Notes: Summary Radioimmunoassays for erythropoietin are limited so far to a few specialized laboratories and this requires transport and storage of samples. We therefore tested the stability of immunoreactive erythropoietin in serum and plasma samples obtained from a uremic and a nonuremic anemic patient. No significant change in the concentration of immunoreactive erythropoietin was found in either serum or plasma samples for up to 14 days of storage. This type of stability was observed no matter whether the samples were stored at room temperature, 4° C, or −20° C. There was no difference between the estimates of erythropoietin in serum and heparinized plasma. Validity of the radioimmunoassay used in this study was demonstrated by parallelism of dilution curves of test specimens and the 2nd International Reference Preparation for erythropoietin and by a close correlation between the immunoreactivity and the bioactivity of the hormone, as assessed in the same samples by the exhypoxic polycythemic mouse bioassay. In conclusion the data obtained clearly indicate that the necessity of storage and transport of clinical samples does not limit the practicability of the radioimmunoassay for erythropoietin.

Notes: Summary Using highly sensitive nucleic acids hybridization techniques, which allow the detection of 0.1–0.5 single copy gene equivalents per cell, DNA from the temporal cortex of seven definite schizophrenics, five persons with schizophrenia-like psychoses, three patients with Huntington's chorea and nine mentally normal individuals were probed with human cytomegalovirus (HCMV) DNA. A clear hybridization signal was obtained with DNA from the temporal lobe of a young schizophrenic patient, whereas DNA from the temporal cortex of controls did not hybridize to the HCMV probe. This finding is in agreement with the cytomegalovirus hypothesis of schizophrenia and hints at the possibility that viral infection of the temporal cortex may in some sporadic cases be a contributing factor to the development of schizophrenic psychoses. There is no indication, however, that infection of the central nervous system with HCMV is an aetiological factor in the great majority of schizophrenic disorders. Clearly further studies, preferably in situ hybridizations of whole brains, are needed to prove or disprove the cytomegalovirus hypothesis of schizophrenia.

Notes: Summary The Salmonella typhimurium LT2 metF gene, encoding 5,10-methylenetetrahydrofolate reductase, has been cloned. Strains with multicopy plasmids carrying the metF gene overproduce the enzyme 44-fold. The nucleotide sequence of the metF gene was determined, and an open reading frame of 888 nucleotides was identified. The polypeptide deduced from the DNA sequence contains 296 amino acids and has a molecular weight of 33 135 daltons. Mung bean nuclease mapping experiments located the transcription start point and possible transcription termination region for the gene. There is a 25bp nucleotide sequence between the translation termination site and the possible transcription termination region. This region possesses a GC-rich sequence that could form a stable stem and loop structure once transcribed (ΔG=-9 kcal/mol), followed by an AT-rich sequence, both of which are characteristic of rho-independent transcription terminators. The nucleotide and deduced amino acid sequences of the S. typhimurium metF gene are compared with the corresponding sequences of the Escherichia coli metF gene. The nucleotide sequences show 85% homology. Most of the nucleotide differences found do not alter the amino acid sequences, which show 95% homology. The results also show that a change has occurred in the metF region of the S. typhimurium chromosome as compared to the E. coli chromosome.

Notes: Summary The nucleotide sequence of the 3.4 kb SphI-G fragment that contained the repressor gene (c) of the temperate Streptomyces phage ϕc31 was determined. Analysis of this sequence revealed a large open reading frame with protein coding character and sequence changes in c gene point and deletion mutants identified this as the coding region of the repressor. Two of the mutants studied had undergone deletions of 1.1 kb and 1.4 kb that had occurred across short direct repeats of 6 bp and 11 bp, respectively. Coupled in vitro transcription-translation experiments using the cloned SphI-G fragment and Streptomyces lividans cell free extracts identified a protein product of approximately 72 kDa, in close agreement with that predicted from the nucleotide sequence. A strongly predicted helix-turn-helix motif that may be involved in DNA binding occurred towards the carboxy-terminus of the amino acid sequence. Initial attempts to clone the SphI-G fragment in Streptomyces failed; using information gained from the sequence analysis a smaller segment of this DNA fragment was cloned in S. lividans and conferred immunity to a clear plaque mutant (c1) of ϕc31.

