Durchtritt und Bindung von 3H-markiertem Ouabain, Digitoxin, Digoxin, Peruvosid und Proscillaridin wurden am isolierten, durchströmten Dünndarm von Ratten und Meerschweinchen in vitro untersucht. 2. Der Durchtritt der Glykoside durch die Darmwand ist proportional der Konzentration. Für die Penetrationsfähigkeit ergibt sich bei der Ratte die Reihenfolge: Digitoxin, Peruvosid, Proscillaridin = Digoxin, Ouabain; für das Meeschweinchen: Proscillaridin, Peruvosid = Digitoxin, Ouabain, Digoxin. 3. Die Bindung von Digitoxin im Darmgewebe ist bei Ratte und Meerschweinchen am größten. Das Verhältnis der Glykosid-Gehalte pro g Gewebe und pro ml Durchströmungsflüssigkeit betrug bei der Ratte für Digitoxin 3,3, für Ouabain 0,18, für Digoxin 0,6, für Peruvosid 1,1 und für Proscillaridin 1,2; beim Meerschweinchen für Digitoxin 4,2, für Ouabain 0,8, für Digoxin 0,16, für Peruvosid 0,9 und für Proscillaridin 1,4. 4. Auch in vivo, an abgebundenen Jejunumschlingen nimmt bei Ratten die resorbierte Menge proportional mit dem Angebot zu. Die Retention hängt vor allem von der mit der Galle ausgeschiedene Menge ab. 5. Es ergab sich kein Anhaltspunkt dafür, daß bei der Resorption der Glykoside ein Prozeß mit begrenzter Kapazität limitierend wird.

Penetration and binding of tritiated ouabain, digitoxin, digoxin, peruvosid and proscillaridin were studied on isolated segments of the small intestine of rats and guinea pigs in vitro. 2. Penetration of glycosides through the intestinal wall is proportional to concentration. In rat intestine the penetration rate follows the order: digitoxin, peruvosid, proscillaridin = digoxin, ouabain; in guinea pig intestine: proscillaridin peruvosid = digitoxin, ouabain, digoxin. 3. In intestinal tissue of rats as well as of guinea pigs binding of digitoxin is highest. The relation of the glycoside content per g intestine to the content per ml perfusion fluid in the rat is 3.3 for digitoxin, 0.18 for ouabain, 0.6 for digoxin, 1.1 for peruvosid and 1.2 for proscillaridin; in the guinea pig 4.2 for digitoxin, 0.8 for ouabain, 0.16 for digoxin, 0.9 for peruvosid and 1.4 for proscillaridin. 4. Also in vivo the absorption of glycosides in tied loops of rat intestine is proportional to the amount offered. The retention of glycosides depends mainly on the excretion via the bile. 5. There is no indication that the absorption of glycosides depends on a process of limited capacity.

Bei Urethan-narkotisierten Ratten wurde eine Jejunumschlinge mit14C-markierten Arzneimitteln in gepufferter isotonischer Kochsalzlösung mit einem pH von 6–8 (Amidopyrin, Anilin, Antipyrin, Benzoesäure, Salicylsäure) und einem pH von 2,2–3,0 (Amidopyrin, Benzoesäure) perfundiert. 2. Wurde die Durchblutung von etwa 1,5 auf 0,2 ml/min · g Feuchtgewicht gesenkt, dann nahm die Resorptionsrate aller Arzneimittel ab. Wurde umgekehrt die Durchblutung von anfänglich niedrigen Werten auf hohe Werte gesteigert, dann stieg die Resorption der nichtionisierten Arzneimittel in entsprechender Weise an, während die Resorption der ionisierten Arzneimittel nicht beeinflußt wurde. Bei gleichbleibender mittlerer Durchblutung (0,6–0,7 ml/min · g) blieb die Resorptionsrate der nichtionisierten Arzneimittel (mit Ausnahme von Amidopyrin) konstant, während sie bei den ionisierten über einen Zeitraum von 60 min um 15–22% abnahm. 3. Das abweichende Verhalten der ionisierten Arzneimittel gegenüber Durchblutungsänderungen wird auf folgenden Mechanismus zurückgeführt: maßgebend für ihre Resorption ist der leicht saure und von der Perfusionslösung weitgehend unabhängige pH-Wert (virtual pH) in einem Bereich unmittelbar an der Mucosaoberfläche (microclimate). Eine verminderte Mucosadurchblutung führt nicht nur zu einer Verringerung der Dränagewirkung (die alle Pharmaka betrifft), sondern auch zu einer hypoxischen Schädigung des Zottenepithels und einer Anderung des virtuellen pH, die nur die Resorption der ionisierten Arzneimittel beeinträchtigt.

Jejunal loops were prepared in anaesthetized rats and perfused with14C-labelled drugs in buffered isotonic saline solutions at pH 6–8 (amidopyrine, aniline, antipyrine, benzoic acid, salicylic acid) and at pH 2.2–3.0 (amidopyrine, benzoic acid). The blood flow in the loops and the absorption rate of the drugs were determined simultaneously. 2. A decrease of the blood flow from about 1.5 to O.2 ml/min X g wet tissue diminished the absorption rate of all drugs, whereas an increase of the blood flow from low to high values caused only an increased absorption rate of the unionized but not of the ionized drugs. When the blood flow was held constant (0.6–0.7 ml/ min X g wet tissue), the absorption rate of the unionized drugs (with the exception of amidopyrine) remained constant, whereas the absorption rate of the ionized drugs decreased by 15–22% within 60 min. 3. The different reaction of the ionized drugs to blood flow alterations is assumed to be due to the following mechanism: the absorption of the ionized drugs is substantially favoured by a “virtual pH” near the mucosal border which is slightly acidic and largely independent of the pH within the gut lumen. A period with insufficient mucosal blood flow results not only in a diminished drainage of the mucosa-which affects the absorption rate of all drugs-, but also in a hypoxic impairment of the epithelium and its capability to maintain the “virtual pH”. This failure affects only the absorption rate of the ionized drugs.

    Notes: Summary An NAD dependent “digitoxose dehydrogenase” has been demonstrated in extracts from rat liver; the Km values of this enzyme have been determined. Polyacrylamide disc gel electrophoresis shows that there are at least four different proteins capable of oxidizing digitoxose. After i.p. injections of digitoxose into normal rats 53.3% of the deoxysugar injected are excreted in the animals' urine as digitoxose, digitoxonic acid or digitoxonic-γ-lactone. The corresponding figure for adrenalectomized rats instead of normal animals is 68.3%-“Digitoxose dehydrogenase” of the rat liver seems to be a constitutive enzyme. The increased excretion of digitoxonic-lactone following adrenalectomy can be interpreted as a “permissive effect” of the adrenal hormones upon digitoxose metabolism beyond the stage of the digitoxonic lactone.

    Description / Table of Contents: Zusammenfassung Es wurde der Einfluß verschiedener Chelatbildner auf die Verteilung und Ausscheidung von trägerfreiem und isotopisch verdünntem Radioeisen bei der Ratte untersucht. Während bei gleichzeitiger Verabfolgung der Chelatbildner die Retention in allen Geweben herabgesetzt wird, führt die nachträgliche Verabreichung zu einer nur geringfügigen Mobilisierung von Depoteisen aus Leber und Milz. Die Wirksamkeit ist im Falle von isotopisch verdünntem Radioeisen größer. Äthylendi-(α-o-hydroxyphenyl)glycin, 2-(β-Aminoäthoxy)cyclohexylamintetraacetat und Desferrioxamin B erwiesen sich als die wirksamsten Verbindungen, wobei jedoch die hohe Toxicität der ersteren die praktische Verwendung ausschließt.
    Notes: Summary The influence of various chelating agents on the distribution and excretion of radioiron, carrier-free or diluted with stable carrier, was studied in rats. Simultaneous administration of chelating agents lowers the retention of radioactive iron in all tissues whereas after delayed treatment a small fraction of storage iron can be removed from liver and spleen. The efficacy is higher in the case of isotopically diluted radioactive iron. The most effective compounds are ethylenedi-(α-o-hydroxyphenyl)glycine, 2-(β-aminoethoxy)cyclohexylaminetetraacetate, and desferrioxamine B. The practical value of the former chelator, however, is excluded by its high toxicity.

La différence entre les manches et les bouts était, remarquable, mais pas entre les os séparates; en les tibiae, qui etait fracturé depuis 7 semaines et guéries, la relation etait identique à celui des tibiae manches normals. On a présumé que la découverte était due à des différences qualitatives plustot que quantitatives entre les procédé de minéralisation dans l'os cortical et trabeculair.
    Abstract: Zusammenfassung Das Verhältnis zwischen der Abnahme von85Sr und47Ca wurde von der Tibia und Femur 24 Std nach der Injektion bei Ratten gemessen. Man fand einen signifikanten Unterschied zwischen den Schaften und den Enden, aber nicht zwischen den verschiedenen Knochen. In 7 Wochen alten, verheilten Tibiafrakturen war das Verhältnis genau so wie in einem normalen Tibiaschaft. Das Resultat berechtigte zu der Annahme, daß der Unterschied in dem Mineralisierungsprozeß zwischen corticalen und spongiösen Knochen wahrscheinlich qualitativ und nicht quantitativ ist.
    Notes: Abstract The ratio between the uptake of85Sr and47Ca was measured in the tibiae and femora of rats 24 h after injection of the tracers. There was a significant difference between shafts and ends but not between the different bones; in healed tibial fractures, 7 weeks old, the ratio was identical to that of normal tibial shafts. The findings were interpreted to be related to qualitative rather than quantitative differences in mineralization between cortical and trabecular bone.

