Notes: Summary 1. Stimulation through concentric electrodes on the surface of a wide cerebellar folium was employed to set up a parallel fibre volley or beam. Serial recording of the field potential was made over a range of depths along microelectrode tracks arranged in a transverse plane across the folium in order to discover the action on Purkinje cells, both those that were on-beam for the parallel fibre volley and those at various distances off-beam. A juxta-fastigial electrode was carefully placed so that an applied stimulus could excite the axons of Purkinje cells distributed across the folium under investigation, the antidromic propagation of impulses thus obtained being utilized to test the effect of parallel fibre volleys upon Purkinje cells. 2. The observations were in accord with the two actions that a parallel fibre volley would be expected to exert on Purkinje cells: a direct excitatory action by the synapses made by parallel fibres with the spines of the Purkinje cell dendrites; an inhibitory action mediated by the stellate and basket cells that themselves are directly excited by the parallel fibre volley. 3. The excitatory synaptic action would result in the two types of responses that were restricted to the narrow zone and superficial location of the parallel fibre volley: active sinks formed by this excitatory synaptic action on the superficial dendrites of Purkinje cells would account for the observed depth profile of extra-cellular slow potentials, a superficial negative wave reversing to a deeper positive wave formed by passive sources on deeper dendrites; superficial synaptic excitation would also account for the facilitation of the propagation of antidromic impulses into the superficial dendrites. 4. The inhibitory synaptic action would result in the two types of responses that were widely dispersed transversely and in depth, far beyond the traject of the parallel fibre volley: a slow positive potential wave with a maximum at a depth usually of 300–400 μ; an inhibitory action on the antidromic invasion of Purkinje cells. The transverse profiles of these two presumed indices of inhibitory action on Purkinje cells apparently revealed that a basket cell may give inhibitory synapses up to 1000 μ laterally from the location of its soma and dendrites. 5. A description is given of the variants in the transverse profiles of the deeper positive waves and of inhibitory actions of a parallel fibre volley that presumably are mediated by basket cells and also by the superficial stellate cells. These physiological findings are correlated with the histologically determined distribution of synapses from a basket cell onto Purkinje cells.

When electrical stimuli were applied to the surface of a cerebellar folium by a local electrode (LOC), there was a propagated potential wave along the folium with a triphasic (positive-negative-positive) configuration. 2. Investigations by microelectrode recording established that this wave is produced by impulses propagating for at least 3 mm and at about 0.3 m/sec along a narrow superficial band or “beam” of parallel fibres. As expected from this interpretation, there was an absolutely refractory period of less than 1 msec and impulse annihilation by collision. 3. Complications occurred from the potential wave forms resulting from the excitation of mossy fibres by spreading of the applied LOC stimulus. These complications have been eliminated by chronically deafferenting the cerebellum. 4. When recording within the beam of excited parallel fibres there was a slow negative wave of about 20 msec duration, and deep and lateral thereto, there was a slow positive wave of approximately the same time course. 5. These potential fields were expressed in serial profile plots and in potential contour diagrams and shown to be explicable by the excitatory and inhibitory synaptic action on Purkinje cells: excitatory depolarizing synapses of parallel fibre impulses on the dendrites; and hyperpolarizing inhibitory synapses of stellate and basket cells respectively on the dendrites and somata. The active excitatory synapses would be strictly on the parallel fibre beam and the inhibitory concentrated deep and lateral thereto, which is in conformity with the axonal distributions of those basket and stellate cells that would be excited by the parallel fibre beam. 6. Complex problems were involved in interpretation of slow potentials produced by a second LOC stimulus at brief stimulus intervals and up to 50 msec: there was a potentiation of the slow negative wave, and often depression of the positive wave deep and lateral to the excited beam of parallel fibres. 7. Often the LOC stimulus evoked impulse discharge from the Purkinje cells, these discharges being inhibited by a preceding LOC stimulus.