Notes: Summary In cyanobacteria, light is harvested by phycobilisomes which are essentially made up of chromophoric proteins called phycobiliproteins. We have characterized two gene clusters (cpcB1, cpcA1 and cpcB3, cpcA3) each encoding the two subunits of phycocyanin (βPC and αPC, respectively), one of the major phycobiliproteins in Calothrix 7601. Downstream from the gene encoding the PCα subunit in cluster 1, an open reading frame was found, cpcE1. These genes are organized in two transcriptional units, namely: cpcB3 A3 and cpcB1 A1 E1. All these genes are transcribed whatever the chromatic light received during cell growth. Consequently, although only one type of “constitutive” PC has been biochemically characterized, we have demonstrated that there are two cpc operons “constitutively” transcribed in this strain. With the previously described red light “inducible” cpcB2 A2 operon, there are three copies of the PC encoding genes in Calothrix 7601. The significance of this newly described multigene family in cyanobacteria is discussed. We have also mapped the 5′ and 3′ termini of the major transcript from the cpc1 operon. Analysis of the 5′ untranslated region of this transcript has revealed alternative secondary structures which are proposed to play a role in the regulation of the expression of this operon.

Notes: Summary The processing of LamB-IGF-1 fusion protein and the export of processed IGF-1 (insulin-like growth-factor-1) into the growth medium was examined in the Escherichia coli host strain, JM101. Several strain or plasmid modifications were tried to increase export of periplasmic (Processed) IGF-1 into the growth medium of JM101. These included: (1) use of a lon null mutant strain to increase accumulation levels of unprocessed LamB-IGF-1 fusion protein; (2) use of an alternative drug resistance marker on the expression plasmid rather than beta-lactamase, thereby reducing any competition for processing of LamB-IGF-1 by signal peptidase; (3) examination of whether phage M13 gene III protein expression caused more periplasmic IGF-1 to be exported into the growth medium due to increased outer membrane permeability; and (4) examination of the effect of E. coli or yeast optimized IGF-1 codons. None of these strain or plasmid modifications caused any significant increase in export of IGF-1 into the growth medium of JM101. Solubility studies of LamB-IGF-1 and processed IGF-1 showed that virtually all of the LamB-IGF-1 and IGF-1 remaining within the cell after a 2 h induction period was insoluble. This implied that only soluble LamB-IGF-1 was processed to IGF-1 and that only soluble IGF-1 was exported into the growth medium. Taken together, the results indicated that LamB-IGF-1 and IGF-1 solubility were the limiting factors in secretion of IGF-1 into the periplasm and export of IGF-1 into the growth medium.

Notes: Summary We have worked out conditions for the study of competence development and genetic transformation in Streptococcus oralis NCTC 11427 (type strain), a species that contains choline in the cell wall. The peak of competence was found at the early exponential phase of growth and the optimal conditions for transformation were achieved with shuttle plasmids prepared from S. pneumoniae or from Escherichia coli serving as donor DNA. Transformation with dye-bouyant density gradient purified plasmid preparations followed first-order kinetics. The pneumococcal amidase can be expressed in S. oralis harbouring a plasmid carrying the lytA gene. This enzyme lysed the cell wall of the transformed cell in the presence of detergents.

