    Notes: Summary Ten myxoid liposarcomas (ML) were studied ultrastructurally in an attempt to determine the histogenesis of this neoplasm and structural associations which might contribute to their relatively benign clinical behavior. The findings were compared with normal and neoplastic adipose tissue. Three cell types were observed, i.e., “primitive” mesenchymal cells, intermediate cell types, and lipoblasts at various stages of development. The principle differences between the lipoblastic elements were the number and the size of intracytoplasmic fat vacuoles, the development of basement membrane-like material, micropinocytotic vesicles along the plasma membrane and the quantity and pleomorphism of mitochondria. The tumor vasculature was complex but consistently demonstrated a multilayered basal lamina. This finding has been described in neoplasms associated with a relatively good prognosis. This study demonstrates that the better differentiated lipoblasts in ML share some features with normal brown fetal fat and hibernoma. It is, thus, suggested that ML may be derived from brown adipose tissue.

    Notes: Summary In granular cell tumors, the granule-containing tumor cells and their processes form rounded complexes surrounded by a common bounding membrane. In the tumor, these complexes are closely related to the peripheral nerves and are seen in the perineurium and in the endoneurium. In addition, there are mixed complexes composed both of Schwann cells and granular tumor cells: the two types of cells are in close contact with each other and are also surrounded by a common bounding membrane. Furthermore, there one contacts between granular tumor cells and axons. These observations strongly suggest the existence of a relationship between Schwann cells and the cells of granular cell tumors. A comparison between the granular tumor cells and the phagocytic forms of Schwann cells reveals striking similarities: the granular cell tumor complexes are comparable with the Büngner bands of phagocytozing Schwann cells. A relationship between granular cell tumors and the phagocytozing form of Schwann cells is therefore assumed.

    Notes: Summary The ultrastructure of eleven thymomas with lymphocytic predominance, one “epitheloid” cell thymoma and two normal human thymuses is described with special reference to “Emperipolesis”. All patients have had myasthenia gravis. The normal human thymus consists of three parts: outer cortex, inner cortex, and medulla. The outer cortex contains mainly lymphoblasts and Metcalf's macrophages within the so-called “Clark-packet's”. The inner cortex consists mainly lymphocytes and interdigitating reticulum cells, and the medulla of epithelial cells, lymphocytes and Hassall's corpuscles. In all cases of lympho-epithelial thymoma and in normal human thymuses there are enormous interdigitations between epithelial (tumor) cells, lymphocytes and macrophages. The “epitheloid” cell thymomas also show findings which suggest an epithelial cell interaction. We have not found intact lymphocytes inside the cytoplasm of normal and/or tumor epithelial cells, macrophages or interdigitating reticulum cells. The intracellular existence of intact lymphocytes has been termed “Emperipolesis” by Humble, Jayne, and Pulvertaft, meaning “internal wandering”. These investigations indicate that “Emperipolesis” is not an adequate term for cellular interaction in normal human thymuses and thymomas. A false impression of intraepithelial location of thymic lymphocytes is created by two-dimensional sections of complex thymic structure. These ultrastructural studies revealed damage to lymphocytes only in macrophages with lymphocytolysis within these cells and accumulation of numerous heterophagic vacuoles containing fragments of lymphocytic debris within them.

    Description / Table of Contents: Zusammenfassung Zwei FÄlle von aggressiver Fibromatose der Schilddrüsenregion werden mit histologischen, immunhistochemischen und elektronenmikroskopischen Methoden untersucht. Mit Hilfe von menschlichem Serum gegen glatte Muskulatur können kontraktile Zellen in sÄmtlichen PrÄparaten, vornehmlich aber in den Randbezirken der Wucherung nachgewiesen werden. Elektronenoptisch werden alle diese Zellen als Myofibroblasten identifiziert. Die zentralen Bezirke der Wucherung zeigen hyalin verdickte und gequollene Kollagenfaserbündel mit nur wenigen zellulÄren Elementen. Somit wird der Myofibroblast als der für die Wucherung charakteristische Zelltyp erkannt. Die sich daraus für Pathogenese und Histogenese ergebenden Schlüsse werden diskutiert.
    Notes: Summary Two cases of aggressive fibromatosis of the thyroid gland region were studied by histological, immunhistological and electron-microscopic methods. Contractile cells can be found in all tissue preparations using human serum containing antibodies to smooth muscle, especially in the periphery of the tumor. These cells have the characteristics of myofibroblasts by electron microscopy. The central areas of the tumor are composed of hyalin thickened bundles of collagen fibres with few cellular elements. The myofibroblast is thus considered to be the characteristic cell type of the tumor, and the pathogenesis and histogenesis of the lesion are discussed.

    Notes: Summary The distribution and localization of glycogen particles in mesenchymal cells, chondroblasts and in young, hypertrophic and degenerating chondrocytes of the tibiotarsal anlage of developing chick embryos were examined ultrastructurally using the periodic acidthiocarbohydrazide-silver proteinate. The distribution of glycogen particles increased with the degree of differentiation and maturation of the chondrogenic cells. Mesenchymal cells showed minimal content of glycogen particles which gradually increased in the chondroblasts. The young chondrocytes further showed increased glycogen particle content compared to chondroblasts. This content reached a maximum level in hypertrophic chondrocytes. This level subsided in degenerating chondrocytes which showed a much reduced distribution of glycogen particles compared to hypertrophic chondrocytes. The morphological nature of glycogen particles depended on the degreee of cellular differentiation. Both mesenchymal cells and chondroblasts contained single particles, while the glycogen particles of chondrocytes aggregated and formed a series of interconnected particles.

    Notes: Summary An initial parenteral injection of adrenalin-thyroxin (1 injection daily for 5 days) followed by a high lipid diet induces an unusual, rapidly occuring atheroma in the rabbit aorta. Previous ultrastructural and histoenzymologic studies of the same model have shown obvious intimal and medial changes, apparently due to the hormonal treatment alone. Repair occurs quickly. The mechanism of atherogenesis is uncertain. The present study reports the occurence of obvious atherosclerosis at later stages in this model, when studied by the same methods. The changes are compared with those induced by hormonal treatment alone, allowing some insight into the additional effects of adrenalin and cholesterol on rabbit aorta: — two sets of 6 experimental animals were killed after one month. In the first group, receiving hormonal treatment alone, the vascular wall is not completely repaired and a change in the histochemical perfusion gradient (elevated level of hydrolase activities of the outer layers of the vessel) remains. In the second group, receiving hormonal treatment and the lipid diet, atheromatous plaques containing lipid containing cells appear at intimal level. These cells develop by migration of myocytes from the medial layer. Cholesterol seems to be the main factor inducing the smooth muscle cell migration and lipid deposition and its proliferative effect is increased by the elevated vascular permeability. — after adrenalin-thyroxin, 6 animals received the lipid diet for one month and were sacrificed after a further month. Atheromatous plaques were still present and contained higher lipid levels. Medial scarring may disturb the parietal gradient and increase the intimal lipid accumulation found.

    Notes: Summary Partial hepatectomy was carried out on Xenopus laevis to investigate its influence on the endocrine system. In addition to other endocrine effects, a marked hypertrophy and stimulation of the interrenal gland was observed. Activated cells contain mitochondria with extended and irregularly coiled tubules embedded in a low electron dense matrix. Hepatectomy induces two phases of proliferation [3 and 35 days postoperative (p.o.)]. After 106 days p.o. giant mitochondria possessing narrow and closely packed, parallel tubules surrounded by an electron dense matrix indicate a phase of inactivation. The smooth endoplasmic reticulum and the Golgi apparatus proliferate after hepatectomy. During activation the high lipid content seen in controls is decreased significantly.

    Notes: Summary The ecchordosis physaliphora, a small gelatinous mass attached to the midline of the clivus, is characterized ultrastructurally by glycogen-laden intracytoplasmic vacuoles, focally distended endoplasmic reticulum and perinuclear cisterns with cytoplasmic invaginations, large clusters of granular endoplasmic reticulum interdigitating with mitochondria, and an abundant extracellular space. These morphologic features are also present in the 9-day embryonal chick notochord and the human chordoma and serve to reaffirm the derivation of the ecchordosis and chordoma from notochordal rests.

    Notes: Summary The present study reports unusual, long, tubular, rod-shaped cytoplasmic inclusions found in the stromal cells of a cerebellar hemangioblastoma, associated with von Hippel-Lindau syndrome. These inclusions have two layers of circular, laminated filaments, and longitudinally arranged filaments; and appear similar to the so-called “Hirano bodies”, which have been previously found in normal and various pathologic conditions of the brain tissue, skeletal muscle cells in myopathy, and interstitial cells of the testis. A review of previous ultrastructural studies of cerebellar hemangioblastoma shows several different types of cytoplasmic inclusions, but none are found to be identical to the present report.

    Notes: Summary A muscle biopsy of a boy of 14 years presenting clinically a benign congenital myopathy showed granular intrasarcoplasmic bodies exhibiting reducing activity. They resemble the bodies described by Brooke and Neville in muscle of two children with severe congenital myopathy which they referred to as “reducing body myopathy”. If the reducing bodies are the characteristic morphological feature peculiar to this newly recognized congenital myopathy, the case reported here would be therefore a benign form of reducing body myopathy. The origin and nature of the granular material forming the bodies is doubtful. The close relationship between this material and the myofilaments may suggest that the granules arise from some myofibrillary component.

    Notes: Summary The ultrastructural characteristics of the cerebellum and olfactory bulb capillaries were studied in 12 human specimen measuring 25–200 mm. Type I capillaries, whose lumina were wider than 8 μ contained no pores or fenestrations. The basement membrane was sometimes discontinuous. In that case, junctions without quintuple-layered zones could be observed. Such capillaries may correspond to primary vessels in which the sinusoid character has disappeared. Elsewhere, the basement membrane was continuous and the interendothelial junctions always contained quintuple-layered zones. Type II capillaries, whose lumina measured between 2 and 8 μ had basement membranes that were either discontinuous or continuous. They were identical to the immature capillaries described previously in numerous species including man. Type III capillaries, rarely observed, were characterized by the presence of several endothelial cells with abundant cytoplasm, which limited the lumina from 0.5–3 μ in diameters. They had continuous basement membranes. Type IV and V capillaries has small or non-patent lumina. The basement membrane was absent or rudimentary. These capillaries appeared to correspond to simple and complex endothelial sprouts. In Types II, III, V and most Type IV capillaries, the interendothelial junctions contained quintuple-layered zones. No mature capillaries were observed. The hypothetical pathways of development of cerebral capillaries and the degree of permeability of immature interendothelial junctions are discussed. The long duration of maturation in man probably accounts for the diversity of capillary formations observed as compared with animals. The co-existence over a long period of time of several capillary types with probably varying permeability may play a significant role in the maturation of the blood-brain barrier.

    Notes: Summary A previously healthy middle aged man died following a 6 month illness which presented with middle ear symptoms, apparently resolved, and then 2 months later manifested as encephalitis. The illness was characterized initially by depression and intellectual deterioration. No family member or working associate was affected. The clinical diagnosis of viral encephalitis was confirmed by brain biopsy but no virus was isolated in the laboratory. Numerous intracisternal toroidal virus-like particles were demonstrated by electron microscopy in the perikarya and dendrites but not in glia. The particles resemble, but are not identical to, the oncornaviruses associated with spontaneous and induced murine neoplasms. The resemblance of these structures to the intracisternal toroidal type “A” virus of murine leukemia is noted and other possible causes for this atypical meningoencephalitis are discussed.

