    Notes: Abstract Antimicrobial treatment of otitis media, especially drug dosing considerations, is largely empiric, with few reported pharmacologic studies of drug distribution into the middle ear. A chinchilla animal model of serous and purulent otitis media has been used for some time to investigate mechanisms of disease pathogenesis. This model was adapted to investigate the penetration of amoxicillin, trimethoprim, and sulfamethoxazole into middle ear effusion. Purulent otitis media was produced by direct middle ear inoculation with type 7F Streptococcus pneumoniae. Serous otitis media was produced by eustachian tube obstruction using silastic sponge or Coeflex cement, but the Coeflex caused an undesirable local inflammatory response. The three antibiotics were administered to chinchillas with serous and purulent middle ear effusion. Plasma and ear fluid drug concentrations were measured by liquid chromatography and demonstrated the value of this model in assessing antibiotic penetration.

    Notes: Abstract Physiological models have often been used to investigate the processes involved in drug targeting. Such a model is used to investigate some aspects of drug targeting, including the pharmacodynamics of therapeutic and toxic effects. A simple pharmacodynamic model is incorporated in a three-compartment pharmacokinetic model. Conventional administration and drug targeting are compared at steady state for the same degree of therapeutic effect. The efficiency of drug targeting is quantified as the ratio (TA) of the rates of administration of free drug or of a drug–carrier complex required to achieve this effect. Also, the ratios of drug concentrations in the toxicity compartment (DTI) or of the consequent degree of toxic effects (TI) are used to compare conventional administration with drug targeting. The kinetic characteristics of the drug–carrier complex, rate of elimination, and rate of free drug release, influence TA but not DTI or TI. The importance of these characteristics depends on the cost and toxicity of the drug–carrier complex or of the carrier alone. The pharmacodynamics of the free drug in both the target and the toxicity compartments have an important influence on TI but not on TA or DTI. As the pharmacological selectivity of the drug increases, so does TI. However, a drug with good pharmacological selectivity may not be suitable for drug targeting. TI is also very dependent on the shape of the effect–concentration curves, particularly that for toxicity. While TA increases as the rate of elimination of free drug from either central or target compartments increases, TI may actually be reduced if release of free drug is not confined to the target compartment.

    Notes: Abstract The pharmacokinetics and bioavailability of scopolamine were evaluated in six healthy male subjects receiving 0.4 mg of the drug by either oral or intravenous administration. Plasma and urine samples were analyzed using a radioreceptor binding assay. After iv administration, scopolamine concentrations in the plasma declined in a biexponential fashion, with a rapid distribution phase and a comparatively slow elimination phase. Mean and SE values for volume of distribution, systemic clearance, and renal clearance were 1.4 ± 0.3 liters/kg, 65.3 ± 5.2 liters/hr, and 4.2 ± 1.4 liters/hr, respectively. Mean peak plasma concentrations were 2909.8 ± 240.9 pg/ml following iv administration and 528.6 ± 109.4 pg/ml following oral administration. Elimination half-life of the drug was 4.5 ± 1.7 hr. Bioavailability of the oral dose was variable among subjects, ranging between 10.7 and 48.2%. The variability in absorption and poor bioavailability of oral scopolamine indicate that this route of administration may not be reliable and effective.

    Notes: Abstract The pharmacokinetics of platinum was investigated in 10 cancer patients treated with a 1-hr infusion of 300 mg/m2 of carboplatin which was given 2–4 days after the administration of 100 mg/kg (20-mg/kg bolus and 80-mg/kg intravenous infusion) of methotrexate. Platinum was analyzed in the samples by flameless atomic absorption spectrophotometry. The concentration vs time data for total platinum in plasma followed a two-compartment model and the mean (and SE) values for β, TBC, V c, and RC were 0.0827 (0.22) hr−1, 2.355 (0.252) liters/hr · m2, 10.74 (0.62) liters/m2, and 2.405 (0.228) liters/hr · m2, respectively. There was no significant change in the creatinine clearance or TBC with repeated treatment. The ultrafilterable platinum which was measured in the plasma of two patients constituted 82 and 11.3% of the total platinum at 1 and 24 hr, respectively, and the data conformed to the one-compartment model. The mean (SE) values for t β, TBC, and V d for free platinum were 1.844 (0.208) hr, 4.583 (1.059) liters/hr · m2, and 11.88 (1.45) liters/m2, respectively. The above data are in good agreement with those reported earlier for platinum following the administration of carboplatin as a single agent. These results suggest that high-dose methotrexate therapy, when administered 2–4 days before carboplatin, does not affect the pharmacokinetics of platinum in the plasma.

