    Notes: Summary The pyloric gland of Styela clava contains large glycogen deposits that are digested by treatment with alpha amylase and depleted by 15 days starvation. The deposits are surrounded by cytoplasmic regions containing smooth endoplasmic reticulum and mitochondria. The cells also have rough endoplasmic reticulum, Golgi cisterns, lysosomes, microvilli, cilia, and lateral infoldings of the plasma membrane. The fine structure of the pyloric cells and the position of tubules between the absorptive epithelium and general circulation suggest that the gland functions as the vertebrate liver in carbohydrate metabolism. The pyloric cells of Styela do not appear to be excretory in a ‘renal’ sense, since there is no infolding of the basal plasmalemma and mitochondria are usually associated only with the glycogen deposits. However, a hepatic-like excretory role is consistent with current findings. In light of the phylogenic affinities of vertebrates and ascidians, it is possible that the pyloric gland is homologous to the liver.

    Notes: Summary Electron microscopic studies on developing follicles of Locusta migratoria show the vitelline membrane to be composed of two ultrastructurally distinguishable components: The vitelline membrane bodies (VMBs) and, in addition, fine granular material, cementing the VMBs together. VMBs form first in the oocyte-near zone within the oocyte-follicle cell space. Subsequently, the second vitelline membrane substance is secreted between the VMBs through apical protrusions of the follicle cells. The possible origin of the VMBs is discussed. Yolk uptake in Locusta seems to occur predominantly by pinocytosis. During oocyte development the oocyte membrane is enlarged by numerous microvilli and folds. In addition pinocytotic vesicles are pinched off. It is supposed that the latter loose their coat and eventually transform into large proteid yolk spheres.

    Notes: Summary In order to localize pineal indoleamine synthesis at the ultrastructural level, an electron microscopic analysis was carried out on rabbit pineal tissue, cultured for several days in a medium containing the inhibitor p-chlorophenylalanine. Furthermore, electron microscopic autoradiography was applied to rabbit pineal tissue cultured in a medium containing the tritium labeled precursor 5-hydroxytryptophan. p-Chlorophenylalanine altered the ultrastructure of the mitochondria in the light pinealocytes only. Incubation with 3H-5-hydroxytryptophan caused a moderate and uniform labeling of the pinealocytes. These data suggest that the synthesis and storage of indoleamines in the rabbit pineal gland do not take place within the conventional membrane-limited cell organelles but more diffusely in the cytosol of the light pinealocytes. The spherical enlargement of the mitochondria in the light pinealocytes after culturing with p-chlorophenylalanine supports the biochemical findings of Hori et al. (1976) that the mitochondria are the carriers of the enzyme tryptophan 5-hydroxylase. The subsequent decarboxylation of 5-hydroxytryptophan to serotonin probably occurs free in the cytosol. The strong labeling of the noradrenergic nerve endings (in contrast to the cholinergic ones) after incubation with 3H-5-hydroxytryptophan demonstrates that the noradrenergic nerve endings in the rabbit pineal gland can, at least in vitro, selectively take up 5-hydroxytryptophan.

    Notes: Summary Coelomocytes of the earthworm, Lumbricus terrestris, were studied by transmission electron microscopy. Four morphological cell types are distinguishable: lymphocytic coelomocytes, granulocytic coelomocytes, eleocytes (chloragogen cells), and inclusion-containing coelomocytes. Within these major categories, several distinct cell types differ and may represent developmental stages. The two types of lymphocytic coelomocytes are small with central nuclei and scanty cytoplasms. Two types of granulocytic coelomocytes differ greatly in shape and content; both have small dark-staining granules that resemble lysosomes. Electrocytes, derived from chloragogen tissue, contain a variety of granules, inclusions and vacuoles. Inclusion-containing coelomocytes appear as two types which may be immature and mature forms. Although these cells resemble those that have been referred to as erythroid cells in other invertebrates, the large inclusion bodies are apparently unrelated to hemoglobin; they can undergo morphologic transformation and be extruded by exocytosis. This information on lymphocytic, granulocytic and inclusion-containing coelomocytes is crucial to understanding more about cellular immunity in the earthworm.

    Notes: Summary Just as in other cyprinids, three zones can be distinguished in the digestive tract of Barbus conchonius. A fat absorptive zone (65–75%), including the intestinal bulb, is followed by a protein absorptive zone (25–35%) and a small ion and water absorptive zone (〈5 %). The main characteristics of these zones are described. Four types of enteroendocrine cells can be distinguished between the intestinal epithelial cells. The number decreases in the caudal direction, and there are very few in the protein absorptive zone. All the enteroendocrine cells are argyrophilic and differ mainly in the size and shape of their secretory granules. Serotonin producing and hence argentaffin cells have not been found. Amine precursor uptake and decarboxylation (APUD) by the enteroendocrine cells of adult fishes has not been observed. The possible functions of the enteroendocrine cells are discussed. (Auto-)phagosomes, common in epithelial cells of the gut of B. conchonius, show similar staining characteristics as the enteroendocrine cells; their function is discussed.

    Notes: Abstract The flagellar root system of zoospores in two species ofChlorosarcinopsis (C. minuta andC. spec.) has been studied in detail. The biflagellate zoospores show a cruciate root system, two of the four microtubular roots containing two microtubules, the other two four microtubules. The flagellar apparatus is otherwise identical with that ofChlamydomonas reinhardi as described byRingo (1967). Evidence is presented that the genusChlamydomonas is characterized by a bilateral symmetric root system (4-2-4-2) rather than a system with four equally numbered roots (i.e. 4-4-4-4). It is suggested that a root system with four identical cruciate roots is not present in any biflagellate algal cell. The taxonomic significance of cruciate root systems in green algae is discussed refering to the identical root systems ofChlorosarcinopsis andChlamydomonas.

    Notes: Summary This is the first morphometric analysis of the cochlear spiral ganglion. Therefore the ganglion was subdivided in precisely defined compartments. We found that the normal, untreated spiral ganglion in guinea pigs contains 58% ganglion cells of the granular type, 26% satellite cells and Schwann cells, 8.6% extracellular space, 6.4% myelinated nerve fibers and 1% ganglion cells of the filamentous type. The granular ganglion cell consists in 83% out of cytoplasm and in 17% out of nuclei. The cytoplasm is containing 13% ribosomes, 12% mitochondria, 10% RER, 2.1% Golgi apparatus, 1.3% lysosomes. Based on these quantitative datas we expect to detect alterations of the normal structure earlier and more precisely under pathophysiological conditions as before.

    Notes: Summary After the electron microscopic examination of tissue samples of juvenile nasopharyngeal angiofibromas obtained from 9 male patients ranging in their age from 7–24 years the problems of cytogenesis and classification are discussed. Besides it is tried to correlate particular morphological findings to certain clinical phenomenons. The vascular component of juvenile nasopharyngeal angiofibromas shows a clear proliferation of the vascular wall cells. Particularly, proliferating pericytes, cells withous peculiar characteristics (“undifferentiated” cells) and cells in various stages of differentiation are to be emphasized. Obviously, vascular wall cells emigrate into the surrounding tissue and transform themselves into small fibroblasts. The second component of juvenile nasopharyngeal angiofibromas is represented by stromal fibroblasts with several cytological variations. Only activated “classical” fibroblasts and fibroblasts with histiocyte-like features reveal the nuclear pattern unique for these growths which is characterized by the combination of protrusions of nuclear membrane with formation of nuclear “blebs” and of dense intranuclear granules. Cells with these nuclear characteristics were considered as preexisting fibroblasts. Thus juvenile nasopharyngeal angiofibromas are formed by the proliferation of two tissue components, namely by the proliferation of vascular wall cells and stromal fibroblasts, and can be conceived as reactive hyperplasias. The swelling body-like and organoid appearance, cytological pecularities, characteristic topographic relations (localization and supplying vessels) and the sex-dependent occurrence speak for a tumor-like hyperplasia of a rudimentary organ unknown till now.

    Notes: Summary The cochlear spiral ganglion of rats consists of 61.8% granular ganglion cells, 22.4% satellite cells and Schwann cells, 9.3% extracellular space, 4.8% myelinated nerve fibers and 1.4% filamentous ganglion cells. The granular ganglion cells consist of 76% cytoplasmic components and 24% nuclei. Their cytoplasm contains 66.3% hyaloplasm, 11.4% mitochondria, 11% free ribosomes, 7.8% rough endoplasmic reticulum, 2.7% Golgi apparatus and 0.8% lysosomes. Comparing with morphometric data in guinea pigs, granular spiral ganglion cells in rats show some species dependent differences in their quantitative cytoarchitecture. They don't differ in their principal architectural pattern, especially in regard to the metabolism for energy and cell performance. The spiral ganglion cells of rats in contrast to those of guinea pigs do not show viral particles. Since it is impossible to evaluate with certainty the influence of viruses on the metabolism of infected cells, in our opinion rats are more suitable experimental animals for morphometric investigations of spiral ganglions.

    Notes: Summary The influence of two fixation buffers on the quantitative cytoarchitecture of the cochlear spiral ganglion in guinea pigs was evaluated morphometrically. After fixation with phosphate buffered 1.3% OsO4 granular spiral ganglion cells lost 45% of their average individual volume as compared to the volume after fixation with s-collidine buffered 1.3% OsO4. Using the two fixatives there was no significant difference of the volume proportion of cell nuclei, mitochondria, lysosomes and rough endoplasmic reticulum per unit volume cytoplasm of the granular spiral ganglion cells. The volume proportion of their ribosomes and their Golgi apparatus per unit volume cytoplasm doubled, the surface of the Golgi apparatus per unit volume cytoplasm increased 3.5fold after fixation with phosphate buffered OsO4. The volume density of the granular ganglion cells decreased by 30%, the volume density of the interganglionar space (= space between granular ganglion cells) showed an increase of 50% using phosphate buffer. Mostly the extracellular space was participating in this relative increase of the interganglionar space. As a result fixation of the spiral ganglion for morphometric studies should be performed using s-collidine buffered OsO4. The morphometric findings underline the presumption of semicompact myelin being a fixation artefact.

    Notes: Summary The fine structure of seminiferous tubules from 5 crabeater, 2 leopard and 2 Ross seals showed that during the nonbreeding season the tubules were essentially similar in possessing spermatogenic and Sertoli cells. However, the tubules of leopard and Ross seals had more primary and secondary spermatocytes and spermatids than the crabeater seals. In general, the tubules were devoid of spermatozoa. The spermatids showed stages of maturation such as Golgi phase of acrosome formation, acrosomal cap formation and condensation of nuclei. Some spermatids degenerated in tubules. Both maturing and degenerating spermatids were closely associated with Sertoli cells. Junctional complexes with plaques of filaments were observed between Sertoli cells and the spermatogenic cells. Sertoli cells, irregular and polygonal, contained highly convoluted nuclei, strands of rough endoplasmic reticulum, smooth endoplasmic reticulum, Golgi complexes, small mitochondria, variable amounts of lipid droplets, lysosomes, lipofuscin granules and highly plicated plasma membranes. In brief, the spermatogenic activity had practically ceased in the testes and the animals probably secreted low levels of testosterone during the nonbreeding season.

    Notes: Summary The ultrastructure of the thyroid gland of the domestic fowl has been investigated and found to be similar to that of mammals. The differences were found at subcellular level in the distribution of the “dark bodies” which were mainly apical and in the sizes of primary lysosomes. These were found to range from 100 to 500 nm in diameter. All organelles described in mammals as being concerned with the production of thyroglobulin and the two hormones thyroxine and triiodothyronine were found to be present.

    Notes: Summary An ultrastructural analysis of the rat lateral hypothalamic area (LHA) was undertaken in order to provide an initial step in the characterization of this complex area which appears to participate in a number of important neural functions. The organization of the normal tuberal LHA was compared to the area following acute and chronic denervating lesions. In the normal animal, the principal features of the LHA are the presence of lateral hypothalamic neurons, a major sagittal pathway (the medial forebrain bundle, MFB) and the interposed neuropil richly populated by a variety of synaptic terminal types. Alterations in the synaptic organization of the LHA following rostral and caudal MFB lesions were most pronounced in animals with acute and chronic caudal lesions. A 10% reduction of synaptic terminals containing 800–1000 Å diameter dense core vesicles and a 10% increase in terminals containing lucent core vesicles was observed in animals with caudal lesions while no significant redistribution of synaptic terminal types occurred with rostral lesions. The preliminary degeneration experiments indicate that identification of the numerous and diverse afferents to the LHA neuropil may be aided by this method but that a detailed and systematic ultrastructural analysis will be required to identify sources of input with certainty.