    Notes: Summary In a woman, aged 52, who had impaired phosphate excretion and low serum calcium levels, abundant calcium deposits (Pseudokalk) were found between the basement membranes of blood vessels in the regions of corpus striatum and nucleus dentatus as well as in the subcortical white matter and centrum semiovale. Calcium deposits were found also outside blood vessels but always in teh vicinity of the basement membrane. These can be phagocytozed by makrophages or astrocytes. Calcium deposits have a characteristic ultrastructure. They are built up of 140–400 Å electron lucent filaments (acid mucopolysaccharids?) within which electron dark segments built up of 40–80 Å units (calcium deposits?) are found. The concentric rings of calcified deposits are of reflection of differing density of aggregation of dark filaments. The growth of the deposits takes place by additional precipitation. In the development of calcification of the cerebral blood vessels in Fahr's disease, the role of high serum phosphate levels, the increased permeability and dysfunction of mesenchymal cells of the vessel walls are discussed.

    Notes: Summary A secondary demyelination process was brought about in the optic tract by unilateral enucleation of 10 adult rats which were sacrificed at 8, 15, 20, 30 and 45 days respectively, after the intervention. The Marchi reaction, which is identifiable by the presence of granular bodies, is positive at all stages, but tends to lessen towards the 45th day. The granular bodies are inside the disintegrating myelin sheath, and gradually fill the space made available by the degenerate axoplasm. The granular bodies are at first bulky and compact before breaking up and disappearing. The remains of the myelin sheath are then resorbed. Satellite cells occur from the 15th day onwards. The interest of the Marchi reaction for the tracing of nervous pathways is confirmed by these findings.

    Notes: Abstract Intracytoplasmic membranes isolated from Rhodospirillum rubrum, mutant strain VI, were extracted with the detergent lauryl dimethyl amine oxide. Subsequently two fractions were isolated, one of which contained reaction centers and the other contained light-harvesting bacteriochlorophyll of the photosynthetic apparatus. The two fractions are compared with unextracted membranes on the basis of protein patterns obtained by different methods of polyacrylamide gelelectrophoresis. Electron micrographs of the light-harvesting bacteriochlorophyll fraction reveal the presence of vesicular membrane structures. The only difference between such membranes and unextracted membranes is identified after freeze etching. While unextracted membrane surfaces are studded with particles extracted membranes exhibit a smooth surface.

    Notes: Abstract Bacteriophage 16-6-12 of Rhizobium lupini has a long, non-contractile tail and a head which is hexagonal in outline. The tail is 140 nm in length, 11 nm in diameter, and carries a short terminal fiber. Analysis of the tail structure by optical diffraction indicates that it is of the helical “stacked disc” type. After phenol-extraction from purified particles, the DNA of phage 16-6-12 can circularize in vitro. No significant difference in contour length was observed between the linear (14.34±0.28 μm) and circular (14.44±0.24 μm) forms of molecules. After partial denaturation with alkali an AT-GC-map was constructed, which shows an asymmetric distribution of AT- and GC-rich regions. It is concluded that this phage DNA can circularize due to the presence of cohesive ends and that it is not circularly permuted.

The following developmental steps were recognized: 1) contact between oogonial and antheridial initials; 2) penetration of the antheridial initial by the oogonial initial; 3) reemergence of the oogonial initial; 4) oogonial expansion; 5) gametangial delimitation and oogonial wall thickening; 6) penetration of the oogonium by the antheridial fertilization tube; 7) oosphere formation; 8) periplasm degeneration and outer oospore wall formation; and 9) inner oospore wall formation. Electron micrographs were obtained of steps 3–9. Steps 1 and 2 were reconstructed from subsequent events. Steps 3–6 are stages of active wall formation with clear indication of intensive dictyosome activity leading to the formation of numerous wall-destined vesicles of two different sizes and electron densities. No vesicles were seen associated with the development of the inner oospore wall; however, by this stage of development the oosphere cytoplasm exhibited an overall intense electron density that obscured fine detail. Cytoplasmic appearance changed enormously during differentiation, from a developing oogonium rich in mitochondria, ribosomes, rough endoplasmic reticulum, dictyosomes and their vesicles, through an oosphere filled with large “finger-print” vacuoles and lipid-like bodies, to a mature oospore with a large central vacuole (ooplast) surrounded by a cortex of numerous lipid-like bodies; other organelles are confined to the interstitial space between these storage bodies.

Elektronemikroskopische Untersuchungen an jungen Zellen zeigten Mesosomen in typisch eingerollter Form. In alternden Zellen konnten stattdessen lose gerollte oder langgestreckte, abgeplattet-tubuläre Membransysteme gefunden werden, die als degenerierende Mesosomen gedeutet wurden. Durch Lysozym- oder Ultraschall-behandlung dieser Zellen konnten tubuläre Strukturen isoliert und durch differentielle Zentrifugation angereichert werden. Elektronenmikroskopische Aufnahmen solcher Anreicherungen zeigten lange, abgeplattete Röhren, die gelegentlich an einem Ende geschlossen erschienen. Ihr Durchmesser betrug 34±5 nm. Sie waren mit einer Substanz ausgekleidet, die durch Trypsin abgebaut werden konnte, wobei eine elektronentransparente Matrix freigelegt wurde. Isolierte Tubuli zeigten in einigen Fällen einen periodischen Feinbau aus ellipsoiden Untereinheiten. Die lichtoptische Diffraktions-analyse ergab ein Gitter von Untereinheiten, die in Schrauben mit einer 27°-Steigung angeordnet sind; die Dimensionen der Elementarzelle betragen 112×56 Å. Die Proteinnatur der Gitterkomponenten wurde aus ihrer Trypsinempfindlichkeit gefolgert. Es wird postuliert, daß diese Proteinkomponenten auf einer tubulären Membranmatrix aufgelagert sind. Form und Feinstrukturparameter unterscheiden die Tubuli deutlich von einer periodisch aufgebauten Schicht der P. rhodos-Zellwand mit tetragonalem Gitter sowie von den “polyheads” und “polysheaths” defekter Bakteriophagen. Ihre mögliche Entstehung aus intakten Mesosomen wird diskutiert.
    Notes: Abstract During a 10 day-incubation on agar surfaces at 30°C, cells of the gram-negative soil bacterium Pseudomonas rhodos pass through three phases distinguishable by physiological and morphological criteria. When viewed by electron microscopy, typically “rolled” mesosomes could frequently be observed in young cells. In aged cells instead, loosely rolled or stretched-out, flattened tubules could be discerned, presumed to be degenerate mesosomes. Tubular flattened structures have been isolated from these cells by lysozyme treatment or sonication and were concentrated by differential centrifugation. Electron micrographs of these preparations showed long, straight tubules which sometimes appeared sealed at one end. Their width was 34±5 nm. They contained a lining of material, which could be digested by trypsin leaving behind an electron-transparent matrix. In rare cases, isolated tubules showed a periodic fine structure composed of ellipsoidal subunits. Optical diffraction analysis yielded a lattice consisting of subunits arranged in helices of pitch-angle 27°; the unit cell dimensions were shown to be 112×56 Å. Owing to their sensitivity to trypsin, components of the regular lattice are supposed to consist of protein. It is postulated that these protein components are layered onto a tubular membrane. These tubules are clearly distinguishable by their shape and fine structure from the periodic structure of a P. rhodos cell wall layer, which exhibits a tetragonal pattern, and also from polyheads and polysheaths of defective bacteriophages. Their possible origin from intact mesosomes is discussed.

    Notes: Abstract Development of microbodies in Candida tropicalis pK 233 was studied mainly by electron microscopical observation. The yeast cells, precultured on malt extract, scarcely contained microbodies and showed very low catalase activity. When the precultured cells were transferred to a n-alkane medium and incubated with shaking, the number of microbodies increased and concomitantly the activity of catalase was enhanced. That is, both the area ratio of microbodies in the cell and the ratio of microbodies to cytoplasm in area increased significantly during the utilization of n-alkanes for 8 hrs. Localization of catalase in the microbodies was demonstrated cytochemically by use of 3,3′-diaminobenzidine, but other organella in the cell, except for vacuoles appearing in the early growth phase and mitochondria, were not stained with this reagent. Microbodies seemed to grow by division. Biogenesis of microbodies in the yeast cells is also discussed.

    Notes: Abstract Results of an investigation of the stability of n-layers of barium stearate, cadmium arachidate and tripalmitin by means of electron microscopy and attenuated total reflection infrared spectroscopy are reported. Odd and even numbered barium stearate n-layers with n=1,2,3.4,5 are found to rearrange spontaneously from a regular film into ultrastructures of irregular, flat islands of varying thickness. The kinetics of the phase transformation of the first layer depends on the substrate, that of n-layers appears to be dependent on n, the temperature, and the surrounding medium. The kinetic behaviour of odd and even numbered layers is distinctly different. Similar studies on cadmium arachidate layers reveal much slower kinetics of the rearrangement process. In the case of tripalmitin n-layers it is shown that electron microscopy and infrared spectroscopy yield valuable complementary information about ultrastructure and molecular structure of the layers in correlation with the rearrangement process, which also occurs with this system. Consequences of the results of this paper for work published in various fields are briefly discussed.

    Description / Table of Contents: Zusammenfassung Die Untersuchungen zur Ultrastrukturpathologie der Colitis ulcerosa wurden an einem Kollektiv von 37 Kolitis-Patienten durchgeführt. Unter den epithelialen Veränderungen fallen vor allem Alterationen der Mikrovilli und der Glykokalyx der oberflächlichen Zylinderzellen auf. Möglicherweise könnten diese als morphologisches Substrat eines zumindest partiell gestörten „Mucosablockes” im Bereich der Oberflächenepithelien (IgA- und “secretory piece”-Mangel) gewertet werden. Hypothetisch wird dieser partiell gestörte „Mucosablock” als wesentliches pathogenetisches Moment bei der Colitis ulcerosa angenommen. Das Entzündungsinfiltrat des Stratum proprium mucosae ist vor allem durch zahlreiche Lymphocyten, Plasmazellen und Makrophagen ausgezeichnet. Aus der hierbei gefundenen engen topographischen Zuordnung dieser Zellen zueinander läßt sich eine funktionelle Kooperation bei dem Autoimmunprozeß vermuten, wie er bei der Colitis ulcerosa diskutiert wird.
    Notes: Summary The ultrastructural pathology of ulcerative colitis was investigated on a group of 37 colitis patients. Among the epithelial changes, the alterations of the microvilli and of the glycocalyx of the surface epithelium are quite evident. These alterations may possibly be understood as a morphological substrate of a partly impaired “mucosa block” of the surface epithelium (IgA- and “secretory piece” deficiency). Hypothetically, this partly impaired “mucosa block” is considered to be an essential pathogenetic moment of ulcerative colitis. The inflammatory infiltrate of the stratum proprium mucosae is characterized in particular by numerous lymphocytes, plasma cells and macrophages. The close topographical interrelation of these cells observed here seems to indicate a functional cooperation in the auto-immunological process, as it is discussed here in connection with ulcerative colitis.

    Notes: Summary The ultrastructure of calcareous deposits in a case of Sturge-Weber disease is described. Concrements were found mainly extracellularly, outside of blood vessels, but there were some also within the vessel walls. The laminated concrements are built up of fine filaments, similar to those seen in the Fahr's disease. Needle-like cristals (calcium apatit) were observed within the concrements. Increased permeability of the altered vessel walls may be responsible for these alterations.