Notes: Summary Several hundred bacterial isolates were screened for bacteriolytic activity by growing them on agar medium containing autoclaved, lyophilized Micrococcus lysodeikticus cells as the substrate. A Bacillus sp. producing the largest lytic zone was selected. A genomic bank of this selected bacterium was constructed in the multi-functional vector pTZ18R, with partial SauIIIA DNA fragments inserted at the SalI restriction site. Screening of 800 colonies of this bank for cell lysis gave 5 recombinants exhibiting lytic activity, as detected by analysis of extracts of sonicated Escherichia coli cells on denaturing polyacrylamide gels containing autoclaved, lyophilized M. lysodeikticus cells as the substrate. One clone (pBH2500), expressed inE. coli strain NM522, was found to code for a lytic enzyme corresponding, in molecular weight, to the 27 kDa Bacillus sp hydrolase. This clone with an insertion of 2.5 kb was then subcloned as a 929 bp EcoRI-SauIIIA fragment in pTZ18R (pBH929) and showed higher cell lytic activity. A unique open reading frame for a protein of 251 amino acids, followed by a putative terminator sequence, was found after a consensus ribosome binding site. A putative leader sequence was identified in the first 37 amino acids. One truncated subclone (pBH703), corresponding to 196 out of 251 residues from the protein N-terminal end, still possessed lytic activity.

Notes: Summary Two non-homologous chloramphenicol (Cm) acetyltransferase (CAT) genes, designated catA and catB, were cloned from Clostridium butyricum type strains and characterized by restriction mapping. Both genes are efficiently expressed in Escherichia coli and Bacillus subtilis. In contrast to analogous genes from staphylococci and bacilli, gene expression is not dependent on induction by Cm. The genes are considered as chromosomal, since no association with endogenous plasmids was detectable. Southern hybridization revealed a homology between catA and the staphylococcal Cm resistance plasmid, pC194. The subunit size of the clostridial CAT enzymes expressed in E. coli was determined as 22.5 kDa (catA) and 24 kDa (catB), respectively. The C. butyricum cat genes provide potentially useful selection markers for the construction of cloning vectors from cryptic clostridial plasmids.

Notes: The time delay of oxygen probe response to the signal from a fermenter makes identification of the volumetric oxygen transfer coefficient kLa by the dynamic method more complicated. A coupled model involving the transient-state oxygen balance of the fermenter together with the dynamic model of the oxygen probe must be then formulated, solved and identified.In this paper two simple models of air-lift loop fermenters have been proposed and a coupled mathematical model of the fermenter - oxygen probe system has been developed. The identification procedure was used to estimate kLa values in the fermenter with internal circulation flow on the basis of experimental measurements. A comparison of evaluated and experimental indications of the probes placed at various heights of the column proves that the model presented gives a possibility of the first-step approximation of kLa in loop fermenters.

Notes: The experimental technique for measurement of microbial culture heat evolution directly in fermenter has been described and its correctness analysed. Heat-to-oxygen ratio, Q0, of synchronized yeast culture in the absence of fermentative metabolism has been found to be practically independent of a cell cycle phase and close to the theoretical constant predicted by the mass-energy balance theory. Notes: A HPLC technique for the analysis of organic acids in the production of α-ketoglutaric acid was developed. The method was applied and optimized for the quantitative determination of citric acid, pyruvic acid, isocitric acid and α-ketoglutaric acid in fermentation solutions.As microorganism the yeast Yarrowia lipolytica and as substrates glucose or paraffins were used.The chromatographic separations were carried out by means of 50 and 100 × 8 mm i.d. glass columns packed with an anion-exchange resin based on an 8% cross-linked polystyrene-divinylbenzene copolymer.The relative errors ranged from 2.1% (α-ketoglutaric acid) to 5.2% (isocitric acid). The percent recovery values varied between 94.4% (isocitric acid) and 107.7% (pyruvic acid).The contents of organic acids in fermentation solutions after the microbial synthesis based on paraffins or glucose were compared.

Notes: SO2emissions arising by burning of coal represent an important ecological problem. Therefore, in international scale it is worked today in the development of techniques for solving this problem. A possibility consists in the microbial conversion of sulphur compounds of coal before its burning into compounds which not produce SO2 by burning. Sulphur is bound in inorganic and organic form in coal. The composition may strongly differ in dependence on the kind of coal and its deposit. For the bioconversion of different sulphur compounds specific microorganisms were used. The paper gives an overview about the situation and the tendencies of the research in this field and also some informations of our own research.