Intracellular recording from Purkinje cells has been employed in investigating the excitatory and inhibitory synaptic action that is exerted on these cells by the mossy fibre input into the cerebellum. 2. These synaptic actions are evoked not directly by the mossy fibres, but probably always through granule cells and their axons, the parallel fibres. The intracellular records conform with the anatomical evidence that the parallel fibres directly exert a powerful synaptic excitatory action on Purkinje cells, and that the inhibitory pathway occurs via an inhibitory interneurone — a basket cell or a stellate cell. Direct stimulation of parallel fibres gives intracellular potentials closely resembling those produced by deep stimulation of mossy fibres. 3. As would be expected, direct stimulation of parallel fibres produces an EPSP with a latency 1 to 2 msec briefer than the IPSP. The IPSP has a duration usually in excess of 100 msec. The EPSP appears to be briefer, though its superposition on the IPSP greatly reduces its apparent duration. Neutralization of the IPSP by appropriate membrane polarization or by intracellular chloride injection reveals an EPSP duration of up to 50 msec. 4. The IPSP is typically affected by polarizing currents; reduced and even inverted by hyperpolarizing currents, and increased by depolarizing currents. The IPSP is converted to a depolarizing response by excess of intracellular chloride. It must therefore be generated by an increased ionic permeability of the inhibitory subsynaptic membrane, chloride ions being importantly concerned. 5. Often small irregular IPSPs can be observed occurring spontaneously, and they react to polarizing currents and to chloride injections in a manner identical to the evoked IPSPs. It is concluded that they are generated by the spontaneous discharges of basket cells. 6. A brief account is given of various spontaneous rhythmic responses of impaled Purkinje cells, and of the effect of synaptic inhibitory action upon them. 7. There is a general discussion of these findings in relation to the various neural pathways and neural mechanisms that have been postulated in the light of the preceding investigations.

Notes: Summary During stimulation of the anterior lobe of the cerebellum, postsynaptic potentials were recorded intracellularly from ipsilateral Deiters neurones of the cat. In the majority of examined cells, the inhibitory postsynapic potentials were induced with short latency; 1.06 msec on the average from lobule III or IV. The latency was longer (1.23 msec) when the lobule V was stimulated, while it was shorter (0.86 msec) from the juxtafastigial region. It follows that the IPSP was produced via a monosynaptic pathway at a conduction velocity of 15 to 20 m/sec. Recording of the extracellular field potentials and focal stimulation within and around Deiters' nucleus further indicated that the inhibitory impulses propagated out of the cerebellum along a remarkable bundle of fibres which terminated within Deiters' nucleus. These results are all explicable by assuming that the cerebellar Purkinje cells are inhibitory in nature and so produce IPSPs monosynaptically in Deiters neurones via the long corticofugal fibres. Monosynaptic EPSPs were also detected in some Deiters neurones. They are considered to be mediated by the other pathways formed of axon collaterals of the cerebellar afferents.

Notes: Chloroform und 1.1-Difluor-1.2.2-trichlor-äthan (F 122)F 122 (= Frigen 122) als Kurzbezeichnung entspr. Frigen, Freon, Kaltron u. ä. Handelsbezeichnungen®. wurden zwischen 100 und 270° mit ClF3 und zum Vergleich mit elementarem Chlor in der Gasphase umgesetzt. Chloroform reagierte sowohl mit Cl2 als auch mit ClF3 leichter als F 122. Bei der Halogenierung von F 122 macht sich der stabilisierende Einfluß von Halogenatomen in α-Stellung zum substituierbaren H-Atom bemerkbar. Chloroform wird durch ClF3 lediglich fluoriert. Bei der H-Substitution von F 122 betrug das Verhältnis Cl: F der aus dem ClF3 stammenden Halogene in den Reaktionsprodukten etwa 1 : 1, so daß der Rest des Fluors als HF verlorenging. Dies gilt bei vielen organischen Verbindungen für die Halogenierung mit ClF3.

Diese Reaktion ergibt eine experimentell einfache Methode zur Überführung von Acetalen und Orthoestern unter Kettenverlängerung in Carbonsäureester. - Offenkettige und cyclische Acetale, die nicht nach (5) umgesetzt werden konnten, sind in den „Sonderbesprechungen“ zusammengefaßt; 1.3-Dioxolan liefert bei der Behandlung mit katalytischen Mengen BF3 und wenig Diazoessigester ein festes, farbloses Polymerisat.