    Notes: Summary The epithelioid cells forming the granulomata of Crohn's disease show striking vacuolation from the coalescing of pinocytotic vesicles with double membrane-bound bodies and their subsequent fusion with lysosomal dense bodies. The fine detail of the granulomata in each individual is uniform but varies from case to case in such a way as to suggest an episodic stimulation of pinocytosis and vacuole formation. The production of such granulomata with these cyclical appearances is consistent with an intermittent stimulus by exogenous antigen or antigen antibody complexes arriving in the diet.

    Notes: Summary Morphologic studies of pituitary neoplasms removed by surgery from 36 human patients revealed 8 chromophobe adenomas which differed clearly from the remaining tumors. The cytoplasm of the adenoma cells failed to stain with PAS, aniline blue, aldehyde fuchsin, aldehyde thionin, orange G or light green, but positively stained granules were found by using erythrosine or carmoisine. Immunoperoxidase technique disclosed the presence of prolactin in the cytoplasm of some adenoma cells. The adenoma cells exhibited distinct ultrastructural features such as well developed rough surfaced endoplasmic reticulum with Nebenkern formation, prominence of Golgi apparatus, presence of misplaced exocytosis as well as pleomorphism of secretory granules with a considerable variation of size ranging from 130 to 500 nm in diameter. Thus, by electron microscopy the adenoma cells showed a close resemblance to prolactin cells of the non-tumorous pituitary glands except for the reduced size and number of secretory granules. These chromophobe adenomas are regarded as representing a distinct pathological entity clearly distinguishable from other forms of pituitary neoplasms. In view of the morphologic findings and the elevation of blood prolactin level (measured in 3 patients) the term, “sparsely granulated prolactin producing pituitary adenoma”, appears to be the most appropriate one to designate these tumors.

Nach intraperitonealer k-Strophanthin-Prämedikation in therapeutischer Dosierung (2 × 10−5 g/kg Körpergewicht) nehmen Zahl und Ausdehnung der Nekrosen statistisch signifikant zu. Diese Befunde sprechen gegen einen präventiven Schutzeffekt des k-Strophanthins und erklären sieh aus dem biochemischen Wirkungsmechanismus der Katecholamine und Glykoside, indem beide Substanzen gleichsinnig eine Erhöhung des mobilisierbaren intracellulären Calcium der Herzmuskelfaser bewirken. Eine Strophanthin-Prämedikation bewirkt bei den durch Arterenol induzierten Myokardfasernekrosen weder im Verteilungsmuster der Nekrosen noch im elektronenmikroskopischen Befund eine Änderung.
    Notes: Summary Female rats with a body weight of approximately 200 g regular show necroses of myocardial fibers after subcutaneous injection of 1-Noradrenaline (2.5 mg/kg body weight). After intraperitoneal premedication with Strophanthin k in a therapeutical dosage (2 × 10−5 g/kg body weight), the increase in number and extent of the necroses is statistically significant. These findings argue against a preventive effect of Strophanthin k and can be explained by the mode of action of catecholamines and glycosides, as both substances, in a like manner, cause an increase in the mobilizable intracellular calcium of the myocardial fiber. Premedication with Strophanthin causes no alteration in the distribution pattern of necroses, nor in the electron-microscope findings in the case of myocardial necroses produced by Noradrenaline.

Die folgenden Mißbildungstypen lassen sich beschreiben: 1. Akrosomfehlbildungen in Kontakt zum Spermatidenkern: bläschenförmige Einschlüsse in der Akrosomblase, asymmetrische Verdickungen der Akrosomkappe, Einfaltungen der akrosomalen Lamina in den Spermatidenkern, teilweise Abhebung des Akrosoms vom Kern, apikale knopfartige Verdickungen des Akrosoms. 2. Akrosomfehlbildungen unabhängig vom Spermatidenkern: das akrosomale Material ist enthalten in kleinen Bläschen, ringförmigen Strukturen, in rundlichen oder schalenartigen Formen. Der Kern solcher Spermatiden hat kein Akrosom. 3. Fehlbildungen des Kerns: Einschlüsse von Bläschen oder Membranen, ausgedehnte Membranbildungen, sehr große Kernvacuolen, fehlende Kondensation des Karyoplasma. 4. Mehrkernige Spermatiden: zwei oder mehr Kerne in einem Perikaryon sind durch ein Akrosom verbunden. 5. Doppelte Akrosomanlagen. 6. Fehlbildungen des Schwanzes: Zerstörung des Axonema, Unterdrückung oder Überzahl von Tubuli und Fasern, doppelte axiale Filamentenbündel, Destruktion der Halsregion, Fehlen der Mitochondrienscheide, Auftreibung der Faserscheide. Mißgebildete Keimzellen treten in unterschiedlichen Prozentsätzen sowohl bei gesunden Männern als auch bei infertilen Patienten auf. Damit ist das Vorkommen fehlgebildeter Keimzellen nicht unbedingt ein Hinweis auf eine bestehende Infertilität. Nur bei bestimmten Mißbildungstypen, wie den rundköpfigen Spermatozoen, ist — wenn alle Spermatozoen im Ejaculat den gleichen Defekt aufweisen — mit Infertilität zu rechnen.
    Notes: Summary Malformed spermatids and spermatozoa in the testes, epididymes, and ejaculates of 29 apparently healthy men and in 171 patients suffering from different andrological diseases were investigated with the electron microscope. The following types of malformations are described: 1) Malformations of the acrosome in contact with the nucleus of the spermatid: Vesicular inclusions in the acrosome vesicle, asymmetric thickenings of the acrosome cap, enfolding of the acrosomal lamina into the spermatid's nucleus, partial separation of the acrosome from the nucleus, apical knob-like thickenings of the acrosome. 2) Malformations of the acrosome independent from the spermatid's nucleus: the acrosomal material is contained in small vesicles, large vacuoles, in ring-like structures, in globular or shell-like laminar structures. The nucleus of such spermatids lacks an acrosome. 3) Malformations of the nucleus: inclusions of vesicles or membranes, excessive packages of membranes, gigantic nuclear vacuoles, lack of condensation of the caryoplasm. 4) Multinucleate spermatids: two or more nuclei within the pericaryon of one spermatid are joined by an acrosome. 5) Duplicate acrosome anlagen. 6) Malformations of the tail: disorganization of the axonema, a lack or excessive numbers of tubules and fibres, doublets of the axial filaments, destruction of the neck region, a lack of the mitochondrial sheath, and enlargement of the fibrous sheath. Malformed germ cells were observed to occur in varying amounts both in apparently healthy men and in those who were apparently infertile. This indicates that germ cell malformations are no absolute indicator of male infertility. However, certain distinct types of malformations (such as round-headed spermatozoa) are definitely associated with infertility, but only if all spermatozoa in the ejaculate of a patient have the same defect.

    Description / Table of Contents: Zusammenfassung Die maligne Hyperthermie ist charakterisiert durch einen rapiden Anstieg der Körpertemperatur, Muskelkontraktionen und zunehmende Hypoxie. Sie ist eine moderne Narkosekomplikation mit einem sehr hohen Letalitätsrisiko von über 60%. Da über die morphologischen Veränderungen bei maligner Hyperthermie sehr wenig bekannt ist, wurden an Muskelbiopsien von 3 Patienten mit maligner Hyperthermie und einem sog. Risikopatienten histologische, histochemische und elektronenmikroskopische Untersuchungen durchgeführt. Histologisch zeigten sich akute Muskelfasernekrosen, außerdem wurden Hinweise für eine latente Myopathie erhoben. Histochemisch ergab sich, daß beide Muskelfasertypen I und II befallen waren. Mit der Phosphorylase-Reaktion konnten auch bei Fasern, die bei üblicher histologischer Bearbeitung unauffällig waren, pathologische Veränderungen nachgewiesen werden. Elektronenmikroskopisch konnten neben einer akuten Rhabdomyolyse eine Erweiterung der Zisternen des sarkoplasmatischen Beticulums und eine eigentümliche Einrollung bzw. Wucherung des Sarkolemms beobachtet werden. Die Befunde unterstützen die pathogenetische Vorstellung anderer Autoren, daß dieses Syndrom, das durch Inhalationsnarcotica und Muskelrelaxantien (Halothan bzw. Succinylcholin) ausgelöst wird, auf der Grundlage eines Defektes caloiumspeichernder Membranen des sarkoplasmatischen Reticulums beruht. Wegen des familiären Auftretens dieser Komplikation sollte angestrebt werden, klinische, laborchemische und morphologische Untersuchungen auch bei den Verwandten solcher Patienten zum Ausschluß einer derartigen latenten Myopathie durchzuführen.
    Notes: Summary Malignant hyperthermia is a rare but severe complication of modern anesthesia, induced by halothane and succinylcholine. The syndrome is characterized by a rapid sustained and extreme rise in body temperature associated with muscular rigidity, tachycardia, tachypnoea and cyanosis. The lethality is about 60%. The present paper describes the histological, histochemical and electron microscopical findings performed on muscle biopsies of 3 patients with malignant hyperthermia (1 patient died) and a so called risk patient. In all patients morphological findings consistent with a pre-existent myopathy were found. Histologioally there were acute necrotic muscular fibers as well as in types I and II, variations in the fiber diameter and centralization of the nuclei. In two cases even fibers that had a normal aspect in HE slides, showed a pathologic pattern after phosphorylase reaction. In addition to acute rhabdomyolysis, electron-microscopic investigations revealed cystic expansion of the cisterns of the sarcoplasmic reticulum with a peculiar proliferation of the sarcolemma. In a degenerating mitochondrium, a crystalline inclusion was identified. These findings support the pathogenetic concept of Britt and coworkers of a functional defect in the calcium release or binding mechanism of sareoplasmic reticulum. Since it is known that malignant hyperthermia has a familial predilection, it seems very important that clinical, biochemical, and morphological investigations be performed such as CPK estimations and muscular biopsies not only of the patients but also of the relatives in order to rule out this type of latent myopathy.

    Notes: Summary After i.v. injection in the rat, d-tubocurarine is taken up and concentrated by the liver. A method is developed for the visualisation of d-tubocurarine inside the liver cell by electron microscopy. Glutaraldehyde fixed liver blocks were immersed in an ammonium molybdate solution; d-tubocurarine was precipitated at sites of high concentration by molybdate, to form an insoluble d-tubocurarine-molybdate complex. This precipitate was found predominantly at the surface of lysosome-like particles, but also inside these organelles. In subcellular fractionation experiments, d-tubocurarine was found with a high relative specific “activity” in the lysosomal fraction, lending support to a lysosomal localisation of d-tubocurarine.