    Notes: Abstract A Phase I study of recombinant interferon-gamma (rIFN-γ) was conducted to determine the toxicity and pharmacokinetics of this lymphokine in acquired immunodeficiency syndrome (AIDS) patients with Kaposi's sarcoma (KS). Sixteen patients with AIDS/KS were entered into a fixed-dose trial at either 0.001, 0.01, 0.1, or 1.0 mg/m2 of rIFN-γ. rIFN-γ was initially administered either as a single 24-hr continuous iv infusion or as a single im injection, followed 4 days later by a 10-day course of daily therapy by the same route. Following a 1-week washout period, this sequence of administration was then repeated, with the drug given by the alternate route. Pharmacokinetic analysis of the 1.0-mg/m2 group revealed that peak serum levels of up to 153 U/ml occurred 2–4 hr after im injection and that steady-state levels of up to 40 U/ml were reached approximately 7–12 hr after beginning iv infusion. Dose-related toxicities in this trial included fever, headache, fatigue, nausea, and hepatitis, all of which were most severe at the two highest doses. Dose-dependent depression of the total white blood-cell (WBC) count, affecting both granulocytes and lymphocytes, was the most common laboratory abnormality. Natural killer (NK)-cell activity was slightly enhanced at a dose of 0.1 mg/m2 but suppressed at 1.0 mg/m2 of drug; monocyte-mediated cytotoxicity, in contrast, was significantly increased only at the highest dose. No dose-related changes were noted in KS lesions, HLA-DR expression by peripheral blood mononuclear cells, lymphocyte blastogenesis, or the ability to culture cytomegalovirus (CMV) from body fluids. We conclude that a maximally tolerated dose (MTD) for this drug is in the range of 0.1–1.0 mg/m2 and that at least modest evidence of systemic immunomodulation may be seen when rIFN-γ is given at doses at or near this MTD.

    Notes: Abstract From a pharmacochemical point of view the existing anti-leprotics as well as possible innovations in the chemotherapy of leprosy are discussed. Of the main anti-leprotics, which are used nowadays — dapsone, rifampicin, clofazimine, isoniazide, ethionamide and prothionamide — the mechanism of action, the main problems in their application and possibilities to develop improved variants are reviewed. Based on the chemistry ofMycobacterium leprae, the target systems for new anti-leprotics are identified. These systems include the cell wall, the catabolism of reactive oxygen species, the metabolisms of carbon sources, the amino acid metabolism and the uptake of iron. Two possible new lead structures from other fields, 4-quinolones and mycobacterial ribonucleotide reductase inhibitors are presented.

    Notes: Terbutaline is a β2-agonist and administered as the racemic mixture. The pharmacokinetics of the separate enantiomers differ with respect to degree of absorption and clearance. In the present study, repeated doses of racemic terbutaline were given to six healthy volunteers. Plasma was analyzed for the concentrations of the two enantiomers. The observed plasma concentrations at steady state differed from those predicted from the values observed after single dose administration of the separate enantiomers. The difference between the observed and predicted values can be tentatively explained by a combined influence of (-)-terbutaline on the absorption of (+)-terbutaline and the influence of (+)-terbutaline on the elimination of (-)-terbutaline. The results have implications for the interpretation of effect/concentration studies with terbutaline, but do not affect the doses used in clinical practice.

    Notes: Abstract The role of histamine in brain function is discussed. A brief review is presented on the three types of histamine receptors with regard to their biochemistry and functions, as well as on specific ligands, both agonists and antagonists. It is concluded that several aspects of the pharmacology of histamine in the central nervous system are still a matter of empirism and speculations. The recent publications on the brain-penetrating H2 antagonist zolantidine and the selective H3 agonists and antagonists are expected to contribute to the knowledge of the histaminergic pathways in the brain. Therapeutical areas within the central nervous system are related to behaviour (including wake-sleep), neuroendocrinal processes, the control of muscle activity and cerebral circulation.

    Notes: Summary A survey of the pharmacokinetic properties of the three prototypical calcium antagonist agents shows that they have in common a very high rate of hepatic first-pass metabolism with, in the case of verapamil and diltiazem, the formation of an active metabolite that affects the dose during chronic therapy. Therefore, the major factor altering the pharmacokinetic properties and the dose of the drug required is the capacity of the liver to metabolize the drug, which in turn depends on the hepatic blood flow and the activity of the hepatic metabolizing systems. Hence liver disease, a low cardiac output, and coadministration of certain drugs inducing or inhibiting the hepatic enzymes, all indirectly affect the pharmacokinetic properties of the calcium antagonists. There are also other potential drug interactions of a kinetic or dynamic nature that may arise. In general, renal disease has little effect on the pharmacokinetics of calcium antagonists.

    Notes: Abstract A pharmacokinetic/pharmacodynamic model of hypersensitivity to adrenergic stimulation following abrupt withdrawal of chronic β blockade was developed. The model employs the Hill equation, a term which describes the competition between isoproterenol and l- propranolol for β receptors, and a kinetic term which characterizes the appearance and disappearance rates of up-regulated β receptors. The model predicted peak chronotropic hyperresponsiveness to isoproterenol 48 hr following abrupt withdrawal of chronic treatment with daily propranolol doses of 160 mg, and a drug half-life of 3.5 hr. The model also predicted that increasing the dose rate and prolonging the half-life of propranolol delayed and decreased the extent of adrenergic hypersensitivity. The time-course of adrenergic hypersensitivity simulated by our model was in excellent agreement with that observed in studies which were published earlier by our laboratory. The model underestimated the extent of adrenergic hypersensitivity. The results of our simulation are consistent with a β agonist-receptor-effector system, which involves spare receptors, amplification of response by second and third messengers, and β agonist-antagonist-induced receptor regulation.