    Notes: Summary Nurse cells develop from diploid cells in the testis. Each cell undergoes a reduction division which leaves the nucleus with half the volume of a normal diploid cell. They send out pseudopodia which form desmosomelike junctions with developing spermatids. The nurse cells detach from the testicular wall, their nuclei degenerate and secretion droplets form in the cytoplasm. The pseudopodia are drawn in as the cytoplasmic secretions swell and the nurse cell becomes spherical. The eupyrene sperm become grouped unilaterally and at this stage are attached to the nurse cell by only the tips of their acrosomes. At maturity the nurse cells with their clumps of attached eupyrene sperm (spermatozeugmata) are released from the testis via ducts into the seminal vesicles, where they are stored prior to copulation. Nurse cells serve similar functions to those of apyrene sperm which are common among the Molluscs. We believe that the nurse cell and apyrene sperm are homologous.

    Notes: Summary Late stages of oogenesis in Acerentomon gallicum Jonescu have been studied by means of light and electron microscopy. Each of the two ovaries of this species consists of a single panoistic ovariole. Late previtellogenic and early vitellogenic oocytes are enclosed in an electron opaque layer, the so-called primary sheath. The precursors for this sheath are most likely synthesized by follicle cells. The yolk develops through autosynthesis, with free ribosomes, dictyosomes and lamellar bodies being involved in the process. Mature yolk spheres contain proteins and polysaccharides. Besides the organelles that take part in vitellogenesis, mitochondria and cisternal stacks of the rough endoplasmic reticulum occur in the ooplasm.

    Notes: Summary The structure of the neurohypophysis of Poecilia latipinna (green molly, sailfin molly) was studied with the electron microscope. Profile diameters of neurosecretory granules in the non-myelinated neurohypophysial nerve fibres were measured and mathematically corrected for error due to section thickness. Six different types of nerve fibres could be distinguished by statistical classification of their granules and by other ultrastructural features. One fibre-type (type B) contained granules with a mean diameter of 85 nm, and the other five types (types Ala, Alb, A2, A3 and A4) all contained granules with mean diameters greater than 100 nm. Synaptic contacts were observed between type B fibres and all the adenohypophysial cell-types, although in the case of the ACTH cells the synapses were separated from the cell membrane by a continuous double basement membrane. Type A fibres were observed to contact the cells of the proximal pars distalis and pars intermedia, but did not form synapses. However, synapses occurred between type A fibres and pituicytes, and between type A fibres and the pericapillary basement membrane in the interior of the neurohypophysis. The possible roles of the different types of nerve fibres in controlling the adenohypophysial cells are discussed in the context of evidence from other teleosts.

    Notes: Summary Topical application of 5 μg of a juvenile hormone analogue (JHA), ZR-615, to female callow adults of Ips paraconfusus induced degeneration of the dorsoventral flight muscles. Within 24h after JHA-treatment the diameter of the myofibrils was reduced to almost half due to the lysis of the peripheral myofilaments. Mitochondria showed conspicuous degenerative changes like swelling, dissolution of the matrix or presence in the matrix of dense filamentous material or myelin-like figures. Degeneration of the mitochondria seemed to take place inside isolation membranes derived from sarcoplasmic reticulum. A number of granular osmiophilic bodies appeared in the sarcoplasm. Three days after JHA-treatment the muscles were very thin and sheath-like. Most of the mitochondria had already degenerated. The dense sarcoplasm contained numerous crystalline bodies. The granular dense bodies were also more frequent. The myofibrils were comprised of only occasional small bundles of myofilaments. The tubules of the T system enclosed an amorphous material. The nuclei and the tracheal system remained intact but they were crowded due to the decreased volume of the muscle. In some specimens degeneration of the myofibrils and mitochondria was completed by the third day. No sign of degeneration was observed in the flight muscles of acetone treated control insects.

    Notes: Summary The mitochondria-rich (chloride) cells have been found to be present in the gill epithelia of four species of stenohaline fresh water teleosts. The cytoplasm of these chloride cells contains an extensive network of cytoplasmic tubules which communicate with intercellular spaces bordering the lateral and basal cell surfaces. Numerous vesicles with fairly electron-dense interiors are also present in the apical cytoplasm of chloride cells. The apical surface of a chloride cell forms an apical pit, but the lumen of the pit does not appear to be in continuity with the interior of the apical vesicles and tubules inside the cell. When Carassius auratus were kept in 100, 200, 300, and 400 mOsm-diluted sea water for a month, no appreciable changes occurred in the number and fine structure of the chloride cells, except for a dilation of the apical vesicles and a slight decrease in diameter of the cytoplasmic tubules in these cells in the fishes kept in 300 and 400 mOsm. These results suggest that chloride cells may be a rather common occurrence in the gill epithelia of stenohaline fresh water teleosts, and may function in ion-transport in these fishes in fresh water environments.

    Notes: Summary The testes of adult male Syrian hamsters underwent involution within six weeks after optic enucleation. The diameter of the seminiferous tubules was 39% less than controls. Sertoli cells, spermatogonia, and primary spermatocytes were still present, but all steps of spermatids were completely absent from the involuted testes. Lipid droplets filled the Sertoli cell cytoplasm and often encroached upon the nucleus. Sertoli cells had sparse mitochondria and smooth endoplasmic reticulum, but Golgi cisternae were abundant. Typical SertoliSertoli junctions attached contiguous Sertoli cells. With lanthanum tracers it was demonstrated that these junctions were impenetrable; therefore, the bloodtestis barrier was deemed intact. Irregularly shaped protrusions often arose from the peritubular tissue and extended inward toward the seminiferous epithelium, often displacing the cytoplasm of the Sertoli cells and spermatogonia. The core of these protrusions consisted of irregular extensions of myoid cell cytoplasm surrounded by the myoid cells' basal lamina. External to the myoid cell basal lamina were bundles of collagen filaments with the basal lamina of the seminiferous epithelium forming the outermost layer of these protrusions. The apices of the Sertoli cells gave rise to numerous leaf-like processes that extended into and obliterated the lumen of the tubules. The Sertoli cell basal cytoplasm often contained phagocytized degenerating germ cells that appeared to give rise to the lipid droplets that filled the Sertoli cell cytoplasm. Acid phosphatase rich lysosome-like organelles were seen fusing with the degenerating germ cells and lipid droplets. The degenerating germ cells also were shown to contain acid phosphatase activity.

Das Epithel ist einschichtig. Regenerationszellen findet man im A- und P-part. “Helle Zellen” (“clear cells”) mit möglicherweise endokriner Funktion kommen lediglich im P-part vor. In vielen strukturellen Parametern gleichen sich die Epithelzellen der verschiedenen Mückenarten. Keine wesentlichen Unterschiede bestehen zwischen Anopheles gambiae (“Wildfänge”) und A. stephensi (Laborstamm). Die Zellstruktur läßt vermuten, daß in beiden Mitteldarmabschnitten sowohl Synthese und Sekretionsvorgänge als auch Resorptionsund Transportprozesse ablaufen können. Daneben enthält der A-part mehr Mikrovilli, einen größeren Anteil an glattem endoplasmatischem Retikulum und an Membranen des basalen Labyrinths. Der P-part ist gekennzeichnet durch einen höheren Anteil an rauhem endoplasmatischem Retikulum (rer) und an Mitochondrien. Als Arbeitshypothese wurde dem A-part aller untersuchten Culiciden die Bildung und Sekretion von Mucus-ähnlichen Substanzen und die Resorption von Zucker zugeordnet. Dementsprechend sollte im P-part sowohl Synthese und Sekretion der peritrophen Membran und der Enzyme für die Blutverdauung als auch zu wesentlichen Teilen Resorption, Transport und temporäre Speicherung von Blutabbauprodukten stattfinden. Strukturelle Unterschiede zwischen den untersuchten Mückenarten bestehen zur Hauptsache im Folgenden: a) weniger Oberflächendichte des rer im P-part von Culex und Anopheles verglichen mit Aedes; erst während und nach der Verdauung des 1. Blutmahls mehr rer in P als in A, b) eine zusätzliche rer Cisterne bildet bei Culex einen “Kernhof”, c) apicale Granula im P-part von Anopheles, d) Reserveprodukte bei Aedes als Lipide gespeichert, bei Anopheles als Kohlenhydrate und Lipide, e) keine Maculae adhaerentes im P-part von Anopheles.
    Notes: Summary The midgut of female Anophelinae and Culicinae comprises a narrow anterior (A-part) and a wide posterior part (P-part, stomach). Both are lined by a singlelayered epithelium, and in both parts regenerative cells are found between the epithelial cells. “Clear cells”, possibly with an endocrine function, are only found in the P-part. The epithelial cells of the mosquito species investigated display close ultrastructural similarities. No significant differences are found between Anopheles gambiae and A. stephensi which differ markedly in origin and habitat. In all species investigated the cellular organization suggests that both midgut parts are able to carry out processes of synthesis and secretion as well as of absorption and transport. On the other hand, in the A-part more microvilli, a higher density of membranes of the smooth endoplasmic reticulum and a tendency for more membranes of the basal labyrinth are seen by comparison with the P-part. The P-part is characterized by more rough endoplasmic reticulum (rer) and mitochondria. As a working hypothesis it is proposed that absorption of sugar-containing solutions and the formation of mucus-like material occurs in A. The P-part is postulated to be responsible for synthesis and release of precursors of the peritrophic membrane and digestive enzymes as well as for the main absorption, transport and temporary storage of metabolites resulting from blood digestion. Ultrastructural differences between mosquito species mainly consist of the following: a) less surface density of rer in the P-part of Culex and Anopheles as compared with Aedes; more rer in P than in A of Anopheles only present from 1st blood meal onwards, b) an additional rer cisterna in Culex forms a “halo” around the nucleus, c) apical granules in the P-part of Anopheles, d) storage deposits — in Aedes primarily as lipids — in Anopheles as carbohydrates and lipids, e) no maculae adhaerentes in the P-part of Anopheles.

    Notes: Summary The distribution of ionic lanthanum (La3+) was studied after perfusion of the isolated rat heart with a buffered salt solution containing lanthanum chloride. We have demonstrated that the localization of lanthanum differed from that shown in previous studies in which hearts were exposed to ionic or colloidal lanthanum during or after fixation. La3+ passed rapidly through the intercellular cleft between endothelial cells. There was relatively little transport by micropinocytosis and no evidence of transendothelial passage through the cytoplasm. Perfusion with a solution containing 1 to 15 mM LaCl3 resulted in localization of La3+ on the entire cytoplasmic membrane system of muscle cells, i.e.: the non-specialized parts of the muscle cell membranes were stained by La3+ as well as the membranes of micropinocytotic vesicles, of the T-system and of the intercalated disc. The cytoplasmic leaflets of gap junctions were especially stained. However, the cell membranes of non-muscle cells were free of lanthanum precipitates. Freeze-fracturing of muscle cells showed concomitant alteration in the distribution of membrane-associated particles. The gap junction membranes also showed changes in particle aggregation. Intracellular binding of La3+ was restricted to muscle cells. Lanthanum was found in the cell nucleus associated with heterochromatin, in the nucleolus, and along the nuclear membrane. The myofibrils were stained mainly over the I-band.