    Notes: Summary The ontogenetic development of the hypothalamic nuclei of the rat was examined from the 16th post-coital day to adult age in both sexes, which were determined separately. The following conclusions have been drawn: 1. The hypothalamus is visible already before the 16th day of gestation. 2. The walls of the third ventricle are first composed of primitive cell layers, from which the migration of neurons proceeds on the 15th or 16th day. At this stage the differentiation of the wall of the third ventricle occurs: the germinal-, mantle-and marginal layers appear. 3. The differentiation of the nuclei starts, with some exceptions, before the 19th day of gestation. 4. Nn. suprachiasmaticus, supraopticus, periventricularis anterior, arcuatus and one part of paraventricularis differentiate from the lateral border of the germinal layer. Nn. ventromedialis, dorsomedialis, hypothalamicus anterior, praeoptici, praemamillaris ventralis, praemamillaris dorsalis, mamillaris medialis, mamillaris lateralis and the greater part of paraventricullaris differentiate from the mantle layer. Lateral nuclei, of which only nucleus tuberomamillarius has been described, differentiate from the marginal layer. 5. Some nuclei which belong to the hypothalamo-hypophyseal area have a peak in their differentiation during the critical period when the function of the hypothalamo-hypophyseal axis also starts. 6. It is stated that the development of the nuclei in the rat hypothalamus compared to that of the other mammals is similar only to that of the mouse. It takes place quite slowly before and after birth. 7. Sexual differences are not to be seen in the development of the nuclei.

    Description / Table of Contents: Résumé Aucun effet diabétogène ni hyperglycémiant aigu de l'hydrochlorothiazide (HCT) n'a été décelé par des déterminations de la glycémie à jeun (avant et 2–8 h après l'administration d'HCT) et de la tolérance au glucose effectuées chez des rats normaux et des rats sensibilisés à un effet diabétogène par une pancréatectomie subtotale ou par l'injection d'une dose subdiabétogène d'alloxane, au cours d'un traitement de 6 semaines par des doses de 50–200 mg/kg · jour d'HCT p. o. — De fortes doses (plus de 3 ml/kg) de N-monométhylamide de l'acide acétique (NMMAA), solvant qui fut utilisé dans la préparation d'une solution injectable commerciale d'hydrochlorothiazide, ont par contre exercé des effets diabétogènes très marqués chez le rat. Toute dose léthale de NMMAA a induit chez cet animal un syndrome diabétique, c'est-à-dire une hyperglycémie progressive avec aeido-cétose métabolique, ressemblant au syndrome diabétique aigu induit par l'injection d'une forte dose de sérum anti-insulinique. — Des fractions de doses léthales administrées quotidiennement pendant plusieurs jours ont exercé des effets diabétogènes et léthaux cumulatifs: la substance ou l'un de ses métabolites toxiques apparaît persister longtemps dans l'organisme du rat.-Des doses subléthales de NMMAA ont induit une hyperglycémie réversible d'une durée de quelques jours. Nous avons donc constaté que le diabète produit par le NMMAA était soit transitoire soit léthal. Aucun signe de toxicité ne s'est manifesté au cours d'un traitement de 6–8 mois par des doses inférieures à 0,5 ml/kg · jour. — Chez les rats intoxiqués par une dose léthale de NMMAA, une corrélation positive très significative a été notée entre les valeurs sériques d'insuline immuno-réactive (IRI) et les valeurs de glycémie. Bien qu'ils aient été, à glycémie comparable, supérieurs à ceux constatés dans le sérum de rats normaux après surcharge de glucose, les taux d'IRI se trouvant dans le sérum des rats intoxiqués n'ont pas été en mesure de s'opposer à l'élévation régulière de la glycémie jusqu'à la mort. — L'effet hypoglycémiant d'insuline cristalline porcine est apparu inhibé chez les rats intoxiqués par le NMMAA, comparé à l'effet observé chez des rats normaux ou en diabète alloxanique.-Utilisant l'augmentation de l'oxydation du premier atome de carbone du glucose-l-C14 par le tissu adipeux épididymaire du rat, in vitro, comme index métabolique de l'activité insulino ïde du sérum (ILA), nous n'avons pas constaté, dans le sérum des rats intoxiqués et en état de forte hyperglycémie, des taux d'ILA significativement supérieurs à ceux présents dans le sérum de rats témoins à jeun. Alors que, chez des rats normaux, la fraction de l'ILA sérique supprimable par sérum anti-insulinique (SILA) s'élevait fortement, comme l'IRI, au cours de l'hyperglycémie induite par une surcharge de glucose, cette fraction SILA n'est pas apparue en quantités décelables dans le sérum des animaux intoxiqués. L'IRI sérique de ces animaux n'a donc semble-t-il pas exercé d'effet «insulin-like» sur le tissu adipeux isolé de rat normal.-Les faits observés amènent à la conclusion que les rats intoxiqués par le NMMAA inactivent et l'insuline endogène et l'insuline exogène. Bien qu'ayant perdu son activité métabolique, l'insuline endogène inactivée reste immunologiquement compétente.
    Abstract: Zusammenfassung Große Dosen von Hydrochlorothiazid (50–200 mg/kg/Tag, oral) über einen Zeitraum von 5–6 Wochen riefen weder eine Steigerung der Nüchternblutzuckerspiegel, noch eine Herabsetzung der Glucosetoleranz bei normalen Ratten oder Ratten hervor, deren Empfindlichkeit gegenüber diabetogenen Substanzen durch eine subtotale Pankreatektomie oder durch subdiabetogene Dosen von Alloxan erhöht worden war. 8 Std. nach einer Einzeldosis von 50 mg/kg Hydroehlorothiazid fand sich keine Blutzuckererhöhung. Dagegen hatten große Dosen (über 3.5 ml/kg) des Lösungsmittels N-Monomethylacetamid (NMMAA), das vorübergehend zur Herstellung eines injizierbaren Hydrochlorothiazidpräparates gedient hat, deutliche diabetogene Effekte bei Ratten. Letale Dosen von NMMAA führten immer zu einem diabetischen Syndrom, d.h. fortschreitende Hyperglykämie mit Ketonämie und metabolischer Acidose, das mit dem diabetischen Syndrom nach Verabreichung großer Mengen von Anti-Insulin Serum große Ähnlichkeit aufwies. Bruchteile der letalen Dosis, die wiederholt an aufeinanderfolgenden Tagen gegeben wurden, summierten sich in ihrer diabetogenen und letalen Wirkung : NMMAA oder seine wirksamen Abbauprodukte scheinen längere Zeit im Organismus zu persistieren. Subletale Dosen von NMMAA lösten eine reversible Blutzuckererhöhung für einige Tage aus. Der durch NMMAA hervorgerufene Diabetes war also entweder reversibel oder tödlich. Die Langzeitbehandlung mit Dosen unter 0.8 ml/kg führte auch nach 6–8 Monaten zu keinerlei toxischen Anzeichen. Bei Ratten, die mit letalen Dosen von NMMAA vergiftet wurden, stiegen die Insulin-und Glucose-Spiegel im Blut gleichsinnig an, wobei das Insulin ähnliche Konzentrationen wie bei normalen Ratten erreichte, bei denen orale oder i.v. Glucosezufuhr zu einer entsprechenden Blutzuckersteigerung geführt hatte.-Das während der NMMAA Hyperglykämie sezernierte Insulin bewirkte also keine Blutzuckersenkung. Im Gegensatz zu der glucoseinduzierten Hyperglykämie bei der normalen Ratte stieg die mit Antiinsulin hemmbare insulinähnliche Aktivität während der NMMAAHyperglykämie nicht auf meßbare Werte an, d.h. das IRI der vergifteten Tiere schien auf normales, isoliertes Fettgewebe keinen insulinähnlichen Effekt auszuüben.-Die blutzuckersenkende Wirkung von exogenem SchweineInsulin war bei mit NMMAA vergifteten Ratten niedriger als bei Normaltieren oder Ratten mit Alloxandiabetes.-Diese Befunde veranlassen zu der Schlußfolgerung, daß mit NMMAA vergiftete Ratten exogenes und endogenes Insulin inaktivieren, wobei das inaktivierte endogene Insulin trotz des Verlustes seiner Stoffwechsel-wirkung immunologisch aktiv bleibt.
    Notes: Summary Large doses of hydrochlorothiazide (50-200mg/kg/day p.o.) given for 5 to 6 weeks did not induce any increase in the fasting blood-sugar concentration, nor any decrease of glucose tolerance in normal rats and in rats “sensitized” toward diabetogenic agents by a subtotal pancreatectomy or by a sub-diabetogenic dose of alloxan. No increase in blood sugar was found in the 8 h following a single oral dose of 50 mg/kg of hydrochlorothiazide. —Large doses (〉 3.5 ml/kg) of the solvent N-monomethyl-acetamide (NMMAA), used at one time in the preparation of one brand of hydrochlorothiazide for injection, on the other hand, exerted marked diabetogenic effects in the rat. Lethal doses of NMMAA always induced a diabetic syndrome, i.e. progressive hyperglycaemia with ketonaemia and metabolic acidosis resembling the diabetic syndrome induced by large doses of antiinsulin serum. Fractions of lethal doses given repeatedly on successive days had additive diabetogenic and lethal effects: the drug or its toxic metabolites appeared to persist for a long time in the organism.-Sublethal doses of NMMAA induced a reversible hyperglycaemia of some days' duration. Thus the diabetes induced by NMMAA was either transitory or lethal. Chronic treatment with doses 〈 0.8 ml/kg/day did not induce any signs of toxicity within 6–8 months. In the rats intoxicated with lethal doses of NMMAA, the serum concentration of immunoreactive insulin (IRI) increased simultaneously with the glycaemia, and attained the same levels as in normal rats with similar blood glucose concentrations established by oral or i.v. loads with glucose.-The insulin secreted during NMMAA hyperglycaemia, thus, did not lower the blood sugar. During NMMAA hyperglycaemia, in contrast to glucose-induced hyperglycaemia in normal rats, the fraction of the insulin-like-activity of the serum suppressed by anti-insulin serum (SILA) did not rise to detectable levels : i. e. the IRI of the intoxicated animals did not appear to exert an insulin-like effect on normal isolated adipose tissue.-The blood-sugar-lowering effect of exogenous porcine insulin was depressed in rats intoxicated with NMMAA in comparison with normal animals or animals with alloxan-induced diabetes.-The findings lead to the conclusion that rats intoxicated with NMMAA inactivate exogenous as well as endogenous insulin. Although losing its metabolic activity, the inactivated endogenous insulin remains immunologically competent.