    Notes: Abstract Myocardial contractility of normotensive and spontaneously hypertensive rabbits was determined following an iv bolus injection of propranolol HCl. Left ventricular pressure and dimension were used to calculate the contractility parameters of (dP/dt) max ,maximum fiber shortening velocity V cf , and the slope of the end systolic pressure-end systolic volume line (ESP-ESV line). Hypertension was induced by a methoxamine HCl iv infusion which mimicked the cardiac effects seen in essential hypertension. Propanolol caused a significant decrease in all contractility parameters (p〈0.05)within 15min after administration, with a peak effect occurring after 30–35 mins. The pharmacokinetics and pharmacodynamics of propranolol were fit using Hill's equation in conjunction with the concentration of drug in the theoretical effect compartment. The normotensive group of rabbits had a calculated EC (50) of 12.7 ng/ml, while the hypertensive group had an EC 50 of 6.9 ng/ml,indicating that the hypertensive rabbits were much more sensitive to the propranolol than the normotensive group. In addition, the normotensive group of rabbits demonstrated a much different pharmacokinetic-pharmacodynamic relationship than that of the hypertensive group, indicating that the hypertensive state of the animal has a significant effect upon the concentration-effect relationship.

    Notes: Abstract The theoretical groundwork for a rate constant formulation of inhaled anesthetic elimination kinetics is discussed. In an effort to simulate recent experimental results a linear flow-limited five-compartment model was used comprising lung, vessel-rich tissue, muscle, nonvisceral fat, and an additional compartment, marrow-visceral fat whose functional existence recently has been experimentally demonstrated. Hypothetical but plausible parameters for the marrow-visceral fat compartment were used. The theoretically predicted values were in good agreement with experimental results suggesting that this model is appropriate for the elimination kinetics of agents that are not metabolized to any significant degree. Simple approximate expressions for the rate constants were also derived and were in reasonable agreement with experimental results. The model was also employed to clarify the effect of anesthetic duration on subsequent elimination kinetics.

    Notes: Abstract A physiologically realistic model of enterohepatic cycling (EHC) which includes separate liver and gallbladder compartments, discontinuous gallbladder emptying and first-order absorption from both an oral formulation and secreted bile (ka po and ka b, respectively) has been developed. The effect of EHC on area under the first-moment curve (AUMC) of drug concentration in plasma and on parameters derived from the AUMC was investigated. Unlike AUC, AUMC is dependent on the time and time-course of gallbladder emptying, increasing as the interval between gallbladder emptying increases. Consequently, mean residence time (MRT) is also a time-dependent parameter. Analytical solutions for MRTiv and MRTpo were derived. Mean absorption time (MAT = MRTpo — MRTivj is also time-dependent, contrary to findings previously published for a model of EHC with a continuous time lag. MAT is also dependent on k a po , k a b and the hepatic extraction ratio. The difference between MRT po s two formulations with unequal k a po values may deviate from the difference in the inverse of their absorption rate constants. Implications for design and interpretation of pharmacokinetic studies include (i) MAT values may be dominated by the time-course of recycling rather than the time-course of the initial absorption, depending on the extent of EHC and (ii) the unpredictable nature of the time of gallbladder emptying will contribute to intrasubject variability in derived parameters during crossover studies. Knowledge of the extent of EHC is invaluable in deciding whether modification of the in vitro release characteristics of an oral formulation will have any effect on the overall time-course of absorption in vivo. Techniques to monitor or control gallbladder emptying may be helpful for reducing variability in pharmaco-kinetic studies for compounds which are extensively cycled in bile.

    Notes: Abstract The impact of assay variability on pharmacokinetic modeling was investigated. Simulated replications (150) of three “individuals” resulted in 450 data sets. A one-compartment model with first-order absorption was simulated. Random assay errors of 10, 20, or 30% were introduced and the ratio of absorption rate (K a )to elimination rate (K e )constants was 2, 10, or 20. The analyst was blinded as to the rate constants chosen for the simulations. Parameter estimates from the sequential method (K e )estimated with log-linear regression followed by estimation of K a and nonlinear regression with various weighting schemes were compared. NONMEM was run on the 9 data sets as well. Assay error caused a sizable number of curves to have apparent multicompartmental distribution or complex absorption kinetic characteristics. Routinely tabulated parameters (maximum concentration, area under the curve, and, to a lesser extent, mean residence time) were consistently overestimated as assay error increased. When K a /K e =2,all methods except NONMEM underestimated K e ,overestimated K a ,and overestimated apparent volume of distribution. These significant biases increased with the magnitude of assay error. With improper weighting, nonlinear regression significantly overestimated K e when K a /K e ,=20. In general, however, the sequential approach was most biased and least precise. Although no interindividual variability was included in the simulations, estimation error caused large standard deviations to be associated with derived parameters, which would be interpreted as interindividual error in a nonsimulation environment. NONMEM, however, acceptably estimated all parameters and variabilities. Routinely applied pharmacokinetic estimation methods do not consistently provide unbiased answers. In the specific case of extended-release drug formulations, there is clearly a possibility that certain estimation methods yield K a and relative bioavailability estimates that would be imprecise and biased.