    Notes: Summary After gentle trypsinization, the pseudostratified columnar Müllerian epithelium that lines the uterine cervix of newborn mice could be separated from the enclosing stromal tissue. Pure epithelial tubes explanted in vitro and were allowed to grow in a standard medium for 3–4 days forming a confluent colony of rather closely-fitting cells. The cell sheet was studied by a preparatory technique that allows examination of a large number of cells with preserved intercellular spatial orientation. Attempts were made to identify cultured cells according to the morphology of cell types in the cervicovaginal epithelium in vivo. Electron micrographs revealed that, close to the explant, the cultured cell sheet exhibited several features similar to the Müllerian epithelium in vivo. Outside these central areas of the colony was a broad transitional zone consisting of thin platelike cells distinguished by an abundance of microfilaments. At the periphery of the colonies, bulky cells possessing microvilli and a vacuolated cytoplasm tended to overlap adjoining platelike cells. These bulky cells had a morphology resembling that of the superficial cells seen in the upper vagina and common cervical canal of immature and diestrous animals. The epithelial development in the cultures apparently simulated the transformation in vivo from a pseudostratified Müllerian epithelium in the newborn to a stratified epithelium resembling that of the uppermost vagina and common cervical canal of immature animals. Judged by morphological and cytochemical criteria, the Müllerian cells in the outgrowth obviously had many changed features. It thus seems questionable whether the cells grown in vitro are comparable with the corresponding cells in vivo when used for experiments requiring the controlled conditions of the culture environment.

    Notes: Summary Oögenesis in the oviparous marine teleost, Blennius pholis L., is examined. Eleven developmental stages are identified by ultrastructural observations when changes in the distributions of the organelles and inclusions are described. An exogenous source for the protein yolk precursors is indicated, but less clear is the endogenous contribution. Changes in the follicle epithelium are described together with the formation of the zona which is considered to be follicular in origin. Two types of follicle cell are distinguished and these probably function differently in the process of zona formation. The zona becomes divided into the externa and interna, the latter probably resulting from the chemical ordering by disulphide bonding of the proteinaceous material of the former.

    Notes: Summary The respiratory epithelium in the lungs of the tortoise (Testudo graeca) has been studied by electron microscopy. The epithelium consists of a mosaic of two different cell types (here called “pneumonocytes”). Type I pneumonocytes are roughly squamous and possess attenuated flanges of cytoplasm which extend over the septal capillaries. Localized cytoplasmic expansions are often present near the periphery of these flanges. Most of the organelles are concentrated in the perinuclear region; the most prominent of these are the mitochondria and osmiophilic inclusions. In contrast, type II pneumonocytes are cuboidal and are richly endowed with organelles including large Golgi complexes, extensive endoplasmic reticulum and numerous inclusion bodies. The morphological evidence suggests that type I pneumonocytes are involved in the secretion of osmiophilic material (presumed to be pulmonary surfactant) and in maintaining the integrity of the air-blood barrier. Type II pneumonocytes appear to be concerned solely with the production of surfactant.

    Notes: Summary Electron microscopical observations show that the cones in the retina of the diurnal Poecilia reticulata shed their membranous outer segment disks. This occurs at the side of the disk which is open to the extracellular space. Shedding is observed in single and twin cones and occurs at any level of the outer segment. The disks are not discarded in packages or as single disks, but are shed in small vesicular portions. This mode of ‘disk shedding’ may explain why in cone outer segments radioactively labelled replacement protein is diffusely distributed.

    Notes: Summary The mammalian pineal is thought to produce an antigonadotropic principle under conditions of reduced photoperiod, constant darkness or blinding by optic enucleation. A number of previous studies on mammalian pineals have suggested that the dense-cored vesicles present in pinealocytes may represent morphological evidence of secretory activity. In the present study the ultrastructure of pinealocytes was studied in adult Charles River CD-1 mice blinded by optical enucleation. By one month following optic enucleation the mean number of dense-cored vesicles in the cytoplasm of pinealocytes adjacent to pericapillary spaces had significantly decreased by 55% when compared with intact controls, and remained at this low level at two months and six months. A relative increase in the proportion of large agranular vesicles and an increased number of large, irregular vacuoles was observed also in the pinealocytic polar processes of blinded mice. When compared to control mice the pinealocytic Golgi regions appeared to be hypertrophied in blinded mice. The apparent stimulation of pinealocytic organelles coupled with the observed decrease in dense-cored vesicles suggest an increased synthesis and release of secretory product.

Die Cornea ist zwar außerordentlich dünn, doch der Kristallkegel ist gut entwickelt. Es fehlt eine klare pigmentfreie Zone zwischen dioptrischem Apparat und Rhabdom. Vereinzelte Pigmentkörner werden lediglich innerhalb der Basallamina angetroffen. Das Rhabdom ist massiv und nimmt rund 50 % des Augenvolumens ein. Es besteht aus rechtwinklig angeordneten Mikrovilli, die einen Durchmesser von 72 nm aufweisen. Querschnitte zeigen die dichte Packung der Rhabdome. Interrhabdomale Lüken für Retinula-Zellplasma sind kaum vorhanden. Nach einer einstündigen Helladaptation wurden keine Feinstrukturveränderungen an den Mikrovilli beobachtet. In allen Retinulazellen traten jedoch in der Nähe der Basallamina Vesikel verschiedenster Art auf. Die sieben Axone eines Ommatidiums verlassen das Auge als gemeinsames Bündel, doch unterhalb der Basallamina vereinigen sich oft mehrere Bündel zu größeren Einheiten.
    Notes: Summary The eye of the deep-sea penaeid shrimp Gennadas consists of approximately 700 square ommatidia with a side length of 15 μn. It is hemispherical in shape and is located at the end of a 1.5 mm long eye stalk. The cornea is extremely thin, but the crystalline cone is well-developed. A clear zone between dioptric structures and the rhabdom layer is absent. A few pigment granules are found within the basement membrane; otherwise they, too, are absent from the eye of Gennadas. The rhabdom is massive and occupies 50 % of the eye. It consists of orthogonally oriented microvilli (the latter measuring 0.07 μm in diameter) and is 75 μm long. In cross sections adjacent rhabdoms, all approximately 8 μm in diameter, form an almost continuous sheet and leave little space for retinula cell cytoplasm. In spite of a one h exposure to light, rhabdom microvilli show no disintegration or disruption of membranes. Vesicles of various kinds, however, are present in all seven retinula cells near the basement membrane. Bundles of seven axons penetrate the basement membrane. On their way to the lamina they often combine and form larger aggregations.

    Notes: Summary Occlusal intradentinal cavities, prepared in normal human premolars and third molars to be extracted for orthodontic reasons, were filled for 7 to 11 days with gutta percha. A superficial pulpitis with localized small abscesses developed in the pulp chamber. Under local anesthesia, 0.2 to 0.3 cc of sterile colloidal carbon was injected in the pulp horn and the teeth were extracted 1 to 3 h later. Lymphatic capillaries could thus be identified in the pulpal tissues. They were characterized by a thin endothelium with occasional large intercellular clefts, absence or incompleteness of basement membrane, absence of pericytes, absence of luminal red blood cells, and presence of a filamentous material between the endothelium and the surrounding collagen fibrils. Moreover, some structural variations were observed.

    Notes: Summary Two types of mitochondria-rich cells were found in the interplatelet areas of the gills of sexually mature, male river lampreys. Type 1 cells (previously referred to as “male glandular cells”) showed some ultrastructural characteristics in common with ion-transporting cells but were readily distinguished by large lipid structures with electron-lucent centres. Type 2 cells were found to be identical to the presumed ion-uptake cells found during other stenohaline freshwater phases of the animal's life history. In the sexually mature female, only Type 2 cells were positively identified. This sexual dimorphism in gill structure is discussed in relation to the possible functions of the lamprey gill, with particular reference to ionic regulation.

    Notes: Summary In this report we describe the time of appearance and ultrastructural features of enteroendocrine cells (EECs) in the human fetal small intestine (SB) between 9 and 22 weeks gestation. Thirteen distinctive EECs were identified in fetal SB. Two of these, not found in normal adult SB, appeared within the stratified epithelium of the proximal SB at 9–10 weeks. They were arbitrarily termed “primitive” and “precursor” cells. As in all fetal EECs, the pale cytoplasm of the “primitive” cell contains a distinctive population of secretory granules (SGs). Primitive cell SGs average 200–330 nm; some have dense cores with lucent halos while others are filled with a homogeneous dense or flocculent material. The SGs of the “precursor” cells are larger, averaging up to 1 μm in diameter and their contents vary in electron density. A third group of cells not described in normal adult SB was arbitrarily termed “transitional” cells. These have two populations of SGs; one resembles the SGs of the “precursor” cells, and the other resembles the SGs of some of the specific adult type EECs. Transitional EC, S, I and G cells are seen. In addition, mature appearing EC, S, G, I, L, D, and D1 cells were identified by 12 weeks of gestation. The “primitive”, “precursor”, and “transitional” cells may represent sequential developmental precursors of adult type EECs.

    Notes: Summary The development of the nerve supply of the pituitary pars intermedia (PI) of C3H mice was studied by electron microscopy. Nerve fibres and terminal structures, most probably adrenergic, first appear in the newborn. The adult innervation pattern is achieved by the end of the first postnatal week. In the adult animal two types of nerve terminals were distinguished; type A (peptidergic or neurosecretory) and type B (adrenergic). The peptidergic fibres were scarce and exhibited no synapse-like contacts. It is suggested that they are of secondary importance in a direct nervous hypothalamic control of PI function. Type B terminals were found throughout the PI. They formed synapse-like contacts with the glandular cells, indicating that the primary innervation is exerted by adrenergic neurons. An autonomous differentiation of the glandular cells and in the adult a combined direct nervous and neurohumoral control of PI function is suggested.

    Notes: Summary An electron microscopic survey of pinealocytes from normal rats revealed a highly organized arrangement of cytoplasmic tubules. Such tubules had been previously observed in normal rats (Lin, 1967) and in rats after melatonin administration or two weeks exposure to darkness (Freire and Cardinali, 1975). In a later publication the presence of the tubules was attributed to experimental manipulation resulting in infertility (Gusek, 1976). The present study resolves the discrepancy in the literature by establishing that the tubular organelle does indeed occur in untreated male rats, but rather rarely.

    Notes: Summary The sheep parotid is a compound tubular gland; its ultrastructure reflects the function of this gland to secrete large amounts of fluid with very little protein. The cells of the secretory tubules possess extensively folded lateral plasma membranes and a fairly large number of mitochondria. Rapid equilibration of water across the epithelium is assured by the close proximity over large areas of intercellular spaces and the wide secretion canaliculi. Numerous long microvilli extend into the latter. Although secretion granules may be quite numerous, there is evidence that many of these granules are not discharged but undergo degradation by lysosomal enzymes. The intercalated ducts are often dilated but excessive distension is probably prevented by bundles of microfilaments in their epithelial cells.

    Notes: Summary The ultrastructure of the tanycyte ependyma in male 160–180 g Wistar albino rats was studied under normal conditions and in experiments involving long-term suppression of ACTH secretion and its long-term stimulation. The former was accomplished by daily (for 8 days) intraperitoneal administration of dexamethasone phosphate at low (5 μg/100 g) and high (100 μg/100 g) concentrations. The effectiveness of suppression of the hypothalamic-hypophyseal-adrenal system in the experimental animals was judged by their reaction to two-minute ether stress (determination of plasma corticosterone) and by the results of measurement of the adrenal weights. Stimulation of ACTH secretion was achieved by bilateral adrenalectomy; the animals were examined on days 8, 10, 14, and 22 following the operation. The results obtained were in agreement with the previously established fact that there is a negative correlation between tanycyte activity and hypophyseal adrenocorticotrophic function (Akmayev and Fidelina, 1974). They also testified to the predominant involvement of the median eminence tanycyte ependyma (beta-tanycytes according to the authors' nomenclature) in these relationships. It is supposed that these correlations are regulated by a feedback mechanism and attest to the involvement of beta-tanycytes in the inhibiting control of hypophyseal adrenocorticotrophic function. The mechanism of this control may be explained alternatively: either the tanycytes transport ACTH-suppressing substances (catecholamines, corticosteroids, ACTH) from the CSF to the hypophyseal portal system or they themselves secrete substances possessing ACTH-suppressive activity. The authors distinguish several types of vesicles in the beta-tanycytes, the number of which changed with experimentally induced shifts in hypophyseal adrenocorticotrophic function. These vesicles are discussed in connection with the transport and secretory activity of the tanycytes and are considered to be a possible substrate of the hypothalamic inhibiting effect on ACTH secretion.