    Description / Table of Contents: Zusammenfassung Die Wirkung von CCl4 auf die Ascorbinsäure-Konzentration in der Leber und die Ascorbinsäure-Ausscheidung mit dem Harn wurde bei erwachsenen Ratten innerhalb der ersten 22 Std nach der Applikation untersucht. In der Frühphase der CCl4-Wirkung kommt es zum Anstieg der Konzentration in der Leber. Der früheste signifikante Anstieg wurde 30 min nach der Applikation beobachtet. Das Maximum wurde dosisabhängig zwischen 90 und 180 min erreicht. Das Anwachsen der Ascorbinsäure-Konzentration in der Leber wird mit einer Aktivitätssteigerung vorhandener an der Ascorbinsäure-Synthese beteiligter Enzyme erklärt. Der Erhöhung der Leber-Ascorbinsäure-Konzentration folgt ein dosisabhängiger Abfall mit einem Minimum 12 Std nach der Applikation, der mit einer gleichfalls dosisabhängigen Erhöhung der Ascorbinsäure-Ausscheidung verbunden ist. Danach kommt es zu einem im zeitlichen Verlauf dosisabhängigen Wiederanstieg der Leber-Ascorbinsäure-Konzentration.
    Notes: Summary The effects of carbon tetrachloride on the concentration of ascorbic acid in the liver and on ascorbic acid excretion in the urine of adult rats were studied over a period of 22 hours after intraperitonal application. During the initial phase of the carbon tetrachloride effect, there results an increase in the concentration of ascorbic acid in the liver. The earliest significant rise was observed 30 minutes after application. The maximal rise came between 90 and 180 minutes, depending on the dose. The increase in the ascorbic acid concentration in the liver is explained by an increase in activity of those enzymes present which participate in ascorbic acid synthesis. The enhancement of hepatic ascorbic acid concentration is followed by a dose-related decrease which reaches a minimum 12 hours after application and which is accompanied by a silmultaneous increase in ascorbic acid excretion. Thereafter in the course of time, there results a renewed dose-dependant increase in the ascorbic acid concentration in liver.

    Notes: Summary The monoamine innervation of the cerebellum of the rat has been studied by both in vivo and in vitro techniques using the histochernical fluorescence method for the demonstration of catecholamines (CA) and certain tryptamines. By way of a pharmacological approach using inter alia protriptyline, which acts mainly by blocking the membrane pump of the noradrenaline (NA) neurons, evidence was obtained that CA nerve terminals in the cerebellum mainly represent NA nerve terminals. These were found to innervate practically all parts of the cerebellar cortex with a patchy innervation pattern and with an innervation of especially the anterior and posterior lobes. The terminals mainly seem to make axodendritic contacts in the molecular and granular layers without any strict localization of the terminal plexus to any special plane of the cerebellar folia. The fibers enter the cerebellum via the inferior cerebellar peduncle and run in the white matter of the cortex cerebelli. Incubation studies with 6-hydroxytryptamine indicate that there exists also a 5-hydroxytryptamine (5-HT) innervation of the cortex cerebelli, although not as pronounced as the NA innervation. The 5-HT nerve terminals are very fine, varicose fibers and innervate mainly the molecular layer, especially of the anterior lobe. The terminals run mainly in the transverse plane of the folium parallel to the surface. Thus, the pattern of innervation of these 5-HT afferents is different from that of the NA nerve terminals. In the uvula, structures which may represent the “rosettes” of the mossy fibers or golgi axon terminals in the granular layer take up and accumulate monoamines after incubation with amine in vitro. The exact nature of these structures remains to be elucidated.The cerebellar nuclei receive a very low to low degree of innervation of NA and 5-HT nerve terminals.

    Notes: Summary In the first experiment rats were trained in black-white discrimination, then injected with 7.5, 10.0, or 15.0 mg/kg of pentylenetetrazol (Metrazol) or saline. Injections were given immediately or 15 min following training. The animals were retested 24 hours later. Improved performance was observed for drug groups, but the extent of improvement was a joint function of the amount of drug and the time at which it was given. Animals given 10 mg/kg showed greater retention when injection was delayed. In a second experiment animals were given spaced trials in a position discrimination task, extending over a period of several days. Following each day's, session, animals were injected with pentylenetetrazol or saline at intervals of 0, 5 or 10 min. A maximum facilitation effect was obtained at the 10-min interval.

    Notes: Summary In 17 male Wistar rats in antidiuresis 10 different nephron segments and arteries are identified with the aid of Lowry's technique, dissected and total-and (Mg++)-adenosintriphosphatase (=ATPase) determined. (Na+K+)-activated ATPase in the distal tubule is four to five times (max. eight times) more active than in the proximal segment. This difference of activity may speak for a high pump mechanism mediated by the way of a (Na+K+)-activated enzyme system in the distal nephron and for a partially passive reabsorption of sodium from the proximal convolution.

    Description / Table of Contents: Zusammenfassung Ausgehend von zahlreichen neurochemischen und neuropathologischen Veröffentlichungen über die 3-Acetylpyridin (3-AP)-Vergiftung der Ratte wurde die bioelektrische Aktivität im Cortex und in subcorticalen Gebieten bei 15 3-AP-vergifteten Ratten studiert. Bei den künstlich beatmeten Tieren wurden mit Hilfe eines stereotaktischen Gerätes rostfreie Stahlelektroden im Hippocampus, Mandelkern, in der Formatio reticularis, im N. septi lateralis und N. caudatus sowie Silberelektroden im Cortex implantiert. Während der akuten 3-AP-Vergiftung konnten weder corticale noch subcorticale bioelektrische Veränderungen beobachtet werden. Vor und während der 3-AP-Vergiftung wurde die Formatio reticularis und der N. caudatus gereizt. Auch hierbei ergaben sich keine Unterschiede. Am Ende der Versuche wurde Cardiazol i.p. injiziert und regelmäßig eine diffuse Krampfaktivität provoziert. Die Befunde werden mit den elektrophysiologischen, neuropathologischen, neurologischen und neurochemischen Beobachtungen anderer Autoren diskutiert mit dem Ergebnis, daß die sog. Rollkrämpfe nach 3-AP-Vergiftung wahrscheinlich in der Hauptsache auf Läsionen der unteren Oliven zurückgeführt werden müssen.
    Notes: Summary Numerous neurochemical and neuropathologioal experimental studies concerning the 3-acetylpyridine (3-AP) poisoning of the rat have been previously published. In this paper the results of experiments on the bioelectrical activity in the cortex and in subcortical areas are presented on 15 rats poisoned with 3-AP. Stainless steel electrodes were implanted stereotactically in the hippocampus, nucleus amygdalae, formatio reticularis, nucleus septi lateralis and nucleus caudatus, and silver electrodes on the cortex of the artificially respirated animals. During the acute 3-AP poisoning neither cortical nor subcortical bioelectrical changes could be observed. The effect of stimulation on the formatio reticularis and the nucleus caudatus was not altered by the 3-AP intoxication. At the end of the experiments metrazol was injected intraperitoneally and in all cases diffuse convulsive discharges could be evoked. The findings are discussed and compared with the observations of other authors. The findings are comparible with the hypothesis that the so called “Rollkrämpfe” after 3-AP poisoning are caused by lesions in the oliva inferior.

Mit dem Proteinaseninhibitor Trasylol konnte sowohl das Ödem vermindert als auch das Überleben verlängert werden. Als günstigste Dosis erwies sich eine Gabe von 25000 KIE/kg kurz vor Tourniquet-Lösen. Danach überlebten von 20 Tieren 13 (65%) die vierfache Zeit von Kontrollen, während sich 7 (35%) völlig erholten. Durch intravenöse Applikation vor Anlegen der Tourniquets konnte zwar die lokale Ödementwicklung gehemmt, die Überlebenszeit aber nicht verbessert werden. Daraus wird auf mindestens zwei Angriffspunkte der Trasylolwirkung geschlossen.
    Notes: Summary In experiments with tourniquet application on both hind legs for 4 h at a temperature of 28°C throughout the experiments the controls succumbed about 2 h after tourniquet release, developing postischemic edema. The development of the edema could be diminished by the protease inhibitor Trasylol as well as the survival time could be prolonged. The optimal effect was seen with the injection of 25000 KIE per kg body-weight i.v. just prior to tourniquet release. In this group of 20 animals 13 (65%) survived four times the time of the controls and 7 (35%) recovered completely. By intravenous injection just prior to the application of the tourniquets the local edema could also be diminished but the survival time could not be prolonged. It is concluded that the Trasylol effect must be brought about by at least two modes of action.