    Notes: Summary The structure-activity relationship of two isomeric 1-aminoadamantanes, 1-C-ethylaminoadamantane (D 174) and 1-amino-3-ethyladamantane (D 175), on membrane anisotropy and the excitability of neurons was studied in the CNS of the rat and in the decerebrated cat. Mass spectrometric analysis showed that after a single, 40 mg/kg dose, D 174 and D 175 were unevenly distributed within the CNS of the rat, moreover the distribution pattern of the two substances was different. As measured by fluorescence depolarization in controls the membrane anisotropy was found to be higher in the older parts as compared with the younger parts of the CNS. After i.p. application of 40 mg/kg the membrane anisotropy was reduced in the cortex by D 174, whereas D 174 and D 175 increased the rigidity in striatal membranes. If cortical membranes were incubated with the substances, the fluidizing effect of D 174 was more prominent than that of D 175. In the decerebrated cat only D 174 in a dose of 5 mg/kg i.p. raised the discharge of spinal alpha-motoneurones significantly. The results suggest that the membrane architecture is more affected by D 174 as compared with D 175 which is reflected by a greater effect on membrane anisotropy as well as on the activity of spinal alpha-motoneurones.

    Notes: Penicillium funiculosum NRRL 13033 produced β-glucosidase and β-xylosidase activities when grown on wheat straw. The addition of some inducers (individually or in combination) to the fermentation medium were tested for the production of both enzymes. The relation of mycelial bound enzyme to cell free enzyme was studied during incubation period of fermentation. The optimum activity of β-glucosidase and β-xylosidase were found to be in the pH 4.5 using phosphate-citrate buffer at 50°C for 60 min and at 55°C for 40 min respectively. β-Glucosidase lost about 40% of its original activity by heating to 65°C for 60 min, while, β-xylosidase activity was found to be nearly stable with the same treatment. Both enzyme activities were greatly inhibited when 1.0% (w/v) of xylose and glucose were added to the assay mixture.

    Notes: A bioelectrode consisting of two parallel noble metal nets and a thin layer of gel-entrapped urease between them permits to determine urea conductimetrically with a response time 3-5 min and reproducibility 5-6% above 60 micromolar concentration of urea. Differential measurement eliminates the interference due to conductive components of the real sample. Evidence has been obtained for extended application of the method by using immobilized microbial cells or tissue slice as biocatalyst in place of the immobilized purified enzyme. It is also suitable for simple kinetical assaying arginase activity in the tissue extracts.

    Notes: The effects of an extracellular microbial glycolipid, the interfacial active lactonic sophoroselipid, and of Triton X-100 on strains of Acinetobacter calcoaceticus are compared. Sophoroselipid diminished growth rates on n-heptadecane. Both surfactants led to the excretion of enzyme activities into the culture medium. Sophoroselipid increased the release of cytoplasmic malate dehydrogenase whereas in presence of Triton X-100 the quinoprotein glucose dehydrogenase was also excreted in large amounts.

The SLP-2 was developed for the control of high biomass concentrations in fermenters and is fitted with a self-cleaning mechanism. The SLP-3 is used for control of very small turbidities, e.g. in brewery products or in drinking water. The SLP-4 is designed for continuous measurement of higher turbidities in (waste) water or in different products flowing through pipe-lines. It may be used alternatively as a submerged probe.

    Notes: The action of thermostable α-amylase produced by Bacillus licheniformis 44MB82 strain on soluble and insoluble starch, amylose and amylopectin at temperatures 30°C and 90°C was studied. The hydrolysis of soluble starch proceeded rapidly for 10 to 15 minutes after which the maltodextrins thus formed were further dissociated. In the course of 60-minutes enzyme treatment mainly glucose, maltose and maltosugars (from G3 to G6) as low molecular weight products were found and the formation of maltcse and maltotriose was increased by the longer treatment. The hydrolysis of insoluble starch and amylopectin proceeded in the same way while the amylose was hydrolysed slowly.

    Notes: The effects of ethanol and Trichoderma reesei cellulase on the saccharification and fermentation processes as well as the tolerance of the cellulase complex for ethanol have been investigated. The studies were conducted with respect to their usefulness in the process of simulataneous saccharification and fermentation of cellulose to ethanol. The following results were obtained. (1) Fermentative activity of Kluyveromyces fragilis yeasts was gradually depressed with increasing intial ethanol concentrations and temperature of fermentation between 35-46°C. (2) Crude cellulase preparation introduced to the culture broth to a final enzyme activity of 0.5 to 2.0 FPU/ml had not distinct effect on the biomass production, ethanol yield, and glucose uptake by yeasts in 48 h fermentation at 43°C. On the other hand, only a negligible decrease in the cellulase complex activity was observed during fermentation process. (3) Saccharification of wheat straw was inhibited by at least 1% w/v ethanol. (4) The enzymes of the cellulase system showed a high stability to exposure to ethanol for 48 h at 43°C.

    Notes: The continuous ethanol production by free and Ca-alginate-entrapped cells of the yeast Saccharomyces cer. IBT H 191 was investigated in a membrane reactor and in a CSTR, respectively. An empiric productivity model for free-cell fermentation was found to be valid for immobilized cells if no external diffusion resistance existed within the reactor. In such a case the fermentative power of the immobilized cells was identical with this one of the free cells. A change of the ethanol-tolerance behaviour of the cells as a result of the cell-immobilization did not occur.