    Notes: Summary Long-term treatment of female rats with the anabolic steroid hormone Methandrostenolone results in a conspicuous increase of intermediate sized, nonmyofibrillar filaments in muscle cells of the left cardiac ventricle, as revealed by electron microscopy. These filaments, measuring 70–110 Å in diameter, form a characteristic network at the Z-level of the sarcomere, either encircling or penetrating the Z-bands, and appear to insert into the nuclear membrane.     Notes: Summary Ultrastructural studies on the Malpighian tubules of Glomeris marginata (Villers) reveal considerable morphological differences between the upper, fluid secreting, segment, and the lower segment which is at present of unknown function. Previous reports have shown that the upper tubule has a high permeability to compounds of high molecular weight. This may be accounted for by the fact that the epithelium shows very extensive intercellular spaces which are linked directly to junctions apparently specialised to provide a low resistance extracellular pathway between the haemocoel and the tubule lumen. Histochemical studies on the localisation of phosphatase enzymes reveal intracellular vesicles with acid phosphatase activity. The basal labyrinth of the lower tubule exhibits considerable alkaline phosphatase activity which is apparently identical in location to the enzyme revealed by two different ATPase localisation techniques.

    Notes: Summary The ultrastructure of the musculature of Lymnaea stagnalis was studied. Each of the six muscle systems of the body wall, previously distinguished in an anatomical study, has its own type of smooth muscle, characterised by the size and number of the myofilaments, number of mitochondria and distribution of smooth endoplasmic reticulum. The visceral musculature comprises both smooth and striated muscle. Cross-striated muscle is found in the heart and proximal aorta, obliquely striated muscle in the buccal mass, gizzard and vas deferens. Myofibroblasts and myoendothelial cells were also distinguished. On the basis of the observations it is concluded that striated muscles in L. stagnalis contract and relax more rapidly and have a higher endurance than smooth muscles, but that the latter can contract over a wider range. Among smooth muscles the head retractor muscle contracts most rapidly, the shell muscle is most powerful and the diagonal muscle is slow to contract but has a relatively high endurance. The latter muscle, together with the horizontal foot muscle, plays a major role in maintenance of the hydrostatic skeleton. A model for the organisation of the smooth muscles is deduced from the ultrastructural observations. It implies that the myosin-paramyosin filaments change their actin filament partners during contraction. This agrees with a model deduced for other smooth muscles on the basis of physiological experiments and X-ray diffraction analysis.

    Notes: Summary The turnover of newly formed proteins in the aortic myocyte was studied by means of quantitative light and electron microscopic radioautography. Adult and young mice were sacrificed at long intervals between 4 h and 108 days after one injection of tritiated leucine. The results showed that secretory activity, important in the post-natal myocyte, persists noticeably in the mature myocyte. The similarity in sedentary protein turnover, noted in both the thoracic and abdominal aortic segments of the adult and young animals, seems to be expressed by the same labelling patterns of the myoplasm. Turnover of exportable protein, which varies with age and anatomical level, appears to be related to the higher rate of elastin synthesis in the thoracic aorta of the young animal.

    Notes: Summary The follicle cells, nurse cells and germinal epithelia, which are closely associated with the oocyte of Agriolimax reticulatus (Müller) during its development in the ovotestis, have been studied using light and electron microscopy. The various secretory, digestive and phagocytic activities of these cells have also been investigated using electron cytochemical tests for oxidisable polysaccharide, acid phosphatase and electron-opaque tracer molecules. The oocyte lies initially between the germinal epithelia and a layer of nurse cells but, as oocyte vitellogenesis proceeds, it becomes encapsulated by a layer of follicle cells. Both the follicle and the nurse cells are active in secretion and digestion and contain Golgi apparatus, granular endoplasmic reticulum and acid phosphatase-rich digestive vacuoles. The significance of these activities is discussed in relation to oocyte vitellogenesis, secondary envelope formation and the digestion and recycling of cellular material.

    Notes: Summary The ultrastructure of the compound eyes of five species of mysids (Crustacea: Mysidacea), Praunus flexuosus, Siriella norvegica, Mysidopsis gibbosa, Neomysis integer and Erythrops serrata, is described. The ommatidia are constructed on a common plan, but there are considerable differences in detail. Common features include the arrangement of the cornea, crystalline cone and the basement membrane. The number of retinular cells differ: in Neomysis and Erythrops there are seven, whereas in the other species there are eight, the eighth cell forming a distal rhabdom, which consequently is lacking in the ommatidia of Neomysis and Erythrops. Another difference is the epirhabdom, which is lacking in Erythrops, but present in the other species. The epirhabdom is an extracellular structure, probably serving as a dioptric element. The pigment arrangement is similar in the first four species. The pigment shield consists of the distal pigment, distal reflecting pigment, proximal pigment (in the retinular cells) and the proximal reflecting pigment. The distal and proximal pigments are dark screening pigments. In addition to these, there are basal red pigment cells, which are mainly located below the basement membrane. In Erythrops there are three kinds of pigment cells: distal pigment cells, distal reflecting pigment cells and basal red pigment cells. Besides the basal red pigment cells, the distal pigment cells contain red pigment granules.

    Notes: Summary Bands of electron-dense material beneath the cell membrane of smooth muscle cells of the guinea-pig taenia coli provide attachment to thin myofilaments and to intermediate (10 nm) filaments; about 50% of the cell membrane is occupied by dense bands in muscle cells transversely sectioned at the level of their nucleus, and between 50 and 100% in smaller cell profiles nearer the cell's ends. In addition to the known cell-to-cell junctions (intermediate contacts), more complex apparatuses anchor muscle cells together, either end-to-end or end-to-side or side-to-side. They consist of elaborate folds, invaginations and protrusions accompanied by large amounts of basal lamina material. In the end-to-end anchoring apparatuses numerous finger-like and laminar processes from the two cells interdigitate. Other muscle cells have a star-shaped profile in the last few microns of their length, or show longitudinal invaginations occupied by a thickened basal lamina and occasionally by collagen fibrils. The septa of connective tissue extend only for a few hundred microns along the length of the taenia. In taeniae fixed in condition of mild stretch the muscle cells form an angle of about 5° with the septa. In muscles fixed during isotonic contraction the angle increases to about 20–22°, and in longitudinal sections the muscle cells appear arranged in a herring-bone pattern. The collagen concentration in the taenia coli is 4–6 times greater that in skeletal and cardiac muscles. These various structures are discussed in terms of their possible role in the mechanism of force transmission.

    Notes: Summary Male rats were divided in two experimental groups. In group I two partes distales of the hypophysis were grafted under the kidney capsule and in group II two complete hypophyses were transplanted. Animals were killed 5 to 22 months after the operation. The grafted tissue was excised and processed for light and electron microscopy. The transplanted pars distalis tissue showed a well developed vascularisation in contrast to the pars intermedia which appeared poorly vascularised. Six different cell types were observed in grafted pars distalis. They correspond to the different types of cells found in the rat pars distalis “in situ”. The predominant cell type in the graft displayed all the morphological characteristics of stimulated prolactotrophs. Pars intermedia cells appeared hypertrophied resembling the MSH cells under stimulation. Two types of syncytial formations were frequently seen. One of them appeared to originate from prolactotrophs and the other from MSH cells. Bodian impregnated fibres and structures resembling either growth cones of axons or typical nerve endings were observed in the pars intermedia of long-term grafted hypophyses. Pituicytes remained as isolated clusters of cells. Canaliculi lined by two or more pituicytes were observed. Saccular formations resembling the hypophyseal cleft appeared in all grafts studied. The present findings suggest that in the male rat the chronically grafted pituitary gland is capable of synthesising most or all the hormones which are known to be produced by the gland “in situ”. Furthermore, prolactin and MSH seem to be the predominant secretion of the transplanted pituitary.

Das Epithel beider Arten ist einschichtig. Die Epithelzellen sind durch eine mäßige sekretorische Aktivität, eine wechselnde Menge von apikalen PAS-positiven Grana und durch apikal und basal gelegene Exo- oder Endocytosevesikel gekennzeichnet. Benachbarte Zellen werden durch “junctional complexes” zusammengehalten. Ihre lateralen Zellmembranen sind stark gewunden und ihre Oberfläche durch Ausläufer vergrößert, eine Organisation, die an ein Transportepithel denken läßt. Vereinzelt vorkommende “helle” Zellen zeichnen sich durch sehr variable cytoplasmatische Einschlüsse aus und sind niemals mit benachbarten Zellen verbunden; möglicherweise sind sie amöboid beweglich. Im gesamten Epithel, mit Ausnahme eines kleinen cranial gelegenen Abschnittes im Uterus von S. atra, sind, abgesehen von einer wahrscheinlich größeren Anzahl abgeflachter Zellen bei trächtigen Weibchen, keine signifikanten morphologischen Veränderungen feststellbar, die in Zusammenhang mit der Trächtigkeit gebracht werden können. Die Ergebnisse werden im Hinblick auf eine mögliche Versorgung der heranwachsenden Jungen von seiten der Mutter diskutiert.
    Notes: Summary The uterine epithelium of the viviparous Salamandra atra and the ovoviviparous Salamandra salamandra was studied in non pregnant and ovulating females and in females during different stages of pregnancy. The epithelium of both species is organized in a monolayer. The epithelial cells are characterized by a moderate secretory activity, a variable amount of apical granules which include PAS-positive material and by some apical and basal exo- or endocytotic vesicles. Adjacent cells are joined by junctional complexes. The lateral surfaces form a tortuous boundary with adjoining cells which suggest that the epithelium is involved in transport. Sporadic light cells possess highly variable cytoplasmic inclusions and are not joined with neighbouring cells. Possibly they represent migratory cells. The entire epithelium, except for a small cranial portion of the uterus in S. atra, undergoes no remarkable morphological changes during the different physiological stages examined except that flattened cells seem to be more numerous in pregnant females. The results are discussed with regard to the possible supply of the developing young by the mother.

    Notes: Summary Using the enzyme peroxidase and ionic lanthanum as tracers, paracellular uptake has been demonstrated in the foot of the slug Agriolimax reticulatus (Mü). Both tracers appeared to pass between adjacent foot epithelial cells and were demonstrated in the zonula adhaerens, the septate desmosomes, and the intercellular spaces which occur beneath the septate junctions. Ferritin, a somewhat larger tracer, was excluded from all these sites. Ionic lanthanum was not normally pinocytosed in short incubation times. The epithelial cells could be induced to endocytose this marker, however, when combined with a variety of proteins. The implications these findings have on the uptake of molluscicides is discussed.

    Notes: Summary A casting technique has been employed to display in three dimensions, the lymphatic microcirculation within the human lymph node. The casting compound filled the marginal sinus, and diffusely permeated the cortical lymphoid parenchyma. However, deep within the lymph node in the medullary region, the medium remained within the limits of the sinus walls. The casts showed well-defined channels appearing similar to vessels. These converged into larger vessels, which drained into efferent lymphatics leaving the node at the hilus. Electron microscopic examination showed that the outer wall of the marginal sinus and the trabecular side of trabecular sinuses had an intact, continuous endothelium with a basement membrane. However, gaps were present in the inner wall of the marginal sinus, as well as in the parenchymal wall of the trabecular sinus. In the medulla, the sinuses were lined by endothelial cells which appeared similar to macrophages. The sinus lining was incomplete and possessed numerous perforations. These observations indicated that sinus walls adjacent to connective tissue served as a barrier to cell movement, but those adjacent to a large lymphoid cell population had gaps, with cells in apparent transit between sinus lumen and parenchyma.

