenzellen des Hirnstamms Vorkommen intrazytoplasmatischer lysosomaler Speicherkörper vom „Zebra“-Typ, in der Milz fanden sich durch zahlreiche, kaum strukturierte Vakuolen geblähte Speicherzellen. Metachromatische Leukodystrophie; im Gegensatz zu Literatur-Befunden waren keine Speicherprozesse im fetalen Hirn und Rückenmark, jedoch starke Speicherungen in den Nierentubuli in Form multilamellärer (zirkulär, parallel oder unregelmäßig geschichteter) Speicherkörper, teils mit Prävalenz in den Zellen mit Mikrovilli, nachweisbar. Fluoreszenzmikroskopisch war metachromatisches Material in Nierentubuli und Sammelrohren, ferner auch in der Leber (Grenze der Glissonschen Dreiecke zum Parenchym) darstellbar. Morbus Niemann-Pick Typ C; große Nervenzellen des Rückenmarks und der Stammganglien enthielten zahlreiche Myelinfiguren-artige Einschlußkörper, die den postnatalen lysosomalen Speicherkörpern in Nervenzellen bei dieser Erkrankung ähneln. Morbus Gaucher; der erst 19 Wochen alte Fet zeigte trotz biochemisch eindeutigen Defekts der Glucocerebrosidase-Aktivität noch keine Speicherphänomene. Die bereits pränatal oft deutliche morphologische Manifestation der Speicherprozesse bei Neurolipidosen zeigt, daß die postnatale Latenz der klinischen Erscheinungen während mehrerer Monate (bisweilen 1–2 Jahre) einem hohen Grad an zellulärer Kompensationsfähigkeit entspricht.
    Notes: Summary After positive prenatal enzymic diagnosis of different neurolipidoses therapeutic abortion was carried out in the 19th to 25th week of pregnancy. Ten delivered fetuses were studied ultrastructurally and in nine of them positive results were obtained, although in some cases one had to accept relatively poor structural conservation of fetal tissues. The ultrastructure of the quantitatively small lipid storage effects qualitatively resembled that of the postnatal stages with some exceptions of localization. In fetal GM2-gangliosidosis type 2 (variant 0) concentric menbranous cytoplasmic bodies were detected in the brain cortex. In Krabbe's disease the myelinated regions of the spinal cord showed scattered storage (globoid) cells, sometimes closely related to blood vessels, which contained isolated or stranded tubular or spicular inclusions. In GM1-gangliosidosis type 1 neurons of the brain stem showed lamellar inclusions structured as zebra bodies, and splenic histiocytes exhibited numerous almost clear cytoplasmic vacuoles. In fetal metachromatic leukodystrophy the CNS including myelinated regions was essentially free of morphologic lipid storage effects. However, many kidney tubules cells contained great numbers of irregular or roughly parallel stacks of membranes. These inclusions may be equivalent to “tuffstone” bodies. In one fetus the bodies were restricted to tubular cells bearing microvilli. Fluorescent microscopy of arcus of the kidney tubule showed excess amounts of metachromatic material. Less of this material was demonstrable in the envelope layer of hepatic Glisson triangles. In the fetus with Niemann-Pick disease type C large neurons of the basal ganglia and the spinal cord were filled with membranous inclusions that were similar to myelin-shaped bodies rather than to solid membranous bodies. The 19-weeks-old fetus with enzymically proven Gaucher disease was free of ultrastructural lipid storage effects. Most but not all of the morphological findings in the fetuses with neurolipidoses were in accordance with published results.

    Notes: Summary The renal effects of histamine, histamine receptor agonists and antagonists were studied in the isolated rat kidney, which was perfused with a synthetic medium at constant perfusion pressure in a single pass system. Histamine induced a concentration-dependent increase of renin release ranging from a two-fold increase at 0.5 μM to a four-fold increase at 10 μM. No change in renal vascular resistance, glomerular filtration rate and sodium excretion occurred. Histamine-H2-antagonists (ranitidine and cimetidine) were more effective to block the response to histamine than was the histamine H1-antagonist diphenhydramine. Histamine-H2-agonists (impromidine and dimaprit, 2.5 μM each) were potent stimulators of renin release, their effect was blunted by H2-antagonists. The histamine-H1-agonist pyridyl-2-ethylamine had a low stimulatory activity at 10 μM final concentration, which may reflect partial H2-agonistic effects. It is concluded that histamine stimulates renin release via H2-receptor activation.

    Notes: Summary The effect of X-ray irradiation on the process of primary mineralization in bone was studied by biochemical and ultrastructural methods. A single dose of 1500R was administered to the head region of rats. The animals were examined immediately after irradiation and 1, 2 and 3 weeks later. Fractions of isolated cells and extracellular matrix vesicles were prepared from the maxillary alveolar bone of irradiated and untreated rats by collagenase digestion and differential centrifugation. The protein content and activities of vesicular phosphatases were determined in both fractions. A continuous decrease in the activity of alkaline phosphatase could be observed in both cell and matrix vesicle fractions during a three-week follow up after irradiation. Acid phosphatase activity decreased only in the vesicle fraction. Transmission electron microscopy of irradiated bone tissue revealed that many matrix vesicles were devoid of intact membranes and apatite crystals. Calcifying nodules were abundant in the matrix without their apparent fusion into larger mineralized structures. It is suggested that irradiation interferes with enzymatic processes associated with primary mineralization.

    Notes: Summary Clinical and histopathological features of two cases of malignant rhabdoid tumor of the kidney are presented. One of these cases was also studied by electron microscopy. Histologically, both tumors consisted of an admixture of undifferentiated polygonal or elongated cells and cells with abundant eosinophilic cytoplasm frequently containing hyaline globular structures. Ultrastructurally, these cytoplasmic inclusions were composed of large masses of actin-size and intermediate-size filaments. The poor prognosis of this type of tumor is emphasized and histological criteria for differential diagnosis from other malignant renal tumors of childhood and adolescence are discussed.