    Notes: We report here the construction of a promoter-probe vector, pRS2, which can be utilized in either Acetobacter methanolicus MB 58 or Escherichia coli due to the presence of broad-host-range replicon RSF 1010. The vector provides several unique restriction sites for promoter cloning as well as resistance markers for the selection of transformants. The promoter-probe vector was constructed by inserting an EcoRI-SalI-polylinker fragment of pUC 19 into EcoRI/SalI digested pMK 16. The resulting plasmid, pRS1, was cloned into the unique EcoRI site of the broad-host-range plasmid RSF 1010.The vector was used to clone promoter-containing sequences derived from the A. methanolicus MB 58 chromosome as well as the E. coli lac-promoter.

    Notes: In the present work, a mathematical model was developed regarding the immobilized living yeast cell reactor for sugar bioconversion to ethanol. The model, composed of a system of ordinary differential equations (ODEs) enables the computation of the paramters involved in the steady state reactor behaviour. Comparing the values computed through the integration of this mathematical model with the experimental data, it has been shown its capacity to describe sufficiently accurate the steady state behaviour of the continuous fixed film bioconversion reactor.

    Notes: Four silica supports differing in pore dimensions were activated by treatment with SiCl4 and then with ethylenediamine to obtain alkylamine groups on the silica surface. Three enzymes, peroxidase from cabbage, glucoamylase from Aspergillus niger C and urease from soybean were immobilized on these supports using glutaraldehyde as coupling agent. It was found that the protein content, the retained enzymatic activity and the storage stability of the silica supported enzymes were considerably affected by support pore size and enzyme molecular weight, the factors which are supposed to alter protein distribution inside the support pores. The highest activity was found for peroxidase and glucoamylase attached to the silica with the widest pores, but their loss in activity during storage was considerable. The urease retained less activity after immobilization, but its storage stability was excellent.

    Notes: The oxygen requirements of Aspergillus awamori as well as the adaptation to it and the aeration of the cultivation medium were determined in the process of glucoamylase synthesis. Under the selected agitation and aeration conditions (impeller-tip speed = 4.2 m/s; aeration 1.5 vvm) the cultivation-medium aeration was analysed by means of dissolved-oxygen-concentration measurement during the course of the process. It was demonstrated that for obtaining the glucoamylase-activity level of 800 U GA/cm3 under the selected conditions and with the fungus applied the dissolved-oxygen concentration at the level of 25% saturation should be maintained. Those findings could serve as auxiliary indexes in the scale-up process of glucoamylase synthesis.

    Notes: A new biotechnological process for sulphide removal is proposed. The process is based on the oxidation of sulphide into elemental sulphur, which can be removed by sedimentation. In this study it was found that elemental sulphur and sulphate are the main oxidation products of the biological sulphide oxidation. The settling characteristics become worse as the sulphide concentration increases, due to polysulphide formation. The start-up phase of this biological system is very short; Only four days are needed to reduce the sulphide concentration of 100 to 2 mg/l at a HRT (Residence time) of 22 minutes. Also some environmental factors were evaluated. The optimal pH is situated in the pH-range 8.0-8.5. Significantly lower conversion rates are found at pH = 6.5 to 7.5 and pH = 9.0, while at pH = 9.5 the sulphide oxidation capacity of the system detoriates. The process temperature was 20°C, although the optimal temperature is situated in the range 25-35°C. No substrate inhibition of sulphide was found at sulphide concentrations up to 100 mg/l.

    Notes: The method described allows the determination of dissolved oxygen in water-inmiscible organic liquids under normal pressure in a temperature range of 0°C to 80°C depending on the boiling point of the organic material. The method can be adapted to modified conditions.The method using the principles of O2-measurement by means of CLARK'S electrode offers an accuracy of 2-6%. The exact knowledge of dissolved molecular oxygen in organic liquids can be of interest for chemical or biotechnological processes.

    Notes: The silver sorption capacity of three methylotrophic bacteria and one gelatinotrophic Pseudomonas strain has been investigated in dependence of the cultivation conditions. Whereas the maximum loading capacity of the microorganisms was not appreciable influenced by the variation of the dilution rate in the range between 0.05 h-1 to 0.5 h-1, a change of the carbon source or the modification of the limitation conditions during the chemostatic cultivation process provokes a strong alteration in the sorption behaviour. The maximum silver loading values were obtained, when the microorganisms are cultivated under N-limitation and the minimum values were measured under P-limitation.