    Notes: Summary The duck interrenal cell possesses ultrastructural characteristics common to other steroid-secreting cells. Lipid droplets and mitochondria are abundant and lie principally at the apical end of the cell. Lipid droplets are not membrane-limited. Cisternae of smooth endoplasmic reticulum that are occasionally continuous with the less abundant rough endoplasmic reticulum are a prominent feature of the interrenal cell. Tubular profiles of rough endoplasmic reticulum often lie tangentially to mitochondria and ribosomes are either free, grouped in polyribosomal clusters, or bound to the endoplasmic reticulum. Mitochondria possess tubular cristae in the inner regions of the gland and frequently contain a paracrystalline array of small 10nm (o.d.) tubules and less frequently a hexagonal array of 40nm trilaminar rings. Other cytoplasmic components include dense bodies, residual bodies, microtubules, microfilaments and specialized single membrane-bound vesicles. Gap junctions, intermediate junctions and interdigitating processes constitute the main intercellular associations. No tight junctions were identified. The single membrane-bound vesicles which are occasionally filled with a low electron-dense, lipid-like material form septate-like “junctions” with the plasma membrane. The septa bridge an intracellular gap of 15–17 nm. The vesicles are usually located near the subendothelial space at the basal and basilateral regions of the cell. Occasionally, vesicles fuse with the plasma membrane. It is suggested that these vesicles represent morphological evidence for the exocytotic release of steroid hormones.

    Notes: Summary Light microscopic autoradiography performed subsequent to intraocular injection of 3H-leucine revealed silver grains (SG) above axons of the optic tract which could be followed into the ventral and caudal portion of the suprachiasmatic nuclei (SCN) and above the contralateral anterior hypothalamic nucleus (AHN). By high resolution photometric measurement and computer processing the labelled areas were analysed, thus yielding statistical data of the relative grain distribution. The highest SG density was found in the ventrolateral part of both SCN (SCvl), confirming earlier reports concerning retinohypothalamic connections. That area exhibiting a cytoarchitecture different from the remaining nucleus was traversed, however, by numerous labelled axons. In the caudal part of both SCN a specific projection field of retinal fibres could be located. Here, almost no traversing fibres contribute to the rather circumscribed marked area. In the ventral part of the contralateral AHN, diffuse labelling well above background levels could be observed. Distinction between bypassing and terminating fibres within the SCvl could not be made using light microscopy. Analysis of SG distribution of the SCvl with electron microscopic autoradiography revealed a specific localization of SG within presynaptic terminals containing clear vesicles and pale mitochondria.

    Notes: Summary The pineal organ of the chicken was investigated electron microscopically during embryonic and post-hatching development with special regard to photosensory and secretory features. Throughout the developmental period both pinealocytes and supporting cells, of which the pineal parenchyma is composed, were rich in ribosomes, granular endoplasmic reticulum and mitochondria, but lacked agranular endoplasmic reticulum. The outer segments of pinealocytes barely showed formation of lamellar structures (disks) at the 17th and 21st day of incubation. Before and after hatching the follicular lumen was often filled with amorphous material presumed to be derived from outer or inner segments. By 15 days after hatching the whorl-like structures were occasionally connected to bulbous outer segments, and their relation appeared similar to that of the adult. Mitochondria disappeared from the inner segments after 21 days of incubation. Dense core vesicles (about 80–120 nm in diameter), regarded as secretory granules, appeared first at the 10th day of incubation in the supranuclear region of the pinealocyte. With the extending of basal processes the dense core vesicles gradually migrated into these processes, attained maximum number one month after hatching and increased further in the adult; they are located both around the Golgi apparatus and in the basal process. These results provide evidence that secretory activity is maintained from embryonic stages to adulthood.

    Notes: Summary The ultrastructure of nerve endings in the musculature of Lymnaea stagnalis was studied. Seventeen different types of axon ending were distinguished according to the size and morphology of the granules or vesicles the contain. Nine types of axon endings form neuromuscular synapses. Some of these types also form axo-somatic synapses on peripheral neurones. One axon may innervate several muscle cells. Furthermore more than one type of axon may innervate one muscle cell. One type of axon ending forms axoaxonic synapses on the presynaptic part of neuromuscular junctions. In the connective tissue near muscle cells seven types of free axon endings were found. These seem to be peripheral neurosecretory endings. Some of these are propably derived from known types of neurosecretory cells in the central nervous system, whereas others appear to originate from peripheral neuronal perikarya. Body wall muscles appear to be innervated by neuromusculatur synapses, whereas in the visceral musculature both neuromuscular junctions and free axon endings are found.

    Notes: Summary The neurotensin-cell is identified immunohistochemically and ultrastructurally by differential counting of endocrine cells in the gut of a primate (Tupaia belangen). Utilizing light microscopy, the EC-cells are identified by the Masson-Fontana silver stain; with the same method the neurotensin cells are not stained. The other endocrine cells have been quantified in the small intestine using the peroxidase-antiperoxidase stain with antisera against glucagon, somatostatin, cholecystokinin, gastrin, secretin, pancreatic polypeptide, gastric inhibitory peptide and neurotensin. In the ileal mucosa of Tupaia, the most frequent endocrine cell is the EC-cell followed by the glucagonoid cell, (L-cell). The immunoreactive neurotensin cell represents the third most frequent endocrine cell in this region. On the ultrastructural level, this third most frequent endocrine cell is a heretofore undescribed cell, the N-cell, containing electron dense secretory granules measuring 335±87 nm in diameter.

    Notes: Summary Immunocytochemically, secretin cells have been demonstrated to occur in the duodenum and jejunum of several mammals. Calculations on the relative frequency of such cells indicate that the bulk of secretin occurs in the jejunum, a fact supporting the view that secretin may be released by physiological stimulants other than hydrochloric acid. Electron microscopical identification of cat and pig secretin cells confirmed their identity with the ultrastructurally defined S cells, and staining experiments revealed that secretin cells were argyrophilic both with the method of Grimelius and with that of Hellerström and Hellman. Secretin cells are detected already in the 17-day old fetal rat duodenum and show a developmental pattern similar to that displayed by the gastrin cells. It is suggested that secretin may play a role in the early regulation of growth of the fetal gastrointestinal tract.

    Notes: Summary An indirect immunofluorescence technique and an anti-chicken LH serum were used to localize cells in the adenohypophyses of drakes at different stages of their breeding cycle, after castration, and after castration combined with thyroxine treatment. Immunofluorescent cells were distributed throughout both lobes of the adenohypophyses from control and experimental birds and were shown to be alcian blue positive, PAS negative, basophiles. Immunofluorescent cells were as numerous in castrated birds as in castrated birds treated with thyroxine. Adjacent thin and semi-thin sections were used to study the cells binding anti-LH serum at light microscope and ultrastructural levels. The cells contained spherical granules with variable densities and diameters ranging between 40 and 280 nm in the rostral (= cephalic) lobe, and between 60 and 260 nm in the caudal lobe. The light microscope and ultrastructural observations showed that the anti-LH serum binds to cells which have been classified by other authors in the Pekin duck, quail and pigeon as TSH producing delta cells. The experimental technique used did not permit a distinction to be made between cells producing FSH and LH.

    Notes: Summary The columnar epithelial cells of the gas gland in the swimbladder of the toadfish, Opsanus tau L., contain lamellar bodies that resemble the lamellar bodies found in epithelial cells of vertebrate lungs. Cytochemical assays indicate that swimbladder lamellar bodies are soluble in chloroform-methanol solution, react with tricomplex flocculation solution (indicating a phospholipid component), exhibit a positive reaction for cholesterol when exposed to digitonin, and contain acid phosphatase. The anterior chamber of the toadfish swimbladder is lined by an extracellular layer. Digitonin-cholesterol crystals are found in this layer when the swimbladder is treated with digitonin. A ruthenium red positive layer is also present in the anterior chamber of the toadfish swimbladder. The structure and cytochemistry of swimbladder lamellar bodies are compared with those of vertebrate lung lamellar bodies. Similarities between the extracellular layer in the swimbladder and the extracellular layer in lungs are also noted.

The seminiferous tubules were devoid of all stages of spermatid development and mature spermatozoa were absent from the tubule lumina. The diameter of the Leydig cells was 25 % less than that of controls. Examination with the electron microscope revealed thick bundles of collagen fibrils interspersed between Leydig cells and surrounding Leydig cells in the blinded hamsters. The Leydig cell nuclei were shrunken and highly infolded. Lipid droplets that were often seen in normal Leydig cells were absent in the involuting Leydig cells. The size of the Golgi complex and the amount of smooth endoplasmic reticulum were reduced. Results of the present experiment confirm that inactivity of the Leydig cells is the reason for the decline in serum testosterone levels in blinded hamsters.

The ultrastructural details concerning these internal membranous structures are shown in detail and their significance is discussed.

    Notes: Summary The fine structure of the secretory epithelial cells of rat's ventral prostate has been studied following organ culture. Culturing with either testosterone or insulin alone, and with the two hormones combined, were carried out to investigate how insulin modifies the action of testosterone on the maintenance of cellular integrity. After 4 days in hormone-free culture, the secretory epithelial cells showed signs of cellular atrophy and regression, involving loss of the apical microvilli, absence of the apical secretory vacuoles, atrophy of the Golgi apparatus, decrease in rough endoplasmic reticulum and the appearance of autophagic vacuoles. The presence in the medium of either testosterone or insulin alone, or combined, prevented cellular atrophy and regression. The best maintenance of cellular integrity was obtained in a culture containing both hormones. The effects of insulin was approximately equivalent to those of testosterone in the maintenance of cellular integrity.

    Notes: Summary The fine structure of the interstitial gland of the rat ovary was studied at estrus and on Days 4, 6, 10, 14 and 18 of pregnancy. At estrus, ovarian interstitial cells have small nuclei with dense irregular clumps of heterochromatin. Mitochondria are small and rod-shaped and have predominantely lamellar cristae. Numerous osmiophilic lipid droplets are present. At Days 4 and 6, nuclear heterochromatin is reduced, and nucleoli are larger and complex. Mitochondria are enlarged and often bizarre-shaped and have tubular cristae. Golgi and smooth endoplasmic reticulum are more conspicuous. At Day 10, prominent ultrastructural features include nuclei with conspicuous heterochromatin, smaller mitochondria with both lamellar and tubular cristae, numerous ribosomes and lipid droplets with decreased osmiophilia. At Days 14 and 18, nuclei have increased heterochromatin, mitochondria are small and have lamellated cristae and an increase in the size and number of lipid droplets occurs. These observations suggest that steroidogenic activity of interstitial cells is highest during the first half of pregnancy and regresses during the last half. It is suggested that the interstitial gland is an important ovarian source of pregnancy hormone(s) during the first half of gestation and that LH may modulate steroidogenic activity in this ovarian component.

    Notes: Summary Red, intermediate and white axial muscle fibres of the sharks Etmopterus spinax and Galeus melastomus were studied by electron microscopy and morphometry. The mitochondrial content is more than thirty percent in red, less than one percent in white, and up to fifteen percent in intermediate fibres. About one third of the mitochondria in red fibres are accumulated close to the sarcolemma. Red fibres contain much glycogen, present as rosettes (alpha particles). Intermediate fibres contain less glycogen (as beta particles). White fibres have scarcely any visible energy reserves. Red fibres contain slightly less (4–5%) of the sarcotubular system than the other fibre types (6–8 %). In all fibre types, the terminal cisternae of the SR are regularly divided by clefts. Triads or dyads are generally positioned at the Z discs, but in Galeus white fibres two dyads may be present, one on each side of the Z disc. The morphology is discussed in relation to current views on the functions of different muscle fibre types.

    Notes: Summary The ultrastructure of mid-gut cells of female Nasonia fed on a diet of 10% sterile sucrose is described. There are extensive alterations in cell organelles, particularly the mitochondria, rough endoplasmic reticulum (R.E.R.) and lipid inclusions, when compared to similar insects fed a normal diet of dipteran pupae. A proportion of the mitochondria found in the apical cell region are enlarged in size and contain electron-dense granules. The remaining mitochondria are smaller, but also contain electron-dense granules. Cytochrome-c oxidase activity appears to be absent from the enlarged mitochondria. The R.E.R. appears reduced in many cells of the 1 day old, sugar-fed insects, however, this component fluctuates throughout the remaining life span. The lipid inclusions prominent in the 1 day old, pupae-fed insects are not present in sucrose-fed females of the same age, but lipid deposition was recorded later in the life span. There are many large residual bodies and cytolysosomes present in the old, sugar-fed insects. These changes in ultrastructure are discussed in relation to diet and longevity.