    Notes: Summary Vacuole-like structures were found in the nuclei of root tip cells ofBrassica napus. The cells containing the unusual nuclear inclusions were found to be adjacent to zones of degenerating cells. Such groups of cells occurred irregularly in the meristematic regions of the young root tips. The possibility that they represent changes which have occurred in old seeds is discussed. The “vacuole-like” structures seen in the cells adjacent to the degenerating zones were bounded by a membranous layer 12 nm thick. This is thicker than most cellular membranes. The “vacuoles” frequently contained inclusions and showed similarities to protein bodies reported elsewhere. The structures are thought to represent rearrangements of cell products which may have accumulated through an imbalance of metabolism in consequence of the imminent cell degeneration.

Both species have an area immediately behind the hyphal apex free of all large organelles and containing a high concentration of vesicles. InA. giganteus only one size class of vesicle is clearly evident, with a mean diameter of 72 nm. InA. nidulans two size classes of vesicle were found, with mean diameters of 75 nm and 31 nm. A Spitzenkörper is evident inA. nidulans as an area very close to the tip containing only the smaller vesicles. InA. giganteus one or more apparently mature Woronin bodies were found within the first 1 μm of some hyphal apices. The possible significance of their presence is discussed.

    Notes: Summary An extensive system of microtubules develops during meiotic prophase in the mossRhynchostegium serrulatum (Hedw.)Jaeg. &Sauerb. Development of the cytoskeleton can be traced to early prophase when the nucleus is acentric and the single plastid divides into four plastids. The cytoskeletal microtubules are associated with equidistant positioning of the four plastids at the distal tetrad poles and with migration of the nucleus to a central position in the sporocyte. The cytoskeleton, which interconnects plastids and encloses the nucleus, contributes to the establishment of moss sporocyte polarity. Just prior to metaphase I evidence of the prophase cytoskeleton is lost as the bipolar metaphase I spindle develops in association with discrete polar organizers located in opposite cleavage furrows between plastids.

    Notes: Summary Generation of movement in benthic diatoms is thought to be intimately associated with secretion at the raphe, a slit in the silica cell wall. The presence and distribution of extracellular substances and their source was investigated cytochemically by transmission electron microscopy. Extracellular material, possibly-acid mucopolysaccharide, was observed consistently within the entire length of the raphe of both valves and also as a sheath enveloping the silica frustule. Such quantities of extracellular material are absent in conventionally fixed motile diatoms. Numerous cytoplasmic vesicles, with fibrillar contents, distributed peripherally but concentrated along the raphe and at the cell poles, react strongly with a polysaccharide specific stain; their distribution in the cell and polysaccharide content suggest these may be the source of raphe and sheath material. Results support the most recent theories on the mechanism of locomotion in outline only; the details cannot be clarified. Localization procedures using alcian blue and silver staining of peroxidised sections are discussed briefly.

Mitochondria, vacuoles, plasma membrane, and vesicles with attached ribosomes from homogenized germlings equilibrated at the 1.2/1.4 M interface in discontinuous sucrose gradients. Accelerated flotation in centrifuged Ficol-sucrose gradients resulted in the additional separation of the mixed membranes into three fractions: one contained predominantly intact mitochondria, another was composed of vacuoles and vesicles coated with ribosomes, and a third was enriched in plasma membranes. Based upon morphometric analysis, these fractions contained 92% mitochondria, 53% vacuoles, and 89% plasma membranes, respectively. The source of vesicles coated with ribosomes was investigated since rapidly growing hyphae ofG. persicaria contained little rough endoplasmic reticulum as compared with other classes of membranes. Reconstruction from electron micrographs of mitochondrial fragmentation and vesiculation suggested that most of the ribosome-coated vesicles originated from disrupted mitochondria rather than from rough endoplasmic reticulum. The study demonstrates the utility of ultrastructural markers to identify membranesin vitro independent of, or as an adjunct to, cytochemical and biochemical markers.

Progressive degeneration of papular cells is observed in stigmas from open flowers and at anthesis papillae may be moribund and collapsed. Fluid emanating from the hollow style flows onto the surface through ruptures in the cuticle at the interpapillar junctions into the interstices at maturity. This secretion stains positively for protein. Stigmas are of the “wet” type. The cuticle overlying the papillar cells is ridged and at the final stages prior to flowering this cuticle becomes detached from the underlying cellulosic wall. The sub-cuticular space so formed is filled with secretion. InAneilema species detachment of cuticle is at the papillar tip and along the lateral walls. InCommelina species the anticlinal walls of adjacent papillae are strongly attached for much of their length and thus detachment of cuticle is restricted to the papillar tip. The cell wall at the tip in both genera may proliferate forming a rudimentary transfer-cell type wall. The secretion is considered to be produced by the papillar cells. It is PAS positive but fails to stain for protein and in both the light and electron microscopes appears heterogenous. Pollen attachment, hydration, germination and early tube growth are very rapid following self-pollination, the pollen tubes entering the neck of the style within ten minutes of attachment. A unique character combination involving pollen and stigmas in these genera indicates a monophyletic origin.

After 24 hours of induction approximately 90% of the myxamoebae had differentiated into microcysts. The kinetics of encystment were not significantly affected by pH or osmolarity of the encystment medium. Early stages of encystment were distinguished by the appearance of autophagic vacuoles and an extracellular “slime-like” sheath. The outer wall layer, consisting of dense fibrils, was unevenly deposited after 4 hours. An electron-lucent, second wall layer appeared between 5–10 hours followed by a densely packed, third wall layer adjacent to the plasma membrane. Wall formation appeared to involve smooth-membraned vesicles of possible Golgi origin. The vesicle contents and outer wall layer reacted with the periodic acid-silver methenamine stain for polysaccharide. The density of intramembrane particles of the protoplasmic fracture face increased during encystment with a gradual formation of aggregates of particles.