Der Ersatz von Eiprotein-N durch Ammonium-N führt in jedem Mischungsverhältnis zu einer Erhöhung des Stickstoffumsatzes. 2. Methioninzulagen zu Ei/NH4 +-Gemischen bleiben ohne Einfluß auf die Stickstoffumsatzgröße. 3. Eine allgemeine Möglichkeit zur Übertragung von Ergebnissen der Rattenversuche auf den Bedarf des Menschen an Stickstoffträgern besteht für die Bilanzmethode nicht.
    Notes: Summary Metabolism trials under conditions of minimum nitrogen input and nitrogen equilibrium yield the same analytical accuracy (reproducibility of results within a standard deviation of ±1.5%) with men and with rats. In contrast to results with men published formerly, experiments with rats show the following: 1. Replacement of egg protein by non-essential nitrogen (diammoniumcitrate) in any proportions leads to increased nitrogen requirements. 2. Addition of methionine to mixtures of egg protein and ammonia is of no influence on the dimension of nitrogen requirements. 3. A possibility for deducing men's nitrogen requirements from minimum balance results with rats (or vice versa) thus apparently does not exist.

Die intensivste, gegenüber einer Dosis von 35000 KIE//kg KG aber nicht viel stärkere Hemmwirkung, wird mit 50000 KIE/kg KG bei intravenöser Injektion kurz vor Tourniquet-Lösen erreicht. Gaben bis zu 2 1/2 Stdvor und 1 Stdnach Lösen des Tourniquets zeigen ebenfalls noch einen gewissen Effekt. Eine deutliche Wirkung ist auch dann zu erzielen, wenn Trasylol vor Abschnüren der Extremität gegeben wird. Dabei ist der Wirkungsmechanismus nicht eindeutig. In Frage kommt, daß die Entstehung gefäßaktiver Peptide verhindert wird oder daß Trasylol direkt permeabilitätsmindernd auf die Gefäße wirkt.
    Notes: Summary Intravenous injection of Trasylol can stop the development of postischemic edema in a proportional dose response relationship between 10000 and 35000 KIE per kg body weight. The most intensive effect was seen with 50000 KIE per kg injected just prior to the release of the tourniquet, but this effect was not much stronger than with 35000 KIE. An effect could still be demonstrated when Trasylol was given until 2 1/2 h prior to or 1 h after tourniquet release. A marked effect could be obtained even if Trasylol was given prior to tourniquet application. So far the mode of action is not clear. It might be possible, that the development of vasoactive peptides is hindered or that Trasylol has a direct diminishing effect on the permeability of the vessels.

    Notes: A process was devised for the synthesis of tris(hydroxymethyl)acetic acid by means of bio-oxidation of pentaerythritol. A flavobacterium able to grow on pentaerythritol was isolated from the soil. Mutants were obtained that, were deficient, in their pentaerythritol oxidation system. These mutants could not grow on pentaerythritol, but when grown on an assimilable carbon source; they oxidized pentaerythritol to tris(hydroxymethyl)acetic acid. This conversion readily took place in a medium consisting of 20 g pentaerythritol, 10 g yeast extract, and 2 g acetic acid (neutralized) per liter. Since the mutants were unable to metabolize tris(hydroxymethyl)acetic acid, theoretical conversion yields were attainable. When pentaerythritol was added stepwise and the acid formed was neutralized continuously, 95-100% yields were obtained in concentrations of the order of 60 g/l.

    Notes: In order to increase the availability of the cell bound protein in Scenedesmus algae, mechanical, enzymatic, and chemical methods of degrading the cell wall structure were investigated.Mechanical treatment involved the use of a ball-mill. The algae suspension together with glass beads was milled in a water-cooled chamber equipped with rotating disks. The enzyme tested was a cellulolytic enzyme (Meicelase) and the chemical employed was hydrogen peroxide.In the ball-mill experiments a complete disintegration was achieved ina disintegrator, working with batches. Trails wwere also performed with a continuous disintegrator and the depedence of disintegration on bead size and flow rate was studied. The disintegration determined by microscropic cell count was compared to the increase of the pepsin digestibility.The meicelase treatment caused a slight increase of the pepsin digestibility, as measured after 3 hr pepsin incubation. No increase of the pepsin disgestibility could be detected with hydrogen peroxide treatment.After the ball-mill disintegration 95% of contaminating bacteria were killed and yields of extractable proteins were higher. The capacity of availble continuous ball-mills is such that they could be used on a pilot-plant scale and the energy cost of disintegration would be of the same magnitude as that of separation.

    Notes: The results achieved by the cultivation of the yeast. Candida lipolytica on gas oil are referred. By using a distillation fraction of gas oil distilling between 180-400°C, containing 10-20% of n-alkanes, the optimal condition for biomass production and deparaffination were estimated for various dilution rates and various amounts of gas oil in the medium. The main factor, which influences the yield coefficient by hydrocarbon fermentation is the polyauxie of the hydrocarbon substrate. The penetration of dispersed hydrocarbons into the yeast cell is demonstrated on electron micrographs and the velocity and reversibility of this process is estimated by using tritium-traced hexadecane.

    Notes: Continuous culture in a cascade of vessels with the addition of supplemental nutrients to any stage permits adjustment of the physiological state of the culture in each stage to best achieve a desired performance goal. The yeast Saccharomyces cerevisiae in two-stage continuous cultivation was selected as a model system. With conditions in the first stage held constant- at a selected glucose concentration in the feed stream, dilution rate for the second stage was varied. Cell numbers, dry weight, glucose concentration, respiration coefficient, and titers of several enzymes were determined. The seed rate was defined as the ratio of glucose concentration in the feeds to stage 1 and to stage 2. At low seed rates, the calculated specific growth rate in the second stage was proportional to dilution rate. At higher seed rates, the specific growth rate based on dry weight behaved differently from that based on cell numbers, and the dependence on dilution rate was not linear.

    Notes: A pilot-scale process for the isolation of an aliphatic, amidase from Pseudomonas aeruginosa has been developed. A constitutive, partially irrepressible mutant was employed to give a high initial enzyme concentration. An existing laboratory isolation procedure has been scaled up and modified particularly by substitution of polyethylene glycol for ammonium sulfate precipitation as the first stage in the conversion of the fractionation to continuous operation. Full recovery of activity was achieved with the modification. The recovery of enzyme from a subsequent chromatographic stage was 85% and the maximum overall purification was 28-fold.

    Notes: It has been shown that alkylbenzenes which do not support growth of Pseudomonas aeruginosa exert a stimulatory or inhibitory effect on the utilization of n-alkanes by this organism. The effect is a function or the number of methyl substitutions to the benzene ring and the concentration of aromatic hydrocarbon. The alkylbenzenes have been shown to exert the stimulatory and inhibitory effect, on all phases of growth. The data suggests different mechanisms involved in the inhibition by benzene and p-xylene but a similar mechanism by all of the stimulatory compounds. Warburg data demonstrates changes in the rate of oxygen uptake in the presence of trimethylbenzene at different stages of the fermentation. The release of water soluble material in the presence of trimethylbenzene suggests a permeability effect on the cells.

    Notes: An oxygen probe is developed, using the principle of the silver-lead galvanic cell, covered with a plastic membrane. It is steam sterilizable while filled with water; after sterilization, water is easily exchanged for the electrolyte, potassium bicarbonate. Flat plastic membranes up to a thickness of about 0.1 mm can be applied. The membrane is secured and stretched in a special way, which guarantees a leakproof seal. The outer shape of the probe has a tulip-form and consists of polished stainless steel, except for the electrode face. The electrode is suitable for laboratory and industrial use and its components are completely standardized.Almost linear response for O2-partial pressures up to atmospheric is obtained with 0.001 in. and 0.002 in. FEP-Teflon membranes. 90% response time is of the order of 10-15 sec for the 0.001 in. membrane, Residual current is almost negligible (〈 1 μA).

    Notes: Microorganisms were continuously cultivated in multistage column consisting of ten perforated plate sections to which medium and air were supplied concurrently from the bottom.At steady state the cell concentration in the various stages was gradationally differentiated from the bottom to the top in the direction of medium flow. RNA content per unit cell concentration at each sage was determined. The cells in the lower stages were higher in RNA content than those from the upper stages. Wash out was observed to occur in the column at dilution rates which do not result in wash out in a single stage chemostat system.A study of the flow characteristics revealed that the overall performance of the plate column was equivalent to that of a multistage system, when hole diameter and hole area to column cross sectional area ratio were properly selected. This was true even in highly aerated conditions. These results indicated that the perforated plates in the column hindred intermixing through the plates, and that each stage functioned as an independent stirred vessel. Industrial and research application of this type fermentor was discussed.

    Notes: Current methodology now makes it possible to establish in culture a variety of mammalian cells which perform organ-specific functions during serial propagation for periods of months or years. This report describes the results of experiments with animal and human cells that produce growth hormone, adrenocortical steroid hormones, thyrocalcitonin, and parathyroid hormone. Within the next decade it should be possible to use cell culture methods for manufacturing purposes to produce hormones and other valuable cellular products which are difficult to obtain in other ways. At first it may be necessary to use neoplastic cells for this purpose; but evidence is accumulating to suggest that it may eventually be possible to establish in culture normal, functional animal and human cells.