Notes: Aromatische Azide und Benzylazid lagern sich an α.β-ungesättigte Carbonester, Ketone und Nitrile so an, daß der elektronenanziehende Substituent in der 4-Stellung des Δ2-1.2.3-Triazolins erscheint. Unter Basenkatalyse gehen die 1-substituierten Δ2-Triazolin-carbonsäure-(4)-methylester vollständig in ringoffene N-substituierte 3-Amino-2-diazo-propionsäureester über, die ihrerseits als aliphatische Diazoverbindungen zur erneuten Cycloaddition an α.β-ungesättigte Carbonester befähigt sind. Aus den Azid-Additionen an Dimethyl-fumarat oder Äthyl-crotonat ließen sich zum Teil nur die offenkettigen Diazokörper oder deren Folgeprodukte isolieren. Bei den Δ2-Triazolinen aus Äthyliden-aceton und Methyl-vinylketon ist die Ringöffnung besonders rasch. - Das aus Phenylazid und Acrylnitril hervorgehende 1-Phenyl-4-cyan-Δ2-triazolin setzt sich unter Triäthylamin-Katalyse mit 29% 3-Anilino-2-diazo-propionitril ins Gleichgewicht.

Das Hydroxylaminderivat von 1 ist 2.3.4.5-Tetrachlor-pyridin-N-oxid, das von 2 eine O-Acyl-hydroxamsäure, die bei der Hydrolyse zu 3.4.5-Trichlor-N-hydroxy-pyridon-(2) cyclisiert. Dem Aldehyd 1, der zugehörigen Carbonsäure und einigen verwandten Verbindungen wird auf Grund ihres Reaktionsverhaltens die cis-Konfiguration zugesprochen.

Notes: Aus p-Benzochinon-bis-dimethylsulfamoylimin (1) und Enaminen bilden sich substituierte 5-Dimethylsulfamoylamino-2-piperidino(morpholino)-2.3-dihydro-indole (2), aus denen sich das Amin in 2-Stellung leicht eliminieren läßt. 1 reagiert ebenfalls mit dem Enolat des Acetessigaldehyds in Dimethylformamid oder Dimethylsulfoxid zu 5-Dimethylsulfamoylamino-1-dimethylsulfamoyl-3-acetyl-indol (3f). Die Schutzgruppe in 1-Stellung am Indolring ist generell leicht abspaltbar, in 5-Stellung gelang die Spaltung noch nicht.

Amino-triazolen (15, 16). - Cyansäure setzt sich mit 2 Moll. 1 zu Bis-aryl-cyanuraten (24) um, während 2 Moll. Rhodanwasserstoffsäure und 1 Mol. 1 2-Aroxy-4.6-dimercapto-s-triazine (26) bilden.

Notes: Die Methode der Kettenverlängerung von Carbonsäuren um 5 oder 6 C-Atome wird auf ihre Eignung zum Aufbau methylverzweigter Carbonsäuren untersucht. Das Studium jedes einzelnen Reaktionsschrittes ermöglicht es, ohne Reinigung von Zwischenstufen mit ∼50% Ausbeute Carbonsäuren aufzubauen, die mit Ausnahme der 3-Stellung in jeder Position eine Methylgruppe tragen können. Von entscheidender Bedeutung ist die Beeinflußbarkeit der Spaltungsrichtung von 2-Methyl-2-acyl-cyclohexanonen durch die Reaktionsbedingungen.