    Notes: Summary Discophrya collini is a suctorian protozoan with contractile tentacles containing a microtubule-lined canal and microfilaments. The effects of a range of cations on tentacle contraction and ultrastructure have been determined. Treatment with 80 mM CaCl2 and 95 mM MgCl2 causes contraction to 28% and 57% of the control length respectively. Re-extension takes over 4 hours in the culture medium, but CaCl2-treated tentacles are re-extended after a 5 minutes treatment with 10−2 M EDTA or 5 × 10−3 M EGTA. CuCl2 causes a significant contraction at 10−5 M (to 77%); LaCl3 at 10−4 M (to 65%); ZnCl2 at 10−2 M (to 65%), but BaCl2, CoCl2, MnCl2, NiCl2, and SrCl2 cause significant changes only at 10−1 M. The cytoplasm of CaCl2-treated cells contains two forms of membraneous structures when viewed in TEM; that of MgCl2-treated cells reveals granular areas of medium electron density. None of these features are seen in control cells. The microtubules of the tentacle canal appear to be intact upon its retraction into the cell with no change occurring in the numbers or relative positions of the microtubules. The tentacle cortex is wrinkled. It is suggested from this and previous work that tentacle contraction may be mediated by a microfilament-based mechanism, and that calcium may be involved.

    Notes: Summary Determining how the orientation and association among organelles are maintained within zoospores of theChytridiales is important to understanding the control of zoospore motility. Zoospores of the aquatic fungi,Chytriomyces aureus andC. hyalinus, contain microbody-lipid globule complexes with an elongate microbody adjacent to the portion of a lipid globule facing the cell's interior and a fenestrated cisterna (the rumposome) opposed to the surface of the lipid globule toward the plasma membrane. Mitochondria are intimately associated with the microbody. Electron microscopy of the microbody-lipid globule complex fixed in glutaraldehyde and osmium tetroxide, with or without tannic acid, reveals cross-linking bridges connecting the rumposome to the plasma membrane, to the microbody, and to microtubules of the rootlet extending from the kinetosome. It is concluded that these bridges are responsible, at least in part, for the consistent location of the microbody-lipid globule complex in the zoospore body. The possible role of the rumposome as a receptor organelle is discussed.

    Notes: Summary Callus-derived suspension cultures of oats dramatically increase the viscosity of the culture media after one month in culture. Colorimetric assays for sugars and protein, as well as measurements of viscosity, suggest that the released material is a long-chain polysaccharide, probably a pectinaceous substance. These cells grow slowly in liquid culture, yet despite their low cell density, they are able to increase the viscosity of the media several fold within seven days after media transfer. Ultrastructural observations show that oat cells have features common to actively-secreting cells; especially evident are numerous dictyosomes with hypertrophied cisternae. Using a combination of filtering and centrifugation techniques we were able to recover large numbers of intact secretory vesicles. The interior of the vesicles stain with periodic acid-silver hexamine, and colormetric analysis of the vesicle pellet for total sugars confirms the presence of polysaccharides in this vesicle fraction. Because of the uniformity of these cells, the high rate of secretion, and the accessability of a large vesicle population, this culture system is'a useful model for studying the secretory process in plant cells.

    Notes: Summary Muscle ultrastructure of a group of subjects possessing extreme hypertrophy was compared with that of a control group which had undergone 6 months of heavy resistance training. Two needle biopsies were taken from triceps brachii of two international calibre powerlifters and five elite bodybuilders. In addition, samples were taken from five healthy volunteers before and after 6 months of training of the elbow extensors. One biopsy was prepared for electron microscopy and analyzed stereologically, and the other was stained for myosin ATPase activity and photographed under the light microscope. Despite large differences in elbow extension strength and arm girth there was no significant difference in fibre areas or percentages of fibre types between the elite group and the trained controls. This suggests that the elite group possessed a greater total number of muscle fibres than the controls did. Mitochondrial volume density of the elite group was similar to that of the control group following training but significantly less (p〈0.05) than the pretraining control measurements. Myofibrillar volume density was significantly lower and cytoplasmic volume density significantly higher in the elite group than in the trained controls. There was a considerably higher incidence of structural abnormalities including central nuclei and atrophied fibres in the elite group than in the control group, which might possibly have been associated with the use of anabolic steroids by the elite group.

Als Vergleichsgruppen dienten Laparotomie, alleinige Pyloroplastik und als obligates Refluxmodell die Resektion nach BII ohne Enteroanastomose. Die Höhe des duodenogastrischen Refluxes war nach SPV mit und ohne Pyloroplastik signifikant niedriger als in der Kontrollgruppe. Tiere mit alleiniger Pyloroplastik zeigten einen signifikant erhöhten Reflux. Die BIIGruppe hatte einen fünffach höheren Wert als die Kontrollgruppe. Aufgrund der Pyloroplastik als Drainageoperation kommt es am Rattenmagen nach SPV nicht zur Retention von Speiseresten, die man nach SPV ohne Pyloroplastik beobachtet. Aber wegen der verzögerten Magenmotilität legt sich eingedickter Speisebrei vor den Magenausgang und reduziert so den duodenogastrischen Reflux. Nach unseren Ergebnissen kann ein vermehrter duodenogastrischer Reflux nach SPV mit und ohne Pyloroplastik keine morphologischen Veränderungen der Magenmucosa verursachen.
    Notes: Summary Using rats in a standardized animal experiment, the extent of duodenogastric reflux was studied eight months postoperatively following SPV with and without pyloroplasty. The reflux was determined by enzymatically testing the concentration of bile acid secretion withdrawn from the stomach intraoperatively. Serving as comparison were laparotomies, simple pyloroplasties, and the obligatory reflux models: resection after Bll without enteroanastomosis. After SPV with and without pyloroplasty, the extent of duodenogastric reflux was less than in the control group. Specimens with simple pyloroplasty displayed a significantly greater reflux. The Bll group had a fivefold higher reflux than the control group. Since pyloroplasty can be considered a drainage operation, there is no retention of food particles in the stomach of the rat following SPV - an occurrence expected after SPV without pyloroplasty. However, due to the retarded stomach motility, the thickened chyme tends to linger at the opening of the stomach, thus reducing the duodenogastric reflux. According to our investigations, an increased duodenogastric reflux following SPV with or without pyloroplasty does not induce changes in the mucosa of the stomach.