    Notes: The degradation of fifteen sugars, sugar acids and related substrates were examined using cellfree extracts of Aspergillus ustus growing on D-glucose, D-mannose, D-galactose or D-gluconate as the only carbon source. D-gluconate was superior for the induction of the enzymes capable for the degradation of some of these substrates. The addition of 0.5% malt extract with D-gluconate to the growth medium or the presence of shaking conditions resulted to an increase in the degradation of those substrates, whileas the incorporation of 0.5% malt extract alone to the medium has no effect. Extracts of D-gluconate-grown mycelia of A. ustus degraded D-gluconate 〉 D-galactonate 〉 1 : 5 gluconolactone and 〉 L-arabonate nonphosphorolytically more effectively. Optimum pH and temperature for the degradation of D-gluconate were found to be 8.0 and 40°C, respectively. Thermal stability studies on the behaviour of D-gluconate dehydratase showed that this enzyme was stable at 50°C and 60°C for 30 and 5 minutes, respectively. Specific activity of this enzyme was increased three times when cell-free extracts were incubated at 60°C for 5 minutes. MgCl2 and CoSO4 were good activators, while CaCl2 p-mercurychlorobenzoate (PMCB), sodium arsenite, ZnSO4, CuSO4, iodoacetic acid, MnCl2 and FeSO4 were potent inhibitors for D-gluconate dehydratase activity. Km was calculated for D-gluconate and found to be 2.5 × 10-2 M.

    Notes: Calcium in soymilk was increased to that of human milk by the addition of 0.2% calcium lactate. It was, however, impossible to raise the calcium content of soymilk to that of cow's milk. The maximum amount of calcium lactate that could be added to soymilk without coagulation of the milk protein was found to be 0.45% which resulted in about 75 mg of Ca/100 ml of soymilk. This amount was approximately 60% of the calcium in cow's milk. Sodium citrate had to be added in combination with calcium lactate to reinforce colloidal stability of the milk. The ratio of Ca to P in the milk containing added 0.45% calcium lactate was comparable to that of cow's milk and at the amount normally consumed daily by infants and children it seemed to meet the requirements of Ca and P.

    Notes: In growth-factor limited fermentations carried out in continuous high-flow rate fermenters the relationship existing between the specific ethanol formation rate and the ethanol concentration was found to change its characteristics from a linear function into a nonlinear one at high biomass concentrations. The deviation from the linear inhibitory behaviour of the cells was the greater the more the biomass concentration increased.A very good correspondence between the experimentally found productivities of ethanol formation and its adequate values obtained by calculation could be attained by using an improved steady-state productivity model, in which the variability of the function v = f(P) had been considered.

    Notes: Studies on the pelletization of fungus Pleurotus ostreatus have established the pellets to differ as to the size, density and chemical composition. Changes in the pellets, taking place during culture growth, are helpful in studying the mode of their development.     Notes: The cellulase complex of the fungus Aspergillus niger (strain CBS 554.65 = ATCC 16 888) was fractionated by gel filtration yielding six pronounced peaks. Only proteins from the fraction corresponding to the first peak (96 kDa) showed β-glucosidase activity vs. the substrate 4-nitrophenyl-β-D-glucopyranoside (pNPG). These proteins have been fractionated by chromatofocusing, yielding two β-glucosidases (I and II) which are shown to be homogeneous in isoelectric focusing experiments (pI = 4.6 and 3.8, respectively). Kinetic experiments with pNPG, MU-glucopyranoside and cellobiose revealed that both types of β-glucosidases behave like aryl-β-glucosidases. β-Glucosidase-I acting on pNPG exhibits a split kinetics characterized by high and low substrateconcentration kinetics which are differentiated by different values of V and of Km. In addition, β-glucosidase-II is shown to be an exo-glucohydrolase as deduced from experiments with MU-cellobiopyranoside.Experimental features should be emphasized; usual soft-gel ion-exchange materials did not work in the chromatofocusing separation of the two β-glucosidases, in contrast to the 10μ-Si 500 = DEAE exchange material (Serva) typically used in HPLC-experiments. Furthermore, protein content determinations based on different procedures yielded widely differing values.

    Notes: The sorption capacity of silver on different biological materials has been investigated depending on physico-chemical pretreatments. The maximum silver loading values measured were compared with the values obtained with nontreated biomasses.The results show an increase of the loading capacity up to a factor of 10 in case of the alkalitreated biomasses. When the biomasses are extracted before being used as adsorbent with a solvent mixture of chloroform/methanol in a ratio of 2:1 the efficiency of the silver adsorbing power can be increased.Beyond that, the ability to adsorb silver can also be influenced when microorganisms are used as biocatalysts in a product synthesis before they are used as adsorbents. A strain of Acetobacter methanolicus possesses 1.8 times higher affinity to silver when it is employed in a process of gluconic acid production before adsorption. Physico-chemical pretreatments influence not only the loading capacity of the biological material, but also the contacting time required for the establishment of the adsorption equilibrium can be considerable reduced.

    Notes: Kluyveromyces bulgaricus protoplast formation was studied by the determination of the remaining intact cells with the Coulter counter, the decrease of the absorbance of the cell suspension upon dilution into an hypoosmotic medium and the liberation of intracellular proteins after protoplast lysis. Due to the scattering of light by cells and ghosts, spectrophotometric readings were not enough specific and led to the underestimation of the reaction rates (about 40% less). The Coulter counter technique and the determination of the liberation of intracellular proteins gave the same rates over a large range of cell concentrations and in the presence of various cell lytic enzymes. The importance of corrected etermination of the time course of the reaction was illustrated by the good correlation obtained between protoplast formation and ATP turnover.