Veränderungen in den akustischen Kernen wurden licht- und elektronenmikroskopisch untersucht. 5–55 Tage nach Schallexposition wurde Phagocytose an degenerierenden N. acusticus Endigungen und von Neuronen zweiter Ordnung beobachtet. Es werden verschiedene Hypothesen diskutiert, eine hinreichend überzeugende Erklärung für diese Veränderungen konnte jedoch nicht gefunden werden.
    Notes: Summary Guinea pigs were exposed to the noise of 40 shots of an alarm pistol held at a distance of about 60 cm. The ventral cochlear nuclei were studied in phase contrast and electron microscopy after both short survival periods and longer periods of up to 55 days survival. Marked degeneration of primary cochlear nerve endings and of synapting secondary neurons of the posterior caudal part of the ventral cochlear nucleus (AVCN) and the octopus cell area (OCA) of the posterior ventral cochlear nucleus (PVCN) was found most distinctly after 5–55 days. As criteria of degeneration of the second neuron of the afferent auditory pathway we used: 1. The loss of the synapting nerve endings, mainly “shrinking”. 2. The formation of huge mitochondria in the second order neurons and their dendrites. 3. The phagocytosis by glial cells of nerve endings, of the second order neurons and of their dendrites. After 5 days survival time no distinct changes were found in the granular cell area of PVCN, where as all stages of degeneration could be found in OCA at this time. In the discussion of these findings it is concluded that additional studies of the morphology of the cochlear nuclei seem necessary, as these may lead to a better understanding of the pathology of hearing following heavy noise exposure.

    Notes: Summary Olfactory epithelium in Tiliqua scincoides scincoides is of a loosely packed pseudostratified type. It receives secretion from the supporting cells and the underlying glands of Bowman. Its surface bears microvilli and cilia from sensory cells and microvilli from supporting cells. The vomeronasal epithelium is also pseudostratified but higher and more closely packed. Its surface carries microvilli from sensory and supporting cells but lacks cilia. Vascular connective tissue penetrates it almost to the epithelial surface but is always outlined by basal cell processes and a basal lamina. There are no secretory cells in or under the sensory epithelium but some cells in the epithelium of the mushroom body contain secretion granules. Sensory cells of both epithelia are bipolar neurons. The perikarya of the vomeronasal cells are more neuronal in character. Axonic processes are similar in both, dendrites are distinctive. Olfactory dendrites end in rounded rods bearing microvilli and cilia of an unusual type. Microvilli with filamentous cores occur on vomeronasal dendrites. There are no cilia, but 2–6 centrioles appear below the cell surface. Basal cells are structurally similar in both epithelia, but axonic processes of olfactory cells are surrounded by supporting cell processes, while vomeronasal axonic processes are surrounded by basal cells before they leave the epithelium. The presence of cilia and microvilli on the surface of the sensory cells is discussed in relation to the physical conditions surrounding them.

    Notes: Summary During nuclear elongation in spermatids of Gambusia affinis, a deep fossa is formed at the base of the nucleus in which the centriolar complex and proximal portion of the flagellum reside. To stabilize the positional relationship between the nucleus and centriolar complex, while nuclear morphogenesis is taking place, a series of microtubules develop which emanate from the centriolar complex and extend to the nuclear envelope lining the fossa. Buttressing microtubules also develop within the nuclear fossa which both originate and insert along the nuclear envelope. These appear to stabilize nuclear shape prior to the time when chromatin condensation has proceeded to the stage where it could lend structural stability to nuclear form. Microtubules develop only after specific nuclear morphogenic events have taken place. It is therefore concluded that the spermatid nucleus is capable of “self-assembly” involving microtubules in a supportive role in addition to stabilizing the nuclear-flagellar relationship in G. affinis. The pattern of nuclear fossa-associated microtubules in G. affinis is significantly different from that observed in other poeciliid teleosts indicating a degree of species specificity with regard to both the timing of appearance and total number of microtubules.

    Notes: Summary Isolated islets of Langerhans from mice were maintained in tissue culture for one week at either a high (28 mM) or a low (3.3 mM) extracellular glucose concentration. Electron microscopic morphometry by means of stereological methods revealed a much greater volume of mitochondria in islet cells cultured at low glucose than in those cultured at high glucose. The former islets also showed a higher activity of the mitochondrial marker enzyme, L-3-hydroxyacyl-CoA-dehydrogenase (E.C.1.1.1.35). These results indicate a true mitochondrial hypertrophy at the low glucose concentration. Although it is known from previous studies that the islet cell metabolism is diminished after low-glucose culture, the present observations of an increased mitochondrial volume probably do not reflect a degenerative process, but rather adaptive changes towards oxidation of energy yielding substrates other than glucose.

    Description / Table of Contents: Zusammenfassung Licht- und elektronenoptische Untersuchungen wurden an Muskelbiopsien von 4 Fällen hypokaliämischer periodischer Paralyse durchgeführt. Die frühesten Veränderungen betreffen nach Meinung der Verfasser das sarkoplasmatische Reticulum: Vacuolisierung des Längssystems und vermutlich Multiplikation des Quersystems. Es wird vermutet, daß die Vacuolenbildung die Folge einer Fusion der Vesikeln des Quersystems darstellt. Destruktion von Myofibrillen und Vergrößerung der, interfibrillären Räume treten erst in späteren Phasen des Krankheitsprozesses auf. Ausdehnung und Intensität dieser Veränderungen bedingen das typische histologische Bild myogener Läsionen bei Fällen m klinisch fixierten Symptomen der Myopathie.
    Notes: Summary Light and electron-microscopic studies were performed on the muscles taken from 4 cases with hypokalemic periodic paralysis. The authors conclude that the earliest changes concern the sarcoplasmic reticulum: vacuolisation of the longitudinal system and probably, multiplication of the transverse system. It is suggested, that vacuoles are formed as a result of fusion of the vesicles of the transverse system. Destruction of myofibrilles and enlargement of the interfibrillar spaces appear at a later period of the disease process. Extensive and intensive changes of that kind present a typical appearance of myogenic changes in histological study observed in the cases with clinically fixed symptomes of myopathy.

Lichtmikroskopisch bestanden die Herde aus knötchenförmigen Anhäufungen von Makrophagen, deren Cytoplasma mit Hämatoxylin-Eosin eine charakteristische schwachblaue und nach Durchführung der Perjodsäure-Schiff-Reaktion eine stark rote Färbung zeigte. Elektronenmikroskopisch fanden sich in den Hirnläsionen Bacillen, die morphologisch den im Darm beschriebenen glichen und die auch dieselben Degenerationserscheinungen aufwiesen. Die Perjodsäure-Schiff-Reaktion färbt Kapselmaterial und Zellwände,die auch nach Untergang der Bacillen noch nachweisbar sind. Diese Beobachtungen werden als weiterer Beweis für die infektiöse Ätiologie der Whippleschen Krankheit angeführt.
    Notes: Summary Among the 21 autopsied cases of Whipple's disease on file at the Armed Forces Institute of Pathology, 4 had neurological symptoms and prominent involvement of the central nervous system, By light microscopy the lesions were composed of nodular aggregates of macrophages having cytoplasm that stained a distinctive pale blue with hematoxylin-eosin and bright red with the periodic acid-Schiff technique. By electron microscopy the cerebral lesions revealed bacilli morphologically identical to and undergoing the same sequence of degenerative changes as those observed in the intestine. The periodic acid-Schiff reaction stains the walls and the capsular material that persists even after intact organisms disappear. These observations further support the infectious nature of Whipple's disease.

Obwohl diese Veränderungen eine Nervenzellerkrankung anzeigen dürften, werden sie wegen ihrer Ähnlichkeit sowie wegen des Vorliegens anderer, spezifischerer Zellveränderungen als Sekundärphänomene gedeutet. Im ersten Fall wird als primäre Läsion eine Verdickung der Basalmembran der Gefäße, im zweiten Fall eine Astrogliaschwellung angenommen. Da diese Strukturen wichtige Transportfunktionen haben dürften, könnten diese Anomalien leicht die neuronale Versorgung stören und dadurch die neuronalen Läsionen verursachen. Die Bedeutung des Glykogens in diesen Strukturen wird diskutiert.
    Notes: Summary The electron microscopic and histochemical findings in biopsies of two cases of subacute dementia have revealed neurons containing numerous lipofuscin bodies and increased acid phosphatase. Although these changes are considered to indicate neuron disease, they are believed to represent a secondary process because of their similarity and because other more distinctive cell alterations are present. In the first case, the primary abnormality is believed to be thickening of the vascular basement membrane, while in the second case, astroglial dilatation is considered a primary change. Since these structures appear to have important transport functions, these abnormalities could easily impair the nutrient supply to neurons and, in this way, produce the neuronal alteration. The significance of glycogen in these tissues is discussed.

Die Anomalien der Markscheiden sowie die Häufigkeit von Desmosomen sind bemerkenswert. Diese Besonderheiten unterscheiden diesen Fall von anderen, bisher beschriebenen chronischen Neuropathien. Trotzdem erscheint es derzeit nicht möglich, eine Aussage über die Spezifität der ultrastrukturellen Befunde am untersuchten Nerven zu treffen.
    Notes: Summary The principal lesion in this case of chronic early infantile neuropathy is a modification of the Schwann cell resulting in the formation of a concentric network of lamellae formed by a double-layered sheet of basement membrane around numerous, large myelinated and unmyelinated fibers. The abnormalities in the myelin sheaths and the frequency of desmosomes are noteworthy. These peculiarities distinguish this case from other chronic neuropathies described up to now. Nevertheless, it is impossible to say at present whether the ultrastructural aspect of the nerve studied here is in any way specific.

    Notes: Summary Four cases of S.S.L.E are reported. The electron microscopy findings show: 1. Tubulary inclusion bodies: They appear in three cases out of four, either in neuronal and glial nucleus or in axis cylinders. Their morphological features are similar to the nucleocapsides of myxoviruses and particularly measles-virus. 2. Other different types of nonspecific inclusions: nuclear bodies, cristalline-like rods and fibrillar bundles which may be considered as the result of a nuclear metabolic disorder, osmiophilic particles which may be seen as the result of a cellular intracytoplasmic hyperactivity. The tubules are found whatever the duration of the disease (3 months up to 5 years) may be. This might indicate that there is no autosterilization of the virus in the C.N.S. during the course of S.S.L.E. According to the most recent papers, such a disease might be interpreted as an infection indirectly induced by measles-virus with an unknown immunologic or metabolic mechanism.

    Description / Table of Contents: Zusammenfassung Bei Kaninchen mit einer experimentell erzeugten Randzonensiderose des Zentralnervensystems konnten elektronenmikroskopisch in der Großhirnrinde eigentümliche Spiralstrukturen beobachtet werden. Diese Spiralen fanden sich vornehmlich im Bereich von subpialen und perivasculären Gliosen vor. Sie bestanden aus äußerst dünnen lamellären Astrocytenausläufern, welche um andere astrocytäre Zellfortsätze oder um Dendriten aufgewickelt waren. Die spiralisierten Astrocytenausläufer wiesen in ihrem Inneren feine Filamente mit einer ziemlich regelmäßigen Anordnung auf. In radiärer Richtung ließen die astrocytären Lamellenspiralen eine periodische Schichtung erkennen, welche etwas an die von echten Markscheiden erinnerte. Es wird in Betracht gezogen, daß sie das submikroskopische Äquivalent der sog. Plaques fibromyéliniques darstellen.
    Notes: Summary Peculiar spiral structures could be observed by electron microscopy in the cerebral cortex of rabbits with an experimentally produced marginal siderosis of the central nervous system. These spirals were found predominantly within areas of subpial and perivascular gliosis. They consisted of extremely thin lamellar astrocyte extensions winding around other astroglial processes or dendrites. The spirally coiled astrocyte extensions exhibited within their interior fine filaments having a rather regular arrangement. In the radial direction, the astrocytic lamellae spirals showed a periodic stratification somewhat resembling that of true myelin-sheaths. They were considered to represent the submicroscopic equivalent of the socalled plaques fibromyéliniques.