    Notes: Pure bacterial cultures can be flocculated by a variety of chemical flocculants. Flocculation of bacteria will assist in their recovery, especially where the cells themselves are of interest, as in microbial protein production. Studies with several genera of bacteria indicate that the mechanism of flocculation is highly complex. Such interacting variables as temperature, ionic environment, physiological age, flocculant, bacterial genus, and surface shear have been observed. Jar test experiments with washed cells indicate that many of the variables are related to the release by the cell of proteins, nucleic acids, or polysaccharides. When released, these polymers may increase the required dosage of flocculant for recovery as in the case of E. coli, or the dosage may decrease as it does for Lactobacillus.

    Notes: A theoretical study was made on the dynamic behavior of a single-vessel continuous fermentation subject to a growth inhibition at, high concentration of the rate limiting substrate. Phase plane plotting and stability analysis showed three steady states to exist; namely, a “washout”; state, a nontrivial stable state, and an unstable state. Whether the system attains a nontrivial steady state or is to be washed-out depends upon the initial values of the cell and/or substrate concentration(s). Since this property is a characteristic feature of the present system, an experimental procedure was suggested to insure a stable operation in practice.

The purification factor was 100-200 × and the overall yield was about 115%.     Notes: Dissolved oxygen and glucose concentrations have been independently maintained at various concentrations for extended periods during growth of Candida utilis in continuous culture. Simultaneous observations of cytochrome concentration, growth rate, rates of uptake of oxygen and glucose and rates of production of CO2, ethanol and acid have been made during steady states at various levels of oxygen and glucose. There is an inverse relationship between dissolved oxygen and cytochrome and between glucose cocentration and cytochrome.Studies of the transient state following a step change from high to low dissolved oxygen show that there is a lag of about 10 hr during which there is no change in the above parameters. This is followed by rapid oscillatory changes in cytochrome content and a change to a more fermentative metabolism.

    Notes: Total lipids were extracted from the cells of Candida utilis grown at a constant population density in continuous culture. At different steady states, the environment was controlled with respect to dissolved oxygen and glucose concentrations, pH and temperature. Gas liquid chromatography was used to follow quantitative and qualitative changes in the fatty acid composition of the cells. Increasing glucose concentration resulted in higher lipid content; high oxygen concentrations increased the level of polyunsaturated fatty acids. The most significant changes in fatty acid composition took place when both glucose and oxygen concentrations were high, and under these conditions the amount, of linolenic acid was at its highest value.

    Notes: On increasing aeration rate in chemostat cultures of E. coli the redox potential (Eh) of the culture changes from -l50 mv to +250 mv. During this period only a slight increase in measured dissolved oxygen concentration is observed.It is suggested that Eh be used as a control variable for aeration in the oxygen limited growth condition, whilst dissolved oxygen concentration be used as control variable in the presence of excess oxygen.Change in cytochrome b1, three tricarboxylic acid cycle enzymes and hydrogenase can be related to culture Eh. These changes are discussed.

    Notes: In beer fermentation, yeast cells are kept in suspension, despite their higher density, by natural agitation created by ascending CO2 bubbles. Yeast cells are unable to nucleate bubbles but instead release CO2 in a soluble form in such a way that the medium tends to become supersaturated.A higher concentration of yeast cells and the presence of solid particles cause the formation of bubbles at the bottom of the fermenter and practically only there. The rising bubbles grow and accelerate by sweeping the CO2 formed throughout the fermenter by the suspended yeast cells, thereby creating a fluid regime. A mathematical expression relating the bubble agitation power to the fermentation parameters was obtained and used to design more efficient fermenter shapes.

    Notes: Mathematical models which can be used to describe batch frowth in fermentations with two liquid phases are developed for systems in which the growth limiting substrate is the dispersed liquid phase. Three special cases are considered assuming pure substrate in the dispersed phase and a decreasing interfacial area due to substrate consumption. In the first, it is assumed that all growth occurs at the surface of the dispersed phase. In the second and third growth occurs at the interface and in the continuous phase. The second case assumes substrate equilibrium between the two phases while the third assumes substrate consumption in the continuous phase is limited by rate of substrate transport to that phase. Since the amount of growth at the interface and substrate transport to the continuous phase depend on the interfacial area, two limiting cases for the decrease of interfacial area with substrate consumption are also considered in this investigation. The first and third models are compared with available experimental data.

Two types of dispersed systems are considered in this investigation. In the first, type, it is assumed that the composition of the dispersed phase is such that, increased substrate utilization results in a decreased substrate concentration with no change in the interfacial area. In the second type of system, the dispersed phase is assumed to be pure substrate; therefore, the substrate concentration in the dispersed phase remains constant but the interfacial area is affected by changes in dilution rate. Three special cases are examined for each type of system in order to examine the effect of the interfacial area, the phase equilibrium constant, and the mass transfer coefficient on system performance. Comparison of two of the models with available experimental data shows fair agreement, between model and data.

    Notes: A microprobe electrode was used to determine dissolved oxygen concentrations near the surface and within a bacterial slime mass supplied with a continuous flow of nutrient solution. With dilute medium, the oxygen profile became level at high concentrations within the film, indicating substrate-limited respiration. More concentrated medium caused the profile to fall to low oxygen concentrations characteristic of oxygen-limited respiration. Oxygen responses to sudden changes in concentration of nutrient medium were measured. Estimates of microbial respiration rate and of diffusivity of oxygen were based on well-known diffusion equations.

    Notes: Three different established strains of mammalian cells were grown in chemically defined medium in large cultures. The degree of proliferation of cells of an established strain from human skin in large stationary cultures was significantly greater in the presence of methylcellulose (medium NCTC 135M) than in its absence (medium NCTC 135). The relatively fragile cells of a derivative of monkey kidney LLCMK2 strain were carried in large stationary cultures through 11 transfer generations during 152 days. The presence of methylcellulose was associated with higher cell population levels, proliferation rates, and cell viability. Cells of this strain utilized glucose at an extremely high rate; during two representative periods the rate averaged 1.2 mg/106 cells/day in cultures on medium 135M and 1.9 mg in medium 135.In a 53-day experiment with mouse fibroblast 2071-L cells, the cells in suspension culture during the first 28 days went through the normal lag, logarithmic plateau, and initial decline phases in medium 135M, and then were transferred to large stationary cultures, where they proliferated for 7 days at uniformly high rates in both medium 135 and medium 135M. It appeared that cells of strain 2071-L in such stationary cultures had no need for Methocel as a protective agent. Glucose utilization rates while these cells were carried in large stationary cultures averaged 2-4 times the rates while they were in suspension cultures: about 0.8 and 0.2 mg/106 cells/day, repectively.

    Notes: A Micrococcus cerificans strain was grown on simple media with n-hexadecane or gas oil as sole carbon sources. Samples of cellular material recovered from hexadecane or gas oil fermentations do not appear to differ significantly in their composition. The protein content varied from 68 to 75%. With the exception of sulfur amino acids the amino acid distribution compares favorably with the FAO standard reference protein.The biological value of cell protein recoveered from hexadecane fermentations was 67 (cascin, 70). In the case of gas oil grown cells, the cell material recovered had to be completely purified in order to improve its protein quality. After fully extraction of undersirable fraction with petroleum ether in a Soxhlet apparatus the biological value observed was 63.

    Notes: A temperature-synchronized, semi-continuous culture and monitoring system is described, with results from use of the system for autotrophically growing Euglena. Outflow from the culture vessel consists of measured samples taken automatically at 2-hour intervals and fixed for later counting. Inflow is by siphon feed, which restores the culture level after each sampling. The interpretation of growth curves obtained from such cultures is discussed from the viewpoint, of division synchrony and cell cycle studies, and some general comparisons are made between batch and continuous cultures for such studies.

    Notes: A fermentation system was designed and constructed to study the growth characteristics of microorganisms at low and high cell concentrations. The technique used to develop high cell densities utilized a rotating microfiltration unit to permit the removal of cell-free product from the fermenter. The fermenter volume and the filter were contained in a single unit composed of a series of concentric cylinders. Annuli contained the fermenter volume while the second outermost cylinder supported a microfiltration membrane. Feed to the system was pumped at constant rates, and the internal pressure built up to a value, which would effect the required filtration rate. The system was operated batchwise and continuously with and without filtration.The anaerobie growth characteristics of Streptococcus faccalus were determined at 37°C and pH 7.0 for batch, continuous, and continuous with filtration modes of operation. The growth characteristics were unchanged when the cell density was increased. Changes in cell yield peer model of glucose consumed were clearly illustrated during thee course of single run by operating the fermenter in the unsteady state with filtration. No consumption of glucose for developed was 40% packed cell volume, a value 45 times larger than could be grown in simple batch culture.

    Notes: The design of a continuous column fermentor with a multiple staging effect is described. The column is divided into four compartments by horizontal perforated plates and is provided with a central agitator shaft driving an impeller in each compartment. A tube at the center of each plate forms a liquid seal around the shaft and also acts as a “downcomer.”The fermentor is normally operated with counter-current flow of gas and medium. Fresh medium is added to the top stage and product is withdrawn from the bottom.The effect of plate and agitator design on fermentor performance was studied in terms of factor such as oxygen transfer rate, gas holdup, and interstage mixing. By proper choice of the design parameters, the fermentor was made to approximate a perfect four-stage cascade in terms of reactor performance.Preliminary experiments were performed with air-water systems, but a more realistic picture of fermentor performance was obtained in experience involving propagation of Escherichia coli. Data for business and substrate concentrations in each stage confirmed the staging effect of the apparatus. The fermentor operated in a stable manner for periods of more than two weeks.