    Notes: Summary The fine structure of the tentacles of the articulate brachiopod Terebratalia transversa has been studied by light and electron microscopy. The epidermis consists of a simple epithelium that is ciliated in frontal and paired latero-frontal or latero-abfrontal longitudinal tracts. Bundles of unsheathed nerve fibers extend longitudinally between the bases of the frontal epidermal cells and appear to end on the connective tissue cylinder; no myoneural junctions were found. The acellular connective tissue cylinder in each tentacle is composed of orthogonal arrays of collagen fibrils embedded in an amorphous matrix. Baffles of parallel crimped collagen fibrils traverse the connective tissue cylinder in regions where it buckles during flexion of the tentacle. The tentacular peritoneum consists of four cell types: 1) common peritoneal cells that line the lateral walls of the coelomic canal, 2) striated and 3) smooth myoepithelial cells that extend along the frontal and abfrontal sides of the coelomic canal, and 4) squamous smooth myoepithelial cells that comprise the tentacular blood channel. Experimental manipulations of a tentacle indicate that its movements are effected by the interaction of the tentacular contractile apparatus and the resilience of the supportive connective tissue cylinder. The frontal contractile bundle is composed of a central group of striated fibers and two lateral groups of smooth fibers which function to flex the tentacle and to hold it down, respectively. The small abfrontal group of smooth myoepithelial cells effects the re-extension of the tentacle, in conjunction with the passive resiliency of the connective tissue cylinder and the concomitant relaxation of the frontal contractile bundle.

    Notes: Summary Isolated rat hearts were perfused with a balanced electrolyte solution containing 1000mM glycerol for 15min and then perfused with normal electrolyte solution for up to 32 min. The perfusion with hypertonic glycerol solution and subsequent washout is termed “glycerol treatment”. Initially, glycerol removal causes swelling and rupture of the T-system in ventricular myocardial cells which correlates temporally with a period of cardiac arrest. Contractility returns during further glycerol removal and concomitant recovery of the T-system is observed. Atomic absorption spectometry and neutron activation analysis were used to measure ventricular sodium, potassium and calcium ion content. There is no apparent correlation between changes in ion content and cardiac arrest or recovery. The water movements were calculated from wet weight, dry weight and inulin space, and confirmed by morphometric analysis of extracellular and intracellular space. It is suggested that the swelling and rupture of the T-system is due to the rapid water movements that were observed during the onset of glycerol removal. Ultrastructural analysis of glycerol-treated atrium from the same hearts shows damage of mitochondria and of the L-system and intracellular edema. The structural changes are correlated with a loss of atrial contraction. As in ventricular myocardium, resumption of contraction is associated with an almost complete recovery from ultrastructural damage.

    Notes: Summary Quantitative ultrastructural morphometric studies were made on the prolactin cells of Poecilia latipinna adapted to freshwater (FW), one-third seawater (1/3 SW) and full-strength seawater (SW), and at various times after transfers between 1/3 SW and FW. In fully-adapted fish the rates of prolactin (PRL) synthesis and PRL release are inversely related to environmental salinity. During adaptation to a new salinity the two rates are temporarily uncoordinated, with release increasing or decreasing more readily than synthesis. Synthesis appears to take 30 h or longer to come into balance with the increased release rate following transfer from 1/3 SW to FW, and 72 h or longer to adjust to the reduction in release rate that follows the reverse transfer. The excess PRL granules that accumulate in the latter situation appear to be removed by lysosomal digestion. As in other teleosts, in fish adapted to the external medium the size of the stored PRL granules is inversely related to external salinity, but this relationship breaks down during adaptation to a new salinity. The stellate cells which penetrate between the PRL cells are more prominent, more extensively ramified, and appear more metabolically active in FW-adapted fish than in the other groups. These cells seem to be closely related in function to the secretory activity of the PRL cells.

    Description / Table of Contents: Zusammenfassung Es wurde die glatte Muskulatur des Antrum ventriculi sowie des Pylorus von 7 Patienten mit einem oder mehreren Ulcera ventriculi untersucht. Vergleichsweise konnten die entsprechenden Regionen des Magens von 5 normalen Organspendern der elektronenmikroskopischen Präparation zugeführt werden. Elektronenoptisch faßbare Veränderungen fanden sich noch 180 mm von der Ulcusläsion entfernt an den glatten Muskelzellen der inneren Rings- und der äußeren Längsmuskulatur sowie an den Gefäßen. Eine regelmäßig anzutreffende Veränderung der glatten Muskelzellen des Antrum und des Pylorus besteht in einer Strukturverdichtung des Kerns, der sarkoplasmatischen Grundsubstanz und der Myofilamente, kombiniert mit einer Vermehrung und Vergrößerung der hypolemmalen Vesikel, wie der dense bodies. Die Befunde werden als Ausdruck einer starken isometrischen Kontraktion der glatten Muskulatur im Antrum und im Pylorus bei Ulcera ventriculi gedeutet. Die geschilderten Befunde ließen sich an normalem Untersuchungsmaterial nicht erheben. Neben für das Magenulcus unspezifischen pathologischen Befunden an den glatten Muskelzellen, wie Anhäufung von Lipofuscin im Endoplasmahof und Lyse der Myofilamente, konnten spezifische Veränderungen gefunden werden. Diese betreffen die nur im Endoplasmahof differenzierten Räume des endoplasmatischen Reticulums. Bei zahlreichen Muskelzellen der Rings- und Längsmuskelschicht kommt eine extrem starke Quellung des endoplasmatischen Reticulums zur Beobachtung. Die Vacuolisation der endoplasmatischen Räume führt zu einer Verdrängung der übrigen Zellorganellen sowie der Myofilamente. Die Capillarendothelzellen weisen degenerative Veränderungen auf, die zu einem Sistieren cytopemptischer Vorgänge führen. In der arteriolären Gefäßstrecke findet sich vielfach eine starke Kontraktion der glatten Muskulatur bis zum völligen Verschluß der Gefäßlichtung. Auch diese Befunde ließen sich nur an den Mägen mit Ulcera ventriculi, nicht dagegen an dem normalen Vergleichsmaterial erheben.
    Notes: Summary The smooth musculature of antrum and pylorus of 7 patients with one or more stomach ulcers was examined by transmission electron microscopy. Corresponding regions of the stomachs of 5 normal organ donors served as control material. Electronmicroscopically visualizable changes were found at distances of up to 180 mm from the site of ulcerous lesion on the smooth muscle cells of the inner circular and outer longitudinal musculature, also on the blood vessels. A regularly occurring modification of the smooth muscle cells of antrum and pylorus consists of a structural condensation of the nucleus, of the sarcoplasmic ground substance and of the myofilaments, combined with augmentation and distension of the hypolemmal vesicles and of the dense bodies. These findings are interpreted as expression of a pronounced isometric contraction of the smooth musculature in the antrum and pylorus in presence of ulcera ventriculi. Such findings are not obtainable on normal material. Besides pathological modifications at the smooth muscle cells which are unspecific for stomach ulcer, such as accumulation of lipofuscin in the endoplasmatic lacunae and lysis of the myofilaments, specific changes were observed. These concern the interstices of the endoplasmic reticulum to be found only in the endoplasmatic lacunae. In numerous muscle cells of the circular and longitudinal muscle layers an extremely marked distension of the endoplasmic reticulum is found. Vacuolization of the endoplasmic cisternae causes displacement of the other cell organelles and of the myofilaments. The capillary endothelial cells show degenerative changes which bring about arrest of cytopemptic processes. In the arteriolar regions a strong contraction of the smooth musculature, at times resulting in complete occlusion of the vasal lumen, is frequently found. These findings, too, are obtainable only on stomachs with ulcera ventriculi and not on normal control material.

Injektion von 100 mg/kg eines Pentosanschwefelsäureesters (SP 54) sind in der Nierenrinde von Ratten schwere degenerative Veränderungen der proximalen Tubuluszellen zu beobachten. Das Tubulusepithel ist flachkubisch umgewandelt, das Tubuluslumen ist angefüllt mit ausgestoßenen Mitochondrien und anderen Zelltrümmern. Am stärksten betroffen ist die Pars contorta des proximalen Tubulus. Weniger stark veränderte Tubuluszellen sind angefüllt mit Cytosomen, die eine spezifische Feinstruktur haben oder gehäuft gegenüber Kontrollen Cytoplasmaprotrusionen in das Tubuluslumen aufweisen. Die akute Tubulusdegeneration ist ein spezifischer Effekt von SP 54. Heparin bewirkt nur eine leichte Schwellung des Tubulusepithels, die häufiger auftritt als bei Kontrolltieren.
    Notes: Summary 24 hrs after the intravenous injection of 100 mg/kg of a Pentosansulfuricacidester (SP 54) severe degenerative alterations are observed in the proximal tubular cells in rat kidney cortex. The tubular epithelium has changed to a single layer of elongated cubic cells. The tubular lumen is filled with cellular debris and extruded mitochondria. The greatest change is seen in the pars contorta of the proximal tubule. Less degenerated tubular cells have a great number of cytosomes with a specific substructure or have cytoplasmatic protrusions into the tubular lumen. This acute tubular degeneration is a specific event after the administration of SP 54 and is not seen after an equimolar dosis of Heparin. Heparin induces only a slight swelling of the tubular epithelium, more frequent seen as in the control animals.

Blutgefäße mit Pericyten, porenhaltige Capillaren sowie die Tubuli der Rinde und des Markes werden nicht von Nervenfasern begleitet. Ganglienzellen wurden in der Rattenniere nicht beobachtet. Periphere Nerven mit einem ein-bis zweischichtigen Perineurium kommen im paravasalen Gewebe der Interlobar- und Arcuata-Arterien vor; sie enthalten neben zahlreichen marklosen Nervenfasern gewöhnlich auch 2–4 markhaltige. Nervenfaser-Bündel ohne perineurale Scheide finden sich im paravasalen Gewebe der Arcuata- und Interlobular-Arterien. Darüber hinaus sind in unmittelbarer Nachbarschaft der großen Arterien (Interlobar-, Arcuata- und Inter-lobular-arterien) und der Vasa afferentia marklose Nervenfasern und freie Axone vorhanden, die auch die proximalen Abschnitte der Vasa efferentia der subcapsulären und intermediären Rindenschicht begleiten. Im Nierenmark werden die juxtamedullären Vasa efferentia und die Arteriolae rectae innerviert; marklose Nervenfasern und freie Axone sind nur bis zur Außen-Innenstreifen-Grenze nachweisbar. Die Innervation der muskelzellhaltigen arteriellen Gefäße erfolgt durch aufgetriebene Axonabschnitte (Neuroeffektor-Zonen), die vorwiegend agranuläre Vesikel enthalten. Diese Strukturen liegen stets an der Grenze von Adventitia und Media bzw. Elastica externa; zwischen den glatten Muskelzellender Media wurden keine vesikelhaltigen Axonabschnitte gefunden. Als minimaler Abstand zwischen den vesikelhaltigen Axonabschnitten und den von einer Basalmembran umschlossenen glatten Muskelzellen (neuromuskuläre Distanz) wurden 600 Å gemessen. Die vorliegenden Ergebnisse werden mit den fluoreszenzmikroskopischen und histochemischen Untersuchungen über die adrenerge und cholinerge Innervation der Niere verglichen. Die sich aus diesem Vergleich ergebenden Probleme und funktionellen Konsequenzen für die Innervation der Niere sowie die Natur der cholinergen Fasern (afferente oder postganglionäre parasympathische Fasern) werden diskutiert.
    Notes: Summary The innervation of the rat kidney is defined by a system which supplies those arterial blood vessels whose walls contain smooth muscle cells and the juxtaglomerular apparatus. Vessels containing pericytes, or those vessels composed of an endothelium only, as well as the tubules of both the cortex and medulla, are not innervated. Furthermore, ganglion cells do not occur in the rat kidney. The nervous apparatus of the rat kidney consists of peripheral vegetative nerves, ensheathed by a perineurium, with 2–4 myelinated fibers running in the paravasal tissue of the interlobar and arcuate arteries, and of nerve bundles without a perineurial sheath in the paravasal tissue of the arcuate and interlobular arteries. Non-myelinated fibers and free axons occur in the immediate vicinity of the great arteries (interlobar, arcuate, and interlobular) and the vasa afferentia. Nerve fibers and free axons are also seen in the vicinity of only the proximal parts of those vasa efferentia which supply the cortical capillary plexus. The arteriolae rectae of the medulla, and their vasa efferentia, from which they arise, are innervated by non-myelinated fibers and free axons which accompany these arterial vessels only to the boundary of the outer and inner stripe of the outer zone of the medulla. The functional innervation of those vessels with smooth muscle cells results from neuro-effector zones which predominantly show agranular vesicles. These structures were never seen between the smooth muscle cells within the media; the minimum neuromuscular distance was 600 Å. The present findings are correlated with the lightmicroscopically demonstrated adrenergic and cholinergic innervation. The resultant problems and functional consequences of the innervation of the kidney, especially the nature of the cholinergic fibers (afferent or post-ganglionic parasympathetic fibers) are briefly discussed.