Notes: From different fungal genera some mutant strains were selected which in cellulase formation exhibit reduced catabolite-repression related to several soluble carbon sources. The cellulase production by these mutant strains on the basis of soluble carbon sources is possible if a fed-batch-technique is used for feeding the substrate.The optimum procedure is an oscillating-fed-batch-technique (OFB) with feed-back-controlled intermittent addition of the substrate. The substrate concentration in the medium is adjusted in such a manner to realize oscillations between a limitation for mycelium growth and non-limiting conditions under a substrate concentration which is yet below the repressing concentration for cellulase formation. A suitable feed-back-parameter for the control of substrate addition is the specific CO2-formation of the mycelium.Using OFB the growth in cellulase activity is nearly proportional to the feeded substrate whereas the growth in mycelium is reduced. The productivity of cellulase formation is enhanced in comparison to continuous substrate addition as well as under limiting and non-limiting conditions for growth of mycelium.

Notes: The oxygen uptake rate (OUR) was studied in a solid state fermentation process of dried citrus peel with the strain Aspergillus niger QH-2 in order to obtain the growth estimation of the microorganism in the system.The relationship between OUR, the maintenance coefficient (m) and the yield for oxygen consumption YO2 allows the estimation of the biomass rate if we consider that both parameters are not constants in some periods of the process.It was estimated that in the first 24th the strain has an specific growth rate of 0.174 h-1 with values for YO2 and m in the order of 2.84 g-cell/g-oxygen and 0.006 g-oxygen/g-cell ·h respectively.

Notes: Simultaneous utilization of methanol and glucose by Hansenula polymorpha (Torulopsis sp.) MH 26 results in an increase in growth yield of up to 18% in dependence of the mixing proportion. The distribution of carbon from 14C-methanol into biomass and carbon dioxide was investigated. The distribution pattern was strongly influenced by the mixing proportion showing that methanol plays an increasing role as an energy donor as the glucose content in the mixture increased. Due to these data and verified by theoretical considerations the effect on growth yield was discussed to be caused by an interconnection in the conversion of the individual substrates. This is attributed to glucose delivering the acceptor for C1-assimilation and resulting in a more efficient utilization of glucose carbon on the one as well as the energy content of methanol on the other hand.

Notes: Industrial processes of cultivation of aerobic microorganisms are characterized by too small transportation rates of oxygen from a gas into a cell. The search for new ways of productivity increase by means of intensification of oxygen mass transfer in the fermentation process is of great importance.The possibilities for use of chelate transition metal complexes (CTMC) capable of binding oxygen in biosynthetic processes have been studied.The formation of labial complex of the studied substances with oxygen is shown by spectrophotometry. The increase of the oxygen delution rate in water in the presence of CTMC is shown by physical absorbtion method. The rate increase of oxygen mass transfer from gas into liquid is calculated on the base of the sulphite method.The use of CTMC in the role of oxygen carriers in the yeast cultivation leads to the biomass increase up to 20%.

Notes: The aim of the investigations was to obtain - from the parent strain Aspergillus niger A.n. 33 - selectants with an increased ability of cellulolytic enzymes biosynthesis. Own selection methods allowed to receive two selectants A.n. 33/2 and A.n. 33/20 characterized by enhanced activities of saccharifying cellulase (respectively 0.11 and 0.14 FPU/cm3), endo-beta-1,4-glucanase (15.4 and 21.8 U/cm3) and cellobiase (0.6 and 1.4 IU/cm3) as compared with the parent strain (FPA - 0.09 IU, CMC - 8.2 U and CB - 0.1 IU/cm3). Moreover, the selectants differed in shape and size of conidial heads, in shape and colour of conidia, as well as in structure and shape of hyphae. Enzyme preparations obtained after ultrafiltration of liquid cultures were characterized by following activites: FPA-4-16 IU, CMC - 900-1800 U, CB - 60-120 IU and xylanase - 250-280 IU/cm3.