    Notes: A method for improving the nutritional value of straw as a substrate for fungal growth by radiation is described. The conditions for fungal growth on a medium containing irradiated straw of winter rye were studied. It was found that the straw substrate improves both the quantity of carbohydrates and the quality of their composition in the nutrient medium. Easier accessibility of the irradiated substrate to enzymatic hydrolysis allows the time bioconversion of cellulose into protein to be reduced and the protein yield to be increased, thus improving the qualitative composition of the fungal biomass. The data obtained suggest a possibility of converting straw into a fodder product by substituting the method of using reagents for pretreatment by a simpler and less labour-consuming method.

    Notes: An attempt is made to give a quantitative estimation of a possible contribution of the dissociation process in the formation of the heterogeneous population structure for the Rhodococcus rubropertinctus (Micobacterium lacticolum) in continuous cultivation. This estimation is realised with the help of a constructed mathematical model of the process. The model equations obtained describe particular biomass changes for R-, S- and M-dissociants and changes for substrate (glucose) concentration within the chemostate in continuous cultivation. Next, contributions to dissociant biomass changes were taken into account: (1) cell reproduction (each dissociant has its own specific growth rate depending on the substrate concentration), (2) cell outflow (in proportion to dissociant biomass) and (3) dissociation (dissociant genotype change during reproduction occurs for each dissociant with its own specific frequency).It is determined that if substrate growth constants are equal the rapidly growing dissociants (possessing maximal μmax) outs the rest of them, and the greater the speed of the flow the less the time to reach a steady-state distribution of the dissociant biomasses. But, at the same time the slowly growing dissociant can become dominant in the population if it has a lower substrate growth constant.

    Notes: A quadrupol mass spectrometer QMS c 300 developed and manufactured from the workshops of the Institute of Nuclear Research of the Hungarian Academy of Sciences, Debrecen, has been cuppled with a 12 l laboratory steril fermenter. The QMS apparatus is provided with 3 different possibilities for sampling. Via a dynamic air input system the discharged air of the fermenter can be analyzed continuously. The static sampling input allows the off-line measurement of gaseous and volatile compounds. Special membran samplers are used for measurements of volatile and gaseous substrates and microbial metabolites in the fermentation broth. The samplers can be adopted in the reactor vessel directly analogous to the pH and pO2-electrodes.

    Notes: Different methods of bubble-free oxygenation are discussed. Today, the surface oxygenation has lost its importance. Technically, membrane oxygenation methods are used in various fermentervolumes although the construction principles are quite complicated. The scale-up-potential is here very limited. On the other hand, the CHEMCELL-oxygenation is a simple method, safe in terms of sterility and applicable to all fermenter sizes common for cell culturing. A great benefit of this principle is that the amount of oxygenation is not dependent from the mixing parameters.

    Notes: The knowledge of the upper limits of growth rate and cell density is necessary to define the maximum possible productivity of a growth process. After a brief survey of the theoretical basis of microbial growth in dependence of mass transfer of oxygen and rheological properties of fermentation medium recent experimental results on the influence of higher pressure in the fermenter, of different substrate and dissolved oxygen concentrations on growth rate and yield coefficients of methanol utilizing bacteria are presented. Also some correlations between cell concentration and viscosity of fermentation broth are considered.

    Notes: The increased interest in large scale production of biologically active molecules, as for example monoclonal antibodys, hormones, proteins and enzymes, has stimulated a rapid development of different methods to cultivate eukaryotic cells. Further progress in this field of modern biotechnology is expected not only from the selection of more productive cell lines and more efficient cultivation techniques, but also from the improvement in new bioreactor design and operation, which guarantees increased productivity per unit volume and reduces the downstream processing. Most important factors for a new reactor design in future will be the energy- and mass transfer, shear stress and scaleability. The provision of an adequate oxygen supply to large scale reactors is the most critical barrier to scale up. If oxygen is limited in a small degree, the result is inhibition of cell density and cellular efficiency in the production of the desired biomolecules. In addition, when methodologies are used which allow high cell densities and high metabolic active cells, the oxygen transfer becomes more important and, at the same time, more difficult. Since direct sparging of air into the cell-containing medium causes problems such as shear forces and foaming, new, efficient methods in bubble free aeration must be utilized. A new aeration system is presented, which is suitable to bubble-free aeration as well to separation of microcarrier-anchered cells from the harvested medium when running in a continuous perfusion mode. The efficiency of the CHEMCELL System regarding aeration and cell retention is demonstrated by the growth kinetics of BHK 21-cells in batch and perfusion mode.

But they have the advantage of high stability and there is no necessity of enzyme isolation and purification.     Notes: The methanol-utilizing yeast Hansenula polymorpha MH 26 is thermotolerant and grows at 40°C and pH 3.5 with a maximum specific growth rate of 0.23 h-1 on methanol.In continuous cultivation the maximum cell yield can be improved of 0.35 (methanol) to 0.44 g dry cells/g methanol (mixtures of methanol and stillage) through additional utilization of the essential growth and nutritive substances of stillage.The utilization of methanol and sucrose (molasses) at various mixtures is simultaneous possible. Investigations of these mixtures with 14C-marked methanol show an increasing incorporation of methanol of about 20-30% against methanol alone. This effect is caused by the increasing metabolization of sucrose to carbondioxide and additional energy delivery for better assimilation of methanol.