Einerseits ließen sich in geeigneten Lösungsmittelsystemen weitere Carbonylsalze [Re(CO)4L2][Me(CO)nL′] (L2 = 2 CO, 2 P(C6H5)3, o-Phenanthrolin; Me = V, Mn, Fe, Co; L′ = CO, NO) fassen, andererseits wurden infolge einer Redoxreaktion zwischen Carbonyl-Kation und Carbonylmetallat gemäß \documentclass{article}\pagestyle{empty}\begin{document}$$ [{\mathop{\rm Re}\nolimits}{\rm (CO)}_{\rm 4}{\rm L}_2]^ \oplus \; + \;[{\rm Me(CO)}_{\rm n}]^\Theta \; \to \;{\rm L}_{\rm 2}{\rm (CO)}_{\rm 3}{\mathop{\rm Re}\nolimits} - {\rm Me(CO)}_{\rm n} \; + \;{\rm CO} $$\end{document} gemischte Metallcarbonyle mit einer einfachen Metall-Metall-Bindung erhalten. Dieses sehr allgemein anwendbare Reaktionsprinzip lieferte hier die Metallcarbonyle (CO)5 Re—Mn(CO)5, (CO)5Re—Co(CO)4 und deren o-Phenanthrolinderivate. IR-Spektren und Struktur der Verbindungen werden diskutiert. - Natrium-pentacarbonylrhenat(-I) bildet mit Chlorameisensäure-äthylester das flüchtige Carboäthoxopentacarbonylrhenium, das von Chlorwasserstoff in Carbonyl-Kation und Äthanol gespalten wird: Anhand der Reaktionsweise kationischer Kohlenoxidkomplexe von Mangan(I) und Rhenium(I) mit Alkoholaten und Hydroxyl-Ionen wird eine neue Anschauung über die Basen-reaktion neutraler Metallcarbonyle mit Alkalilaugen entwickelt. Als einleitender Schritt wird der nuceophile Angriff eines Hydroxyl-Ions am C-Atom eines CO-Liganden postuliert.

Bei [BX4]⊖-Verbindungen ist δ11B sowohl von der Elektronegativität als auch von Anisotropie-Effekten der Liganden bestimmt, während bei den dreibindigen Bor-Verbindungen die π-Wechselwirkung einen entscheidenden Beitrag zur chemischen Verschiebung beisteuert. δ11B wird hauptsächlich von den ‚Erstliganden‘ bestimmt, d. h. jenen Atomen, die direkt an das Boratom gebunden sind. Sie ist auf Grund empirisch ermittelter Faktoren vorauszusagen und besitzt deshalb analytischen Wert.

Notes: Das aus 3-Methylen-bicyclo[3.3.1]nonanon-(7) (1) durch Lithiumalanat-Reduktion erhältliche 3-Methylen-bicyclo[3.3.1]nonanol-(7) (2) geht mit Säuren in 1-Methyl-2-oxa-adamantan (3) über. 1 erleidet mit Quecksilberacetat einen ähnlichen Ringschluß unter Bildung von 3-Hydroxy-1-acetoxymercurimethyl-2-oxa-adamantan (4), das in 3-Hydroxy-1-methyl-2-oxa-adamantan (6) überführbar ist. Diese Verbindung entsteht auch bei der Einwirkung von Methyllithium oder Methylmagnesiumjodid auf Bicyclo[3.3.1]nonandion-(3.7). Analog reagieren auch andere metallorganische Verbindungen.

Notes: Bei der Umsetzung von Cyanharnstoff mit ω-Brom-acetophenon oder einigen p-Substitutions-produkten entstehen in Abhängigkeit von den Reaktionsbedingungen 4-substituierte 1-Cyanimidazolone-(2) 1 bzw. 5-substituierte 2-Amino-oxazole 8. Der nucleophile Angriff des ambidenten Cyanharnstoff-Anions auf die polarisierte C—Br-Bindung der α-Bromketone erfolgt also in beiden Fällen über das N-Atom. Die 1-Cyan-imidazolone-(2) lassen sich leicht zu 1-Carbamoyl-imidazolonen-(2) 3 bzw. unter gleichzeitiger Decarboxylierung zu Imidazolonen-(2) 2 verseifen. - Setzt man bei obiger Reaktion die doppelte Menge Cyanharnstoff ein, so erhält man mit den ω-Brom-acetophenonen sowie mit 2-Brom-butanon, α-Brom-propiophenon und Desylbromid die entsprechend substituierten 1-Cyanguanyl-imidazolone-(2) 4.