    Description / Table of Contents: Zusammenfassung Eine Modifikation der Methode zur ultrastrukturellen Darstellung von Natrium wird beschrieben, die eine Perfusion mit leicht alkalischer Mischung von Hydroxydipaldehyd, Phosphatpuffer und Kalium-Pyro-Antimonat verwendet. Im normalen Gehirn werden keine Natrium-Deposite angetroffen. Das spricht dafür, daß im Gehirn keine intraoder extracelluläre Konzentration über 150 mM vorliegt. Bei Vorliegen hoher NaCl-Konzentration sieht man ein großes, dichtes Präcipitat, das nach direkter Injektion in den extracellulären Räumen, Neuronen und Astrocyten lokalisiert werden kann. Im Mark der vergifteten Ratten findet sich ähnlich wie nach direkter Natrium-Injektion ein diskretes, gleichfalls als Natrium aufgefaßtes Präcipitat an der Oberfläche von Oligodendrogliazellen, aber nicht innerhalb dieser Zellen. Ein elektronendichtes Deposit unklarer Herkunft findet sich in den Myelinspalten, die nach akuter TET-Vergiftung auftreten. Der Wechsel in Charakter und Lokalisation dieser elektronendichten Partikel mahnt zur Vorsicht in ihrer Interpretation bei histochemischen Untersuchungsmethoden.
    Notes: Summary A modification of the technique for the ultrastructural demonstration of sodium is described which involves a perfusion with a slightly alkaline mixture of hydroxyadipaldehyde, phosphate buffer and potassium pyroantimonate. No sodium deposits are found in normal brain indicating that no intra or extracellular concentration exceeding 150 mM is present in the brain. In the presence of high concentration of NaCl, a large dense precipitate can be seen which after a direct injection can be localized to extracellular spaces, neurons and astrocytes. In the white matter of tin poisoned rats, as well as after the direct injection of sodium, a discrete precipitate, also believed to represent sodium, is noted at the surface of oligodendrocytes but not within these cells. An electron dense deposit of dubious origin is present in the myelin clefts which occur as a result of acute triethyltin intoxication. The variations in the character and localization of these electron dense particles emphasize the caution which is required in their interpretation following histochemical procedures.

    Notes: Summary An electron microscopic study of the corpus callosum of normal and swayback lambs has revealed morphological evidence of normal myelinogenesis in both groups of animals. Axons wrapped by two cell processes were frequently encountered. Reactive changes were found in axons in the swayback lambs. The significance of these findings is discussed.

    Description / Table of Contents: Zusammenfassung Es wird über licht- und elektronenoptische Untersuchungen an Muskel-und Hirnbiopsien eines Falles von spongiöser Degeneration des ZNS berichtet. Die in der grauen und weißen Hirnsubstanz enthaltenen Vacuolen entsprechen a) Spalten in den Markscheiden infolge Aufsplitterung an der intraperiodischen Linie und b) geschwollenen Astrocytenperikaryen und-fortsätzen. Ungewöhnliche Mitochondrien mit Gehalt an kristallinem Material fanden sich nur in Astrocyten. Die ultrastrukturellen Befunde entsprechen denen des Hirnödems. Es wird angenommen, daß die Astroglia eine primäre Rolle in der Flüssigkeitsansammlung spielt, während die Markscheidenschwellung als eine Sekundärläsion aufgefaßt wird. Die mögliche Bedeutung abnormer Astrocyten-Mitochondrien wird diskutiert.
    Notes: Summary Light and electron microscopy study of skeletal muscle and cerebral biopsies from a case of spongy degeneration of central nervous system is reported. The multiple vacuoles present in cerebral gray and white matter correspond to (a) clefts within myelin sheaths resulting from splitting at the intraperiod line and (b) swollen astrocytic perikarya and processes. Unusual mitochondria containing crystalline-like material were observed only in astrocytes. The ultrastructural findings are consistent with cerebral edema. It is suggested that the astrocytes play a primary role in the fluid accumulation while the myelin swelling is a secondary lesion. The possible role of the abnormal astrocytic mitochondria is discussed.

It is suggested that this appearence, which has not been seen in any other condition, may be due to the forcing of myofibrillar material into the nucleus during its migration to the centre of the fibre invaginating but not breaching its membrane.

    Description / Table of Contents: Zusammenfassung Die häufig diskutierte, (Mit)beteiligung von Herpes simplex-Virus (HSV) bei bestimmten Alterationen im ZNS sowie der hohe Neutropismus des HSV führte zur Frage, inwieweit sich HSV auch in in vitro gezüchteten, spezifischen Zellen vom Nerven-system vermehren und züchten läßt, und ob sich hierbei licht-und elektronenmikroskopisch charakteristische Alterationen nachweisen lassen. Es zeigte sich, daß sich das HSV in allen verschiedenen angezüchteten spezifischen Zellen (von Plexus chorioideus des Rhesusaffen, Kaninchenspinalganglien, menschlichen Oligodendrogliomen, Meningeomen sowie fibrillären und protoplasmatischen Astrocytomen) kultivieren und vermehren läßt. Hierbei kommt es in den spezifischen Zellen nicht nur lichtmikroskopisch zu charakteristischen cytopathischen Veränderungen, sondern auch biologisch zu einer signifikanten Vermehrung des Virus in diesen Zellen. Auch elektronenmikroskopisch lassen sich viele der charakteristischen Bildungs-, Reifungs- und Ausschleusungsstadien des Virus in den Kernen und im Cytoplasma der Zellen beobachten. Die Besonderheiten der elektronenmikroskopisch erhobenen ultrastrukturellen Befunde werden ausführlich beschrieben, und die sich daraus abzuleitenden Möglichkeiten werden im Hinblick auf die biologischen Eigenheiten des HSV bei in vivo-Infektionen, aber auch im Hinblick auf die Deutung lichtmikroskopisch histochemischer und immunhistologischer Befunde nach in vivo-Infektionen mit diesem Virus ausführlich diskutiert.
    Notes: Summary The participation of herpes simplex virus (HSV) in certain alterations of the CNS has been partially questioned and frequently discussed. Therefore, the question arose whether HSV can also be cultivated and multiplied in vitro in CNS specific cells, and if so, whether characteristic alterations and structures can be observed by means of light and electron-microscopic examinations. Our data demonstrate that HSV multiplies in all different specific cells originating from plexus chorioideus of rhesus monkeys, from spinal ganglia of rabbit, from human oligodendroglioma, meningeoma as well as from fibrillar and protoplasmatic astrocytoma. Characteristic cytopathic alterations of these specific cells and significant multiplication of the virus in these cells are to be found. Many of the characteristic forms of formation, maturation and release of HSV can be seen by electronmicroscopic examinations. The particular ultrastructural data observed by electron microscopy are described in detail and the resulting possibilities are broadly discussed not only with regard to the biologic particularity of HSV in in vivo infection but also in view of the interpretations, deriving from histochemical and immunohistological data obtained by light microscopy after in vivo infection.

    Description / Table of Contents: Zusammenfassung Die Reaktion von kolloidalem Eisen mit Sialomucinen (Hale stain) wurde zur elektronenmikroskopischen Darstellung der neuraminsäurehaltigen Ganglioside in den subcellulären Fraktionen des Gehirns angewandt. Zunächst wurde mit Hilfe tritiummarkierter Ganglioside überprüft, ob bei der Gewinnung subzellulärer Fraktionen Ganglioside nicht unspezifisch subcellulär adsorbiert werden. Eine nennenswerte Adsorption konnte ausgeschlossen werden. Das Verfahren wurde zuerst auf die aus Meerschweinchengehirnen isolierten Fraktionen angewandt. Da Nervenendigungen, Mitochondrien und Synapsenbläschen Hale-positiv reagierten, dienten Enzyminkubationen mit Neuraminidase und Hyaluronidase zur Differenzierung von anderen Hale-positiven sauren Substanzen. Schließlich wurde die Methode auf die cytoplasmatischen multilamellären Körperchen (MCB) übertragen, die aus Gehirnen an Tay-Sachs'scher Krankheit Verstorbener isoliert wurden. Aus den Versuchen konnte abgeleitet werden, a) daß die Hale-Färbung auch Ganglioside anfärbt, b) daß Ganglioside auf der Oberfläche der Membran von den multilamellären Körperchen und mit hoher Wahrscheinlichkeit auch auf der Membran der Nervenendigungen lokalisiert sind, c) daß die Synapsenbläschen eine positive Reaktion ergeben, die jedoch nicht mit Sicherheit auf vorhandene Ganglioside zurückgeführt werden kann, d) daß die äußere Mitochondrienmembran Hale-positive Substanzen enthält, deren Natur unbekannt ist.
    Notes: Summary The reaction of colloidal ferric oxide with sialomucins (Hale stain) was applied to sialic acid-containing gangliosides in subcellular fractions of the brain, in order to investigate their localization by electron microscopy. Prior to all experiments, a check was made, by means of tritium-labeled ganglioside, to confirm that the ganglioside content in the subcellular particles was not the result of an unspecific adsorption during the isolating procedure. No considerable unspecific adsorption could be registered. Hale stain was first applied to subcellular fractions obtained from guinea-pig brains. Since nerve-endings, mitochondria and synaptic vesicles gave a positive staining reaction, incubations with neuraminidase and hyaluronidase were carried out in order to achieve a differentiation. Finally, the method was applied to the membranous cytoplasmic bodies (MCB) which were isolated from the brain of a patient with Tay-Sachs disease. It was concluded that: a) Hale stain stains gangliosides also, b) gangliosides are localized in the membrane of membranous cytoplasmic bodies and also very probably in the membrane of nerve-endings, c) synaptic vesicles give a Hale-positive reaction which cannot with certainty be attributed to gangliosides, d) the outer membrane of mitochondria contains Hale-positive substances, the nature of which is not known.

    Description / Table of Contents: Zusammenfassung Eine kombinierte licht- und elektronenmikroskopische Untersuchung am normalen N. suralis von 7 Menschen im Alter von 15–59 Jahren wurde vorgenommen. Qualitative und quantitative Beobachtungen an Schwann-Zellen und Fibroblasten, markhaltigen und marklosen Fasern wurden an isolierten Faszikeln durchgeführt. Schwannzellen überwiegen gegenüber Fibroblasten im Verhältnis von etwa 9:1. Die meisten Schwannzellen (etwa 80%) liegen an marklosen Fasern. Die Faktoren, welche die Dichte dieser Zellen pro Querschnittsareal beeinflußen, werden diskutiert. Einige ultrastrukturelle Befunde an bemarkten Fasern werden beschrieben und ihre Zahl pro mm2 sowie die Häufigkeitsverteilung ihrer Dicke wird angegeben. Eine indirekte Methode zur Bestimmung der mittleren Internodienlänge für jede der Markfasergrößenpopulationen an Querschnitten von Faszikeln normaler Nerven durch Bestimmung des Verhältnisses der markhaltigen Fasersegmente zu ihrer Kernzahl wird vorgeschlagen. Die Ultrastruktur der marklosen Nervenfaern wird beschrieben und die Identifizierung dieser Axone mit extremen Durchmessern diskutiert. Ihre Dichte und Größenfrequenzhistogramme sind die ersten, die am Menschen durch systematische elektronenoptische Untersuchungen veröffentlicht werden. Das mittlere Verhältnis von marklosen zu bemarkten Fasern ist etwa 3,7:1 und schwankt in den Faszikeln der Einzelindividuen.
    Notes: Summary A combined light and electron microscope study of the normal sural nerve in 7 people aged 15–59 years is reported. Qualitative and quantitative studies of the Schwann cells and fibroblasts, myelinated and unmyelinated fibres are made in isolated fascicles. Schwann cells predominate over fibroblasts in the ratio of about 9-1. Most Schwann cells, almost 80%, are attached to unmyelinated fibres. Factors influencing the densities of these cells per cross sectional area are discussed. Some ultrastructural features of the myelinated fibres are described and their numbers per sq.mm and frequency distribution of their sizes are produced. An indirect method is proposed for assessing the mean internodal length for earch of the myelinated fibre size populations in cross sections of fascicles of normal nerves by estimating the proportion of myelinated segments cut through their nucleus. The ultrastructure of unmyelinated fibres is described and the identification of axons of extreme diameter is discussed. Their densities and size frequency histograms are the first to be reported in man by systematic electron microscope studies. The average ratio of unmyelinated to myelinated fibre density is about 3.7:1 though it varies in the fascicles of the different individuals. The implications of axonal diameter in the presence of myelin are commented on.