    Notes: Basis sets ranging in size from (16, 10, 7) to (20, 14, 11) have been derived for the atoms Y-Cd. Separate sets represent the energy optimized wave functions for each of the s2dn, s1dn+1, and s0dn+2 configurations. The energies from the largest sets are within 3 mhartrees of the values obtained in numerical Hartree-Fock calculations. Reasonable Hartree-Fock s2dn- s1dn+1 and s2dn- s0dn+2 excitation energies may be obtained either using the largest basis sets, or using d-orbitals optimized for the s0dn+2 configurations. The basis sets are slightly unbalanced in favor of the s-functions and in disfavor of the d-functions, but various alternative basis sets may be derived by combining parts of the five parent sets. The convergence of radial expectation values is discussed.

    Notes: A method for fitting a smooth ribbon representation of DNA structures is proposed. Following a review of the relevant definitions for classical linear helical DNA and generalizations to curving helices, a parameterization of smooth ribbons is given, which leads to tractable expressions. In addition it suggests a new way to define twist, tilt, and roll for a base step that is free of the ambiguities caused by noncommutativity of finite rotations. A least squares fitting criterion for ribbons is then proposed. In some cases the optimal ribbon with respect to this criterion is not unique. This problem is analyzed, and the circumstances in which it can occur are specified. To resolve the nonuniqueness problem, a variational description of the optimal ribbon is proposed, namely the ribbon of lowest elastic energy achieving a specified level of fit with respect to the least squares criterion. The appropriate level of fit is decided using distances of backbone atoms from fitted ribbon axes. Theoretical tests of the fitting methodology are presented, and as a sample application a smooth ribbon is fit to an existing experimental structure.

    Notes: Molecular recognition in Rebek's cleft was studied with the MM2 force field. A new computational protocol was used to determine the free energies of substrate binding to this topologically unique model receptor. The energies and structural features of substrate-receptor complexes reveal that molecular recognition involves ammonium ion binding to the interior of the cleft and aromatic π-stacking on the exterior of the cleft.

    Notes: A procedure that finds the most probable conformational states of a protein chain is described. Single-residue conformations are represented in terms of four conformational states, α, ∊, α*, and ∊*. The conformation of the entire chain is represented by a sequence of single-residue conformational states; the distinct conformations in this representation are called “chain-states.” The first article in this series described a procedure that computes tripeptide conformational probabilities from the amino acid sequence using pattern recognition techniques. The procedure described in this article uses the tripeptide probabilities to estimate the probabilities of the chain-states. The chain-state probability estimator is a product of conditional and marginal probabilities (obtained from the tripeptide probabilities), with a penalty factor to eliminate conformations containing α-helices and ∊-strands of excessive length. The probability estimator considers short-range conformational information, medium-range sequence information and some simple long-range information (through the restrictions on helix and strand lengths). Energy minimization calculations can be carried out in the region of conformational space corresponding to a particular chain-state. By selecting the most probable chain-states, the search can be focused on the most probable, or “important,” regions of the conformational space. These energy calculations are described in the third article of the series. The complete procedure described by the three articles is called PRISM, for pattern recognition-based importance sampling minimization.

    Notes: A procedure that generates random conformations of a protein chain, and then applies energy minimization to find the structure of lowest energy, is described. Single-residue conformations are represented in terms of four conformational states, α, ∊, α*, and ∊*. Each state corresponds to a rectangular region in the φ, ψ map. The conformation of an entire chain is then represented by a sequence of single-residue conformational states. The distinct “chain-states” in this representation correspond to multidimensional rectangular regions in the conformational space of the whole protein. A set of highly-probable chain-states can be predicted from the amino acid sequence using the pattern recognition procedure developed in the first two articles of this series. The importance-sampling minimization procedure of the present article is then used to explore the regions of conformational space corresponding to each of these chain-states. The importance-sampling procedure generates a number of random conformations within a particular multidimensional rectangular region, sampling most densely from the most probable, or “important,” sections of the φ, ψ map. All values of φ and ψ are allowed, but the less-probable values are sampled less often. To achieve this, the random values of φ and Φ are generated from bivariate gaussian distributions that are determined from known X-ray structures. Separate gaussian distributions are used for proline residues in the α and ∊ states, for glycine residues in the α, ∊, α*, and ∊* states, and for ordinary residues involved in 29 different tripeptide conformations. Energy minimization is then applied to the randomly-generated structures to optimize interactions and to improve packing. The final energy values are used to select the best structures. The importance-sampling minimization procedure is tested on the avian pancreatic polypeptide, using chain-states predicted from the amino acid sequence. The conformation having the lowest energy is very similar to the X-ray conformation.

    Notes: Formulas are derived for analytical first and second energy derivatives with respect to nuclear coordinates in molecular mechanics force fields employing lone-pair pseudoatoms. These derivatives may further be used for the calculation of normal modes and vibrational frequencies while properly accounting for the presence of pseudoatoms. The equations are applied using the MM2 force field to calculate the vibrational spectrum of methanol to illustrate the applicability of the method. The results are compared to both experiment and a numerical approximation in which small masses are assigned to the lone-pair pseudoatoms.