Portal circulation was established using the host's renal vessels and ureter for bile drainage: renal vein to inferior vena cava end to end, using continuous 10-0 nylon; renal artery to portal vein using a stent and bile duct to ureter using a stent. The new model provides low mortality, short operation time and ischemia time and was successful in more than 100 transplants for immunological studies.

Es wurde überprüft, inwieweit hiernach im Vergleich zu einer nicht-vagotomierten Kontrollgruppe Schleimhautveränderungen auftreten, die unter Exposition mit dem Carcinogen MNNG zeitlich früher und häufiger zur Carcinomentstehung prädisponieren. Als mögliche wichtige cocarcinogene Faktoren wurden zusätzlich die intragastrale Bakterienflora und Nitrit-Konzentration der einzelnen Versuchsgruppen untereinander verglichen. Es zeigte sich, daß der Schweregrad gefundener Magenschleimhautveränderungen, sowie das Ausmaß der bakteriellen Besiedlung und die Nitrit-Konzentration im Magensaft neben der durch Vagotomie angestrebten Säurereduktion vor allem durch Störung der Magenmotilität oder einen enterogastrischen Reflux verstärkt wird. Die Ergebnisse der Untersuchungen sprechen dafür, daß nach Vagotomie, soweit eine ausreichende Magenentleerung gesichert und diese nicht durch einen enterogastrischen Reflux (Gastroenterostomie) erkauft wird, das Risiko für ein Operationsfolgecarcinom gering anzusehen ist.
    Notes: Summary To assess the possible risk of cancer in the operated stomach, the influence of truncal vagotomy combined with different operative drainage procedures (pyloroplasty, gastro-enterostomy without entero-anastomosis, Roux-en-Y gastro-enterostomy) was studied in animal experiments. The aim of the study was to determine whether, compared to controls an earlier and higher incidence of precancerous epithelial lesions or cancers in the stomach occurs in vagotomized animals after oral administration of carcinogens (MNNG). To consider conditions, that may predispose to in vivo formation of carcinogenicN-nitroso compounds, bacterial flora and intragastric nitrite concentration were measured and compared in the various operative groups. The severity of induced epithelial lesions and the quantity of bacterial growth and nitrite concentration in the gastric juice were influenced, apart from the reduction of acid output after vagotomy, mainly by prolonged gastric stasis or the degree of enterogastric reflux. The results suggest that there is probably no increased risk of cancer following vagotomy, if sufficient gastric clearance is guaranted and enterogastric reflux is largely excluded.

    Notes: Summary After feeding young rats a diet deficient in vitamin A, we examined the inner ear with the electron microscope. There were changes in the cuticle of the outer and inner hair cells. Furthermore, there were changes in the reticular system of the intermediate zone and massive degenerative changes in the ganglion cells of the VIII nerve. In a second experiment with older animals we found no significant changes in the sensory cells, though there was new bone formation in Rosenthal's canal and damage to the ganglion cells, of a lesser extent than was evident in the first experiment, however. In a further clinical study, we carefully chose human subjects suffering from alcoholic liver disease who also had a negative history of ear infection, noise exposure, head injury and use of streptomycin. Normal auditory function in the family was also a criterion. A decreased auditory function associated with low vitamin A levels was found in these patients. Those with liver disease showed not only a significant auditory dysfunction in the higher frequencies, but as well a poorer performance in the tone decay test. They were compared to a control group with normal hepatic, renal and thyroid status.

    Notes: Summary Mechano-electric transduction and neuronal transmission were studied in sensory systems ascending from and descending to single receptor cells of the labyrinth organs in submammalian vertebrates. The animals were young crocodiles (Caiman crocodilus), geckos (Gekko gecko, Tarentola mauritanica), and turtles (Pseudemys scripta elegans, Chinemys reevesii). Intracellular receptor potentials from the apical region of the hair cell (or from the ciliary surface) were recorded in the ampullar, macular, and papillar sensory cells. These single-cell responses are, within limits, proportional to stimulus amplitude, frequency, or phase and are bidirectional in that they show depolarization by kinociliopetal stereociliar displacement and hyperpolarization by kinociliofugal displacement. Synaptic potentials (presynaptic from the basal region of the hair cell, postsynaptic from the contacting nerve endings) were recorded in the utricular, saccular, and lagenar neuroepithelia with electron-optic localization of the in situ fixed microelectrode tip. As local excitatory or inhibitory processes, respectively, they follow the stimulus and receptor potential with latency and with nonlinear distortion. Action potentials (spikes), as synchronized by the excitatory synaptic potentials, were recorded from single nerve fibers or bipolar cells, related to ampullar, macular, or papular receptor units. Unit responses and synaptic potentials were recorded from the first, second, and following centripetal and central neurons of the ascending systems, or from neurons of the descending systems in the brain stem or from centrifugal neurons. Such records were achieved during adequate mechanical or acoustical stimulation of the different receptor systems, with additional electrical stimulation, uni- or bilaterally. Thus, the influence of centripetal-centrifugal bilateral interaction on the receptor functions was measured, as inhibition or disinhibition, respectively. The input-output relations of these sequential stages of information transfer were plotted as histograms of different types, as characteristic curves, power spectra, or by correlation operations, with or without feedback, from the different systemic levels.