    Description / Table of Contents: Zusammenfassung Nach experimenteller CO-Vergiftung fanden sich schwerere und ausgedehntere Läsionen in der weißen als in der grauen Hirnsubstanz. Zwischen akuter und chronischer CO-Vergiftung ergaben sich keine wesentlichen qualitativen Unterschiede der Hirnveränderungen. Die Läsionen nach chronischer Intoxikation waren jedoch schwerer als nach akuter Vergiftung. Die Nervenzellen zeigten leicht dilatiertes oder fragmentiertes EPR und Golgi-Komplexe, doch boten die Mitochondrien meist keine pathologischen Veränderungen. Bei chronischen Fällen fand sich gelegentlich eine deutliche Schwellung des EPR von Oligodendrogliazellen. Die Capillaren blieben intakt und die perivasalen Astrogliaendfüßchen waren nicht geschwollen. Im Mark kam es zu teilweisem Ausfall der normalen Markscheidenbestandteile. Die Marklamellen zeigten mäßige Exfoliation sowie eine Umwandlung in homogene oder strukturlose Substanz. Bemarkte Axone waren teilweise kontrahiert und zeigten auf Querschnitten unregelmäßige Form. Gelegentlich fand sich eine Vergrößerung der axonalen Mitochondrien und Zerstörung ihrer inneren Parallelstrukturen. Als Hauptfaktoren in der Pathogenese der Hirnveränderungen nach experimenteller CO_Vergiftung werden zerebrale Anoxie infolge CO-Hämoglobinbildung, cytotoxische Wirkung von CO auf das Hirngewebe und eine Störung der Blutzirkulation angenommen.
    Notes: Summary In experimental carbon monoxide poisoning, more marked and widespread pathological changes in the brain were seen in the white matter as compared with the gray matter. No essential difference, in the appearance of cerebral alterations, was detected between acute and chronic carbon monoxide poisoning. It was confirmed that the changes resulting from chronic poisoning showed a higher degree of severity as compared with those resulting from acute poisoning. Nerve cells with slightly dilated or fragmented endoplasmic reticulum and Golgi complex in the gray matter were found infrequently. However, most mitochondria did not show any kind of abnormality in such cells. A remarkable dilatation of the endoplasmic reticulum of oligodendrocytes was sometimes seen in chronic cases. Capillaries remained intact and astrocytic perivascular foot-plates were not swollen. In the white matter, the normal constituents of the myelin sheath were partially lost and the myelin lamellae showed moderate exfoliation, or a change into homogenous or structureless substance. Myelinated axons were partially contracted and showed an irregularity in shape in transverse sections. Enlargement of occasional axonal mitochondria and the destruction of their internal parallel structures were observed. Cerebral anoxia caused by carboxyhaemoglobin, together with the cytotoxic action of carbon monoxide on the brain tissue and the disturbance of blood circulation in the brain are suggested as the chief factors in the pathogenesis of the cerebral alterations induced by experimental carbon monoxide poisoning.

Es wird vermutet, daß Natriumacid den Wasserstoffwechsel beeinträchtigt, wobei es eine osmotische Ausgleichstörung im Gehirn erzeugt.
    Notes: Summary Histotoxic hypoxia was experimentally produced in the rat brain by sodium azide intoxication. In spite of the absence of light microscopic alterations, electron micrographs showed the early changes in the cerebral cortex and subcortex which consisted of widening of the intercellular space and swelling of the pericapillary astrocytic end-feet. However, ferritin-albumin mixture, used for the tracer of protein transport, did not show any increase in the capillary permeability. Sodium azide was considered to disturb the water metabolism, producing osmotic unbalance in the brain.

Bei jedem vergifteten Tier wurde die Feinstruktur des Gehirns elektronenmikroskopisch untersucht. Die Befunde waren folgende: 1. Verschmelzung der Membranen zwischen Axonen in den Nervenendigungen sowie zwischen Axonen und Dendriten wurden in der Großhirnrinde, im limbischen System, im Thalamus und Hypothalmus beobachtet; ferner wurde eine sich vom Cytoplasma bis zur Nervenendigung erstreckende Membranstruktur in diesen Hirnbereichen festgestellt. 2. Im Frontal-, Parietal-, Temporal- und Occipitalhirn sowie im limbischen System fanden sich vermehrt membranbegrenzte Vesikel im Cytoplasma der Nervenzellen und verdicktes und vermehrtes ER in den Nervenfasern. In Anbetracht klinischer Symptome der Patienten mit Amphetaminvergiftung wurde angenommen, daß die Verschmelzung von Membranen zwischen Nervenfortsätzen und das Auftreten der Membranstrukturen in allen Teilen des Nervenzellcytoplasmas vom Perikaryon bis zur Nervenendigung die wichtigsten Befunde sind, die darauf hindeuten, daß die Vergiftungsvorgänge einen wesentlichen Einfluß auf das Leitungssystem der Nerven ausüben können.
    Notes: Summary In 8 male albino strain guinea pigs, 1 mg/kg of methamphetamine HCl was injected daily for 7 months to 1 year to produce chronic methamphetamine intoxication and to study the fine structure of the brain electron microscopically. The following results were obtained: 1. Coalescence of membranes, between axons at the nerve endings and between axons and dendrites, was found in the cortex of the cerebrum and limbic system, in the thalamus and hypothalamus. Membranous structures were found in all portions of the cytoplasm of the nerve cells. 2. Within the cytoplasm of the nerve cell, coated vesicles were increased in number with hypertrophy and increase of endoplasmic reticulum within nerve fibres in the frontal, parietal, temporal and occipital regions, and in the limbic system. In view of these findings, as well as the clinical neurological symptoms in patients with methamphetamine poisoning, the coalescence of membranes between nerve processes and the appearance of membranous structures in all portions of nerve cell cytoplasm from body to nerve endings appear to represent the most important findings, they probably produce a profound disturbance in the system of nerve conduction and transmission.

Die Untersuchungen erfolgten 12 und 120 Std nach einer örtlichen Quetschungsverletzung. Die ersten bemerkbaren Veränderungen finden sich im Axon. Nodale und paranodale Anhäufungen von Mitochondrien, multivesikulären und lamellären Körpern, wie auch kleinen blasen- und röhrenartigen Bildungen sind teilweise in den Knoten sichtbar und am deutlichsten nach 24–36 Std erkennbar. Gleichzeitig erfolgt eine Aufsplitterung der Neurofilamente und Neurotubuli, die ihre Liniengestaltung verlieren und sich zusammenballen. Die Zone der erhöhten Dichte gerade unterhalb des nodalen Axolemmas bleibt erhalten. Veränderungen im Myelin beginnen etwas später und bestehen in einem vesikulären Verfall der Endomyelinlamellen und einer Trennung der Endomyelinschlaufen vom Axolemma durch Schwannzellenfortsätze. Dieser Vorgang schien mit einem Zurückziehen des Myelins vom Knoten im Zusammenhang zu stehen. Schwannzellenfortsätze erstrecken sich auch so weit, daß sie das nodale Axon bedecken, wobei sie die nodalen Schwannzellenfortsätze vom Axolemma trennen. Das Endstadium ist die Unterbrechung des nodalen Axons und die Verschmelzung der Myelinenden als Teil der Ovoidbildung.
    Notes: Summary Observations are reported on the ultrastructural alterations at the nodes of Ranvier in the rat sural nerve during the course of Wallerian degeneration. These were examined between 12 and 120 hours after a localized crush injury. The earliest detectable changes are in the axon. Nodal and paranodal accumulations of mitochondria, multivesicular bodies, lamellar bodies and small vesicular and tubular profiles are seen at a proportion of the nodes and are most evident at 24–36 hours. Concomitantly with this, the neurofilaments and neurotubulus fragment, lose their alignment and clump together. The zone of increased density just beneath the nodal axolemma is preserved. Changes in the myelin begin slightly later and consist of vesicular breakdown of the terminal myelin lamellae, and separation of the terminal myelin loops from the axolemma by Schwann cell processes. The latter event appeared to be associated with retraction of the myelin from the node. Schwann cell processes also extend to cover the nodal axon, separating the Schwann cell nodal processes from the axolemma. The final stage is the interruption of the nodal axon and the fusion of the ends of the myelin as part of ovoid formation.

    Description / Table of Contents: Résumé Dans la leucodystrophie métachromatique, les lipides anormaux s'accumulent dans les lysosomes des cellules gliales et des phagocytes. Les inclusions lipidiques les moins organisées sont les plus riches en phosphatases acides. Celles dont l'ultrastructure est prismatique ont une activité enzymatique très faible et semblent plus proches de corps résiduels que de lysosomes actifs. Il semble que l'absence d'arylsulfatase lysosomiale induise la persistance des sulfatides accumulés dans les lysosomes alors que les autres lipides provenant de la dégénérescence myélinique, seraient normalement catabolisés. Un remaniement progressif de l'ultrastructure du contenu des lysosomes en est la conséquence.
    Notes: Summary In metachromatic leucodystrophy abnormal lipids are stored within the lysosomes of the glial cells and phagocytes. The lipid inclusions which are the less structured are the richest in acid phosphatases. Those of which the ultrastructure is prismatic show a very low enzymatic activity and are more similar to residual bodies than to active lysosomes. It appears that the lack in lysosomal arylsulfatase leads to an accumulation of the sulfatide moiety of the stored lipids while the other lipids originating from the myelin breakdown, would be normally catabolized. This leads to a progressive transformation of the ultrastructure of the lysosome contents.