    Notes: This paper illustrates the type of conclusions which may be reached by the use of an electrode for measuring oxygen tension in fermentation broths. The critical oxygen level, based on oxygen uptake, and the minimum oxygen tension necessary for maximum antibiotic synthesis for two commercial antibiotic fermentations, capreomycin and cephalosporin C, were studied. Capreomycin yields were depressed at an oxygen tension which was slightly below the critical oxygen level. Cephalosporin C synthesis was depressed at an oxygen tension higher than the critical oxygen level.

    Notes: The oxygen transfer coefficient has been investigated in S. noursci and S. lavendulae fermentation broths obtained from fermentors of different operating volumes (61., 30001., 20,0001.). Fermentors had KLas values ranging from 1.0 to 17.0 min-1, calculated from sulphite oxidation rates. The dynamic measurement of the volumetric oxygen transfer coefficient. (KLa) has been performed in the different fermenting systems. As the fermentation progressed, especially in the first stages, KLa values have decreased in both fermentations and in each system of fermentors. In order to characterise the whole fermenting system an average KLas was calculated from the obtained KLa values. The average KLa grew with increasing KLas values and ranged from 0.03 to 3.72 min-l. Some factors possibly having an influence on the, change of KLa have been studied.The oxygen transfer coefficients of the broths have been measured in falling films and ranged from 0.05 to 0.4 cm min-1. The flow conditions have been characterized by Reynolds numbers of broths varying between 1.0 and 60.0. The average thickness of the falling films have been measured and plotted against Reynolds number. The Re+ which is the breaking-point of the plot increased as the fermentation proceeded. In the region of Re+ the values of the oxygen transfer coefficient increased rapidly. An approximate correlation could be established between the Re+ and the physical properties of fermentation broth.

    Notes: The presence of microorganisms have been shown to increase by over 40% the mass transfer rates from small oxygen bubbles at low Reynolds number flow. This increase was found to be due only to the microbe cells as inert particles disrupting the quasi-static liquid surface film surrounding the gas bubble and thus decreasing the gas-liquid interfacial resistance. The observed increase in oxygen mass transfer rates was not dependent on cell viability, no effect was noted due to cell-liquid interfacial resistance, nor was the phenomenon due to altering the physical properties of the liquid during cell propagation. These results were obtained in a unique plexiglass apparatus designed for observing under a microscope a small (0.4 mm dia.) stationary oxygen bubble collapsing into a flowing fluid. The oxygen bubble was injected by a small hypodermic needle and the fluid was suspensions of the yeast Candida intermedia, the bacterium Pseudomonas ovalis, 0.3μ alumina, as well as base points of cell free broth and pure water. Several well-known chemical inhibitors of oxidative phosphorylation were used to limit cell oxygen uptake. Calculations of oxygen mass transfer rates were compared with the semi-empirical model of Frössling, the circulating sphere model of Levich, and the rigid sphere concentration boundary layer model of Fried-lander, the latter two showing strong Reynolds number dependence that may be due to radial fluid motion.

    Notes: The physiology of Aspergillus nidulans strain 224 has been studied under conditions of batch- and glucose-limited chemostat-culture and the effect of different steady state growth rates and dissolved oxygen tensions (DOT) examined. Measurements of the specific activities of selected glucose enzymes, the extent of oxygen uptake inhibition by glycolytic inhibitors, and radiorespirometric analyses were made in order to follow the variations in glucose catabolism, which occurred under these conditions. Greatly increased activity of the hexosemonophosphate (HMP) pathway was found during: (i) exponential growth of batch cultures; (ii) at near maximum specific growth rates (μ = 0.072 hr-1) (DOT = 156 mm Hg); and (iii) at low DOT levels (〈30 mm Hg) (μ = 0.050 hr-1) in chemostat cultures. These changes in glucose eatabolism have been discussed in terms of the biosynthetic demands of the fungus under the influence of changing growth pressures. Preliminary studies also have been made of transition state behavior following stepwise alteration of the DOT. A new steady state was established after 4-5 culture doublings during which period an “overshoot” in HMP pathway activity occurred; these kinetics are indicative of a derepression of certain glucose enzymes. Low molecular weight phenols are synthesized during the exponential phase in batch cultures and these are further metabliized to a major secondary metabolite, melanin, at the onset of stationary phase conditions. The kinetics of tyrosinase production in steady state chemostats differs from those that might be predicted for an enzyme associated solely with secondary metabolism. A primary physiological role for this oxidase in Aspergillus nidulans has been postulated.

    Notes: Streptomyces peucetius var. caesius, obtained from S. peucetius, the daunomycin producing microorganism, by mutagenic treatment, differs from the parent culture by the color of the vegetative and aerial mycelia and by its antibiotic, producing ability. S. peucetius var. caesius accumulates adriamycin in submerged and aerated culture on a medium containing glucose, brewer's yeast, and inorganic, salts both in shake flasks and in stirred fementers. Isolation of the product is performed by solvent extraction, chromatography on buffered cellulose columns, and crystallization as the hydrochloride. The new antitumor agent, adriamycin, is the 14-hydroxv derivative of daunomyein.

    Notes: We have studied the influence of strain of organism, temperature, and medium on the production of the antileukemic intracellular enzyme L-asparaginase by E. coli B grown in shaken flasks. Five strains of E. coli B exhibited wide differences in their capacities to synthesize the EC-2 form of L-asparaginase active against leukemia. For the most productive strain, when grown in a casein hydrolysate medium, maximal production of L-asparaginase occurred at 25°C. At this temperature, the organism required glycerol, glucose, or other mono-saccharides to synthesize L-asparaginase. Synthesis was stimulated when glycerol was used in place of glucose, but not in its presence. The effect of glycerol on L-asparaginase synthesis was most evident when the cells were grown at 37°C, rather than at 25°C. With 0.25% glucose, cells had a specific activity of 409 I.U./g; with glycerol cells had a specific activity of 553 I.U./g. At 25°C, both cell and L-asparaginase synthesis were increased by the use of 0.25% glycerol resulting in only a slight increase in specific activity of the cells. The addition of zinc, copper, manganese, iron, L-asparagine, L-glutamine, or L-aspartic acid had no effect on L-asparaginase synthesis in the casein hydrolysate medium. L-aspartic acid (10-2 M) enhanced L-asparaginase synthesis in a synthetic medium that lacked these metals or L-asparagine, L-glutamine, or L-aspartic acid; cells grown under these conditions had a specific activity of 90 I.U./g.In the casein hydrolysate medium, cell morphology was correlated with temperature of incubation.

    Notes: Some results of our studios on transformation of steroids by mixed culture fermentation are presented in this paper. Arthrobacter simplex was paired in turn with each of the following: Streptomyces roseochromogenes, Curvularia lunata, Absidia coerulea, and Aspergillus ochraceus. The steroid substrates examined for multiple transformation were 16α-hydroxy-cortexolone, 16α-hydroxy-cortexolone 16,17-acetonide, 9α-fluorohydrocortisone, 9α-fluorohydrocortisone 21-acetate, and 9α-fluorohydrocortisone 21-hemisuccinate. The effects of media, steroid substrate, and microbial interaction in a mixed culture on the induction and repression of steroid transforming enzymes were unique to each case studied. The reaction mechanism of the multiple steroid transformation was also found to vary from one mixed culture system to another. Two different reaction mechanisms were observed, namely, consecutive and parallel. In the former, one of the two enzymatic reactions always preceded the other, while in the latter, two different enzyme reactions occurred simultaneously, thereby giving rise to two different intermediates. Multiple transformation of steroids by a single step mixed culture fermentation has potential economic advantages.

    Notes: This investigation used the glucose oxidase system to simulate oxygen transfer rate in fermentation broths. It was demonstrated that the fungal preparation contained sufficient lactonase activity so that D-glucono-δ-lactone did not accumulate and that the rate of production of gluconic acid was proportional to the oxygen uptake rate. Enzyme concentrations of 1.5-2 g/1 were found adequate to determine oxygen absorption rates in shake flasks while maintaining the dissolved oxygen concentration of low levels. The apparent Michaelis constant for oxygen, Km(O2), was found to be 27% saturation with air; this value along with experimentally determined uptake rates could be used to calculate dissolved oxygen concentration in lieu of using a dissolved oxygen probe. Enzyme concentrations of 5 g/l were sufficient to give linear acid production and low dissolved oxygen concentrations in a bench-scale fermenter with no foaming or enzyme deactivation. The method is considered more valid and easier to employ than previously utilized techniques such as sulfite oxidation. Extension of the system to evaluating aeration effectiveness and scaleup of fermentation equipment is discussed.

    Notes: Growth kinetics of heterogeneous populations of sewage origin were studied in completely mixed reactors of the once-through type at a high concentration of incoming substrate, 3000 mg/l glucose, and in systems employing cell feedback or sludge recycle at an incoming substrate concentration of 1000 mg/1 glucose. The recycle flow rate employed was 25% of the incoming feed flow, and the concentration of cells in the recycle was maintained as closely as possible at 150% of the cell concentration in the reactor. Studies were made at various dilution rates. Throughout these studies, batch experiments using cells grown at the various dilution rates were made to determine ks and μm values. As in previous studios using heterogeneous populations, the relationship between specific growth rates μ and substrate concentration S was represented better by the Monod equation than by any other which was tested. The growth “constants” μm, ks, and Y were found to fall in the same general range as those determined in previous studies in once-through systems operated at 1000 mg/l glucose. It was observed that cell recycle, even at the relatively low concentration factor employed in these studies, greatly enhanced the flocculating and settling characteristics of the cells.