    Notes: The complexation of Cue2+ with 1, 8-diamino-3, 6-diaza-2, 7-octanedione (—N, N′-diglycyl-1, 2-ethanediamine, DED) and with 1, 9-diamino-3, 7-diaza-2, 8nonanedione (—N, N′-diglycyl-1, 3-propanediamine, DPD) has been studied by potentiometric and by spectrophotometric titration. With both ligands L the complexation to Cue2+ leads to relatively complicated equilibria with CuLH3+, CuL2+, CuLH-2, and dimeric Cu2L24+ complexes. With DED, another dimeric species, Cu2L2H-22+, is formed in addition. Independent numerical treatment of spectrophotometric and poteritiometric titrations was used to obtain a satisfactory model for the complexation and to test the relative discriminatory power of the two methods. Titrations of glycine ethylamide (GEA) were used as an additional test and as a model for DED and DPD. It was shown that in each case spectrophotometric titrations give results of similar reproducibility and have a discriminatory power equal to or better than potentiometric titrations, provided that optimum mathematical algorithms are used in the numerical treatment.

A Simple Synthesis of Azulenopseudophenalenons4, 6, 8-Trimethylazulene (3) reacts after metalation with lithiumdiisopropyl-amide in ether with bromoacetic acid to the 6, 8-dimethylaltulene-4-propionic acid (4), which undergoes cyclization to the 7, 9-dimethyl-4, 5-dihydro-3H-benz [cd]-azulene-3-one (5) in the presence of p-toluenesulfonic acid; oxidation of 5 with 2, 3-dichloro-5, 6-dicyanobenzoquinone yields 7, 9-dimethyl-3H-benz [cd]azulene-3--one (1b). Alkylation of 1b with triethyloxonium tetrafluoroborate in CH2C12 gives the 3-ethoxy [cd]benzazulenium tetrafluoroborate (6).

    Notes: The dienone-dianion derivatives 1 react with all types of electrophiles tested (alkyl halide, silyl chloride, ester, ketone, aldehyde, epoxide) to give β, γ-unsaturated carbonyl compounds of type A (see Formulae 2-6, 13, 14 and Tables 1-5). The α- and β-hydroxyalkylation products obtained from 1a-1d can be converted to tetra-hydrofuran and tetrahydropyran derivatives 7 and 16, respectively (Tables 1 and 2), those from the sulfur analogues 1e and 1f to ketene thioacetals 9 and to dienone derivatives 10 and 12. The t-butyl and α-hydroxy-ketones are cleaved to give nitriles, amides, carboxylic acids and esters (Formulae 16-25). The reagents 1 allow to synthesize products with distant functional groups in one step (cf. 1,8-diketones 14 and Formulae 26-30); they correspond to the d5-synthons 31-33; in Table 6, they are compared with other d5-reagents.

This explains contradictory reports in the literature and, in the case of the reductions with potassium in which pinacol coupling does not compete, proves that the reductions involve bimolecular reactions between two ketyls, or between the ketone and the ketyl or the ketone dianion.

    Notes: Investigation of the formation of complex reaction products in the gas-phase system O3/NO2/(Z)-2-butene by combination of linear reactors with IR. matrix and microwave Stark Spectroscopy is reported. Besides the polyatomic products observed earlier in the gas-phase ozonolysis of (Z)-2-butene, the following products were identified; N2O5, HNO3, HNO4, CH3NO2, CH3ONO, CH3COONO2 and CH3COO2NO2 (peroxyacetyl nitrate, PAN). Matrix IR. spectra of N2O5, HNO3. CH3COONO, CH3COONO2 required for reference purposes are presented. It is shown that PAN-formation occurs already in the absence of light. A reaction scheme is proposed for explanation of the observed complex NOx-containing products, which assumes methyldioxirane as a central intermediate. Particular reaction steps of the scheme will be discussed, including thermochemical estimates of reaction enthalpies.

The structures of 2 and 3 were confirmed by converting the diazonium chlorides derived from 5-fluoro-2-nitroaniline (5) and from 2-fluofo-5-nitroaniline (8) to 5-fluoro-2-nitrobenzene-sulfonyl chloride (6) and 2-fluoro-5-nitrobenzenesulfonyl chloride (9), respectively, followed by hydrolysis of 6 to 5-fluoro-2-nitrobenzenesulfonic acid (7), and of 9 to 2-fluoro-5-nitrobenzenesulfonic acid (10), and by final reduction. Compound 10 was also obtained by sulfonation of 1-fluoro-4-nitrobenzene (11) with oleum.

    Notes: The radical anion of 1,1,2.2,9,9,10, 10-octafluoro[2.2]paracyclophane (1) has been generated by electrolytic reduction of 1 in 1,2-dimethoxyethane (tetrabutylammonium perchlorate as the supporting salt). The hyperfine coupling constants of the eight 19F-nuclei and the eight protons, af = 3.35 and aH ≈ 0.10 mT, are qualitatively reproduced by INDO. calculations. According to these calculations, the singly occupied orbital of 1⊖ can be represented by an Ag-combination of two ‘symmetric’ benzene LUMO's, with a substantial transfer of spin population into the 2s-AO's of the F-atoms. Line-width alternation in the ESR. spectrum of 1⊖ indicates an ion pairing of the radical anion with its counter-ion Bu4N⊖. The energy barrier to the migration of the cation between two equivalent sites at the radical anion is determined as 14±2 kJ/mol.

I. cis-Civetone and polymorphous α- and β-forms of his 2,4-dinitrophenylhydrazonecis-Civetone (C17H30O) forms tetragonal plastic crystals, space groupe 141; a = 9.95(4), c = 32.79(1) Å; Z = 8. The plastic phase exists in a wide temperature range and 731 reflexions could be collected at 153 K. The highly disordered structure model was obtained by the use of direct methods. The molecules appear as ring-shaped diffuse electron-density distributions located in special position.Two polymorphous crystalline forms were isolated for the 2,4-dinitrophenyl-hydrazone of cis-civetone (DNPHCC). Both forms are triclinic, space group P1. Z = 2 (α-Form: a = 6.279(5), b = 12.605(8), c = 15.253(10) Å, α = 105.49(7). β = 100.31 (6), γ = 91.23(7)°; β-Form: a = 7.950(2). b = 8.405 (2). c = 18.233(4) Å, α = 100.28(2), β = 92.29(3), γ = 94.18(2)°). The structures were solved by direct methods and refined to R = 0.11. Each polymorph is associated with a different quinquangular conformation of the macrocycle. In the crystals the intermolecular interactions between macrocycles and aromatic substituents are minimized, the DNPH group being oriented in a face-to-face arrangement across a centre of symmetry.Empirical force field calculations show that the overall intluence of the DNPH moiety on the attached cycle does not significantly modify its conformation with regard to that of the ketone itself.