Notes: D,L-Ethionine was added in varying concentrations (0.1-1 mM) to two cell suspension cultures of Ruta graveolens. Growth, alkaloid formation and activities of some shikimate pathway-specific enzymes in these cultures were estimated. Also the effect of ethionine on shikimate pathwayspecific enzymes under in vitro conditions was followed. Growth is only slightly inhibited in supplemented cultures. Alkaloid formation is drastically reduced in a low-producing and to a lesser extent in the high producing cell line by ethionine. Activities of DAHP synthase, chorismate mutase, and anthranilate synthase in the presence of ethionine are in different Ruta strains to a varying degree affected.

Notes: A biobattery of a new type in which pH difference between two media serves as a source of free energy is described. The ability of some algae and other water plants to change markedly the pH of their surroundings according to the light conditions can be used to maintain two solutions at widely differing pH values. Notes: It is shown that Ca- and Al-pectate gel beads prepared by use of high molecular weight polygalacturonic acid (viz. polygalacturonic acid having a high STAUDINGER index) are well suited for cell immobilization. The pectate beads are much more insensitive to those ions and chemical agents which destructively act on alginate beads (such as phosphate, citrate, gluconate, lactate as well as a high excess of sodium, potassium, and/or ammonium ions), even without addition of gelling cations to the liquid phase.

Notes: A fungal strain suitable for protein enrichment of starchy raw materials was selected by evaluation of the growth rate, results of protein biosynthesis in solid state fermentation (SSF) and assessment of fungal biomass. The strain Aspergillus oryzae A.or. 11 selected for further studies was characterized by the radial growth rate Kr of about 300 μm/h and by the specific growth rate μ of about 0.100 h-1. Fungal biomass contained about 32% of crude protein in dry matter. The digestibility of this protein in vitro was close to 75%. Protein analysis for amino acids showed that the content of exogenous amino acids approached that in protein of the FAO standard. As a result of 34 - h culture of this strain in solid medium, net protein increased by about 5.0 g/100 g of starting medium d.m. at the cost of decomposition of about 20 g carbohydrates.

Notes: A method for the photometric measurement of dehydrogenase activities with the redox dye phenazine methosulphate at a fixed biomass concentration was applied for the investigation of idiophasic product synthesis. It was found that in citric acid fermentation by Yarrowia lipolytica which is characterized by a well defined separation into tropho-and idiophase the magnitude of product synthesis rates corresponds to the level of dehydrogenase activity at the end of the trophophase. In cellulase production by Trichoderma reesei the tropho-and idiophase are not well defined but a rise of dehydrogenase activity in the course of the idiophase is likely to correspond to an increase of the cellulase formating rate. The significance of the relation of dehydrogenase activity to production rates for monitoring industrial fermentations is discussed.

Notes: The extend of non-enzymatic browning and ascorbic acid loss during air drying of a slab are predicted by computer, utilizing moisture and temperature dependencies of reaction kinetics together with simulated data for either averaged moisture contents. Results show that the extend of quality loss is a function of the drying parameters.The computer model is useful for the optimization of quality retention during dehydration of biotechnological products and foods.

Notes: A fed-batch cultivation method was elaborated which 1. warrants simultaneous consumption of glucose and formate, 2. is easy to employ and provides reproducible results, 3. gives information about the role of formate in yeast growth, 4. results in almost identical conditions for parallel cultures on glucose and glucose/formate. The method was applied to Hansenula polymorpha and Saccharomyces cerevisiae. Relevant results were obtained and were comparable to those found previously under chemostatic conditions. Thus, the method is considered suitable for comparative investigations of yeast growth on glucose/formate to demonstrate the role of formate as an additional energy donor.