    Description / Table of Contents: Zusammenfassung Groß- und Kleinhirnrinde, Plexus chorioideus und Rückenmark von Mäusen wurden nach intracerebraler Inoculation einer Hirnemulsion mit Virus der Encephalitis japonica (JEV) elektronenoptisch untersucht, um den Ort der JEV-Replikation zu bostimmen. 72 Std nach der Inoculation bei Beginn der encephalitischen Symptome enthielten 70 bis 80% aller Rindenneurone und Vorderhornzellen viele sphärische Partikel, die meist im zarten endoplasmatischen Reticulum (EPR) und vereinzelt im granulären EPR lokalisiert waren. Die Einzelpartikel zeigten eine gleichförmige Substruktur aus einem elektronendichten zentralen Hof von 25–30 mμ Durchmesser, einer äußeren, weniger elektronendichten Zone und einer äußersten Grenzmembran von 40 mμ Durchmesser. 96 Std nach der Inoculation zeigte das Cytoplasma der Rinden- und Vorderhornneurone sehr viele Vacuolen und Vesiceln. Partikel wurden weit verstreut in den Vacuolen und Vesiceln sowie erstmals im ERP der Sternzellen und Purkinjezellen angetroffen, allerdings in geringerer Zahl. Keine derartigen Partikel wurden in Kontrolltieren und normalen Mäusegruppen angetroffen. Sogenannte eosinophile intranucleäre Einschlüsse in Epithelzellen des Plexus chorioideus zeigten keine derartigen Partikel im Kern oder in den cytoplasmatischen Bläschen. Nachdem keine als JEV identifizierbaren Partikel in Glia- und Endothelzellen nachzuweisen waren, wird angenommen, daß das JEV echt neurotrop ist und sich im EPR der Nervenzellen repliziert.
    Notes: Summary Cerebral and cerebellar cortices, choroid plexus and spinal cord of mice, inoculated intracerebrally with a brain emulsion containing Japanese encephalitis virus (JEV), were studied electronmicroscopically to determine the cell type and the site of JEV replication. 72 hours after inoculation, when the mice began to show encephalitic symptoms, 70 to 80% of all cortical neurons and anterior horn cells contained many spherical particles mostly located in the smooth endoplasmic reticulum and a few in the granular endoplasmic reticulum. The individual particles demonstrated an uniform substructure consisting of an electron dense central core of 25–30 mμ diameter, an outer less electron dense zone and an outermost limiting membrane of 40 mμ diameter. 96 hours after inoculation, the cytoplasm of cortical neurons and anterior horn cells was observed to contain very many vacuoles and vesicles. Particles were found widely scattered throughout the vacuoles and vesicles, and were observed for the first time in the endoplasmic reticulum of the stellate neurons and in Purkinje cells, though fewer. No such particles were observed in control and normal mouse groups. So-called eosinophic intranuclear inclusions of epithelial cells of choroid plexus failed to show any particles in their nuclei or cytoplasmic vesicles. Considering that no particulate matter, identifiable as JEV, was identified within any of the glial cells or endothelium in this examination, it was concluded that JEV was really neurotropic and replicates in the endoplasmic reticulum of the neurons.

L'aspect des stries Z, en section transversale, varie en effet avec la fixation: réseau de ponts «obliques» (par rapport à l'alignement quadratique régulier des terminaisons des filaments fins) après fixation osmiée, réseau de ponts «transversaux», isolé ou superposé au premier réseau, après fixation aldéhydique initiale. L'aspect des bâtonnets en section transversale varie également avec la fixation; la disposition quadratique des filaments constitutifs des bâtonnets est visible après l'une et l'autre fixation, mais un réseau quadratique régulier, de 75 Å de côté, n'est clairement apparent qu'après fixation aldéhydique. Le point important est que ce dernier aspect diffère complètement du réseau «oblique» des stries Z normales, et peut être rapproché au contraire du réseau «transversal» de la strie Z obtenu après fixation aldéhydique; l'hypothèse est ainsi soulevée de la constitution des ponts dans les bâtonnets aux dépens d'un seul des deux constituants morphologiques de la strie Z. La topographie des bâtonnets a été également étudiée: tantôt périphérique, au sein de masses sarcoplasmiques latérales, tantôt centrale, au sein de petits foyers de désintégration myofibrillaire. La structure des plaques motrices visibles dans les préparations était normale. La spécificité de la lésion élémentaire et l'autonomie de ce type de myopathie sont ensuite discutées.
    Notes: Summary Ultrastructural study of an observation of “Nemaline Myopathy” has been focused on structural relationships between rods and normal Z-bands in different conditions of fixation. The Z-band structure studied in transversal section, varies with the fixation: network of “oblique” (relative to the regular quadratic pattern of the thin filament endings) bridges after osmic fixation; network of “transversal” bridges, alone or associated to the first network, after aldehydic fixation. The rod structure, in transversal section, is also varying with the fixation: a quadratic disposal of the filamentary content is visible after both fixations, but a regular quadratic network, 75–85 Å side, is clearly appearent only after aldehydic fixation. This last network differs from the “oblique” network of the normal Z-band; it can be compared to the “transversal” one obtained in the Z-band after aldehydic fixation. The hypothesis that bridges in the rods are formed only by one of the two normal morphological components of the Z-band, is raised. Topography of the rods in muscle fibers has been studied: either in peripheral sarcoplasmic masses, or in small central-located foci of myofibrillar disintegration. The end-plates structure was normal. Specificity of the elementary lesion, and autonomy of that type of myopathy is discussed.

    Description / Table of Contents: Zusammenfassung 17 chirurgisch entfernte Meningiome vom endotheliomatösen und 5 vom fibromatösen Typ wurden elektronenmikroskopisch untersucht. Unterschiede im Entwicklungsgrad der cytoplasmatischen Fortsätze und der Beziehungen zwischen den Plasmamembranen der Tumorzellen wurden in den endotheliomatösen Meningeomen beobachtet. In 4 fibromatösen Tumoren sind die zelligen Areale von bindegewebigen Räumen umgeben. Auch wenn die meisten Tumorzellen helle Zellen sind und sich nicht wesentlich von den Tumorzellen der endotheliomatösen Meningiome unterscheiden, ist die histologische Anordnung der fibromatösen klar von derjenigen der endotheliomatösen Meningeome zu trennen. Ausschlaggebend dafür ist die starke Entwicklung des Bindegewebes. Die Dunkelzellen lassen sich in zwei Typen gliedern: Typ I wird in vier Tumoren mit stark ausgebildetem Bindegewebe angetroffen. Die Analyse der inneren Zellstruktur sowie das Vorliegen von Übergängen zwischen hellen und dunklen Zellen machen es wahrscheinlich, daß diese nur verschiedene Typen einer einzigen Ursprungszelle darstellen. Die Bedeutung dieser verschiedenen Erscheinungsformen der Meningiomzellen wird diskutiert. Die Ultrastruktur der Tumorzellen ähnelt derjenigen der normalen arachnoidalen Zellen. Rückschlüsse auf die blastodermale Herkunft der normalen und tumoralen meninigalen Zellen auf Grund ihrer ultrastrukturellen Merkmale erscheinen verfrüht.
    Notes: Summary Seventeen surgically removed meningiomas of the endotheliomatous and five of the fibromatous type were investigated with an electron microscope. Differences of the development of cytoplasmic processes and the relations between plasma membranes of blastomatous cells were observed in endotheliomatous meningiomas. In four of the fibromatous tumors the cell groups were surrounded by spaces of connective tissue. Although most of the tumor cells were light cells and are not essentially different from the tumor cells of the endotheliomatous meningiomas, the histological structure of fibromatous meningiomas is clearly distinguished from the endotheliomatous type, because of the greater amount of connective tissue. The dark cells may be divided into two types: the first was found in those four tumors, where the connective tissue is well developed, while the second one only occurred in one of the tumors. The analysis of the inner cell structure as well as the presence of interstages between dark and light cells makes it probable that dark and light cells are different types of one original cell. The cause of this different appearance of the menigioma cells is discussed. The fine structure of the tumor cells showed a great similarity with that of normal arachnoidal cells. Conclusions on the blastodermic origin of normal and blastomatous meningial cells on the basis of ultrastructural characteristics, however, seem to be premature.

Die Tiere wurden am 1., 3., 5., 8., 14. 21. und 28. Tag nach der Operation getötet. Das Gebiet des epileptogenen Herdes wurde elektronenmikroskopisch untersucht. Die frühesten Veränderungen bestanden in ödem. Später traten Veränderungen der Astrocytenfortsätze auf, die sich an die Basalmembranen der Capillaren anlegten; die Intercellularräume erweiterten sich, füllten sich mit elektronenoptisch dichten Massen und entwickelten ein “plasmatisches Infiltrat”. Allmählich breitete sich dieses Exsudat weiter um die Capillaren aus; es trat an Orten auf, wo die Astrocytenfortsätze und die Dendriten schwer geschädigt waren und erfüllte die weiteren Intercellularräume des Neuropils. Die Nervenzellen zeigten eine auffallende Widerstandsfähigkeit gegenüber diesem pathologischen Prozeß. Manchmal waren sie umschlossen von Exsudatmassen und zum Teil oder ganz der zarten neuralen und Astrocytenfortsätze beraubt, von denen sie normalerweise umgeben sind. Unsere Befunde werden als das ultrastrukturelle Korrelat der Schols'schen “plasmatischen Infiltration” interpretiert.
    Notes: Summary In 25 adult rats an epileptogenic cobalt-gelatine focus was produced in a standard region of the cerebral cortex. The animals were killed successively on the 1st, 3rd, 5th, 8th, 14th, 21st and 28th day after operation and the region of the epileptogenic focus was examined electron microscopically. The earliest changes were those of oedema. Later there occurred disturbances of the astrocytic processes which cling to the basement membranes of the capillaries; the intercellular spaces widened and filled with rather E. M. dense masses and a “plasmatic infiltrate” developed. Gradually this exudative substance spread even further from the capillaries. It appeared in places where the astrocytic processes were severely demaged and the dendrites were partly dissipated and it filled out the wide intercellular spaces of the neuropil. The nerve cells showed a striking resistance toward this pathological process. Sometimes they floated in the exudative masses but were partly or totally deprived of the fine neural and astrocytic processes which normally surround them. Our findings are interpreted as the ultrastructural correlate of Scholz's “plasmatic infiltration”.