Notes: 5.5′- und 6.6′-Diaza-indigo ließen sich durch Alkalischmelze des N-[3-Carboxy-pyridyl-(4)]-glycins bzw. des N-[4-Carboxy-pyridyl-(3)]-glycins und nachfolgende Luftoxydation darstellen. Die Pyridylglycin-o-carbonsäuren (19 bzw. 10) konnten durch Hydrierung des Kondensationsproduktes aus den entsprechenden Aminopyridin-o-carbonsäuren mit Glyoxylsäureester erhalten werden. Die klassische Umsetzung der Aminopyridin-o-carbonsäuren mit Chloressigsäure führte zu Pyridinbetain-Verbindungen.

Notes: 2.4.6-Triphenyl-4H-pyran reagiert mit Dichlorcarben (aus Natrium-trichloracetat) unter Addition an eine der Doppelbindungen. Mit 4H-Pyran erhält man neben dem einfachen und zweifachen Additionsprodukt (8 und 10) auch ein Einschiebungsprodukt in die CH-Bindung an C-4 (9). In ähnlicher Weise reagiert auch 4H-Thiopyran. Die Einschiebungsprodukte 9 bzw. 17 sind äußerst empfindlich. Die Umwandlung in ein Derivat des Oxepins bzw. Thiepins gelang in keinem Fall. Im Anhang werden die NMR-Spektren von Pyrylium- und Thiopyryliumfluoroborat mitgeteilt.

Die 5-fach koordinierten Stibin-haltigen Carbonyl-Kationen bilden mit Kaliumalkoholaten Carboalkoxocarbonyle, RO2CMe(CO)L3 (R = CH3, C2H5), die bei der Spaltung mit Chlorwasserstoff in Hydridokomplexe, HMe(CO)L2Cl2, übergehen. Die IR-Spektren werden hinsichtlich der Struktur der Verbindungen diskutiert.

Notes: Für die Synthese von Phosphorsäure-o-phenylenester-chlorid bzw. -bromid (2a bzw. 2b) wird ein neues Verfahren beschrieben. 2a läßt sich in die zugrundeliegende Säure, den Phosphorsäure-o-phenylenester (5) überführen, dessen Struktur u. a. durch Reaktion mit Diazoverbindungen gesichert wird. - Weiterhin wird das Verhalten von 2a gegenüber äthern, Acetalen und Estern beschrieben. Die aus 2a und Carbonsäuren bzw. Dimethylformamid erhältlichen gemischten Anhydride bzw. Säureamidkomplexe sind zur übertragung von Acylresten geeignet.

Notes: Bei der Behandlung eines Gemisches von 2-Methyl-1.2-dihydro-papaverin und 2-Methyl-1.2-dihydro-barbonin mit verdünnter Säure entstehen nicht zwei, sondern vier 3.4-Dihydroisochinoliniumsalze. Dies ist ein Beweis, daß die prototrope Umlagerung von 2-Methyl-1.2-dihydro-papaverin nicht intra-, sondern intermolekular abläuft. Die Struktur der Umlagerungsprodukte wird durch Synthese gesichert.

Notes: Die Synthese von Fünfring-Siebenring-Aromaten-π-Komplexen des Chroms durch Ringaustausch von C6H6 in C5H5CrIC6H6 (1) mit C7H7R (R = H, CH3, C6H5)/AlCl3 bzw. C7H7BF4 wird beschrieben. C5H5Cr0C7H7 (4) und sein Kation [C5H5CrIC7H7]+ (2) werden an Hand physikalischer Daten und chemischer Eigenschaften mit C5H5V0C7H7 sowie dem isomeren System Cr0(C6H6)2/[CrI(C6H6)2]+ verglichen. Durch Metallierung von C5H5Cr0C7H7 mit Amylnatrium und Umsetzung mit C6H5CHO sind C5H5Cr0C7H6COC6H5 (B) und C6H5COC5H4Cr0C7H7 (C) zugänglich. [C5H5CrIC7H7]J (6) reagiert mit LiC6H5 außer unter Reduktion zum Cr0-Komplex auch unter Addition zu C5H5CrIC7H7C6H5(7).