Cette formation endothéliochoriale est composée: d'un endothélium maternel hypertrophié et de cellules géantes déciduales; d'une couche de «substance inerte interstitielle»; du trophoblaste constitué d'une assise syncytiale continue (syncytiotrophoblaste) et d'une assise cellulaire discontinue (cytotrophoblaste); d'un endothélium foetal accompagné de son environnement mésenchymateux. Les capillaires foetaux envahissent progressivement le syncytiotrophoblaste. L'hypertrophie de l'endothélium maternel diminue en fin de gestation; de ce fait la distance, séparant le sang maternel du sang foetal, peut atteindre en certains points 2 μ. L'endothélium maternel, le syncytiotrophoblaste et l'endothélium foetal présentent des signes ultrastructuraux d'un rôle de transfert. La «substance inerte inerte interstitielle», qui sépare les tissus d'origine maternelle des tissus d'origine foetale, est constante mais d'épaisseur variable; elle est spécifique des structures endothéliochoriales. Les cellules géantes déciduales, dont le nombre diminue en fin de gestation comportent parfois un ou plusieurs «corps glycogéniques». Le syncytiotrophoblaste, siége d'activité de biosynthése de stéroïdes, est riche en ergastoplasme, ce qui autorise à postuler la synthése d'hormones protéiques.
    Notes: Summary The ultrastructure of feline placental labyrinth has been studied from the 45th to the 63rd day of gestation. The endotheliochorial structure is composed of: a thick hypertrophied maternal endothelium with giant decidual cells; an “interstitial inert substance”; a continuous syncytial layer (syncytiotrophoblast) and a discontinuous cellular layer (cytotrophoblast) of trophoblast; and a thin foetal endothelium with its surrounding mesenchyme. Foetal capillaries increasingly invade the syncytiotrophoblast. Maternal endothelial hypertrophy is reduced in the last days of gestation. Thus the interval between maternal and foetal bloods may in some areas become 2 μ, in late pregnancy. Foetal endothelium, syncytiotrophoblast, and maternal endothelium demonstrate ultrastructural features of transfer function. The “interstitial inert substance”, which separates maternal from foetal tissue was always evident though with variable thickness. The giant decidual cells which are reduced in late pregnancy, show one or several “glycogen bodies”. The syncytiotrophoblast, where some steroïd biosynthesis has been demonstrated, has an extensive rough endoplasmic reticulum suggestive of protein hormone synthesis activity.

    Notes: Summary Testes of newborn mice not older than one day were transplanted into the anterior chamber of one eye of an adult male white Holtzman rat. The events observed in the Leydig cells of the grafted testes can be divided into two periods. Period one is marked by differentiation; period two, by dedifferentiation. In period one, the lipid and glycogen inclusions disappear, possibly owing to the gonadotropins of the host. In period two, which starts after day 15 post-grafting, the lipids and glycogen reappear; the Leydig cells now contain residual bodies, whereas the S.E.R. disappears. Immunological mechanisms can be implicated; it is suggested that the Leydig cells dedifferentiate because the host's gonadotropins may be incapable of evoking and/or activating the genetic memory of the grafted Leydig cells.

Six stades du développement embryonnaire sont observés 2, 3, 4, 5, 7 et 8 jours après la date présumée de fécondation. Dans les 3 premiers cas, on compte respectivement 2, 8 et 24 blastoméres. A 5 jours, il y a 30 à 40 cellules et un nombre dépassant 60, ultérieurement. Des cellules trophoblastiques primitives se différencient à 7 jours et une cavité de segmentation en forme de croissant apparaît au 8 ème jour. La zone pellucide est toujours présente à 8 jours, la période de préimplantation semblant plus longue chez le babouin que chez l'homme. Des particules de type C sont décrites dans la zone pellucide et les espaces périvitellin et interblastomériques. La transformation des mitochondries, des changements dans la répartition des ribosomes, des corps multivésiculaires, pseudocristallins et figures myéliniques, des nucléoles et amas granuleux intra-nucléaires s'observent chez le babouin comme chez certains autres Mammifères; par contre, les faisceaux fibrillaires cytoplasmiques sont absents. L'étude des changements de l'ultrastructure provoqués par des hormones et drogues permettrait de mieux évaluer la nocivité de ces substances pendant les premiers stades du développement embryonnaire des Primates.
    Notes: Summary The baboon preimplantation stages were examined using light and electron microscopy. Six cases were studied at 2, 3, 4, 5, 7 and 8 days estimated fertilization age. The first 3 specimens were composed of 2, 8 and 24 blastomeres respectively. At 5 days, 30 to 40 cells were counted and more than 60 cells in later stages. Primitive “trophoblast cells” differentiate at 7 days and a crescentic blastocoele appears at 8 days. Shedding of the zona pellucida is not observed in the 7 and 8 day specimens. The preimplantation period is longer in the baboon than in man. C-type viruses are observed in the zona pellucida, in the perivitelline and interblastomeric spaces. Microvilli and caveolae cover the periphery of the baboon conceptus. As in many other mammals, transformation of the mitochondria, changes in the ribosomes distribution, multivesicular bodies, myelin figures, nucleoli and intranuclear clusters of granules are described in the baboon. Cytoplasmic fibrous strands are not present as in the mouse. Experiments on the influence of hormones and drugs on ultrastructural changes would help to evaluate the importance of biohazards during the early development of primates.

    Notes: Summary The thin limbs of the loops of Henle in the mouse kidney have been investigated by conventional electron microscopy. Resulting from light microscopic investigations, a distinction in the epithelia of short and long loops can be demonstrated. Ultrastructurally, the thin limbs (descending) of short loops are composed of a uniformly thin and simple epithelium. In contrast, long loops (thin descending and ascending) are composed of three different epithelial types which are representative of a distinctly more complex epithelial system. Two epithelial types were observed in the thin descending limbs of long loops and the third type was observed in the ascending thin limbs. Based upon these findings it is suggested that the thin descending limbs of short and long loops of Henle in the mouse kidney cannot perform the same functions in the renal concentrating mechanism.

    Notes: Summary The myosatellite cells in the axial muscles of the sharkGaleus melastomus is found more frequently in the red muscle fibers than in the white fibers. The total length of the cell extensions is about 100 μm. In about 20% of all cases, the extensions were present in pairs, and it is concluded that the satellite cell processes bifurcate. Processes towards the intercellular space and towards the interior of the muscle fiber are also described. The cytoplasm of the satellite cells contain glycogen, Golgi elements, lysosome-like vesicles, granular endoplasmic reticulum and microtubules. It is concluded that the morphology of the satellite cells indicates a more active function than that of a dormant reserve myoblast.

    Notes: Summary The fine structure of the blood vessels was studied in the developing brain of chick embryo. The blood vessels were present in the embryos first examined (4 days after incubation) and their number increased in subsequent stages. The endothelial cells were generally large and showed junctional complexes, many microvilli and a large number of cytoplasmic organelles. Many tubular bodies and coated vesicles were also present. The tubular bodies were sometimes noted near the Golgi apparatus, suggesting their origin from this organelle. The mitochondria in the endothelial cells were generally larger than those in the surrounding neuropil. An ill-defined basement membrane-like substance was noted outside some endothelial cells on the 11th day and well formed basement membrane was present in the vessels of 18 day old embryos. A complete subpial astrocytic basement membrane was present from the early stage studied (4 days).

    Notes: Summary The fine structure of the blood vessels of an apparently primary intracerebral malignant lymphoma was compared with those in another malignant lymphoma in a retroperitoneal lymph node. The essential features of the blood vessels in both cases were the same. They resembled the blood vessels found in normal lymph nodes and were both distinctly different from those seen in normal brain. It was concluded that the direction of differentiation of the blood vessels which arise to nourish the neoplastic tissue is determined by the nature of the tumor rather than the vessels from which they originate.

    Notes: Summary Histochemical studies of type II myoclonus bodies demonstrated that they were composed of a saccharide-lipid complex with a small amount of protein. In particular, they contained phospholipid, ganglioside and sphingomyelin. The ultrastructure of the bodies revealed that they were bound by a membrane. They were diffusely stippled or homogeneous. They were not formed of filaments or lamellae. Some correlation with microbody or lysosomal structures in association with degeneration of the neurone was suggested.

Basement membrane covers the outer aspect of the cortex, the outer layer of which is formed by interlacing astrocyte processes of varying size. The processes are closely applied with occasional desmosomes between them. Projections of astrocyte processes of different dimensions covered by basement membrane extend at irregular intervals from the cortex into the subarachnoid space.

Parts of the nerve were demyelinated, sometimes extensively so and from these regions the oligodendrocytes had disappeared and there was proliferation of astrocytes. Parallel bundles of fibres with a “railway line” formation, occurred in the cytoplasm of the astrocytes. Phagocytes infiltrated the damaged nerve bundles and the fibrous septa between them. Osmiophilic particles occurred in the astrocytes, capillary endothelial cells and in the phagocytes. The collagen fibres of some septa were widely separated presumably by fluid. b) Devic's Disease. This condition occurred in a patient with active pulmonary tuberculosis. The nerve was extensively demyelinated and showed absence of oligodendrocytes, proliferation of astrocytes and infiltration by macrophages. Some astrocytes possessed Rosenthal fibres. Intranuclear inclusions occurred in the astrocytes and electron dense cytoplasmic inclusions in the capillary endothelial cells and the macrophages. c) Disseminated Sclerosis. Parts of the nerve were partially and other parts completely demyelinated. Oligodendrocytes were absent from the completely demyelinated zones but were present in the partially demyelinated zones. In and around the demyelinated regions, there was proliferation of astrocytes and they frequently exhibited the “railway line” patterns in their cytoplasm. Phagocytes were frequent amongst the degenerating myelin and the proliferating astrocytes and also in the fibrous septa. Some macrophages presented intranuclear inclusions having a “corn on the cob” appearance.

The walls formed terminal loops limiting up to 4 adjacent tubules; transverse extensions were present between adjacent walls.     Notes: Summary Wistar rats of both sexes and the same litter were exposed to chronic lead intoxication from birth untill sacrifice 9 months later. Lead was administered as 0.4% solution of lead nitrate in drinking water. Samples from the parietal brain cortex were examined electron microscopically following intracardiac perfusion with paraformaldehyde-glutaraldehyde solution. Similar changes were observed in the microglial cells and the vascular pericytes whereas all the other tissue elements appeared intact. Both cell types hypertrophied, the microglia assumed characteristic spindle or rod shape, the cell organelles increased, the microglial endoplasmic reticulum widened strongly and a large number of lipid inclusions appeared. The latter consisted of large lipid droplets of varying size and shape, containing multiple zones of low density, and a dense component with a coarse granular structure. The similarity in the response of both cell entities to the lesion as well as some probable functions of microglial cells and their relationship to vascular pericytes are discussed.