    Description / Table of Contents: Résumé On a étudié la formation de l'hydroxyapatite de calcium cristallin à partir de solutions d'ions de calcium et de phosphate et l'inhibition de la croissance de cristaux de l'hydroxyapatite de calcium au moyen de polyphosphonates et de polyphosphates. Les polyphosphonates, éthane hydroxy-1-diphosphonate-1,1 de disodium et dichlorométhanediphosphonate de disodium, sont inhibiteurs efficaces contre la croissance de cristaux de l'hydroxyapatite de calcium. Les polyphosphates sont aussi inhibiteurs efficaces contre la croissance de cristaux de l'hydroxyapatite de calcium tant que le niveau exigé de polyphosphate intact est présent dans le système. Cependant, à cause de leur instabilité hydrolytique, qui est soulignée par une température élevée, valeur de pH basse, et certaines enzymes, la concentration du polyphosphate diminue avec le tempsin vitro, et son activité comme inhibiteur est perdue. Au contraire aux polyphosphates, les polyphosphonates sont hydrolytiquement stables. Les polyphosphonates sont chimiosorbés sur la surface des microcristallites de l'hydroxyapatite de calcium, ainsi empêchant l'occurrence d'autre croissance de cristaux semblable à l'action d'autres poisons connus de croissance de cristaux. On propose l'extension de cette action sur la formation de l'apatite et cette stabilité des polyphosphonates aux applications médicales et dentaires concernant le metabolisme pathologique de calcium et de phosphate.
    Abstract: Zusammenfassung Die Bildung des kristallinen Calciumhydroxyapatit aus Lösungen, welche Calcium- und Phosphationen enthalten, und die Hemmung der Bildung von kristallinen Calciumhydroxyapatit durch Polyphosphonate und Polyphosphate wurden untersucht. Polyphosphonate, Dinatriumäthan-1-hydroxyl-1,1-diphosphonat und Dinatriumdichloromethandiphosphonate verhindern das Kristallwachstum des Calciumhydroxyapatits. Die Polyphosphate verhindern ebenfalls das Kristallwachstum des Calciumhydroxyapatits, solange die notwendige Konzentration des nicht hydrolysierten Polyphosphats vorhanden ist. Wegen ihrer hydrolytischen Unbeständigkeit, die durch hohe Temperatur, niedrige pH und bestimmte Enzyme erhöht wird, vermindert sich jedoch die Konzentration des Polyphosphats allmählichin vitro, und ihre Hemmungsaktivität geht verloren. Im Gegensatz zu den Polyphosphaten sind die Polyphosphonate hydrolytisch beständig. Die Polyphosphonate werden an der Oberfläche der Mikrokristallite des Calciumhydroxyapatits chemisorbiert und verhindern, wie andere bekannte Kristallwachstumsgifte, auf diese Weise weiteres Kristallwachstum. Die Beständigkeit der Polyphosphonate und ihre Chemisorption an dem Apatit empfehlen ihren Gebrauch in der ärztlichen und zahnärztlichen Praxis, soweit sie den pathologischen Calcium- und Phosphatstoffwechsel betreffen.
    Notes: Abstract The formation of crystalline calcium hydroxyapatite from solutions of calcium and phosphate ions and the inhibition of calcium hydroxyapatite crystal growth by polyphosphonates and polyphosphates have been studied. The polyphosphonates, disodium ethane-1-hydroxy-1,1-diphosphonate and disodium dichloromethane diphosphonate, are effective inhibitors of calcium hydroxyapatite crystal growth. The polyphosphates are also effective inhibitors of calcium hydroxyapatite crystal growth as long as the required level of intact polyphosphate is present in the system. However, because of their hydrolytic instability, which is enhanced by high temperature, low pH, and certain enzymes, the concentration of the polyphosphate decreases with timein vitro, and its activity as an inhibitor is lost. In contrast to the polyphosphates, the polyphosphonates are hydrolytically stable. The polyphosphonates are chemisorbed on the surface of the microcrystallites of calcium hydroxyapatite and, in the manner of other known crystal growth poisons, thus prevent further crystal growth. The stability of the polyphosphonates and their chemisorption on apatite suggest their use in medical and dental applications involving pathological calcium and phosphate metabolism.

Membranovesiculäre Körperchen finden sich ebenfalls in den Nervenzellperikaryen; typische granuläre Lipofuscinkörperchen werden aber nicht gesehen. Die Astrocyten der Rinde enthalten eine ähnliche Art von Einschlüssen, von denen aber die Einschlüsse in den Astrocyten der Marksubstanz völlig verschieden sind. Die Lokalisation der Aktivität von saurer Phosphatase in allen Einschlußtypen läßt an ihre mögliche lysosomale Natur denken. Die biochemische Analyse deckt keine spezifischen Abweichungen auf.
    Notes: Summary Electron microscopic, histochemical and biochemical studies of a case of juvenile lipidosis have been presented. The neuronal inclusions were mostly unit membranebound with fingerprint-like patterns. Membrano-vesicular bodies were also found in the neuronal perikarya, but “typical” granular lipofuscin bodies were not seen. Cortical astrocytes contained a similar type of inclusion but the inclusions in the astrocytes in the white matter were quite different. The localization of acid phosphatase activity in all types of inclusions suggests their possible lysosomal nature. Biochemical analysis failed to pin-point any specific abnormalities.

Elektronenoptisch bestanden die Einschlußkörper aus einer Zusammenballung von körnigen Gebilden, die in einem unregelmäßigen Netz von vermehrten Neurofilamenten verstreut lagen. Viele eosinophile Hiranosche Körper fanden sich in nächster Nachbarschaft von Nervenzellen. Diese Körper bestanden aus verflochtenen Schichten von geperlten oder glatten Filamenten. Die mögliche Lokalisation der Hiranoschen Körper innerhalb von degenerierenden Axonendigungen wird diskutiert.
    Notes: Summary Brain tissue from an autopsied case of Pick's disease was studied by light and electron microscopy. Intraneuronal argentophilic Pick bodies were most numerous in the pyramidal cells of the hippocampus. By electron microscopy, these inclusions consisted of a conglomeration of granules enmeshed in an irregular network of proliferated neurofilaments. There were also multiple eosinophilic juxtaneuronal structures (Hirano bodies). By electron microscopy these consisted of interwoven sheaves of beaded and continuous filaments. The possible location of the Hirano bodies within degenerating axonal terminals is discussed.

    Notes: Summary Electron microscopic post mortem examination of the lenticular nuclei of two human cases who died in hepatogenic coma is reported. Lesions were found to be swelling and vacuolation of astrocytes. In addition an excess of lipofuscin and lipid like substances were present in the same cells. No changes were found in myelinated fibres, neurons and small blood vessels.

Es wird angenommen, daß diese Veränderungen durch segmentale Demyelinisation verursacht wurden.
    Notes: Summary Observations have been made on 10 consecutive nerve biopsies from patients with diabetic neuropathy. 1 patient showed the typical appearances of hypertrophic neuropathy on light and electron microscopy. 5 displayed typical hypertrophic changes visible only on electron microscopy and minor abnormalities of a similar nature were seen in 2 others. It was considered that they were likely to have resulted from recurrent segmental demyelination.

    Description / Table of Contents: Zusammenfassung Histochemische und elektronenoptische Befunde eines Patienten mit hypertrophischer Neuritis von 25 Jahren Dauer mit Beginn der Erkrankung in der Kindheit werden mitgeteilt. Die erhobenen Befunde sowie andere jüngste Beobachtungen bilden die Grundlage einer Diskussion der verschiedenen pathogenetischen Hypothesen der “Zwiebelschalen”-Strukturen in den Nerven von Patienten mit hypertrophischer Neuritis. Die Zwiebelschalenbildung wird als unspezifische Manifestation einer chronischen segmentalen Entmarkung infolge primärer oder erworbener Anomalie der Schwann-Zellen interpretiert.
    Notes: Summary Histochemical and electron microscopic data are presented from a patient with hypertrophic neuritis of 25 years duration with onset in childhood. The present observations and those of recent investigators form the basis of a discussion of various pathogenetic hypotheses for “onion bulb” structures in the nerves from patients with hypertrophic neuritis. It is concluded that the onion bulb lesion is a nonspecific manifestation of chronic segmental demyelinization resulting from a primary or acquired abnormality of Schwann cells.

    Description / Table of Contents: Zusammenfassung Die Köpfe von insgesamt 17 Meerschweinchen beiderlei Geschlechts wurden mit 17 MeV-Betastrahlen homogen bestrahlt. Die Einzeitbestrahlungen erfolgten mit Dosiswerten von 500–5000 R und einer Dosisleistung von 100 R pro Minute. Sämtliche Tiere wurden 24 Std nach der Bestrahlung getötet. Durch PAS- und Astrablau-Reaktionen konnten Globuli, die Mucoproteide und saure Mucopolysaccharide enthalten und vorzugsweise in der weißen Substanz vorkommen, nachgewiesen werden. Die regionäre Verteilung ist von der Dosierung abhängig. Von 2000 R an finden sich in der marknahen Großhirnrinde und Medulla oblongata geschrumpfte und nekrotische Ganglienzellen. Ebenso verhalten sich die Purkinje-Zellen. Diese Veränderungen werden im ultrastrukturellen Bereich bestätigt. Das Ödem findet sich in den Astrocyten; der extracelluläre Raum der grauen Substanz des Cortex ist nicht erweitert.
    Notes: Summary The heads of 17 guinea pigs of both sexes were irradiated homogenously by 7 MeV-beta irradiation. The dosage of the one-time irradiation was 500–5000 R with a dosage of 100 R/minute. All the animals were killed 24 hrs after the irradiation. By means of PAS- and Astrablue reactions globuli containing mucoproteids and acid mucopolysaccharides could be detected mainly in the white matter. Their regional pattern depends on the dosage. Beginning from 2000 R, shrinked and necrotic nerve cells are seen in the deep cerebral cortex and medulla oblongata. Similar lesions are seen in the Purkinje cells. These lesions are confirmed by ultrastructural findings. Edema is found in astrocytes. The extracellular spaces of the cortical gray matter are not enlarged.

    Description / Table of Contents: Zusammenfassung Das basische Polypeptid Melittin bewirkt die Freisetzung von β-Glucuronidase, Kathepsin, saurer Phosphatase und alkalischer Phosphatase aus isolierten lysosomalen Granula polymorphkerniger neutrophiler Leukocyten und aus intakten Leukocyten vom Rind. An polymorphkernigen Leukocyten setzt Melittin außerdem Lactat-Dehydrogenase und Glutamat-Pyruvat-Transaminase frei. Das mitochondriale Enzym (GPT) wird zeitlich später solubilisiert als Kathepsin. Elektronenoptisch zeigen isolierte Granula nach Einwirkung von Melittin Verringerung des Durchmessers, Verdichtung der Matrix und Membranbrüche. Intakte Zellen werden degranuliert. Es kommt zur Auflösung lysosomaler Granula in cytoplasmatischen Vacuolen. Ein Mastzellen-lysierendes Polypeptid aus Bienengift war in Konzentrationen bis zu 2 · 10−5 M/l an isolierten Granula unwirksam. Mit Hilfe der Änderung der Lichtdurchlässigkeit von Granulasuspensionen wurden andere basische Polymere auf ihre membranschädigende Wirkung untersucht. Ihre Wirkungsstärke war von der Basizität und der Kettenlänge abhängig.
    Notes: Summary The cationic polypeptide Melittin effects the liberation of acid and alkaline phosphatase, β-glucuronidase and cathepsin from isolated lysosomal granules of polymorphonuclear neutrophilic bovine leucocytes as well as from intact leucocytes. In addition Melittin releases LDH and Glutamate-Pyruvate-Transaminase. The mitochondrial enzyme (GPT) is solubilized later than cathepsin. Electron microscopy of isolated granules, after treatment with Melittin shows a reduction of their diameter, an increase in density of the matrix and membrane fractures. Intact cells are degranulated and there occurs a lysis of lysosomal granules in cytoplasmic vacuoles. Another mast cell-degranulating polypeptide from bee venom in concentrations up to 2×10−5 M/l was ineffective on isolated granules. Other cationic polymers were studied with regard to their membrane-damaging effect by means of changes in optical density of suspended granules. The intensity of their effect was dependent on the number of cationic charges and on chain length.

Elektronenoptisch kommt es nach Einwirkung von Mersalyl zur Vacuolisierung und Abnahme der elektronenoptischen Dichte der Matrix. Höhere Konzentrationen führen zur Ruptur der Granulamembran. Blockiert man in der Membran von intakten Granula 1 Mol SH pro 22000 g Granulaprotein mit p-Chlormercuribenzoat, so kommt es zur Erhöhung der Permeabilität der Partikel für Enzymproteine.
    Notes: Summary A number of mercury sulfhydryl-reagents act on isolated lysosomal granules of bovine polymorphonuclear leucocytes in concentrations of 10−3 to 10−7 M/l to cause a decrease of optical density and the liberation of acid phosphatase, cathepsin and cationic polypeptides. Electron microscopy of isolated granules after treatment with mersalyl showed a decrease in electron density of the matrix and vacuolisation. Concentrations up to 10−3 M/l destroyed the particles and ruptured their membranes. The number of sulfhydryl groups was estimated at 1 Mol SH per 22000 g of granule protein. Blocking these groups with p-chlormercuribenzoate causes an increase in membrane permeability for enzymes of the lysosomal particles.