    Notes: A mathematical model for the prediction of product formation in batch processes is presented. The suggested procedure allows production of a prognose at any given moment of a run concerning the product concentration at any future moment of the same experiment. The series of subsequent prognoses thus produce a trend, and it is assumed that an analysis of the trend may provide information, which is useful in the evaluation of different decision alternatives in pilot plant batch experiments.

    Notes: A high intensity light system (HILIS) was designed and constructed to define the environmental parameters affecting production of algae. The HILIS incorporates the basic concepts of an aerobic fermenter for heterotrophic cells with high intensity illumination for photosynthetic studies. Of nine parameters considered, temperature and light intensity studies using Chlorella 71105 have been completed. Total illumination was varied from 25,000 to 300,000 lumens (30 times intensity of sunlight as measured at earth's surface) in 7.7-1, culture. The effect of illumination upon growth was measured as cell concentration and total daily algal production when operating the HILIS as a continuous system at a dilution rate of 0.91 per day.Growth may be expressed as a long function of illumination. A maximum algal concentration of 25.5g/l., dry weight basis, was attained at 300,000 lumens.

    Notes: A description is given of the design and operation of high-power magnetic drives developed to enable shaft seals and glands to be dispensed within deepculture vessels, in tissue homogenizers, and in mixing and filling processes where sterility is essential. The drives operate at speeds of 300 to 2000 rpm in volumes of 300 1. to 10 ml with clearances up to 16 mm between the pole faces of the magnets.Two types of drive are described, one in which the driving and driven magnets form an integral unit on the lid of a vessel: such vessels are used for transporting material. To intiate stirring, it is only necessary to connect a motor directly, or through a cable-drive, to the magnetic-drive assembly. In the other type of unit the driving magnet is attached permanently to the driving motor. Locating pins on the base of the motor and corresponding sockets on the lid of the vessel ensure that when the motor is in position, the driving and driven magnets are located correctly in relation to one another.The design of these drives is based on the use of multipole, ceramic magnets. The advantages of their use in such units, compared with metal magnets, are discussed. Earlier magnetic drives are also discussed and explanations offered for the difficulties formerly met in scaling up.

    Notes: Experiments were performed on a cellulose acetate ultrafiltration membrane (HF-200, ABCOR Inc., Cambridge, Mass.) to test its efficacy in concentrating and purifying a crude enzyme (trypsin) preparation. Studies were also made to determine the influence of inorganic salts, pressure, and temperature on the rate of ultrafiltration for this membrane. The results showed reductions in the rates will be encountered due to the presence of inorganic salts. However, the reduced rates were still sufficiently high to make this method extremely attractive. Operating at filtration pressures above 75 psi at, 20 to 30°C for this membrane does not show any beneficial effect in terms of ultrafiltration rates. However, at 10°C there were continual increases in the filtration rates up to 100 psi. Concentration and purification studies with trypsin yielded a concentration factor of 8.35 and a purification factor 2.35. It was shown concretely that the purification of the enzyme was due to the passage of low molecular weight proteins (below 20,000) through the membrane. Enzyme activity slightly greater than 90% was obtained: 70% was found in the concentrate and 20% in the filtrate. It is concluded that membrane ultrafiltration is an ideal simple, rapid, and economical method for the recovery of biological active substances.

    Notes: A procedure for measuring the rate of heat production from a fermentation has been developed. The method is based on measuring the rate of temperature rise of the fermentation broth resulting from metabolism, when the temperature controller is turned off. The heat accumulation measured in this manner is then corrected for heat losses and gains. A sensitive thermistor is used to follow the temperature rise with time. This procedure is shown to be as accurate as previous methods but much simpler in execution. Using this technique, the rate of heat production during metabolism was found to correlate with the rate of oxygen consumption. Experiments were performed using bacteria (E. coli and B. subtilis), a yeast (C. intermedia), and a mold (A. niger). The substrates investigated included glucose, molasses, and soy bean meal. The proportionality constant for the correlation is independent of the growth rate, slightly dependent on the substrate, and possibly dependent On the type of organism growth. This correlation has considerable potential for predicting heat evolution from the metabolism of microorganisms on simple or complex substrates and providing quantitative parameters necessary for heat removal calculations.

    Notes: A multistage tower laboratory fermentor has been constructed consisting of eight compartments separated by sieve plates. Flow of substrate and air is concurrent from the bottom to the top of the column. It, was hoped that this system could be used to reproduce, simultaneously on a continuous basis, eight distinct phases of a batch growth curve. It was believed that the extent of batch curve simulation would depend upon the character of hydraulic mean residence time of broth in the column and in the individual compartments. The expected relationship did not occur. Rather it was found that growth in the column involved residence time characteristics not only for the fluid but also for the microorganisms, and for the growth limiting substrate. Depending upon the column operation, these could be distinct and different.The purpose of this investigation was to study the residence time distribution (RTD) of the continous (fluid) and dispersed (microorganisms) phases for model systems as well as for a yeast fermentation. Various degrees of flow nonideality, i.e., fluid blackflow and dispersed phase sedimentation, were noticed. The former seems to be due to interaction of the concurrent gas and liquid flow; it is particularly dependent upon void area of the sieve plate holes. Sedimentation is probably a function of plate design as well as cell size and density. It wa concluded that for a particular plate design the gas hold-up wass controlled by superficial air velocity and was the main parameter governing the differences between dispersed and continous phase(Rt1). This conclusion was supported by a computeraided styudy utilizing a mathematical model of fluid flow to fit the growth kinetics and cell distribution observed experimentally throughout the fermentor.Some advantages of foam control in the tower fermentor by surface active compounds are mentioned. Also, suggestions are made for carrying out fermentations that have two liquid phases, such as a hydrocarbon fermentation. The possibility of closely approximating plug-flow conditions in the multistage tower fermentor, a necessary condition for batch growth simulation, is discussed from a practical point of view.

    Notes: A mixed culture of methanol oxidizing bacteria has been cultivated on simple inorganic salts medium supplemented with methanol. Optimal growth occurred at 31°C, pH 6.0-6.3, and a methanol concentration between 1 and 2 ml/1, of medium. The maximum yield was 4.5 g dw/I and the mean generation time 3.2 hr.It was estimated that 41% of methanol carbon was converted into cell-carbon, and that 73% of the inorganic nitrogen was converted to organic nitrogen.

    Notes: The current knowledge concerning the biosynthesis of chloramphenicol is discussed. Cultures of Streptomyces sp. 3022a fed 14C-shikimie acid incorporated the label to the same extent into phenylalanine, tyrosine, and chloramphenicol. Of possible precursors of the phenylpropanoid nucleus of this antibiotic only p-aminophenylalanine and DL-threo-p-amino phenylserine specifically labeled chloramphenicol. On the basis of these results a pathway for the biosynthesis of chloramphenicol is presented. The lack of specific incorporation of 15N-nitrogen from a competitive feeding experiment in which both l5N-nitrate and 14N-DL-serine were fed to growing cultures suggests that both the amido- and the nitro-nitrogen atom present in this antibiotic are derived from a common pool. Studies on the enzyme, DAHP synthetase, show that in streptomyces sp. 3022a it is not subject to feed back inhibition by either phenylalanine, tyrosine, or chloramphenicol.

    Notes: 9α-Hydroxy-19-norandrostenedionc (9α-hydroxy-Δ4-estrene 3, 17-dione) (IV) was prepared by fermentation of 19-norandroslenedione with Corynespora melanis or Norcardia restriclus. When incubated with a growing culture of Arthrobacter simplex or its acetone-dried cells, IV was converted to 9α-hydroxyestronc (VII) and 9-keto-9, 10-secoestrone (VI). 9α-Hydroxyestrone undergoes spontaneous as well as enzymic dehydration to form Δ9(11)-estrone (IX). Both VI and IX have been isolated and identified as such while VII was isolated as its 3-acetate.

    Notes: A protease, excreted by a sporogeneous strain of B. megaterium, growing exponentially in a minimum glucose ammonium medium, was isolated. It is a neutral endopeptidase, stabilized by Ca++, inhibited by o-phenanthroline, but not by di-isopropylfluorophosphate. The specificity, studied on insulin B-chain, glucagon, cytochrome c, and dipeptides substrates, indicated the need for a dipeptide backbone with both substituted amino and carboxyl groups. A requirement was observed for a nonpolar lateral chain in the amino acid whose amino group was involved in the peptide bond (Leu, Phe, Ala, He, Val). Rates of hydrolysis varied also with the amino acid whose carboxyl group was involved (e.g., His 〉 Ser 〉 Ala 〉 Gly). In complex medium, supplemented with Yeast Extract, the biosynthesis of the protease was repressed during growth, but the same enzyme was excreted during sporulation. The repression was apparently of the same nature as that controlling sporulation during and after growth (e.g., repression by a mixture of amino acids or high concentration of glucose). An asporogeneous mutant showed a normal product ion of protease under all conditions, and a low intracellular protease turnover after growth. A mutant unable to produce protease showed a normal sporulation and a high protein turnover. This protease, here termed megapeptidase, seems to be a typical growth enzyme, not related to either the sporulation process or to the protein turnover after growth.