Notes: Investigations were conducted into the potential use of enzyme hydrolysed cassava whey for ethanol production by Saccharomyces cerevisiae Aspergillus niger grown on whct bran was used as crude enzyme source to saccharify the whey starch. The whey with an initial HCN concentration of 54.0μg/ml was fermented at pH 4.5 and 30°C in a one-step process to produce ethanol. A maximum ethanol concentration of 4.5% (v/v) was obtained in 120 h with a decrease in HCN level to 4.0 μg/ml. In a two-stage fermentation, in which the raw whey was pre-hydrolysed and under the same fermentation conditions, the unsterilized hydrolysate yielded alcohol content of 5.5% (v/v), while the sterilized hydrolysate gave higher alcohol yield, 7.5% (v/v), in 48 h. No HCN was detected in the fermented liquour at the end of the two-stage process.

Notes: The relative constancy of ATP content per unit of dry biomass in different microorganisms enables to use the specific, sensitive and quick luciferin-luciferase method for ATP assay for indirect quantitation of microbial biomass by measuring the ATP content in biomass samples.The aim of this work was to investigate the feasibility of using the ATP concentration for the monitoring of the growth of Escherichia coli K 12 W 3350 in different cultivation conditions. Statistically significant high correlation coefficients (r 〉 0.9) between the ATP content (ATP/ml) and the other growth characteristics examined showed that the ATP content of the culture is as suitable characteristic as the optical density of the culture, total cell number per ml or total cell volume per ml for the monitoring of the growth of E.coliin batch culture in mineral medium with glucose at different growth temperatures (27-42 °C) as well as in fed-batch culture.

Notes: This paper describes our experience with the use of high performance liquid chromatography in the analysis and preparation of several NAD-dependent dehydrogenases and oxygen-dependent oxidases. The chromatographic materials tested were from Pharmacia (Sweden), LKB (Sweden) and Lachema (Czechoslovakia), the columns were attached to the fast protein liquid chromatographic (FPLC) system from Pharmacia. The preparative use of high performance ion exchange, molecular sieve and hydrophobic interaction chromatographies as well as of chromatofocusing made it possible to prepare tens of milligrams of completely pure enzymes in several hours. In most cases a combination of two high performance methods was sufficient to yield a homogeneous enzyme. The purified enzymes were used as analytical reagents for determining the concentrations of several metabolites and activities of some enzymes. A biotechnological application of immobilized alcohol dehydrogenase for the production of reduced nicotineadenine dinucleotide from the oxidized form of the coenzyme is discussed in a greater detail.

Notes: The detailed thermal decomposition characteristics of several amino acids in different fase state - dry and moist crystals (Trp, Arg HCl, Lys HCl), water solutions (Trp) are determined. It is found that thermodecomposition of Trp in water solutions is inhibited by Na2S2O3 and oxyethylidendiphosphonic acid.The method of finding optimal control parameters (temperatur of moist crystals, heating surfaces, drying agent) during amino acid drying on the bases of their thermodecomposition kinetics is given. The received results can be used for technolodical optimization of different stages of amino acid extraction and concentration processes.

Notes: Suitable and optimized chromatographic separation systems for HPLC analyses of mono-, di- and oligomeric carbohydrates, organic acids (e.g. gluconic acid, α-ketoglutaric acid), phospholipids (PE, LPE, LPC, PC, SPH) and neutral lipids (squalene, cholesterol) are demonstrated.Applications of HPLC technique are separation examples of sugars from hydrolyzed starches which were isolated from potatos, calculations of organic acids in fermentation mediums and determinations of neutral lipids and phospholipids which were isolated from microbial biomass.The liquid chromatographic separations are based on self-packed highly efficient (approximately 80 000 theoretical plates per meter, N/m) glass columns.

Notes: The gelpermeation chromatography of sodium ligncsulfonates with bidistilled water as an eluent is influenced by the volume expansion of the polyelectrolyte, by the negative changes of the gels, and by the inclusion effect so markedly that true conclusions on the molecular weight distribution of sodium lignosulfonate cannot be drawn. Using electrolytes as an eluent these irregularities can be avoided. Polydispersity of these macromolecules however prevents separation into discrete peaks. As a result of this behaviour changes of molecular weight distribution are only poorly recognized. Therefore, on studying biotransformation of lignosulfonic acids we purpose to use gels by which one part of the lignosulfonates is excluded, and biotransformation may be controlled by the ratio of excluded part of the lignosulfonates to that part retarded by the gels.