    Notes: The interaction of solvents and of unidentate ligands such as N3-, SCN-, OCN- and OH- with the Co2+-, Ni2+ and Cu2+-complexes of 1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetradecane (TMC) have been studied by Spectrophotometric and calorimetric techniques. The spectra in different solvents (Table 2) show that the Ni2+- and probably also the Cu2+-complex with TMC exist as square planar or pentacoordinate species or as a mixture of both, depending on the donor properties of the solvent. The [Co(TMC)]2+-complex is pentacoordinate in all the solvents studied.Ternary complexes [M(TMC)X]n+ are also formed by the unidentate ligands X = N3-, OCN-, OH-, F- and NH3 and their stability constants have been determined. Interesting is the high selectivity of [Ni(TMC)]2+ towards the addition of a further donor (Table 3). Only small ligands such as those listed above form stable adducts, whereas the larger ones such as imidazole or pyridine do not. This is a consequence of the special structure of the complex and of the trans-I-(RSRS)- conformation of the ligand in these complexes. Since the four methyl groups are all on the side of the macrocycle to which the additional unidentate ligand binds, steric interaction between the four methyl groups and the larger ligands prevents the formation of the adducts.The calorimetric measurements show that the stability of the complexes [M(TMC)X]n+ is due to both an enthalpic and entropic contribution which differ in their magnitude (Table 4). This indicates that several antagonistic factors are important in determining the overall stability.

Mixed crystals were prepared with cis-civetone as second component. Accordingly the diffuse structural model obtained for the disordered phase of the cis-isomer [1] is also regarded as representative of that of trans-civetone.The crystal structure of trans-civetone 2,4-dinitrophenylhydrazone (DNPHTC; C23H34N4, Monoclinic, Space group P21/c, a=8.364(2), b=7.971(1), c=36.025(8) Å, β=91.44 (2)°) was solved by direct methods and refined to a final R of 0.09 (Rw = 0.021). The macrocycle adopts a sexangular conformation [23*434*44*], and its mean plane is approximatively perpendicular to that of the aromatic substituent. Empirical force field calculations have shown that the DNPH substituent has very little influence on the macrocycle conformation. The intermolecular interactions take place essentially between groups of the same type: macrocycle…macrocycle and DNPH…DNPH.

Competitive methylene blue sensitized photo-oxygenation experiments allowed the determination of relative rates of photo-oxygenation of 1-benzyl-3,4-dihydroisoquinolines, Substituents were shown to affect both the equilibrium concentration of the tautomeric enamine and the overall photo-oxygenation rate. After correcting for differences in enamine concentration, the relative rate data provided a diagnostic probe of the reaction mechanism, which involves transfer of charge in the rate-limiting step.

    Notes: Stereoselective syntheses of 2exo, 3exo-bis (chloromethyl)-5-[(Z)-chloromethylidene]- (9), 2exo, 3exo-bis (chloromethyl)5-[(E)-chloromethylidene]- (10) and 2exo, 3exo-bis(chloromethyl)-5-[(E)-methoxymethylidene]-6-niethylidene-7-oxa-bicyclo[2.2.1]heptane (13) are presented. Double elimination of HCI from 9, 10 and 13 yielded 2-[(Z)-chloromethylidene]- (14), 2-[(E)chloromethylidene]- (15) and 2-[(E)-methoxymethylidene]-3,5,6-mmethylidene-7-oxabicycio[2.2.1]heptane (18), respectively, without loss of the olefin configuration. Ethylene tetracarbonitrile (TCE) and N-phenyltriazolinedione (NPTAD) added to these new exocyclic dienes and tetraenes preferentially onto their exo-face. The same face selectivity was observed for the cycloadditions of TCE to the (Z)- and (E)-chlorodienes 9 and 10, thus realizing a case where the kinetic stereoselectivity of the additions is proven not to be governed by the stability of the adducts. The exo-face selectivity of the Diels-Alder additions of dienes grafted onto 7-oxabicyclo [2,2.1]heptanes contrasts with the endo-face selectivity reported for a large number of cycloadditions of dienes grafted onto bicyclo[2.2.1]heptane skeletons.

    Notes: The rates of photo-oxidation of exocyclic S-cis-butadienes grafted onto bicyclo-[2.2.1]heptanes and 7-oxabicyclo[2.2.1]heptanes (1-6) are dependent upon remote modifications of the bicyclic skeletons. They correlate with the rates of Diels-Alder additions of these dienes to strong dienophiles. The 2,3-dimethylidenenorbornane (1), 5,6-dimethylidene-2-norbornene (2) and 2,3-dimethylidene-7-oxanorbornane (3) gave the corresponding endo-peroxides (3,6-dihydro-1,2-dioxines) 7-9 in good yield. The 2, 3, 5, 6-tetramethylidene-7-oxanorbornane (4) gave the mono-endo-pe-roxide 6, the latter did not react with a second equivalent of oxygen. Similarly, 5, 6-dimethylidene-7-oxa-2-norbornene (5) was unreactive toward photo-oxidation. Thermal isomerization of the endo-peroxides 7 and 9 gave, the trans-diepoxides 10 and 14, respectively, with high stereoselectivity. The endo-peroxides 6, 7 and 9 were cleanly isomerized into the corresponding α, β-unsaturated γ-hydroxy aldehydes in the presence of catalytic amounts of Rh2(CO)4Cl2.