    Notes: The stability of foam formed during fermentation is decisively affect ed by the nature of the nutrient media used. In froth-flotation models, (a) the foam formation time, characteristic of the tendency to foam, and (b) foam subsistence time, characteristic of the stability of foams formed, have been studied. With the utilization of these two parameters, the stability of foam from aqueous solution of several surface active components of nutrient media has been noted as a function of concentrations. Further, but, without attempting completeness, the viscosity enhancing effect of carbohydrate components, and the effect of the subsistence time of their foam, upon the stability of foam have been studied together with the correlation between “standing” time after sterilization and tendency to foam. Taking soy-bean meal as a model, the stability of foam films in function of pi I, at constant concentration, has been studied. It seems that though a proper control of the factors mentioned, nutrient media with a low tendency of foaming can be formulated.

    Notes: Cultivation of Norcardia sp., Mycobacterium phlei, and Candida lipolytica in inorganic salt solution containing n-alkanes C10-C20 as solo carbon and energy source was investigated. Generation times of 0.5-7.0 hr were typical during the exponential growth phase. The final cell concentrations (dry weight) were usually 9-26 g/l with n-alkane mixtures ranging from n-decane through n-eicosane. A linear dependence was found between the production of cell mass and the consumption of n-alkanes. The rest concentration of n-alkanes in the cell mass is in all experiments smaller than 0.5% (w/w). Cell yields were Ysub 60-142% and for Ye 50-97% based on n-alkane utilization. In one case, with the Nocardia NBZ 23, the substrate specifity on hydrocarbons and on a n-alkane mixture C10-C20 was studied. The cell mass recovered from the fermentations contained 47.8-57.7% carbon, 5.6-9.95% nitrogen, 7.2-9.4% hydrogen, 35-62% crude protein, and 6-36% lipid. Cellular protein and lipid synthesized by an organism is influenced by the type of nitrogen source. The amino acid, glucosamine, muramic acid, 2,6-diaminopimelinic acid, and fatty acid distribution in organisms grown on n-alkanes compared with a corresponding fermentation on glucose as sole carbon source were also estimated.

    Notes: Mathematical models which can be used to describe batch growth in fermentations with two liquid phases are developed for systems in which the growth limiting substrate is dissolved the dispersed liquid phase. In the models, the possibilities of growth occurring at the surface of the dispersed phase and in the continuous phase are considered. It is assumed that the composition of the dispersed phase is such that substrate utilization from it causes little of no change, in interfacial area. Three special cases are examined. In the first, it is assumed that all growth occurs at the surface of the dispersed phase. In the second and third, it is assumed that growth occurs both at the interface and in the continuous phase. The second case assumes that substrate equilibrium is continuously established between the two phases while the third assumes substrate consumption in the continuous phase is limited by rate of transport of substrate to that phase. Comparison of the first model with available experimental data shows good agreement between model and data.

    Notes: A number of improvements have been made in a totally-automated antibiotic bioassay machine previously described. The new machine accepts unmeasured, untreated, opaque suspensions of fermentation beers three times faster (120 samples per hour) and supplies printed potencies sooner (in just over two hours). Whereas the original machine employed a self-cleaning filter and used disposable two milliliter beakers, this version involves a batch-dialysis scheme for effecting sample purification, and provides for automated cleaning of incubation chambers.In operation, a measured, portion of thoroughly-mixed fermentation beer is automatically diluted and transferred into one side of an incubation chamber, the two halves of which are separated by a dialysis membrane. The other half is filled with inoculated media. During the two hour incubation at 37°, dialyzable antibiotic limits growth of the inoculum in proportion to its concentration. After incubation, the turbidity of the inoculum is simultaneously read by an online computer and plotted on a strip chart recorded. The computer suplies printed potency values and sample identification on site, while the recording provides the operator with an analog record of turbidity. Fiber optics are employed in the turbidmetric readout, and an electric typewrite provides the printout.

Semicontinuous growth of E. coli K-12 MO7 yielded 77 kg of harvested cells in four days. Phenol extraction and ethanol precipitation recovered a crude tRNA material that was further purified by DKAE-cellulose chromatography in runs of 1 × 106 A260 units each on a 6 × 30 in. column using a 240 1, gradient elution. The purified tRNAs were partially concentrated and resolved into three groups.

    Notes: Streplomyces griscus var. X-2455 produces an antibiotic complex which is active in vitro against a number of gram-positive and gram-negative bacteria and in mice against systemic infections caused by K, pneumoniae and D, pneumoniac. In view of the favorable chemotherapeutic index and the broad in vitro spectrum of crude concentrates, isolation of the pure antibiotic complex and the individual constituents was undertaken. The antibiotics referred to as Ho 5-2667, Ro 7-7730, and Ho 7-7731 can be differentiated by tle, ultraviolet light absorption spectra, and in vitro antibacterial activities. They all contain iron and may be classified as sideromycins.From antibiotic concentrates an antibacterially inactive substance was isolated and identified as N-acetyltyramine.

    Notes: A brief review of the development of the research on erythorbic acid fermentation was presented. A previously proposed scheme of the acid biosynthesis has been proved to be correct. D-glucono-γ-lactone dehydrogenase was purified to 50 fold and compared with other lactone dehydrogenases. For the purpose of commercial development, screening and mutagenie treatments of strains and studies on fundamental cultural conditions were carried out. Penicillium, but no other genera, was obtained as a producer. The experiments of ultraviolet irradiation and various cultural conditions were successful in elevating the yield of the acid over 40% in jar-fermentor to glucose supplied. The continuous multibed extraction system of anion-exchange resin was developed and a yield of 19.2% of the acid from fermentation broth was obtained.

    Notes: Bromination of the thiazole ring by the enzyme chloroperoxidase, produced by the fungus Caldariomyces fumago, was demonstrated. Both 2-acetoacetamido-4-methylthiazole and 2-acetamidothiazole were brominated on C-5 of the thiazole ring in the presence of chloroperoxidase, bromide, and hydrogen peroxide in 0.06 M Phosphate solution at pH 3.0. No reaction occurred in the absence of enzyme.

    Notes: Red kidney bean (Phascolus vulgaris) cells, derived from roof, callus, were grown in suspension culture in shake flasks and in laboratory fermentors using batch and continuous batch culture techniques. The medium contained casein hydrolysate, sucrose, inorganic salts, vitamins, and growth hormones. In continuous batch culture yields of up to 171 g wet weight, (8.5 g dry weight) per liter were obtained in 7 days. Organic nitrogen was used preferentially. Growth on nitrate was considerably slower than on organic nitrogen sources. Indole acetic and naphthalene acetic acids were not essential for good growth of the cells whereas kinetin and 2, 4-D were. The optimum pH for growth was about p11 4.5. The presence of amylase and peroxidase was detected in culture filtrates. Amylase activity was low in either the presence or the absence of starch in the medium. Peroxidase production could be related directly with growth of the culture. Maximum peroxidase yield, as measured by the guaiacol method and expressed as horse radish peroxidase, was 1.25 × 10-8 M.

    Notes: Batch experiments were run using heterogeneous populations to determine whether a hyperbolic equation of the type suggested by Monod could be used to depict the relation between specific growth rate, μ, and NH3-N concentration when ammonia N was the growth-limiting nutrient. The heterogeneous populations employed were developed from sewage seed grown on glucose at various levels of nitrogen and various dilution rates in completely mixed continuous flow reactors. It was found that the hyperbolic function could be used. Values of μm in the range of 0.4-0.7 hr-1 were observed, and values of Ks, in general, ranged from 1.5 to 4.0 mg/l. Variation in the values of these growth “constants” did not follow any discernible pattern related to past growth history (i.e., COD:N ratio or dilution rate at which the cells were previously grown).

    Notes: Heterogeneous populations of sewage origin were grown continuously at, dilution rates from 1/12 hr-1 to dilute-out (1/1 hr-1) using glucose (1000 mg/l) as carbon source and three concentrations of NH3-N as the nitrogen source (COD:N = 70:1, 40:1, and 25:1). The effects of nitrogen level and growth rate (dilution rate) on substrate removal, biological solids production, cellular carbohydrate and protein, and NH4+-N in the effluent were examined. It was found that the optimum level of nitrogen supplementation for the synthetic nitrogen-deficient waste employed should not be based solely on the desired effluent quality with respect to COD removal but should include due consideration of reactor detention time (or dilution rate) and the allowable (or desirable) level of nitrogen leakage in the effluent.

    Notes: A synthetic waste (with glucose as carbon source) devoid of a source of nitrogen was purified in a laboratory scale pilot plant by a new modification of the activated sludge process. The process makes use of a separate carbon assimilation (oxidative assimilation) phase and an endogenous phase in which ammonia is added to a portion of the settled sludge and non-nitrogenous products stored in the cells in the assimilation phase are converted to protein. It was found that sludge so treated, when recycled to the assimilation tank, could carry out continuous oxidative assimilation of the waste. Various COD:N ratios were studied. At the highest, 70:1, 90% purificaton efficiency was achieved.

    Notes: Continuous saccharification of Solka Floc (cellulose pulp) in single and four-vessel stirred-tank reactor systems has been possible employing enzymes obtained directly from submerged fermentation of Trichoderma viride QM 6a. Studies on the effect of modification of the solid substrate, enzyme stability, substrate concentration, and the influence of reducing sugar concentration on the rate of hydrolysis are reported. While susceptibility of substrate to digestion is not affected by heating alone, it is strikingly increased by heating plus grinding, or by grinding following heating. Batch and steady state continuous saccharification experiments have yielded more than 5% reducing sugar in the effluent with a dilution rate of 0.025 hr-1 at 50°C, at a substrate level of 10%. An average glucose concentration of 3.4% has been obtained in the effluent of a continuous saccharification using 5% substrate at the same dilution rate and temperature.