Notes: In the late 70's an Austrian Consortium consisting of the STEYRERMÜHL-Papierfabriks- und Verlags AG, the Voest-Alpine AG and the Universities of Graz was formed to develop a new process for the production of monosaccharides from renewable carbohydrate sources, in particular lignocellulosic material. After detailed examination of various hydrolytic processes it was decided to intensify work on enzymatic hydrolysis. The composition of this group brought the great advantage that it could focus not only on one problem but could also research into the total process. This included pretreatment, enzyme production, hydrolysis, by-product utilization and energy supply.The raw materials studied most thoroughly on our program were waste paper and wheat straw, since these lignocellulosics are available in large quantities in Austria.Until now all endevour to produce ethanol out of lignocellulosic biomass on an industrial scale failed due to uneconomical production. Thus it is evident that the economy of such a process is very dependent not only on the yield of sugar, which is strongly affected by the respective pretreatment, but also on the utilization of the hemicellulose. Summarizing it can be said that the cellulose has to be degregated to a high extent and that the hemicellulose must be utilized to reach a feasible project.

Notes: S. cerevisiae cells immobilized in alginate beads show in many cases an increase of mean single cell volume during long-time fermentations (successive batch cycles). The biomass loading capacity of the gel beads is characterized by a maximum volume but not by a maximum number of cells occupying the gel volume. In our system this loading capacity, i.e. the maximum volume fraction of cells per volume of beads, amounted to about 0.54. As a more important result it must be stated that the specific product formation rate in the case of fermentations negligibly influenced by diffusion hindrance is related to the total surface of the viable cells but not to their total number, total volume or total dry weight.

Notes: Ability of deacetylation of Arthrobacter citreus (ATCC 11 624) was studied on three different types of substrates: 4,27-Di-O-acetyl-withaferin A, calycotomine derivatives and isoindolo/1,2-b//3/benz-azepine-5-one derivative. Only O-acetyl groups were cleaved by this strain, N-acetyl group was not attacked. This deacetylation was not regioselective; by hydrolysis of racemic acetates there was only slight stereospecificity observed. As this reaction took a short time and afforded high yields under moderate conditions it may be useful in organic chemistry and/or in the structure elucidation of natural products.

Notes: Yeasts were chemostatically cultivated on C2-substrates. Experimental yield figure were 0.77 g/g and 0.57 g/g on ethanol, 0.37 g/g and 0.29 g/g on acetate and ammonium and nitrate, respectively, as the nitrogen sources. Growth on hexadecane/ammonium yielded 0.94 g/g. Simultaneous utilization of formate lead to an increase of growth yield on acetate and hexadecane but not on ethanol. The experimental data were compared to theoretical figures. These results show that growth yield on ethanol/ammonium is carbon-limited, on ethanol/nitrate is NADPH-limited and on acetate with both nitrogen sources is energy-limited. With hexadecane/ammonium growth yield is energy-limited too. According to these results ethanol seems to be the only substrate from which an excess in energy can be obtained.

Notes: A laboratory version of orgoro; a low alcohol plantain beverage was found to possess a better odour, taste and colour after 7 days of fermentation compared with a native brew. The pH of the native brew decreased from 3.90 to 1.93 after 21 days of fermentation while the filterable acidity increased from 0.100 to 0.157%. The laboratory brew showed a decrease of pH from 4.60 to 2.72 and an increase in filterable acidity from 0.250 to 0.485%. Both brews had alcohol contents of 0.109 and 0.197% (v/v) respectively after this period. The total sugars content, soluble protein and total nitrogen contents of the native brew were 2.0 mg/ml, 1.3 mg/ml and 2.05 mg/ml respectively while values for the laboratory brew were 5.0 mg/ml, 2.2 mg/ml and 4.0 mg/ml respectively.