    Notes: Synthesis and Circular Dichroism of Optically Active Carotenoid ModelsThe synthesis of the following optically active carotenoidic model compounds are described: ( - )-(3,S,3′S)-3,3′-diisopropenyl-16,17,18,16′,17,18′-hexanor-β,β-carotene (1), (3R,3′R)-19,20,19′,20′-Metranor-zeaxanthin (2) and (6R,6′R)-19,20, 19′,20′-tetranor-ε,ε-carotene (3). These compounds were synthesized for the following reasons: (1) the presence of methyl groups at C(1), C(1′), C(5), C(5′) of cyclic carotenoids profoundly affects the torsional angle of the C(6), C(7)- and C(6′), C(7′)-bonds. Sign and magnitude of this angle are, according to recent theories [4] [5], responsible for a helical chromophore and for strong conservative [4] Cotton effects. CD. measurements of 1 give experimental support to these state- 1 exhibits weaker and less temperature dependent Cotton effects. Of more significance, the shape of the curve is no longer conservative, as expected. This constitutes experimental evidence for the contention that the β-endgroups and the polyene chain indeed form an inherently dissymmetric chromophore in optically active β, β-carotene derivatives; (2) the slightly S-shaped form of the polyene chain of carotenoids, shown by X-ray analyses [12] [13], is mainly ascribed to the presence of the methyl groups in the chain. Models 2 and 3 therefore are assumed to be linear. CD. studies of these compounds should consequently give information about the influence of deviation from Linearity and planarity of the polyene on the CD. spectra of carotenoids. CD measurements of 2 and 3 show that the lack of methyl groups does not alter the general type of the curve. Only the intensity and to some extent the position of the Cotton effects are influenced. Carotenoids with the ε-endgroup possess inherently symmetric but asymmetrically distorted chromophores.The assumption that non-conservative CD. spectra could become conservative upon cooling [4] is experimentally confirmed by model 3.The rule stating that pairs of all-(E) and mono-(Z) isomers of carotenoids with only one cyclic endgroup should have CD. spectra with the same sign [5] is disproved by the CD. spectra of four stereoisomeric rubixanthins (s. Fig. 5).The UV./VIS. spectrum of 3, λmax 447 (ε 216000), 418 (ε 189000) exhibits the highest molecular extinction ever reported for a carotenoid.

Single crystals are obtained by zone melting at 258 °K. The high degree of disorder and the ambiguity of space group have precluded a structural determination by X-ray diffraction methods.The crystal structure of muscone 2,4-dinitrophenylhydrazone (DNPHM; C22H34N4O4, triclinic, space group P¯, a = 8.015 (4), b = 8.235 (2), c = 17.409 (4) Å. α = 102.69 (4), β; = 93.86 (3), γ = 96.03 (3)° Z = 2) was solved by direct methods and refined to a final R of 0.155 (Rω = 0.102). The macrocycle shows static disorder, which could be partially resolved by analysis of the anisotropy of the atomic vibrational parameters. Eight possible conformers with reasonable geometries emerged from the subsequent refinements and were used as starting models in force-field calculations, which converged to four quinquangular conformations ([25233]; [34233]; [21423*4]; [3423*4]) with similar energies. In the molecular packing, the interactions between macrocycles and substituents are minimized. They are comparable to those described for the cis- and trans-civetone DNPH derivatives [1] [2].

    Notes: Reactions of [2N]cyclophanes (N = 2,⃛6) with solvated electrons in 1,2-di-methoxyethane at 193 K have been studied by ESR. and ENDOR. spectroscopy. All but the two most highly bridged cyclophanes (N = 5 and 6) are reduced to paramagnetic species under these conditions. Whereas the radical anions of [2.2]-paracyclophane and [23](1,2,4)- and [24](l,2,4,5)cyclophanes are sufficiently persistent to be characterized by their hyperfine data, those of the remaining five cyclophanes undergo a rapid cyclization to the radical anions of 4,5,9,10-tetrahydropyrenes. These have been identified as the unsubstituted tetrahydropyrene (from [2.2]-metacyclophane and [23](l,2,3)cyclophane), the 2,7-dimethyl-derivative (from [23](1,3,5)- and [24](l,2,3,5)cyclophanes) and the 1,8-dimethyl-derivative (from (24l,2,3,4)cyclophane). The persistence of the cyclophane radical anions seems to depend on the numbers, nmeta and npara, of the meta-and para-positions of the bridging ethano groups in the two benzene rings. The prerequisite for the radical anion to be persistent is nmeta≤npara.

Under rearomatization of the central frame protonation of 4 yields the salt 4c. The reaction of 4 with dicyanoketene gives the ‘push-pull-substituted’ 5, and with 9-carbonylfluorene the fulvalene derivative 7, which can be protonated by CF3CO2H at C(9″) to the salt 8. The reaction of 2-methylthio-l,3-dithiolium jodide 9 with the keto-enol mixture 2 yields 10, which, on protonation at the carbonyl group by CF3CO2H, gives the salt 11 under rearomatization. The spectral data of the new compounds 4, 4c, 5, 7, 8, 10 and 11 are reported and discussed.

    Notes: The water exchange of [V(H2O)6]Cl2 in aqueous solution has been studied as a function of temperature and pressure (up to 250 MPa), by measuring the 17O-FT-NMR. line-widths of the free water resonance at 8.13 MHz. The kinetic parameters obtained are K298m = 87±4 s-1, ΔH* = +61.8 ± 0.7 kJ mo1-1 and ΔS* = -0.4±1.9 J mol-1 K-1. A pressure-independent volume of activation ΔV* = -4.1±0.1 cm3 mol-1 is obtained, suggesting an associative interchange (Ia) mechanism for this early divalent metal ion.