Two muscle biopsies obtained before and after treatment with large doses of steroids, as well as autopsy tissue, demonstrated widespread degeneration of myofibres associated with cytoplasmic tuboreticular structures resembling paramyxovirus nucleocapsids in capillary endothelial cells and fibroblasts. Aggregates of spherical particles and “simple and granular” nuclear bodies suggested a virus-related or associated etiologic agent. In addition, interstitial cells of mesenchymal origin, probably related to macrophages or histiocytes were present. These contained numerous rodlike profiles or phagolysosomal bodies which resembled “Birbeck” granules. Inflammatory response was very scant. It would appear that polymyositis probably represents a primary reaction to antigemic substances and a morphologic manifestation of altered host macrophage response.

    Description / Table of Contents: Résumé Des cals expérimentaux de neuf jours, formés au niveau de radius de jeunes rats, sont traités par la méthode calcium-cobalt de Gomori (1939) pour la mise en évidence ultrastructurale de la phosphatase alcaline afin d'étudier son rôle éventuel dans le dépôt du calcium. L'activité enzymatique apparait initialement sous forme de précipités globulaires en dehors de la membrane cellulaire de jeunes chondroblastes hypertrophiques. Ce précipité donne ensuite naissance à des corps sphériques de phosphatase alcaline qui se forme près de la cellule. Ces corps sphériques s'observent dans une zone intermédiaire plus éloignée. Une formation de cristaux en aiguilles (apparemment une calcification) se développe dans des corps isolés ou agrégés, laissant voir nettement leurs limites, même lorsque la calcification est plus avancée au point qu'on ne peut plus distinguer des cristaux individuels. Au niveau des coupes témoins, traitées de façon identique mais sans substrat ou avec de l'E.D.T.A., on n'observe ni précipité enzymatique ou corps sphériques. L'aspect des dépôts cristallins dans des corps qui contiennent de la phosphatase alcaline ne peut s'expliquer que par l'existence d'une association étroite entre enzymes et calcification.
    Abstract: Zusammenfassung Neun Tage alter experimenteller Kallus an Radii von jungen Ratten wurde mit Gomori's (1939) Calcium-Kobalt Methode untersucht, um die Verteilung der alkalischen Phosphatase und ihre Beziehung zur Calciumablagerung ultrastrukturell zu demonstrieren. Enzymaktivität zeigte sich zuerst als globulares Präzipitat außerhalb der Zellmembran von Knorpelzellen im Beginn der Hypertrophie. Aus dieser Präzipitatschicht entstanden dann gerundete Körperchen, die sich von der Zelle abtrennten. Solche Körperchen wurden auch in größerer Entfernung von der Zelle beobachtet, d.h. in einer Zwischenzone zwischen benachbarten Zellen. Nadelförmige Kristalle, wahrscheinlich von Calcium-Salzen, wurden in einzelnen oder aggregierten Körperchen beobachtet. Die äußere Zone der Körperchen blieb jedoch deutlich sichtbar, selbst dann, wenn der Calciumgehalt derart zugenommen hatte, daß einzelne Kristalle nicht länger erkennbar waren. In Kontrollen, die in gleicher Weise behandelt waren, aber ohne Substrat oder mit Zufügung von EDTA, wurden weder Präzipitate noch Körperchen beobachtet. Das Auftreten von Calciumablagerungen in alkalischer Phosphatase enthaltenden Körperchen scheint kaum anders erklärbar als durch eine enge funktionelle Verbindung zwischen Enzym und Calciumablagerung.
    Notes: Abstract Nine day old experimental calluses in radii of young rats were treated with Gomori's (1939) calcium-cobalt method to demonstrate ultrastructurally the presence of alkaline phosphatase in a search for its possible role in the desposition of calcium. Enzyme activity first appeared as globule-like precipitates outside the cell membrane of early hypertrophic cartilage cells. This precipitate layer then seemed to give rise to spherical bodies of alkaline phosphatase which occur at a slight distance from the cell. The spherical bodies were also observed further away from the cell in an intermediate zone between neighboring cells. Needle-like crystal formation, apparently calcification, occurred inside single or aggregated bodies, leaving their peripheral rim clearly visible, even when calcification had increased to such an extent that individual crystals could no longer be recognised. In controls, treated in the same way but without substrate, or with EDTA, no enzyme precipitate or spherical bodies were seen. The appearance of crystalline deposits in bodies which contain alkaline phosphatase seems difficult to explain on any other basis than that there is a close functional association between the enzyme and calcification.

    Notes: Summary A comparison was made between morphological and biochemical estimates of extracellular and intracellular fluid spaces in rat brain cortex slices incubated under different conditions. By light microscopy the periphery of the slices was found to be more swollen than the center; this regional difference was verified biochemically in unfixed tissue. The electronmicroscopic evaluation of intra- and extracellular fluid spaces was accordingly based upon findings in a preselected area. Due to intracellular penetration of inulin in rat brain cortex slices the biochemically, determined extracellular and intracellular spaces were obtained by compartmental analysis of the inulin space. The concordance between the biochemical and the morphological findings was good: Both methods showed that the extracellular space increased during the incubation to a considerable magnitude after one hr. and that this extracellular space was reduced by excess potassium, glutamate, anoxia or incubation at 0°. Under the same conditions the biochemically determined intracellular space was increased. This cellular swelling was confirmed morphologically and found to comprise mainly glia cells after exposure to excess potassium, predominantly neurons after incubation at 0° and both cell types after anoxia or addition of glutamate.

    Notes: Summary After administration of the anorectic drug fenfluramine (40 mg/kg per day) to rats (s. c. or i. p.) and guinea pigs (p.o.) over periods of 2 to 11 weeks, electron microscopic examination was performed on lungs, liver, lymphatic tissues and peripheral blood of both species, and on adrenal glands and ovaries of rats. In rats, fenfluramine caused the following alterations (listed according to the duration of treatment): Formation of abnormal lamellated inclusions in lymphocytes and plasma cells of lymphatic tissues, appearance of “foam cells” in lung alveoli; formation of abnormal lamellated inclusions in corpus luteum, lymphocytes of peripheral blood, and in adrenal cortex. In guinea pigs, the same alterations were found in lymphocytes of spleen and peripheral blood, in hepatocytes and in lung. The present observations support the concept of a generalized phospholipidosis induced by amphiphilic compounds. The potency of fenfluramine to induce a lipidosis is, however, considerably less pronounced than that previously demonstrated for the anorectic drug chlorphentermine. This difference is suggested to be due mainly to the lower degree of amphiphilia of fenfluramine.

    Notes: Summary Lymphocytes of rat lymph nodes have previously been revealed as reliable and sensitive indicators of a particular drug side effect, notably a generalized phospholipidosis, inducible by a compound of amphiphilic character (chlorphentermine). The purpose of the present study was to examine the effects of other amphiphilic compounds upon rat lymphocytes. After oral administration of various tricyclic antidepressants (iprindole, imipramine, clomipramine, 1-chloro-amitriptyline, 1-chloro-10,11-dehydro-amitriptyline, noxiptiline, amitriptyline), or of two neuroleptic drugs (chlorpromazine, thioridazine) to rats, the popliteal lymph nodes were examined with the electron microscope. After a single application of either iprindole (50 mg/kg), imipramine (100 mg/kg), or clomipramine (150 mg/kg) small proportions of lymphocytes were found to contain abnormal lamellated cytoplasmic inclusions. The size and number of inclusions and the number of affected lymphocytes increased with further treatment. Similar observations were made after treatment with the chlorinated amitriptyline derivatives. On the other hand, only small numbers of lymphocytes were affected by noxiptiline, amitriptyline, chlorpromazine and thioridazine, even after prolonged treatments (up to 13 weeks) with high doses (up to 125–175 mg/kg). The present ultrastructural finding are interpreted as representing lipidosislike cellular alterations, yet of highly varying degrees. The large quantitative differences are tentatively suggested to be due to differences in the rate and mode of metabolism of the drugs, and due to differing degrees of amphiphilia of the compounds applied.

Although narrower, they correspond in gross appearance and ultrastructure to previously describedScenedesmus bristles. Openings, bordered by a series of props, are unlike those ofScenedesmus culture 614. Additional props are observed scattered independently on the cell wall; ridges are composed of a linear row of props. Sections of cells, or cell walls, reveal an additional prop, situated inside the openings; these props are composed of several tubules. Possible extrusion of bristles through these tubules, as well as the origin of the bristle from the cavity and vesicles immediately under the opening are discussed.

Small plasmodia cease streaming and round up. Food vacuoles collect in the lower center of the cytoplasmic mass. As the cytoplasm rises the food vacuolar contents are excluded from the plasmalemma and become the stalk core. A continuous, fibrous peridium and stalk tube enclose cytoplasm and stalk core respectively. Capillitial formation just precedes spore cleavage. Sporophore development is marked by autophagic activity and calcium deposition. Stalks of dried herbarium specimens of seven additional species have been examined. A mature stalk morphology very similar toProtophysarum with recognizable remnants of microorganismal food material is seen in all of them. It is thought that this marker is indicative of non-stemonitaceous stalk development.

    Notes: Abstract A strain ofSchizophyllum commune carrying a mutation in theB-mating factor (B-mut) shows septal dissolution when grown at 30° C for 2 to 3 days. The septa are intact if the organism is grown at 25° C for the same time, but begin to break down within 1 h after transfer to 30° C. At the ultrastructural level the dolipore swelling is the first part of the septal apparatus to be degraded, closely followed by the disorganization of the parenthesomes. A progressive thinning of the septal cross-wall produces an enlargement of the septal aperture sufficient to allow the passage of nuclei. It appears that degradative enzymes are probably carried to the site of septal dissolution in vesicles derived from endoplasmic reticulum in the area of the septal apparatus.

    Notes: Abstract Nitrogen starvation, effected by incubating a culture ofAnabaena cylindrica in a medium free from combined nitrogen and under an atmosphere of 1% CO2 in argon, leads to rapid and characteristic changes in the appearance, structure and function of the alga. Change of colour, due apparently to a decrease in the amounts of nitrogenous pigments, is accompanied by a structural transformation of vegetative cells: cyanophycin granules and polyhedral bodies disintegrate, lipid and glycogen accumulate, and large membrane-bound spaces form by means of thylakoid swelling and vesiculation. The rate of heterocyst differentiation and nitrogenase activity is increased. These changes are fully reversed on addition of ammonia to the culture. It appears that thylakoids reform by coalescence of small vesicles assembled in the intrathylakoidal space. Rapid ammonia assimilation is indicated by ample formation of cyanophycin granules in vegetative cells and of “plugs” in the heterocysts.

    Notes: Abstract Complexes of twisted ribbons composed of ordered arrays of microtubules are identified in close association with the plasmalemma and the surfaces of some organelles in senescent cells of photoheterotrophically cultured Chlamydomonas dysosmos. The ribbon complexes occur throughout the cytoplasm, and do not appear related to the flagellar insertions. The component microtubules are approximately 26 nm in width, exhibiting a center-to-center spacing of about 44 nm. Additional cytoplasmic microtubules are often closely related to the tubular complexes. A detailed description of their fine structure is presented here which tends to support the ascribed function of microtubules in maintaining the structural integrity of the protoplasm.

    Notes: Abstract The structural and functional symptoms of molybdenum deficiency inAnabaena cylindrica grown in a medium without combined nitrogen and thus dependent on fixation of elemental nitrogen, resemble those brought about by nitrogen starvation. However, the substantially increased rate of heterocyst differentiation in this culture is not accompanied by a corresponding increase in nitrogenase activity; on the contrary, enzyme activity is severely impaired in the absence of molybdenum. When the supply of molybdenum, or of ammonia, is restored, the alga recovers rapidly. Vanadium exerts an inhibitory effect upon nitrogen-fixing ability of the alga, and its presence in the molybdenum-deficient culture results in the amplification of the symptoms of mlybdenum deficiency.

    Notes: Abstract Microbody-like organelles occur in the cytoplasm of two chloromonadophycean algae,Vacuolaria virescens Cienkowsky andGonyostomum semen Diesing. Microbodies ofVacuolaria andGonyostomum have a granular matrix which lacks a crystalloid core; they are often present in close association with elements of the endoplasmic reticulum. The occurrence of microbodies in other algae is briefly reviewed.