The expression of pTGH201 encoded genes and the kinetics of plasmid loss differ significantly from the behaviour of pBR322 plasmid.

Notes: Comparative investigations for the possibilities of milk coagulation by the action of soluble and immobilized (on DEAE-cellulose by adsorption) bacterial enzyme preparation “syrenin” in periods from 30-120 minutes at 5°C are carried out. The milk clotting activities of both preparations are equalized after 40 minutes. The laboratory experiments for producing a bulgarian white brine cheese by the immobilized “syrenin” show a tendency to decrease the losses of substance into the whey and to increase the curdle yields in comparison with these made by soluble preparation.

Notes: In this paper we studied the conditions for the production of β-glucosidase from T. reesei QM9414 in batch cultures using milled and sieved wheat straw as sole carbon source. High β-glucosidase production in the presence of wheat straw, a more realistic substrate than commercial cellulose, was obtained.The influence of particle size of wheat straw on β-glucosidase production in cell-free, cell and cell-wall extracts was studied. The particle size of wheat straw notably influenced enzyme production in cell and extramycelial extracts but it was less important with respect to the cell wall bound enzyme.β-glucosidase production was studied along of the fermentation. The results suggest a close relation between β-glucosidase from cell extract and extramycelial broth; geneticin levels of inhibition of β-glucosidase biosynthesis in both fractions were similar, a fact that suggests a common origin for the enzyme.Kinetic parameters for β-glucosidase from cell free and cell extracts were Vmax = 0.28 μmol/min/mg, KM = 0.91 mM and Vmax = 0.095 μmol/min/mg, KM = 0.39 mM respectively. Kinetic parameters for β-glucosidase from cell-wall could not be calculated because experimental data did not fit the different monosubstrate equations.

Notes: The role of the microbial secondary metabolism in the cytodifferentiation of producer strains and in the ecological system is reviewed. The production of bioactive metabolites such as antibiotics seems to play a role in long-term strategies of adaptation to environmental changes. This is reflected by characteristic features of regulation of the secondary metabolism such as catabolite repression, lack of uniform regulatory mechanisms and limited specificity of enzymes involved. The examples given may show that advantages have been taken from the knowledge of special regulatory features with regard to strain improvement and the designing of the metabolite spectrum.

Notes: Changes in chemical composition and morphology of wheat straw during a 5-step treatment with delute aqueous sodium hydroxide of increasing (step 1 to 3) and then decreasing (step 4 and 5) NaOH concentration are described varying straw pretreatment, maximum NaOH concentration and total time of contact between straw and lye. These structural changes are correlated to increase in swelling, in-vitro degradability by cellulase and digestibility as revealed by feeding tests with sheep. Results are discussed on the basis of the processes of swelling, delignification and pentosan extraction, and the technological advantages of this multi-step treatment for enhancing the nutritional value of straw are discussed in comparison to a one-step treatment with subsequent lye-removal by pressing.

Notes: Potato starch hydrolysis products were analyzed for glucose, maltose, higher oligomeric carbohydrates (to a degree of polymerization, dp 11) and 5-hydroxymethylfurfural (HMF) using high-performance liquid chromatography.For quick qualitative and quantitative analyses short glass columns [12] of high efficiency were useful.The hydrolyses were carried out by means of enzymes (e.g. α- and β-amylase) or mineral acids. For the acid degradation procedures hydrochloric acid, sulfuric acid and phosphoric acid of different concentrations (0.1…2.0 N) during times ranging from 5 to 60 min at temperatures ranging from 100 to 140°C were used.Maximum glucose contents (163.4 g/l and 169.3 g/l) were found after 40 to 50 min of hydrolysis in 0.1 N hydrochloric acid heated to 120°C. These values are equivalent to 78.9% or 81.7% glucose yield referred to the initial potato starch amount, respectively. The calculated HMF concentrations were 140 and 180 mg/l.