    Notes: A total synthesis of racemic 3-deoxy-7,8-dihydromorphine ((±)-2) and 4-me-thoxy-ALmethylmorphinan-6-one ((±)-3) is described. The key intermediate was 2,4-dihydroxy-N-formylmorphinan-6-one (11), obtained from 3,5-dibenzyloxy-phenylacetic acid (4) in 41.8% overall yield. Bromination of 11, and treatment with aqueous NaOH-solution afforded, after N-deblocking and reductive N-methylation with concomitant removal of the aromatic bounded Br-atom, the morphinanone 14. Elimination of the HO-C(2) group in 14 was accomplished by hydrogenolysis of its N-phenyltetrazolyl ether 15, to give 3-deoxy-6,0-didehydro-7,8-dihydromorphine (16). Reduction of 16 with L-Selectride at low temperature provided (±)-2 in high yield. The ether 15 directly afforded, under more vigorous reduction conditions, 4-hydroxy-N-methylmorphinan-6-one (17). and after O-methylation of 17, the methyl ether (±)-3 was obtained. A (1:l)-mixture of 4-hydroxy-2-methoxy-N-methylmor-phinan-6-one (28) and its 2-hydroxy-4-methoxy isomer 30 svere obtained by Grewe-cyclization of a mono-methoxylated aromatic precursor similar to that which afforded 11. The 2,4-dioxygenated N-methylmorphinan-6-ones 29, 31 and 38 were also prepared and characterized.

    Notes: The synthesis of new 2-oxo-2,8-dihydro-[l,2,4]-oxadiazoo-[2,3-a] pyrimidine-carbamates 3 described in this paper is based upon a regioselective heterocyclization of the pyrimidine-2,6-bis (alkoxycarbonylamino) N-oxides 4. Structural and mechanistic aspects of that cyclizalion are discussed.

    Notes: The rate constants for 3-substituted adamantyl p-toluenesulfonates 3a-3k in ethanol/water 80:20 correlate well with the respective inductive substituent constants σIq. The reaction constant ρ for the toluenesulfonates 3 is 10% larger than for the corresponding bromides 2, indicating somewhat more charge separation in the activation of the toluenesulfonates. Evidence is presented that stabilization of the resultant 1-adamantyl cations by induction involves graded 1,3-bridging, which is favored when the substituent is an electrofugal group, and that stabilization by n-electron donors involves C, C-hyperconjugation. Rate ratios for the toluenesulfonates 3 and the bromides 1 exceed 103 and are almost independent of the 3-substituents. The implications of this are discussed in the light of current hypotheses.

    Notes: The syntheses of optically active t-butyl 2-(p-tolylsulfinyl)propionate and -butyrate (2a and 2b, respectively) are described, and it is shown that aldol-type condensation of the corresponding enolates is limited by steric hindrance. Optically active (2a) reacted, however, in high yield with aliphatic aldehydes and lead to 80% of asymmetric induction.

    Notes: Total synthesis of α-disaccharidyl-2,5,6-trideoxy-streptamines related to aminoglycoside antibiotics derived from maltose and lactoseThe synthesis of the hitherto unknown 4-O-substituted α-linked pseudo-tri-saccharides 1,2 and 3 is described. The key intermediates required in the reaction sequence are the glycals 12, 13 and 14, prepared from maltose, lactose and the chiral ditosyloxy-cyclohexanol 15, readily accessible from quinic acid. The pivotal step in the reaction's scheme - a stereospecific α-glycosylution procedure -, is an acid-catalyzed addition of the alcohol 15 to the glycals 12, 13 and 14 leading to the corresponding 2,3-unsaturated trisaccharides. The latter compounds were transformed to the target α-glycosides of 2,5,6-trideoxystreptamine 1, 2 and 3 which were devoid of significant antibacterial activity. These results indicate that the presence of an ether linkage at the position 4′ is probably critical for antibacterial activity.

Hydrolysis of the crude primary products furnishes α-alkylated cyclohexanones of 〉 90% e. e. (3, Scheme 3). Their (2S,1′R)-configuration was deduced by chemical correlation with l-cyclohexyl-l-phenyl-ethane and from an X-ray crystal structure analysis of (-)-(2R,3S,6′R1,l″S′)-3-methyl-N-[6′-(2″-nitro-l″-phenylethyl)-l′-cyclohexenyl]-2-phenylmorpholine (lla, Scheme 5 and Fig. 2). - The relative topicity of reactant approach with the prolinol derivative (see II) is specified as lkul-l,4. The steric course and the mechanism of the reaction are discussed.

    Notes: The reduction of a series of 1,2-dimethyl-3-indolyl heteroaryl ketones, where the heteroaryl components are 2-benzoxazolyl, 2-behzpthiuzolyl. 1-methyl-2-benz-imidazolyl, 4- and 2-pyridyl, in aprotic and prone solution are described by means of DC. and AC. polarography, cyclic voltammetry and coulometry.In acetonitrile a two-step reduction mechanism with a reversible first and an irreversible second one-electron transfer has been observed, confirmed by AC.-polarographic and cyclovoltammetric measurements. The reduction potentials, between -1.377 and -1.787 V for the first step, and between -1.937 and -2.250 V (vs. Ag/AgCl) for the second, could not be correlated with the LUMO energies from HMO calculations which was explained as being due to a distorted configuration of the π systems.In protic media (ethanol/water) reduction occurs in an irreversible one-step mechanism. Measurements of the pH dependence of the half-wave potential in the pH* range 0-12 and the evaluated transfer coefficients show that the reduction step represents the transfer of two electrons and two protons yielding secondary alcohols and no dimerization products under any condition. These findings are supported by coulometric and macroscale electrolysis of the ketones; the products isolated were characterized by IR. and 1H-NMR. data.

    Notes: The silylation of the surface of sodium borosilicate glass was investigated under conditions that might be applied to the fabrication of ion-selective microelectrodes of the liquid membrane type such as are used in physiology, Silylation was carried out for 15 min at different temperatures and with six different silanes. The hydrophobicity was measured and found to be greatest with reaction temperatures of 200-350°, the effectiveness of the different silylating agents being in the (increasing) order: trimethylchlorosilane, tributylchlorosilane, (dimethylamino)trimethylsilane, hexamethyldisilazane, dimethyldichlorosilane, bis(dimethylamino)dimethylsilane. The results give evidence of a catalytic effect of basic amine groups and show that the reactivity of the surface of freshly drawn glass is increased by acid leaching.
